Effects of electrochemical deposition time on the phase, morphology, bioactivity of hydroxyapatite (HA) coating and the bonding strength with the substrate were investigated adopting alkali-treatment of titanium alloy followed by electrochemical deposition. Results show that the electrochemically deposited coatings prepared on the surface of alkali-treated titanium alloy are composed of flake HA phases, and the electrochemical deposition time has no effect on the phase composition of the coatings, but will bring impact to the coating thickness. After 60 min deposition, the obtained HA coatings is very compacted and has the bonding strength with the substrate up to 17.85 MPa. It is shown that the HA coatings obtained after each deposition time have good bioactivity. In the simulated body fluids (SBF), CO32- substitutes PO43- in the HA into apatite to form steamed bun-shaped carbonated hydroxyapatite(CHA),with the diameter of 7-8 μm. After being immersed in SBF for 3 days,the CHA can be spread all over the surface of HA.

采用碱预处理钛合金表面再电化学沉积的方法,研究了电化学沉积时间对羟基磷灰石(HA)涂层物相、形貌、生物活性及其与钛合金基体间结合强度的影响。结果表明,碱预处理钛合金表面制备的电化学沉积涂层主要由片状HA相组成,电化学沉积时间对涂层物相组成没有影响,但会影响涂层厚度;沉积时间60 min时得到的HA涂层较致密,与基体的结合强度达17.85 MPa。各沉积时间下得到的HA涂层均具有较好的生物活性,在模拟体液(SBF)中,CO32-置换HA中的PO43-进入磷灰石中形成类骨磷灰石CHA,呈馒头状,直径7~8 μm,且在SBF中浸泡3 d,CHA可铺满HA表面。

采用碱预处理钛合金表面再电化学沉积的方法,研究了电化学沉积时间对羟基磷灰石(HA)涂层物相、形貌、生物活性及其与钛合金基体间结合强度的影响。结果表明,碱预处理钛合金表面制备的电化学沉积涂层主要由片状HA相组成,电化学沉积时间对涂层物相组成没有影响,但会影响涂层厚度;沉积时间60 min时得到的HA涂层较致密,与基体的结合强度达17.85 MPa。各沉积时间下得到的HA涂层均具有较好的生物活性,在模拟体液(SBF)中,CO32-置换HA中的PO43-进入磷灰石中形成类骨磷灰石CHA,呈馒头状,直径7~8 μm,且在SBF中浸泡3 d,CHA可铺满HA表面。

Effects of electrochemical deposition time on the phase, morphology, bioactivity of hydroxyapatite (HA) coating and the bonding strength with the substrate were investigated adopting alkali-treatment of titanium alloy followed by electrochemical deposition. Results show that the electrochemically deposited coatings prepared on the surface of alkali-treated titanium alloy are composed of flake HA phases, and the electrochemical deposition time has no effect on the phase composition of the coatings, but will bring impact to the coating thickness. After 60 min deposition, the obtained HA coatings is very compacted and has the bonding strength with the substrate up to 17.85 MPa. It is shown that the HA coatings obtained after each deposition time have good bioactivity. In the simulated body fluids (SBF), CO32- substitutes PO43- in the HA into apatite to form steamed bun-shaped carbonated hydroxyapatite(CHA),with the diameter of 7-8 μm. After being immersed in SBF for 3 days,the CHA can be spread all over the surface of HA.

