OBJECTIVE To establish the HPLC fingerprint and multi-component quantitative determination method of Glehniae Radix, and study the dispersion in quality of Glehniae Radix of different regions and residual root bark ratio by chemical pattern recognition. METHODS HPLC method was used to establish the fingerprint and determine the contents of seven active components. The chromatographic column was ShimNex CS C18(4.6 mm×250 mm, 5 μm), the mobile phase was methanol-0.4% phosphoric acid water, gradient elution was conducted at a volume flow rate of 1.0 mL·min-1, the column temperature was maintained at 30 ℃, and the detection wavelength was set at 204 and 248 nm. Fingerprint similarity was used to evaluate different regions of Glehniae Radix. Cluster analysis, principal component analysis and discriminant analysis were used to analyze the quality and the differential components of Glehniae Radix of different residual root bark ratio. RESULTS A total of 18 common peaks were found in the fingerprints of 60 batches of Glehniae Radix fingerprints from different regions, and seven peaks were identified, including facarindiol, panaxynol, xanthotoxin, bergapten, imperatorin, isocorallol and isoimperatorin. The similarity of 60 batches of samples was 0.814-0.996. The multi-component contents of the 190 batches of Glehniae Radix were divided into two groups by cluster analysis, and the samples with residual root bark ratio ≤1/3 and >1/3 were significantly distinguished. The results of PCA and HCA were consistent. The different quality markers were panaxynol and facalindiol. CONCLUSION The established HPLC fingerprint and multi-component content determination method are simple, stable, and reliable, which can provide a basis for the quality control, overall evaluation and standardized processing methods of Glehniae Radix.

目的 建立北沙参高效液相色谱(HPLC)指纹图谱及多成分定量方法,结合化学模式识别,评价不同产地、不同带皮程度对北沙参质量的影响。方法 采用HPLC法建立北沙参指纹图谱与7种指标成分含量的测定方法。采用指纹图谱相似度评价不同产地北沙参质量,应用聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘法(OPLS-DA)评价不同带皮程度北沙参质量,进行差异性成分分析。结果 60批不同产地北沙参指纹图谱共有18个峰,指认了7个峰,各批次相似度在0.814~0.996。聚类分析将190批北沙参分为2类,显著区分残皮程度≤1/3及>1/3的样品,PCA与HCA结果一致,差异性质量标志物为人参炔醇、法卡林二醇。结论 建立的HPLC指纹图谱和多成分含量测定方法操作简便,稳定可靠,可为北沙参的质量控制、整体评价及规范产地加工方式提供依据。

目的 建立北沙参高效液相色谱(HPLC)指纹图谱及多成分定量方法,结合化学模式识别,评价不同产地、不同带皮程度对北沙参质量的影响。方法 采用HPLC法建立北沙参指纹图谱与7种指标成分含量的测定方法。采用指纹图谱相似度评价不同产地北沙参质量,应用聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘法(OPLS-DA)评价不同带皮程度北沙参质量,进行差异性成分分析。结果 60批不同产地北沙参指纹图谱共有18个峰,指认了7个峰,各批次相似度在0.814~0.996。聚类分析将190批北沙参分为2类,显著区分残皮程度≤1/3及>1/3的样品,PCA与HCA结果一致,差异性质量标志物为人参炔醇、法卡林二醇。结论 建立的HPLC指纹图谱和多成分含量测定方法操作简便,稳定可靠,可为北沙参的质量控制、整体评价及规范产地加工方式提供依据。

OBJECTIVE To establish the HPLC fingerprint and multi-component quantitative determination method of Glehniae Radix, and study the dispersion in quality of Glehniae Radix of different regions and residual root bark ratio by chemical pattern recognition. METHODS HPLC method was used to establish the fingerprint and determine the contents of seven active components. The chromatographic column was ShimNex CS C18(4.6 mm×250 mm, 5 μm), the mobile phase was methanol-0.4% phosphoric acid water, gradient elution was conducted at a volume flow rate of 1.0 mL·min-1, the column temperature was maintained at 30 ℃, and the detection wavelength was set at 204 and 248 nm. Fingerprint similarity was used to evaluate different regions of Glehniae Radix. Cluster analysis, principal component analysis and discriminant analysis were used to analyze the quality and the differential components of Glehniae Radix of different residual root bark ratio. RESULTS A total of 18 common peaks were found in the fingerprints of 60 batches of Glehniae Radix fingerprints from different regions, and seven peaks were identified, including facarindiol, panaxynol, xanthotoxin, bergapten, imperatorin, isocorallol and isoimperatorin. The similarity of 60 batches of samples was 0.814-0.996. The multi-component contents of the 190 batches of Glehniae Radix were divided into two groups by cluster analysis, and the samples with residual root bark ratio ≤1/3 and >1/3 were significantly distinguished. The results of PCA and HCA were consistent. The different quality markers were panaxynol and facalindiol. CONCLUSION The established HPLC fingerprint and multi-component content determination method are simple, stable, and reliable, which can provide a basis for the quality control, overall evaluation and standardized processing methods of Glehniae Radix.

