OBJECTIVE To establish a highly specific TaqMan real-time PCR method for the identification of Periplaneta americana medicinal materials and Periplaneta americana powder.METHODS According to the characteristics of genome sequence of Periplaneta americana, specific primers and probes suitable for real-time PCR detection were designed, and the probe concentration, cycle times and annealing temperature were optimized to establish the best TaqMan real-time PCR method of Periplaneta americana. RESULTS The specific primers and probes were only specifically amplified from Periplaneta americana medicinal materials and Periplaneta americana powder, and no fluorescence curves were amplified from Blaptica dubia, Eupolyphaga Sinensis W. Blattella germanica and Aspongopus chinensis Dallas. CONCLUSION The established TaqMan real-time PCR method can accurately and rapidly identify Periplaneta americana and common counterfeit products, providing a more specific technical reference for the identification of Periplaneta americana.

目的 本研究旨在建立一种具有高特异性的TaqMan探针实时荧光聚合酶链式反应(PCR)方法,用于美洲大蠊药材以及中药饮片美洲大蠊粉的鉴别。方法 依据美洲大蠊基因组序列的特点,设计出适用于实时荧光PCR检测的特异性引物和探针,并对探针浓度、循环数和退火温度进行优化,建立能够快速鉴别美洲大蠊的TaqMan探针实时荧光PCR最佳方法,进一步验证该方法的特异性和灵敏度。结果 研究表明特异性引物和探针仅对美洲大蠊药材以及美洲大蠊粉有特异性扩增,对杜比亚蟑螂、土鳖虫、德国小蠊和九香虫均未扩增出荧光曲线。结论 所建立的TaqMan探针实时荧光PCR法能够准确、快速对美洲大蠊及常见混伪品进行鉴别,为美洲大蠊的鉴别提供一种专属性更强的技术手段参考。

目的 本研究旨在建立一种具有高特异性的TaqMan探针实时荧光聚合酶链式反应(PCR)方法,用于美洲大蠊药材以及中药饮片美洲大蠊粉的鉴别。方法 依据美洲大蠊基因组序列的特点,设计出适用于实时荧光PCR检测的特异性引物和探针,并对探针浓度、循环数和退火温度进行优化,建立能够快速鉴别美洲大蠊的TaqMan探针实时荧光PCR最佳方法,进一步验证该方法的特异性和灵敏度。结果 研究表明特异性引物和探针仅对美洲大蠊药材以及美洲大蠊粉有特异性扩增,对杜比亚蟑螂、土鳖虫、德国小蠊和九香虫均未扩增出荧光曲线。结论 所建立的TaqMan探针实时荧光PCR法能够准确、快速对美洲大蠊及常见混伪品进行鉴别,为美洲大蠊的鉴别提供一种专属性更强的技术手段参考。

OBJECTIVE To establish a highly specific TaqMan real-time PCR method for the identification of Periplaneta americana medicinal materials and Periplaneta americana powder.METHODS According to the characteristics of genome sequence of Periplaneta americana, specific primers and probes suitable for real-time PCR detection were designed, and the probe concentration, cycle times and annealing temperature were optimized to establish the best TaqMan real-time PCR method of Periplaneta americana. RESULTS The specific primers and probes were only specifically amplified from Periplaneta americana medicinal materials and Periplaneta americana powder, and no fluorescence curves were amplified from Blaptica dubia, Eupolyphaga Sinensis W. Blattella germanica and Aspongopus chinensis Dallas. CONCLUSION The established TaqMan real-time PCR method can accurately and rapidly identify Periplaneta americana and common counterfeit products, providing a more specific technical reference for the identification of Periplaneta americana.

