Objective:

To establish the ultra high performance liquid chromatography (UPLC) fingerprint of Qiye Shen’an dropping pills,and to determine the contents of 8 index components by quantitative analysis of multi-components by single marker at the same time.

Methods:

Waters CORTECS® T3 (150 mm×2.1 mm,1.6 μm) chromatography column was used for gradient elution with acetonitrile -0.1% phosphoric acid as mobile phase,flow rate was 0.40 mL·min-1,column temperature was 35 ℃,detection wavelength was 203 nm.

Results:

The UPLC fingerprint of Qiye Shen’an dropping pills was established,8 common peaks were established,and the similarity of 15 batches of samples was greater than 0.99. Ginsenoside Rb1,ginsenoside RC,panax notoginseng saponin FC,ginsenoside Rb2,ginsenoside Rb3,ginsenoside Rd,panax notoginseng saponin Fe and panax notoginseng saponin Fd were determined by quantitative analysis of multi-components by single marker method. The contents of 8 components in 15 batches were between 7.6-10.0 mg per pill. The average recoveries were 99.8%,100.6%,99.8%,101.2%,102.1%,101.9%,99.7% and 100.2% respectively,and the RSDs were 2.3%,2.2%,2.0%,1.5%,1.7%,1.2%,2.4% and 2.6% respectively.

Conclusion:

The UPLC fingerprint of Qiye Shen’an dropping pills and the content determination method of quantitative analysis of multi-components by single marker method established in this study are simple to operation,and have good repeatability,stability and reliability,and can provide a more comprehensive basis for the quality control of Qiye Shen’an dropping pills.

目的:

建立七叶神安滴丸的超高效液相色谱(UPLC)指纹图谱鉴别方法,同色谱条件下完成一测多评法同时测定8个指标成分的含量。

方法:

采用Waters CORTECS®T3(150 mm×2.1 mm,1.6 μm)色谱柱,以乙腈-0.1%磷酸水为流动相进行梯度洗脱,流速0.40 mL·min-1,柱温35 ℃,检测波长203 nm。

结果:

建立七叶神安滴丸UPLC指纹图谱,确立了15个共有峰,15批样品相似度均大于0.99。并对其含量较高的8个成分人参皂苷Rb1、人参皂苷Rc、三七皂苷Fc、人参皂苷Rb2、人参皂苷Rb3、人参皂苷Rd、三七皂苷Fe和三七皂苷Fd采用一测多评的方法进行含量测定,15批次样品每丸8个成分含量之和在7.6~10.0 mg。8个成分在相应浓度范围内与峰面积的线性关系良好,平均加样回收率分别为99.8%、99.7%、99.1%、99.4%、101.8%、102.0%、99.2%和100.2%,RSD分别为2.3%、2.2%、2.0%、1.5%、1.7%、1.2%、2.4%和2.6%。

结论:

本研究建立的七叶神安滴丸的UPLC指纹图谱及一测多评的含量测定方法,操作简单,重复性好,稳定可靠,可为七叶神安滴丸的质量控制提供更加全面的依据。

目的:

建立七叶神安滴丸的超高效液相色谱(UPLC)指纹图谱鉴别方法,同色谱条件下完成一测多评法同时测定8个指标成分的含量。

方法:

采用Waters CORTECS®T3(150 mm×2.1 mm,1.6 μm)色谱柱,以乙腈-0.1%磷酸水为流动相进行梯度洗脱,流速0.40 mL·min-1,柱温35 ℃,检测波长203 nm。

结果:

建立七叶神安滴丸UPLC指纹图谱,确立了15个共有峰,15批样品相似度均大于0.99。并对其含量较高的8个成分人参皂苷Rb1、人参皂苷Rc、三七皂苷Fc、人参皂苷Rb2、人参皂苷Rb3、人参皂苷Rd、三七皂苷Fe和三七皂苷Fd采用一测多评的方法进行含量测定,15批次样品每丸8个成分含量之和在7.6~10.0 mg。8个成分在相应浓度范围内与峰面积的线性关系良好,平均加样回收率分别为99.8%、99.7%、99.1%、99.4%、101.8%、102.0%、99.2%和100.2%,RSD分别为2.3%、2.2%、2.0%、1.5%、1.7%、1.2%、2.4%和2.6%。

结论:

本研究建立的七叶神安滴丸的UPLC指纹图谱及一测多评的含量测定方法,操作简单,重复性好,稳定可靠,可为七叶神安滴丸的质量控制提供更加全面的依据。

Objective:

To establish the ultra high performance liquid chromatography (UPLC) fingerprint of Qiye Shen’an dropping pills,and to determine the contents of 8 index components by quantitative analysis of multi-components by single marker at the same time.

