Objective: To establish a rapid analytical method for chemical components in complex traditional Chinese medicine (TCM) based on an integrated strategy combining ultra high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UHPLC-Q TOF MS/MS) with feature-based molecular networking(FBMN), so as to systematically characterize the chemical components in Sishen pills. Methods: MS data for Sishen pills were acquired using UHPLC-Q TOF MS/MS, with separation performed on on a Waters HSS T3 column(2.1 mm×100 mm, 1.8 µm). The mobile phase was consisted of a gradient elution of 0.1% formic acid in water(A) and acetonitrile (B) at a flow rate of 0.3 mL · min-1. The electrospray ionization (ESI) source was used for both positive and negative ion modes, with a scan range of m/z 80 to 1 500. The data were uploaded to the Global Natural Product Social Molecular Networking (GNPS) to create a FBMN for Sishen pills. Results: A total of 131 compounds were identified in Sishen pills, by analyzing retention time, accurate molecular mass, MS2 fragmentation characteristics, comparison with reference standards and self-established database, as well as utilizing FBMN for predicting structural similarity of compounds. These compounds included 27 alkaloids, 63 flavonoids, 11 phenylpropanoids, 14 phenols, 8 terpenoids, and 8 other compounds. Conclusion: In this study, the chemical components of Sishen pills are quickly and comprehensively characterized, which layes a foundation for the effective materials and quality control research of Sishen pills. The study also provides insights for the rapid analysis of chemical constituents in other TCM formulas.

目的:建立基于超高效液相色谱-四极杆飞行时间质谱(UHPLC-Q TOF MS/MS)结合特征分子网络(feature-based molecular networking,FBMN)整合策略的中药复杂体系化学成分快速分析方法,对四神丸中的化学成分进行系统表征。方法:采用UHPLC-Q TOF MS/MS对四神丸进行质谱数据采集,采用Waters HSS T3色谱柱(2.1 mm×100 mm,1.8 µm)进行分离,以0.1%甲酸水(A)-乙腈(B)为流动相进行梯度洗脱,流速0.3 mL · min-1,电喷雾离子源(ESI),正、负离子模式扫描,扫描范围m/z 80~1 500,并上传至Global Natural Product Social Molecular Networking(GNPS),以创建四神丸的FBMN。结果:通过分析保留时间、精确分子质量、二级质谱碎片特征,结合对照品、自建数据库及相关文献信息比对,以及FBMN关联性预测结构相似性化合物,在四神丸中共鉴定出131个化合物,包括27个生物碱类、63个黄酮类、11个苯丙素类、14个酚类,8个萜类化合物以及8个其他化合物。结论:本研究对四神丸的化学成分进行了快速全面的表征,总结了主要化学成分的裂解规律及特征离子,为四神丸的药效物质基础和质量控制研究奠定了基础,同时为其他中药复方化学成分的快速分析提供借鉴。

