Objective: To establish an ultra-performance liquid chromatography-triple quadrupole mass spectrometry(UPLC-QQQ MS/MS) method for the simultaneous determination of 14 active components in Qiyu Sanlong decoction,including L-argnine, monotropein, deacetyl asperulosidic acid, rutin, peimisine, calycosin-7-O-β-D-glucoside,caffeic acid, solasonine, solamargine, p-coumaric acid, ferulic acid, calycosin, astragaloside Ⅳ and astragaloside Ⅰ. Methods: The chromatographic separation experiment was performed on an ACQUITY UPLC BEH C18 column(2.1 mm×100 mm, 1.7 μm), with gradient elution of 0.1% formic acid aqueous solution (A)-acetonitrile (B) as mobile phase at the flow rate of 0.2 mL · min-1, injection volume of 5 μL, and column temperature of 35 ℃. The ion source was an electrospray ionization source (ESI), the scanning mode was simultaneous scanning of positive and negative ions, and the monitoring mode was multiple reaction monitoring. Results: The 14 active components revealed good linearity within their respective ranges (r>0.995 0), RSDs of precision and repeatability were below 2%, RSDs of stability were below 3%, and the average recoveries ranged from 88.4% to 108.5% with the RSDs ranging from 0.020%to 3.6%. The content ranges of the aforementioned 14 components in 10 batches of self-prepared Qiyu Sanlong decoction were as follows (in μg · g-1): 667.28-785.78, 165.72-197.27, 196.32-275.60, 17.60-26.52, 4.68-10.75,279.12-388.05, 26.00-47.57, 385.52-442.77, 288.00-358.82, 629.88-839.02, 86.67-125.83, 51.58-65.83, 25.50-37.53, and 55.50-76.13. Conclusion: The UPLC-QQQ MS/MS method established in this study is rapid, accurate,sensitive and repeatable, which can provide a reference for the quality control of Qiyu Sanglong decoction.

