Objective:

To establish a liquid-liquid extraction method for the determination of genotoxic impurities N-nitrosodimethylamine(NDMA) and N-nitrosodiethylamine(NDEA) in chondroitin sulfate sodium by gas chromatography-mass spectrometry (GC-MS).

Methods:

The chromatographic column was Thermo TG WAXMS (30 m × 0.25 mm,0.25 μm). The column temperature was maintained at 40 ℃ for 1 min,increased to 240 ℃ at 25 ℃·min-1 and maintained for 2 min. The inlet temperature was 220 ℃. The carrier gas was high-purity helium,and the flow rate was 1.0 mL·min-1. The ion source of mass spectrometry was EI source. The electron energy was 70 eV. The ion source temperature was 280 ℃ and the transmission line temperature was 240 ℃. Injection volume was 1 μL.

Results:

The LOD of NDMA was 0.64 ng·mL-1. Experiment showed a good linear relationship between peak area and concentration in the range of 4-16 ng·mL-1,and the correlation r was 0.999 3. The LOD of NDEA was 0.176 ng·mL-1 with a good linear relationship between peak area and concentration within the range of 1.1-4.4 ng·mL-1. The correlation r was 0.999 6. NDMA and NDEA were not detected in chondroitin sulfate sodium.

Conclusion:

The method is simple,specific and can be used to determine the genotoxic impurities NDMA and NDEA in chondroitin sulfate sodium.

目的:

建立一种液液萃取法,采用气相色谱-质谱联用(GC-MS)技术,测定硫酸软骨素钠原料中的基因毒杂质N-亚硝基二甲胺(NDMA)与N-亚硝基二乙胺(NDEA)。

方法:

目的:

建立一种液液萃取法,采用气相色谱-质谱联用(GC-MS)技术,测定硫酸软骨素钠原料中的基因毒杂质N-亚硝基二甲胺(NDMA)与N-亚硝基二乙胺(NDEA)。

方法:

采用Thermo TG-WAXMS色谱柱(30 m×0.25 mm,0.25 μm);柱温在40 ℃保持1 min,以25 ℃·min-1升至240 ℃,保持2 min;进样口温度为220 ℃;载气为高纯氦,流速为1.0 mL·min-1。质谱的离子源为EI源;电子能量为70 eV;离子源温度为280 ℃;传输线温度为240 ℃;进样量为1 μL。

结果:

NDMA的检测限为0.64 ng·mL-1,在4~16 ng·mL-1范围内峰面积与浓度呈良好的线性关系,相关系数r为0.999 3。NDEA的检测限为0.176 ng·mL-1,在1.1~4.4 ng·mL-1范围内峰面积与浓度呈良好的线性关系,相关系数r为0.999 6。硫酸软骨素钠中均未检出NDMA与NDEA。

结论:

该法操作简便,专属性强,可以用于硫酸软骨素钠中的基因毒杂质NDMA与NDEA的测定。

Objective:

To establish a liquid-liquid extraction method for the determination of genotoxic impurities N-nitrosodimethylamine(NDMA) and N-nitrosodiethylamine(NDEA) in chondroitin sulfate sodium by gas chromatography-mass spectrometry (GC-MS).

Methods:

The chromatographic column was Thermo TG WAXMS (30 m × 0.25 mm,0.25 μm). The column temperature was maintained at 40 ℃ for 1 min,increased to 240 ℃ at 25 ℃·min-1 and maintained for 2 min. The inlet temperature was 220 ℃. The carrier gas was high-purity helium,and the flow rate was 1.0 mL·min-1. The ion source of mass spectrometry was EI source. The electron energy was 70 eV. The ion source temperature was 280 ℃ and the transmission line temperature was 240 ℃. Injection volume was 1 μL.

Results:

The LOD of NDMA was 0.64 ng·mL-1. Experiment showed a good linear relationship between peak area and concentration in the range of 4-16 ng·mL-1,and the correlation r was 0.999 3. The LOD of NDEA was 0.176 ng·mL-1 with a good linear relationship between peak area and concentration within the range of 1.1-4.4 ng·mL-1. The correlation r was 0.999 6. NDMA and NDEA were not detected in chondroitin sulfate sodium.

