Objective: To prepare monoclonal antibodies (MAbs) that can specifically recognize genistein(Gen), and to establish a specific and rapid quantitative method for Gen using indirect competitive enzyme-linked immunosorbent assay (ELISA). Methods: Gen was coupled with carrier protein by the Mannich method to synthesize artificial antigens. Femal Balb/c mice aged 6-8 weeks old were immunized by Gen-BSA. After the booster immunization, splenocytes of the mice were collected and fused with SP2/0 myeloma cells under the action of 50% polyethylene glycol (PEG). Subclones were screened by the limiting dilution method to obtain specific MAbs and an indirect competitive ELISA method was established. Results: Two hybridoma cell lines (C1 and C3)were isolated successfully with the titers of 1 : 32 000, 1 : 40 000, respectively. Under the optimized conditions,the indirect competitive ELISA based on C3 for Gen showed a half maximum inhibition concentration (IC50) of 16.15 ng · mL-1 and detection ranges of 3.78-55.85 ng · mL-1 with cross-reactivity for biochanin A and irisolidone of 4.96% and 2.40%, respectively, negligible cross-reactivity with other Gen analogs including apigenin and 4-oxo-4H-1-benzopyran-2-carboxylic acid. The recovery test showed that the recovery rate was between 85% and 110%, precision tests showed RSD of inter-assay and inter-assay was less than 10%. Conclusion: In this study,Gen-specific MAbs are prepared successfully. An sensitive and accurate indirect competitive ELISA based on MAb for Gen is developed. The developed method can meet the requirements for immunoassay.

