OBJECTIVE To establish content determination method of polyethylene glycol (PEG) in mupirocin ointment. METHODS The content determination method of PEG was performed on TSKgel G2000SWXL column at 35 ℃ using refractive index detector at 35 ℃, 0.1 mol·L-1 NaNO3(pH 3.0) was used as mobile phase and the flow rate was 1.0 mL·min-1,and the injection volume was 20 μL. RESULTS The content determination method of three different PEG in mupirocin ointment was established. The type and content of PEG in mupirocin ointment from different manufacturers were determined. In this method, the limits of detection (LOD) for PEG400, PEG3350, and PEG4000 were all 0.01 mg·mL-1. The limits of quantitation(LOQ) were all 0.02 mg·mL-1, and the ranges of linearity were all 0.2 to 5.0 mg·mL-1. The average recoveries were 99.3%、99.8% and 100.2%, respectively (n=3). CONCLUSION The established method is efficient, rapid, accurate and sensitive, which can be used as the content determination method of PEG in mupirocin ointment.

目的 建立莫匹罗星软膏中辅料聚乙二醇(PEG)的含量测定方法。方法 色谱柱为TSKgel G2000SWXL(7.8 mm×30 cm,5 μm),流动相为0.1 mol·L-1硝酸钠溶液(用磷酸调节pH值为3.0)-乙腈(95∶5),示差折光检测器,检测器温度为35 ℃,柱温为35 ℃,流速为1.0 mL·min-1,进样体积为20 μL。结果 建立了3种不同PEG的含量测定方法,并对不同厂家莫匹罗星软膏中PEG的种类及含量进行了测定。此方法中,PEG400、PEG3350及PEG4000的检测限均为0.01 mg·mL-1,定量限均为0.02 mg·mL-1,线性范围均为0.2~5.0 mg·mL-1,平均回收率分别为99.3%、99.8%和100.2%(n=9)。结论 所建立的PEG含量测定方法高效、快速、准确、灵敏,可对莫匹罗星软膏中不同种类PEG的含量进行控制。

目的 建立莫匹罗星软膏中辅料聚乙二醇(PEG)的含量测定方法。方法 色谱柱为TSKgel G2000SWXL(7.8 mm×30 cm,5 μm),流动相为0.1 mol·L-1硝酸钠溶液(用磷酸调节pH值为3.0)-乙腈(95∶5),示差折光检测器,检测器温度为35 ℃,柱温为35 ℃,流速为1.0 mL·min-1,进样体积为20 μL。结果 建立了3种不同PEG的含量测定方法,并对不同厂家莫匹罗星软膏中PEG的种类及含量进行了测定。此方法中,PEG400、PEG3350及PEG4000的检测限均为0.01 mg·mL-1,定量限均为0.02 mg·mL-1,线性范围均为0.2~5.0 mg·mL-1,平均回收率分别为99.3%、99.8%和100.2%(n=9)。结论 所建立的PEG含量测定方法高效、快速、准确、灵敏,可对莫匹罗星软膏中不同种类PEG的含量进行控制。

OBJECTIVE To establish content determination method of polyethylene glycol (PEG) in mupirocin ointment. METHODS The content determination method of PEG was performed on TSKgel G2000SWXL column at 35 ℃ using refractive index detector at 35 ℃, 0.1 mol·L-1 NaNO3(pH 3.0) was used as mobile phase and the flow rate was 1.0 mL·min-1,and the injection volume was 20 μL. RESULTS The content determination method of three different PEG in mupirocin ointment was established. The type and content of PEG in mupirocin ointment from different manufacturers were determined. In this method, the limits of detection (LOD) for PEG400, PEG3350, and PEG4000 were all 0.01 mg·mL-1. The limits of quantitation(LOQ) were all 0.02 mg·mL-1, and the ranges of linearity were all 0.2 to 5.0 mg·mL-1. The average recoveries were 99.3%、99.8% and 100.2%, respectively (n=3). CONCLUSION The established method is efficient, rapid, accurate and sensitive, which can be used as the content determination method of PEG in mupirocin ointment.

