Objective:

To establish an HPLC method for the determination of six related substances in N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine.

Methods:

The analysis was conducted on YMC Triart C18(250 mm×4.6 mm,3μm) column,the mobile phase was consisted with 0.1% trifluoroacetic acid in water(A) and 0.1% trifluoroacetic acid in acetonitrile(B)at the flow rate of 1.0 mL·min-1. The column temperature was set 30 ℃,the detection wavelength was 265 nm and the injection volume was 10 μL.

Results:

N-Fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine had good separation from the adjacent impurity peaks;The resolution of impurity1- 6 was greater than 1.5,and showed a good linear relationship (r≥0.999) in the corresponding mass concentration range,the detection limit of impurity 1-6 was 0.03 μg·mL-1,and the quantitative limit was 0.06 μg·mL-1,the average recovery rate (n=9) of impurity 1-6 was in the range of 97.6%-98.8%. The results of the three batches of N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine showed that the contents of impurity 2,impurity 4 were <0.2% and <0.1%,respectively,and the other four impurities were not detected,and content of the total impurity was <1%.

Conclusion:

This method has good resolution,high sensitivity and strong specificity,and is suitable for the determination of related substances in N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine.

目的:

建立高效液相色谱同时测定N-芴甲氧羰基-O-叔丁基-L-苏氨酸中6个特定杂质的方法。

方法:

采用YMC Triart C18(250 mm×4.6 mm,3 μm)色谱柱,以0.1%三氟乙酸水溶液为流动相A,以0.1%三氟乙酸乙腈溶液为流动相B,流速1.0 mL·min-1,梯度洗脱,检测波长265 nm,柱温30 ℃,进样体积10 μL。

结果:

N-芴甲氧羰基-O-叔丁基-L-苏氨酸与相邻杂质峰的分离良好;6个杂质分离度均大于1.5;且在相应质量浓度范围内呈现良好的线性关系(r≥0.999);6个杂质检测限和定量限分别约为0.03 μg·mL-1和0.06 μg·mL-1;6个杂质的平均回收率(n=9)在97.6%~98.8%范围内。3批N-芴甲氧羰基-O-叔丁基-L-苏氨酸测定结果显示,杂质1的含量<0.2%,杂质4的含量<0.1%,其他4种杂质未检出,总杂含量<1%。

结论:

本方法分离度好,灵敏度高,专属性强,适用于N-芴甲氧羰基-O-叔丁基-L-苏氨酸中有关物质的检测。

目的:

建立高效液相色谱同时测定N-芴甲氧羰基-O-叔丁基-L-苏氨酸中6个特定杂质的方法。

方法:

采用YMC Triart C18(250 mm×4.6 mm,3 μm)色谱柱,以0.1%三氟乙酸水溶液为流动相A,以0.1%三氟乙酸乙腈溶液为流动相B,流速1.0 mL·min-1,梯度洗脱,检测波长265 nm,柱温30 ℃,进样体积10 μL。

结果:

N-芴甲氧羰基-O-叔丁基-L-苏氨酸与相邻杂质峰的分离良好;6个杂质分离度均大于1.5;且在相应质量浓度范围内呈现良好的线性关系(r≥0.999);6个杂质检测限和定量限分别约为0.03 μg·mL-1和0.06 μg·mL-1;6个杂质的平均回收率(n=9)在97.6%~98.8%范围内。3批N-芴甲氧羰基-O-叔丁基-L-苏氨酸测定结果显示,杂质1的含量<0.2%,杂质4的含量<0.1%,其他4种杂质未检出,总杂含量<1%。

结论:

本方法分离度好,灵敏度高,专属性强,适用于N-芴甲氧羰基-O-叔丁基-L-苏氨酸中有关物质的检测。

Objective:

To establish an HPLC method for the determination of six related substances in N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine.

Methods:

The analysis was conducted on YMC Triart C18(250 mm×4.6 mm,3μm) column,the mobile phase was consisted with 0.1% trifluoroacetic acid in water(A) and 0.1% trifluoroacetic acid in acetonitrile(B)at the flow rate of 1.0 mL·min-1. The column temperature was set 30 ℃,the detection wavelength was 265 nm and the injection volume was 10 μL.

Results:

N-Fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine had good separation from the adjacent impurity peaks;The resolution of impurity1- 6 was greater than 1.5,and showed a good linear relationship (r≥0.999) in the corresponding mass concentration range,the detection limit of impurity 1-6 was 0.03 μg·mL-1,and the quantitative limit was 0.06 μg·mL-1,the average recovery rate (n=9) of impurity 1-6 was in the range of 97.6%-98.8%. The results of the three batches of N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine showed that the contents of impurity 2,impurity 4 were <0.2% and <0.1%,respectively,and the other four impurities were not detected,and content of the total impurity was <1%.

