Objective: To compare the chemical composition changes in Kehuang before and after fermentation using multi-component content determination combined with chemometric analysis. Methods: HPLC was employed using a ZORBAX Eclipse Plus C18 column (4.6 mm×250 mm, 5 μm). The mobile phase consisted of acetonitrile and 0.1% phosphoric acid aqueous solution, applied with gradient elution. The flow rate was set at 1.0 mL · min-1,the column temperature was maintained at 30 ℃, the detection wavelength was 203 nm, and injection volume was 10 μL. This method was applied to analyze the chemical compositions of Kehuang before and after fermentation and to establish their corresponding chemical fingerprint profiles. Quantitative determination was conducted for 11 chemical constituents, including chlorogenic acid, cimifugin, notoginsenoside R1, baicalin, berberine, quercetin,baicalein, wogonin, emodin, chrysophanol, and physcion. Furthermore, chemometric methods such as hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least squares discriminant analysis (OPLS-DA) were employed to distinguish and compare the samples before and after fermentation, with the aim of identifying key differential compounds. Results: Chemical fingerprint profiles were established for Kehuang before and after fermentation, with 22 common peaks identified. The fingerprints had good specificity and can be used for quality evaluation of Kehuang. The results of multi-component content determination and chemometrics analysis revealed significant differences in the content of notoginsenoside R1 (1.023-2.927 mg · g-1),scutellarein (0.125-0.568 mg · g-1), and berberine hydrochloride (2.151-3.068 mg · g-1) among the crude Kehuang powder, fermented Kehuang powder, and the content of Kehuang capsules. Notoginsenoside R1, scutellarein and berberine were identified as differential markers between pre- and post-fermentation of Kehuang, which could serve as quality indicators for distinguishing and identifying Kehuang before and after fermentation. Conclusion: The chemical composition of Kehuang changes significantly after fermentation, and the differential compounds have been clearly identified, which provides an analytical basis for quality control in the production of Kehuang capsules.

目的:采用多成分含量测定结合化学计量学方法比较克癀发酵前后的化学成分变化。方法:采用ZORBAX Eclipse Plus C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,流速1.0 mL · min-1,柱温30 ℃,检测波长203 nm,进样量10 μL。对克癀发酵前后所含的绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚11个化学成分进行含量测定;建立克癀发酵前后的化学指纹图谱;应用聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘判别分析(OPLS-DA)等化学计量学方法,对克癀发酵前后成分进行区分与比较,筛选主要差异性成分。结果:建立了克癀发酵前后的化学指纹图谱,共标定了22个共有峰,所建立的化学指纹图谱专属性良好,可用于克癀的质量评价;多成分含量测定和化学计量学结果表明,克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末中三七皂苷R1(1.023~2.927 mg · g-1)、黄芩素(0.125~0.568 mg · g-1)和盐酸小檗碱(2.151~3.068 mg · g-1)的含量有明显差异;确认三七皂苷R1、黄芩素和小檗碱为克癀发酵前后的差异性成分,可以作为区分和鉴别克癀发酵前后的质量标志物。结论:经过发酵的克癀化学成分发生了明显变化,明确了其差异性成分,为克癀胶囊生产质量控制提供了分析依据。

Objective: To compare the chemical composition changes in Kehuang before and after fermentation using multi-component content determination combined with chemometric analysis. Methods: HPLC was employed using a ZORBAX Eclipse Plus C18 column (4.6 mm×250 mm, 5 μm). The mobile phase consisted of acetonitrile and 0.1% phosphoric acid aqueous solution, applied with gradient elution. The flow rate was set at 1.0 mL · min-1,the column temperature was maintained at 30 ℃, the detection wavelength was 203 nm, and injection volume was 10 μL. This method was applied to analyze the chemical compositions of Kehuang before and after fermentation and to establish their corresponding chemical fingerprint profiles. Quantitative determination was conducted for 11 chemical constituents, including chlorogenic acid, cimifugin, notoginsenoside R1, baicalin, berberine, quercetin,baicalein, wogonin, emodin, chrysophanol, and physcion. Furthermore, chemometric methods such as hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least squares discriminant analysis (OPLS-DA) were employed to distinguish and compare the samples before and after fermentation, with the aim of identifying key differential compounds. Results: Chemical fingerprint profiles were established for Kehuang before and after fermentation, with 22 common peaks identified. The fingerprints had good specificity and can be used for quality evaluation of Kehuang. The results of multi-component content determination and chemometrics analysis revealed significant differences in the content of notoginsenoside R1 (1.023-2.927 mg · g-1),scutellarein (0.125-0.568 mg · g-1), and berberine hydrochloride (2.151-3.068 mg · g-1) among the crude Kehuang powder, fermented Kehuang powder, and the content of Kehuang capsules. Notoginsenoside R1, scutellarein and berberine were identified as differential markers between pre- and post-fermentation of Kehuang, which could serve as quality indicators for distinguishing and identifying Kehuang before and after fermentation. Conclusion: The chemical composition of Kehuang changes significantly after fermentation, and the differential compounds have been clearly identified, which provides an analytical basis for quality control in the production of Kehuang capsules.

