Objective:

To develop the first batch of traditional Chinese medicine chemical reference substance 2-acetate regaloside A used in standard of Baihe (Juandan) Peifang Keli.

Methods:

The structure was elucidated by nuclear magnetic resonance spectrometry, mass spectrometry, ultraviolet spectroscopy and infrared spectroscopy. Purity analysis, determination of related substance and investigation of other physical and chemical parameters were carried out by using analytical different techniques such as high performance liquid chromatography, gas chromatography, coulometry and ignition residue method. On the above basis, the mass balance was used for value determination, and the 1H quantitative nuclear magnetic resonance method was applied for further verification.

Results:

The structure of the traditional Chinese medicine chemical reference substance was determined as 2-acetate regaloside A, and it was identified as a new compound by SciFinder search. The calibration value by mass balance method was 99.31%, and result of 1H quantitative nuclear magnetic resonance method was 100.06% (n=3, RSD=0.47%).

Conclusion:

The accurate development and calibration of the first batch of 2-acetate regaloside A fully ensures the implementation of the standard for Baihe (Juandan) Peifang Keli.

目的:

研制标定百合(卷丹)配方颗粒标准项下首批中药化学对照品2-乙酰王百合苷A。

方法:

目的:

研制标定百合(卷丹)配方颗粒标准项下首批中药化学对照品2-乙酰王百合苷A。

方法:

采用核磁共振波谱、质谱、紫外光谱和红外光谱技术进行结构鉴定,采用高效液相色谱法、气相色谱法、库仑法、炽灼残渣法等分析技术进行纯度分析、有关物质测定及其他理化参数考察,在此基础上采用质量平衡法进行定值,同时采用氢核磁共振定量法加以佐证。

结果:

首批中药化学对照品结构鉴定为2-乙酰王百合苷A,经SciFinder检索确定为新化合物,质量平衡法定值结果为99.31%,核磁共振波谱定量法测定结果为100.06%(n=3),RSD为0.47%。

结论:

首批中药化学对照品2-乙酰王百合苷A的研制标定保障了百合(卷丹)配方颗粒标准的实施。

Objective:

To develop the first batch of traditional Chinese medicine chemical reference substance 2-acetate regaloside A used in standard of Baihe (Juandan) Peifang Keli.

Methods:

The structure was elucidated by nuclear magnetic resonance spectrometry, mass spectrometry, ultraviolet spectroscopy and infrared spectroscopy. Purity analysis, determination of related substance and investigation of other physical and chemical parameters were carried out by using analytical different techniques such as high performance liquid chromatography, gas chromatography, coulometry and ignition residue method. On the above basis, the mass balance was used for value determination, and the 1H quantitative nuclear magnetic resonance method was applied for further verification.

Results:

The structure of the traditional Chinese medicine chemical reference substance was determined as 2-acetate regaloside A, and it was identified as a new compound by SciFinder search. The calibration value by mass balance method was 99.31%, and result of 1H quantitative nuclear magnetic resonance method was 100.06% (n=3, RSD=0.47%).

Conclusion:

The accurate development and calibration of the first batch of 2-acetate regaloside A fully ensures the implementation of the standard for Baihe (Juandan) Peifang Keli.