目的:制备可特异性识别金雀异黄酮(Gen)的单克隆抗体,建立特异、快速定量Gen的间接竞争酶联免疫吸附分析(ELISA)方法。方法:通过曼尼希法将Gen和载体蛋白偶联,合成人工抗原。免疫6~8周龄的雌性Balb/c小鼠,冲击免疫后取小鼠脾细胞与SP2/0骨髓瘤细胞,在50% PEG的作用下进行细胞融合。采用有限稀释法筛选阳性亚克隆,获取特异性单克隆抗体,建立间接竞争ELISA法。结果:试验成功筛选出C1和C3 2株抗Gen单克隆抗体,效价分别为1 : 32 000、1 : 40 000。利用C3抗体优化间接竞争ELISA反应条件,检测抗体的灵敏度IC50为16.15 ng · mL-1,线性范围(IC20~IC80)为3.78~55.85 ng · mL-1。交叉反应结果显示,Gen单克隆抗体与鹰嘴豆芽素A、尼泊尔鸢尾异黄酮的交叉反应率分别为4.96%和2.40%,而与芹菜素、4-苯并吡喃酮2-羧酸的交叉反应率均<0.1%。回收率在85%~110%,批内和批间RSD均<10%。结论:本研究制备了Gen特异性的单克隆抗体,用其建立了Gen间接竞争ELISA检测方法,灵敏度高,符合免疫分析的基本要求。

目的:制备可特异性识别金雀异黄酮(Gen)的单克隆抗体,建立特异、快速定量Gen的间接竞争酶联免疫吸附分析(ELISA)方法。方法:通过曼尼希法将Gen和载体蛋白偶联,合成人工抗原。免疫6~8周龄的雌性Balb/c小鼠,冲击免疫后取小鼠脾细胞与SP2/0骨髓瘤细胞,在50% PEG的作用下进行细胞融合。采用有限稀释法筛选阳性亚克隆,获取特异性单克隆抗体,建立间接竞争ELISA法。结果:试验成功筛选出C1和C3 2株抗Gen单克隆抗体,效价分别为1 : 32 000、1 : 40 000。利用C3抗体优化间接竞争ELISA反应条件,检测抗体的灵敏度IC50为16.15 ng · mL-1,线性范围(IC20~IC80)为3.78~55.85 ng · mL-1。交叉反应结果显示,Gen单克隆抗体与鹰嘴豆芽素A、尼泊尔鸢尾异黄酮的交叉反应率分别为4.96%和2.40%,而与芹菜素、4-苯并吡喃酮2-羧酸的交叉反应率均<0.1%。回收率在85%~110%,批内和批间RSD均<10%。结论:本研究制备了Gen特异性的单克隆抗体,用其建立了Gen间接竞争ELISA检测方法,灵敏度高,符合免疫分析的基本要求。

Objective: To prepare monoclonal antibodies (MAbs) that can specifically recognize genistein(Gen), and to establish a specific and rapid quantitative method for Gen using indirect competitive enzyme-linked immunosorbent assay (ELISA). Methods: Gen was coupled with carrier protein by the Mannich method to synthesize artificial antigens. Femal Balb/c mice aged 6-8 weeks old were immunized by Gen-BSA. After the booster immunization, splenocytes of the mice were collected and fused with SP2/0 myeloma cells under the action of 50% polyethylene glycol (PEG). Subclones were screened by the limiting dilution method to obtain specific MAbs and an indirect competitive ELISA method was established. Results: Two hybridoma cell lines (C1 and C3)were isolated successfully with the titers of 1 : 32 000, 1 : 40 000, respectively. Under the optimized conditions,the indirect competitive ELISA based on C3 for Gen showed a half maximum inhibition concentration (IC50) of 16.15 ng · mL-1 and detection ranges of 3.78-55.85 ng · mL-1 with cross-reactivity for biochanin A and irisolidone of 4.96% and 