Conclusion:

This method has good resolution,high sensitivity and strong specificity,and is suitable for the determination of related substances in N-fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine.

多肽类临床药物是治疗多种疾病的大宗基础性药物,该类药物约占全世界总临床药物的5%~8%左右。多肽类药已经用于各种疾病的治疗[1-6]。多肽药物的合成工艺主要为经典的液相合成和固相合成,其中固相合成技术日趋成熟[7-8]。保护氨基酸是固相合成多肽的起始物料之一,其质量对多肽的质量有重要的影响。吴一凡[9-10]等在审评中发现申请人在进行固相合成多肽的研发时多关注于合成策略及保护氨基酸的投料比、裂解剂的选择,而对起始物料-保护氨基酸的研究不够全面,具体表现为保护氨基酸的纯度检测方法专属性差(如仅仅使用TLC法进行杂质控制)或限度过于宽松,无法有效控制起始物料的质量,也无法有效保证原料药的质量。多肽合成过程中,由起始物料引入的杂质所产生的相关杂质在纯化过程中可能与目标肽共洗脱,因此杂质很难减少或消除。为了尽量减少对终产物的潜在影响,这类杂质的限度应严格控制[11]。国家药品监督管理局2023年出台的《化学合成多肽药物药学研究技术指导原则》[12]中指出,由于保护氨基酸中的杂质可能伴随化学反应而发生杂质传递和转化,继而增加多肽药物的提纯难度,应增加必要的杂质研究,同时制定合理的控制限度。应结合保护氨基酸的合成工艺确定杂质谱组成。由此可见,保护氨基酸中杂质控制非常重要。
N-芴甲氧羰基-O-叔丁基-L-苏氨酸(N-Fluorenylmethoxycarbonyl-O-tert-butyl-L-threonine),是保护氨基酸中的一种化合物,其制备工艺比较复杂,结合工艺路线及反应机理,N-芴甲氧羰基-O-叔丁基-L-苏氨酸中可能含有的杂质为Fmoc-Thr-OH(杂质1)、Fmoc-β-Ala-OH(杂质2)、Fmoc-β-Ala-Thr(tBu)-OH(杂质3)、Fmoc-Thr(tBu)-Thr(tBu)-OH(杂质4)、Fmoc-β-Ala-β-Ala-OH(杂质5)、Fmoc-Ala-Ala-OH(杂质6),结构式见图1,其中,杂质1为侧链保护基丢失产生,杂质2、杂质3、杂质4为引入Fmoc保护基时产生的杂质,杂质5、杂质6为起始原料引入。多肽药物质量控制的文献多有报道[13-15],但保护氨基酸质量控制的文章鲜有报道[16-18],未检索到同时测定N-芴甲氧羰基-O-叔丁基-L-苏氨酸中6个杂质的文献。本文开发了一种高效液相色谱方法,同时测定了N-芴甲氧羰基-O-叔丁基-L-苏氨酸中的6个杂质,为保护氨基酸中杂质控制提供参考。
Thermo Ultimate 3000高效液相色谱仪配Chromeleon Console工作站(Thermo公司);MS205DU十万分之一电子天平(Mettler Toledo公司);Milli-Q超纯水机(默克密理博公司)。
N-芴甲氧羰基-O-叔丁基-L-苏氨酸(批号GLS221008-36202、GLS220719-36202、GLS220524-36202-1)和杂质1对照品(批号GLS220606-36201,含量93.26%)、杂质2对照品(批号GLS210529-35003,含量99.39%)、杂质3对照品(批号GLS220605-35068,含量99.07%)、杂质4对照品(批号GLS210302-36242,含量98.68%)、杂质5对照品(批号GLS220322-35073,含量96.65%)、杂质6对照品(批号GLS220927-J10867,含量98.52%),均购自吉尔生化(上海)有限公司;三氟乙酸(色谱级,TCI公司),乙腈(色谱级,Merck公司),纯化水(自制)。