首批中药化学对照品的研制标定首先须确证化学结构,并对其纯度、有关物质、理化性质和均匀性进行考察,这些检查符合要求后,方可进行定值研究,此外还需要进行稳定性考察[1]。众所周知,核磁共振(nuclear magnetic resonance,NMR)技术是目前有机化合物结构解析的一种重要工具,中药化学对照品的结构鉴定即主要通过解析NMR、质谱、紫外光谱和红外光谱得以实现[2-5]。纯度分析主要采用高效液相色谱或气相色谱;有关物质主要包括水分、溶剂残留和无机杂质,通常分别采用库伦水分测定仪、气相色谱和炽灼残渣测定[6-9]。中药化学对照品的定值采用国际通用的质量平衡法。同时,还需要采用至少1种不同原理的可靠方法进行定值佐证,以氢核磁共振波谱定量法(1H qNMR)最为常用,且多采用内标法[10-14]
中药化学对照品通常以已知化合物为主,本文报道的首批中药化学对照品2-乙酰王百合苷A(化学结构见图1),经SciFinder检索为新化合物,其来源是百合科植物卷丹Lilium lancifolium Thumb. 的干燥肉质鳞叶,系供百合(卷丹)配方颗粒特征图谱与含量测定项用。2-乙酰王百合苷A的准确定性与定值研究为充分保障有关标准的顺利实施提供了重要的物质基础。
Waters 2998高效液相色谱仪(Waters公司),METTLER TOLEDO C30SX库仑水分滴定仪(Mettler公司);Agilent 7890B气相色谱仪(Agilent公司);MFLC-16/12马弗炉(泰斯特公司);Agilent 8453紫外可见分光光度计(Agilent公司);Thermo iN10MX傅里叶变换显微镜红外光谱仪(Thermo公司);Bruker Ascend 600 MHz型核磁共振仪(Bruker公司);Thermo Scientific Q Exactive高分辨质谱仪(Thermo公司);Mettler TOLEDO XPE105(精度0.01 mg)、XS105(精度0.01 mg)电子天平(Mettler公司),X-显微熔点测定仪(控温型)(北京泰克仪器有限公司);SHIMADZU DTG-60热重差热同步测定仪(岛津公司);Rudolph Autopol IV旋光仪(Rudolph公司)。
2-乙酰王百合苷A对照品原料购自桂林纽斯达医药科技开发有限公司;乙腈为色谱纯,购于赛默飞世尔科技公司,水为超纯水;CD3OD(批号PR-29983/042718ME1)购于Cambridge Isotope Laboratories Inc.公司,1,4-二硝基苯(99%;批号D0819)购于东京化成工业株式会社。
样品为白色粉末;[α]-16.2°(c 0.51,CH3OH)。UV光谱(CH3OH)显示最大吸收λmax 201、212、228、313 nm,与文献[15]报道的王百合苷A和B主要紫外吸收峰基本一致。它的IR(溴化钾压片)光谱显示羟基(3 542、3 464、3 333 cm-1)、羰基(1 729、1 701 cm-1)、双键(1 636 cm-1)和苯环(1 603、1 516 cm-1)的存在。HR ESI-MS给出准分子离子峰m/z 441.141 30 [M-H]- (计算值:441.139 6),结合13C NMR谱,确定该化合物分子式为C20H26O11
1H NMR(600 MHz,CD3OD)谱图(表1)显示高场区1个甲基信号δ 2.01(3H,s,-COCH3),低场区显示1组反式双键氢信号δ 7.64 (1H,d,J=15.6 Hz,H-7’)和δ 6.35 (1H,d,J=15.6,H-8’),1组1,4-二取代苯环氢信号δ 7.48 (2H,dd,J=6.6,1.8 Hz,H-2’,6’)和δ 6.83 (2H,dd,J=6.6,1.8 Hz,H-3’,5’)以及1个糖基的端基氢信号δ 4.52 (1H,d,J=8.4 Hz,H-1”)。13C NMR (CD3OD,125 MHz)谱图(表2)显示20个碳信号,包括乙酰基碳信号δ 172.2 (COCH3)、20.9(COCH3),1组对羟基桂皮酰基碳信号δ127.1 (C-1’)、131.3 (C-2’)、116.8 (C-3’)、161.4 (C-4’)、116.8(C-5’)、131.3 (C-6’)、147.1 (C-7’)、114.6 (C-8’)和168.8 (C-9’),1组葡萄糖基信号δ 104.7(C-1”)、75.0 (C-2”)、78.0 (C-3”)、71.5 (C-4”)、77.9 (C-5”)、62.7 (C-6”),以及3个连氧碳信号δ 64.0 (C-1)、72.1 (C-2)、68.8 (C-3)。葡萄糖基端基氢偶合常数J=7.8 Hz,说明葡萄糖基是β-D-型。这些数据与文献[15]报道的王百合苷A(regaloside A)和王百合苷B(regaloside B)的核磁波谱数据相似,表明三者结构十分相近。通过异核单量子(HSQC)谱,对上述碳氢结合信号进行了归属。在异核多键相关(HMBC)谱图显示糖基的端基氢δ 4.32 (H-1”)与δ 68.8 (C-3)存在远程相关信号;δ 4.06 (H-3)和3.83 (H-3)与δ 64.0 (C-1)、72.1 (C-2)和104.7 (C-1”)均存在远程相关信号;δ 5.34 (H-2) 与δ 68.8 (C-3)和172.2 (COCH3)均存在远程相关信号;δ 4.52 (H-1)和4.36 (H-1)与δ 68.8 (C-3)、72.1 (C-2)和168.8 (C-9’)均存在远程相关信号;δ 2.09 (CH3CO)与δ 172.2 (COCH3)存在远程相关信号(见图2),从而进一步确定了样品结构。与王百合苷A相比,样品结构为王百合苷A的2位羟基乙酰化产物。2-乙酰王百合苷A与王百合苷B为同分异构体,主要结构差异在于乙酰基与葡萄糖基的取代位置不同。