2.40%, respectively, negligible cross-reactivity with other Gen analogs including apigenin and 4-oxo-4H-1-benzopyran-2-carboxylic acid. The recovery test showed that the recovery rate was between 85% and 110%, precision tests showed RSD of inter-assay and inter-assay was less than 10%. Conclusion: In this study,Gen-specific MAbs are prepared successfully. An sensitive and accurate indirect competitive ELISA based on MAb for Gen is developed. The developed method can meet the requirements for immunoassay.

金雀异黄酮(genistein,Gen)是从豆科植物中发现的一种异黄酮,具有和17β-雌二醇相似的分子结构,可与雌激素受体结合,发挥微弱的雌激素作用[1]。近年来,有研究人员利用Aβ25-35诱导乳鼠海马神经元构建细胞损伤模型,发现Gen对大鼠海马神经元损伤具有保护作用[2]。张静等[3]发现,Gen可提高视网膜细胞抗炎和抗氧化能力,抑制视网膜细胞凋亡,从而起到对大鼠视网膜损伤的保护作用。Gen的生物学活性研究逐渐成为热点,但关于Gen的快速定量检测方法,目前尚不明确。传统的仪器检测方法主要包括高效液相色谱、液质联用色谱、气相色谱等,这些检测需要购置昂贵的仪器设备,实验步骤烦琐,耗时长,成本高[4-5]。林胜军等[6]利用高效液相色谱-串联质谱法测定大豆样本中Gen的含量,质量浓度在2.0~200 ng · mL-1范围内具有良好的线性,但需要经过柱分离、梯度洗脱和负离子电离等一系列操作,步骤烦琐,经济成本较高,不适用于大量样本的快速检测。酶联免疫吸附分析(enzyme-linked immunosorbent assay,ELISA)基于抗原和抗体的特异性结合,是一种成本低、高效、灵敏、快速的检测方法,适用于大量样本的现场快速检测,为Gen定量检测提供了一种新的方法。本研究利用人工抗原免疫小鼠,制备特异性识别Gen的单克隆抗体,旨在建立灵敏度高、特异性好的间接竞争ELISA检测方法,为Gen的生物学活性及药理研究提供有效工具。
卵清蛋白(OVA) (纯度 98%,批号O12IS228371),上海源叶生物科技有限公司;牛血清白蛋白(BSA)(纯度≥98%,批号WXBD8494V),购自Sigma-Aldrich公司;Gen、吗啉乙磺酸、鹰嘴豆芽素A、芹菜素,上海阿拉丁生化科技股份有限公司;尼泊尔鸢尾异黄酮,上海麦克林生化科技股份有限公司;无水乙醇、磷酸氢钠、磷酸二氢钾、氯化钠、氯化钾、浓硫酸、聚乙二醇(PEG)、NN-二甲基甲酰胺(DMF),国药集团化学试剂有限公司;弗氏佐剂、链球菌蛋白G、次黄嘌呤/胸腺嘧啶脱氧核苷培养基添加物(hypoxantin/thymidin,HT)、次黄嘌呤/氨基蝶呤/胸腺嘧啶脱氧核苷培养基添加物(hypoxantin/aminopterin/thymidin,HAT)、4-苯并吡喃酮2-羧酸、辣根过氧化物酶(HRP),Sigma公司;单克隆抗体亚型鉴定试剂盒,ThermoFisher公司;3,3’,5,5’-四甲基联苯胺(TMB),BioFx公司;SP2/0骨髓瘤细胞,美国细胞培养物收藏中心(ATCC);羊抗鼠IgG,武汉云克隆科技股份有限公司。
6只6~8周龄的雌性Balb/c小鼠购自湖北省疾病预防控制中心,喂养和实验操作严格按照《实验动物管理条例》执行。本研究动物实验在武汉云克隆动物有限公司动物实验中心进行,实验动物使用许可证号:SYXK(鄂)2023-0135。动物实验方案符合3R原则,获得武汉云克隆动物有限公司伦理审查委员会批准。
4SMR16酶标仪(波长范围 450~630 nm),武汉优尔生生命科学装备有限公司;UV2600紫外分光光度计,岛津公司;Microfuge 20台式高速离心机,Beckman公司;96孔酶标板,购自Corning公司;FA604A天平(精度为0.1 mg),上海精天电子仪器有限公司;MCO-15AC二氧化碳细胞培养箱,Panasonic公司。
