OBJECTIVE To establish the HPLC fingerprints, determine the contents of chlorogenic acid, liquiritin, lobetyolin, liquiritigenin, glycyrrhetinic acid, atractylenolide Ⅲ, atractylenolide Ⅱ, atractylenolide Ⅰ, 10-gingerol in Lizhong Pills, and lay the foundation for its quality control. METHODS A Eclipse Plus C18 column was used with acetonitrile-0.1% aqueous phosphoric acid as the mobile phase in gradient elution at a flow rate of 0.8 mL·min-1. The detection wavelengths were set at 215 nm and 254 nm, and the column temperature was maintained at 30 ℃. Through similarity evaluation, combined with chemical pattern recognition, the fingerprints of 21 batches of Li Zhong Pills were evaluated and analyzed, and 9 index components were quantitatively determined. RESULTS The fingerprints of 21 batches of Lizhong Pills were established, with 29 peaks identified, the similarity of each batch was greater than 0.9, and 21 batches of samples from 3 manufacturers could be clustered into 3 categories, with 16 compounds differing between groups. Nine components showed good linear relationships within their respective linear ranges (r≥0.998 7), the average recoveries ranged from 95.34% to 104.41%, and the RSDs ranged from 0.95% to 2.95%. CONCLUSION The fingerprint and quantitative determination method of Lizhong Pills is simple, accurate and reproducible, and can be used for the evaluation of the overall quality of Lizhong Pills.

目的 建立理中丸高效液相色谱(HPLC)指纹图谱并测定绿原酸(chlorogenic acid)、甘草苷(liquiritin)、党参炔苷(lobetyolin)、甘草素(liquiritigenin)、甘草酸(glycyrrhetinic acid)、白术内酯Ⅲ(atractylenolide Ⅲ)、白术内酯Ⅱ(atractylenolide Ⅱ)、白术内酯Ⅰ(atractylenolide Ⅰ)和10-姜酚(10-gingero)的含量,为其质量控制提供依据。方法 采用Eclipse Plus C18色谱柱,以乙腈体积分数-0.1%磷酸水溶液为流动相,梯度洗脱,流速0.8 mL·min-1,检测波长215和254 nm,柱温30 ℃;通过相似度评价,结合化学模式识别,对21批理中丸指纹图谱进行评价及分析,并对9种指标成分进行定量测定。结果 21批理中丸指纹图谱中共标定29个共有峰,各批次的相似度均大于0.9,3个厂家21批样品可聚为3类,组间差异化合物有16个。9种成分在各自线性范围内线性关系良好(r≥0.998 7);平均加样回收率95.34%~104.41%,相对标准偏差(RSD)0.95%~2.95%。结论 建立的理中丸指纹图谱及多成分定量测定方法简单、准确、重现性较好,可用于理中丸整体质量评价。