目的:建立超高效液相色谱-三重四极杆串联质谱(UPLC-QQQ MS/MS)法测定芪玉三龙汤中14个活性成分(L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ)的含量。方法:色谱分析采用Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱,流速0.2 mL · min-1,进样量5 μL,柱温35 ℃。质谱分析采用电喷雾离子源(ESI),正负离子模式扫描,多反应监测模式。结果:14个活性成分在各自浓度范围内线性关系良好,r均>0.995 0;精密度和重复性RSD均<2%,稳定性RSD<3%;平均加样回收率为88.4%~108.5%,RSD为0.020%~3.6%;10批自制芪玉三龙汤中上述14个成分的含量范围分别为667.28~785.78、165.72~197.27、196.32~275.60、17.60~26.52、4.68~10.75、279.12~388.05、26.00~47.57、385.52~442.77、288.00~358.82、629.88~839.02、86.67~125.83、51.58~65.83、25.50~37.53、55.50~76.13 μg · g-1结论:本研究基于UPLC-QQQ MS/MS建立的芪玉三龙汤多种活性成分的含量测定方法具有快速准确,灵敏度高,专属性好的特点,可为芪玉三龙汤的质量控制提供参考。

目的:建立超高效液相色谱-三重四极杆串联质谱(UPLC-QQQ MS/MS)法测定芪玉三龙汤中14个活性成分(L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ)的含量。方法:色谱分析采用Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱,流速0.2 mL · min-1,进样量5 μL,柱温35 ℃。质谱分析采用电喷雾离子源(ESI),正负离子模式扫描,多反应监测模式。结果:14个活性成分在各自浓度范围内线性关系良好,r均>0.995 0;精密度和重复性RSD均<2%,稳定性RSD<3%;平均加样回收率为88.4%~108.5%,RSD为0.020%~3.6%;10批自制芪玉三龙汤中上述14个成分的含量范围分别为667.28~785.78、165.72~197.27、196.32~275.60、17.60~26.52、4.68~10.75、279.12~388.05、26.00~47.57、385.52~442.77、288.00~358.82、629.88~839.02、86.67~125.83、51.58~65.83、25.50~37.53、55.50~76.13 μg · g-1结论:本研究基于UPLC-QQQ MS/MS建立的芪玉三龙汤多种活性成分的含量测定方法具有快速准确,灵敏度高,专属性好的特点,可为芪玉三龙汤的质量控制提供参考。

Objective: To establish an ultra-performance liquid chromatography-triple quadrupole mass spectrometry(UPLC-QQQ MS/MS) method for the simultaneous determination of 14 active components in Qiyu Sanlong decoction,including L-argnine, monotropein, deacetyl asperulosidic acid, rutin, peimisine, calycosin-7-O-β-D-glucoside,caffeic acid, solasonine, solamargine, p-coumaric acid, ferulic acid, calycosin, astragaloside Ⅳ and astragaloside Ⅰ. Methods: The chromatographic separation experiment was performed on an ACQUITY UPLC BEH C18 column(2.1 mm×100 mm, 1.7 μm), with gradient elution of 0.1% formic acid aqueous solution (A)-acetonitrile (B) as mobile phase at the flow rate of 0.2 mL · min-1, injection volume of 5 μL, and column temperature of 35 ℃. The ion source was an electrospray ionization source (ESI), the scanning mode was simultaneous scanning of positive and negative ions, and the monitoring mode was multiple reaction monitoring. Results: The 14 active components revealed good