Gen偶联抗原的合成采用曼尼希法[7]。BSA偶联物制备方法如下:向10 mL冻干瓶中加入BSA 20 mg,加入 2%吗啉乙磺酸溶液1 mL溶解。称取Gen 5 mg,溶解于无水乙醇 0.3 mL。将Gen溶液加入BSA溶液中,再加入40%甲醛溶液50 µL 混匀。将冻干瓶盖好且避光,放入摇床中振荡反应 20 h,设定温度为25 ℃。反应结束后,取出反应液,使用0.01 mol · L-1磷酸盐缓冲液(PBS,配制方法:称取十二水合磷酸一氢钠 7.06 g、二水合磷酸二氢钠 0.52 g、氯化钠 10.5 g,使用0.1 mol · L-1的稀盐酸调节pH至7.2,加水定容至1 000 mL)进行透析,即得。OVA偶联物制备方法如下:向10 mL冻干瓶中加入OVA 20 mg,加入2%吗啉乙磺酸溶液1 mL溶解;称取 Gen 5 mg,溶解于0.3 mL无水乙醇;将Gen溶液加入OVA溶液中,再加入40%甲醛溶液30 µL 混匀,将冻干瓶盖好且避光,放入摇床中振荡反应20 h,设定温度为25 ℃,反应结束后,取出反应液,使用0.01 mol · L-1 PBS进行透析,即得。合成的抗原使用紫外光谱扫描法进行鉴定,鉴定方法如下:使用1×PBS分别配制质量浓度为0.5 mg · mL-1的BSA、OVA、Gen、Gen-BSA(以BSA计)、Gen-OVA(以OVA计)溶液,基线调零后在200~700 nm波长范围内对以上5种溶液进行紫外光谱扫描,记录不同波长下的吸收度,分析数据并组合紫外光谱图,利用组合图分析Gen是否与BSA、OVA偶联。偶联成功的紫外光谱图见图1
图1所示,Gen在263 nm处有特征吸收,而Gen-BSA和Gen-OVA在271 nm附近出现宽吸收峰,相对于Gen吸收峰有所红移,提示Gen和载体蛋白成功偶联。
首次免疫时将Gen-BSA与弗氏完全佐剂按照1 : 1等体积混合。选取4只6~8周龄的雌性Balb/c小鼠,分别使用乳化后的抗原进行腹部多点皮下注射免疫,21 d后开展第2次免疫,取与弗氏不完全佐剂混合后的Gen-BSA,对接受首次免疫的小鼠进行皮下注射免疫;分别在第42天和第63天进行第3、4次腹腔免疫;第77天进行冲击免疫。每只小鼠免疫剂量为100 μg(以BSA计)。取小鼠脾细胞与SP2/0骨髓瘤细胞,按5 : 1(数量配比)比例,在50% PEG的作用下融合,置于选择性培养基中培养,取细胞上清,使用间接ELISA法筛选阳性杂交瘤细胞。采用有限稀释法筛选阳性亚克隆。准备2只6~8周龄雌性Balb/c小鼠,按照每只小鼠0.5 mL的剂量腹腔注射灭菌石蜡油,7 d后注射筛选的单克隆杂交瘤细胞株,待小鼠腹部隆起且体积趋于稳定时采集腹水,用蛋白G亲和层析法进行腹水纯化并初步检测效价。共分离2株单克隆抗体,命名为C1和C3。使用单克隆抗体亚型鉴定试剂盒对单克隆抗体进行亚型鉴定,鉴定结果见图2
本研究筛选获得2株稳定分泌抗Gen抗体的杂交瘤细胞株(C1和C3)。经亚型鉴定,2株单克隆抗体均为IgG1。以4 μg · mL-1 Gen-OVA(以OVA计)包被酶标板,将腹水进行系列倍比稀释,采用间接ELISA对腹水进行效价测定,以450 nm处的吸收度为2.0时对应的腹水稀释度初步作为腹水的效价。C1、C3的效价分别为1 : 32 000、1 : 40 000,选择C3作为建立间接竞争ELISA方法的检测抗体。
采用方阵滴定法[10-11]初步筛选抗原包被浓度和对应的抗体稀释度的组合,具体操作如下:分别使用0.01 mol · L-1 PBS将Gen-OVA稀释为0.125、0.25、0.5、1、2、4 µg · mL-1 6个浓度梯度(以OVA计),包被酶标板,4 ℃孵育过夜。洗涤后加入1%明胶200 μL,37 ℃封闭1 h,每孔加入用0.01 mol · L-1 PBS倍比稀释(1 : 4 000、1 : 8 000、1 : 16 000、1 : 32 000、1 : 64 000、1 : 128 000、1 : 256 000)的单克隆抗体50 µL。37 ℃反应1 h后洗涤,加入HRP标记的羊抗鼠 IgG 100 µL,37 ℃反应40 min。洗涤后每孔加入TMB 100 µL,10 min后加入终止液。在450 nm波长下测定各孔吸收度,结果如表1所示。选择了2组包被浓度和抗体稀释度组合:2 μg · mL-1、64 000和1 μg · mL-1、32 000,这2个孔的吸收度在2.0左右,且与邻孔差异明显,是包被抗原浓度和抗体稀释度较好的组合。
