OBJECTIVE To establish a small column centrifugation combined with HPLC method for the determination of encapsulation efficiency for aptamer-modified cantharidin (CTD) liposomes targeting nano-system carrier system (Apt-CTD-NLP), and investigate the pharmaceutical characteristics. METHODS The surface of CTD-Lip was modified by chemical self-assembly technology by aptamer to construct Apt-CTD-NLP, and its pharmaceutical properties were characterized. CTD-Lip was made by the thin film dispersion method. Using the recovery rate of blank liposomes column and the elution rate of free CTD as indexes, the optimum centrifugation conditions of small column were investigated by single factor, and the encapsulation rate of Apt-CTD-NLP was quantitatively determined by HPLC. RESULTS Apt-CTD-NLP was regularly round and spherical, and the mean particle size, PDI and Zeta potential were (101.0±1.058) nm, (0.208±0.012) and (-1.9±0.397) mV, and had a sustained release effect. The optimal conditions for separation of free CTD by dextran gel column were as follows: 400 μL loading volume, standing adsorption for 25 min, centrifugation for 5 min at 1 000 r·min-1, and elution with deionized water for 3 times (500 μL, 1 000 r· min-1×5 min). The average encapsulation rate of three batches of Apt-CTD-NLP was (94.56±0.80)%. CONCLUSION For lipophilic small molecule drug liposomes, column centrifugation combined with HPLC can quickly and effectively separate liposomes from free drugs, and determine the encapsulation rate of Apt-CTD-NLP.

目的 建立小柱离心联合高效液相色谱法(HPLC)测定适配体修饰的斑蝥素 (CTD) 脂质体靶向纳米载药体系(Apt-CTD-NLP)的包封率,并进行药剂学性质研究。方法 采用薄膜分散法制备CTD-NLP,其表面通过化学自组装技术用适配体进行修饰,构建Apt-CTD-NLP并进行理化性质表征。以空白脂质体柱回收率与游离CTD洗脱率为指标,单因素考察优化小柱分离条件,用HPLC定量测定Apt-CTD-NLP的包封率。结果 Apt-CTD-NLP呈规则圆球形,平均粒径、多分散系数(PDI)、Zeta电位分别为 (101.0±1.058) nm、(0.208±0.012)、(-1.9±0.397) mV,且具有缓释作用。葡聚糖凝胶小柱分离游离CTD的最佳条件:400 μL上样量,静置吸附25 min,1 000 r·min-1离心5 min,去离子水洗脱3次 (500 μL,1 000 r·min-1离心 5 min)。3批Apt-CTD-NLP平均包封率为 (94.56±0.80)%。结论 对于亲脂性小分子药物脂质体,小柱离心联合HPLC可快速有效地分离脂质体与游离药物,测定Apt-CTD-NLP的包封率。

目的 建立小柱离心联合高效液相色谱法(HPLC)测定适配体修饰的斑蝥素 (CTD) 脂质体靶向纳米载药体系(Apt-CTD-NLP)的包封率,并进行药剂学性质研究。方法 采用薄膜分散法制备CTD-NLP,其表面通过化学自组装技术用适配体进行修饰,构建Apt-CTD-NLP并进行理化性质表征。以空白脂质体柱回收率与游离CTD洗脱率为指标,单因素考察优化小柱分离条件,用HPLC定量测定Apt-CTD-NLP的包封率。结果 Apt-CTD-NLP呈规则圆球形,平均粒径、多分散系数(PDI)、Zeta电位分别为 (101.0±1.058) nm、(0.208±0.012)、(-1.9±0.397) mV,且具有缓释作用。葡聚糖凝胶小柱分离游离CTD的最佳条件:400 μL上样量,静置吸附25 min,1 000 r·min-1离心5 min,去离子水洗脱3次 (500 μL,1 000 r·min-1离心 5 min)。3批Apt-CTD-NLP平均包封率为 (94.56±0.80)%。结论 对于亲脂性小分子药物脂质体,小柱离心联合HPLC可快速有效地分离脂质体与游离药物,测定Apt-CTD-NLP的包封率。

OBJECTIVE To establish a small column centrifugation combined with HPLC method for the determination of encapsulation efficiency for aptamer-modified cantharidin (CTD) liposomes targeting nano-system carrier system (Apt-CTD-NLP), and investigate the pharmaceutical characteristics. METHODS The surface of CTD-Lip was modified by chemical self-assembly technology by aptamer to construct Apt-CTD-NLP, and its pharmaceutical properties were characterized. CTD-Lip was made by the thin film dispersion method. Using the recovery rate of blank liposomes column and the elution rate of free CTD as indexes, the optimum centrifugation conditions of small column were investigated by single factor, and the encapsulation rate of Apt-CTD-NLP was quantitatively determined by HPLC. RESULTS Apt-CTD-NLP was regularly round and spherical, and the mean particle size, PDI and Zeta potential were (101.0±1.058) nm, (0.208±0.012) and (-1.9±0.397) mV, and had a sustained release effect. The optimal conditions for separation of free CTD by dextran gel column were as follows: 400 μL loading volume, standing adsorption for 25 min, centrifugation for 5 min at 1 000 r·min-1, and elution with deionized water for 3 times (500 μL, 1 000 r· min-1×5 min). The average encapsulation rate of three batches of Apt-CTD-NLP was (94.56±0.80)%. CONCLUSION For lipophilic small molecule drug liposomes, column centrifugation combined with HPLC can quickly and effectively separate liposomes from free drugs, and determine the encapsulation rate of Apt-CTD-NLP.