Methods:

Waters CORTECS® T3 (150 mm×2.1 mm,1.6 μm) chromatography column was used for gradient elution with acetonitrile -0.1% phosphoric acid as mobile phase,flow rate was 0.40 mL·min-1,column temperature was 35 ℃,detection wavelength was 203 nm.

Results:

The UPLC fingerprint of Qiye Shen’an dropping pills was established,8 common peaks were established,and the similarity of 15 batches of samples was greater than 0.99. Ginsenoside Rb1,ginsenoside RC,panax notoginseng saponin FC,ginsenoside Rb2,ginsenoside Rb3,ginsenoside Rd,panax notoginseng saponin Fe and panax notoginseng saponin Fd were determined by quantitative analysis of multi-components by single marker method. The contents of 8 components in 15 batches were between 7.6-10.0 mg per pill. The average recoveries were 99.8%,100.6%,99.8%,101.2%,102.1%,101.9%,99.7% and 100.2% respectively,and the RSDs were 2.3%,2.2%,2.0%,1.5%,1.7%,1.2%,2.4% and 2.6% respectively.

Conclusion:

The UPLC fingerprint of Qiye Shen’an dropping pills and the content determination method of quantitative analysis of multi-components by single marker method established in this study are simple to operation,and have good repeatability,stability and reliability,and can provide a more comprehensive basis for the quality control of Qiye Shen’an dropping pills.