目的 建立理中丸高效液相色谱(HPLC)指纹图谱并测定绿原酸(chlorogenic acid)、甘草苷(liquiritin)、党参炔苷(lobetyolin)、甘草素(liquiritigenin)、甘草酸(glycyrrhetinic acid)、白术内酯Ⅲ(atractylenolide Ⅲ)、白术内酯Ⅱ(atractylenolide Ⅱ)、白术内酯Ⅰ(atractylenolide Ⅰ)和10-姜酚(10-gingero)的含量,为其质量控制提供依据。方法 采用Eclipse Plus C18色谱柱,以乙腈体积分数-0.1%磷酸水溶液为流动相,梯度洗脱,流速0.8 mL·min-1,检测波长215和254 nm,柱温30 ℃;通过相似度评价,结合化学模式识别,对21批理中丸指纹图谱进行评价及分析,并对9种指标成分进行定量测定。结果 21批理中丸指纹图谱中共标定29个共有峰,各批次的相似度均大于0.9,3个厂家21批样品可聚为3类,组间差异化合物有16个。9种成分在各自线性范围内线性关系良好(r≥0.998 7);平均加样回收率95.34%~104.41%,相对标准偏差(RSD)0.95%~2.95%。结论 建立的理中丸指纹图谱及多成分定量测定方法简单、准确、重现性较好,可用于理中丸整体质量评价。

OBJECTIVE To establish the HPLC fingerprints, determine the contents of chlorogenic acid, liquiritin, lobetyolin, liquiritigenin, glycyrrhetinic acid, atractylenolide Ⅲ, atractylenolide Ⅱ, atractylenolide Ⅰ, 10-gingerol in Lizhong Pills, and lay the foundation for its quality control. METHODS A Eclipse Plus C18 column was used with acetonitrile-0.1% aqueous phosphoric acid as the mobile phase in gradient elution at a flow rate of 0.8 mL·min-1. The detection wavelengths were set at 215 nm and 254 nm, and the column temperature was maintained at 30 ℃. Through similarity evaluation, combined with chemical pattern recognition, the fingerprints of 21 batches of Li Zhong Pills were evaluated and analyzed, and 9 index components were quantitatively determined. RESULTS The fingerprints of 21 batches of Lizhong Pills were established, with 29 peaks identified, the similarity of each batch was greater than 0.9, and 21 batches of samples from 3 manufacturers could be clustered into 3 categories, with 16 compounds differing between groups. Nine components showed good linear relationships within their respective linear ranges (r≥0.998 7), the average recoveries ranged from 95.34% to 104.41%, and the RSDs ranged from 0.95% to 2.95%. CONCLUSION The fingerprint and quantitative determination method of Lizhong Pills is simple, accurate and reproducible, and can be used for the evaluation of the overall quality of Lizhong Pills.

