OBJECTIVE To develop a method for the determination of dracorhodin in Draconis Sanguis scald ointment based on the concept of quality by design (QbD), and to study its preparation process for quantitative transfer. METHODS Using the content of dracorhodin as key quality attribute, the key analytical parameters in preparation of test solution were selected through Plackett-Burman design. The relationship between key quality attribute and key analysis parameters was evaluated through Box-Behnken design. According to characteristics of the medicinal material, intermediate and finished product of scald ointment, the different methods for determining content of dracorhodin were established, and the law of quantity transfer was studied. The preparation conditions of test solution for content determination of Draconis Sanguis scald ointment were as follows: lanolin was removed by petroleum ether, extraction solvent was 7% phosphoric acid methanol solution, and extraction was carried out in a water bath at 60 ℃ for 30 min. The chromatographic conditions were as follows: the mobile phase was consisted of acetonitrile and 0.05 mol·L-1 sodium dihydrogen phosphate solution using a gradient elution. Temperature was 40 ℃ and flow rate was 1.0 mL·min-1, at a detector wavelength of 440 nm. RESULTS The feasibility of preparation process of Draconis Sanguis scald ointment was proven. The transfer rate of dracorhodin from medicinal material to finished product was over 90%. CONCLUSION The analytical method developed based on concept of QbD could accurately determine content of dracorhodin in Draconis Sanguis scald ointment, which is conducive to product quality evaluation and process value transfer research.

目的 基于质量源于设计(quality by design,QbD)理念研究血竭烫伤膏中血竭素含量测定方法,并对成型工艺进行量值传递研究。方法 以血竭素含量为关键质量属性,通过Plackett-Burman设计筛选供试品制备中的关键分析参数;通过Box-Behnken设计评价关键质量属性与关键分析参数间关系。针对烫伤膏药材、中间体、成品的特性,建立不同的血竭素含量测定方法,并进行量值传递研究。血竭烫伤膏含量测定中供试品制备条件:加石油醚除去羊毛脂,以7%磷酸甲醇溶液为提取溶剂,60 ℃水浴提取30 min。色谱条件:以乙腈-0.05 mol·L-1磷酸二氢钠溶液为流动相,梯度洗脱;流速1.0 mL·min-1;检测波长440 nm;柱温40 ℃。结果 通过分析可知血竭烫伤膏制备工艺可行,药材到成品过程中血竭素转移率达90%以上。结论 基于QbD理念开发的血竭烫伤膏分析方法能够准确测定血竭素含量,利于产品质量评价与工艺量值传递研究。

目的 基于质量源于设计(quality by design,QbD)理念研究血竭烫伤膏中血竭素含量测定方法,并对成型工艺进行量值传递研究。方法 以血竭素含量为关键质量属性,通过Plackett-Burman设计筛选供试品制备中的关键分析参数;通过Box-Behnken设计评价关键质量属性与关键分析参数间关系。针对烫伤膏药材、中间体、成品的特性,建立不同的血竭素含量测定方法,并进行量值传递研究。血竭烫伤膏含量测定中供试品制备条件:加石油醚除去羊毛脂,以7%磷酸甲醇溶液为提取溶剂,60 ℃水浴提取30 min。色谱条件:以乙腈-0.05 mol·L-1磷酸二氢钠溶液为流动相,梯度洗脱;流速1.0 mL·min-1;检测波长440 nm;柱温40 ℃。结果 通过分析可知血竭烫伤膏制备工艺可行,药材到成品过程中血竭素转移率达90%以上。结论 基于QbD理念开发的血竭烫伤膏分析方法能够准确测定血竭素含量,利于产品质量评价与工艺量值传递研究。

OBJECTIVE To develop a method for the determination of dracorhodin in Draconis Sanguis scald ointment based on the concept of quality by design (QbD), and to study its preparation process for quantitative transfer. METHODS Using the content of dracorhodin as key quality attribute, the key analytical parameters in preparation of test solution were selected through Plackett-Burman design. The relationship between key quality attribute and key analysis parameters was evaluated through Box-Behnken design. According to characteristics of the medicinal material, intermediate and finished product of scald ointment, the different methods for determining content of dracorhodin were established, and the law of quantity transfer was studied. The preparation conditions of test solution for content determination of Draconis Sanguis scald ointment were as follows: lanolin was removed by petroleum ether, extraction solvent was 7% phosphoric acid methanol solution, and extraction was carried out in a water bath at 60 ℃ for 30 min. The chromatographic conditions were as follows: the mobile phase was consisted of acetonitrile and 0.05 mol·L-1 sodium dihydrogen phosphate solution using a gradient elution. Temperature was 40 ℃ and flow rate was 1.0 mL·min-1, at a detector wavelength of 440 nm. RESULTS The feasibility of preparation process of Draconis Sanguis scald ointment was proven. The transfer rate of dracorhodin from medicinal material to finished product was over 90%. CONCLUSION The analytical method developed based on concept of QbD could accurately determine content of dracorhodin in Draconis Sanguis scald ointment, which is conducive to product quality evaluation and process value transfer research.