《神农本草经》记载斑蝥可攻毒蚀疮、逐瘀散结,研究表明其有效成分斑蝥素 (cantharidin,CTD) 具有多靶点、多途径、多机制抗恶性肿瘤作用[1-3],但其毒副作用大、靶向性低、水溶性差,临床应用严重受限[4-6]。脂质体生物相容性好、易于修饰,是FDA批准用于临床的第一个纳米载药系统[7-9]。适配体(aptamer,Apt ) 是能高特异识别靶标,且免疫原性低的单链寡聚核苷酸,成为最佳靶向分子之一[10-11]。因此,本研究采用HepG2肝癌细胞的适配体通过化学自组装技术修饰到脂质体表面,构建适配体修饰的斑蝥素脂质体靶向纳米载药体系 (Apt-CTD-NLP),精准识别肝癌细胞,增强抗肝癌效果,降低CTD的毒副作用。包封率是评价脂质体处方工艺和包封状况的重要指标[12],是降低CTD毒副作用的重要因素。
脂质体包封率常用测定方法包括透析法、超滤离心法、超速离心法等[13-15]。采用透析法测定时,由于包封药物泄漏导致结果偏低,且游离的CTD被透析介质稀释几十倍,必须经过浓缩才能用高效液相色谱法(HPLC)检测。超滤离心法则由于“浓差极化”使得药物易附着在超滤膜上导致测定结果不准,且成本高[16]。超速离心法则由于脂溶性药物沉降于油水界面和离心管底部,导致测量结果偏高[17]。小柱离心联合HPLC基于葡聚糖凝胶色谱的“分子筛”原理,在外加离心力作用下实现游离药物的快速分离,葡聚糖凝胶用量少、成本低,洗脱液用量少不会过度稀释样品,可以直接用HPLC进行定量测定[12,17]
在Apt-CTD-NLP靶向纳米载药体系包封率的测定过程中,建立了葡聚糖凝胶小柱联合HPLC定量测定包封率的方法。以空白脂质体柱回收率与游离CTD洗脱率为指标单因素考察优化小柱分离条件,建立了Apt-CTD-NLP包封率的测定方法。用去离子水洗脱3次就可以完全分离游离药物和脂质体。该法能高效分离体系中游离CTD、简便易行、测定结果可靠,为后续进一步深入研究Apt-CTD-NLP的体内外抗肝肿瘤作用提供实验基础。

CTD (批号:20080601,纯度大于98%,武汉欣欣佳丽生物科技有限公司);CTD对照品 (批号:110783-202206,纯度大于96.4%,中国食品药品检定研究院);Apt [批号:1926742116,生工生物工程 (上海) 股份有限公司];葡聚糖凝胶 SephadexG-50 (批号:17004201,瑞典 Pharmacia公司);大豆卵磷脂 (批号:579010-1150055-13)、硬脂酰基磷脂酰乙醇胺-聚乙二醇2000(DSPE-PEG2000,批号:588200-2190092-01)、DSPE-PEG2000-马来酰亚胺(DSPE-PEG2000-Mal,批号:115CCG1663) (德国Lipoid公司);磷酸盐缓冲液 (PBS,批号:23045283,广州赛国生物科技有限责任公司);二氯甲烷(批号:20220516,分析纯,太仓沪试剂有限公司);胆固醇 (批号:20181116,上海国药集团化学试剂有限公司); 甲醇为色谱纯。
精密称取2 mg CTD、60 mg大豆卵磷脂、5 mg胆固醇、12 mg DSPE-PEG2000及945 μL DSPE-PEG2000-Mal (0.04 mg·mL-1) 置于250 mL茄型瓶,加入10 mL二氯甲烷,超声溶解 (功率:200 W,频率:400 kHz,5 min),55 ℃恒温水浴,减压旋蒸除去二氯甲烷,形成均匀透明的脂质薄膜,加入10 mL PBS (pH 7.4), 35℃恒温水浴,搅拌水化1 h,探头超声10 min(总功率100 W,超声2 s,间隔2 s),经过0.22 μm 微孔滤膜过滤,即得带有淡蓝色乳光的斑蝥素脂质体靶向纳米载药体系(CTD-NLP),同法制得NLP。按n(DSPE-PEG2000-Mal)-n适配体=10∶1的比例将CTD-NLP与适配体在4℃、N2保护下孵育24 h,制成带有淡蓝色乳光的Apt-CTD-NLP (图1),同法制得适配体-空白脂质体 (Apt-NLP),4℃储藏待用。
