Objective:

To establish a method and validate its feasibility for quality evaluation of Erycibes Caulis and its processed products,and analyze the effect on the contents of the seven active ingredient before and after processing.

Methods:

The contents of components were determined by ultra performance liquid chromatography (UPLC). Chlorogenic acid was chosen as the internal reference substances,the relative correction factors (RCFs) of neochlorogenic acid scopolin,scopoletin,isochlorogenic acid B,isochlorogenic acid A,isochlorogenic acid C and to chlorogenic acid were established. The contents of these seven active ingredients were determined by external standard method and quantitative analysis of multi-components by single-marker (QAMS) method. The method was evaluated by comparison of the quantitative results between external standard method and quantitative analysis of multi-components by single-marker (QAMS).

Results:

The peaks of neochlorogenic acid,scopolin,chlorogenic acid,scopoletin,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in the sample showed good linear relationship between 0.028-1.420 μg·μL-1,0.019-0.956 μg·μL-1,0.027-1.324 μg·μL-1,0.014-0.720 μg·μL-1,0.017-0.824 μg·μL-1,0.010-0.500 μg·μL-1 and 0.013-0.672 μg·μL-1,respectively. The average recoveries of them(n=6)were as follows 98.9%,99.0%,100.6%,101.2%,100.8%,101.7% and 100.5%,with the RSDs of 1.1%,1.7%,1.5%,1.6%,0.55%,1.6% and 1.5%,respectively. It was showed that no significant difference was found in the quantitative results of seven ingredients by external standard method and QAMS method. The content of 5 kinds of organic acids (neochlorogenic acid,chlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C) in Erycibes Caulis with different processing technology showed the same change trend. The sample of Y5 (boiled products with licorice sauce and brine) had the highest contents,and the sample of Y10 (dried products Ⅱ soaked in licorice sauce and brine) had the lowest. The content of scopoline was the highest in Y5(3.53 mg·g-1),while the lowest was Y10(0.31 mg·g-1). The content of scopoletin in Y7(dried products Ⅰ with licorice sauce) was the highest(1.48 mg·g-1) and that in Y3(boiled products Ⅱ with licorice sauce) was the lowest(0.30 mg·g-1).

Conclusion:

The RCFs established in the QAMS methods with chlorogenic acid as the internal reference substances is accurate and feasible. It can be used to control the quality of Erycibes Caulis. Both heating and excipients preparation have a certain effect on the active ingredients in Erycibes Caulis.

目的:

建立一测多评法同时测定丁公藤及其炮制品中7个活性成分含量,并分析炮制对其影响。

方法:

采用超高效液相色谱法(UPLC),以绿原酸为参照物,建立新绿原酸、东莨菪苷、东莨菪内酯、异绿原酸B、异绿原酸A和异绿原酸C的相对校正因子。分别采用外标一点法和一测多评法测定上述7个活性成分含量,比较计算值与实测值的差异,以验证一测多评法的准确性和和可行性。

结果:

新绿原酸、东莨菪苷、绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C分别在0.028~1.420、0.019~0.956、0.027~1.324、0.014~0.720、0.017~0.824、0.010~0.500和0.013~0.672 μg·μL-1范围内呈现良好线性关系,平均加样回收率(n=6)分别为98.9%、99.0%、100.6%、101.2%、100.8%、101.7%和100.5%,RSD分别为1.1%、1.7%、1.5%、1.6%、0.55%、1.6%和1.5%。采用校正因子计算的7个活性成分含量值与外标法实测值之间无显著差异。不同丁公藤饮片中新绿原5个有机酸(新绿原酸、绿原酸、异绿原酸B、异绿原酸A、异绿原酸C)含量变化趋势基本一致,Y5(甘草汁-盐水煮品)含量最高,Y10(甘草汁-盐水晒品Ⅱ)含量最低,东莨菪苷含量以Y5含量最高(3.53 mg·g-1),Y10最低(0.31 mg·g-1)。东莨菪内酯含量以Y7(甘草汁晒品Ⅰ)含量最高(1.48 mg·g-1),Y3(甘草汁煮品Ⅱ)含量最低(0.30 mg·g-1)。