linearity within their respective ranges (r>0.995 0), RSDs of precision and repeatability were below 2%, RSDs of stability were below 3%, and the average recoveries ranged from 88.4% to 108.5% with the RSDs ranging from 0.020%to 3.6%. The content ranges of the aforementioned 14 components in 10 batches of self-prepared Qiyu Sanlong decoction were as follows (in μg · g-1): 667.28-785.78, 165.72-197.27, 196.32-275.60, 17.60-26.52, 4.68-10.75,279.12-388.05, 26.00-47.57, 385.52-442.77, 288.00-358.82, 629.88-839.02, 86.67-125.83, 51.58-65.83, 25.50-37.53, and 55.50-76.13. Conclusion: The UPLC-QQQ MS/MS method established in this study is rapid, accurate,sensitive and repeatable, which can provide a reference for the quality control of Qiyu Sanglong decoction.

芪玉三龙汤是治疗非小细胞肺癌的名老中医验方,疗效确切,临床应用已有20余年[1]。该方由黄芪、龙葵、天龙、地龙、泽漆、白花蛇舌草、莪术、玉竹、薏苡仁、川贝母等10味中药组成,诸药合用共奏益气养阴、化痰祛瘀、解毒消积之功。现代药理学研究[2-4]表明,芪玉三龙汤抑制非小细胞肺癌的机制可能是与其干预mTOR/Beclin1/LC3信号轴相关分子和Bax、cleaved Caspase-3和Bcl-2蛋白表达,以及通过PD-1信号通路增强Th1免疫反应有关。对芪玉三龙汤的药效物质基础和质量控制进行深入全面的研究,对推动其临床应用和相关制剂的开发,具有深远的意义。
课题组前期利用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q TOF MSE)快速辨识了芪玉三龙汤中166个化学成分[5],然后通过体内药物分析,分别在大鼠血浆、尿液和粪便中识别了101、85、93个芪玉三龙汤的原型成分和代谢产物[6-8],这些成分构成了芪玉三龙汤抑制非小细胞肺癌的潜在药效物质基础,最后通过“谱-效”关系研究,筛选出14个芪玉三龙汤抑制非小细胞肺癌的活性成分[9-10]。对芪玉三龙汤活性成分进行定量研究有助于控制复方制剂的质量,前期尝试运用UPLC法测定芪玉三龙汤中咖啡酸、对香豆酸、芦丁、毛蕊异黄酮和毛蕊异黄酮-7-O-β-D-葡萄糖苷5个成分的含量,但分离效率低,分析时间长。超高效液相色谱-三重四极杆串联质谱(UPLC-QQQ MS/MS)具有选择性好、专属性强和灵敏度高等优势,已广泛应用于中药复方的多组分定量分析[11-13]。基于此,本研究对芪玉三龙汤中的14个活性成分进行含量测定,旨在为芪玉三龙汤的全面质量控制提供科学方法和依据。
黄芪(批号220401、230701、230702)、龙葵(批号210601、220401、221101)、天龙(批号211201、220308002、230301)、地龙(批号220601、220901、230201)、泽漆(批号210701、210902、230701)、白花蛇舌草(批号211201、220501、221001)、莪术(批号220801、230201、230501)、玉竹(批号210701、220601、221201)、薏苡仁(批号A220113、A221211、A230320)、川贝母(批号210501、230201、2304030),以上10味中药饮片均购于北京同仁堂(合肥),由安徽中医药大学中药资源中心杨青山副教授鉴定。