取1 mL Apt-CTD-NLP用PBS定容于10 mL量瓶中,准确移取10 μL滴入铜网中,静置24 h干燥,用透射电子显微镜观察其形态。用激光粒度仪测定Apt-CTD-NLP的粒径、Zeta电位及多分散系数(PDI)。结果见图2图3,Apt-CTD-NLP为规则的圆球形,分散均匀,粒径分布在 (101.0±1.058) nm,平均PDI为 (0.208±0.012),平均Zeta电位为 (-1.9±0.397) mV。结果表明,Apt-CTD-NLP粒径大小利于在肿瘤部位的聚集,PDI数值小于0.3符合药典标准[18],表明Apt-CTD-NLP分布均匀,分散性好。
采用透析法评价Apt-CTD-NLP、CTD-NLP的体外释放。将2 mL脂质体加入透析袋中,加入300 mL 体积分数0.25%Tween 80的PBS(pH 7.4)缓冲液,于37 ℃、360 r·min-1恒温振荡48 h,并于0.5、1、2、7、12、24、48 h准确量取1 mL透析介质(同时补充等量的释放介质)测定CTD质量浓度,计算释放率,结果见图4。CTD-NLP、Apt-CTD-NLP在30 min内释放率均未超过40%,在1~2 h之间Apt-CTD-NLP、CTD-NLP装载的CTD均迅速释放,2 h后缓慢释放,12 h累积释放率均超过80%,48 h的累积释放率分别为87.95%、90.63%,结果表明制备得到的Apt-CTD-NLP、CTD-NLP具有缓释作用,适配体的修饰并未影响CTD的释放。
WondaSil C18 Superb 色谱柱(4.6 mm×250 mm,5 μm)。流动相:水-甲醇 (23∶77);流速1.0 mL·min-1;柱温:30 ℃;检测波长:233 nm;进样量:20 μL。
CTD对照品和供试品的制备:分别精密称取CTD对照品、供试品20 mg,置于10 mL量瓶中,以色谱甲醇溶解定容,分别制成2.00 mg·mL-1的CTD对照品和供试品,4 ℃储存备用。
脂质体供试品的制备:准确量取1 mL空白脂质体置于10 mL量瓶中,甲醇定容,超声破乳30 min (功率为200 W,频率为400 kHz),静置24 h,即得空白脂质体破乳液,同法制得Apt-CTD-NLP破乳液。
分别取2.00 mg·mL-1 CTD对照品溶液、2.00 mg·mL-1 CTD供试品溶液、空白脂质体破乳液和Apt-CTD-NLP破乳液,按“2.2.1”项色谱条件检测,记录色谱图,见图5。结果表明,在“2.2.1”项色谱条件下,辅料对CTD的含量测定无干扰,此条件可用于CTD的定量检测。
分别准确量取2.00 mg·mL-1 CTD对照品溶液0.125、0.5、1.25、2.5、3.75 mL置于5 mL的量瓶,色谱甲醇定容配成 0.05、0.20、0.50、1.00、1.50 mg·mL-1的标准溶液,0.22 μm微孔滤膜过滤,进样HPLC检测,采用外标法对CTD对照品的质量浓度进行测定,记录峰面积。以对照品质量浓度为横坐标 (x,mg·mL-1),峰面积为纵坐标 (y) 得到线性回归方程y=905.69x+1.986,r=0.999 (n=6),表明CTD质量浓度与峰面积在0.05~2.00 mg·mL-1有良好的线性关系。
分别准确量取2.00 mg·mL-1 CTD对照品溶液0.125、2.5 mL置于5 mL的量瓶,用色谱甲醇定容配成0.05、1.00 mg·mL-1,取低、中、高质量浓度 (0.05、1.00、2.00 mg·mL-1) 进样HPLC检测,重复进样5次,记录峰面积,见表1。结果表明,进样量为20 μL,低、中、高浓度的峰面积RSD值 (n=5) 分别为1.48%、0.13%、0.10%,表明进样精密度良好。
