OBJECTIVE To analyze the impurity profile of irbesartan and its preparations by LC-MS/MS. METHODS ZORBAX SB-C18 column (4.6 mm×150 mm, 3.5 μm) was used for the separation of the related substances with a mixture of 0.1% formic acid solution (adjusted to pH 3.5 with ammonia) and acetonitrile (62∶38) as the mobile phase by isocratic elution. The structures of the related substances were speculated by ESI-TOF-MS/MS and verified further by reference substances. RESULTS Irbesartan and its related substances were separated under the established chromatographic condition, and a total of 16 related substances were detected. The structures of 10 related substances were verified by reference substances. CONCLUSION The established LC-MS/MS method is effective for separation and identification of the related substances of irbesartan and its preparations, and the results obtained are valuable for its quality control and manufacturing process.

目的 采用液相色谱-串联质谱联用技术(LC-MS/MS)分析厄贝沙坦及其制剂的杂质谱。方法 采用色谱柱ZORBAX SB-C18(4.6 mm×150 mm,3.5 μm),以体积分数0.1%甲酸溶液(用氨水调节pH值至3.5)-乙腈(体积比62∶38)为流动相,等度洗脱,对厄贝沙坦及其制剂中有关物质进行分离,采用电喷雾-飞行时间质谱法(ESI-TOF-MS/MS)测定各有关物质分子结构信息,并结合对照品质谱结构信息确证有关物质结构。结果 厄贝沙坦与其有关物质在该色谱条件下分离良好,共检出16个有关物质,通过对照品确证其中10个有关物质的结构。结论 本实验建立的LC-MS/MS方法能够有效分离鉴定厄贝沙坦及其制剂中有关物质,为其质量控制和工艺优化提供参考依据。

目的 采用液相色谱-串联质谱联用技术(LC-MS/MS)分析厄贝沙坦及其制剂的杂质谱。方法 采用色谱柱ZORBAX SB-C18(4.6 mm×150 mm,3.5 μm),以体积分数0.1%甲酸溶液(用氨水调节pH值至3.5)-乙腈(体积比62∶38)为流动相,等度洗脱,对厄贝沙坦及其制剂中有关物质进行分离,采用电喷雾-飞行时间质谱法(ESI-TOF-MS/MS)测定各有关物质分子结构信息,并结合对照品质谱结构信息确证有关物质结构。结果 厄贝沙坦与其有关物质在该色谱条件下分离良好,共检出16个有关物质,通过对照品确证其中10个有关物质的结构。结论 本实验建立的LC-MS/MS方法能够有效分离鉴定厄贝沙坦及其制剂中有关物质,为其质量控制和工艺优化提供参考依据。

OBJECTIVE To analyze the impurity profile of irbesartan and its preparations by LC-MS/MS. METHODS ZORBAX SB-C18 column (4.6 mm×150 mm, 3.5 μm) was used for the separation of the related substances with a mixture of 0.1% formic acid solution (adjusted to pH 3.5 with ammonia) and acetonitrile (62∶38) as the mobile phase by isocratic elution. The structures of the related substances were speculated by ESI-TOF-MS/MS and verified further by reference substances. RESULTS Irbesartan and its related substances were separated under the established chromatographic condition, and a total of 16 related substances were detected. The structures of 10 related substances were verified by reference substances. CONCLUSION The established LC-MS/MS method is effective for separation and identification of the related substances of irbesartan and its preparations, and the results obtained are valuable for its quality control and manufacturing process.