Conclusion:

The method is simple,specific and can be used to determine the genotoxic impurities NDMA and NDEA in chondroitin sulfate sodium.

N-亚硝基二甲胺(N-nitrosodimethylamine,NDMA)与N-亚硝基二乙胺(N-nitrosodiethylamine,NDEA)属于N-亚硝胺类化合物,N-亚硝胺类杂质是基因毒性杂质同时具有高致癌风险,在人用药品技术要求国际协调理事会(The International Council for Harmonisation of Technical Requirements for Pharmaceutical for Human Use,ICH)ICH M7(R1)指南中列为基因毒性杂质中的“关注队列”(cohort of concern)[1]。2018年7月,欧洲药品管理局在缬沙坦原料中检测出了NDMA,接着又在其他药物中检测出N-亚硝胺类杂质,如二甲双胍、雷尼替丁等,引起各国的高度重视。国家药品监督管理局(NMPA)、欧洲药品管理局(EMA)、美国食品药品监督管理局(FDA)均发布了人用药中N-亚硝胺类杂质控制的指导意见[2-4],指出NDMA与NDEA每日最大用量分别不得超过96与26.5 ng。
硫酸软骨素钠是动物软骨组织中提取制得的硫酸化链状糖胺聚糖钠盐,主要来自猪、鸡、牛、鲨鱼等动物的透明软骨[5]。硫酸软骨素钠具有抗关节炎、抗氧化、抗动脉粥样硬化、调节肠道菌群等作用[6],其制备过程主要包括软骨降解、核心蛋白水解、去蛋白、回收以及纯化等[7]
动物来源生产的硫酸软骨素钠的提取主要是在水溶液中进行[8],而NDMA与NDEA作为消毒副产物极易在水中残留[9],且各种预处理去除工艺均不能完全去除NDMA与NDEA[10]。FDA行业指南[4]中也特别提到原料药(active pharmaceutical ingredients,API)生产中的工艺用水存在含有亚硝胺盐的可能性,从而会导致API生产受到亚硝胺污染。同时,作为硫酸软骨素钠来源的猪、鸡、牛等动物的饲料可能含有亚硝酸盐类物质,这些物质有可能残留在动物体内,进一步生成亚硝胺[11-12],会将NDMA与NDEA引入硫酸软骨素钠中。但是,关于硫酸软骨素钠中NDMA与NDEA的质量控制在现有研究中却未见报道。
本文根据硫酸软骨素钠易溶于水,不溶于乙醇、丙酮或冰醋酸等有机溶剂[13]的特点,开发了一种液液萃取法,采用气相色谱-质谱联用(GC-MS)技术测定硫酸软骨素钠原料中的基因毒杂质NDMA与NDEA,并对市面上3个生产厂家的原料开展检测,以期为硫酸软骨素钠质量安全控制体系的完善提供参考。
Trace 1300GC-ISQ7000气相色谱-质谱联用仪(Thermo Scientific公司);BT25S十万分之一电子天平,BSA224S万分之一电子天平(赛多利斯公司);Milli-Q IQ 7000去离子水发生装置(Millipore公司);WH-2涡旋混合仪(上海楚定分析仪器有限公司);KH-300DB数控超声波清洗器(昆山禾创超声仪器有限公司)。
标准物质:NDMA(1 000 μg·mL-1,批号A2102018)和NDEA(2 000 μg·mL-1,批号A1909202),规格均为每支1 mL,溶剂均为甲醇,均来自坛墨质检-标准物质中心。
色谱纯:二氯甲烷(TEDIA公司)、三氯甲烷[永华化学科技(江苏)有限公司]、正己烷(TEDIA公司),甲醇(默克公司)。
硫酸软骨素钠分别来自S、X和Z公司,共5批,各批号分别命名为S1、X2、X3、Z4、Z5。