结论:

以绿原酸为参照物建立的一测多评法可用于丁公藤饮片质量评价,加热和加辅料炮制均对丁公藤的化学成分产生了一定影响。

目的:

建立一测多评法同时测定丁公藤及其炮制品中7个活性成分含量,并分析炮制对其影响。

方法:

采用超高效液相色谱法(UPLC),以绿原酸为参照物,建立新绿原酸、东莨菪苷、东莨菪内酯、异绿原酸B、异绿原酸A和异绿原酸C的相对校正因子。分别采用外标一点法和一测多评法测定上述7个活性成分含量,比较计算值与实测值的差异,以验证一测多评法的准确性和和可行性。

结果:

新绿原酸、东莨菪苷、绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C分别在0.028~1.420、0.019~0.956、0.027~1.324、0.014~0.720、0.017~0.824、0.010~0.500和0.013~0.672 μg·μL-1范围内呈现良好线性关系,平均加样回收率(n=6)分别为98.9%、99.0%、100.6%、101.2%、100.8%、101.7%和100.5%,RSD分别为1.1%、1.7%、1.5%、1.6%、0.55%、1.6%和1.5%。采用校正因子计算的7个活性成分含量值与外标法实测值之间无显著差异。不同丁公藤饮片中新绿原5个有机酸(新绿原酸、绿原酸、异绿原酸B、异绿原酸A、异绿原酸C)含量变化趋势基本一致,Y5(甘草汁-盐水煮品)含量最高,Y10(甘草汁-盐水晒品Ⅱ)含量最低,东莨菪苷含量以Y5含量最高(3.53 mg·g-1),Y10最低(0.31 mg·g-1)。东莨菪内酯含量以Y7(甘草汁晒品Ⅰ)含量最高(1.48 mg·g-1),Y3(甘草汁煮品Ⅱ)含量最低(0.30 mg·g-1)。

结论:

以绿原酸为参照物建立的一测多评法可用于丁公藤饮片质量评价,加热和加辅料炮制均对丁公藤的化学成分产生了一定影响。

Objective:

To establish a method and validate its feasibility for quality evaluation of Erycibes Caulis and its processed products,and analyze the effect on the contents of the seven active ingredient before and after processing.

Methods:

The contents of components were determined by ultra performance liquid chromatography (UPLC). Chlorogenic acid was chosen as the internal reference substances,the relative correction factors (RCFs) of neochlorogenic acid scopolin,scopoletin,isochlorogenic acid B,isochlorogenic acid A,isochlorogenic acid C and to chlorogenic acid were established. The contents of these seven active ingredients were determined by external standard method and quantitative analysis of multi-components by single-marker (QAMS) method. The method was evaluated by comparison of the quantitative results between external standard method and quantitative analysis of multi-components by single-marker (QAMS).

Results:

The peaks of neochlorogenic acid,scopolin,chlorogenic acid,scopoletin,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in the sample showed good linear relationship between 0.028-1.420 μg·μL-1,0.019-0.956 μg·μL-1,0.027-1.324 μg·μL-1,0.014-0.720 μg·μL-1,0.017-0.824 μg·μL-1,0.010-0.500 μg·μL-1 and 0.013-0.672 μg·μL-1,respectively. The average recoveries of them(n=6)were as follows 98.9%,99.0%,100.6%,101.2%,100.8%,101.7% and 100.5%,with the RSDs of 1.1%,1.7%,1.5%,1.6%,0.55%,1.6% and 1.5%,respectively. It was showed that no significant difference was found in the quantitative results of seven ingredients by external standard method and QAMS method. The content of 5 kinds of organic acids (neochlorogenic acid,chlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C) in Erycibes Caulis with different processing technology showed the same change trend. The sample of Y5 (boiled products with licorice sauce and brine) had the highest contents,and the sample of Y10 (dried products Ⅱ soaked in licorice sauce and brine) had the lowest. The content of scopoline was the highest in Y5(3.53 mg·g-1),while the lowest was Y10(0.31 mg·g-1). The content of scopoletin in Y7(dried products Ⅰ with licorice sauce) was the highest(1.48 mg·g-1) and that in Y3(boiled products Ⅱ with licorice sauce) was the lowest(0.30 mg·g-1).