厄贝沙坦(irbesartan),化学名为2-丁基-3-[4-[2-(1H-四氮唑-5-基)苯基]苯甲基]-1,3-二氮杂螺-[4,4]壬-1-烯-4-酮,分子式为C25H28N6O,结构见图1。厄贝沙坦作为一种血管紧张素Ⅱ受体拮抗剂,临床主要用于原发性高血压以及合并高血压的2型糖尿病肾病的治疗,已上市产品有片剂、分散片、胶囊剂等剂型。此外,还可与氢氯噻嗪制成的复方片剂用于单药未能控制的高血压患者的治疗,两者协同降压,改善患者用药依从性[1]
2020年版《中国药典》及《欧洲药典》10.0版等厄贝沙坦标准中有关物质项下仅收载了已知杂质Ⅰ(impurity Ⅰ),结构见图2。本研究建立了适用于厄贝沙坦及其制剂中有关物质检测的液相色谱-串联质谱联用技术(LC-MS/MS)方法,通过对厄贝沙坦及其制剂在强制破坏和加速稳定性试验中的有关物质进行分离和结构鉴定,共检出包括已知杂质Ⅰ在内的16个杂质,结构见图2。通过购买对照品及合成对照品,对其中的10个杂质的结构进行了确证,再结合生产工艺对各有关物质的结构和来源进行系统分析,为厄贝沙坦原料药及其制剂的产品质量控制和生产工艺优化提供参考依据。
SCIEX ExionLC高效液相色谱仪、SCIEX Triple TOF 5600质谱仪、Analyst数据分析软件(美国AB SCIEX公司); XSE-205DU十万分之一电子天平、pH计(美国Mettler公司)。
厄贝沙坦对照品(批号100607-201804,含量100%)、杂质Ⅰ对照品(批号101312-201902,含量100%)(中国食品药品检定研究院);杂质Ⅲ对照品(批号2208-072A7,含量99%)、杂质Ⅶ对照品(批号3069-034A4,含量99%)、杂质Ⅷ对照品(批号2963-061A1,含量95%)(TLC);杂质ⅩⅣ对照品(批号05S-TBK-71-1,含量97%)、杂质ⅩⅥ对照品(批号05S-PAM-99-7,含量95%)(PANPHY);杂质Ⅳ对照品(批号20220916,含量98%)、杂质Ⅴ对照品(批号20220912,含量98%)、杂质Ⅸ对照品(批号20220908-1,含量98%)、杂质Ⅺ对照品(批号20220908-2,含量98%)均来源于自制;甲醇和乙腈均为色谱纯;其他试剂均为分析纯;超纯水由Millipore超纯水系统制备。
本研究实验样品均来自于2022年国家药品抽检项目,其中厄贝沙坦原料药来自于7家生产企业,厄贝沙坦制剂(厄贝沙坦片、厄贝沙坦分散片、厄贝沙坦胶囊)来自于20家生产企业。
采用ZORBAX SB-C18(4.6 mm×150 mm,3.5 μm)色谱柱,以体积分数0.1%甲酸溶液(用氨水调节pH值至3.5)-乙腈(62∶38)为流动相,等度洗脱,流速为1.0 mL·min-1,柱温30 ℃,检测波长为220 nm,进样体积为10 μL。
采用电喷雾正离子化模式(ESI+),离子源温度(TEM)650 ℃,气帘气流速(CUR)206.84 kPa,雾化气流(GS1)344.74 kPa,辅助加热气(GS2)344.74 kPa,离子源喷雾电压(ISVF)5 500 V,去簇电压(DP)80 V,碰撞电压(CE)10 V,扫描范围(m/z)100~800;二级质谱去簇电压(DP)100 V,碰撞电压(CE)35 V,碰撞电压扩展(CES)15 V,扫描范围(m/z)100~800。
取厄贝沙坦原料药和制剂适量(约相当于厄贝沙坦100 mg),精密称定,置100 mL量瓶中,加甲醇使厄贝沙坦溶解并稀释制成每1 mL中约含厄贝沙坦1 mg的溶液,滤过,取续滤液。
分别取厄贝沙坦对照品和厄贝沙坦各杂质对照品适量,精密称定,加甲醇溶解并稀释制成每1 mL中各约含10 μg的溶液。