血竭烫伤膏是临床用于治疗烧烫伤的医院制剂,由血竭、没药、冰片、煅石膏等原料组成,具有活血化瘀、清热解毒、生肌敛疮等功效。由于血竭烫伤膏的生产工艺一直沿用传统方法,缺乏现代质量评价方法,限制了其发展和应用。血竭作为该处方的君药,也是传统名贵中药,对其进行质量评价有助于对血竭烫伤膏的质量评价与工艺控制[1-2]。血竭已被《中国药典》2020年版收录,以血竭素为指标成分,并建立了液相色谱含量测定方法[3]。前期实验研究发现,直接将血竭药材供试品制备方法套用于血竭烫伤膏中血竭素含量测定方法中,存在含量测定不准确问题,影响成品质量评价与工艺量值传递研究。
液相色谱分析方法的开发主要包括供试品溶液制备和色谱条件参数,大多研究倾向于色谱条件参数优化,以满足色谱法含量测定要求,但供试品溶液的制备将直接影响含量测定结果准确性。质量源于设计(quality by design,QbD)理念是先进的药品质控理念,也可指导中药及中成药分析方法的开发[4-5]。本研究将QbD理念运用于血竭烫伤膏中血竭素含量测定方法的供试品溶液制备优化中,采用筛选试验设计(Plackett-Burman design,PBD)筛选影响血竭素含量的关键分析参数,然后运用响应面设计(Box-Behnken design,BBD)建立可靠的分析方法。此外,还将针对血竭烫伤膏药材、中间体、成品特点,选择合适的分析方法进行量值传递规律的研究。
Agilent 1260型高效液相色谱仪(美国安捷伦公司);XS105型十万分之一电子天平,ME204型万分之一电子天平(德国梅特勒公司);DK-S26型恒温水浴锅(上海精宏实验设备有限公司);KQ-500型数控超声波清洗器(昆山市超声仪器有限公司);118B型摇摆式型高速中药粉碎机(瑞安市永历制药机械有限公司);MK-2270M型微波灭菌设备(青岛海尔微波炉制品有限公司);Fisco系列弗鲁克均质混合成套反应设备(上海弗鲁克流体机械制造有限公司)。
血竭药材购于绍兴震元中药饮片有限公司,产地印度尼西亚,经浙江省中药研究所有限公司周丹英正高级工程师鉴定为棕榈科植物麒麟竭(Daemonorops draco Bl.)果实渗出的树脂经加工制成;医药级羊毛脂购于天津市化学试剂三厂;血竭烫伤膏中间体、血竭烫伤膏由浙江省中药研究所有限公司自行制备。血竭素高氯酸盐对照品(批号110811-202108,中国食品药品检定研究院);甲醇、乙腈(色谱纯);甲醇、磷酸(分析纯);纯净水(杭州娃哈哈集团有限公司)。
根据《中国药典》2020年版一部血竭项下血竭素含量测定方法,乙腈-0.05 mol·L-1磷酸二氢钠的配比为50∶50时,血竭素色谱峰出现杂峰干扰,当二者配比为40∶60时能达到定量分离度要求。以十八烷基硅烷键合硅胶为填充剂(Diamonsil C18,4.6 mm×250 mm,5 μm);以乙腈(A)-0.05 mol·L-1磷酸二氢钠溶液(B)为流动相,梯度洗脱(0~20 min,40%~40A%;20~25 min,40%~80%A;25~30 min,80%~40%A);流速1.0 mL·min-1;柱温40 ℃;检测波长440 nm;理论板数按血竭素峰计算应不低于4 000。
取血竭素高氯酸盐对照品10 mg,精密称定,置50 mL棕色量瓶中,加3%磷酸甲醇溶液使溶解,并稀释至刻度,摇匀,精密量取4 mL,置25 mL棕色量瓶中,加甲醇至刻度,摇匀,即得(血竭素质量=血竭素高氯酸盐质量/1.377)。
取本品5.0 g,精密称定,置50 mL离心管中,精密加入石油醚(60~90 ℃)a(mL),b ℃水浴加热,适当搅拌使其融化,离心,弃去上清液。残渣加磷酸体积分数为c(%)的磷酸甲醇溶液d(mL)转移至圆底烧瓶,称定质量,回流时间e(min),水浴温度f ℃,冷水浴冷却,再称定质量,用磷酸甲醇溶液补足减失的质量,摇匀,静置g h,取上清液滤过,取续滤液,即得。