理中丸出自张仲景《伤寒杂病论》,理中者,理中焦之气,以交于阴阳也。上焦属阳,下焦属阴,而中焦则为阴阳相交之处。理中汤一方,乃温中以交通上下焦之阴阳[1]。为《中国药典》2020年版一部收载的中药复方制剂,由党参、土白术、炙甘草、炮姜4味药组成[2],方中以白术为君,大补中宫之土。干姜辛热,能暖中宫之气,为臣药。但辛燥太过,恐伤脾中之阴血,复以甘温濡润之党参为佐药,刚柔相济。炙甘草为使药,与白术干姜辛药同用,便可化周身之阳气;与党参同用,可化周身之阴血。如此则中焦之气旺,阴邪即灭,中焦大振,自然体健身安。临床上可用于治疗脾胃虚寒、呕吐泄泻、胸满腹痛、消化不良等[3]
高效液相色谱(HPLC)指纹图谱作为一种高效、准确的药物质量控制方法,其高分辨率和高灵敏度能够有效地分离和测定中药中的复杂成分,已经在药物研发和生产过程中得到了广泛应用[4-5]。通过比较不同批次药物的指纹图谱,可以确保药物质量的稳定性,为患者提供安全、可靠的药品。该方现行质量标准中仅收载了对甘草的含量测定,而对理中丸中多个化学成分的同时测定鲜有报道。本实验建立理中丸HPLC指纹图谱同时测定绿原酸(chlorogenic acid)、甘草苷(liquiritin)、党参炔苷(lobetyolin)、甘草素(liquiritigenin)、甘草酸(glycyrrhetinic acid)、白术内酯Ⅲ(atractylenolide Ⅲ)、白术内酯Ⅱ(atractylenolide Ⅱ)、白术内酯Ⅰ(atractylenolide Ⅰ)和10-姜酚(10-gingerol)的含量,并进行方法学考察,通过相似度评价,结合聚类分析(CA)、主成分分析(PCA)及正交偏最小二乘法-判别分析(OPLS-DA)等化学模式识别进行分析[6],更全面地挖掘指纹图谱数据中的信息,快速筛选质量差异成分,以期为有效控制该制剂的质量控制、药效评估提供参考。
Waters e2695型液相色谱仪(美国Waters公司);XS105 DualRange型十万分之一天平(瑞士梅特勒-托利多公司);SBL-10DT型超声波恒温清洗机(宁波新芝生物科技股份有限公司)。
对照品绿原酸(批号:RDD-L00701908029,98.00%,成都瑞芬思德丹生物科技有限公司);对照品甘草苷(批号:14310,91.50%)、党参炔苷(批号:16550,≥98.00%)、甘草素(批号:14207,≥98.00%)、甘草酸(批号:16277,95.40%)、白术内酯Ⅲ(批号:16202,≥98.00%)、白术内酯 Ⅱ(批号:14498,≥98.00%)、白术内酯 Ⅰ(批号:14232,≥98.00%)和10-姜酚(批号:15456,≥98.00%)(上海诗丹德生物技术有限公司)。理中丸(规格:每丸重9 g)21批,编号S1~S21,具体见表1。乙腈为色谱纯;磷酸和甲醇为分析纯;水为纯化水;其他试剂为分析纯。
采用Eclipse Plus C18色谱柱(4.6 mm×250 mm,5 μm);乙腈(A)-体积分数0.1%磷酸溶液(B)为流动相;按表2进行梯度洗脱;检测波长为215 nm(绿原酸、甘草苷、党参炔苷、甘草素、白术内酯Ⅲ、白术内酯Ⅱ和10-姜酚)、254 nm(甘草酸、白术内酯 Ⅰ);柱温为30 ℃;流速为0.8 mL·min-1;进样量为10 μL。
分别精密称取绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内酯Ⅲ、白术内酯Ⅱ、白术内酯Ⅰ和10-姜酚对照品9.65、9.02、9.22、11.11、9.90、10.38、9.84、9.02和9.85 mg,置于10 mL量瓶中,用无水甲醇溶解并定容至刻度,4 ℃下保存备用。
将适量剪碎的理中丸与等比例硅藻土混匀,研成粉末。精密称定其粉末2 g于150 mL具塞锥形瓶中,加入体积分数70%甲醇溶液25 mL,称重并记录。超声提取45 min后,冷却并再次称重。用原溶解液补充损失的质量,摇匀,过0.45 μm有机微孔滤膜,即得。
按照本品处方工艺及比例,以“2.2.2”项下方法,分别制得缺炮姜、缺党参、缺土白术、缺甘草、缺党参和土白术的5种阴性样品,即得。
精密称定理中丸粉末(S5)2 g,按相应溶液制备方法和最佳色谱条件,连续进样6次,记录色谱图,以5号峰(甘草苷)为参照峰,计算各共有峰峰面积和相对保留时间的相对标准偏差(RSD)分别小于2.98%和0.076%,表明该方法精密度良好。
精密称定理中丸粉末(S5)2 g,按相应溶液制备方法和最佳色谱条件,平行制备6份样品溶液进样,记录色谱图,以5号峰(甘草苷)为参照峰,计算各共有峰峰面积和相对保留时间的RSD分别小于2.96%和0.086%,表明该方法重复性良好。
