Objective:

To establish an high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method for the simultaneous determination of 45 additives in oral liquid preparations of traditional Chinese medicine.

Methods:

The samples were ultrasonically extracted by acetonitrile-methanol (9:1) (containing 0.1% formic acid). The separation was carried out on an Agilent Eclipse Plus C18 chromatographic column (150 mm×3.0 mm,1.8μm) using gradient elution of methanol -5 mmol·L-1 ammonium acetate. The compounds were scanned and detected simultaneously by electrospray ionization(ESI)ion source in both positive ion and negative ion mode under dynamic multiple reaction monitoring. The retention time and ion ratio were used for qualitative analysis and the external standard method was adopted for quantification.

Results:

The good linear relationship of peak area was observed for the 45 additives with correlation coefficients ≥0.992 in the concentration range of 5-2 000 ng·mL-1,and the limits of quantitation were between 0.2 mg·kg-1 and 4.0 mg·kg-1 under the above chromatographic and mass spectrometric conditions. The average recoveries of the blank samples at different added levels ranged from 75.4% to 118.4% with RSDs of 0.70%-9.8%. The method was used to detect 20 batches of oral liquid preparations of traditional Chinese medicines purchased from pharmacies. Benzoic acid was detected in 6 batches with the contents of benzoic acid from 0.13% to 0.27%,and sorbic acid,trans-cinnamic acid,molasses,ethyl 4-hydroxybenzoate,acesulfame potassium,dehydroacetic acid and saccharin sodium were detected in 7 batches of samples respectively.

Conclusion:

The established high-throughput detection method is sensitive,simple and fast in pre-treatment,with high accuracy,stable recovery and reduced detection cost effectively. It can be used for the simultaneous rapid screening of multiple additives in oral liquid preparations of traditional Chinese medicine.

目的:

建立并优化了高效液相色谱-串联质谱(HPLC-MS/MS)同时测定中药口服液体制剂中45种添加剂化合物的分析方法。

方法:

样品经乙腈-甲醇(9:1)(含0.1%甲酸)超声提取,Agilent Eclipse Plus C18(150 mm×3.0 mm,1.8 μm)色谱柱进行分离,以甲醇和5 mmol·L-1乙酸铵为流动相梯度洗脱,采用电喷雾离子源正、负离子同时扫描,动态多反应监测模式,保留时间和离子丰度比进行定性分析以及外标法定量。

结果:

在优化的色谱-质谱条件下,45种添加剂化合物在5~2 000 ng·mL-1范围内线性良好(r≥0.992);方法定量限为0.2~4.0 mg·kg-1;各待测物在空白样品中不同加标水平下的平均回收率为75.4%~118.4%;RSD为0.70%~9.8%。应用该方法对从药房购买的20批中药口服液体制剂样品进行检测,其中6批次样品检出苯甲酸,含量为0.13%~0.27%,7批次样品分别检出山梨酸、肉桂酸、甜蜜素、4-羟基苯甲酸乙酯、安赛蜜、脱氢乙酸和糖精钠。

结论:

建立的高通量检测方法灵敏、前处理简便、快速、准确度高、回收率稳定,能有效降低检测成本,可用于中药口服液体制剂中多种添加剂化合物的同时快速筛查。

目的:

建立并优化了高效液相色谱-串联质谱(HPLC-MS/MS)同时测定中药口服液体制剂中45种添加剂化合物的分析方法。

方法:

样品经乙腈-甲醇(9:1)(含0.1%甲酸)超声提取,Agilent Eclipse Plus C18(150 mm×3.0 mm,1.8 μm)色谱柱进行分离,以甲醇和5 mmol·L-1乙酸铵为流动相梯度洗脱,采用电喷雾离子源正、负离子同时扫描,动态多反应监测模式,保留时间和离子丰度比进行定性分析以及外标法定量。

结果:

在优化的色谱-质谱条件下,45种添加剂化合物在5~2 000 ng·mL-1范围内线性良好(r≥0.992);方法定量限为0.2~4.0 mg·kg-1;各待测物在空白样品中不同加标水平下的平均回收率为75.4%~118.4%;RSD为0.70%~9.8%。应用该方法对从药房购买的20批中药口服液体制剂样品进行检测,其中6批次样品检出苯甲酸,含量为0.13%~0.27%,7批次样品分别检出山梨酸、肉桂酸、甜蜜素、4-羟基苯甲酸乙酯、安赛蜜、脱氢乙酸和糖精钠。

结论:

建立的高通量检测方法灵敏、前处理简便、快速、准确度高、回收率稳定,能有效降低检测成本,可用于中药口服液体制剂中多种添加剂化合物的同时快速筛查。

Objective:

To establish an high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method for the simultaneous determination of 45 additives in oral liquid preparations of traditional Chinese medicine.