三七为五加科人参属植物,是常用的活血化瘀中药,随着市场需求的增加,综合利用三七各部位,充分开发利用三七具有重要意义。三七茎叶是三七的活性部位之一,有研究表明其主要活性成分三七茎叶总皂苷,具有益气安神、活血止痛、止血、安神之功。七叶神安滴丸为独家生产的品种,其主要成分为三七叶总皂苷,用于心气不足、失眠、心悸、胸痹、心痛或肿瘤、痈肿疮毒及出血症,临床上还用于神经衰弱、偏头痛、失眠等症的治疗,尤其对失眠有显著疗效[1-5]
中药成分复杂,单一指标成分难以全面评价中药质量优劣。指纹图谱具有整体性特点,可在整体上表征中药化学成分,是目前中药制剂质量的有效手段之一。一测多评法实现了采用1个对照品同时测定多个成分含量的目的,解决了中药对照品昂贵、稀缺等问题,同时也简化操作,可多维度控制中药的质量[6-11]。指纹图谱和一测多评含量测定方法相结合,可以很好地解决中药多成分定性定量问题,以及保证产品质量和批间的一致性。
七叶神安滴丸现行质量标准收载于国家药品标准,该标准规定了皂苷类成分的理化鉴别和人参皂苷Rb1、Rb3液相色谱鉴别,以及HPLC法测定人参皂苷Rb3的含量,质控指标单一,不能全面反映产品质量。研究发现,三七茎叶中含有较为丰富的达玛烷型皂苷类成分,在抗抑郁、抗焦虑、调节血脂、保护心脑血管等方面有较好的活性,主要包括人参皂苷Rb1、Rb3、Rc及三七皂苷Fa、Fc、Fe等[12],因此仅采用单一成分评价其质量,缺少科学性。文献已对三七地上地下部分多种成分采用HPLC、UPLC(MS)等检测手段进行了研究[13-20]。本研究对七叶神安滴丸的质量标准进行了全面提高完善,增加了指纹图谱测定项,采用一测多评方法,以人参皂苷Rb3为对照,测定了三七叶中人参皂苷Rb1、人参皂苷Rc、三七皂苷Fc、人参皂苷Rb2、人参皂苷Rb3、人参皂苷Rd、三七皂苷Fe、三七皂苷Fd 8个皂苷类成分的含量,其中人参皂苷Rc、三七皂苷Fc、三七皂苷Fe、三七皂苷Fd、三七皂苷Fe为三七地上部位特有成分,因此建立的方法可用于控制七叶神安滴丸制剂的一致性和质量评价,为三七茎叶类制剂质控指标的建立提供了参考。
超高效液相色谱仪Waters ACQUITY H-CLASS 2台,UltraPerformance LC 1台,Empower3工作站;Waters CORTECS®T3色谱柱,规格均为150 mm×2.1 mm,1.6 μm,柱号分别为01123905015414、01183034215310、01183034215334;十万分之一电子天平(梅特勒-托利多公司)。
对照品人参皂苷Rb1(批号110704-202028,纯度93.1%),人参皂苷Rb2(批号111715-201203,纯度93.8%),人参皂苷Rb3(批号111686-202005,纯度96.5%),人参皂苷Rd(批号111818-201603,纯度92.1%),购自中国食品药品检定研究院。人参皂苷Rc(批号DSTDR001301,纯度99.48%),三七皂苷Fc(批号DSTDR019501,纯度99.58%),三七皂苷Fe(批号DST2010025-037,纯度98.82%),三七皂苷Fd(批号DSTDS013601,纯度99.42%),购自成都德思特生物技术有限公司。
样品共由企业提供,共15批次,为S1~S15。
乙腈为色谱纯,甲醇、磷酸为优级纯,水为超纯水。
采用Waters CORTECS®T3 (150 mm×2.1 mm,1.6 μm)色谱柱,以乙腈(A)-0.1%磷酸水(B)为流动相,梯度洗脱(0~7.0 min,30%A;7.0~11.5 min,30%A→33%A;11.5~16.0 min,33%A→43%A;16~20.0 min,43%A→55%A;20.0~23.0 min,55%A→95%A;23.0~26.0 min,95%A),检测波长203 nm,柱温35 ℃,流速0.40 mL·min-1,进样量1 μL。
取对照品人参皂苷Rb3适量,精密称定,加甲醇制成每1 mL含0.6 mg的溶液,即得。
取重量差异项下的本品6丸,精密称定,置具塞锥形瓶中,精密加入甲醇20 mL,密塞,称量,超声(功率250 W,频率33 kHz)处理10 min,放冷,再称量,用甲醇补足减失的量,摇匀,滤过,取续滤液,即得。
取同一供试品溶液(S12),按照“2.1”项色谱条件进样,仪器精密度6针之间的相似度为1.000,不同序列号色谱柱相似度在0.998~1.000,不同品牌仪器相似度在0.996~0.999,表明方法精密度良好。