对照品L-精氨酸(批号MUST-23090501,纯度99.59%)、水晶兰苷(批号MUST-21102315,纯度99.86%)、去乙酰基车叶草苷酸(批号MUST-22020810,纯度98.17%)、芦丁(批号MUST-22111214,纯度99.04%)、毛蕊异黄酮-7-O-β-D-葡萄糖苷(批号MUST-20031920,纯度99.86%)、咖啡酸(MUST-23061118,纯度99.82%)、澳洲茄碱(批号MUST-20042004,纯度99.61%)、澳洲茄边碱(批号MUST-19102621,纯度99.43%)、对香豆酸(批号MUST-20050603,纯度99.96%)、阿魏酸(批号MUST-23072910,纯度99.99%)、毛蕊异黄酮(批号MUST-19120901,纯度99.84%)、黄芪甲苷(批号MUST-19091308,纯度99.89%)和黄芪皂苷Ⅰ(批号MUST-20042906,纯度99.10%),购于成都曼思特生物科技有限公司;贝母辛(批号B20082,纯度98.00%),购于上海源叶生物科技有限公司。甲酸(Sigma-Aldrich公司)和甲醇、乙腈(Merck公司)均为质谱纯,水为超纯水,其余试剂均为分析纯。
ACQUITY UPLC超高效液相色谱仪串联Xevo TQS三重四极杆质谱仪(Waters公司),KQ-500DB型数控超声波清洗器(昆山市超声仪器有限公司),BP211D十万分之一电子天平(Sartorius公司),5430R低温高速离心机(Eppendorf公司),Milli-Q Gradient A10超纯水机(Millipore公司),RE-3000A型旋转蒸发仪(上海亚荣生化仪器厂),ALPHA1-2LD真空冷冻干燥机(Christ公司)。
称取方中诸药,使用水煎法制备中药汤剂。每副药材134 g(黄芪30 g、龙葵20 g、天龙6 g、地龙6 g、泽漆6 g、白花蛇舌草20 g、莪术10 g、玉竹10 g、薏苡仁20 g、川贝母6 g),在10倍纯净水(1 340 mL)中浸泡1 h,武火煮沸后文火煎煮1.5 h,提取液通过4层纱布过滤,残渣加入8倍纯净水(1 072 mL),武火煮沸后文火煎煮40 min,提取液再次通过4层纱布过滤;合并2次煎煮液,50 ℃真空旋转蒸发浓缩至一定体积,放置于-80 ℃冰箱预冻,然后真空冷冻干燥,得芪玉三龙汤冻干粉,于干燥器中避光保存。取冻干粉0.1 g,精密称定,置100 mL具塞锥形瓶中,精密加入90%甲醇水50 mL,密塞,摇匀,称量,超声(功率250 W,频率40 kHz)提取30 min,待冷却后,再称量,用90%甲醇水补足减失的量。在4 ℃、3 000 r · min-1的条件下离心10 min,取上清液涡旋混匀,过0.22 μm微孔滤膜,即得,4 ℃下保存。
精密称取L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ的对照品各约5.00 mg,分别置5 mL量瓶中,用甲醇溶解并稀释至刻度,配制成1.00 mg · mL-1的对照品储备液,-20 ℃下保存备用。精密吸取各对照品储备液适量,涡旋混匀,过0.22 μm微孔滤膜,即得混合对照品溶液,4 ℃下保存备用。
按照“2.1.1”项下方法制备缺芪玉三龙汤方中诸药的阴性样品溶液,4 ℃下保存。
采用Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm)色谱柱,以0.1%甲酸水(A)-乙腈(B)为流动相,梯度洗脱(0~2 min,5%B;2~3 min,5%B→45%B;3~5 min,45%B→60%B;5~5.5 min,60%B→67%B;5.5~6 min,67%B→70%B;6~7.5 min,70%B→75%B;7.5~9 min,75%B→80%B;9~10 min,80%B→86%B;10~12 min,86%B;12~14 min,86%B→5%B;14~15 min,5%B),流速0.2 mL · min-1,进样量5 μL,柱温35 ℃。
离子源为电喷雾离子源(ESI),扫描方式为正负离子同时扫描,监测模式为多反应监测模式(MRM),MassLynx 4.2软件(Waters)作为质谱工作站对质谱数据进行采集和处理。正负模式下毛细管电压均为2.0 kV,离子源温度500 ℃,脱溶剂气流量1 000 L · h-1,数据采集时间15 min。14个待测成分质谱参数详见表1
分别取“2.1”项下供试品溶液、混合对照品溶液和阴性样品溶液适量,按“2.2”项下色谱、质谱条件进样测定,记录各成分MRM色谱图(图1)。样品与对照品中各待测成分的色谱峰保留时间一致,阴性样品相同保留时间处无色谱峰出现,表明方法专属性良好。
精密吸取“2.1.2”项配制的混合对照品溶液适量,用甲醇逐级稀释配制为10个系列工作浓度的混合对照品溶液,按“2.2”项下色谱、质谱条件进样测定。以各对照品溶液的质量浓度(X,ng · mL-1)为横坐标,相应的峰面积Y为纵坐标,进行线性回归,得线性方程、相关系数和线性范围。以信噪比(S/N)为3和10时的浓度确定各成分的检测限(LOD)和定量限(LOQ)。结果(表2)表明,各成分在相应的浓度范围内线性关系良好(r>0.995 0)。
