Objective:

To establish a method for determination of 7 mycotoxin contaminants in Jianqu by ultra high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).

Methods:

The samples were extracted with acetonitrile aqueous solution (containing 0.1% formic acid) ,and purified by QuEChERS extraction salt bag and Oasis PRiME HLB solid phase extraction column. The column of UPLC was WATERS HSS T3(100 mm×2.1 mm,1.8 μm). Mobile phase was acetonitrile-5 mmol·L-1 ammonium formate solution (containing 0.1% formic acid) with gradient elution. Ionization mode was electrospray ionization(ESI) with positive mode. The work mode was multiple-reaction monitoring mode(MRM).

Results:

The developed method provided a good linearity for the 7 mycotoxins with their respective linear rangers. The correlation(r) ranged from 0.999 1 to 0.999 9. The average recoveries ranged from 83.5% to 113.8%,RSDs of 2.1% to 5.2%. Among 10 samples were selected for analysis,aflatoxin B1 was detected from 3 samples at concentration of 1.32 μg·kg-1 to 6.15 μg·kg-1. The results indicated that there were serious safety risks in the Jianqu products.

Conclusion:

This paper establishes a method for the determination of 7 mycotoxins in Jianqu. The residual of mycotoxins in Jianqu can be rapid ly detected by UPLC-MS/MS which is suitable for the risk monitoring of mycotoxins in Jianqu. This article reveals the current situation of mycotoxins contamination in Jianqu,providing a research basis for the safety control of traditional Chinese medicine. It is recommended that the relevant departments should strengthen the corresponding supervision work.

目的:

建立超高效液相色谱-串联质谱法(UPLC-MS/MS法)快速测定中药建曲中7个真菌毒素的残留。

方法:

样品用含0.1%甲酸的乙腈水溶液超声提取,再用QuEChERS提取盐包和Oasis PRiME HLB固相萃取柱处理,采用WATERS HSS T3(100 mm×2.1 mm,1.8 μm)色谱柱分离,流动相A为乙腈,流动相B为5 mmol·L-1甲酸铵水溶液(含0.1%甲酸),梯度洗脱;采用电喷雾(ESI)正离子离子源电离,工作模式为多反应监测模式(MRM)。

结果:

7个真菌毒素在各自的线性范围内呈良好的线性关系,相关系数在0.999 1~0.999 9。平均回收率为83.5%~113.8%,RSD在2.1%~5.2%。10批样品中有3批样品检出黄曲霉毒素B1,其含量最高达到了6.15 μg·kg-1,最低的为1.32 μg·kg-1。结果提示建曲制品存在严重的安全风险。

结论:

本文建立了建曲中7个真菌毒素的测定方法,通过超高效液相色谱-串联质谱法可以快速检测建曲中的真菌毒素残留量,可适用于建曲中真菌毒素的风险监测。本文揭示了建曲中存在真菌毒素污染的现状,为中药的安全性控制提供了研究基础,建议有关部门加强相应的监管工作。

目的:

建立超高效液相色谱-串联质谱法(UPLC-MS/MS法)快速测定中药建曲中7个真菌毒素的残留。

方法:

样品用含0.1%甲酸的乙腈水溶液超声提取,再用QuEChERS提取盐包和Oasis PRiME HLB固相萃取柱处理,采用WATERS HSS T3(100 mm×2.1 mm,1.8 μm)色谱柱分离,流动相A为乙腈,流动相B为5 mmol·L-1甲酸铵水溶液(含0.1%甲酸),梯度洗脱;采用电喷雾(ESI)正离子离子源电离,工作模式为多反应监测模式(MRM)。

结果:

7个真菌毒素在各自的线性范围内呈良好的线性关系,相关系数在0.999 1~0.999 9。平均回收率为83.5%~113.8%,RSD在2.1%~5.2%。10批样品中有3批样品检出黄曲霉毒素B1,其含量最高达到了6.15 μg·kg-1,最低的为1.32 μg·kg-1。结果提示建曲制品存在严重的安全风险。

结论:

本文建立了建曲中7个真菌毒素的测定方法,通过超高效液相色谱-串联质谱法可以快速检测建曲中的真菌毒素残留量,可适用于建曲中真菌毒素的风险监测。本文揭示了建曲中存在真菌毒素污染的现状,为中药的安全性控制提供了研究基础,建议有关部门加强相应的监管工作。

Objective:

To establish a method for determination of 7 mycotoxin contaminants in Jianqu by ultra high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).