Methods:

The samples were ultrasonically extracted by acetonitrile-methanol (9:1) (containing 0.1% formic acid). The separation was carried out on an Agilent Eclipse Plus C18 chromatographic column (150 mm×3.0 mm,1.8μm) using gradient elution of methanol -5 mmol·L-1 ammonium acetate. The compounds were scanned and detected simultaneously by electrospray ionization(ESI)ion source in both positive ion and negative ion mode under dynamic multiple reaction monitoring. The retention time and ion ratio were used for qualitative analysis and the external standard method was adopted for quantification.

Results:

The good linear relationship of peak area was observed for the 45 additives with correlation coefficients ≥0.992 in the concentration range of 5-2 000 ng·mL-1,and the limits of quantitation were between 0.2 mg·kg-1 and 4.0 mg·kg-1 under the above chromatographic and mass spectrometric conditions. The average recoveries of the blank samples at different added levels ranged from 75.4% to 118.4% with RSDs of 0.70%-9.8%. The method was used to detect 20 batches of oral liquid preparations of traditional Chinese medicines purchased from pharmacies. Benzoic acid was detected in 6 batches with the contents of benzoic acid from 0.13% to 0.27%,and sorbic acid,trans-cinnamic acid,molasses,ethyl 4-hydroxybenzoate,acesulfame potassium,dehydroacetic acid and saccharin sodium were detected in 7 batches of samples respectively.

Conclusion:

The established high-throughput detection method is sensitive,simple and fast in pre-treatment,with high accuracy,stable recovery and reduced detection cost effectively. It can be used for the simultaneous rapid screening of multiple additives in oral liquid preparations of traditional Chinese medicine.

