Objective: To develop an inductively coupled plasma mass spectrometry(ICP-MS) method for the determination of Cd, Pb, As, Hg, Co, V, Ni and Se in the crude drug of lanthanum carbonate. Methods: Samples were diluted with 3% nitric acid and directly injected. The matrix effect was eliminated by standard addition method and matrix matching method. Lanthanum oxide was added into the standard solution as the matrix.Compare the linearity, repeatability, accuracy, quantification limit, detection limit and sample determination results of the two methods by methodological validation and sample determination. Results: The linear ranges of ICP-MS standard addition method and matrix matching method were 0-30 ng · mL-1, the correlation coefficients for all elemental impurities were good(r>0.998). Limits of detection were between 0.003 9-0.22 ng · mL-1 and 0.009 1-0.72 ng · mL-1. The RSD of repeatability(n=6) were between 1.5%-4.9% and 5.1%-6.9%. The recoveries were between 87.0%-98.3% and 83.4%-114.4%. The contents of 8 elemental impurities in the crude drug of lanthanum carbonate were basically same. Conclusion: The established two methods can effectively eliminate the matrix effect. They were both simple, rapid, sensitive, accurate and applicable for the elemental impurity control of lanthanum carbonate.

目的:建立电感耦合等离子体质谱(ICP-MS)法测定碳酸镧原料药中Cd、Pb、As、Hg、Co、V、Ni、Se共8种元素杂质的含量。方法:样品经3%硝酸溶解后直接进样,采用标准加入法和氧化镧基体匹配法2种方法进行测定,结合在线内标校正消除基质效应,同时对比分析2种方法的测定结果。结果:2种方法在质量浓度0~30 ng · mL-1范围内,8种元素线性均良好(r均>0.998),检测限分别为0.003 9~0.22 ng · mL-1和0.009 1~0.72 ng · mL-1,重复性RSD分别为1.5%~4.9%和5.1%~6.9%,加标回收率分别为87.0%~98.3%和83.4%~114.4%,3批碳酸镧原料药中8种元素杂质含量测定结果无显著差异。结论:ICP-MS标准加入法和基体匹配法简单快速,灵敏度高,准确性好,可有效消除基质效应,适用于碳酸镧原料药中元素杂质的质量控制。

目的:建立电感耦合等离子体质谱(ICP-MS)法测定碳酸镧原料药中Cd、Pb、As、Hg、Co、V、Ni、Se共8种元素杂质的含量。方法:样品经3%硝酸溶解后直接进样,采用标准加入法和氧化镧基体匹配法2种方法进行测定,结合在线内标校正消除基质效应,同时对比分析2种方法的测定结果。结果:2种方法在质量浓度0~30 ng · mL-1范围内,8种元素线性均良好(r均>0.998),检测限分别为0.003 9~0.22 ng · mL-1和0.009 1~0.72 ng · mL-1,重复性RSD分别为1.5%~4.9%和5.1%~6.9%,加标回收率分别为87.0%~98.3%和83.4%~114.4%,3批碳酸镧原料药中8种元素杂质含量测定结果无显著差异。结论:ICP-MS标准加入法和基体匹配法简单快速,灵敏度高,准确性好,可有效消除基质效应,适用于碳酸镧原料药中元素杂质的质量控制。

Objective: To develop an inductively coupled plasma mass spectrometry(ICP-MS) method for the determination of Cd, Pb, As, Hg, Co, V, Ni and Se in the crude drug of lanthanum carbonate. Methods: Samples were diluted with 3% nitric acid and directly injected. The matrix effect was eliminated by standard addition method and matrix matching method. Lanthanum oxide was added into the standard solution as the matrix.Compare the linearity, repeatability, accuracy, quantification limit, detection limit and sample determination results of the two methods by methodological validation and sample determination. Results: The linear ranges of ICP-MS standard addition method and matrix matching method were 0-30 ng · mL-1, the correlation coefficients for all elemental impurities were good(r>0.998). Limits of detection were between 0.003 9-0.22 ng · mL-1 and 0.009 1-0.72 ng · mL-1. The RSD of repeatability(n=6) were between 1.5%-4.9% and 5.1%-6.9%. The recoveries were between 87.0%-98.3% and 83.4%-114.4%. The contents of 8 elemental impurities in the crude drug of lanthanum carbonate were basically same. Conclusion: The established two methods can effectively eliminate the matrix effect. They were both simple, rapid, sensitive, accurate and applicable for the elemental impurity control of lanthanum carbonate.