称取供试品适量(约相当于厄贝沙坦100 mg),加5 mL 1 mol·L-1的氢氧化钠溶液,放置2 h,加5 mL 1 mol·L-1的盐酸溶液中和,用甲醇稀释并定容至100 mL,混匀,滤过,取续滤液。
厄贝沙坦分子式为C25H28N6O,理论精确相对分子质量为428.232 5,正离子模式下[M+H]+峰理论精确相对分子质量为429.239 7。参考文献[2]推测其裂解规律见图3,二级质谱主要理论裂解碎片包括[C25H29N4O]+(m/z)401.233 6、[C25H28N3O]+(m/z)386.222 7、[C14H11N4]+(m/z)235.097 8、[C14H11N2]+(m/z)207.091 7、[C11H19N2O]+(m/z)195.149 2、[C13H10N]+(m/z)180.080 8、[C13H8N]+(m/z)178.065 1。厄贝沙坦对照品溶液LC-MS/MS典型图谱见图4,一级质谱和二级质谱实验结果与理论值匹配一致。
厄贝沙坦杂质Ⅰ分子式为C25H30N6O2,理论精确相对分子质量为446.243 0,正离子模式下[M+H]+峰理论精确相对分子质量为447.250 3。推测其裂解规律[3]图3,二级质谱主要理论裂解碎片包括[C25H31N4O2]+(m/z)419.244 2、[C20H23N6O]+(m/z)363.192 8、[C16H15N6]+(m/z)291.135 3、[C14H14N5]+(m/z)252.124 4、[C14H11N4]+(m/z)235.097 8、[C14H11N2]+(m/z)207.091 7、[C11H18NO2]+(m/z)196.133 2、[C14H10N]+(m/z)192.080 8、[C13H10N]+(m/z)180.080 8、[C10H18NO]+(m/z)168.138 3。厄贝沙坦杂质Ⅰ对照品溶液LC-MS/MS典型图谱见图5,一级质谱和二级质谱实验结果与理论值匹配一致。
研究结果[4]表明,厄贝沙坦原料药及其制剂在高温、强酸和强氧化条件下较稳定,主要在强碱性条件下易发生降解反应。厄贝沙坦在碱性条件下主要降解为保留时间约7.7 min的已知杂质Ⅰ,与杂质Ⅰ对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。此外,还可降解为保留时间约3.1 min的杂质Ⅱ和保留时间约5.4 min的杂质Ⅳ,杂质Ⅳ通过对照品进行了结构确证,LC-MS典型总离子流色谱图见图6
厄贝沙坦杂质Ⅱ一级质谱精确相对分子质量为447.250 3,软件分析匹配分子式为C25H30N6O2,与已知杂质Ⅰ为同分异构体,理论精确相对分子质量为446.243 0,正离子模式下[M+H]+峰理论精确相对分子质量为447.250 3。通过二级质谱主要裂解碎片[C11H21N2O2]+(m/z)213.159 6、[C14H11N2]+(m/z)207.091 5、[C14H11]+(m/z)179.085 1,推测其结构与杂质Ⅰ结构类似,同为厄贝沙坦的二氮杂螺开环产物[4-6],LC-MS/MS质谱图和裂解规律见图7
厄贝沙坦杂质Ⅳ一级质谱精确相对分子质量为336.182 2,软件分析匹配分子式为C19H21N5O,理论精确相对分子质量为335.174 6,正离子模式下[M+H]+峰理论精确相对分子质量为336.181 9。通过二级质谱主要裂解碎片[C14H14N5]+(m/z)252.124 8、[C14H11N4]+(m/z)235.097 7、[C14H11N2]+(m/z)207.091 6、[C13H10N]+(m/z)180.080 5,推测其结构与文献[7-8]报道碱降解杂质结构一致,LC-MS/MS质谱图和裂解规律见图8。杂质Ⅳ与经核磁确证结构的合成对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