目的:建立基于超高效液相色谱-四极杆飞行时间质谱(UHPLC-Q TOF MS/MS)结合特征分子网络(feature-based molecular networking,FBMN)整合策略的中药复杂体系化学成分快速分析方法,对四神丸中的化学成分进行系统表征。方法:采用UHPLC-Q TOF MS/MS对四神丸进行质谱数据采集,采用Waters HSS T3色谱柱(2.1 mm×100 mm,1.8 µm)进行分离,以0.1%甲酸水(A)-乙腈(B)为流动相进行梯度洗脱,流速0.3 mL · min-1,电喷雾离子源(ESI),正、负离子模式扫描,扫描范围m/z 80~1 500,并上传至Global Natural Product Social Molecular Networking(GNPS),以创建四神丸的FBMN。结果:通过分析保留时间、精确分子质量、二级质谱碎片特征,结合对照品、自建数据库及相关文献信息比对,以及FBMN关联性预测结构相似性化合物,在四神丸中共鉴定出131个化合物,包括27个生物碱类、63个黄酮类、11个苯丙素类、14个酚类,8个萜类化合物以及8个其他化合物。结论:本研究对四神丸的化学成分进行了快速全面的表征,总结了主要化学成分的裂解规律及特征离子,为四神丸的药效物质基础和质量控制研究奠定了基础,同时为其他中药复方化学成分的快速分析提供借鉴。

Objective: To establish a rapid analytical method for chemical components in complex traditional Chinese medicine (TCM) based on an integrated strategy combining ultra high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UHPLC-Q TOF MS/MS) with feature-based molecular networking(FBMN), so as to systematically characterize the chemical components in Sishen pills. Methods: MS data for Sishen pills were acquired using UHPLC-Q TOF MS/MS, with separation performed on on a Waters HSS T3 column(2.1 mm×100 mm, 1.8 µm). The mobile phase was consisted of a gradient elution of 0.1% formic acid in water(A) and acetonitrile (B) at a flow rate of 0.3 mL · min-1. The electrospray ionization (ESI) source was used for both positive and negative ion modes, with a scan range of m/z 80 to 1 500. The data were uploaded to the Global Natural Product Social Molecular Networking (GNPS) to create a FBMN for Sishen pills. Results: A total of 131 compounds were identified in Sishen pills, by analyzing retention time, accurate molecular mass, MS2 fragmentation characteristics, comparison with reference standards and self-established database, as well as utilizing FBMN for predicting structural similarity of compounds. These compounds included 27 alkaloids, 63 flavonoids, 11 phenylpropanoids, 14 phenols, 8 terpenoids, and 8 other compounds. Conclusion: In this study, the chemical components of Sishen pills are quickly and comprehensively characterized, which layes a foundation for the effective materials and quality control research of Sishen pills. The study also provides insights for the rapid analysis of chemical constituents in other TCM formulas.