碳酸镧是新型非含钙磷结合剂,通过与胃肠道中的磷结合成不溶性复合物发挥作用,可以避免高钙血症等不良反应发生,临床上用于治疗慢性肾衰竭等患者的高磷酸盐血症[1]。碳酸镧原料药主要是将氯化镧或氧化镧与碳酸盐或碳酸氢盐进行化学反应后,经纯化制得[2-3],制备工艺中所接触的试剂、试药、金属管道等不可避免地会引入各种元素杂质,因此,分析与研究碳酸镧原料药中元素杂质的残留具有重要意义。
电感耦合等离子体质谱(ICP-MS)法能同时测量多种元素,具有线性范围宽,分析速度快,灵敏度高等特点,适用于复杂基体中痕量元素的分析[4]。碳酸镧中镧元素的第二电离能低于氩的第一电离能,在氩等离子体高温光源中极易电离并产生双电荷[5],基体电离会消耗大量能量,导致待测元素电离效率大大降低,基体效应明显,内标回收率显著降低,不利于元素杂质的测定。任锟等[6]通过高基体(HMI)进样方式对碳酸镧原料中元素杂质进行含量分析,HMI模式[7]是通过引入稀释气(氩气),降低气溶胶密度,使等离子体能够耐受高基体样品,但该方法对仪器性能要求较高。标准加入法和基体匹配法也是有效降低基体干扰的方法,目前在生物、医药领域有着越来越多的应用[8-11]
本研究参考ICH Q3D[12]元素杂质指南,分别采用ICP-MS标准加入法和基体匹配法,对碳酸镧中ICH Q3D规定的1类元素杂质(Cd、Pb、As、Hg)、2A类元素杂质(Co、V、Ni)和2B类元素杂质(Se)进行考察,方法学研究结果表明,2种方法简单快速,灵敏度高,准确性好,均可有效消除基质效应,可用于碳酸镧原料药中元素杂质的质量控制。
Agilent 7900电感耦合等离子质谱仪(安捷伦公司);Milli-Q超纯水系统(Millipore公司)。
硝酸(批号1121100,Trace Metal Grade),Fisher Chemical公司;试验用水均为超纯水。
ICH/USP Target Elements Standard A(批号00123-32767,含As 15 μg · mL-1,Cd 5 μg · mL-1,Hg 30 μg · mL-1,Pb 5 μg · mL-1)、ICH/USP Target Elements Standard B(批号0012663343,含Co 50 μg · mL-1,Ni 200 μg · mL-1,V 100 μg · mL-1,Se 150 μg · mL-1)、内标标准溶液(批号59-046CRY2,含Li、Sc、Ge、Rh、In、Tb、Lu、Bi各10 μg · mL-1),Agilent公司;氧化镧(批号K2006227,含量99.999%),阿拉丁公司。
3批碳酸镧原料药收集自浙江国邦药业有限公司,批号依次为220101、220102、220103。
以氩气为载气、氦气为碰撞气,等离子功率1 550 W,载气流量1.05 L · min-1,补偿气流量 0.1 L · min-1,进样深度8 mm,蠕动泵速0.10 r · s-1,预混室温度2 ℃,积分时间0.3 s,扫描/重复次数100,检测模式为He mode。基体匹配法目标元素与内标元素见表1。内标元素通过T型三通管在线引入等离子体。
取碳酸镧原料药1 g,置1 000 mL量瓶中,加入硝酸30 mL溶解,用超纯水稀释至刻度,摇匀,即得标准加入法用稀释剂。取氧化镧0.62 g,置1 000 mL量瓶中,同法制备基体匹配法用稀释剂。
分别取ICH/USP Target Elements Standard A、ICH/USP Target Elements Standard B各625 μL,置25 mL量瓶中,加入标准加入法用稀释剂稀释至刻度,摇匀,作为标准曲线储备液。分别取标准曲线储备液0、60、100、200、300、400 μL,置50 mL量瓶中,用标准加入法用稀释剂稀释至刻度,摇匀,作为0 J、0.3 J、0.5 J、1.0 J、1.5 J、2.0 J标准曲线溶液(J为各元素杂质的限度浓度)。同法制备基体匹配法用标准曲线溶液。
精密称取碳酸镧原料药50 mg,置50 mL量瓶中,用3%硝酸溶液稀释至刻度,摇匀,即得。
取内标标准溶液125 μL,用3%硝酸溶液稀释至50 mL,摇匀,即得。