美洲大蠊(Periplaneta americana)为昆虫纲有翅亚纲蜚蠊目(Blattaria)蜚蠊科(Blattidae)大蠊属(Periplaneta)昆虫,俗名“蟑螂”。作为传统中药材,美洲大蠊已具有上千年用药历史,最早记载于《神农本草经》[1]。现代研究表明具有抗炎、抗肿瘤、增强机体免疫能力[2]等功效,虽暂时未被录入2020年版《中国药典》,但已被湖南、云南、四川等多省中药材标准单独收录。美洲大蠊常以干燥虫体为中药原料,其制剂也逐渐应用于临床[3],市场上有不少与美洲大蠊形态特征近似的品种,如德国小蠊(Blattella germanica)、土鳖虫(Eupolyphaga Sinensis W.)及杜比亚蟑螂(Blaptica dubia)等,容易出现用药掺假现象,药材的质量均一性和稳定性难以得到保障。美洲大蠊粉为美洲大蠊干燥虫体经炮制而成的中药饮片,能够健脾消疳,活血通脉,利水消肿,敛疮生肌。因美洲大蠊常以粉末形式入药,通过外观形状特征难以辨别其真伪,而药材的真伪优劣会直接影响到用药的安全性和治疗的有效性,因此,需要提升与完善美洲大蠊质量控制。
现在已研究的美洲大蠊鉴别方法有性状鉴别、显微鉴别、光谱鉴别、色谱鉴别[4]和DNA条形码分子鉴别[5]等。但对美洲大蠊鉴别方法、质量评价方式、质量控制标准没有统一规定,各省中药材标准对美洲大蠊的鉴别,多集中于美洲大蠊中丙氨酸的薄层色谱法鉴别,鉴别方式专属性不强。
随着分子生物学技术的不断发展,分子鉴定技术在中药材鉴定领域得到了广泛应用,如DNA条形码、聚合酶链式反应(PCR)、PCR-RFLP等技术都在药用植物中有所应用[6]。实时荧光PCR鉴别方法是基于特异性单核苷酸多态性(SNP)位点差异,设计特异性引物及探针,因不同物种间DNA具有高度保守的特性,该方法具有专属性较强、结果准确、耐用性好等特点[7]
本研究采用TaqMan探针实时荧光PCR技术,基于物种特异性核DNA片段序列,设计具有高特异性的引物和探针,在实际样品检测中主要用于美洲大蠊的定性检测。该方法不仅能够鉴别美洲大蠊成分,还可区分市场中流通的美洲大蠊混伪品。同时该方法也可用于中药饮片美洲大蠊粉的鉴别,增强美洲大蠊粉质量标准的可控性,实现了美洲大蠊及常见混伪品的快速鉴别,同时为完善美洲大蠊的质量评价体系提供技术参考。

血液/细胞/组织基因组DNA提取试剂盒 [天根生化科技(北京)有限公司,DP304-03];BestarTMqPCR Master Mix(美国DBI公司,P-2025259),引物和探针(上海生工生物工程有限公司);无水乙醇、三氯甲烷均为分析纯。
GenBank数据库中下载美洲大蠊以及常见混伪品的COI序列,与测序获得的序列共同使用BioEdit软件进行同源对齐。寻找发现520,571,634 bp等多个差异性SNP位点(图1),如美洲大蠊在520 bp处为C,其他物种为T,且种内高度保守。在种内保守、种间差异区域设计引物和探针,其中探针5'端标记荧光素基团FAM,3'端标记淬灭基团BHQ1,通过Primer Premier 5.0软件设计美洲大蠊特异性鉴别引物和TaqMan探针(表1),引物和探针位置见图1,引物和探针委托上海生工生物工程有限公司合成。
分别取美洲大蠊和杜比亚蟑螂等药材,用体积分数75%乙醇擦拭干净,晾干,置乳钵中研磨成极细粉。取样品粉末约40 mg,置1.5 mL离心管中,按照血液/细胞/组织基因组DNA提取试剂盒使用说明提取DNA,用超微量紫外分光光度计测定DNA的浓度,吸光度在0.01~1.5之间,测量结果可靠,DNA的A260/A280比值在1.8~2.0之间,表明提取纯度较高,没有蛋白质或RNA的干扰,后置于-20 ℃冰箱备用。取蜚蠊对照药材研磨成极细粉,同方法制备成阳性对照溶液。