Methods:

The samples were extracted with acetonitrile aqueous solution (containing 0.1% formic acid) ,and purified by QuEChERS extraction salt bag and Oasis PRiME HLB solid phase extraction column. The column of UPLC was WATERS HSS T3(100 mm×2.1 mm,1.8 μm). Mobile phase was acetonitrile-5 mmol·L-1 ammonium formate solution (containing 0.1% formic acid) with gradient elution. Ionization mode was electrospray ionization(ESI) with positive mode. The work mode was multiple-reaction monitoring mode(MRM).

Results:

The developed method provided a good linearity for the 7 mycotoxins with their respective linear rangers. The correlation(r) ranged from 0.999 1 to 0.999 9. The average recoveries ranged from 83.5% to 113.8%,RSDs of 2.1% to 5.2%. Among 10 samples were selected for analysis,aflatoxin B1 was detected from 3 samples at concentration of 1.32 μg·kg-1 to 6.15 μg·kg-1. The results indicated that there were serious safety risks in the Jianqu products.

Conclusion:

This paper establishes a method for the determination of 7 mycotoxins in Jianqu. The residual of mycotoxins in Jianqu can be rapid ly detected by UPLC-MS/MS which is suitable for the risk monitoring of mycotoxins in Jianqu. This article reveals the current situation of mycotoxins contamination in Jianqu,providing a research basis for the safety control of traditional Chinese medicine. It is recommended that the relevant departments should strengthen the corresponding supervision work.