依据参考文献确定血竭素含量测定方法的潜在关键分析参数,先进行单因素试验,再结合文献设置关键分析参数与设置水平[3,6-8]。单因素试验结果显示磷酸甲醇体积分数为0、3%、6%时血竭素含量测定结果为0.013 1%、0.017 2%、0.019 4%,提取时间为10、20、30 min时血竭素含量测定结果为0.187%、0.198%、0.206%,提取温度为40、50、60 ℃时血竭素含量测定结果为0.185%、0.019 7%、0.020 8%。试验过程中考虑石油醚的用量与温度、磷酸甲醇溶液体积、样品制备后的测定时间也对含量测定结果有一定影响。以供试品溶液制备中石油醚体积(a)、基质水浴溶解温度(b)、磷酸甲醇溶液体积分数(c)、溶剂体积(d)、提取时间(e)、提取温度(f)、放置时间(g)为7个潜在关键分析参数,以血竭素含量(Y)为关键属性质量。
利用Design Expert 8.0软件,通过PBD试验筛选关键分析参数,其中各因素的高水平对应代码1,低水平对应代码-1。PBD试验分析因子及水平表见表1,PBD试验设计与结果见表2。数学模型为Y=0.012 472-2.89×10-5a+8.33×10-6b+4.67×10-4c+1.33×10-6d+4.00×10-5e+8.83×10-5f+5.56×10-6g。模型P<0.05,具有显著性;R2=0.981 4,${R}_{adj}^{2}$=0.948 9,回归性良好。PBD试验方差分析见表3,分析参数筛选的Pareto分析见图1。结果可知bcdefg均对关键属性质量具有正影响作用,a具有负影响作用。其中磷酸甲醇溶液体积分数(c)、提取时间(e)、提取温度(f)对关键属性质量具有显著性(P<0.05),可作为关键分析参数进一步优化。其他因素为非关键分析参数,根据实验情况定为石油醚体积为25 mL,基质水浴溶解温度40 ℃,磷酸甲醇体积50 mL,放置时间0 h。
基于PBD试验,利用Design Expert 8.0软件,通过BBD试验对磷酸甲醇溶液体积分数(A)、提取温度(B)、提取时间(C)3个关键分析参数进行优化。BBD响应面分析因子及水平表见表4,其中各因素的高、中、低水平分别对应代码1、0、-1。
血竭素含量测定的BBD试验方案及结果见表5,方差分析见表6,各因素的三维响应面图见图2。该模型回归方程Y=-0.038 38+2.37×10-3A+1.38×10-4B+1.59×10-3C+5.56×10-7AB-1.67×10-6AC-1.67×10-6BC-1.53×10-4A2-5.67×10-7B2-1.20×10-5C2。模型的显著性检验具有统计学意义,P<0.000 1;R2=0.995 4,${R}_{adj}^{2}$=0.989 5,表明模型与数据拟合程度较高。可知磷酸甲醇溶液体积分数、提取温度对烫伤膏中血竭素含量的测定具有显著性(P<0.05),而提取温度与提取时间对血竭素含量具有交互性影响。模型给出的最优解为7.44%磷酸甲醇溶液,63.38 ℃下水浴提取32.37 min,血竭素含量预测值为0.023 1%。
提取时间为不显著因素,故以30 min作为定值,建立磷酸甲醇溶液体积分数与提取温度的分析参数设计空间,以高于0.022 6%(11~15组实验的平均值)为分析目标。为提高设计空间准确性,加入置信水平α=0.05的置信区间,优化的设计空间结果通过Overlay plot展示,见图3。亮黄色部分即为优化后的设计空间,满足血竭素测定含量大于0.022 6%。其中可优选分析参数范围为提取时间30 min,磷酸甲醇溶液体积分数7%~8%,提取温度60~65 ℃,即橙色部分。