精密称定理中丸粉末(S5)2 g,按相应溶液制备方法,室温下于0、2、4、8、12、24 h在最佳色谱条件进样测定,记录色谱图,以5号峰(甘草苷)为参照峰,计算各共有峰峰面积和相对保留时间的RSD分别小于2.82%和0.079%,表明溶液在24 h内稳定性良好。
取21批理中丸样品(编号:S1~S21)按“2.1”项下方法,记录色谱图,将所得21批样品色谱图数据导入《中药色谱指纹图谱相似度评价系统》(2012年版),以S5样品的色谱图为参照图谱,对21批样品选择吸收信号较强、峰形明显、峰宽适中、稳定且分辨率高的峰为共有峰进行多点校正和标记(Mark)峰匹配,生成理中丸的对照特征图谱(R)和21批理中丸样品的HPLC叠加指纹图谱(图1),共确认29个共有峰(215 nm),通过与对照品相比,指认出了9个色谱峰,其中1号峰为绿原酸、5号峰为甘草苷、6号峰为党参炔苷、8号峰为甘草素、9号峰为甘草酸、17号峰为白术内脂 Ⅲ、23号峰为白术内脂 Ⅱ、27号峰为白术内脂 Ⅰ、28号峰为10-姜酚。计算S1~S21样品与对照特征图谱的相似度,分别为0.994、0.973、0.967、0.969、0.979、0.972、0.978、0.975、0.974、0.973、0.950、0.935、0.951、0.923、0.923、0.909、0.915、0.962、0.961、0.958、0.959,均>0.90,相似度良好,可知各批理中丸样品的质量比较稳定。
将21批理中丸样品的29个共有峰面积导入SIMCA 14.1软件中,以峰面积结果为变量,采用Ward法计算的距离用于将理中丸样品进行聚类(图2),结果显示,当度量距离为1.0×107时,21批理中丸样品可分为3大类:S2~S10为第一类、S18~S21为第二类、余下批次为第三类。这表明即使在指纹图谱相似度较好的情况下,不同批次药材质量仍存在一定差异。
将21批理中丸样品的29个共有峰面积导入到SIMCA 14.1软件中,以峰面积结果为变量进行PCA分析,得到21批理中丸样品的PCA得分图(图3),图3可以看出,21批样品可以分为3类,其中S2~S10聚为第一类,S18~S21聚第二类、余下批次为第三类。与聚类分析结果一致。
将21批样品中29个共有峰面积导入SIMCA 14.1软件,自动拟合建立OPLS-DA模型,结果该模型累积解释能力参数R2X(自变量矩阵)与R2Y(因变量矩阵)分别为0.746、0.952,预测能力参数Q2=0.848,表明所构建OPLS-DA模型拟合程度高,具有较高的解释能力,且稳定性和预测能力良好。OPLS-DA得分图见图4,该分类结果说明同一厂家生产的不同批次的样品质量一致性较好,但不同厂家之间样品的质量一致性存在明显差异。采用置换检验(n=200)对该模型进行验证,结果见图5,随着置换保留度的下降,R2Q2下降,回归线呈向上的趋势,表明所构建的OPLS-DA模型未出现过拟合现象,具有较好的预测能力,可用于进一步的数据分析。不同批次理中丸样品的变量重要性投影值(variable importance in projection,VIP)分析结果见图6,筛选出导致样品分类影响较大的差异性成分,VIP>1.0的峰有16个:峰5、峰21、峰9、峰27、峰25、峰15、峰11、峰14、峰18、峰13、峰19、峰8、峰26、峰17、峰7、峰16,其中峰5为甘草苷、峰8为甘草素、峰9为甘草酸、峰17为白术内脂 Ⅲ、峰27为白术内脂 Ⅰ,说明这16个成分为影响样品分类差异的主要成分。
取理中丸粉末(S5)适量,按相应溶液制备方法制备对照品溶液、样品溶液以及阴性样品溶液,结果见图7~8,在最佳色谱条件下进样测定,发现各成分色谱峰均具有良好的分离度、峰形对称性,并且与相应对照品一一匹配,表明该方法专属性良好,能够准确、有效地进行成分分析和鉴定。
精密吸取各对照品溶液适量,置于10 mL量瓶内,用无水甲醇定容至刻度,摇匀,配制成质量分别为绿原酸 25.627 5 μg·mL-1、甘草苷 107.289 0 μg·mL-1、党参炔苷6.325 2 μg·mL-1、甘草素 38.080 0 μg·mL-1、甘草酸 217.235 0 μg·mL-1、白术内脂 Ⅲ 71.204 0 μg·mL-1、白术内脂Ⅱ4.821 5 μg·mL-1、白术内脂Ⅰ 22.100 0 μg·mL-1、10-姜酚 48.265 0 μg·mL-1的混合对照品溶液,逐级稀释成系列质量浓度,在最佳色谱条件下进样测定,记录峰面积(Y),并与相应的溶液浓度(X)建立标准曲线。结果见表3,r≥0.998 7,由表3可知9种成分在各自浓度范围内线性关系良好。