莫匹罗星软膏为莫匹罗星按2%配以聚乙二醇(polyethylene glycol, PEG)基质制成的水溶性软膏,为局部外用抗生素,通过抑制细菌蛋白和RNA的合成发挥抑菌作用[1],临床上主要用于治疗革兰阳性球菌引起的原发性皮肤感染及湿疹合并感染[2-3]。莫匹罗星软膏中的辅料为不同比例的PEG,用以调节软膏剂黏度。目前市售莫匹罗星软膏的辅料分别为PEG400和PEG3350或PEG400和PEG4000按照一定比例混合。
莫匹罗星软膏中不同种类PEG的含量以及比例不仅影响样品的性状、黏度以及稳定性,还会对药物释放以及经皮渗透产生较大影响,故有必要对不同厂家制剂中PEG含量及比例进行考察,作为莫匹罗星软膏剂质控指标之一,也可用以评价各厂家制剂处方的合理性。PEG因不含共轭结构,在紫外光区无吸收,因此无法采用紫外检测器进行测定。目前对PEG的检测通常采用示差折光检测器(RID)[4-6]以及蒸发光散射检测器(ELSD)[7-8]。使用ELSD时,要求流动相中不含有非挥发性缓冲盐,且待测物浓度和峰面积需分别取自然对数后成线性[9],定量较为复杂,因此本研究采用高效分子排阻色谱法结合示差折光检测器,建立了莫匹罗星软膏中PEG的含量测定方法并进行了方法学验证,在此基础上,对不同厂家莫匹罗星软膏中的PEG进行了定性及定量分析。
Waters e2695高效液相色谱仪(美国Waters公司),配备2414 RI Detector,Empower 3工作站;Sartorius MSA225S-1CE-DI电子天平(德国 Sartorius);Milli-Q 超纯水仪(美国Millpore公司);TSKgel G2000SWXL (7.8 mm×300 mm,5 μm, 日本东曹株式会社)。
莫匹罗星原料药(国内厂家1);莫匹罗星软膏:9厂家共27批,其中国内厂家8家,共24批,参比制剂1家,共3批。PEG400(国内厂家1);PEG3350(国内厂家2);PEG4000(国内厂家1);甲醇、乙腈(色谱纯);磷酸、硝酸钠(分析纯)。
流动相为0.1 mol·L-1硝酸钠(磷酸调节pH值至3.0):乙腈(95∶5),示差折光检测器,检测器温度为35 ℃,柱温为35 ℃,流速为1.0 mL·min-1,进样体积为20 μL。
供试品溶液:取莫匹罗星软膏约50 mg,置于25 mL量瓶中,用流动相溶解并稀释至刻度。
定位溶液:分别取PEG400、PEG3350、PEG4000、莫匹罗星原料约50 mg,置于25 mL量瓶中,用流动相溶解并稀释至刻度。
取两个不同厂家生产的莫匹罗星软膏按照“2.2”项下配制供试品溶液及定位溶液,按照“2.1”项下色谱条件进样,结果汇总见表1
分别精密称取PEG400、PEG3350、PEG4000各适量,制备50 mg·mL-1的溶液为储备液。精密量取储备液,稀释至不同质量浓度(分别为0.2、0.5、1.0、2.0、3.0、5.0 mg·mL-1)的线性溶液进样,以质量浓度(mg·mL-1)为横坐标,以峰面积为纵坐标进行线性回归。按照信噪比(S/N)分别为10和3确定方法的定量限和检测限。PEG400线性方程的相关系数为0.999 7,定量限浓度为0.02 mg·mL-1,检测限浓度为0.01 mg·mL-1;PEG3350线性方程的相关系数为0.999 7,定量限浓度为0.02 mg·mL-1,检测限浓度为0.01 mg·mL-1;PEG4000线性方程的相关系数为1.000 0,定量限浓度为0.02 mg·mL-1,检测限浓度为0.01 mg·mL-1
取市售莫匹罗星软膏(国内厂家1)适量,加入PEG3350、PEG400对照品溶液,配制成低、中、高3个浓度的加标供试品溶液,每个浓度平行配制3份;取市售莫匹罗星软膏(国内厂家6)适量,加入PEG4000对照品溶液,配制成低、中、高3个浓度的加标供试品溶液,每个浓度平行配制3份。按照“2.1”项下色谱条件进行测定,计算回收率,结果见表2~4
进样精密度:取PEG400、PEG3350、PEG4000对照品溶液连续进样6次,计算峰面积的相对标准偏差(RSD)分别为0.3%、0.2%和0.2%,均小于1%。