气相色谱:色谱柱采用Thermo TG-WAXMS(30 m×0.25 mm×0.25 μm,固定液为聚乙二醇);采用程序升温(起始柱温为40 ℃,保持1 min,以25 ℃·min-1升至240 ℃,保持2 min);进样口(温度为220 ℃),载气为高纯氦气,流速为1.0 mL·min-1,不分流进样,进样量1 μL。
质谱:采用电子轰击离子化(EI),电离电压为70 eV,离子源温度280 ℃,质谱传输线温度240 ℃,MS四极杆温度150 ℃,T-SIM模式。NDMA与NDEA的定量离子m/z分别为74和102,定性离子m/z分别为42、43和42、44。
精密量取NDMA、NDEA各1.0 mL,分别置于100 mL量瓶中,用甲醇稀释至刻度,摇匀,制得10 μg·mL-1NDMA和20 μg·mL-1NDEA对照品储备液。
精密量取NDMA与NDEA标准储备液适量,用二氯甲烷稀释并制成每1 mL含NDMA 8 ng、含NDEA 2.2 ng的混合对照品溶液,摇匀,即得。
精密称取硫酸软骨素钠1.2 g置具塞离心管中,精密加入水4.0 mL,涡旋使分散,超声(频率为40 kHz、功率为100 W)至硫酸软骨素钠完全溶解,放至室温,再加入二氯甲烷4 mL,涡旋5 min,静置分层,取二氯甲烷层,即得。
取具塞离心管,精密加入水4.0 mL,二氯甲烷4 mL,涡旋5 min,静置分层,取二氯甲烷层,即得。
取混合对照品溶液、二氯甲烷、空白溶剂、硫酸软骨素钠供试品液,进样测定,记录色谱图。选择离子(T-SIM)色谱图见图1,专属性结果见图2。由图2可知,该方法的专属性较强。
取混合对照溶液,连续进样5针,记录色谱图。测定NDMA的方法精密度试验结果RSD为2.6%,NDEA的方法精密度试验结果RSD为3.2%。从总体上来看,仪器稳定,精密度良好,可以进行样品分析。
精密量取NDMA与NDEA标准储备液适量,用二氯甲烷稀释并制成每1 mL含NDMA为400 ng、含NDEA为110 ng的混合对照储备溶液,摇匀。
精密量取上述混合对照品储备溶液适量,用二氯甲烷分别稀释并配制成含NDMA 4、6、8、12、16 ng·mL-1与NDEA 1.1、1.6、2.2、3.3、4.4 ng·mL-1系列浓度的标准曲线溶液,摇匀,进样测定,记录色谱图,以质量浓度X为横坐标,峰面积Y为纵坐标进行线性回归。得NDMA和NDEA回归方程:
结果表明,NDMA和NDEA质量浓度分别在4~16 ng·mL-1和1.1~4.4 ng·mL-1与峰面积呈良好的线性关系。
以信噪比S/N≥3为检测限,以信噪比S/N≥10为定量限。NDMA的检测限为0.64 ng·mL-1,定量限为1.60 ng·mL-1;NDEA的检出限为0.176 ng·mL-1,定量限为0.44 ng·mL-1
精密量取NDMA与NDEA对照品储备液适量,分别用水稀释并制成低(含NDMA 4 ng·mL-1与NDEA 1.1 ng·mL-1)、中(NDMA 8 ng·mL-1与NDEA 2.2 ng·mL-1)、高(NDMA 16 ng·mL-1与NDEA 4.4 ng·mL-1)3个浓度的回收率混合对照品储备溶液,摇匀,备用。
精密称取硫酸软骨素钠(批号X3)1.2 g,置具塞离心管中,分别精密加入低、中、高3个浓度的回收率混合对照储备溶液4.0 mL,在“2.2.3”项从涡旋使分散开始操作,各平行制备3份,即得低、中、高3个浓度的回收率溶液。同时取“2.2.2”项的混合对照品溶液,进样测定,记录色谱图,结果见表12,表明回收率满足测定要求。
