Objective:

To establish a quantitative 1H-NMR (qHNMR) method for the determination of dexzopiclone tablets content.

Methods:

Proton signal at δ 6.08 of 1,3,5-trimethoxybenzene and δ 8.54,8.38,8.12,7.78 of dexzopiclone were served as the internal standard and quantitative peaks,respectively. DMSO-d6 was employed as the solvent. The qHNMR spectra were acquired at 298 K with 10 s relaxation delay and 32 scanning times.

Results:

The content of dexzopiclone was estimated to be 2.94% (RSD=0.15%)by qHNMR method,which indicated that 3.00 mg of dexzopiclone was in one tablet. The result was consistent with the description of the label.

Conclusion:

qHNMR method can be used to determine the content of dexzopiclone. The method is accurate,simple and efficient.

目的:

建立定量核磁共振氢谱(qHNMR)测定右佐匹克隆含量的方法。

方法:

以1,3,5-三甲氧基苯δ 6.08处质子信号为内标峰,以右佐匹克隆δ 8.54、8.38、8.12、7.78处质子信号为定量峰,以DMSO-d6为溶剂,在恒温298 K,弛豫延迟时间10 s,扫描次数32次条件下采集氢谱。

结果:

定量核磁共振氢谱法测定右佐匹克隆片中右佐匹克隆平均含量为2.94%,即平均每片中含右佐匹克隆3.00 mg,RSD为0.15%,与药物说明标示的量基本一致。

结论:

定量核磁共振氢谱可用于药物中右佐匹克隆含量的测定,并且具有快速、准确以及操作简单等优势。

目的:

建立定量核磁共振氢谱(qHNMR)测定右佐匹克隆含量的方法。

方法:

以1,3,5-三甲氧基苯δ 6.08处质子信号为内标峰,以右佐匹克隆δ 8.54、8.38、8.12、7.78处质子信号为定量峰,以DMSO-d6为溶剂,在恒温298 K,弛豫延迟时间10 s,扫描次数32次条件下采集氢谱。

结果:

定量核磁共振氢谱法测定右佐匹克隆片中右佐匹克隆平均含量为2.94%,即平均每片中含右佐匹克隆3.00 mg,RSD为0.15%,与药物说明标示的量基本一致。

结论:

定量核磁共振氢谱可用于药物中右佐匹克隆含量的测定,并且具有快速、准确以及操作简单等优势。

Objective:

To establish a quantitative 1H-NMR (qHNMR) method for the determination of dexzopiclone tablets content.

Methods:

Proton signal at δ 6.08 of 1,3,5-trimethoxybenzene and δ 8.54,8.38,8.12,7.78 of dexzopiclone were served as the internal standard and quantitative peaks,respectively. DMSO-d6 was employed as the solvent. The qHNMR spectra were acquired at 298 K with 10 s relaxation delay and 32 scanning times.

Results:

The content of dexzopiclone was estimated to be 2.94% (RSD=0.15%)by qHNMR method,which indicated that 3.00 mg of dexzopiclone was in one tablet. The result was consistent with the description of the label.

Conclusion:

qHNMR method can be used to determine the content of dexzopiclone. The method is accurate,simple and efficient.

