Objective:

To analyze the difference of chemical constituents in rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province.

Methods:

Ultra-high performance liquid chromatography-quadrupole/electrostatic field orbitrap high resolution mass spectrometry(UPLC-Q-Orbitrap HRMS) was developed to determine the chemical constituents in rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province. The data were analyzed by principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). The structures of chemical markers in rhizomes and leaves of Polygonatum sibiricum were identified based on accurate primary mass spectrometry and secondary mass spectrometry fragment ion,combined with the reference map,software database searching and related literature.

Results:

A total of 45 compounds were identified,there were 26 chemical ingredients with significant differences distinguished by the method of OPLS-DA,including 10 amino acids,6 flavonoids,3 organic acids,2 saccharides,1coumarin and 4 alkaloids.

Conclusion:

The chemical markers of amino acids,organic acids and alkaloids are mainly distributed in rhizomes,and the chemical markers of flavonoids are mainly concentrated in leaves part of the plant. It is suggested that the potential multiple utilization of rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province.

目的:

分析陕产黄精根茎和叶中化学成分的差异。

方法:

采用超高效液相色谱-四级杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)法检测陕产黄精根茎和叶的化学成分,所得数据采用主成分分析(PCA)法与正交偏最小二乘判别分析(OPLS-DA)处理,陕产黄精根茎和叶的差异指标成分,通过精准的一级质谱质荷比和二级质谱碎片离子,同对照品图谱和软件数据库搜索,以及相关文献报道成分进行判别分析。

结果:

共鉴定得到45个成分,通过OPLS-DA,筛选出根茎和叶之间差异性化学成分26个,包括氨基酸类10个,黄酮类6个,有机酸类3个,糖类2个,香豆素类1个,生物碱类4个。

结论:

陕产黄精氨基酸类、有机酸类、生物碱类差异成分主要分布于根茎部,而黄酮类差异成分主要集中于叶部,为陕产黄精资源的综合利用提供了理论依据。

目的:

分析陕产黄精根茎和叶中化学成分的差异。

方法:

采用超高效液相色谱-四级杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)法检测陕产黄精根茎和叶的化学成分,所得数据采用主成分分析(PCA)法与正交偏最小二乘判别分析(OPLS-DA)处理,陕产黄精根茎和叶的差异指标成分,通过精准的一级质谱质荷比和二级质谱碎片离子,同对照品图谱和软件数据库搜索,以及相关文献报道成分进行判别分析。

结果:

共鉴定得到45个成分,通过OPLS-DA,筛选出根茎和叶之间差异性化学成分26个,包括氨基酸类10个,黄酮类6个,有机酸类3个,糖类2个,香豆素类1个,生物碱类4个。

结论:

陕产黄精氨基酸类、有机酸类、生物碱类差异成分主要分布于根茎部,而黄酮类差异成分主要集中于叶部,为陕产黄精资源的综合利用提供了理论依据。

Objective:

To analyze the difference of chemical constituents in rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province.

Methods:

Ultra-high performance liquid chromatography-quadrupole/electrostatic field orbitrap high resolution mass spectrometry(UPLC-Q-Orbitrap HRMS) was developed to determine the chemical constituents in rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province. The data were analyzed by principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). The structures of chemical markers in rhizomes and leaves of Polygonatum sibiricum were identified based on accurate primary mass spectrometry and secondary mass spectrometry fragment ion,combined with the reference map,software database searching and related literature.

Results:

A total of 45 compounds were identified,there were 26 chemical ingredients with significant differences distinguished by the method of OPLS-DA,including 10 amino acids,6 flavonoids,3 organic acids,2 saccharides,1coumarin and 4 alkaloids.

Conclusion:

The chemical markers of amino acids,organic acids and alkaloids are mainly distributed in rhizomes,and the chemical markers of flavonoids are mainly concentrated in leaves part of the plant. It is suggested that the potential multiple utilization of rhizomes and leaves of Polygonatum sibiricum from Shaanxi Province.