以方阵滴定法确定的抗原包被浓度和对应的抗体稀释度的组合进行间接竞争ELISA,方法如下:分别使用0.01 mol · L-1 PBS将Gen-OVA稀释为1、2 μg · mL-1(以OVA计),包被酶标板。明胶封闭后每孔加入倍比稀释(1 : 32 000、1 : 64 000)的单克隆抗体50 µL。Gen溶液浓度设置为0、5、10、20、40、80 ng · mL-1,每孔加50 µL。37 ℃反应1 h后洗涤。显色终止后在450 nm波长下测定各孔吸收度。绘制标准曲线(图3),以Gen溶液浓度为0的孔测得的450 nm处的吸收度为A0,不同Gen溶液浓度在450 nm处的吸收度为A,计算抑制率为50%时对应的竞争物浓度,即IC50。以A0接近2.0、IC50最低所对应的包被浓度作为抗原的最佳包被浓度。结果如下:当包被浓度为1 μg · mL-1,抗体稀释度为1 : 32 000时,A0为2.116,IC50为19.99 ng · mL-1;当包被浓度为2 μg · mL-1,抗体稀释度为1 : 64 000时,A0为1.895,IC50为23.58 ng · mL-1。最终确立最佳包被浓度为1 μg · mL-1,抗体稀释度初步确定为1 : 32 000。
以最佳包被浓度包被酶标板,对应的抗体稀释度为基础,等差设计6个抗体稀释度,进行间接竞争ELISA。绘制标准曲线,计算IC50。以A0接近2.0、IC50最低时对应的抗体稀释度作为最佳抗体稀释度,结果如图4所示。最终选择1 : 36 000为最佳抗体稀释度。
使用确立的最佳包被浓度和最佳抗体稀释度,分别改变一抗孵育条件(37 ℃ 30 min,37 ℃ 1 h),二抗孵育条件(37 ℃ 30 min,37 ℃ 1 h)以及TMB显色时间(10、15 min),在450 nm波长下测定各孔吸收度,计算IC50,对间接ELISA反应条件进行优化。选择IC50相对较低的一组作为优化后的试验条件:一抗孵育条件为37 ℃ 30 min,二抗孵育条件为37 ℃ 1 h,TMB显色时间为10 min,见表2
根据确立的最佳抗原包被浓度和抗体稀释度,参照“2.3.2”项下试验步骤,使用0.01 mol · L-1 PBS将Gen制备成0、5、10、20、40、80 ng · mL-1 6个质量浓度的溶液,进行间接竞争ELISA试验。以Gen浓度的对数值为横坐标,各浓度对应的结合率A/A0A是不同浓度Gen在450 nm处的吸收度)为纵坐标,使用GraphPad Prism绘制标准曲线(图5),计算IC50。IC50表示灵敏度,表示Gen与检测抗原竞争的半抑制浓度;以IC20~IC80作为线性检测范围。结果间接竞争ELISA抑制曲线线性良好,IC50为16.15 ng · mL-1。标准曲线的线性检测范围为3.78~55.85 ng · mL-1(IC20~IC80),检测限为1.63 ng · mL-1(IC10)。
分别取鹰嘴豆芽素A、尼泊尔鸢尾异黄酮、芹菜素、4-苯并吡喃酮2-羧酸0.5 mg,溶解于1 mL DMF中,制备成溶液。使用0.01 mol · L-1 PBS分别配制质量浓度为1.0、10、100 ng · mL-1和1、10 μg · mL-1的溶液,使用建立的间接竞争ELISA法进行检测,计算交叉反应率,结果见表3图6
结果显示,该方法与Gen的反应率为100%,对鹰嘴豆芽素A有弱交叉反应,其交叉反应率为4.96%;对尼泊尔鸢尾异黄酮有弱交叉反应,其交叉反应率为2.40%;对芹菜素、4-苯并吡喃酮2-羧酸的交叉反应率均小于0.1%,表明本研究建立的Gen间接竞争ELISA方法具有较高的特异性。