Conclusion:

The RCFs established in the QAMS methods with chlorogenic acid as the internal reference substances is accurate and feasible. It can be used to control the quality of Erycibes Caulis. Both heating and excipients preparation have a certain effect on the active ingredients in Erycibes Caulis.

丁公藤是2020年版《中华人民共和国药典》(简称《中国药典》)收载品种,为旋花科植物丁公藤Erycibe obtusifolia Benth.或光叶丁公藤Erycibe schmidtii Craib的干燥藤茎[1],其性味辛、温,有小毒。具有祛风除湿,消肿止痛功效,在许多地区多以配制酒剂,内服或外用于临床[2]。在少数民族地区,丁公藤除生用外,还有甘草汁-盐水浸泡、蒸后晒干炮制方法,且记载经炮制可降低毒性[3]。近年来对丁公藤的研究逐渐增多,发现丁公藤含香豆素类、生物碱,β-谷甾醇等多个成分[4-8],现代药理研究表明东莨菪苷为其镇痛、抗炎作用的活性成分[49-10],绿原酸具有抗肿瘤、抗病毒和抗菌等活性[11]。在丁公藤的中成药应用方面,有引起剥脱性皮炎、过敏性休克等不良反应报道[12-13],这是否与药材生用或饮片炮制不当有关?或与其所含化学成分变化有关?未见有其炮制方法对化学成分影响的相关报道。在对照品缺乏的情况下,近年采用一测多评法(QAMS),能够大大降低检测成本,故在中药饮片及中成药质量控制应用越来越多[14-17]。因此本试验采用超高效液相色谱法(UPLC),以绿原酸为参照物,运用一测多评法建立新绿原酸、东莨菪苷、东莨菪内酯、异绿原酸B、异绿原酸A和异绿原酸C的相对校正因子,分析比较炮制前后上述7个化学成分含量的变化,以期为丁公藤的炮制方法及临床运用提供科学的理论和实验依据。

对照品:新绿原酸(批号MUST-21030108)、东莨菪苷(批号PS010518)、绿原酸(批号PS08072303)、东莨菪内酯(批号PS010525)、异绿原酸B(批号MUST-21030602)、异绿原酸A(批号MUST-21102611)、异绿原酸C(批号MUST-21081010),纯度均≥98.0%,均由成都普思生物科技股份有限公司提供。
蜂蜜(批号182404Y2),来自上海冠生园蜂制品有限公司;丁公藤(批号17082101),甘草(批号20190807),均购自南宁市一心药店,经广西中医药大学梁子宁教授鉴定丁公藤为旋花科植物丁公藤Erycibe obtusifolia Benth.的干燥藤茎、甘草为豆科植物甘草Glycyrrhiza uralensis Fisch.的干燥根及根茎。
甲醇、乙腈均为色谱纯,来自Fisher公司;水为娃哈哈纯净水,其他试剂均为分析纯。
取甘草100 g,加入10倍蒸馏水,加热至沸腾,保持微沸30 min,过滤,药渣继续加入8倍量蒸馏水,同上法重复操作1次,合并2次所得甘草汁,浓缩至1 000 mL,即得。
取丁公藤样品,除去杂质,洗净,润透,切片,干燥[1],即得。
取丁公藤生品50 g,加入甘草汁30 mL,浸泡24 h,加水500 mL煮4 h至近干,取出晒干,即得。
取丁公藤生品50 g,取“2.1”项甘草汁15 mL,用水按1:1稀释后加入丁公藤中,浸泡24 h,加入水500 mL煮4 h至近干,取出晒干,即得。
取丁公藤生品50 g,加入盐水(即2.5 g盐溶于80 mL水),浸泡24 h,加入500 mL水煮4 h至近干,取出晒干,即得。
取丁公藤生品50 g,加入甘草汁30 mL和盐水(2.5 g盐溶于12.5 mL水),浸泡24 h,加水500 mL煮4 h至近干,取出晒干,即得。
取丁公藤生品50 g,加入盐水(同“2.2.4”),浸泡24 h,晒干,即得。
取丁公藤生品50 g,取“2.1”项甘草汁15 mL,用水按1:1稀释后加入丁公藤中,浸泡24 h,晒干,即得。
取丁公藤生品50 g,加入甘草汁30 mL,浸泡24 h,晒干,即得。
取丁公藤生品50 g,加入甘草汁30 mL,加入盐水(同“2.2.4”),浸泡24 h,晒干,即得。