按照“2.2.2”项下方法平行制备供试品溶液(S12)6份,按照“2.1”项色谱条件进样分析,每2个供试品之间比较相似度为1.000,表明方法重复性良好。
取同一供试品溶液(S12),分别于0、1、2、4、8、16、24 h进样,记录色谱图。每2针之间相互比较相似度为1.000,表明供试品溶液在24 h内稳定性良好。
采用3批次样品(S10、S11、S12),分别用3根同品牌不同柱号色谱柱,3台超高效液相色谱仪(①H-CLASS;②H-CLASS;③UltraPerformance LC),测定指纹图谱,相似度测定结果RSD均小于1.4%。
使用“中药色谱指纹图谱相似度评价系统(2012.130723版)”进行数据处理,共确定了15个共有峰,其中共有峰峰面积占总峰面积的95%以上。人参皂苷Rb3色谱峰分离度较好,峰面积较稳定,作为S峰,经过与对照品比对,指认出8个成分。对照指纹图谱见图1
以21批次三七叶总皂苷生成的对照图谱为对照图谱进行匹配,计算15批七叶神安滴丸的相似度,相似度均>0.99;样品之间的相互比较,均大于0.98,表明样品不同批次间质量稳定,测定方法准确可靠。见图2
采用校准曲线相对斜率法及浓度法2种方法,以人参皂苷Rb3为对照,测定其他7个指标成分相对于人参皂苷Rb3的校正因子。将8个皂苷类成分混合对照品溶液注入超高效液相色谱仪,提取各成分校准曲线的斜率,以人参皂苷Rb3斜率/待测成分斜率得出校正因子;再分别制成不同浓度的对照品溶液,采用同一台仪器,不同色谱柱和同一色谱柱不同仪器进行测定,考察测定成分在不同色谱柱和不同仪器上的重现性,结果RSD均小于5%,并得出校正因子;经比较,校正因子斜率法和浓度法校正因子测定结果RSD均在2.0%以下。最终确定了7个皂苷类成分相对于人参皂苷Rb3校正因子和相对保留时间,可用于一测多评方法测定七叶神安滴丸中8个皂苷类成分。见图3
分别精密吸取对照品溶液(含人参皂苷Rb1 0.152 6 mg·mL-1,人参皂苷Rc 0.384 6 mg·mL-1,三七皂苷Fc 0.410 1 mg·mL-1,人参皂苷Rb2 0.132 7 mg·mL-1,人参皂苷Rb3 0.783 6 mg·mL-1,人参皂苷Rd 0.070 8 mg·mL-1,三七皂苷Fd 0.224 2 mg·mL-1,三七皂苷Fe 0.505 7 mg·mL-1)0.2、0.4、0.8、1.2、1.5、2.0、2.5 μL,注入UPLC仪;记录色谱图,以进样量为横坐标(X),峰面积为纵坐标(Y),计算回归方程。结果见表1
精密吸取同一供试品溶液(S12)1 μL,连续重复进样6次,8个成分峰面积的RSD<2%,表明方法精密度良好。
精密吸取同一供试品溶液(S12)1 μL,分别于配制后0、1、2、4、8、16、24 h,依法测定,结果表明,供试品溶液在24 h内基本稳定,峰面积的RSD<2%。
按“2.2.2”方法,对同一批号样品(S12),平行制备供试品溶液6份,进行测定,结果8个成分含量分别为9.045 3、25.284 0、27.943 5、9.277 4、48.391 9、4.504 4、20.072 2、50.564 6 mg·g-1,RSD分别为2.8%、2.6%、2.6%、2.6%、2.5%、2.6%、2.5%、2.5%,表明方法重复性良好。
采用加样回收法,精密称取已知含量的同一批样品(S12,含人参皂苷Rb1 9.045 3 mg·g-1、人参皂苷Rc 25.284 0 mg·g-1、三七皂苷Fc 27.943 5 mg·g-1、人参皂苷Rb29.277 4 mg·g-1、人参皂苷Rb3 48.391 9 mg·g-1、人参皂苷Rd 4.504 4 mg·g-1、三七皂苷Fe 20.072 2 mg·g-1、三七皂苷Fd 50.564 6 mg·g-1)0.1、0.15、0.30 g,加入7个混合对照品溶液(含人参皂苷Rb1 0.305 2 mg·mL-1、人参皂苷Rc 0.769 2 mg·mL-1、三七皂苷Fc 0.820 2 mg·mL-1、人参皂苷Rb2 0.265 4 mg·mL-1、人参皂苷Rd 0.141 6 mg·mL-1、三七皂苷Fe 0.448 4 mg·mL-1、三七皂苷Fd 1.011 4 mg·mL-1) 2.5、4.0、8.0 mL,人参皂苷Rb3对照品溶液(浓度为1.475 5 mg·mL-1)2、4、8.0 mL,2种混合对照品溶液(含三七皂苷Fe 0.561 3 mg·mL-1、三七皂苷Fd 1.371 4 mg·mL-1) 1.0、2.0、4.0 mL,加甲醇至20 mL,称量,超声处理(功率250 W,频率33 kHz)10 min,用甲醇补足减失的量,摇匀,滤过,取续滤液,平行制备3份,依法测定,结果表明本方法具有良好的准确度,数据见表2