根据BBD试验最优选择以及分析参数设计空间,最终选择的供试品制备方法如下:取本品5.0 g,精密称定,置50 mL离心管中,精密加入石油醚(60~90 ℃)25 mL,40 ℃水浴加热,适当搅拌使其融化,离心,弃去上清液。残渣加体积分数为7%磷酸甲醇溶液50 mL转移至圆底烧瓶,称定质量,60 ℃水浴提取30 min,冷水浴冷却,再称定质量,用磷酸甲醇溶液补足减失的质量,摇匀,取上清液滤过,取续滤液,即得。对上述优选分析参数进行验证,血竭素含量测定结果为(0.022 9±0.004)%(n=3),与最优条件下预测值(0.023 1%)的一致性较好。
取血竭阴性样品5.0 g,照供试品溶液制备方法制备,作为阴性供试品溶液。分别精密吸取对照品溶液、供试品溶液、阴性供试品溶液注入液相色谱仪,测定。结果见图4,阴性供试品在血竭素对照品出峰处无干扰,专属性符合要求。
取血竭素高氯酸盐对照品,精密称定,用甲醇分别制成每1 mL各含95.81、47.91、23.95、11.98、5.988 μg的血竭素溶液,测定。以血竭素峰面积与浓度作线性回归处理,回归方程为y=19.731x+7.200 3,r2=0.999 9,血竭素在5.988~95.81 μg·mL-1内线性关系良好,符合要求。
取同一份对照品溶液,连续测定6次,血竭素峰面积相对标准偏差(RSD)为0.70%,精密度符合要求。平行制备6份供试品溶液,测定,血竭素含量RSD为1.35%,重复性符合要求。取同一份供试品溶液,分别于0、2、4、6、8、12、24 h测定,血竭素12 h稳定性RSD为2.19%,稳定性符合要求。24 h稳定性RSD为4.31%,应现配现测。
取本品6份,每份约2.5 g,精密称定,分别精密加入血竭素高氯酸盐对照品,制备供试品溶液,测定见表7。血竭素加样回收率均在99.7%~106.0%之间,RSD为2.17%,符合90%~108%要求。
称取三批样品各2份,每份约5 g,精密称定,按“2.3.4”项下方法制备,注入液相色谱仪,测定。结果显示,3批供试品中血竭素含量为0.022 8%、0.022 1%、0.022 6%,一致性较好。
按处方量将血竭等药材混合,粉碎成细粉(中间体1),微波灭菌(中间体2),备用。取适量羊毛脂等基质加热混合均匀,保温,加入药粉,混合搅拌,放冷,即得成品。
药材供试品溶液制备:取血竭药材适量,研细,取0.1 g,精密称定,置具塞锥形瓶中,精密加入体积分数为3%磷酸甲醇溶液10 mL,密塞,称定质量,60 ℃水浴提取30 min,放冷,再称定质量,用3%磷酸甲醇溶液补足减失的质量,摇匀,滤过,测定。
中间体供试品溶液制备:取中间体1.5 g,精密称定,置具塞锥形瓶中,精密加入体积分数为7%磷酸甲醇溶液50 mL,密塞,称定质量,60 ℃水浴提取30 min,放冷,再称定质量,用7%磷酸甲醇溶液补足减失的质量,摇匀,滤过,测定。
成品供试品溶液制备:按BBD试验优化所建立“2.3.4”项下方法。
比较不同工艺点样品中血竭素含量的变化情况,药材、中间体1、中间体2、成品的量值传递研究结果见图5与见表8。工艺过程中血竭素损失率为粉碎环节>微波灭菌环节>混合搅拌环节,但损失率均小于6%。血竭素含量降低的主要原因在于温度控制,为减少损失,粉碎工艺在满足粒度的情况下多次粉碎,减少持续粉碎时间;微波灭菌工艺药粉铺薄,短时间多次加热,尽量避免局部过热;混合搅拌温度控制在60 ℃,搅拌时间小于30 min。三批成品血竭素含量的平均值为0.022 5%,较理论值0.024 3%下降7.4%。