钛合金(Ti6Al4V)是目前人体植入物的常用材料之一[1-2],但钛合金是生物惰性材料,植入后无法像生物活性材料一样与人体骨骼形成良好的化学结合,因此,需要对钛合金进行改性处理,提高其表面生物活性。目前钛合金表面改性的首选材料为羟基磷灰石(HA)[3],在钛合金表面涂覆一层HA涂层得到复合材料,既保留了钛合金的优良力学性能,又具有HA良好的生物相容性[4]
电化学沉积是一种常用的在钛合金表面制备HA涂层的方法,其原理是在含有Ca2+和H2PO4-等离子的电解液中,通过调控电压、电流密度、电解液浓度、电解液pH值、沉积温度及沉积时间等工艺参数,使电解液在电场的作用下产生大量OH-,阴极附近pH值增大,磷酸钙盐的过饱和度迅速上升,从而在阴极金属表面得到磷酸钙盐涂层[5]。然而,在钛合金表面直接涂覆HA涂层会出现结合强度较低(约4.5 MPa)的问题[6],为了改善涂层质量、提高涂层和钛合金基体的结合强度,通常需要对基体表面先进行预处理,再进行电化学沉积。
本文采用先碱预处理Ti6Al4V试样、再电化学沉积的方法制备HA涂层,研究电化学沉积时间对HA涂层物相、形貌、生物活性以及HA涂层与基体间结合强度的影响。
采用线切割法将Ti6Al4V板材切成25 mm×25 mm×3 mm的正方形薄片并在边缘打孔用于系绳,然后置于HF∶HNO3∶H2O=1∶3∶10(体积比)的溶液中腐蚀3~5 min,至表面光亮。将处理好的钛合金片依次用P200、P400、P600、P800水砂纸打磨;然后将钛合金试样依次放入丙酮、无水乙醇、去离子水中超声清洗10 min,再将试样放入60 ℃干燥箱中干燥。最后,将试样置于8 mol/L的氢氧化钠溶液中于60 ℃浸泡48 h,并用去离子水冲洗试样30 s,随后将试样置于37 ℃恒温干燥箱中备用。试验试剂均为国药集团分析纯试剂。
电化学沉积是通过电场作用,使电解液中的正、负离子发生定向迁移,并与电极发生氧化还原反应,在电解液与电极的接触面形成涂层的方法。将0.042 mol/L Ca(NO32·4H2O、0.025 mol/L NH4H2PO4、1.5×10-3 mol/L EDTA-2Na溶于去离子水,制得所需电解液。使用DP3020型电源为电化学沉积设备,以经过碱处理的钛片为阴极、铂片为阳极,采用恒流模式,在温度90 ℃,电流密度2.50 mA/cm2,沉积时间分别为15 min、30 min、60 min、90 min条件下制备涂层。
模拟体液(SBF)配制[1]:将称量好的NaCl、NaHCO3、KCl、K2HPO4·3H2O、MgCl2·6H2O、1 mol/L HCl、CaCl2、Na2SO4及(CH2OH)3CNH2按顺序逐一加入去离子水中,然后加入适量1 mol/L HCl调节溶液pH值至7.42,制得SBF。
将电化学沉积试样放入配置好的SBF中,温度设为36.5 ℃,每天更换一次SBF,3 d后取样,研究HA涂层经SBF浸泡后的生物活性。
采用FEI Guanta FEG450型场发射扫描电子显微镜(SEM)表征碱预处理后试样表面微观形貌;采用D8 ADVANC型X射线衍射仪(XRD)分析试样物相;采用KBr压片法,使用VERTEX70v型傅里叶红外光谱仪分析HA样品红外光谱(FTIR);采用Senterra R200-L型色散型激光拉曼显微镜光谱仪表征涂层化学组分;根据GB 23101.4—2008[7]测试HA涂层与基体之间的结合强度,采用E7胶将涂层与基体黏接,试验设备为Z050TH型万能材料试验机,拉伸速率1 mm/min。