定量核磁技术用于定量分析的依据是化学环境不同的氢原子核的共振峰面积只与氢原子数有关,不需特定的对照品,快速准确,特异性强,特别适合无对照品的新药含量测定,已先后被美国药典、欧洲药典、日本药局方和2020年版《中华人民共和国药典》(简称《中国药典》)收录[1-8]
右佐匹克隆化学名为(+)-(7S)-6-(5-氯-2吡啶基)-7-[(4-甲基哌嗪-1-基)甲酰氧基]-5,6-二氢吡咯并[3,4-b]吡嗪- 5-酮(见图1),是吡咯环酮类短效催眠药,用于治疗失眠症。霍美娣等[9]对其化学结构进行了确证,刘惠军等[10]对佐匹克隆原料及片剂有关物质的HPLC测定进行了研究。《中国药典》采用电位滴定法(通则0701)及紫外-可见分光光度法(通则0101)测定右佐匹克隆含量,采用高效液相色谱法(通则0512)测定有关物质[11]。目前,定量核磁共振氢谱法测定右佐匹克隆含量未见报道。本文以1,3,5-三甲氧基苯为内标,建立了基于定量1H-NMR测定右佐匹克隆片中右佐匹克隆含量的方法。研究了溶剂、内标物及NMR实验参数对测量结果的影响,为该物质的含量测定提供了一种新思路,建立了一种简单、方便、快捷的测定右佐匹克隆含量的方法。
Bruker Avance Ⅲ HD 700 MHz NMR谱仪(Bruker公司);高效液相HPLC型号 Agilent1260(安捷伦科技有限公司),YMD-Pack ODS-A(250 mm×4.6 mm,5 μm);MestReNova 14.3.0数据处理软件(Mestrelab Research S.L.公司);ME204E万分之一电子天平(梅特勒公司)。
对照品1,3,5-三甲氧基苯(批号510137-201601,含量99.9%)、右佐匹克隆(批号100871-200801,含量100%),均购自中国食品药品检定研究院。右佐匹克隆片(商品名:伊坦宁,成都康弘药业集团股份有限公司,批号3D006,规格3 mg·片-1);DMSO-d6 (99.9%,CIL公司),色谱乙腈(北京迈瑞达公司),其他试剂均为分析纯。
分别精密称取一定量的1,3,5-三甲氧基苯和右佐匹克隆片待测样品,置于同一离心管,加入DMSO-d6 0.6 mL,超声(功率70 W)溶解30 min,离心取上清液转移至5 mm核磁管中。
采用zg30脉冲序列,谱宽1 409 7.74 Hz(谱仪对应的质子共振频率为70 0.18 Hz)、中心频率为490 1.26 Hz、θ=30°、时间域数据点为64 k,测定温度为298 K,延迟时间为10 s,采样次数为32次,每份样品平行测定5次取平均值,采用线宽因子为0.3 Hz的窗函数对原始的自由感应衰减(FID)信号进行傅里叶变换得到频域谱,对谱图进行手动相位和基线校正及化学位移定标处理,同时对照参考文献[9]对右佐匹克隆的1H、13C NMR信号进行归属,见表1
采用下列公式[2]计算待测样品中右佐匹克隆的含量。
Ix为右佐匹克隆片待测样品定量峰的积分面积,IIS为1,3,5-三甲基苯内标定量峰的积分面积;NxNIS分别为右佐匹克隆片待测样品和1,3,5-三甲基苯内标定量峰包含的质子数(Nx=4,NIS=3);MxMIS分别为右佐匹克隆待测样品和1,3,5-三甲基苯的相对分子质量(Mx=388.81,MIS=168.19);mxmIS分别为右佐匹克隆待测样品和1,3,5-三甲基苯内标的称样量;PxPIS分别为右佐匹克隆待测样品和1,3,5-三甲基苯内标的质量分数(PIS=99.9%)。
按“2. 1”项方法制备供试品溶液,在“2. 2”项条件下进行检测,并将1H-NMR谱中δ 8.54、8.38、8.12、7.78信号作为其定量峰,1,3,5-三甲氧基苯1H-NMR谱中δ 6.08的信号作为其内标的定量峰,按上述公式计算,每个定量峰积分面积计算的平均值作为结果,测得待测样品中右佐匹克隆的含量均值为2.94%,折算成质量即平均每片右佐匹克隆片(伊坦宁)中含右佐匹克隆3.00 mg,RSD为0.15%,见表2
取同一待测样品溶液分别在1 d内的第0、6、12和24 h进行NMR测定,结果显示待测样品中右佐匹克隆与1,3,5-三甲氧基苯内标的峰面积之比的RSD为0.090%(n=4)。表明待测样品的DMSO-d6溶液室温放置24 h稳定。