随着QbD理念在制药工艺中的成功应用,将分析方法作为一个工艺环节,在QbD理念指导下开发并优化分析参数范围,求取多维设计空间,已成为新研究热点[9-12]。血竭烫伤膏中血竭素含量测定方法的关键在于供试品制备方法优化,由于辅料的加入以及血竭素不稳定的性质,供试品制备影响因素较多。本研究首先以血竭素含量为分析方法的关键质量属性,通过文献、经验、结合单因素试验、PBD试验筛选出分析方法中供试品制备的关键分析参数为磷酸甲醇溶液体积分数、提取温度、提取时间。再通过BBD试验建立关键分析参数和关键质量属性间的二次多项式模型,获得最优关键分析参数为提取时间32.37 min,磷酸甲醇溶液7.44%,提取温度63.38 ℃;设计空间为提取时间30 min,磷酸甲醇溶液体积分数7%~8%,提取温度60~65 ℃。最终选定的参数:提取时间为30 min,磷酸甲醇溶液为7%,提取温度为60 ℃,血竭素含量测定结果为(0.022 9±0.004)%。
血竭中的主要成分为血竭素、血竭红素、去甲基血竭素及血竭黄烷等化学成分[13]。其中血竭红素作为“结合型血竭素”,酸性环境下可发生降解,逐渐转化为血竭素[7]。用酸化方法测定的血竭素含量偏高,实际是血竭红素与血竭素的总含量[14]。《中国药典》2020年版收载的血竭药材中血竭素含量测定方法[3],测得血竭药材的血竭素含量为0.841%~1.043%,与研细程度与振摇力度有关;后考察3%磷酸甲醇,室温超声30 min,测定结果为1.096%;3%磷酸甲醇,60 ℃回流30 min,测定结果为1.215%。通过《中国药典》2020年版方法测定血竭素含量偏低,与文献报道相符。Hu等[6],Li等[7]均研究发现依据《中国药典》2020年版方法测定的数据不稳定,且偏低。通过单因素考察磷酸甲醇溶液体积分数、回流时间、回流温度,确定药材中血竭素含量测定方法为3%磷酸甲醇,60 ℃回流30 min。
研究过程中发现若中间体直接按药材方法测定,则血竭素转移率与理论情况不相符,推测是中间体粉末有煅石膏等成分,会降低提取液酸强度,影响血竭素稳定性,导致测定结果偏低。考察了3、5、7、9%磷酸甲醇体积分数的影响,确定中间体中血竭素含量测定方法为7%磷酸甲醇,60 ℃回流30 min。制剂中含有羊毛脂等辅料,由于羊毛脂在甲醇中具有一定溶解度,若按中间体方法直接提取,滤液过微孔滤膜后放置一段时间便有羊毛脂析出,进行液相分析会对仪器和色谱柱造成较大损伤。根据羊毛脂和血竭素的溶解特性,考察了石油醚(30~60 ℃)、石油醚(60~90 ℃)、正己烷、二氯甲烷,结果显示石油醚(30~60 ℃)、石油醚(60~90 ℃)、正己烷均能除去羊毛脂,保留血竭素,含量测定结果基本一致;二氯甲烷能除去羊毛脂,但会溶解血竭素。考虑安全性,故确定石油醚(60~90 ℃)为除羊毛脂的溶剂。
血竭烫伤膏的质量研究从内容上可以分为药材质量标准、过程质量标准和产品质量标准。其中,药材质量标准是良好质量的源头,过程质量标准是良好质量的保证,产品质量标准是良好质量的最终体现。若量值传递研究药材、中间体、成品的质量分析方法共用一套含量测定方法,所测结果未必真实体现血竭烫伤膏工艺过程引起的血竭素变化规律。本研究依据样品的特性,建立了血竭烫伤膏各环节相应的含量测定方法,避免分析方法的误差影响工艺量值传递的判断。依据《浙江省医疗机构应用传统工艺配制中药制剂备案管理实施细则》中提出原粉入药的转移率一般要求在90%以上,本研究三批产品均满足此指导要求,说明现有的血竭烫伤膏制备工艺是可行、稳定的[15]
本研究运用QbD理念建立血竭烫伤膏中血竭素含量测定方法,引入PPD和BBD试验设计,科学筛选关键分析参数,提升分析方法开发过程的结构化和系统化。并通过血竭素量值传递研究明确血竭等原料药材在被加工制成血竭烫伤膏过程中质量变化,实现工艺的稳定性,从而进一步提升血竭烫伤膏的药效质量。