随着医药行业的快速发展,各种中药口服液体制剂产品日益增多。由于中药口服液体制剂中含有的大量水体基质容易滋生细菌引起腐败变质[1-2],为了抑菌杀菌和延长保质期,常常会加入一些防腐剂和抗氧化剂如山梨酸、尼铂金类、苯扎氯铵类、叔丁基羟基茴香醚等[3-4]。此外,由于中药本身的苦味加上工艺中酸碱试剂的加入产生的刺激性气味,会极大降低中药口服液体制剂的综合口感,为了改善其口感,尤其在一些儿童口服液体制剂中通常会加入一些如阿斯巴甜、安赛蜜、甜菊糖苷等甜味剂作为矫味剂[5]。虽然在中药口服液体制剂生产过程中允许使用添加剂,但2020年版《中华人民共和国药典》(简称《中国药典》)四部中明确规定了部分添加剂的允许使用限量,但未明确其检测方法[6]。添加剂使用不当或滥用,不但直接影响中药口服液体制剂的口感和储存效果,更直接影响其质量与疗效,也对人体健康和用药安全构成潜在威胁[26-10]
目前,添加剂的检测研究主要集中在化妆品、食品和工业产品领域中,或者只是针对单一添加剂种类进行研究,鲜有对中药口服液体制剂中多种添加剂的研究报道[10-14]。因此,急需建立1套中药口服液体制剂中多种添加剂的高通量筛查技术,以解决此类药品监管薄弱问题。高效液相色谱-质谱联用法(LC-MS/MS)结合了液相色谱对复杂机体化合物的高分离能力以及质谱独特的选择性、灵敏度、相对分子质量及结构信息于一体的特点,使其具有应用范围广、检测灵敏度高、分析周期短、定性定量准确以及能有效避免假阴性、假阳性等优点[415-16]。本研究以中药口服液中常用添加剂为研究对象,建立了能同时检测45种添加剂的高通量筛查方法。该方法灵敏度高,准确、快速,可用于中药口服液体制剂中添加剂的快速筛查,为药品监管提供了强有力的技术支持。
Agilent 1290-6495超高效液相色谱-三重四极杆串联质谱仪,Agilent公司;X3R高速离心机,Thermo Fisher Scientific公司;IRM IDH30超声仪,IRM Technology GmbH公司;IKA MS3涡旋混合器,IKA公司。甲酸、乙腈、甲醇、乙酸铵均为色谱纯,Thermo Fisher Scientific公司;实验用水为Milli-Q超纯水。
混合对照品1:苯甲酸、山梨酸、纳他霉素、脱氢乙酸、邻苯基苯酚混合对照品溶液,浓度均为1 000 μg·mL-1;北京曼哈格生物科技有限公司。
混合对照品2:肉桂酸、4-氯-3,5-二甲基苯酚(对氯间二甲酚)混合对照品溶液,浓度均为1 000 μg·mL-1,天津阿尔塔科技有限公司。
混合对照品3:三氯卡班、4-氯-3-甲基苯酚(对氯间甲酚)、苄氯酚、溴氯芬、4-羟基苯甲酸甲酯、4-羟基苯甲酸乙酯、4-羟基苯甲酸丙酯、4-羟基苯甲酸异丙酯、4-羟基苯甲酸丁酯、4-羟基苯甲酸异丁酯、三氯生、2-溴-2-硝基-1,3丙二醇(布罗波尔)、2,4-二氯苯氧乙酸、苯氧乙酸、甲基异噻唑啉酮、甲基氯异噻唑啉酮、碘丙炔醇丁基氨甲酸酯、氯咪巴唑、苄索氯铵、十四烷基二甲基苄基氯化铵、十六烷基二甲基苄基氯化铵、4-羟基苯甲酸戊酯、十二烷基二甲基苄基氯化铵、4-羟基苯甲酸苯酯、4-羟基苯甲酸苄酯、噻苯咪唑、月桂酰精氨酸乙酯盐酸盐混合对照品溶液,浓度均为100 μg·mL-1,北京曼哈格生物科技有限公司。
混合对照品4:纽甜、阿斯巴甜、糖精钠、甜蜜素、安赛蜜、没食子酸丙酯、丁基羟基茴香醚、阿力甜混合对照品溶液,浓度均为1 000 μg·mL-1,天津阿尔塔科技有限公司。
混合对照品5:甜菊糖苷、瑞鲍迪甙A、爱德万甜混合对照品溶液,浓度均为1 000 μg·mL-1,天津阿尔塔科技有限公司。
实验样品:中药口服液体制剂,购自药房。
分别准确吸取混合对照品1溶液0.025 mL、混合对照品2溶液0.01 mL以及混合对照品3溶液0.05 mL,置于同一10 mL量瓶中,用甲醇稀释至刻度,摇匀,作为混合对照品中间液A;准确吸取混合对照品4溶液0.1 mL以及混合对照品5溶液0.2 mL,置于10 mL量瓶中,用甲醇稀释至刻度,摇匀,作为混合对照品中间液B。
称取混合均匀的样品1 g(精确至0.001 g),置50 mL离心管中,加入乙腈-甲醇(9:1)(含0.1%甲酸) 15 mL,涡旋混匀5 min,超声(功率500 W,频率40 kHz)提取10 min,置于高速离心机中9 000 r·min-1离心5 min,将上清液转移至20 mL量瓶中,用乙腈-甲醇(9:1)(含0.1%甲酸)定容至刻度,摇匀,取上清液过0.22 μm滤膜,续滤液供液相色谱-串联质谱仪分析。