PCR反应体系:总体积为20 μL,包括Bestar TMq PCR Master Mix 10 μL,探针1 μL,上下游引物各1 μL,模板DNA溶液0.5 μL,灭菌超纯水6.5 μL。平行设3个重复,Ct值取3个重复的平均值作为最终结果。
实时荧光PCR反应程序:95 ℃预变性3 min;95 ℃变性10 s,60 ℃退火34 s,40个循环。
实验应同时满足以下要求:空白对照无荧光对数增长,相应的Ct值>40.0或无法确定;阳性对照有荧光对数增长,且相应的Ct值小于30.0。
在符合以上要求的情况下,结果判定:无荧光对数增长,或样品与阳性对照的Ct值之差(ΔCt)绝对值>4.0,可判定结果为阴性,即未检出美洲大蠊;有荧光对数增长,且ΔCt绝对值≤4.0,可判定结果为阳性,即检出美洲大蠊。
将“1.4”项下提取的美洲大蠊样品和其他样品的DNA作为模板。按照“1.5”项反应体系和条件进行扩增,以水作为空白对照,验证引物和探针对美洲大蠊样品扩增的特异性。
选取美洲大蠊DNA样本,用灭菌超纯水进行逐级5倍梯度稀释,共稀释成6个浓度梯度,每个梯度设3个平行实验,按照“1.5”项中反应体系和条件扩增,测试该方法的灵敏度。
Ct值比较法是实时荧光PCR的一种相对定量方法,通过标准品和样品的Ct值之差(即ΔCt)来计算相对量。PCR呈指数扩增,Ct值是到达设定的阈值荧光信号时所经历的循环数,样品中的目的DNA所占比例用T表示(公式1)。

在杜比亚蟑螂中掺入不同比例的蜚蠊对照药材,蜚蠊掺入比例分别为3%、5%、10%、30%、50%。利用优化后的PCR扩增条件进行扩增,并根据Ct值比较法进行相对定量测定。
采用建立的方法和条件对收集到的样品进行实时荧光PCR反应,以验证该方法在实际检测工作中的使用价值。
引物和探针的选择是本实验的关键步骤,将测序获得美洲大蠊的COI序列在NCBI数据库进行BLAST同源性比对分析,测序结果与美洲大蠊(Periplaneta americana, GenBank序列号MF149498.1)相似度达到100%,同源性分析表明测序结果准确可靠,能够鉴别美洲大蠊。利用本研究设计的引物探针对蜚蠊对照药材、美洲大蠊以及杜比亚蟑螂等不同药材的DNA进行实时荧光PCR检测,结果表明引物和探针仅对蜚蠊对照药材和美洲大蠊药材有S型荧光扩增曲线,杜比亚蟑螂、土鳖虫等药材均无S型荧光扩增曲线,说明本研究设计的引物和探针用于美洲大蠊检测具有良好的特异性和有效性,可以用于下一步实验,见图2
为提高扩增效率,保证PCR反应完全,分别对探针浓度(2.5和5 μmol·L-1)、循环数(35和40个循环)和退火温度(60 和65 ℃)进行优化。
当探针浓度为5 μmol·L-1时,实时荧光扩增曲线强度较高;增加循环数,实时荧光扩增效率及PCR反应完成度明显提高;退火温度是指引物和模板结合时候的温度,当退火温度为65 ℃时,荧光曲线出现折线形且无荧光对数增长,分析原因可能因为65 ℃与当前引物中GC含量差异较大,不能成功扩增,结果见图3
通过考察影响实时荧光PCR反应的关键因素,最终确定美洲大蠊实时荧光PCR反应体系:总体积为20 μL,Bestar TMq PCR Master Mix 10 μL,探针(5 μmol·L-1)1 μL,上下游引物(2.5 μmol·L-1)各1 μL,模板DNA溶液0.5 μL,灭菌超纯水6.5 μL。
用灭菌超纯水将提取的美洲大蠊DNA(初始浓度为168 ng·μL-1)5倍梯度稀释成6个系列梯度,稀释后浓度分别为0.053 76~168 ng·μL-1,取0.5 μL作为模板用于灵敏度检测,结果表明DNA原液及稀释液样品均得到典型的扩增曲线。以扩增曲线的Ct值为纵坐标,相应浓度的log值为横坐标,绘制标准曲线(图4)。所得线性方程为y=-3.585 6x+31.011,标准曲线相关系数为0.997 8,具有良好的线性关系,扩增效率为90.06%,表明该方法具有较高灵敏度和有效性。