方法学验证要求参考USP<233>[13],线性相关系数应≥0.99,回收率3个浓度水平限度(n=3)应为70%~150%,重复性RSD(n=6)不得过20%。
取“3.2”项下系列标准曲线溶液,进样分析。标准加入法以各元素响应值为纵坐标,元素质量浓度为横坐标,绘制标准曲线;基体匹配法以待测元素响应值与相应内标元素响应值的比值为纵坐标,元素质量浓度为横坐标,绘制标准曲线。结果见表23。2种测定方法质量浓度在0~40 ng · mL-1内,8种元素杂质线性均良好(r均>0.998),可满足测定要求。
分别取2种测定方法下的空白溶液(稀释剂)连续测定11次,以3倍空白响应值标准偏差所对应的浓度计算各测定方法下元素的仪器检测限(ng · mL-1),10倍空白响应值标准偏差所对应的浓度计算各测定方法下元素的仪器定量限(ng · mL-1),结果见表23
ICH Q3D[12]将药品既定每日允许暴露量(permitted daily exposure,PDE)的30%定义为控制阈值,作为元素杂质水平显著性的衡量指标。根据ICH Q3D附录的方法1,以药品的日摄入量为10 g 计算药品中每种组分的目标元素通用允许浓度,根据各元素杂质的PDE值,计算各元素杂质的限度,结果见表23。各元素检测限和定量限均远低于相应限度浓度的30%(0.3 J),2种方法灵敏度均良好。
精密称取碳酸镧原料药样品(批号220101)50 mg,共6份,分别置50 mL量瓶中,分别加入标准曲线储备液200 μL,用3%硝酸溶液稀释至刻度,作为重复性试验的供试溶液。按“2”项下仪器参数进行测定,计算回收率及RSD,结果见表4。2种方法的RSD(n=6)为1.5%~6.9%,表明精密度均良好;各元素12份1.0 J加标样品回收率的RSD(n=12)为4.7%~8.4%,表明2种测定方法结果基本一致。
精密称取碳酸镧原料药样品(批号220101)50 mg,共9份,分别置50 mL量瓶中,分别加入标准曲线储备液100、200、300 μL,用3%硝酸溶液稀释至刻度,作为低、中、高水平回收率测定用供试溶液,每个浓度配制3份。按“2”项下仪器参数进行测定,计算2种方法的加标回收率,结果见表5。0.5 J~1.5 J浓度水平下,标准加入法测得各元素的回收率为87.0%~99.7%,RSD(n=9)为1.8%~7.7%;基体匹配法测得各元素的回收率为83.4%~114.4%,RSD(n=9)为3.6%~7.2%。结果表明,2种方法准确度均良好。
取3批碳酸镧原料药,按“3.3”项下方法配制供试品溶液,取空白溶液、标准曲线溶液和供试品溶液,通过蠕动泵的三通阀在线注入电感耦合等离子体质谱仪,记录响应值。标准加入法取上述3种溶液,以标准曲线法计算供试品溶液中各元素杂质含量。结果见表6。基体匹配法取上述3种溶液,与内标溶液同时在线进样测定,以内标校正的标准曲线法计算供试品溶液中各元素杂质含量。结果见表6。基体匹配法测得碳酸镧原料中8种元素杂质的含量均小于检测限;标准加入法测定碳酸镧原料中8种元素杂质的含量,除Pb和Se有检出外,其他元素杂质含量均小于检测限。2种方法测得的各样品中元素杂质含量均低于ICH规定限度。
ICH-Q3D[12]规定,1类元素杂质(Cd、Pb、As、Hg)和2A类元素杂质(Co、V、Ni)是口服给药途径药品必检项,因此,本研究对1类、2A类元素作分析测定。此外,采用安捷伦Mass Hunter软件自带的半定量监测模式,对ICH Q3D规定的24种元素进行扫描,发现Se有微量检出,同时Se也是La的伴生元素,故将Se列入检测元素,以评估本品质量。
碳酸镧中高比例的镧元素,会产生严重的基体效应,当采用ICP-MS标准曲线法进行试验,用3%硝酸作为稀释剂配制标准曲线溶液,发现测定样品时,内标回收率显著降低(低于70%),样品中待测元素测定结果均偏高,导致结果不准。标准加入法通过在样品中加入系列已知浓度待测元素的标准曲线溶液,绘制标准曲线进行测量,并通过该曲线计算样品中待测元素浓度,可以校正基体干扰,降低基体效应带来的测量误差。基体匹配法是基于标准加入法原理,使标准曲线溶液在与供试品溶液相匹配的基体中进行分析,本研究中通过在溶剂中加入氧化镧,配制成镧浓度与样品基本一致的空白溶液和系列标准曲线溶液,有效消除了镧对各元素响应的干扰。方法学考察结果表明,以上2种测定方法灵敏度、准确度均能满足分析要求。