黄精为百合科植物滇黄精Polygonatum kingianum Coll. et Hemsl.、多花黄精Polygonatum cyrtonema Hua、黄精Polygonatum sibiricum Red.的干燥根茎[1],是临床常用的补气养阴类中药,始载于《名医别录》[2],被列为上品。黄精味甘,性平,归脾、肺、肾经,具补气养阴、健脾、润肺、益肾的功效[3],是我国大量使用的药食两用品种之一。
黄精早在《抱朴子内篇》[4]就有记载,“服其花胜其实,服其实胜其根,但花难多得”;《本草经集注》[5]记载,“根、叶、花、实皆可饵服”;《食疗本草》[6]记载:“根、叶、花、实,皆可食之”。而在现代,仅以黄精根茎作为药用部位,国内外学者对黄精根茎进行了系统的研究[7-9],而对非药用部位叶、花、实的研究则较少。相应地,目前黄精的资源利用主要是根茎,而其余部分则完全废弃。在陕卫中医药发[2020]24号文件中明确规定,黄精属于45种秦药品种下属的10种区域特色中草药之一,为了进一步促进黄精资源的综合利用,本实验拟比较陕产黄精根茎和非药用部位叶的化学成分差异。
植物代谢组学是采用高通量分析技术,以代谢组群指标分析为基础,从而对中药的代谢成分进行无差别、整体、全面的分析[10]。近几年,植物代谢组学技术广泛应用于中药研究,如不同药用部位的鉴别[11-12],药用植物最佳采收期确定[13-14],不同基原的鉴别[15-16],中药炮制机制研究[17-18],不同产地和生长方式药材的品质差异[19-20]等。本实验基于UPLC-Q-Orbitrap HRMS代谢组学技术对陕产黄精根茎和叶的化学成分进行对比研究,为阐明陕产黄精根茎和叶的成分差异以及进一步的资源利用奠定基础。
Vanquish超高效液相色谱仪(Thermo公司),Q Exactive HFX高分辨质谱仪(Thermo公司),梅特勒MS105/A型十万分之一电子天平(北京明宸中寰科技有限公司),H2-16KR台式高速冷冻离心机(湖南可成仪器设备有限公司),SK-1型快速混匀器(武汉格莱莫检测设备有限公司),FW135型中草药粉碎机(天津市泰斯特仪器有限公司),KQ5200E型超声波清洗器(昆山超声仪器有限公司),真空干燥箱(天津泰斯特仪器有限公司)。
对照品哌啶酸(批号 N25HS202420)、L-精氨酸(批号 H11M10Y82633)、L-苯丙氨酸(批号N26GB169364)、1-氨基环丙烷羧酸(批号 D03GB170196)、DL-焦谷氨酸(批号YM0327JA14)、L-缬氨酸(批号S05J12I135887)、D-别异亮氨酸(批号B13J9D52846)、L-色氨酸(批号M20IB210217)、异牡荆素(批号A29HB193210)、异荭草素(批号J24IB220998)、柠檬酸(批号N07IB231318)、苹果酸(批号 A09IB222542)、原儿茶酸(批号O23HB197783)、棉籽糖(批号 S02F12G139355)、D-阿拉伯糖(批号A10GB144812)、对香豆酰酪胺(批号Y30S11W126554)、异牡荆素-2”-O-葡萄糖苷(批号D09HB203568)、牡荆素2”-O- 葡萄糖苷(批号A29HB193210)、甜菜碱(批号DJ0615YA13)均购自上海源叶生物科技有限公司,纯度均≥98%。色谱纯甲醇、乙腈(安谱公司),色谱纯甲酸、异丙醇(默克公司);纯净水(Milli-Q®HX 7000纯水仪,默克公司)。
黄精药材样品于2021年10月9号采自陕西省汉中市略阳县五龙洞中川坝村黄精GAP种植基地,包括采自基地不同位置的栽培黄精6株。所采集样品经陕西中医药大学药学院胡本祥教授鉴定为百合科植物黄精Polygonatum sibiricum Red.全株。黄精根茎和叶为实验室自制。