取Gen 0.5 mg溶于1 mL DMF,配制成溶液。使用0.01 mol · L-1 PBS将Gen溶液浓度稀释成500、50 ng · mL-1。选取150 µL来源于家兔的血清、血浆各3份,分别加入质量浓度为50 ng · mL-1的Gen 溶液17 µL,配制终浓度为20 ng · mL-1 的加标样品;取100 µL血清、血浆各3份,分别加入质量浓度为50 ng · mL-1的Gen 溶液67 µL,配制终浓度为20 ng · mL-1 的加标样品;取150 µL血清、血浆各3份,分别加入质量浓度为50 ng · mL-1的Gen 溶液16.7 µL,配制终浓度为5 ng · mL-1 的加标样品。进行间接竞争ELISA试验,计算回收率和RSD,结果见表4,3次检测的RSD低于12%,血清和血浆添加Gen的回收率在85%~110%,表明准确度良好。
使用0.01 mol · L-1 PBS将Gen配制成5、20、50 ng · mL-1 3种浓度的溶液,重复测定3次,记录并计算测定结果并测定RSD。结果显示,当Gen质量浓度为5 ng · mL-1时,3次测定的质量浓度分别为4.82、5.15、4.90 ng · mL-1,平均值为4.96 ng · mL-1,RSD为3.5%;当Gen质量浓度为20ng · mL-1时,3次测定的质量浓度分别为21.07、20.83、21.54 ng · mL-1,平均值为21.15 ng · mL-1,RSD为1.7%;当Gen质量浓度为50 ng · mL-1时,3次测定的质量浓度分别为52.39、49.52、51.69 ng · mL-1,平均值为51.20 ng · mL-1,RSD为2.9%。同一样本重复检测的RSD均小于10%,说明重复性试验结果良好。
使用0.01 mol · L-1 PBS将Gen配制成5、20和50 ng · mL-1 3种浓度的溶液,由A、B 2名实验人员在不同时间进行检测,每人独立测试3次,记录并计算测定结果。结果见表5,不同人员在不同时间点检测样本,RSD均不超过10%,表明该方法的批间精密度符合要求。
Gen是广泛存在于大豆中的黄酮类化合物,由于其结构特殊,可与雌二醇竞争结合雌激素受体后表现出双向调节作用,对于雌激素水平偏低所致的疾病,比如更年期综合征、骨质疏松等具有保护作用[12~13]。近年来,Gen的药理作用机制受到广泛关注。多项研究表明,Gen在机体内除了可发挥雌激素类似作用外,还在神经元损伤、细胞衰老等多个方面有重要作用[14~16]。精准定量和组织原位分析对Gen作用机制的研究具有十分重要的意义。
间接竞争ELISA是针对小分子化合物常用的分析方法。以特异性单克隆抗体为基础建立的免疫分析技术具有抗干扰性强和灵敏度高的特点,在当代药物分析和药理机制领域具有广泛的应用[17]。抗体的性质直接决定了所建方法的灵敏度和特异性,也是试剂盒检测效果的关键因素。Gen为小分子半抗原,不具备免疫原性。本研究采用曼尼希法将Gen和BSA偶联,合成人工抗原,免疫小鼠,筛选单克隆抗体。利用人工抗原免疫小鼠获得了腹水效价为1 : 40 000的单克隆抗体,利用方阵滴定初步确定抗原包被浓度和抗体稀释度。为了获得更加准确的抗原包被和抗体稀释组合,进行间接竞争ELISA试验,计算不同条件下的IC50,最终确定最佳包被浓度为1 μg · mL-1,抗体稀释度确定为1 : 36 000。还对间接竞争ELISA试验条件进行了一系列的优化,最终确定一抗、二抗及TMB的最佳反应时间。
本实验建立的间接竞争ELISA检测方法检测IC50为16.15 ng · mL-1,线性检测范围为3.78~55.85 ng · mL-1(IC20~IC80),检测限为1.63 ng · mL-1,说明该方法对Gen的检测具有较高的灵敏度。同时,为了验证其特异性,选取了鹰嘴豆芽素A、尼泊尔鸢尾异黄酮、芹菜素等与Gen结构相近的类似物进行交叉反应试验,发现选用单克隆抗体除了与鹰嘴豆芽素A、尼泊尔鸢尾异黄酮有弱交叉反应外,与其余2个物质的交叉反应均小于0.1%,证实建立的检测方法特异性良好。为进一步证实该方法对Gen的检测效果,选取来源于家兔的血清和血浆,制备低、中、高3种不同Gen浓度的加标样品进行回收试验,回收率在85%~110%,说明该检测方法的准确性较好。精密度试验显示批内和批间试验的RSD在10%以下,提示该方法检测Gen精密度良好。
本研究使用人工抗原免疫小鼠,制备特异性识别Gen的单克隆抗体,可为研发其他免疫学检测试剂提供基础材料。挑选优质的单克隆抗体建立间接竞争ELISA检测方法,可快速、准确地对Gen进行定量检测,为药理机制的研究提供有效的工具,并可为多种相关疾病的研究提供助力。