厄贝沙坦原料药中共检出9种杂质(含量≥0.001%),a~g共7家原料药生产企业厄贝沙坦杂质种类基本一致,典型色谱图见图9,杂质含量有差异,加速试验(温度40 ℃、相对湿度75%)2个月各杂质含量见表1。除已知杂质Ⅰ以外,原料药供试品溶液还检出杂质Ⅴ、杂质Ⅷ、杂质Ⅹ、杂质Ⅻ、杂质ⅩⅢ、杂质ⅩⅣ、杂质ⅩⅤ和杂质ⅩⅥ。其中,杂质Ⅴ、杂质Ⅷ、杂质ⅩⅣ和杂质ⅩⅥ通过对照品进行了结构确证。
厄贝沙坦杂质Ⅴ提取离子流图色谱峰保留时间约6.2 min,一级质谱精确相对分子质量为459.250 4,软件分析匹配分子式为C26H30N6O2,理论精确相对分子质量为458.243 0,正离子模式下[M+H]+峰理论精确相对分子质量为459.250 3。通过二级质谱主要裂解碎片[C12H21N2O2]+(m/z)225.159 4、[C14H11N2]+(m/z)207.091 2、[C13H10N]+(m/z)180.080 1,推测其结构为厄贝沙坦与甲醛反应生成的羟甲基化产物[9-11],LC-MS/MS质谱图和裂解规律见图10。杂质Ⅴ与经核磁确证结构的合成对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
杂质Ⅷ提取离子流图色谱峰保留时间约7.3 min,一级质谱精确相对分子质量为415.224 1,软件分析匹配分子式为C24H26N6O,理论精确相对分子质量为414.216 8,正离子模式下[M+H]+峰理论精确相对分子质量为415.224 1。通过二级质谱主要裂解碎片[C24H26N3O]+(m/z)372.206 1、[C14H11N2]+(m/z)207.091 5、[C10H17N2O]+(m/z)181.133 5,推测其为文献[12-13]报道的厄贝沙坦脱去端甲基结构,LC-MS/MS质谱图和裂解规律见图11。杂质Ⅷ与对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
厄贝沙坦杂质Ⅹ提取离子流图色谱峰保留时间约8.5 min,一级质谱精确相对分子质量为427.224 2,软件分析匹配分子式为C25H26N6O,与厄贝沙坦杂质ⅩⅢ为同分异构体,理论精确相对分子质量为426.216 8,正离子模式下[M+H]+峰理论精确相对分子质量为427.224 1。通过二级质谱主要裂解碎片[C25H26N3O]+(m/z)384.207 5、[C14H11N2]+(m/z)207.091 8、[C11H17N2O]+(m/z)193.133 8,参考文献[9]推测其为厄贝沙坦环戊烷基脱氢成双键结构,LC-MS/MS质谱图和裂解规律见图12
厄贝沙坦杂质Ⅻ提取离子流图色谱峰保留时间约9.5 min,一级质谱精确相对分子质量为431.255 4,软件分析匹配分子式为C25H30N6O2,理论精确相对分子质量为430.248 1,正离子模式下[M+H]+峰理论精确相对分子质量为431.255 4。通过二级质谱主要裂解碎片[C19H22N5]+(m/z)320.186 8、[C14H11N4]+(m/z)235.097 8、[C14H11N2]+(m/z)207.091 7、[C10H18N]+(m/z)152.143 2,推测其为厄贝沙坦的二氮杂螺环加氢结构,二氮杂螺环羰基可与环戊烷基位置发生异构化,与厄贝沙坦杂质ⅩⅥ为同分异构体,LC-MS/MS质谱图和裂解规律见图13