按照“2.1.1”项条件制备Apt-CTD-NLP,准确量取1 mL Apt-CTD-NLP置于10 mL的量瓶,用甲醇定容破乳,HPLC重复进样检测5次,CTD的峰面积分别为531.4、530.8、528.2、530.5、533.2,平均峰面积为530.82,RSD为1.61%,表明重复性良好。
分别准确量取2.00 mg·mL-1 CTD对照品溶液0.125、2.5 mL置于5 mL的量瓶,用色谱甲醇定容配成0.05、1.00 mg·mL-1测试液。取1 mL低、中、高质量浓度(0.05、1.00、2.00 mg·mL-1)分别与等量的空白脂质体混匀,按照“2.2.1”项条件进样HPLC检测,记录峰面积,见表2。结果表明,在“2.2.1”项色谱条件下,3份低、中、高浓度的回收率均在90%~110%之间,表明此方法的回收率合格。
称取3 g Sephadex G-50葡聚糖凝胶浸于60 mL去离子水24 h,充分溶胀后填充至2.5 mL注射器 (底部放适量脱脂棉),静置至底端不再滴水,将其放到10 mL小管一起置于离心管中,1 000 r·min-1斜角离心1 min使凝胶脱水形成表层平整、高度约为2 cm的小柱。
准确量取100 μL空白脂质体,用PBS (pH 7.4) 定容于5 mL量瓶中,以PBS为空白对照,200~450 nm进行紫外扫描,测得空白脂质体的最大吸收波长为254 nm。故选择254 nm作为定量测定空白脂质体的吸收波长。
准确量取100、150、200 μL空白脂质体分别均匀加入Sephadex G-50葡聚糖凝胶小柱,静置吸附25 min,1 000 r·min-1离心5 min,去离子水洗脱2次(每次500 μL,1 000 r·min-1,离心5 min),收集洗脱液。以去离子水作为空白对照,于254 nm 测定空白脂质体 (A0) 及各次洗脱液 (Ax) 的吸光度,按公式1计算柱回收率。累积柱回收率分别为(98.37±0.26)%、(100.22±0)%、(100.96±0)%,结果表明,去离子水洗脱2次可将空白脂质体完全洗脱,累积小柱回收率可达到98%以上(公式1)。因此,可以用去离子水做洗脱剂。

准确量取3份150 μL空白脂质体均匀加至葡聚糖凝胶小柱顶部,分别吸附10、15、25 min,1 000 r·min-1离心5 min,用去离子水洗脱2次 (每次500 μL,1 000 r·min-1 ,离心5 min),收集洗脱液。于254 nm 测定洗脱液吸光度,按公式1计算柱回收率。静置吸附10、15、25 min的柱回收率分别为 (42.26±0)%、(94.46±0.19)%、(102.87±0)%,结果表明静置吸附25 min时,空白脂质体已完全洗脱,因此选择静置吸附时间为25 min。
准确量取3份150 μL空白脂质体均匀加至葡聚糖凝胶小柱顶部,静置吸附25 min,分别用700、1 000、1 500 r·min-1离心5 min,再用去离子水洗脱2次 (每次500 μL,1 000 r·min-1 ,离心 5 min),收集洗脱液,在254 nm处测定吸光度。按照公式1计算柱回收率。离心速度为700、1 000、1 500 r·min-1的柱回收率分别为 (95.60±0)%、(102.10±0.19)%、(103.44±0.19)%,离心速度为1 500 r·min-1时会造成柱子断裂,因此离心速度选用1 000 r·min-1
准确量取3份150 μL空白脂质体均匀加至葡聚糖凝胶小柱顶部,静置吸附25 min,1 000 r·min-1分别离心1、3、5 min,去离子水洗脱2次(每次500 μL,1 000 r·min-1,离心5 min),收集离心液,在254 nm处测定吸光度,按照公式1计算柱回收率。柱回收率分别为 (71.70±0)%、(98.90±0.38)%、(102.80±0.38)%。结果表明,离心时间越长,柱回收率越高。离心5 min时空白脂质体完全过柱,故离心时间定为5 min。