明代薛己《内科摘要》中将二神丸与五味子散两方合为四神丸,为治疗泄泻的名方[1],四神丸以肉豆蔻、五味子、吴茱萸和补骨脂为主药,再辅以大枣、生姜[2-3],具有温肾散寒、涩肠止泻的功效[4],临床上常用于治疗脾肾阳虚型泄泻、结肠炎以及肠易激综合征等[5-6]。四神丸中化学成分主要含有苯丙素类、黄酮类和生物碱类等[7-8]。目前对于四神丸复方的化学成分研究,仅仅采用LC-MS对其中几种活性成分定量分析[9],不足以阐明物质基础,评估其药效价值。因此创建快速可靠的分析方法全面表征其化学成分至关重要。
超高效液相色谱-四极杆飞行时间质谱(UHPLC-Q TOF MS/MS)具有高选择性、高灵敏度和高准确度等优势[10],广泛用于中药复方化学成分的鉴定[11-12]。Global Natural Product Social Molecular Networking(GNPS)集群数据库近年来被广泛使用,是基于质谱技术所建立的“可视化”分子网络[13]。特征分子网络(feature-based molecular networking,FBMN)在运用MS/MS数据构建分子网络的基础上,导入峰表,引入保留时间和实际m/z,解决了GNPS分子网络在鉴定同分异构体方面的局限性[14-15]。借助FBMN,可进一步表征出四神丸中潜在的未知化合物。
本实验采用UHPLC-Q TOF MS/MS技术,与FBMN相结合,对四神丸化学成分进行全面表征,为其药效物质基础和质量控制提供参考依据。
四神丸(批号20210302,山西华康药业股份有限公司);补骨脂素(批号16092810)、异补骨脂素(批号17062204)、新补骨脂异黄酮(批号16112510)、补骨脂二氢黄酮甲醚(批号18101705)、槲皮素(批号16111114)、山柰酚(批号17032911),纯度均大于99%,均购于成都曼斯特生物科技有限公司;乙腈和甲酸为色谱纯,赛默飞世尔科技有限公司。
Exion LC™ AD超高效液相色谱仪、X500R QTOF型质谱仪,AB Sciex公司;SB-5200DT超声波清洗机,宁波新芝生物科技股份有限公司;PX124ZH1/10万分之一电子天平,奥豪斯仪器有限公司;KDC-140HR高速冷冻离心机,安徽中科中佳科学仪器有限公司。
分别取补骨脂素、异补骨脂素、新补骨脂异黄酮、补骨脂二氢黄酮甲醚、槲皮素、山柰酚、齐墩果酸适量,用70%乙腈溶液配成质量浓度约为0.5 µg · mL-1的混合对照品溶液。
取四神丸适量,研细过100目筛,取1.0 g,精密称定,置具塞锥形瓶中,分别精密加入70%乙腈溶液50 mL,密塞,超声(360 W,40 kHz)提取30 min,摇匀,滤过,取续滤液,12 000 r · min-1高速离心10 min,过0.22 µm微孔滤膜,即得。
采用Waters HSS T3色谱柱(2.1 mm×100 mm,1.8 µm),以0.1%甲酸超纯水(A)-乙腈(B)为流动相,梯度洗脱(0~5 min,5%B→25%B;5~25 min,25%B→75%B;25~30 min,75%B→95%B;30~31 min,95%B→5%B;31~35 min,5%B),流速0.3 mL · min-1,柱温35 ℃,进样量1 µL。
采用电喷雾离子(ESI)源正负离子分别扫描,离子化电压为-4 500 V(负离子)、5 500 V(正离子),离子源温度为450 ℃,喷雾气(Gas 1)379 kPa,辅助加热气(Gas 2)379 kPa,气帘气(CUR)207 kPa,去簇电压(DP)60 V。采用数据依赖性扫描(IDA模式):1个MS循环包含1个TOF MS扫描(累积时间0.25 s)和至多10个子离子扫描(累积时间0.09 s)。信息关联采集(IDA)触发条件:强度阈值100 cps,开启动态背景扣除(DBS)。一级质谱(TOF MS)扫描范围为m/z 80~1 500,子离子扫描范围为m/z 50~1 250,二级碰撞能量(CE)为40 eV,碰撞能量滚动区间(CEs)15 eV。
通过查阅四神丸各单味药化学成分,建立化合物数据库,采用SCIEX OS软件进行峰提取,比对精确分子量、二级质谱碎片等信息。采用MS Dial软件将原始的LC-MS/MS数据进行提峰处理,利用代谢组学软件One-MAP中的转换功能将LC-MS/MS数据转换成mgf格式,将峰表和mgf格式文件上传到GNPS数据库(https://gnps.ucsd.edu),在Feature Networking模块建立FBMN,采用cytoscape 3.8.0软件对建立的FBMN进行图像处理。根据已鉴定化合物与其相邻节点的相似度,结合m/z和相对分子质量差异,推测未知化合物结构。
根据四神丸各单味药化学成分研究报道,构建“四神丸化学成分数据库”,包括化合物名称、分子式、化学结构、精确分子量和二级质谱碎片等化学信息。数据库包含151个已知成分,包括来源于吴茱萸的27个生物碱成分和1个萜类成分(柠檬苦素),补骨脂中的56个黄酮类成分和11个苯丙素类成分,肉豆蔻中的19个苯丙素类成分,五味子中的9个苯丙素类成分,大枣中的3个萜类成分和生姜中的25个酚类成分。