中药炮制是我国独特的制药技术,发酵作为中药的一种炮制方法,在增效减毒及改变药性方面具有特殊意义。通过发酵,可产生新的化学成分或药理作用,如六神曲、半夏曲、淡豆豉、红曲等发酵中药制品,在临床应用广泛[1-3]。克癀胶囊由三七、黄连、黄芩、白花蛇舌草和甘草等16味中药加入红曲发酵制成,可用于湿热邪气积聚、淤血阻塞经络等病症[4]。现代临床研究表明,克癀胶囊在抗肝纤维化[4]、治疗非酒精性脂肪肝[5]和肝癌[6]等肝脏疾病中疗效显著。
目前,克癀胶囊研究多集中于药效评价、作用机制等方面[7-10],对其多指标化学成分分析的研究较少,未收录于2020年版《中华人民共和国药典》。此外,克癀胶囊质量标准以薄层色谱定性鉴别为主,没有指标成分的含量测定,尤其是发酵前后化学成分变化规律及差异性成分尚不明确,无法全面表征其中间体及成品的整体质量。因此,本研究采用HPLC法,对15批次大生产规模的克癀粉末发酵前后及胶囊的化学成分进行指纹图谱分析,建立绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚11个化学成分的含量测定方法,再运用聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘判别分析(OPLS-DA)等化学计量学方法,寻找克癀粉末发酵前后及胶囊的差异性成分,为后期克癀胶囊生产质量控制和质量标准建立提供参考依据。
岛津LC-20A型高效液相色谱仪(岛津公司),低速台式离心机L420-A(湖南湘仪实验室仪器开发有限公司),ME204E电子万分之一天平(梅特勒-托利多仪器有限公司),KQ-500DE型超声波清洗机(昆山市超声仪器有限公司)。
克癀原药材粉末、克癀发酵粉末和克癀胶囊均由科兴生物制药股份有限公司提供,样品信息见表1。对照品绿原酸(批号B20782)、大黄素甲醚(批号B20242)、三七皂苷R1(批号B21099)、盐酸小檗碱(批号B21449)、大黄酚(批号B20238)、大黄素(批号B20240)、升麻素苷(批号B21157)、黄芩苷(批号B20570)、槲皮素(批号B20527)购自上海源叶生物科技有限公司,质量分数均≥98%;汉黄芩素(批号15363)、黄芩素(批号15727)购自上海诗丹德标准技术服务有限公司,质量分数均≥98%;甲醇、乙腈为色谱纯,均购于天津市彪仕奇科技发展有限公司;水为超纯水(Milli-Q默克),其他试剂均为分析纯。
取克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末(过65目筛)各约0.3 g,精密称定,分别置于50 mL具塞锥形瓶中,精密加入甲醇25 mL,再次精密称定,超声处理(500 W、40 kHz)30 min,静置,放冷,称量,用甲醇补足减失的量,于4 000 r · min-1离心15 min,取上清液,过0.22 μm微孔滤膜,取续滤液,即得各样品的供试品溶液。
精密称取绿原酸、升麻素苷、三七皂苷R1、黄芩苷、盐酸小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚的对照品适量,置于5 mL棕色量瓶中,加甲醇溶解并定容至刻度,配制成质量浓度分别为360、610、1 590、680、410、420、590、470、330、370和1 020 μg · mL-1的混合对照品溶液,并于4 ℃条件下保存,备用。
采用ZORBAX Eclipse Plus C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~15 min,15%A→20%A;15~50 min,20%A→38%A;50~60 min,38%A→42%A;60~70 min,42%A→60%A;70~80 min,60%A→90%A;80~95 min,90%A),流速1.0 mL · min-1,柱温30 ℃,进样量10 μL,检测波长203 nm。
取克癀胶囊内容物粉末(JN1)适量,按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件连续进样6次,以黄芩苷(6号峰)为参照峰,指纹图谱中22个共有峰的结果显示,各共有峰相对保留时间的RSD<2.0%,相对峰面积的RSD<2.8%,表明所用仪器精密度良好。