不同电化学沉积时间下所得涂层的XRD图谱见图1。由图1可知,所有涂层含有HA(Ca10(PO46(OH)2)和Ti两种物相,说明通过电化学沉积法在碱预处理钛合金表面制备得到了HA涂层。电化学沉积15~90 min得到的涂层衍射峰位置与HA标准卡片衍射峰位置基本吻合。电化学沉积时间15 min时,HA相初步生成,Ti的特征衍射峰较强,HA相的衍射峰较弱,同时还存在部分Ca8H2(PO4)·5H2O相;HA相衍射峰显著宽化,最强峰为位于2θ=25.9°处的(002)晶面衍射峰,而不是HA标准衍射卡片中2θ=31.74°处的(211)晶面衍射峰,说明部分HA晶粒沿c轴方向择优生长[8],且尺寸较小,结晶度较低[9]。随着电化学沉积时间延长,中间相Ca8H2(PO4)·5H2O[10]衍射峰强度逐渐降低,HA相衍射峰强度逐渐增大、峰宽变窄,Ti衍射峰强度逐渐降低,沉积时间60 min时,HA相衍射峰最强,Ti衍射峰较弱,Ca8H2(PO4)·5H2O衍射峰基本消失。沉积时间90 min时,HA相和Ti衍射峰均无明显变化。随着电化学沉积时间变化,谱峰强度有所变化,说明沉积量有所变化[8]。沉积时间15 min时,由于HA涂层很薄,Ti衍射峰很明显;沉积时间60 min时,Ti衍射峰很弱,说明此时得到的HA涂层已具有一定厚度;继续延长沉积时间,涂层厚度不再增加,Ti衍射峰强度变化已不明显。在此过程中,(002)晶面衍射峰一直是最强峰,表明在电化学沉积过程中HA沿c轴方向择优生长的趋势一直存在。
不同电化学沉积时间下HA涂层的红外光谱(FTIR)如图2所示,拉曼(Raman)光谱见图3。谱图中3 445 cm-1和1 650 cm-1处分别为水的伸缩和弯曲振动峰,562 cm-1、603 cm-1、1 061 cm-1处出现了PO43-的振动吸收峰,可认为是HA中羟基和磷酸根的振动造成的。随着沉积时间延长,红外谱峰和拉曼谱峰均有增强的趋势,证明随着沉积时间延长,HA沉积量增加[8],但涂层物相组成并不发生变化,这与XRD分析结果一致。沉积时间过长时,HA涂层靠近内层的晶体与周围同层晶粒及外层晶粒竞争生长,靠近外表面的晶体因溶液中钙离子及磷酸根基团的消耗使溶液饱和度下降,成核受到抑制[11],此时HA生长速度明显变慢。FTIR和Raman谱图中PO43-的振动吸收峰没有明显的分裂现象,说明得到的HA结晶度不高[12-14]
综合以上分析,电化学沉积时间60 min时,涂层物相组成和厚度基本稳定。
图4为不同电化学沉积时间下HA涂层的SEM微观形貌。沉积时间15~60 min时,HA以片状为主;沉积时间90 min时,HA形态主要为针状。与针状HA相比,片状HA具有更致密的排列结构,且层片间通常以一定的取向关系相互配合,可有效降低单位面积界面能,具有更高的结合强度[15]。沉积时间较短时,得到的HA层片较小;随着沉积时间延长,片尺寸和厚度增加,表明HA涂层的形成包括形核和长大两个方面。沉积60 min时,HA涂层片均匀致密,沉积效果好。
电化学沉积HA涂层得到复合材料的最终目的是植入人体,这就要求涂层与基体间具有一定的结合强度。HA涂层断裂后断面的XRD图谱如图5所示。可以看到,拉伸后样品断口处存在HA和Ti的衍射峰。与拉伸前样品相比,HA衍射峰强度减弱而Ti衍射峰强度明显增强,由此可以断定断裂发生在涂层内部(HA涂层内部的结合强度较涂层与基体间的结合强度弱),故拉伸试验得到的结合强度值可以很好地表征HA-Ti复合材料的结合强度。