在参考文献[21]的基础上改良方法来制备供试品溶液。将采集到的6批黄精新鲜样品分为根茎和叶两部分,将根茎鲜切成片后和叶一起放入50 ℃烘箱烘干,将干品粉碎后过4号筛,分别称取黄精根茎和叶粉末0.1 g,精密称定,分别加入预冷水(将灭菌去离子水在4 ℃冰箱放置过夜后拿出,放冰上30 min即可作为预冷水使用)100 μL,涡旋60 s,加入预冷甲醇-乙腈溶液(1:1)400 μL,涡旋60 s,低温超声(功率1 kW,频率20 kHz)30 min,2次,-20 ℃放置1 h沉淀蛋白,12 000 r·min-1,4 ℃离心20 min,取上清液真空干燥后复溶于200 μL 30%乙腈,涡旋,14 000 r·min-1,4 ℃离心15 min,取上清液,用0.22 μm微孔滤膜滤过,取续滤液,即得。
另取上述供试品溶液各10 μL混合,制备质量控制样本,用于检测仪器的稳定性。
分别精密称取各对照品适量,精密称定,加甲醇溶解制成质量浓度均为1 mg·mL-1的各对照品储备液。再分别取各对照品储备液适量,置于10 mL量瓶中,用甲醇稀释成含各对照品1.0 μg·mL-1的混合对照品溶液,涡旋混合,进样前用0.22 μm微孔滤膜过滤。
色谱柱为Waters HSS T3 (100 mm×2.1 mm,1.8 μm),流动相为0.1%甲酸-水溶液(A)和0.1%甲酸-乙腈-异丙醇(B),梯度洗脱(0~2 min,90%A;2~6 min,90%A→40%A;6~15 min,40%A),流速0.3 mL·min-1,柱温40 ℃,进样室温度4 ℃,进样量2 μL。
离子源为ESI源,正/负离子检测模式,鞘气压力275.8 kPa,辅助气为氮气,压力68.95 kPa;离子喷雾电压3 000~2 800 V;离子传输管温度320 ℃;辅助气温度350 ℃;扫描模式为Full-scan MS2模式,一级扫描范围m/z 70~1 050,二级扫描范围m/z 200~2 000,一级分辨率70 000,二级分辨率17 500,MS/MS模式时碰撞能为60 eV。
原始数据用代谢组学处理软件Progenesis QI(Waters Corporation,Milford,USA)进行基线过滤、保留时间校正、峰对齐、峰识别、积分和归一化等预处理,主要参数设置如下:保留时间范围为0~17 min,m/z 70~1 050,保留时间为0.1 min,m/z允许偏差为5×10-6,最终得到1个保留时间、质荷比和峰强度的数据矩阵。将所得数据导入Simca-p14.0软件进行无监督的主成分分析(PCA),初步观察黄精根茎和叶的聚集情况,再根据t检验(P<0.05),正交偏最小二乘法判别分析(OPLS-DA)模型得到的变量权重值(variable important in projection,VIP≥2),差异倍数(Fold Change,FC≥2或≤0.5),寻找差异化学成分,从整体到部分,更全面分析陕产黄精根茎和叶化学成分的差异。OPLS-DA模型通过CV-ANOVA验证,P<0.05说明该模型预测性能强,差异具有统计学意义。
具有代表性的黄精根茎和叶的UPLC-Q-Exactive HFX基峰离子流图如图12所示,直观显示其根茎和叶的化学成分之间存在明显差异。将对照品图谱、数据库搜索(HMDB、Mass Bank、Thermo mzcloud、PubChem)和相关文献报道的化合物,同检测到的化合物的保留时间、相对分子质量、碎片离子等信息进行对照,共鉴定出45个代谢物,其中包括氨基酸类14个,黄酮类11个,有机酸类9个,糖类3个,香豆素类1个,皂苷类2个,生物碱类5个。结果见表1
图12表明陕产黄精根茎和叶的化学成分存在差异,为确定黄精不同部位的化学成分差异,将质谱的原始数据导入Progenesis QI软件进行保留时间校正、峰对齐、峰识别、积分和归一化等预处理,将处理所得数据集矩阵导入Simca-p 14.0软件中,使用PCA对数据进行处理,建立黄精根茎和叶的PCA得分图,并进行整体趋势观察分析,结果见图3
图3可以看出,陕产黄精的根茎和叶被t[1]轴区分明显,根茎位于正半轴,叶位于负半轴。叶有2个样品位于第三象限,4个样品位于第四象限;根茎有3个样品位于第一象限,2个样品位于第二象限,1个样品位于坐标轴上。从图3可知,陕产黄精的根茎和叶得到了明显的区分,表明陕产黄精不同部位所含化学成分差异显著,本实验所建立方法可以很好地表征根茎和叶的成分差异。