取丁公藤生品50 g,取“2.1”项甘草汁15 mL,用水按1∶1稀释后加入丁公藤中,浸泡24 h,放入蒸笼内隔水蒸4 h,取出晒干,即得。
取丁公藤生品50 g,取“2.1”项甘草汁15 mL,用水按1∶1稀释后加入丁公藤中,浸泡24 h,炒干,即得。
取丁公藤生品50 g,加入盐水(2.5 g盐溶解并稀释至40 mL),浸泡24 h,炒干,即得。
取丁公藤生品50 g,加入甘草汁15 mL,加入盐水(同“2.2.15”),浸泡24 h,炒干,即得。
色谱柱ACQUITY UPLC BEH-C18柱(50 mm×2.1 mm,1.7 μm),以0.1%磷酸(A)-乙腈(B)为流动相,梯度洗脱(0~4 min,10%B→10%B;4.01~8 min,10%B→15%B;8.01~10 min,15%B→20%B;10.01~16 min,20%B→25%B;16.01~18 min,25%B→45%B;18.01~20 min,45%B→45%B),流速0.2 mL·min-1,检测波长338 nm,柱温是30 ℃,进样量4 μL。在上述的条件下,新绿原酸、东莨菪苷、绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C得到了很好的分离,相邻峰之间的分离度均大于1.5,以绿原酸色谱峰计算理论塔板数≥5 000。结果见图1
精密称取对照品新绿原酸35.5 mg、东莨菪苷23.9 mg、绿原酸33.1 mg、东莨菪内酯18.0 mg、异绿原酸B 20.6 mg、异绿原酸A 12.5 mg、异绿原酸C 16.80 mg,置于同一25 mL棕色量瓶中,加80%甲醇溶解并定容至刻度,即得混合对照品储备液。精密量取混合对照品储备液0.10、0.5、1.0、2.0、3.0、5.0 mL至量瓶中,加80%甲醇定容至5 mL,即得系列混合对照品①~⑥溶液。
取丁公藤粉末(过40目筛)约1.0 g,精密称定,置100 mL磨口锥形瓶中,精密加入80%甲醇25 mL,在90 ℃下加热回流30 min,取出放冷后过滤,滤渣再精密加入80%甲醇25 mL同上法提取1次,合并2次滤液,用旋转蒸发仪浓缩后转移至25 mL量瓶,加80%甲醇定容至刻度,13 000 r·min-1离心10 min,取上清液即得。
分别精密吸取系列混合对照品不同浓度溶液各4 μL,注入高效液相色谱仪,按“2.3”项的色谱方法进行测定,以对照品的进样浓度(X,μg·μL-1)为横坐标,峰面积Y为纵坐标,绘制标准曲线,结果见表1。结果显示,各成分在各自浓度的范围内呈良好的线性关系。
精密吸取“2.4.1”混合对照品溶液中⑥号溶液4 μL,在“2.3”项色谱条件下,连续进样6次,计算各峰峰面积的RSD,结果分别为0.090%、0.030%、3.80%、0.59%、2.90%、1.60%、1.20%,说明试验所用的仪器有良好精密度。
按“2.4.2”项下方法,平行制备6份丁公藤Y11供试品溶液。在“2.3”色谱条件下进行检测,记录样品溶液新绿原酸、东莨菪苷、绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C的峰面积,计算样品各成分的含量。上述7个活性成分含量依次为1.97、0.63、0.73、0.76、4.91、0.44、0.87 mg·g-1,RSD依次为1.9%、1.8%、2.9%、3.3%、1.5%、3.6%、2.2%。证明本研究所建立的方法重复性良好。