取同一份“2.1.2”项下配制的混合对照品溶液适量,按“2.2”项下色谱、质谱条件连续进样。1 d内连续进样6次,记录各成分峰面积并计算RSD,测定日内精密度,L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ峰面积的RSD分别为0.56%、0.58%、0.66%、0.88%、1.0%、0.52%、0.13%、0.91%、0.50%、0.55%、0.51%、0.54%、1.6%、0.96%;连续测定3 d,每天连续进样6次,测得日间精密度,上述14个成分峰面积的RSD分别为0.91%、0.62%、0.70%、0.96%、0.81%、0.58%、0.97%、1.4%、0.67%、1.7%、1.2%、0.76%、1.8%、1.2%,均<2.0%,表明仪器精密度良好。
取同一批样品(批号20230910),按“2.1.1”项下方法平行制备6份供试品溶液,按“2.2”项下色谱、质谱条件进样,记录各成分峰面积,计算含量及RSD。L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ的含量分别为695.48、177.85、284.36、29.08、11.39、366.98、30.98、385.02、394.03、529.17、113.46、59.57、31.16和86.47 μg · g-1,RSD分别为0.59%、1.1%、0.88%、1.4%、1.5%、0.92%、1.0%、0.83%、1.1%、1.2%、0.69%、0.19%、1.3%、0.86%,均<2.0%,表明方法重复性良好。
取“2.1.1”项下供试品溶液(批号20230910)一份,在室温条件下放置0、2、4、6、8、10、12、24 h,按“2.2”项下色谱、质谱条件进样,记录24 h内不同时间点各成分的峰面积并计算RSD。结果L-精氨酸、水晶兰苷、去乙酰基车叶草苷酸、芦丁、贝母辛、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、澳洲茄碱、澳洲茄边碱、对香豆酸、阿魏酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ峰面积的RSD分别为1.6%、1.2%、1.3%、1.8%、1.6%、1.3%、2.6%、1.6%、2.7%、2.1%、2.6%、1.6%、1.3%、1.2%,均<3.0%,表明供试品溶液于室温下放置24 h稳定性良好。
精密称取9份已知含量的芪玉三龙汤冻干粉样品(批号20230910),以分析方法中样品测定的浓度为100%计,按样品测定浓度的50%、100%、150%加入一定量的混合对照品,每种浓度分别平行制备3份,按“2.2”项下色谱、质谱条件进样检测,记录峰面积,计算含量和回收率。结果见表3,平均加样回收率为88.4%~108.5%,RSD为0.020%~3.6%,表明该方法准确度良好。
取10批自制芪玉三龙汤冻干粉样品,每批次按“2.1.1”项下方法平行制备3份供试品溶液,按“2.2”项下条件进样检测,结果见表4(单位μg · g-1中的g为芪玉三龙汤冻干粉的质量)。结果显示,精氨酸和对香豆酸在芪玉三龙汤中的含量最高,平均含量分别为693.98 μg · g-1和711.12 μg · g-1,批间RSD分别为5.0%和11.7%;水晶兰苷、去乙酰基车叶草苷酸、毛蕊异黄酮-7-O-β-D-葡萄糖苷、澳洲茄碱、澳洲茄边碱和阿魏酸的含量较高,平均含量分别为176.97、240.40、325.15、420.96、324.08和105.11 μg · g-1,批间RSD分别为4.8%、9.1%、9.8%、5.0%、7.8%和12.0%;芦丁、贝母辛、咖啡酸、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ的含量较低,平均含量分别为20.53、6.85、36.51、58.62、28.65和66.56 μg · g-1,批间RSD分别为14.4%、34.6%、20.4%、8.6%、12.8%和10.4%,其中,贝母辛不仅含量最低(平均含量为6.85 μg · g-1),而且批间含量差异大(批间RSD为34.6%)。