厄贝沙坦杂质ⅩⅤ提取离子流图色谱峰保留时间约19.6 min,一级质谱精确相对分子质量为443.255 4,软件分析匹配分子式为C26H30N6O,理论精确相对分子质量为442.248 1,正离子模式下[M+H]+峰理论精确相对分子质量为443.255 4。通过二级质谱主要裂解碎片[C26H30N3O]+(m/z)400.238 3、[C14H11N2]+(m/z)207.091 4、[C13H10N]+(m/z)180.080 4,参考文献[9]报道推测其为厄贝沙坦延长端甲基结构,LC-MS/MS质谱图和裂解规律见图16
杂质ⅩⅥ提取离子流图色谱峰保留时间约22.9 min,一级质谱精确相对分子质量为431.255 4,软件分析匹配分子式为C25H30N6O2,与厄贝沙坦杂质ⅩⅡ为同分异构体,理论精确相对分子质量为430.248 1,正离子模式下[M+H]+峰理论精确相对分子质量为431.255 4。通过二级质谱主要裂解碎片包括[C19H20N5]+(m/z)318.172 2、[C19H19N2]+(m/z)275.155 1、[C14H11N4]+(m/z)235.098 2、[C14H11N2]+(m/z)207.091 5,推测其结构为厄贝沙坦杂质ⅩⅡ的二氮杂螺环羰基位置异构,LC-MS/MS质谱图和裂解规律见图17。杂质ⅩⅥ与对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
厄贝沙坦制剂中共检出14种有关物质(≥0.001%),A~T共20家制剂生产企业厄贝沙坦杂质种类和含量有差异,典型色谱图见图18,加速试验(温度40 ℃、相对湿度75%)6个月各杂质含量见表2。除强制降解溶液和原料药中检出各杂质外,制剂供试品溶液中还检出杂质Ⅲ、杂质Ⅵ、杂质Ⅶ、杂质Ⅸ和杂质Ⅺ。其中,杂质Ⅲ、杂质Ⅶ、杂质Ⅸ和杂质Ⅺ通过对照品进行了结构确证。
厄贝沙坦杂质Ⅲ提取离子流图色谱峰保留时间约4.1 min,一级质谱精确相对分子质量为373.177 3,软件分析匹配分子式为C21H20N6O,理论精确相对分子质量为372.169 9,正离子模式下[M+H]+峰理论精确相对分子质量为373.177 1。通过二级质谱主要裂解碎片[C14H11N4]+(m/z)235.098 4、[C14H11N2]+(m/z)207.091 6、[C13H10N]+(m/z)180.080 5,参考文献[9]推测其为厄贝沙坦脱去丁基结构,LC-MS/MS质谱图和裂解规律见图19。杂质Ⅲ与对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
厄贝沙坦杂质Ⅵ提取离子流图色谱峰保留时间约6.7 min,一级质谱精确相对分子质量为445.234 9,软件分析匹配分子式为C25H28N6O2,理论精确相对分子质量为444.227 4,正离子模式下[M+H]+峰理论精确相对分子质量为445.234 7。通过二级质谱主要裂解碎片[C11H19N2O2]+(m/z)211.144 6、[C14H11N2]+(m/z)207.091 5、[C11H19N2O]+(m/z)195.1491、[C13H10N]+(m/z)180.080 8,推测其为厄贝沙坦杂质Ⅷ羟甲基化产物,LC-MS/MS质谱图和裂解规律见图20
厄贝沙坦杂质Ⅶ提取离子流图色谱峰保留时间约7.0 min,一级质谱精确相对分子质量为403.224 2,软件分析匹配分子式为C23H26N6O,理论精确相对分子质量为402.216 8,正离子模式下[M+H]+峰理论精确相对分子质量为403.224 1。通过二级质谱主要裂解碎片[C23H26N3O]+(m/z)360.207 4、[C14H11N4]+(m/z)235.098 2、[C14H11N2]+(m/z)207.091 8、[C13H10N]+(m/z)180.080 7,推测其为厄贝沙坦上环戊烷基开环结构,LC-MS/MS质谱图和裂解规律见图21。杂质Ⅶ与对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。