百合属植物中富含苯丙素甘油糖苷类成分,由于最初从Lilium regale(岷江百合,又名王百合)中分离得到,因此俗称为王百合苷(regalosides),其结构通常在糖基化甘油骨架的C1或C2位发生酯化,且2S-构型最常见,但2R-构型不太常见[16-20]。鉴于此,初步推论2-乙酰王百合苷A结构中C2位构型与王百合苷A、B一致,为(2S)-1-O-对香豆酰基-2-O-乙酰基-3-O-β-D-吡喃葡萄糖基甘油酯苷。
按照2020年版《中华人民共和国药典》四部高效液相色谱法(通则0512),采用Agilent ZORBAX SB-C18 (250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)(0~20 min,18% A;20~30 min,18% A→90% A;30~35 min,90% A→18% A;35~45 min,18% A)为流动相梯度洗脱,流速1 mL·min-1,检测波长312 nm,样品以甲醇为溶剂,制成质量浓度为2 mg·mL-1的供试品溶液,进样分析,记录色谱图至主峰保留时间的3倍。在上述色谱条件下,进样20 μg,除主峰外,可见极少量杂质峰检出,归一化纯度为99.66%,1%自身对照纯度为99.69%。均匀性初检归一化纯度(n=3)分别为99.66%、99.65%、99.62%,RSD为0.021%。
采用库仑法卡氏水分仪测定样品中水分,结果分别为0.155 8%、0.142 4%、0.140 1%、0.158 9%、0.139 5%,均值以0.15%(n=5)计,RSD=6.3%。
N,N-二甲基甲酰胺溶解样品,制备质量浓度为4 mg·mL-1的供试品溶液;以无水甲醇为对照。色谱条件:按照2020年版《中华人民共和国药典》四部气相色谱法(通则0521),采用Thermo TG-624毛细管柱(30 m×0.32 mm,1.80 μm)色谱柱,检测器为FID,高纯氮为载气,程序升温(起始温度40 ℃,终止温度为220 ℃,升温速率为10 ℃·min-1),进样体积为1 μL。在上述色谱条件下,取供试品溶液进样分析,外标法计算样品中残留溶剂甲醇含量分别为0.183 2%、0.183 2%、0.176 4%、0.176 5%,均值以0.18%(n=4)计,RSD为2.4%。
按照2020年版《中华人民共和国药典》四部炽灼残渣检查法(通则0841),采用铂金坩埚,于600 ℃炽灼,测得样品炽灼残渣为0.05%。
按照质量平衡法计算公式:
含量=自身对照纯度×(100%-水分-残留溶剂-炽灼残渣),计算2-乙酰王百合苷A含量为99.69%×(100%-0.15%-0.18%-0.05%)=99.31%。
采用90°脉冲,在恒温(25 ℃)下获取1H核磁共振谱[具体试验参数设置:采集(AQ)时间2.75 s,弛豫时间(D1)30 s,扫描次数(NS)32次],结果如图3所示。以1,4-二硝基苯为内标,以样品氢信号(δ 6.79)与内标氢信号(δ 8.45),按照公式
计算核磁共振波谱定量法计算结果为100.06%(n=3),RSD为0.47%。公式中 As为2-乙酰王百合苷A的信号面积,ns为2-乙酰王百合苷A参与含量计算的氢原子数(ns=2),Ms为2-乙酰王百合苷A的分子量(Ms=442.41 g·mol-1),Ar为内标1,4-二硝基苯的信号面积,nr为内标中包含的氢原子数(nr=4),Mr为内标物质的分子量(Mr=168.11 g·mol-1),ms为称取2-乙酰王百合苷A的质量,mr为称取的内标物质的质量,Pr为内标的质量百分含量(Pr=99.0%)。
随机抽取12瓶样品,每瓶称样2次,每次称样量约10 mg,用甲醇溶解制成每1 mL含1 mg的溶液,按照“2.2”项纯度分析条件进样,以峰面积/称样量进行方差分析,结果F值为0.45,<F(11,12)(2.72),表明样品均匀。
设置不同取样时间点0、5、10 d,每个时间点分别取2份样品,每份取样约10 mg,用甲醇溶解制成每1 mL含1 mg的溶液,按照“2.2”项纯度分析条件进样,以峰面积/称样量进行t检验,结果|b1|(3784.0)<t0.951*s(b1)(34 099.6),表明样品在考察期间未观察到不稳定性。
样品经核磁共振波谱、质谱、紫外光谱、红外光谱以及比旋度等技术鉴定其结构为2-乙酰王百合苷A。经高效液相色谱自身对照法分析纯度满足定量用中药化学对照品的要求。经库伦法、气相色谱法、炽灼残渣分别测定样品中水分、残留溶剂以及无机杂质等有关物质。经国际通用的质量平衡法定值,同时采用核磁共振波谱定量法辅助定值,进一步验证定值结果的准确性。经均匀性、稳定性评估结果均符合要求。综合上述结果,鉴定样品为-乙酰王百合苷A,可以作为含量测定用对照品使用、分发。含量以99.3%计。
中药化学对照品通常为已知化合物,本文报道的2-乙酰王百合苷A经Scifinder检索为新化合物,其结构经1H NMR、13C NMR、HSQC、HMBC、HRMS、UV、IR等谱学技术和旋光度解析确定,从源头上保障了首批中药化学对照品的结构准确性。此外,在进行定值研究时,本文除采用国际通用的质量平衡法定值外,进一步采用1H qNMR法加以佐证,2种方法含量测定结果基本吻合,能够更好地保证2-乙酰王百合苷A对照品的赋值准确性。2-乙酰王百合苷A首批对照品的研制标定为相关品种的质量控制提供了强有力的物质基础。