色谱柱为YMC Triart C18柱(250 mm×4.6 mm,3 μm),流动相A为0.1%三氟乙酸水溶液,流动相B为0.1%三氟乙酸乙腈溶液,洗脱程序见表1,检测波长为265 nm,流速为1.0 mL·min-1,柱温为30 ℃,进样量为10 μL。
乙腈-水(50:50)为空白溶剂。
精密称定杂质1~6对照品各约12.5 mg,分别置25 mL量瓶中,加空白溶剂溶解并定容,摇匀,制成各杂质质量浓度均为0.5 mg·mL-1的杂质对照品储备液。
精密量取“2.1.2”项各杂质对照品储备液各100 μL,置同一10 mL量瓶中,用空白溶剂稀释并定容至刻度,摇匀,制成各杂质质量浓度均为5 μg·mL-1的混合对照品溶液。
取“2.1.2”项各杂质对照品储备液各100 μL,分别置10 mL量瓶中,用空白溶剂稀释至刻度,摇匀,即得。
N-芴甲氧羰基-O-叔丁基-L-苏氨酸约25 mg,精密称定,置50 mL量瓶中,加适量空白溶剂溶解,再加入“2.1.2”项各杂质储备液250 μL,用空白溶剂稀释至刻度,摇匀,即得。
N-芴甲氧羰基-O-叔丁基-L-苏氨酸约25 mg,精密称定,置50 mL量瓶中,加空白溶剂溶解并稀释至刻度,摇匀,即得。
精密量取供试品溶液0.5 mL,置100 mL量瓶中,用空白溶剂稀释至刻度,摇匀,即得。
取空白溶剂、各杂质定位溶液、供试品溶液、供试品加标溶液,按“2.1”项下色谱条件进行测定,并记录色谱图。结果如图2所示,空白溶剂对N-芴甲氧羰基-O-叔丁基-L-苏氨酸有关物质的测定无干扰,供试品溶液及供试品加标溶液中N-芴甲氧羰基-O-叔丁基-L-苏氨酸主峰与相邻杂质分离度、各个杂质间分离度均大于1.5,专属性良好。
精密称取N-芴甲氧羰基-O-叔丁基-L-苏氨酸约10 mg,共6份,各置20 mL量瓶中,分别进行酸、碱、氧化、光照及高温破坏试验。①未破坏溶液:加空白溶剂溶解并稀释至刻度,摇匀;②高温破坏溶液:于105 ℃条件下放置48 h,取出放冷,加空白溶剂溶解并稀释至刻度,摇匀;③光照破坏溶液:置复合光照箱内(25 ℃,5 000 lx),放置12 d,取出,加空白溶剂溶解并稀释至刻度,摇匀;④酸破坏溶液:加入1 mol·L-1盐酸溶液1 mL,摇匀,室温放置20 h后加入1 mol·L-1氢氧化钠溶液1 mL中和,加空白溶剂稀释至刻度,摇匀;⑤碱破坏溶液:加入1 mol·L-1氢氧化钠溶液1 mL,摇匀,室温放置2 min后加入1 mol·L-1盐酸溶液1 mL中和,加空白溶剂稀释至刻度,摇匀;⑥氧化破坏溶液:加入5%过氧化氢溶液2 mL,摇匀,室温放置1 h,加空白溶剂稀释至刻度,摇匀。将上述溶液分别按“2.1”项下色谱条件进行测定,并记录色谱图,空白溶剂、供试品加标溶液、供试品溶液、强降解试验色谱图见图1图2
结果显示,N-芴甲氧羰基-O-叔丁基-L-苏氨酸对酸、高温、光照条件较稳定,杂质个数、杂质总量无明显变化;对碱、氧化条件下有不同程度的降解,各有关物质及主峰均可完全分离,经峰纯度考察,主峰均为纯峰,以未破坏溶液的主峰含量为100%,计算上述各破坏条件下主药的物料平衡情况,结果显示物料守恒。说明本方法专属性好,能有效检出N-芴甲氧羰基-O-叔丁基-L-苏氨酸中的降解产物。
分别称取杂质1~杂质6对照品适量,加入溶剂溶解后逐步稀释,按“2.1”项下色谱条件进行测定,以信噪比S/N≥3计算检测限(LOD),以信噪比S/N≥10计算定量限(LOQ)。结果表明杂质1~杂质6的LOD均约为0.03 μg·mL-1。LOQ均约为0.06 μg·mL-1。取LOQ溶液,连续进样6针,计算杂质1~杂质6峰面积的RSD(n=6)均小于5.0%,表明本方法灵敏度较高。
N-芴甲氧羰基-O-叔丁基-L-苏氨酸适量,精密称定,加溶剂溶解并稀释制成每1 mL中约含0.5 mg的溶液,得N-芴甲氧羰基-O-叔丁基-L-苏氨酸储备溶液。
按“2.2”项下配制供试品溶液及各杂质对照品储备液,配制成6个线性溶液。取“2.3.2”项下定量限溶液及6个线性溶液,按“2.1”项下色谱条件进行测定,并记录色谱图。以浓度C为横坐标、峰面积A为纵坐标,进行线性回归,得回归方程。结果见表2N-芴甲氧羰基-O-叔丁基-L-苏氨酸、杂质1~杂质6的相关系数r均大于0.990 0,各有关物质在相应的浓度范围内线性关系良好。