厄贝沙坦杂质Ⅸ提取离子流图色谱峰保留时间约7.6 min,一级质谱精确相对分子质量为473.265 9,软件分析匹配分子式为C27H32N6O2,与厄贝沙坦杂质Ⅺ为同分异构体,理论精确相对分子质量为472.258 7,正离子模式下[M+H]+峰理论精确相对分子质量为473.266 0。通过二级质谱主要裂解碎片[C25H29N6O]+(m/z)429.239 5、[C13H23N2O2]+(m/z)239.175 7、[C14H11N2]+(m/z)207.092 0、[C11H19N2O]+(m/z)195.149 6,参考文献[10]推测其结构为厄贝沙坦与乙醛反应生成的羟乙基化产物,LC-MS/MS质谱图和裂解规律见图22。杂质Ⅸ与经核磁确证结构的合成对照品溶液的保留时间、一级和二级质谱结果匹配一致,确证结构。

本研究建立了液-质联用色谱技术用于检测厄贝沙坦及其制剂中有关物质的方法,共检测出厄贝沙坦有关物质16个,除杂质Ⅰ结构已知外,其他15个杂质均根据LC-MS/MS数据推测结构。通过合成对照品对杂质Ⅳ、杂质Ⅴ、杂质Ⅸ和杂质Ⅺ的结构进行确证,通过购买对照品对杂质Ⅲ、杂质Ⅶ、杂质Ⅷ、杂质ⅩⅣ和杂质ⅩⅥ的结构进行确证。厄贝沙坦及其制剂中有关物质详细信息见表3
厄贝沙坦制剂的每日最大剂量为0.3 g,参考人用药品技术要求国际协调理事会新原料药中的杂质和新药制剂中的杂质的指导原则(ICH Q3A、ICH Q3B),厄贝沙坦原料药降解产物的鉴定限度为0.1%,厄贝沙坦制剂降解产物的鉴定限度为0.2%。厄贝沙坦原料药及其制剂的加速试验中,上述16个杂质中,仅制剂中已知杂质Ⅰ和杂质Ⅴ含量超过了0.2%,其余杂质含量均低于0.1%。本研究对杂质Ⅴ结构进行了确证,并参考人用药品技术要求国际协调理事会基因毒性杂质指导原则(ICH M7)的要求,采用Derek Nexus和 Sarah Nexus对其遗传毒性(致突变性)进行了预测,结果均为阴性,因此建议规定厄贝沙坦制剂中杂质Ⅴ含量不得过0.2%。现有厄贝沙坦制剂标准中规定杂质Ⅰ含量不得过0.2%,其他单个杂质含量不得过0.2%,现有标准较为合理。
根据稳定性试验结果数据分析,厄贝沙坦在高温高湿条件下杂质Ⅲ、杂质Ⅵ、杂质Ⅶ、杂质Ⅷ、杂质Ⅹ、杂质Ⅻ、杂质ⅩⅢ、杂质ⅩⅣ、杂质ⅩⅤ和杂质ⅩⅥ含量基本保持稳定,推测以上10个杂质均为工艺杂质。其中杂质Ⅲ、杂质Ⅵ、杂质Ⅶ为厄贝沙坦制剂中检出杂质,推测为制剂工艺杂质,与制剂生产过程中的干燥工艺有关。各工艺杂质均保留了厄贝沙坦的主要结构,结构差异均集中在二氮杂螺环上的基团,并且存在多个同分异构体。
厄贝沙坦制剂在高温高湿条件下杂质Ⅰ、杂质Ⅴ、杂质Ⅸ和杂质Ⅺ含量均有明显增加趋势,推测均为制剂降解杂质。此外,杂质Ⅰ、杂质Ⅱ和杂质Ⅳ为碱降解杂质,共计6个降解杂质。杂质Ⅴ结构上比厄贝沙坦增加了羟甲基,推测其是厄贝沙坦与甲醛加成反应生成的产物[10]。杂质Ⅸ和杂质Ⅺ是在厄贝沙坦结构上增加了羟乙基,推测其是厄贝沙坦与乙醛加成反应生成的产物,并且在反应时发生二氮杂螺环异构化,生成两个同分异构体。
根据降解杂质结构与来源分析,提示厄贝沙坦原料药与制剂生产企业在生产过程中需要关注高温、高湿和碱性等影响因素对厄贝沙坦稳定性产生的影响。对于厄贝沙坦制剂过程中以及辅料[15]中可能引入的甲醛和乙醛残留需要严格控制,避免厄贝沙坦和甲醛和乙醛反应生成相应的杂质产物。