精密吸取丁公藤Y11供试品溶液4 μL,按“2.3”项色谱条件,分别在0、1、3、6、12、24 h进样,考察成分的稳定性,结果7个成分峰面积的RSD为2.0%、1.1%、0.79%、1.8%、0.55%、1.3%、1.9%,表明供试品溶液24 h内稳定。
称量已知含量的丁公藤Y9粉末6份,每份约0.5 g,精密称定。采用加样回收试验,分别精密加入适量新绿原酸、东莨菪苷、绿原酸、东莨菪内酯、异绿原酸B、异绿原酸A、异绿原酸C对照品。依“2.4.2”项下的方法来制备供试品溶液,按“2.3”项色谱条件进行测定,平行测定6份,计算加样回收率。结果见表2,表明该方法准确度良好。
试验考察采用Agilent 1290 Infinity Ⅱ、Waters ACQUITY、Nexera X2 LC-30AD 3种超高效液相色谱系统和ACQUITY BEH-C18色谱柱(100 mm×2.1 mm,1.8 μm)、Ultimate XB-C18(100 mm×2.1 mm,1.8 μm)、Shim-pack Velox C18 (100 mm×2.1 mm,1.8 μm)3种色谱柱时的相对校正因子。相对校正因子f=Ai×Cs/As×Ci,式中As为待测成分s的峰面积,Cs为质量浓度;Ai为内参物i的峰面积,Ci为质量浓度。分别计算新绿原酸(A)、东莨菪苷(B)、东莨菪内酯(C)、异绿原酸B(D)、异绿原酸A(E)、异绿原酸C(F)与内参物绿原酸(S)的相对校正因子,结果见表3。结果表明,在不同的色谱柱及高效液相色谱仪下中具有较好的重现性,能够用于上述7个成分的含量测定。
本研究采用各待测成分间的相对保留值(待测成分与内参物调整保留时间之比)作为定位标准,并在不同品牌高效液相色谱系统和不同色谱柱下对该参数进行考察,结果表明,不同仪器和色谱柱下各成分间的相对保留值波动较小,RSD在0.10%~0.30%,见表4。此结果说明,在对照品短缺的情况下,利用相对保留值进行色谱峰定位,可根据内参物的保留时间,通过结合各色谱峰的紫外吸收特征及相对保留值能够较准确目标峰的峰位置。因此选用各成分相对保留值这一参数作为丁公藤中上述7个目标色谱峰的定位指标比较合理。
分别称取丁公藤不同炮制工艺饮片粉末(40目)约1.0 g,精密称定,依法制备供试品溶液,分别精密吸取对照品溶液与供试品溶液4 μL,注入超高效液相色谱仪,依法测定,分别采用外标法(EMS)和QAMS法测定不同炮制工艺样品中7个活性成分含量,结果见表5图1。经t检验比较,结果显示外标法实测含量值与QAMS计算的含量值无显著性差异,由此说明一测多评法可用于丁公藤饮片的多成分质量评价研究。
试验前期以同一样品粉末(生品,40目,1 g)对丁公藤的超声提取(功率200 W,频率40 kHz)与回流提取方法进行进样比较,结果显示回流提取法峰面积较高,提取效果最好,因此确定为回流提取法。在此基础上进一步对不同浓度的提取溶剂(50%甲醇、80%甲醇、100%甲醇)、提取温度(65、70、80和90 ℃)、提取次数(1、2、3次)、溶剂用量(15、25、35 mL)进行考察,结果显示80%甲醇,提取温度90 ℃下回流提取2次,各成分峰面积最大,说明含量较其他条件提取高,最终确定为试验供试品溶液的制备方法。
采用一测多评校正因子进行样品含量计算,由于在不同波长下偏差较大,如何选定适合的波长对降低测定误差很关键。结合紫外光谱及液相紫外全波长扫描结果,本研究分别在323、324、330、345、338 nm波长下对7个成分进行测定,结果发现上述7个成分在338 nm处具有较大吸收,基线平稳,且色谱峰分离度均不小于1.5,达到色谱分析要求,因此选用338 nm为检测波长。