按照“2.3”项下检测条件进样,获得UHPLC-Q TOF MS正、负离子模式下四神丸样品的基峰离子流(BPI)色谱图(图1),通过与对照品比对、检索相关文献及自建数据库对其进行化学成分表征,结合FBMN,共鉴定出133个化学成分,主要有生物碱类、苯丙素类、黄酮类及其同分异构体等(表1),随后选择具有代表性的化学结构类型分析其裂解规律,总结特征离子,汇总结果见表2
生物碱类化合物主要来源于吴茱萸,分别以吲哚类生物碱吴茱萸酰胺(evodiamide)和喹啉类生物碱1-methyl-2-[(Z)-6-undecenyl]-4(1H)-quinolone为例。在正离子扫描模式下,化合物54的准分子离子峰为m/z308.175 5[M+H]+,分子式为C19H21N3O,二级质谱图显示碎片离子m/z为106.065 4、116.049 6、134.060 0,主要通过a键、b键和c键3种方式断裂(图2-A),参考相关文献[16]推测其为吴茱萸酰胺。此外,还通过数据库以及文献检索[16]鉴定了其他吲哚类生物碱(化合物59、62、64、122),总结其特征离子有m/z 106.06、116.04。
化合物98准分子离子峰为m/z312.231 8[M+H]+,分子式为C21H29NO,二级质谱图显示m/z159.068 3、173.083 5、186.091 4、200.105 2、228.137 9,裂解方式主要是化合物的支链裂解,以丢失最大烃基最为常见,根据二级质谱及参考相关文献[13],推测其为1-Methyl-2-[(Z)-6-undecenyl]-4-(1H)-quinolone(图2-B)。化合物122准分子离子峰为m/z 342.278 8[M+H]+,其分子式为C21H29NO,二级谱图显示m/z为159.059 3、173.083 3、186.091 0、200.106 7、326.409 9,二级谱图显示化合物124与化合物100具有相同的母核,裂解方式相似,根据二级质谱及参考相关文献[16],推测其为二氢吴茱萸卡品碱(dihydroevocarpine)。
喹啉类生物碱大多产生支链裂解的特征离子(m/z 159.07、173.03、186.09、200.10),结合参考文献[17],共鉴定6个同类型的化合物(化合物60、62、64、73、76、86)。此外,构建了生物碱在正离子模式下的FBMN(图2-C),鉴定了5个生物碱类化合物节点,生物碱类化合物主要以喹啉为母核,其中化合物110(m/z 314.247)与已鉴定化合物122(C23H35NO m/z 342.278)之间存在m/z 28.031的质量差,生物碱支链丢失C2H4,推测其分子式为C21H31NO,结合特征离子鉴定化合物110为1-Methyl-2-undecyl-4(1H)-quinolone。化合物61(m/z 258.186)与化合物110(m/z 314.247)之间存在m/z 56.061(C4H8)的质量差,推断其分子式为C17H23NO,结合特征离子鉴定化合物61为青花椒碱(schinifoline)。以此类推,可表征相似结构的系列化合物(61、103、114)。最终,通过文献报道[18-19]、数据库比对结合FBMN,共表征27个生物碱类化合物,包括5个以吲哚为母核的生物碱(图2-A)和22个以喹啉为母核的生物碱(图2-B)。
黄酮类化合物主要来源于中药补骨脂,在正离子扫描模式下,化合物88(21.58 min)与化合物108(24.29 min)分子离子峰均为m/z339.158 7[M+H]+,互为同分异构体[20-21],分子式均为C21H22O4,进一步分析其二级质谱,发现化合物88与化合物108均产生m/z283.096 9、271.096 2、269.080 5的碎片离子。但不同的是:化合物88主要发生逆狄尔斯-阿德尔(RDA)反应,产生m/z219.101 5碎片,随后丢失异戊烯基,产生m/z151.038 8,再丢失羰基(CO),产生m/z123.044 0碎片(图3-A);而化合物108主要通过a键、b键和c键3种方式裂解(图3-B),产生m/z147.043 9,177.044 2,219.101 3的二级裂解碎片。进一步与对照品保留时间进行比对,化合物88和108分别为补骨脂二氢黄酮甲醚(bavachinin)和4’-甲氧基补骨脂查尔酮(4’-O-menthylbroussochalcone),两者的裂解规律如图3所示。
化合物55在负离子扫描模式下,其准分子离子峰m/z321.113 1 [M-H]-,分子式为C20H18O4,二级质谱图显示m/z为135.009 0、145.029 5、237.055 9、265.050 4、278.058 3,通过参考相关文献[22-23],推测其为新补骨脂异黄酮(neobavaisoflavone),为异黄酮类化合物。