按“2.2”项下配制各杂质对照品储备溶液及对照品溶液,取N-芴甲氧羰基-O-叔丁基-L-苏氨酸约25 mg精密称定,置50 mL量瓶中,先加适量空白溶剂溶解,再分别加入各杂质储备溶液250 μL,用溶剂稀释至刻度,摇匀,作为重复性溶液,平行配制6份,分别进样。结果显示,6份重复性溶液中杂质1~杂质6含量的平均值(n=6)分别为0.59%、0.50%、0.50%、0.56%、0.49%、0.49%,RSD分别为1.6%、1.0%、0.82%、1.1%、1.1%、1.3%,说明重复性良好。
更换分析人员及检测仪器,于不同日期同法试验6次,与重复性试验6个数据结果一并计算RSD。结果显示,12份加标溶液中杂质1~6含量的平均值(n=12)分别为0.59%、0.50%、0.49%、0.56%、0.48%、0.49%,RSD(n=12)分别为1.1%、1.6%、0.76%、0.68%、0.67%、0.79%,说明精密度良好。
按“2.2”项方法配制各杂质对照品储备液、混合对照品溶液、供试品溶液。
精密量取各杂质对照品储备液适量,加入供试品溶液中,分别配制限度浓度80%、100%、120%的回收率溶液,每个浓度平行配置3份。精密量取各回收率溶液0.5 mL,分别置100 mL量瓶中,用溶剂稀释至刻度,摇匀,作为回收率自身对照溶液。
取对照品溶液、供试品溶液、各浓度回收率溶液及回收率自身对照溶液,分别进样,计算各杂质的回收率及RSD;分别用外标法及加校正因子的自身对照法计算各准确度溶液中特定杂质含量,比较2种算法的差距。结果表明,杂质1~6的平均回收率(n=9)分别为98.6%、98.7%、98.8%、97.6%、98.3%、98.1%,RSD(n=9)分别为3.0%、0.68%、0.96%、2.1%、1.1%、0.88%,2种算法结果无明显差异。
考察供试品溶液、对照品溶液、供试品加标溶液于室温(25 ℃)条件下放置0、4、8、12、20 h的稳定性。结果显示,供试品溶液、对照品溶液、供试品加标溶液于室温条件下放置20 h内基本稳定。
在“2.1”项下色谱条件的基础上,分别改变柱温(±2 ℃)、流速(±0.1 mL·min-1),更换不同批号的色谱柱,其他色谱条件不变,考察方法的耐用性。分别按外标法以及加校正因子的自身对照法计算供试品加标溶液中各特定杂质含量。结果表明,当各条件微小改变时,供试品加标溶液中各杂质的含量基本一致,RSD均小于2.0%,表明本法耐用性良好。外标法及加校正因子的自身对照法分别计算供试品加标溶液中特定杂质含量,结果基本一致,表明加校正因子的自身对照法可准确用于本品有关物质计算。
取3批N-芴甲氧羰基-O-叔丁基-L-苏氨酸原料药,依法测定,结果如表3所示,检测结果符合拟定的有关物质限度标准。
采用二极管阵列检测器(DAD)对N-芴甲氧羰基-O-叔丁基-L-苏氨酸、杂质1~6进行全波长扫描,结果显示,N-芴甲氧羰基-O-叔丁基-L-苏氨酸、杂质1~6均在265 nm附近有最大吸收,因此选择265 nm作为有关物质的检测波长。流动相采用三氟乙酸水溶液和三氟乙酸乙腈溶液体系,通过调整梯度洗脱程序,实现了N-芴甲氧羰基-O-叔丁基-L-苏氨酸与相邻杂质、各杂质之间的基线分离。在方法学验证中对外标法和加校正因子的自身对照法进行了对比,从结果可以看出,外标法和加校正因子的自身对照法均可以准确地测定N-芴甲氧羰基-O-叔丁基-L-苏氨酸中的6个杂质的含量,结果无明显差异。因外标法需用到杂质对照品,杂质对照品难获得,而加校正因子的自身对照法不需杂质对照品,操作简单,因此,可以用加校正因子的自身对照法对N-芴甲氧羰基-O-叔丁基-L-苏氨酸中的6个杂质进行检测。
本研究建立了加校正因子的自身对照法同时检测N-芴甲氧羰基-O-叔丁基-L-苏氨酸中6个特定杂质的方法,灵敏度高,专属性强,准确度高,适用于N-芴甲氧羰基-O-叔丁基-L-苏氨酸中有关物质的检测,为多肽药物起始物料保护氨基酸质量研究提供了依据。