重复性:取两个厂家生产的莫匹罗星软膏按照“2.2”项下分别平行配制6份供试品溶液,测定其中PEG的含量(%,PEG的量占处方百分比)。国内厂家5的莫匹罗星软膏中PEG400含量RSD为0.3%,PEG3350含量RSD为0.7%;国内厂家8生产的莫匹罗星软膏中PEG400含量RSD为0.4%,PEG4000含量RSD为0.8%。
中间精密度:更换实验人员,于不同实验日期取两个厂家生产的莫匹罗星软膏按照“2.2”项下分别平行配制6份供试品溶液,测定其中PEG的含量。国内厂家5的莫匹罗星软膏中PEG400含量RSD为0.5%,PEG3350含量RSD为0.6%;国内厂家8生产的莫匹罗星软膏中PEG400含量RSD为0.6%,PEG4000含量RSD为0.8%。
采用建立的方法对9个厂家27批莫匹罗星软膏中不同PEG含量进行测定,结果见表5
以《中国药典》2020年版四部PEG400相对分子质量及其分布测定法项下色谱条件[10]为基础,流动相为0.1 mol·L-1的硝酸钠(含0.02%抑菌剂,测得其pH为5.7),示差折光检测器;检测器温度为35 ℃,柱温为35 ℃,流速为1.0 mL·min-1,进样量为20 μL,对该方法(以下简称PEG400方法)进行优化。
取“2.2”项下溶液,按照PEG400方法的色谱条件分别进样,结果见图1
图1可知,PEG3350与PEG4000色谱峰部分重叠,PEG400与莫匹罗星色谱峰(相对分子质量500.62)部分重叠。所使用色谱柱TSK G2000SWXL其分离机制为反相液相色谱及分子排阻色谱相结合[11],相对分子质量接近的化合物保留时间较为接近,故PEG3350与PEG4000在此色谱系统中无法达到基线分离,但莫匹罗星软膏中PEG3350与PEG4000不会同时使用,仅包含两者中的一种,两者不同时测定,不会产生相互干扰。
以PEG400与莫匹罗星色谱峰的分离度为指标对色谱条件进行优化,包括流速、柱温、流动相中有机相比例以及流动相pH值等。结果见图2~4以及表6。由图2可知,调节流速或柱温后,分离效果并无明显改善,分离效果与原条件基本一致,因此流速与柱温在分子排阻色谱法中并非影响分离效果的关键因素。由图3可知,在流动相中加入不同比例的甲醇或乙腈后,分离效果均较PEG400方法更差。PEG400方法中含有0.02%的抑菌剂,因抑菌剂对于人体具有明确的毒性,考虑向流动相中加入一定比例的有机相替代抑菌剂发挥抑菌作用。同时TSK柱能耐受的有机相比例通常小于20%,因此向流动相中加入5%的乙腈,继续调整其他因素以改善分离效果。由图4可知,在将硝酸钠溶液的pH值由5.7用磷酸调节为3.0并与乙腈按照95∶5的比例混合作为流动相时取得良好的分离效果,因此流动相的pH值为影响分离的关键因素,随着pH值降低,莫匹罗星的保留时间变长,PEG的保留时间基本不变,从而莫匹罗星与PEG400的分离度逐渐增大。由于TSK柱耐受的流动相pH范围一般不低于2.5,因此确定硝酸钠溶液的pH值为3.0。最终确定的色谱条件如下:流动相为0.1 mol·L-1硝酸钠(pH值3.0)-乙腈(95∶5),示差折光检测器,检测器温度为35 ℃,柱温为35 ℃,流速为1.0 mL·min-1,进样体积20 μL。不同色谱条件优化结果对比见表6
表5可知,参比制剂以及国内厂家1~5的制剂均采用的是PEG3350和PEG400,但是两种PEG比例相差较大,国内厂家1和2的PEG400/PEG3350约为4.2,而参比制剂及国内厂家5的比例约为1.5。国内厂家6~8采用的是PEG4000和PEG400,国内厂家6和7的PEG400/PEG4000均为4.0左右,国内厂家8为2.5。此结果说明国内厂家之间及与参比制剂之间处方存在较大差异,这就造成了不同厂家软膏剂的黏度相差较大,进而可能导致不同厂家软膏剂流变学特征、释放及经皮渗透的差异,从而影响软膏剂的有效性。
本研究采用分子排阻色谱法结合示差折光检测器建立了莫匹罗星软膏剂中3种不同PEG含量测定的方法,经方法学验证表明此方法专属性、精密度及准确度均较好。采用此方法对9个厂家莫匹罗星软膏剂中不同PEG进行了检则,结果显示,与参比制剂相比,国内大部分厂家PEG400含量均较高,导致国内厂家制剂黏度小于参比制剂,黏度过小,使得软膏剂从包装中挤出会较为容易,但其在皮肤上的黏附性以及在储存和运输过程中的稳定性均会降低,进而影响产品的有效性和安全性。故采用适当的方法对软膏剂的辅料组成成份进行测定,可对软膏剂的质量进行控制。