此外,通过FBMN鉴定9个黄酮类化合物节点。如图4所示,分子网络图Ⅰ、Ⅱ、Ⅲ均为黄酮类化合物,分子网络图Ⅰ主要结构类型为异黄酮类化合物,其中节点m/z 389.176[M-H]-与已鉴定的化合物55(C20H18O4,m/z 321.113 [M-H]-)之间存在m/z 68.063的质量差,其差值对应C5H8,故推测化合物96的分子式为C25H26O4,结合二级质谱信息鉴定为次苷酸查尔酮(corylifol A)。分子网络图Ⅱ主要结构类型为2个二氢黄酮类和1个查尔酮,分子网络图Ⅲ主要结构类型为黄酮苷类化合物。最终,通过文献报道[24-25]、与对照品和自建数据库比对以及FBMN推测,共表征出63个黄酮类,包括10个二氢黄酮类、20个异黄酮类、10个查尔酮类化合物、23个黄酮及黄酮醇类化合物。
在正离子扫描模式下,化合物36(tR=11.82 min)和化合物40(tR=12.24 min)m/z187.038 8 [M+H]+,分子式均为C11H6O3,二者互为同分异构体,进一步对比两者的二级裂解碎片离子,均发生丢失CO2后再丢失CO产生m/z 115.054 4[M+H-CO2-CO]+的碎片离子,丢失2个CO后产生m/z 131.049 1 [M+H-2CO]+的碎片离子。进一步与对照品保留时间比对,表征化合物36为补骨脂素(psoralen),化合物40为异补骨脂素(isopsoralen)。主要结构类型为呋喃香豆素[26],可产生m/z 为77.03、115.05、131.04的特征离子。
化合物111在正离子扫描模式下,其准分子离子峰m/z417.226 8 [M+H]+,分子式为C24H32O6,二级质谱碎片离子有m/z402.203 3、386.208 4、347.147 8、316.130 3、285.111 8,其中m/z285.111 8 [M+H-C5H10-2CH3O]+为母离子417丢失C5H10后再丢失2个甲氧基后产生的碎片离子,参考相关文献[27]推测其为可能为五味子甲素(deoxyschisandrin),其可能裂解方式如图5-A所示。五味子中的木脂素类成分,其母核为联苯环辛烯型木脂素,故多具有相似的裂解规律,表现为甲氧基和C5H10的丢失。
在正离子模式下的FBMN中,主要分析了2个主要的分子网络图,其中包括5个生物碱类成分(图2-C)、5个苯丙素类成分(图5-B)。其中苯丙素类化合物的分子网络图的相对分子质量在m/z 434.255~387.180,主要结构为联苯环辛烯型木脂素类化合物,根据已鉴定的五味子甲素(deoxyschisandrin)新鉴定4个同类化合物。由于香豆素类化合物与黄酮类化合物的结构相似,所以化合物46(C20H16O5m/z 337.106)与黄酮类化合物聚集在同一个分子笼中。最终,共表征11个苯丙素类化合物,其中有1个来自补骨脂,2个来自肉豆蔻[28-30],8个来自五味子[31]
酚类化合物主要来源于中药生姜[30-32],而萜类来源于吴茱萸[33],共表征出14个酚类化合物和8个萜类,通过比对自建数据库和查阅相关文献[34],生姜中的姜酚类化合物大多失去而生成子离子m/z135的化合物以及丢失H2O和CH3O。例如,化合物104在正离子扫描模式的准分子离子峰为m/z291.159 0[M+H]+,分子式为C17H22O4,其二级质谱图显示m/z 273.148 2[M+H-H2O]+m/z177.054 5[M+H-C4H8-CO-CH3]+m/z149.059 2[M+H-C4H8-2CO-CH3]+,通过参考文献及分析其裂解规律,推测其为可能为1-脱氢-6-姜酮(1-dehydro-6-gingerdione),其可能裂解方式如图6所示。
目前,已有研究对二神丸和四神丸单味药材[35]化学成分进行表征,然而对于四神丸复方制剂化学成分研究尚未见文献报道。本研究采用UHPLC-Q TOF-MS/MS技术,对四神丸化学成分进行全面、快速表征。分别采用50%甲醇、70%甲醇、70%乙腈溶液对四神丸化学成分进行提取,结果显示无论是正离子还是负离子模式下,70%乙腈溶液提取的峰数目较丰富,峰强度较高,因此,选择70%乙腈溶液进行样品预处理。
本研究采用UHPLC-Q TOF-MS技术与FBMN相结合对四神丸提取物中的化学成分进行了全面表征,共鉴定131个化学成分,其中根据对照品、数据库及文献比对鉴定93个,采用FBMN进一步表征了38个化合物。通过总结各类化合物及其亚类的特征离子及中性丢失碎片,明确其裂解规律。FBMN可以将具有相似二级质谱信息的化合物聚集成1个分子笼,每个节点代表1个化合物,在传统分子网络的基础上,减轻冗余节点,鉴别同分异构体。本研究借助FBMN,进一步表征了31个未在四神丸或其单味药中报道的化合物。四神丸中表征的化合物类型包括27个生物碱类、63个黄酮类、11个苯丙素类、14个酚类,8个萜类化合物以及8个其他化合物。其中生物碱来源于吴茱萸,黄酮来源于补骨脂和肉豆蔻,苯丙素类化合物来源于补骨脂、五味子、肉豆蔻,酚类化合物来源于生姜和补骨脂,糖类化合物来源于大枣[36]。根据以上四神丸化学成分的分析表明,生物碱、黄酮和苯丙素类为四神丸复方制剂中的主要化学成分类型。本研究可为四神丸的药效物质基础和质量控制研究奠定基础,也为中药复杂体系物质基础的阐明提供方法参考。