取克癀胶囊内容物粉末(JN1),平行6份,按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件检测,以黄芩苷(6号峰)为参照峰,指纹图谱中22个共有峰的结果显示,各共有峰相对保留时间的RSD<1.8%,相对峰面积的RSD<2.3%,表明方法重复性良好。
取同一份克癀胶囊内容物粉末(JN1),按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件,分别于0、2、4、8、12、18和24 h进样检测,以黄芩苷(6号峰)为参照峰,指纹图谱中22个共有峰的结果显示,各共有峰相对保留时间的RSD<2.5%,相对峰面积的RSD<2.9%,表明供试品溶液于室温放置24 h稳定性良好。
按“2.1.1”项下方法,分别制备克癀原药材粉末(YYC1~YYC5)、克癀发酵粉末(FJ1~FJ5)和克癀胶囊内容物粉末(JN1~JN5)的供试品溶液,按“2.2”项下色谱条件分别进样测定,得到HPLC色谱图。将上述色谱图导入《中药色谱指纹图谱相似度评价系统(2012版)》,以YYC1号样品的色谱图为参照图谱,采用平均数法,时间窗宽度设置为0.10 min,经多点校正后,再Mark峰匹配,生成指纹图谱叠加图谱(图1)。最终标定出22个共有峰,其中6号峰(黄芩苷)稳定性好,峰面积较大,故选该峰作为参照峰。将色谱图与混合对照品色谱图进行比对,指认出11个共有峰,分别为绿原酸(1号峰)、升麻素苷(3号峰)、三七皂苷R1(5号峰)、黄芩苷(6号峰)、小檗碱(8号峰)、槲皮素(10号峰)、黄芩素(12号峰)、汉黄芩素(14号峰)、大黄素(17号峰)、大黄酚(18号峰)和大黄素甲醚(19号峰)。
15批样品的色谱图与对照图谱(R)的相似度评价结果见表2,表明各批克癀药材粉末、克癀发酵粉末和克癀胶囊内容物粉末的相似度有明显的差异性,所建立的指纹图谱方法可用于克癀发酵前后的质量控制。
精密量取“2.1.2”项下的混合对照品溶液,按“2.2”项下色谱条件进样,以对照品的质量浓度X为横坐标,峰面积Y为纵坐标进行线性回归,结果见表3。绿原酸、升麻素苷、三七皂苷R1、黄芩苷、盐酸小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚在各自的线性范围内线性关系良好。
取克癀胶囊内容物粉末(JN1),按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件,连续进样6次,记录峰面积,计算其RSD,结果供试品溶液中绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚、大黄素甲醚峰面积的RSD分别为1.7%、0.47%、1.8%、1.3%、1.7%、1.8%、1.8%、1.1%、1.3%、0.72%和1.1%,表明仪器精密度良好。
取克癀胶囊内容物粉末(JN1),按“2.1.1”项下方法制备供试品溶液,室温放置,按“2.2”项下色谱条件,分别于0、4、8、12、16、24 h进行测定,记录峰面积,计算其RSD,结果供试品溶液中绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚、大黄素甲醚峰面积的RSD分别为1.8%、1.5%、2.5%、1.9%、0.69%、1.1%、2.3%、1.6%、2.0%、1.9%和2.1%,表明供试品溶液在24 h内稳定性良好。
取同批克癀胶囊内容物粉末(JN1),按“2.1.1”项下方法,平行制备6份供试品溶液,按“2.2”项下色谱条件分别进样,记录峰面积,计算绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱(以盐酸小檗碱计)、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚、大黄素甲醚的平均含量分别为0.304 2、0.223 2、1.064 0、3.014 0、2.189 1、0.222 7、0.317 1、0.146 1、0.021 8、0.045 9、0.045 8 mg · g-1,RSD分别为1.9%、2.6%、1.8%、1.7%、0.75%、2.0%、2.1%、0.87%、2.5%、0.66%和1.6%,表明该方法重复性良好。