精密量取NDMA与NDEA对照品储备液适量,用水稀释并制成含NDMA 8 ng·mL-1与NDEA 2.2 ng·mL-1的加标混合对照品储备溶液,摇匀,备用。精密称取硫酸软骨素钠(批号X3)1.2 g,置具塞离心管中,分别精密加入加标混合对照品储备液4.0 mL,在“2.2.3”项从涡旋使分散开始操作,平行配制6份,即得重复性试验供试溶液;另一名分析人员于不同自然日,按“重复性试验供试溶液”配制方法,平行配制6份,即得中间精密度供试溶液。同时分别取“2.2.2”项的混合对照品溶液,进样测定,记录色谱图。结果表明,重复性试验中,NDMA和NDEA的平均回收率分别为85.3%和105.7%,RSD分别为2.3%和3.4%;中间精密度试验中,NDMA和NDEA的平均回收率分别为88.3%和107.1%,RSD分别为2.3%和3.8%。说明该方法重复性、精密度良好。
取混合对照品溶液和“2.3.5”项中浓度回收率溶液,分别在0、1、3、5、7、9、11、15、19、23、24 h测定峰面积。并以0 h峰面积为标准进行计算。对照品溶液在24 h内,NDMA和NDEA峰面积的RSD分别为1.9%和2.4%;中浓度回收率溶液在23 h内,NDMA和NDEA峰面积的RSD分别为2.4%和3.9%。结果表明混合对照品溶液在24 h内,中浓度回收率溶液在23 h内稳定性良好。
采用单因素法,参照“2.1”项测定条件,分别改变其中的升温速率(25±2)℃·min-1、流速(1.0±0.1)mL·min-1、进样口温度(220±10)℃,取X3批次样品6份,按“2.2.3”项,制备硫酸软骨素钠供试品溶液,进样测定NDMA和NDEA的含量,记录色谱图,并对测定结果进行比较。在各测定条件下,NDMA和NDEA均未检出,表明在色谱条件有小的变动时,测定结果不受影响。
按“2.2.3”项制备硫酸软骨素钠供试品溶液,进样测定,记录色谱图。结果3个企业的5批硫酸软骨素钠中均未检出NDMA与NDEA。
欧洲医药管理局(EMA)在2020年6月25日颁布的《Nitrosamine impurities in human medicinal products》[14]中规定:NDMA的PDE值为96.0 ng·d-1,NDEA的PDE值为26.5 ng·d-1
硫酸软骨素钠片的每日最大用量为3.6 g,硫酸软骨素钠注射液的每日最大用量为80 mg,因此,以硫酸软骨素钠片的每日最大用量3.6 g来计算NDMA和NDEA的限度值。通过溶解度试验得知,每1 mL水中能溶解0.3 g硫酸软骨素钠,因此本研究中的限度为:0.3 g·mL-1的硫酸软骨素钠溶液中含NDMA不得超过8 ng·mL-1,含NDME不得超过2.2 ng·mL-1
采用单因素法依次优化萃取溶剂(二氯甲烷、三氯甲烷、正己烷)和涡旋时间(3、5、10、20 min)2个影响因素。结果见图3图4。结果表明,萃取效果最好的是二氯甲烷与三氯甲烷,NDMA与NDEA的萃取回收率分别在85.0%~115.0%,RSD均<5%。由于NDMA与NDEA在正己烷中萃取效率低,而三氯甲烷为易制毒试剂,不容易购买,因此选择二氯甲烷作为萃取溶剂。
萃取时,萃取溶剂和水相充分接触,是萃取的必要条件之一,因此实验考察了不同涡旋时间对萃取回收率的影响。结果表明,涡旋5 min后萃取回收率达到平台期,NDMA与NDEA的萃取回收率分别为86.6%和105.9%,RSD<5%。考虑到工作效率,选择涡旋5 min作为萃取条件。
本研究建立了一种液液萃取法,采用气相色谱-质谱联用(GC-MS)技术,测定硫酸软骨素钠中的基因毒性杂质NDMA与NDEA。本研究选择二氯甲烷作为萃取溶剂,避免了N,N-二甲基甲酰胺、N-甲基吡咯烷酮等酰胺类高沸点溶剂的使用,这些溶剂在高温下容易产生亚硝胺类杂质[2],干扰测定。通过液液萃取,避免原料药如雷尼替丁等因检测温度过高而产生的亚硝胺类降解杂质[15],干扰测定。目前,已报道的大多数化合物中亚硝胺类杂质测定采用LC-MS/MS技术[16-18],而GC-MS技术的使用,可以降低成本,节约资源。
综上所述,本方法为高水溶性原料,如硫酸软骨素钠、盐酸二甲双胍、盐酸雷尼替丁等需要进行亚硝胺类杂质检测的样品提供了新的研究思路。