取3批(S10、S11、S12)样品,分别用3根同品牌不同柱号色谱柱,3台超高效液相色谱仪,对8个成分及总量进行测定,结果RSD<6.0%。
取15批样品,按“2.2.2”项下方法制备供试品溶液,照“2.1”项下条件进样测定,计算得8个成分含量,结果见表3
将15批次样品8个成分的含量测定结果进行皮尔逊相关性分析,得到相关性矩阵,见表4。显著性检验结果P<0.05,各变量之间有显著统计学意义,P<0.01,各变量之间有极其显著统计学意义;当变量为0.8~1.0,0.6~0.8时,相关性为极强相关和强相关。从相关系数矩阵表中可以看出七叶神安滴丸中各成分的相关系数均>0.7,且显著性均P<0.002,说明各成分呈高度相关。
对15批次样品8个成分的含量测定结果进行主成分分析,见图4。提出第一个主成分,可解释87.3%的变量。在得分图(Scores)和载荷图(Loadings)中可以看出,在第一个成分(水平方向)中,与其他7个成分呈正相关,载荷图中各变量的位置影响各样本在得分图中的分布,离Y轴越偏右方向,说明成分含量越高。得分图中相近的样本具有相似的属性。七叶神安滴丸样本S2中人参皂苷Rc、Rb1、Rb2、Rb3和三七皂苷Fc含量较高,样品S13~S15较低;样本S1中人参皂苷Rd和三七皂苷Fd、Fe含量最低。样本S1、S13~S15含量较为接近,样本S3、S5、S6、S10~S12含量较为接近,样本S2、S4、S7~S9含量较为接近。
研究考察了多个品牌和型号的色谱柱10余种,最终选用了Waters CORTECS®T3型号色谱柱(150 mm×2.1 mm,1.6 μm)。对流动相比例也进行了考察,考察流动相A:乙腈;流动相B:①水、②0.1%磷酸水溶液、③0.2%磷酸水溶液,经比较色谱图没有明显差异,最终流动相B选择了0.1%磷酸水溶液。根据使用色谱柱填料的极性,试用不同的洗脱程序,以得到最好的分离效果,比较各色谱柱的分离效果,最终选用正文中的梯度洗脱。比较了柱温30、35、40 ℃时色谱图的分离情况,35 ℃效果最佳。
取滴丸6粒,精密称定,分别精密加入甲醇、90%甲醇水、80%甲醇水、70%甲醇水、60%甲醇水、50%甲醇水,经过比较,无明显差异,最终选用甲醇作为提取溶剂。
经过文献查询[12],皂苷类和三七皂苷类成分,本研究共购得18个人参皂苷和三七皂苷,通过定位发现三七叶中成分与三七花相近,与三七根及茎所含皂苷类成分相差较远,即三七地上和地下部分成分差异比较大,三七根及根茎含三七皂苷R1、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1含量较高,人参皂苷Rb3含量较低,而三七叶皂苷中几乎不含有三七皂苷R1、人参皂苷Rg1、人参皂苷Re,人参皂苷Rb1含量较低,人参皂苷Rb3含量较高。本研究适当调整流动相极性比例,经过定位确定9个皂苷类成分可以用于定量研究,分别为人参皂苷Rb1、人参皂苷Rc、三七皂苷Fc、人参皂苷Rb2、人参皂苷Rb3、人参皂苷Rd、三七皂苷Fe、三七皂苷Fd、20-((S)-人参皂苷F2。后经不同仪器和不同色谱柱比较,发现20-(S)-人参皂苷F2被分离成2个色谱峰,最终确定人参皂苷Rb1、人参皂苷Rc、三七皂苷Fc、人参皂苷Rb2、人参皂苷Rb3、人参皂苷Rd、三七皂苷Fe 8个成分做为指控指标。
采用3批次(S10、S11、S12)样品,分别采用一测多评方法和对照品方法计算8个成分的含量及总量,结果RSD均小于5.0%。
七叶神安滴丸的配伍组成较为简单,作为中药成方,在具有镇痛、镇静、改善睡眠等等方面具有良好的效果,目前七叶神安滴丸市场需求量扩大,应用前景较好。但七叶神安滴丸的质量控制还较为落后,因此提高质量控制水平对提高产品质量,保证用药安全意义重大。本研究采用UPLC法建立了七叶神安滴丸指纹图谱,并用一测多评的方法同时测定其中8个成分的含量,该方法简便易操作,重复性好,能更全面、准确地控制七叶神安滴丸的质量,整体反映了其化学成分特征,且选用的测定成分和活性相关,为其质量综合评价提供了科学依据。