Agilent Eclipse Plus C18(150 mm×3.0 mm,1.8 μm)色谱柱。柱温35 ℃,进样体积5 μL,流动相A:5 mmol·L-1乙酸铵-水溶液,流动相B:甲醇,梯度洗脱(0~1.0 min,10%B;1~3.0 min,60%B;3.0~6.0 min,60%B;6.0~18 min,95%B;18~22 min,95%B),流速0.3 mL·min-1
电喷雾离子源(ESI);正、负离子模式同时扫描,动态多反应监测模式(DMRM),毛细管电压3.0 kV,干燥气温度250 ℃,干燥气流量11 L·min-1,雾化气压力0.31 MPa,鞘气温度300 ℃,鞘气流量11 L·min-1。45种添加剂化合物监测离子对及碰撞能量(CE)参数见表1
分别精密量取一定体积的混合对照品中间液A和B,配制成45种目标化合物浓度在5~2 000 ng·mL-1之间的系列混合对照品溶液进行测定,以目标组分的峰面积Y对相应的浓度X(ng·mL-1)绘制标准曲线,其r均大于0.992。以信噪比(S/N)≥10的作为方法的定量限(LOQ),其线性方程和定量限见表2
分别取质量浓度约为10、20、50 ng·mL-1的各混合对照品溶液(各化合物浓度有所差别与其定量限相一致),按“1.4”项下色谱条件分别连续测定6次,结果(表3)45个目标化合物峰面积的RSD均<5.0%,表明仪器精密度良好。
取质量浓度约为50 ng·mL-1的各混合对照品溶液,分别放置0、2、4、8、12及24 h后,按“1.4”项下色谱条件分别进样进行测定,通过峰面积计算45个化合物的RSD均<5.0%,表明在24 h内各化合物稳定性较好,其结果见表3
取市售经检查不含上述45种添加剂的中药口服液作为空白基质,考察了口服液空白基质和添加45个化合物组分后的色谱图。结果如图1~图3所示,表明中药口服液基质中存在的物质对本方法化合物基本没有干扰。
在空白样品中分别添加低、中、高3个浓度化合物,每个添加水平重复测定6次,计算平均加标回收率和日内精密度,结果见表4。45个化合物各添加水平的平均回收率为75.4%~118.4%;RSD为0.70%~9.8%,均能满足定量分析的要求。
采用本实验建立的方法,对从药房购买的20批中药口服液体制剂样品进行检测,其中3批次清热解毒口服液、1批次桔梗清咽合剂、1批次气血康口服液以及1批次射麻口服液样品中检出苯甲酸,含量为0.13%~0.27%;2批次小儿解表口服液样品中检出山梨酸、肉桂酸、甜蜜素;1批次抗病毒口服液、1批次甘草口服液、1批次强力枇杷露样品中检出4-羟基苯甲酸乙酯和安赛蜜;1批次清热解毒口服液、1批次小儿清热止咳口服液样品检出脱氢乙酸和糖精钠,其中不少样品的标签与说明书中均未注明含有相关添加剂化合物,其余样品未检出添加剂化合物。
实验中选择采用电喷雾离子源(ESI),正、负离子同时扫描模式对45个化合物化合物进行了一级质谱全扫描和子离子扫描分析,筛选出响应较高的碎片离子作为目标化合物的定量、定性离子,同时筛选出最佳碰撞能量值。为了避免样品检测中出现假阳性的风险,在满足质谱响应和峰形的前提下,应尽可能多地采集一些目标化合物的碎片离子,从提高检测的准确性。由于目标化合物较多以及对应的碎片离子也较多,采用传统的多反应监测方法(MRM)可能会使目标化合物碎片离子的色谱峰采集点不够,造成峰形差和灵敏度低,影响定性和定量结果准确。因此,实验中采用了动态多反应监测方法(DMRM),该模式采用保留时间和保留时间窗口2个参数,可以实现以目标化合物岀峰时间为中心,在相应保留时间窗口范围内仅对单一目标化合物进行扫描,确保目标化合物色谱峰完整性的同时,提高了检测灵敏度和定性定量的准确性[17]。45种添加剂总离子流图见图2,定量离子对色谱图见图3
研究中考察了不同提取试剂、混合比例以及不同甲酸浓度对目标化合物提取效果的影响,单独采用甲醇或者乙腈作为提取试剂时,甲基异噻唑啉酮、纳他霉素、苯甲酸、噻苯咪唑等化合物的峰形较差,综合考虑各化合物峰形及分离情况,采用乙腈-甲醇(9:1)提取。同时,进一步考察了甲酸浓度对目标化合物提取效果的影响,不加甲酸时山梨酸、对氯间甲酚、对氯间二甲酚、邻苯基苯酚等化合物响应差;甲酸浓度过高,安赛蜜、糖精钠、甜蜜素的峰形差,无法准确定量分析,考虑到各目标化合物的峰形以及响应,最终采用0.1%甲酸。
采用高效液相色谱-串联质谱(HPLC-MS/MS)技术,建立能同时检测中药口服液液体制剂中45种添加剂的高通量筛查方法,解决了目前中药口服液体制剂中添加剂一次性检测化合物少、检验方法不完善的缺陷,该方法操作简便、准确,检测成本较低,极大提高了中药口服液体制剂中多种添加剂的定性定量检测效率,能快速反映产品中添加剂的实际添加情况,对监测药品中添加剂的使用情况具有重要的意义。