选择OPLS-DA模型进行分析是因其能够增强在PCA模型中观察到的根茎和叶的分离情况,因为OPLS-DA模型在为样品组间的差异提供最相关变量方面具有更大的潜力。采用OPLS-DA进行比较,OPLS-DA模型验证结果表明,其未过度拟合(R2X=0.973,R2Y=1,Q2=1,P<0.05),质量良好。其OPLS-DA得分图如图4-A所示,可看出陕产黄精根茎和叶更明显区分开来。S-plot图能够通过对变量的量级与其可靠性进行比较来预测差异成分,主要用于从组学数据中筛选出指定的代谢物。S-plot图中的每1个点代表1个变量,在S-plot(图4-B)图中位于“S”两端的变量属于关键变量。
根据t检验(P<0.05)、变量权重值VIP≥2,结合差异倍数FC≥2或≤0.5进行筛选,结合已鉴定成分,明确黄精根茎和叶的26个差异指标成分,见表2。质谱图积分面积可反映成分的相对含量,将26个差异指标成分进行手动积分,纵坐标为峰面积,绘制出陕产黄精根茎和叶代谢差异成分的相对含量图(图5),不同部位化学成分的量之间差异显著。根据对差异成分的分析可知,氨基酸类、有机酸类、生物碱类差异成分在根茎中含量高,黄酮类差异成分在叶中含量更高。
陕产黄精是2020年版《中国药典》规定的我国传统中药黄精(Polygonati Rhizoma)的3种基源植物黄精(Polygonatum sibiricum Red.)之一,研究表明,黄精主要含有多糖、皂苷、黄酮等多种化学成分,不同的化学成分作用也具有差异性。如黄精中多糖类成分具有提高免疫力、抗氧化、降血糖、抗骨质疏松等作用[22-25],黄酮类成分具有降血糖、抗疲劳、抗炎、抗氧化等作用[2426-28]
本实验采用基于UPLC-Q-Orbitrap HRMS的代谢组学技术,对陕产黄精根茎和叶化学成分进行了比较研究,从根茎和叶中分别指认化学成分45个,包括氨基酸类成分14个,黄酮类成分11个,有机酸类成分9个,糖类成分3个,香豆素类成分1个,皂苷类成分2个,生物碱类成分5个。t检验、多元统计结合FC值分析结果显示,黄精的根茎和叶虽然含有相似的化学成分,但氨基酸、有机酸等初级代谢产物以及黄酮、皂苷、生物碱等次级代谢产物的相对含量存在较大差异。黄精根茎中L-精氨酸、1-氨基环丙烷羧酸、DL-焦谷氨酸、精氨酸、L-色氨酸、对香豆酰酪胺、阿魏酰酪胺、(e)-n-[2-hydroxy-2-(4-hydroxyphenyl)ethyl]-3-(4-hydroxyphenyl)prop-2-enamide、(e)-n-[2-hydroxy-2-(4-hydroxyphenyl)ethyl]-3-(4-hydroxy-3-methoxyphenyl)prop-2-enamide等的含量高于叶中的含量,而黄精叶中异牡荆素、异荭草素、牡荆素2”-O-葡萄糖苷、异牡荆素、异肥皂草苷、异牡荆素-2”-O-葡萄糖苷、原儿茶酸、甜菜碱等的含量高于茎中的含量,而L-缬氨酸、D-别异亮氨酸、柠檬酸、苹果酸、棉籽糖在根茎和叶中的含量无明显差异。同时,黄精不同部位之间作用的差异是否与氨基酸类、黄酮类、有机酸类、皂苷类等的含量有关,值得进一步研究。
文献研究结果显示[22-25],黄精中多糖、皂苷、黄酮等成分与黄精的药效作用密切相关。本实验研究结果表明,氨基酸类、有机酸类、生物碱类差异成分在根茎中含量高,黄酮类差异成分在叶中含量更高。按照中医传统用药经验,黄精入药部位仅是根茎,叶作为非药用部位往往被舍弃。黄精一般生长4年以上,仅采挖根茎而将叶部分扔掉,将造成巨大的资源浪费。基于本研究的结果,可将黄精的叶用于黄酮类成分的提取及相关研究,从而更合理、高效地开发利用黄精资源。
本研究的不足之处在于未对黄精的果实和花所含成分进行系统研究,从而对黄精根茎、叶、花、果实的差异成分进行综合判定。项目组计划下一步将对黄精花和果实的成分进行系统研究,作为对该论文的有益补充。