准确量取100、200、400 μL空白脂质体分别均匀加至葡聚糖凝胶小柱顶部,静置吸附25 min,1 000 r·min-1离心5 min,去离子水洗脱2次 (每次500 μL,1 000 r·min-1 ,离心5 min),收集洗脱液,在254 nm处测定吸光度。按照公式1计算。柱回收率分别为 (102.99±0)%、(99.04±0.19)%、(100.96±0)%,实验结果显示,上样量对柱回收率的影响相对较小,但研究发现上样量过大会导致游离药物与包封药物无法分离,故采用400 μL脂质体。
准确量取1 mL Apt-NLP和2.00 mg·mL-1 CTD溶液涡旋混匀制成物理混合物。准确量取400 μL物理混合物均匀加至葡聚糖凝胶小柱顶部,按照最佳分离条件,静置25 min,去离子水洗脱15次 (每次500 μL,1 000 r·min-1,离心5 min),收集各次洗脱液,置于2 mL量瓶中用甲醇定容。以甲醇为空白对照组,于254 nm处测定Apt-NLP的吸光度并计算其含量。按照“2.2.1”项色谱条件进样HPLC测定CTD含量。以洗脱次数为横坐标,分别以Apt-NLP、CTD的含量为纵坐标,绘制物理混合物的洗脱曲线,见图6。结果表明,既定的洗脱条件洗脱15次均未检测到CTD,洗脱3次就可以将物理混合物中的CTD和Apt-NLP完全分离。
按照“2.1.1”项条件制备3批Apt-CTD-NLP,准确量取2 mL Apt-CTD-NLP于10 mL量瓶,按照“2.2.2”项下条件破乳,进样HPLC测定并计算出CTD的总质量浓度ρ0。取同等体积的Apt-CTD-NLP,按照按最佳分离条件分离Apt-CTD-NLP和游离的CTD,收集洗脱液于10 mL量瓶,按照“2.2.2”条件破乳,采用HPLC测定并计算出包封的CTD质量浓度ρ1。按公式2计算3批Apt-CTD-NLP的包封率分别为94.29%、95.65%、93.75%,平均包封率为(94.56±0.80)%,RSD值为1.04%。

包封率是评价脂质体纳米载药体系质量的重要指标,包封率的大小会直接影响用药剂量和药物疗效[19]。本研究用小柱离心联合HPLC测定Apt-CTD-NLP的包封率是 (94.56±0.98)%,有望在后续Apt-CTD-NLP抗肝肿瘤作用的研究中,提高CTD抗肝肿瘤效果,降低CTD对其他正常细胞的毒副作用。
在探索Sephadex G-50葡聚糖凝胶小柱分离游离CTD条件时,本研究对空白脂质体进行200~450 nm紫外波长扫描,确定其最大吸收波长为254 nm。通过小柱离心前后空白脂质体的吸光度比值 (柱回收率) 作为评价指标,单因素考察优化小柱分离条件,建立了Apt-CTD-NLP包封率的测定方法。研究中发现有机溶剂易损坏葡聚糖凝胶的立体网状结构发生皱缩塌陷,导致药物无法分离,因此,本研究用去离子水做洗脱剂,且溶解CTD时尽量降低供试品中甲醇的用量,以确保最佳分离效果。离心转速、离心时间、吸附时间、上样量都会影响分离效果,因此在探索时需要对这些因素进行考察。本研究用小柱离心联合HPLC法测定包封率的最佳分离条件是400 μL上样量,静置吸附25 min,1000 r·min-1离心5 min,只需洗脱3次就可以完全分离游离药物。与传统葡聚糖凝胶柱色谱法相比,小柱离心联合HPLC测定法不仅成本低,而且更简便快速,能有效地测定CTD包封率。
综上所述,本研究建立了葡聚糖凝胶小柱离心联合HPLC定量检测Apt-CTD-NLP包封率的方法,并进行了系统的方法学研究。结果表明,该法简便快速,为后续进一步深入研究Apt-CTD-NLP的体内外抗肝肿瘤作用奠定实验基础。