图6为不同电化学沉积时间下HA涂层与基体间的结合强度。随着电化学沉积时间延长,涂层与基体间的结合强度先增加后减小,电化学沉积60 min时结合强度达17.85 MPa,是钛合金表面未处理涂层结合强度(4.5 MPa)的3.97倍[6],且明显高于等离子喷涂单一涂层的结合强度值(13.8 MPa)[16]。其机理如下:钛合金在一定浓度氢氧化钠溶液中浸泡一定时间后,其表面润湿性增加[17-18],即经碱预处理的试样表面具有较好的亲水性,可促进HA形核;经氢氧化钠预处理的钛合金表面呈三维网状结构[19]。电化学沉积过程中,HA可在基体孔洞中形核、生长,并嵌入基体中,使得HA涂层与基体间具有较高的结合强度。
随着电化学沉积时间延长,HA涂层厚度增加,涂层与基体间结合强度增加。但电化学沉积过程中会发生析氢反应,氢气泡在溶液中会经历形核、长大、最终脱附的过程,气泡逸出时对涂层具有冲刷作用,没有逸出的气泡会在涂层内部形成空穴,影响涂层质量,所以气泡越大,对涂层造成的缺陷越多。随着电化学沉积时间进一步延长,大量氢气泡在电极表面堆积,涂层变得十分疏松,疏松的涂层干燥后更容易形成裂纹,导致涂层与基体间结合强度降低。
为了在人体内环境中诱导生成类骨磷灰石(CHA)及成骨细胞,钛合金表面沉积HA涂层所得到的生物复合材料需具有一定生物活性。具有良好生物活性的材料表面能很快诱导生成类骨磷灰石(CHA)及成骨细胞。采用SBF浸泡试验,以在SBF中诱导形成CHA的速度[20]来衡量HA的生物活性。不同电化学沉积时间下HA涂层在SBF中浸泡3 d后的表面形貌如图7所示,电化学沉积60 min所得HA涂层经SBF浸泡后的FTIR结果如图8所示。经SBF浸泡后,HA涂层上均出现馒头状颗粒,有研究认为这种颗粒状物质为通过SBF诱导生成的CHA[21]。经SBF浸泡后,在871 cm-1和1 400~1 450 cm-1处出现了CO32-的特征吸收峰,其他位置为PO43-及OH-的特征吸收峰;1 400~1 450 cm-1处的CO32-的特征吸收峰裂解为1 420 cm-1和1 463 cm-1两个吸收峰,以区别于碳酸盐中的CO32-单峰或自由基CO32-,这是CO32-置换HA中的PO43-进入磷灰石结构的重要标志[22]。生物体骨磷灰石中也有CO32-存在,人体骨骼中CO32-含量为2.8%~8.0%[23],由此可知,通过SBF诱导生成了与人骨成分更加接近的磷灰石。
综合SEM和FTIR分析可知,SBF浸泡3 d后HA涂层诱导生成了CHA,说明本试验得到的HA涂层能较快地诱导CHA形成,即具有较好的生物活性。电化学沉积时间15~60 min时,随着电化学沉积时间延长,SBF诱导生成的CHA尺寸呈长大趋势,电化学沉积60 min后经SBF浸泡得到的CHA直径为7~8 μm;电化学沉积进一步延长至90 min时,涂层表面趋于平整,并出现疏松的颗粒和较大的裂纹,会降低与骨组织的结合。因此,电化学沉积时间60 min时CHA生长效果较好。
1)在钛合金表面通过先碱预处理、后电化学沉积的方法可以制备得到HA涂层。电化学沉积时间对涂层物相组成没有影响,但会影响涂层厚度。
2)沉积时间60 min时可得到片状HA,其与基体的结合强度达17.85 MPa,是未预处理钛合金表面HA涂层结合强度的3.97倍。
3)各沉积时间下得到的HA涂层均具有较好的生物活性,在SBF中CO32-置换HA中的PO43-进入磷灰石中形成类骨磷灰石CHA,呈馒头状,直径7~8 μm,且在SBF中浸泡3 d后CHA即可铺满HA表面。