试验采用超高效液相色谱法(UPLC)比较丁公藤炮制前后东莨菪内酯、新绿原酸、东莨菪苷、绿原酸、异绿原酸B、异绿原酸A、异绿原酸C的含量,建立了丁公藤及其炮制品的多指标质量评价方法。经炮制后,不同丁公藤饮片中新绿原酸含量为:Y5>Y2>Y4>Y12>Y11>Y15>Y13>Y16>Y1、Y9>Y8、Y14>Y6>Y7>Y10>Y3;绿原酸含量结果为Y5>Y2>Y4>Y11>Y12>Y15>Y3>Y13>Y16>Y14>Y1、Y9>Y6>Y8>Y7>Y10;异绿原酸B含量为:Y5>Y3>Y2>Y11>Y4>Y12>Y15>Y16>Y6>Y1、Y9>Y8>Y13>Y7>Y14>Y10;异绿原酸A含量为:Y5>Y2>Y11>Y4、Y12>Y15>Y3、Y16>Y6>Y1、Y9>Y8>Y13>Y7、Y10、Y14;异绿原酸C含量:Y5>Y2>Y11>Y15>Y4>Y16>Y6>Y3>Y1、Y9>Y8>Y13>Y14>Y7、Y10。进一步分析发现,上述5个有机酸含量变化趋势基本一致。Y5(9.89 mg·g-1)、Y2(3.67 mg·g-1)、Y4(2.81 mg·g-1)、Y12(2.12 mg·g-1)中新绿原酸含量分别是Y1(0.48 mg·g-1)的20.6、7.7、5.9、4.4倍,其中Y3(0.06 mg·g-1)最低,仅有Y1的12.5%。与Y2相比,仅甘草汁用量由6%减少为3%,含量相差60多倍;对绿原酸含量分析发现,Y5(3.81 mg·g-1)、Y2(1.39 mg·g-1)、Y4(0.9 mg·g-1)分别是Y1(0.22 mg·g-1)的18.3、6.3、4.1倍,Y10最低,仅有Y1(生品)的2%;各炮制品中异绿原酸B含量,以Y5(18.53 mg·g-1)最高,其次是Y3(14.97 mg·g-1)、Y2(6.35 mg·g-1),分别是Y1(0.82 mg·g-1)含量的22.6、18.3和7.7倍,Y10(0.50 mg·g-1)含量最低,仅有生品的50%;Y5(1.60 mg·g-1)中异绿原酸A含量最高,较Y1(0.1 mg·g-1)增加了15倍。Y2(0.63 mg·g-1)则增加了5.3倍,而Y7、Y10、Y14含量最低,均为0.06 mg·g-1;对异绿原酸C含量比较发现,Y5(3.24 mg·g-1)、Y2(1.19 mg·g-1)、Y11(0.87 mg·g-1)分别是Y1(0.23 mg·g-1)含量的14.3、5.2、3.78倍。Y7、Y10(0.12 mg·g-1)含量最低,约为Y1的50%。
另外,对同为煮法炮制的Y2~Y5有机酸总含量比较,结果为Y5(37.05 mg·g-1)>Y3(15.95 mg·g-1)>Y2(13.22 mg·g-1)>Y4(9.43 mg·g-1)>Y1(1.85 mg·g-1),说明甘草汁与盐水2种辅料一同炮制时比单一辅料(盐水或甘草汁)炮制更有利于成分的溶出,且当药材与甘草比例为100∶6时较100∶3时各成分溶出含量高,说明辅料比例对有机酸成分的溶出有影响;对不同工艺炮制方法分析,发现蒸法炮制样品(Y11~Y13)与炙法炮制样品(Y14~Y16)的2种辅料炮制比单一辅料炮制更有利于有效成分溶出与煮法基本一致,而经浸泡后晒干处理的炮制品(Y6~Y10)的变化与其相反,分析原因可能与煮法、蒸法、炙法的炮制过程均有加热,说明加热有利于其有效物质溶出;当炮制辅料相同情况下,以甘草汁-盐水炮制为例,分析5个有机酸总含量变化趋势为:Y5(37.05 