本研究的测定对象为芪玉三龙汤中14个化学成分,其主要结构有环烯醚萜类、生物碱类、皂苷类、黄酮类、苯丙素类和氨基酸类等。当采用LC-UV检测时,各类待测成分紫外最大吸收波长不同,且皂苷类成分如黄芪甲苷、黄芪皂苷Ⅰ的紫外吸收弱,灵敏度低;当使用蒸发光散射检测器时,其他成分的灵敏度则无法达到要求。由于芪玉三龙汤中含有的化学成分较为复杂,UPLC需要设计较长的洗脱时间才能完成样品中复杂组分的分离,实验耗时且溶剂消耗量大。UPLC-QQQ MS/MS选择性好,专属性强,可以有效分离和定量分析芪玉三龙汤中多种化学成分。QqQ-MS/MS的MRM模式通过2次质量筛选分别选择待测成分的准分子离子和产物离子,进而精准锁定待测组分进入质量分析器,极大提升了检测灵敏度,且降低了分离的难度,适用于中药(复方)等复杂样品中活性成分的含量测定及药代动力学研究。因此,本研究选择UPLC-QQQ MS/MS建立测定芪玉三龙汤中14个活性成分含量的方法。
课题组前期利用UPLC-Q TOF-MS对芪玉三龙汤作了体内外的物质基础研究[5-8],随后建立10批芪玉三龙汤的指纹图谱,以共有峰峰面积为“谱”,以抑制A549细胞增殖、水平迁移、立体迁移和侵袭能力为“效”,通过灰色关联度分析(GRA)、偏最小二乘回归和反向传播神经网络模型结合平均影响值算法建立谱效关系,筛选出去乙酰基车叶草苷酸、毛蕊异黄酮-7-O-β-D-葡萄糖苷、咖啡酸、对香豆酸、澳洲茄碱、澳洲茄边碱等为芪玉三龙汤抑制非小细胞肺癌的活性成分群[9]。进一步以肺腺癌模型大鼠血浆中不同时间点中芪玉三龙汤共有成分为“谱”,以代谢组学获得的内源性生物标志物为“效”,通过GRA和典型相关分析建立谱效关系,预测出贝母辛、对香豆酸、咖啡酸、阿魏酸、毛蕊异黄酮、黄芪皂苷Ⅰ和澳洲茄胺等的代谢物为芪玉三龙汤抑制非小细胞肺癌的药效物质[10]
在上述通过谱效关联性分析筛选出的药效物质中,毛蕊异黄酮、毛蕊异黄酮-7-O-β-D-葡萄糖苷、黄芪甲苷、黄芪皂苷Ⅰ来源于君药黄芪,澳洲茄碱和澳洲茄边碱来源于君药龙葵,芦丁来源于臣药泽漆,水晶兰苷、去乙酰基车叶草苷酸和对香豆酸来源于佐药白花蛇舌草,阿魏酸、咖啡酸来源于佐药玉竹,贝母辛来源于使药川贝母。据文献报道,黄芪甲苷通过AMPK信号通路诱导M1巨噬细胞极化[14],芦丁阻碍A549细胞与胶原蛋白Ⅰ、Ⅳ型和纤连蛋白的黏附[15],毛蕊异黄酮-7-O-β-D-葡萄糖苷增强p53基因在细胞质中的表达[16],对香豆酸、毛蕊异黄酮、澳洲茄碱和澳洲茄边碱调控Bcl-2、Bax等与凋亡相关蛋白的表达[17-20],从而抑制肺癌细胞生长、侵袭、迁移与增殖扩散,诱导其凋亡;去乙酰基车叶草苷酸和水晶兰苷具有抗炎镇痛,缓解氧化应激的作用[21-22];咖啡酸和阿魏酸具有抗氧化、抗炎、抗肿瘤等活性[23-24];贝母辛具有明显的抗肿瘤活性及低毒性[25]。因此,基于课题组前期谱效关系研究结果以及文献调研,选择水晶兰苷、澳洲茄碱、黄芪甲苷等14个活性成分进行含量测定,以期合理地控制芪玉三龙汤质量。
首先分别在正负离子模式下对14个待测成分进行全扫描,确定各成分检测模式。实验过程中发现,负离子模式下黄芪皂苷Ⅰ与甲酸结合生成的准分子离子峰[M+HCOOH-H]-比[M-H]-的丰度高且稳定,故选择[M+HCOOH-H]-作为其准分子离子峰,其余化合物选择[M-H]-或[M+H]+。然后调整去簇电压,使各成分的准分子离子峰响应最高,接着进行碰撞诱导解离反应,获得二级碎片离子;最后通过调整碰撞能量,选择信号稳定且强度较高的碎片离子作为产物离子。最终得到14个成分的最佳质谱参数,详见表1
由芪玉三龙汤中14个活性成分的含量测定结果可知,水晶兰苷、去乙酰基车叶草苷酸、毛蕊异黄酮-7-O-β-D-葡萄糖苷、澳洲茄碱、澳洲茄边碱和阿魏酸的平均含量均达到100 μg · g-1以上,RSD均小于15%,表明这些成分在复方中含量高,且批间差异小,其含量较高的原因可能是它们为白花蛇舌草、黄芪、龙葵、玉竹的主要化学成分,且上述4味药材在复方中的用量占比较高(分别约占芪玉三龙汤的14.9%、22.4%、14.9%和7.5%)。芦丁、咖啡酸、贝母辛、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ的平均含量较低,均低于100 μg · g-1;其中芦丁、毛蕊异黄酮、黄芪甲苷和黄芪皂苷Ⅰ的批间RSD均低于15%,表明批间差异小;而咖啡酸和贝母辛的批间RSD分别达到20.4%和34.6%,提示这2个成分的批间差异较大。咖啡酸的批间差异可能与其结构中邻苯二酚结构的强还原性有关,导致其在加工与储存期间易发生氧化反应,进而含量呈现批间差异。贝母辛的批间差异可能更多与川贝母饮片产地、种植采收、加工储存等外部因素有关,通过影响生物碱的生物合成与积累,最终加剧批间含量差异。
本研究建立的UPLC-QQQ MS/MS法快速、准确,其专属性、精密度、重复性、稳定性等均通过方法学验证,能够用于芪玉三龙汤中14个活性成分的含量测定,有利于控制芪玉三龙汤质量,为确保芪玉三龙汤临床用药的有效性与安全性提供了数据支撑。