取“2.2.1”项下混合对照品溶液连续进样6次,测得绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内脂 Ⅲ、白术内脂 Ⅱ、白术内脂 Ⅰ、10-姜酚峰面积RSD分别为2.98%、2.16%、2.62%、2.46%、2.70%、2.84%、1.62%、2.97%、2.63%,表明仪器精密度良好。
精密称定理中丸样品粉末(S5)2 g,按相应溶液制备方法和最佳色谱条件,平行制备6份样品溶液进样,测得绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内脂 Ⅲ、白术内脂 Ⅱ、白术内脂 Ⅰ、10-姜酚峰面积RSD分别为2.79%、1.54%、1.58%、2.58%、1.85%、2.11%、2.62%、2.62%、2.96%,表明本方法具有良好的重复性。
精密称定理中丸样品粉末(S5)2 g,按相应溶液制备方法,室温下于0、2、4、8、12、24 h在最佳色谱条件进样测定,测得绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内脂 Ⅲ、白术内脂 Ⅱ、白术内脂 Ⅰ、10-姜酚峰面积RSD分别为2.50%、1.57%、2.39%、2.77%、0.98%、1.89%、2.65%、1.95%、2.62%,表明溶液在24 h 内稳定性良好。
取同一批已知含量的理中丸粉末(S5)6份,每份约1.00 g,按照对照品量与样品中含量1∶1的比例加入各对照品,按相应溶液制备方法和最佳色谱条件,测定并计算加样回收率,结果见表4
取21(S1~S21)批理中丸样品,按相应溶液的制备方法和最佳色谱条件,进样测定,计算各成分含量,结果见表5
本研究分别考察了不同流动相体系(甲醇-水,乙腈-体积分数0.1%磷酸水溶液)、不同波长(215、237[7]、254、276 nm[8])、不同流速(0.8、1.0 mL·min-1),对HPLC方法进行优化,以获得最佳的分离效果和分析性能。通过对不同提取方法(加热回流提取、超声提取)、不同提取溶剂(无水乙醇、体积分数30%甲醇、50%甲醇、70%甲醇、无水甲醇、水)和不同提取体积(10、25、50 mL)的考察,最终确定样品提取方法为25 mL体积分数70%甲醇超声处理45 min。以乙腈-水(含0.1%磷酸)为流动相,梯度洗脱,检测波长为215和254 nm,柱温30 ℃,此时各成分色谱峰分离度、峰形良好,基线平稳。
针对理中丸处方组成饮片,参考《中国药典》2020年版含量测定的指标成分结合理中丸相关研究,白术具有健脾益气、燥湿止汗等功效,化学成分主要是由白术内酯Ⅰ、白术内酯Ⅱ和白术内酯Ⅲ等构成,且白术内酯Ⅰ具有调节胃肠道作用[9],白术内酯Ⅱ的在大鼠肠道中吸收部位主要是十二指肠段,白术内酯Ⅲ在所有肠段中的吸收率最高[10];甘草中黄酮类成分(甘草素)多具有抗氧化、提高机体免疫力、保护心脑血管、抗炎止痛等作用[11],绿原酸具有抗氧化、抗菌以及治疗代谢类疾病等药理作用[12];上述药味所含的成分与理中丸的功效息息相关,故选择以上成分作为含量测定的指标成分。
本研究建立的理中丸HPLC指纹图谱,共确定了29个共有峰,与对照品比较确定了其1号峰、5号峰、6号峰、8号峰、9号峰、17号峰、23号峰、27号峰和28号峰、分别为绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内脂 Ⅲ、白术内脂 Ⅱ、白术内脂 Ⅰ、10-姜酚。21批理中丸指纹图谱相似度均在0.900以上,表明理中丸批次间一致性良好,化学成分基本一致,整体质量稳定。结合化学识别模式分析可知,21批理中丸样品可聚为3类,表明理中丸中9种成分的含量与生产厂家有关,造成明显聚类的原因可能与不同厂家生产时所用的投料药材质量差异以及生产工艺差异有关,进一步通过OPLS-DA中的VIP值可以判断出,影响各批次间差异的主要峰有16个,这可能与不同产地药材的含量差异、运输及贮存过程中发生变化有关[13]。含量测定结果中甘草酸的含量最高,其次是甘草素,同一厂家不同批次含量差异较小,不同厂家成分含量差异较大,这可能与生产过程中原材料的来源、品质、加工炮制方法以及工艺控制等因素有关[14-15]
本实验建立理中丸HPLC指纹图谱,并同时测定绿原酸、甘草苷、党参炔苷、甘草素、甘草酸、白术内脂 Ⅲ、白术内脂 Ⅱ、白术内脂 Ⅰ、10-姜酚的含量,能较好地反映药品整体质量状况,再采用化学模式分析筛选出不同批次样品之间的差异,该方法准确可靠,重复性好,为理中丸的质量控制提供参考。