取已知含量的克癀胶囊内容物粉末(JN1),平行6份,每份约0.15 g,精密称定,分别加入绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚、大黄素甲醚对照品适量,按“2.1.1”项下方法制备供试溶液,进样测定,计算样品加样回收率。结果绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱(以盐酸小檗碱计)、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚、大黄素甲醚的平均加样回收率(n=6)分别为93.0%、99.5%、97.4%、94.2%、97.0%、98.0%、93.3%、99.4%、98.8%、99.2%和98.1%,RSD分别为0.96%、1.9%、2.3%、2.4%、2.9%、2.7%、2.4%、2.9%、2.2%、2.7%和2.5%,表明该方法加样回收率良好。
按“2.1.1”项下方法制备5批克癀原药材粉末(YYC1~YYC5)、5批克癀发酵粉末(FJ1~FJ5)和5批克癀胶囊内容物粉末(JN1~JN5)样品的供试品溶液,按“2.2”项下色谱条件进行分析,分别测定绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱(以盐酸小檗碱计)、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚的含量。克癀原药材粉末、克癀发酵粉末、克癀胶囊内容物粉末和混合对照品的HPLC如图2所示,各样品的成分含量测定结果见表4
由测定结果可知,克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末中均含有所测定的11个化学成分。其中,黄芩苷的质量分数在克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末中均最高,分别为3.536~4.019、3.611~3.773和2.987~3.189 mg · g-1;质量分数最低的是大黄素,分别为0.020~0.030、0.034~0.045和0.021~0.023 mg · g-1。从各成分的RSD可知,绿原酸、三七皂苷R1、黄芩素、汉黄芩素和大黄素甲醚在克癀原药材粉末、发酵粉末和胶囊内容物粉末中存在较明显的差异。
为了进一步对克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末进行比较,明确三者之间的差异,寻找质量标志物,在多成分定量分析的基础上,运用HCA、PCA和OPLS-DA等化学计量学方法,以所测定的11个化学成分含量为变量,对克癀原药材粉末、发酵粉末和克癀胶囊内容物粉末进行分析与比较。
表2数据导入SPSS 26.0软件,以组间连接法结合平方欧氏距离对克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末进行HCA,结果见图3。由HCA结果可知,当平方欧氏距离为5时,克癀胶囊内容物粉末(JN1~JN5)聚为一类,克癀发酵粉末(FJ1~FJ5)聚为一类,克癀胶囊原药材粉末(YYC1~YYC5)聚为另一类;当平方欧氏距离>5时,所有样品主要聚为两大类,克癀胶囊内容物粉末和克癀发酵粉末为一类,克癀原药材粉末为另一类。提示克癀原药材粉末与克癀发酵粉末、克癀胶囊内容物粉末化学成分之间有差异。
采用SIMCA 14.1.0软件,以测得的11个成分含量测定结果为变量进行PCA,结果见图4。由PCA结果可知,前2个主成分的累积贡献率为99.1%。从所得的方差贡献率来看,建立的模型累计解释能力参数R2X为0.996,预测能力参数Q2为0.989,说明所建立的PCA模型的区分度和预测程度都较好。按照PCA结果,将15批粉末样品划分为3类:FJ1~FJ5为第Ⅰ类,JN1~JN5为第Ⅱ类,YYC1~YYC5为Ⅲ类。PCA的结果与HCA结果一致。