真菌毒素是由产毒真菌在一定的条件下产生的有毒次级代谢产物,其具有致畸、致突变、致癌、抑制免疫等毒性[1-3],对人体的健康造成严重危害,并且广泛存在于食品、中药材、饲料、果蔬等中[4-5]。目前发现的真菌毒素共有400多个,其中对人体危害比较大的主要有黄曲霉毒素、脱氧雪腐镰刀烯醇(DON,又称呕吐毒素)、伏马菌素、赭曲霉毒素、玉米赤霉烯酮等[6]。部分中药材在种植、加工、炮制、存储的过程中很有可能遭到各种真菌的污染,产生各种真菌毒素,残留真菌毒素的中药材被患者服用后可能产生各种中毒症状,严重的会导致患者机体损伤甚至死亡[5]。因此真菌毒素的残留污染严重的影响了中药材的质量和用药安全,关于中药材中真菌毒素污染的研究也成为了学者研究的热点,监管部门也加强了对于中药材中真菌毒素的限度检测工作。2010年版《中华人民共和国药典》(简称《中国药典》)首次制订了中药材中真菌毒素的测定方法,并且规定了桃仁、酸枣仁、胖大海、陈皮和僵蚕5种中药材的黄曲霉毒素最大限量值,2020年版《中国药典》将规定黄曲霉毒素最大限量值的中药材扩大至24种。
建曲又名泉州神曲,为赤小豆、苦杏仁、青蒿、辣蓼、苍耳草等21味药粉碎后与麦麸和面粉等混合经过发酵工艺制备得到的曲剂中药,其具有解表和中的功效,主治寒热头痛,食滞阻中,呕吐胀满[7-8]。因为建曲是经过发酵制备而成的,所以在制备过程中可能会滋生各种真菌和细菌,从而产生各类生物毒素。本课题组前期经过研究发现,建曲中存在被椰毒假单胞菌污染,从而残留生物毒素米酵菌酸的现象[9]。而目前关于建曲中各类真菌毒素检测的相关研究并未见到报道,《中国药典》也并未对建曲制定限量标准,因此亟需建立建曲中真菌毒素的监测方法。
目前真菌毒素的检测方法主要有高效液相色谱法(HPLC)[10]、液相色谱-质谱法(LC-MS)[11]、薄层色谱法(TLC)[12]、酶联免疫吸附法(ELISA)[13]等。其中TLC法前处理简单,但灵敏度和专属性较差。ELISA法使用较为安全,但不太稳定,存在假阳性、假阴性,且定量准确性较差。HPLC法前处理简单,准确性较高,但专属性较差。LC-MS法具有稳定性高、准确性高、专属性强、检测通道多等特点,特别适合于样品中多成分同时测定,目前是真菌毒素检测的首选方法[11]。本文建立超高效液相色谱-串联质谱法快速测定建曲中的伏马菌素B1,脱氧雪腐镰刀菌烯醇,黄曲霉毒素B1、B2、G1、G2、M1共7个真菌毒素,本方法专属性强,分析时间短,前处理简单,可为建曲中真菌毒素残留的风险监控提供研究基础。
Waters UPLC Xevo TQ-S超高效液相色谱-质谱联用仪,含有超高效液相色谱仪、电喷雾离子化源和三重四极杆质量分析器(沃特世公司);METTLER AE-200型万分之一电子天平和METTLER XS105DU型十万分之一电子天平(梅特勒-托利多公司);SK5200HP型超声波提取器(上海科导公司);MILLI-Q超纯水制备仪(密理博公司);色谱柱为WATERS HSS T3(规格100 mm×2.1 mm,1.8 μm);Oasis PRiME HLB柱(3 cc/60 mg)固相萃取小柱(WATERS公司);分散固相萃取纯化管包括gSO4 100 mg ,PSA 50 mg,C18 100 mg(安谱公司)。
伏马菌素B1对照品(批号SZBE295XV,浓度为50 μg·mL-1)购自Sigma公司;黄曲霉毒素M1对照品(批号O3430095,浓度为10.5 μg·mL-1)购自安谱公司;脱氧雪腐镰刀菌烯醇对照品(批号349508,浓度为200 μg·mL-1)购自O2Si公司;黄曲霉毒素混合对照品(批号352733,黄曲霉毒素B1、B2、G1、G2浓度分别为0.9、0.3、0.9、0.3 μg·mL-1)购自O2Si公司;甲醇和乙腈购自默克公司,均为色谱纯;甲酸购自ANAQUA CHEMICAL SUPPLY公司,为质谱纯;水为MILLI-Q制备的超纯水;QuEChERS提取盐包(包括4 g硫酸镁,1 g氯化钠,1 g柠檬酸钠,0.5 g柠檬酸氰二钠)购自安谱公司;建曲样品(批号201805058、20171104、20180101、0180212、20171106、20170806、20180611、20180307、20170914、20170608)购自泰州市中药材市场。