按确定的最佳方法和条件对美洲大蠊进行重复性和精密度测定,平行提取6份美洲大蠊药材,平均Ct值为26.751,RSD为1.35%,同一份美洲大蠊DNA样品,设6个复孔,平均Ct值为26.575,相对标准偏差(RSD)为1.07%,实验结果表明该方法具有良好的重复性和精密度。
在杜比亚蟑螂中掺入不同比例的蜚蠊对照药材进行实时荧光PCR方法检测限分析,通过Ct值之差的方法计算掺伪比例,以计算值与预期值比值的百分数作为回收率,蜚蠊掺伪定量检测的回收率在90.93%~104.81%之间(表2)。
理论上当ΔCt绝对值为4时,掺伪比例为6.25%,当ΔCt绝对值为5时,掺伪比例为3.13%。实际检测中,运用Ct值比较法对不同比例的蜚蠊对照药材掺伪品进行了相对定量检测,计算结果与预期的结果相仿,蜚蠊掺入比例为3%时仍能稳定检出,掺入比例为5%时,其ΔCt绝对值在4~5之间(表2)。综合考虑到药材及饮片的市场现状和实验环境等因素带来的误差,将ΔCt绝对值≤4作为检出美洲大蠊的判断依据。
在实际检测工作中,将实验室收集的4批美洲大蠊药材、10批中药饮片美洲大蠊粉进行DNA提取,然后按照上述建立的美洲大蠊TaqMan探针实时荧光PCR方法进行检测。结果表明,4批美洲大蠊药材和10批美洲大蠊粉均有荧光扩增曲线,且与阳性对照的Ct值之差绝对值≤4.0 (表3),检测结果为阳性,表明本研究建立的荧光PCR方法可以准确地用于市售美洲大蠊以及美洲大蠊粉的实际检测。另一方面,也将实验室收集到的市场上常见混伪品进行检测,结果显示,杜比亚蟑螂、土鳖虫、德国小蠊和九香虫则无荧光扩增曲线,检测结果为阴性,这也进一步表明本研究所建立的检测方法具有良好的特异性(图5)。本研究所建立的实时荧光PCR检测方法快速、可靠且稳定,可以用于美洲大蠊药材以及中药饮片美洲大蠊粉的快速检测。
美洲大蠊作为一种传统动物药,在临床上有其独特疗效,具有抑菌[8]和促进伤口愈合方面[9]等显著疗效。近几年,国内外学者对美洲大蠊的关注日益增多,国内报道较国外多,但主要集中在其疗效机制以及药理作用[10]。随着需求量的增加,对美洲大蠊质量控制的首要任务是品种鉴定和真伪鉴别,但目前尚未见统一的质量控制标准,已有的鉴定方法未能结合美洲大蠊自身特性,专属性较低,因此,需要完善并提高其质量评价体系,进一步推进美洲大蠊药材质量评价的科学与有效性,保障临床用药的安全性。
对美洲大蠊进行研究时,应考虑动物药的特殊性,在传统鉴别方法的基础上,结合现代新方法新技术建立专属性较强的鉴定方法。分子生物学在中药领域的应用逐渐广泛,实时荧光定量PCR作为基因检测的重要手段,以不同物种间遗传信息的差异作为研究点,具有较高的特异性和灵敏度,受DNA杂质的影响较小[11]。动物药的分子生物学鉴别多采用线粒体基因序列COI片段[12-14],物种水平的鉴定效率较高。
本研究设计出有效性和专属性较强的引物和探针,通过引物、探针组合的设计,建立了一种具有高度可复制性和良好特异性的美洲大蠊TaqMan探针实时荧光PCR检测方法,并对该检测方法的关键参数进行了优化和验证,使得方法具有较高的灵敏度。利用PCR技术的特异性强、自动化程度高、检测周期短等特点,该方法不仅能对完整的美洲大蠊药材进行鉴定,还能对其破碎的组织、粉末等进行准确有效的鉴定,可避免传统PCR的污染问题,提高鉴别水平,在实际样品检测中主要应用于美洲大蠊的定性检测。实验通过制备不同比例的蜚蠊对照药材掺伪品,并运用Ct值比较法对这5种比例的自混掺伪样进行了相对定量检测,计算结果与预期的结果相仿,证明本方法有较高的精确性和较好的实用性,也可用于美洲大蠊的相对定量检测。实验结果表明,该方法可以成功地用于鉴定美洲大蠊,并且能够实时监测实验结果,操作更加简单快捷,大大缩短了检测周期,还可应用于检测中药饮片美洲大蠊粉,具有较强的实用价值。