为进一步区分克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末之间的差异性标志化合物,以11个成分含量测定结果为变量,借助SIMCA 14.1.0软件,建立OPLS-DA模型(图5)。该模型中,自变量拟合指数R2X=0.999,因变量拟合指数R2Y=0.998,模型预测指数Q2=0.997,说明所建立的OPLS-DA模型可很好地用于克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末之间的分析。15批样品较好地分为3类,且与HCA、PCA的结果一致。进行置换检验(n=200),结果如图6所示,Q2回归线与纵轴的相交点小于0,说明模型未出现过拟合,模型预测能力好且验证有效,能够用于克癀原药材粉末、发酵粉末和胶囊内容物粉末的整体差异性分析。
通过预测变量重要性投影值(VIP)进一步筛选出克癀发酵前后的主要标志性成分,见图7。以VIP>1为标准,筛选出差异较大的成分,其贡献大小依次为三七皂苷R1、黄芩苷、黄芩素、小檗碱,这在区分克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末方面发挥了重要作用。如图8所示,三七皂苷R1、黄芩素和小檗碱的含量在克癀原药材粉末、克癀发酵粉末和克癀胶囊内容物粉末中存在显著差异(P<0.000 1)。其中,三七皂苷R1和小檗碱呈现出递减的趋势,黄芩素在克癀发酵粉末中的含量最高,而黄芩苷在克癀原药材粉末和克癀发酵粉末之间并无显著性差异,由此可认为,三七皂苷R1、黄芩素和小檗碱是克癀原药材粉末、发酵粉末和胶囊内容物粉末的差异性化合物。
为了明确克癀胶囊粉末发酵前后的成分变化规律,本文采用HPLC法对克癀胶囊粉末发酵前后及胶囊进行指纹图谱分析及多成分含量测定。克癀胶囊的成分多而复杂,在分析前参考相关文献[7-811],考察了甲醇-水、甲醇-0.01%磷酸水溶液、乙腈-水、乙腈-0.01%磷酸水溶液等体系的梯度洗脱分离,结果显示,以乙腈-0.01%磷酸水溶液作为流动相,能使克癀胶囊中的各成分得到更好的分离,且基线平稳。
本研究采用HPLC法建立了15批克癀发酵前后的指纹图谱,确定了22个共有峰,指认11个化学成分。各样品间指纹图谱相似度良好,均在0.994以上。通过指纹图谱分析可知,15批克癀原药材粉末、发酵粉末和胶囊内容物粉末的峰面积大小不一,表明三者的成分含量之间存在一定差异性。为了找出其中的差异性化合物,本研究进一步测定了克癀胶囊发酵前后绿原酸、升麻素苷、三七皂苷R1、黄芩苷、小檗碱、槲皮素、黄芩素、汉黄芩素、大黄素、大黄酚和大黄素甲醚11个共有成分的含量,结果以黄芩苷的含量最高。
为了进一步寻找克癀原药材粉末、发酵粉末和胶囊内容物粉末中的质量标志物,在含量测定的基础上,应用HCA、PCA及OPLS-DA等化学计量学方法对克癀原药材粉末、发酵粉末和胶囊内容物粉末进行了区分与比较。结果显示,基于11个化学成分的定量结果,可以将克癀发酵前后进行较好地区分。通过11个化学成分的VIP值筛选和含量分析可知(图78),三七皂苷R1、黄芩素和小檗碱3个成分为克癀原药材粉末、发酵粉末和胶囊内容物粉末之间的差异性化合物,可作为克癀发酵前后的质量控制指标,也可作为区分克癀发酵前后的指标性化合物。
图8中4个成分含量分析显示,黄芩素的含量在克癀胶囊发酵粉末中显著(P<0.000 1)高于原药材粉末,黄芩苷与其来源于同种药材,但在发酵粉末与原药材粉末之间并无显著性差异。相关研究显示,黄芩中所含的黄芩素天然含量极低,但黄芩素表现出比原苷黄芩苷更优越的药理活性,黄芩苷转化为黄芩素极大地提高了其脂溶性和疏水性,使得黄芩苷更容易穿过胃黏膜,且研究表明,黄芩苷在体内发挥药效需要转化为黄芩素后才能实现[12-14]。因此,本研究最终未将含量最高的黄芩苷作为克癀发酵前后的质量控制指标之一,而是选择黄芩素,这对克癀胶囊发酵前后的综合质量评价更具有参考意义。
本研究采用HPLC法对克癀发酵前后化学成分进行了分析,建立了克癀发酵前后化学指纹图谱和含量测定方法;采用化学计量学方法对克癀发酵前后进行了比较,确定了三七皂苷R1、黄芩素和小檗碱为质量标志物。所建立的分析方法准确,结果稳定,可用于克癀的质量控制和评价,并为其提供了方法保障和数据支撑。