采用WATERS HSS T3(规格100 mm×2.1 mm,1.8 μm)色谱柱,流动相A为乙腈,流动相B为5 mmol·L-1甲酸铵水溶液(含0.1%甲酸),梯度洗脱(0~1 min,10%A;1~2 min,10%A→50%A;2~3.2 min,50%A→70%A;3.2~4 min,70%A→50%A;4~4.5 min,75%A→100%A;4.5~5 min,100%A),流速0.4 mL · min-1,柱温35 ℃,进样量5 μL。
离子源ESI(电喷雾电离源),正离子模式,毛细管电压3.5 KV,锥孔电压20 V,离子源温度150 ℃,脱溶剂气温度350 ℃,脱溶剂气流速650 L·h-1,扫描方式为多反应监测模式(MRM),检测离子参数见表1,对照品和样品的色谱图见图1图2
分别吸取适量体积的黄曲霉毒素混合对照品、伏马菌素B1对照品、黄曲霉毒素M1对照品和脱氧雪腐镰刀菌烯醇对照品至量瓶中,加甲醇超声(250 W,40 kHz)定容,制成对照品储备液(其中,伏马菌素B1为1 000 ng·mL-1,脱氧雪腐镰刀菌烯醇为4 000 ng·mL-1,黄曲霉毒素B1为18 ng·mL-1,黄曲霉毒素B2为6 ng·mL-1,黄曲霉毒素G1为18 ng·mL-1,黄曲霉毒素G2为6 ng·mL-1,黄曲霉毒素M1为21 ng·mL-1),于棕色试剂瓶中-18 ℃避光保存,测定当天用含0.1%甲酸的乙腈水溶液稀释得到系列对照品溶液,现配现用。
取已粉碎后的建曲样品1 g,精密称定,置50 mL的离心管中,加入水5 mL和含0.1%甲酸的乙腈10 mL,涡旋1 min,超声(250 W,40 kHz)10 min,再加入QuEChERS提取盐包大力振摇1 min,离心(5 000 r·min-1)5 min。取上清液2 mL上样Oasis PRiME HLB柱(无需活化直接上样),先弃去一部分流出液后再接收。取流出液1 mL于50 ℃下旋转蒸干,用初始流动相溶解旋蒸后的残渣并定容至1 mL,再经过0.22 μm的微孔滤膜过滤,即得。
上述色谱-质谱条件下,对照品的7个真菌毒素的MRM定量离子图见图1,样品的7个真菌毒素的MRM定量离子图见图2。从图中可知7个真菌毒素分离良好,样品的图谱表明样品基质成分不干扰真菌毒素的检测。
取“2.2.1”项下的对照品储备液用初始流动相依次稀释,得到7个真菌毒素的系列标准工作溶液(5个浓度点),分别精密吸取系列标准工作溶液5 μL,按“2.1.1”项下色谱条件测定,以真菌毒素的峰面积(Y)作为纵坐标,以浓度(X)为横坐标,进行线性回归,得线性回归方程,结果见表2。7个真菌毒素的线性相关系数(r)均大于0.999 0,表明各毒素在各自的线性范围内均呈良好的线性关系。
精密称取检测结果为阴性的样品(取约1 g),在其中加入各真菌毒素对照品适量,按照“2.2.2”项下的试验方法制备供试品溶液,每个待测组分均添加高、中、低3个加标水平,每个加标水平平行6份,再按“2.1.1”项下色谱条件测定,用外标法计算回收率,结果见表2。各待测组分的回收率平均值在83.5%~113.8%,RSD≤5.2%(n=6)。说明该方法对于建曲中7个真菌毒素的测定,具有良好的准确度和精密度,符合真菌毒素污染分析的要求。
将加标样品溶液用初始流动相逐级稀释,按“2.1.1”项下色谱条件测定,以信噪比(S/N)=3确定检测限,以信噪比(S/N)=10确定定量限,结果见表2
取10个批次的建曲样品,按照“2.2.2”项下方法分别制备供试品溶液,再按“2.1.1”项下色谱条件测定,用外标曲线法定量,分别计算各批次样品中7个真菌毒素的含量。结果有3批检出了黄曲霉毒素B1,含量分别为1.32、6.15和1.58 μg·kg-1,其余的真菌毒素均未检出。