精密称取内标物1,3,5-三甲氧基苯29.80 mg,用DMSO-d6 1 mL溶解,得到29.80 mg·mL-1的内标液;精密称取右佐匹克隆对照品25.10 mg,精密加入内标液0.1 mL,用DMSO-d6制成质量浓度为3.138、4.701、6.275、7.844、9.413、10.981、12.550 mg·mL-1的一系列对照品溶液。按照“2. 2”项条件下进行1H-NMR分析。以右佐匹克隆对照品与1,3,5-三甲氧基苯的峰面积之比Y为纵坐标,右佐匹克隆对照品与1,3,5-三甲氧基苯的质量比X为横坐标进行线性回归,得回归方程(n=7):
右佐匹克隆对照品与内标物的质量比在0.421~1.684与峰面积之比呈线性关系。
色谱条件:安捷伦1260高效液相色谱仪,YMD-Pack ODS-A(250 mm×4.6 mm,5 μm)色谱柱,流动相为水-乙腈(90:10→5:95)进行梯度洗脱(流动相体系中含有十万分之一的甲酸,洗脱时间30 min),检测波长280 nm,流速1 mL·min-1,柱温25 ℃,进样量5 μL。
溶液的制备:精密称取右佐匹克隆对照品12.62 mg,用甲醇溶解并配制成浓度为0.505、0.252、0.126、0.063、0.032、0.016 mg·mL-1的标准品溶液;精密称取右佐匹克隆待测样品30 mg,用甲醇配制成浓度为1.500 mg·mL-1的待测液。
用HPLC外标法测定待测样品中右佐匹克隆的含量为3.04%,测定结果与qHNMR方法基本一致。
合适的溶剂除了对样品和内标均有良好的溶解性外,其残留的信号峰应不干扰所分析样品的信号峰。右佐匹克隆易溶于二甲亚砜或氯仿,较易溶于醋酸,难溶于甲醇、丙酮或乙腈,极难溶于乙醚或异丙醇,几乎不溶于水。试验中用3种常用的氘代试剂进行提取,发现CDCl3提取成分复杂,谱图略显杂乱;DMSO-d6溶剂中右佐匹克隆信号峰清晰,分辨率好;CD3OD溶剂提取谱图清晰,由于溶剂效应信号分离度不如DMSO-d6,综合考虑右佐匹克隆的溶解度,本文最终确定用DMSO-d6为氘代试剂,见图2
在核磁定量实验中,延迟时间(d1)必须设定的足够长(≥5T1),才能使原子核完全弛豫,确保被积分的信号强度与原子核数目呈正比[12]。为考量d1对实验结果的影响,针对同一待测样品,分别将d1设置为1、2、5、10、15、20 s,结果表明:当d1≥10 s后,待测样品及内标的定量峰的积分面积不再变化[2],为同时满足d1 ≥ 5T1 且节省采样时间,本文选择d1 为10 s。
ns的多少直接影响NMR的信噪比:ns越大,信噪比越高,定量越准确。对于qHNMR,一般要求信噪比(S/N)≥250[13]。但ns越大,采样时间也越长。为考量ns对实验结果的影响,针对同一待测样品,分别将ns设定为8、16、32和64。结果表明:700 MHz液氦超低温探头灵敏度较高,S/N都能达到要求,综合考量测试时间及定量结果的准确性,本文选择ns为32。
理想的内标应满足以下条件:(1)化学惰性不与样品反应;(2)非挥发性、稳定、溶于常规使用的NMR试剂;(3)内标的定量峰与待测样品的定量峰分离良好,分子量与待测样品接近等。待测样品及内标的定量峰均应选择不受其他信号干扰的孤立峰,若化合物中多个信号均符合上述条件,最好采用其积分面积之和,取其平均值作为结果以减小误差。处于不同化学环境的质子NMR弛豫时间不同,在qHNMR实验中,待测样品和内标最好选择处于相似化学环境的质子作为定量峰。由图3可知,右佐匹克隆位于δ 8.54、8.38、8.12、7.78信号峰相对独立,与1,3,5-三甲氧基苯位于δ 6.08的信号均无重合。因此本文选定δ 8.54(d,1H)、8.38(d,1H)、8.12(dd,1H)、7.78(s,1H)为待测样品的定量峰,δ 6.08(s,3H)为内标1,3,5-三甲氧基苯的定量峰。
HPLC外标法所需实验时间长并且高度依赖标准品,而qHNMR则无需标准品,降低了对标准品依赖程度,对样品无破坏性,操作简便,重现性好等优点,已广泛用于药物的常量分析。在文献调研的过程发现各国药典尚未收载右佐匹克隆qHNMR含量测定方法也未见文献报道,故对其研究具有重要意义。本文基于1H-NMR技术,以1,3,5-三甲氧基苯为内标、DMSO-d6为溶剂,对氢核磁共振定量法进行了方法学研究,核磁定量方法的测定结果与HPLC法基本一致,与药品说明书吻合。本文所建立的qHNMR方法操作简便,快速准确,不破坏样品,为右佐匹克隆含量测定提供了一种新的手段,也为其质量控制研究提供了实验依据。