mg·g-1)>Y11(8.92 mg·g-1)>Y16(4.09 mg·g-1)>Y1(1.85 mg·g-1)>Y9(1.76 mg·g-1),此结果显示煮法较蒸法、炙法含量高,而浸泡后晒干(Y9)的炮制方法会使成分含量降低,可能是煮法处理时将药物煮至近干,一方面减少了成分的损失,另一方面可能加热时间长,使部分成分发生了水解或分解生成相应有机酸而使其含量远高于其他炮制方法。蒸法处理时将浸泡后剩余的辅料丢弃,可能是造成其含量偏低的原因。炙法含量较蒸法低,可能原因是其炮制温度较低、加热时间较短所致。而加辅料浸泡后晒干的含量较生品低,说明浸泡使其成分降低。上述结果显示经蒸、煮、炙炮制后,药物中新绿原酸、绿原酸、异绿原酸B、异绿原酸A、异绿原酸C含量增加,说明蒸、煮、炒等加热处理可能使丁公藤中部分成分发生了转化,在相同条件下有利于上述5个有机酸类成分溶出,从而使其含量增加。或可能是辅料甘草或食盐水中甘草酸、氯化钠等成分,促进了新绿原酸、异绿原酸类等成分溶出而使其含量增加。另外,通过比较同样炮制工艺丁公藤中上述5个有机酸含量变化,发现随着甘草汁用量增加,其含量出现了增加的趋势,说明辅料甘草汁的用量比例对绿原酸有较大影响。而浸泡可在一定程度上造成上述成分的损失,因此在水处理时应该注意少泡多润以减少成分损失,需注意辅料甘草汁比例对丁公藤有机酸类成分的影响。
由表6结果可知,东莨菪苷含量高低顺序为Y5>Y15>Y4>Y2>Y12>Y13>Y14>Y8>Y6>Y7>Y11>Y3>Y1、Y9、Y10。进一步分析发现,经炮制处理后,东莨菪苷的含量以Y5含量最高,是Y1含量的11.4倍,其次是Y15、Y4、Y2,Y1、Y9、Y10最低(0.31 mg·g-1)。东莨菪内酯含量以Y7(1.48 mg·g-1)含量最高,Y14、Y10、Y4、Y16、Y13略高于Y1(1.08 mg·g-1)、Y9;Y8、Y12、Y15、Y2、Y11、Y5略低于生品,Y3(0.3 mg·g-1)含量最低。分析比较可发现,炮制对东莨菪内酯的影响与东莨菪苷的趋势变化相反,说明经长时间高温水煮后,东莨菪内酯可能与药物或辅料中糖类成分发生了缩合反应,生成东莨菪苷或其他成分使其含量降低,具体转化机制还有待进一步研究。
综合以上结果分析可知,辅料甘草汁、盐水制对丁公藤的化学成分有一定影响,除东莨菪内酯含量降低外,甘草汁-盐水共煮(Y5)的炮制方法显著提高了其他6个化学成分的含量。但因各炮制品批次较少,还有待进一步增加炮制品批次,对炮制后丁公藤质量进行深入研究。2020年版《中国药典》仅收载了丁公藤生品的炮制方法,并仅以东莨菪内酯为指标进行质量检查,不利于其饮片质量控制,且传统中医药理论认为丁公藤有小毒,这必将影响其临床安全,因此十分有必要进一步对丁公藤炮制工艺进行深入研究。本研究在工艺规范的前提下,建立一测多评法同时测定丁公藤中新绿原酸、绿原酸、东莨菪苷、东莨菪内酯、异绿原酸A、异绿原酸B、异绿原酸C等7个有效成分含量测定方法,建立其限量要求,对于丁公藤饮片的质量控制具有一定意义,因此建议在省市炮制规范中予以明确同时增加相应指标成分含量限度。
本试验的方法简单、快速、准确,可用于丁公藤多指标成分含量测定,为更好控制丁公藤饮片的质量提供参考。加热和加辅料制均对丁公藤的化学成分产生了一定影响,以甘草汁-盐水(Y5)共煮法影响最为明显。