实验中分别比较了7个真菌毒素在负离子和正离子电离模式下的响应值,结果各组分在正离子模式下的响应值均大于负离子,因此本文最终采用正离子模式作为质谱的电离模式。
液质联用仪中待组分的响应值和峰形与所采用的流动相息息相关,在正离子电离模式下流动相中添加适量的挥发性酸,可以增加环境中的H+,促进待测组分的电离,在流动相中添加适量的挥发性盐可改善待测组分的峰形,提高分离的效果。因此,实验分别考察了乙腈-10 mmol·L-1甲酸铵体系、乙腈-0.1%甲酸体系、乙腈-10 mmol·L-1甲酸氨(含0.1%甲酸)体系和乙腈-5 mmol·L-1甲酸氨(含0.1%甲酸)体系下的响应值和分离效果。结果显示,乙腈-10 mmol·L-1甲酸铵体系下,各黄曲霉毒素响应值较高,但伏马菌素B1和脱氧雪腐镰刀菌烯醇的响应值很弱;乙腈-0.1%甲酸体系下,伏马菌素B1和脱氧雪腐镰刀菌烯醇的响应值较强,但黄曲霉毒素的响应值较弱;乙腈- 10 mmol·L-1甲酸氨(含0.1%甲酸)体系下各黄曲霉毒素和伏马菌素B1的响应值较强,但脱氧雪腐镰刀菌烯醇的响应值较弱;乙腈-5 mmol·L-1甲酸氨(含0.1%甲酸)体系下,各黄曲霉毒素和伏马菌素B1和脱氧雪腐镰刀菌烯醇的响应值均较高,且峰形良好,因此采用乙腈-5 mmol·L-1甲酸氨(含0.1%甲酸)体系作为本文的流动相。
QuEChERS提取法分为2步,第1步样品经超声提取后加入盐包,离心后待测液分层,杂质在下层水层中,第2步取上层有机液,加入C18和PSA等吸附剂除杂[14-15]。实验中分别考察了3种净化方式的前处理方法,分别为QuEChERS两步法(图3中标为C18/PSA)、QuEChERS一步法结合C18固相萃取柱(图3中标为C18小柱)和QuEChERS一步法结合PRiME HLB固相萃取柱(图3中标为PRiME HLB小柱)。其中QuEChERS两步法前处理过程为,第一步样品中加入5 mL水和10 mL的含0.1%甲酸的乙腈超声后,再加入提取盐包,剧烈震荡后离心。第二步在分散固相萃取纯化管(包含MgSO4、PSA和C18粉末)中加入离心后的上清液,涡旋后除去杂质。QuEChERS一步法结合固相萃取柱前处理过程为在第一步试剂提取后加入提取盐包,剧烈震荡离心后,上清液经过固相萃取柱除杂,流出液氮气吹干后用流动相复溶。图3可见,QuEChERS两步法中伏马菌素B1和脱氧雪腐镰刀菌烯醇的回收率较差,不到60%,C18固相萃取柱对伏马菌素B1、脱氧雪腐镰刀菌烯醇、黄曲霉毒素B1、B2、G1、M1的回收率较好,在78.2%~92.3%,而对黄曲霉毒素G2的回收率较低,为72.3%。PRiME HLB固相萃取柱对7个真菌毒素的回收率均较高,回收率在85.6%~102.6%,相对于其他2种净化方式,净化效果好,且操作简单、前处理时间段、效率高,适合大批量样品检测。因此选用QuEChERS一步法结合PRiME HLB固相萃取柱的净化方法来处理样品。
本文在建曲中检测到了黄曲霉毒素B1的存在,实际样品测定中,10批次建曲样品有3批次样品检出了黄曲霉毒素B1,真菌毒素污染率为30%,这提示目前建曲中真菌毒素的污染水平较高,亟需制定相关的法律法规,并开展建曲中真菌毒素的质量监控。目前2020年版《中国药典》中并没有关于建曲中真菌毒素的限量标准规定,而建曲中的原材料含有大量的麦麸和面粉,因此可以参照《GB 2761-2017食品安全国家标准食品中真菌毒素限量》中谷物及其制品项下的小麦粉、麦片、其他去壳谷物食品类别的限度标准来判定有无超标风险,标准中规定黄曲霉毒素B1的限量值为5.0 μg·kg-1。检测结果为阳性的3批建曲样品中黄曲霉毒素B1的残留量分别为1.32、6.15和1.58 μg·kg-1,参照上述标准可以判断有1批样品的黄曲霉毒素B1超标。
本文建立了建曲中7个真菌毒素的检测方法,通过超高效液相色谱-串联质谱法可以快速检测建曲中的7个真菌毒素残留量。本方法快速、简便,准确性高,可适用于建曲中真菌毒素的风险监测。本文揭示了建曲中存在黄曲霉毒素B1污染的现状,为中药的安全性控制提供了研究基础,建议有关部门加强相应的监管工作。