北沙参为伞形科植物珊瑚菜(Glehnia littoralis Fr.Schmidt ex Miq.)的干燥根。味甘、微苦,具有养阴清肺、益胃生津的功效,用于治疗肺热燥咳,劳嗽痰血,热病津伤、咽干口渴等症[1]。《神农本草经》列其为上品,为我国常用大宗中药材之一[2]。北沙参主要化学成分为香豆素类、木脂素类、聚炔类,其次有挥发油、多糖、氨基酸等[3-6]。香豆素类化合物具有抗炎、抗菌、镇痛等作用,聚炔类化合物具有提高免疫、抗菌等作用[7-11],为北沙参的主要功效成分。山东莱阳、河北安国与内蒙古赤峰为北沙参主产地,辽宁、江苏、浙江、广东、福建等地也有分布[12-13],文献报道不同产地间北沙参存在质量差异,但研究尚不完善[13-15]。《中国药典》2020年版一部规定,北沙参有两种产地加工方式,其一为除去须根,洗净,稍晾,置沸水中烫后,除去外皮,干燥;其二为洗净直接干燥[1]。为使北沙参外形美观、易干燥、不易生虫等,市场流通的北沙参多为去皮后干燥[13,16]。同时,北沙参作为常用蒙药(查干-扫日劳),传统蒙医为不去皮使用,认为去皮会影响其止咳效果[17]。因此,亟须探究不同产地、加工方式对北沙参主要功效成分构成及含量的影响。
北沙参现行质量标准收载于《中国药典》2020年版,仅规定了性状、鉴别项,缺少有效的质量控制手段。文献也仅测定单一类别成分含量,无法全面评价北沙参产地间、加工方式间的质量差异。本研究建立了北沙参高效液相色谱(HPLC)指纹图谱方法,并可同时测定北沙参中主要的2种聚炔类成分(法卡林二醇、人参炔醇)[5]、5种香豆素类成分(花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素)[11]的含量,以此评价北沙参不同产地间质量差异,并考察不同加工方式对质量的影响,归纳北沙参关键质量评价指标,为评价流通领域北沙参整体质量状况提供基础数据,为优化加工方式、完善质量评价方法提供理论依据。
UltiMate3000超高效液相色谱仪、Orbitrap elite轨道离子阱质谱仪(Thermo Fisher公司,美国);Waters e2695高效液相色谱仪(Waters公司,美国);LC-20A高效液相色谱仪(SHIMADZU公司,日本);Agilent 1260高效液相色谱仪(Agilent公司,美国);XPE26电子天平(Mettler Toldeo公司,瑞士);AE240电子天平(Mettler Toldeo公司,瑞士);Milli-Q Integral 10型超纯水仪(Millipore公司,美国)。
对照品:法卡林二醇(批号:A02GB143892,含量≥98%)、人参炔醇(批号:A01GB143895,含量≥70%)(上海源叶生物);花椒毒素(批号:DST210407-062,含量≥98%)、佛手柑内酯(批号:DST210409-012,含量≥98%)、异珊瑚菜素(批号:DST210322-158,含量≥98%)(上海德斯特生物);欧前胡素(批号:110826-201616,含量以99.6%计)、异欧前胡素(批号:110827-201611,含量以99.4%计)(中国食品药品检定研究院)。
甲醇为色谱纯;水为实验室自制超纯水;其他试剂均为分析纯。
190批次北沙参样品,覆盖了河北、山东、内蒙古3个主产地及其他产地,样品信息见表1,均经河北省药品医疗器械检验研究院段吉平主任药师鉴定为伞形科植物珊瑚菜(Glehnia littoralis Fr. Schmidt ex Miq.)的干燥根。收集的北沙参样本以带少量残皮的居多,且均一性较高,少部分为带有较多残皮的样本,未见不去皮样本。为了便于分析差异,将北沙参按残留皮比例划分为2个等级(表2),示例图见图1
以十八烷基硅烷键合硅胶为填充剂;流动相为甲醇(A)-0.4%磷酸水(B);洗脱梯度:0~15 min,65%~90% A;15~30 min,90%~92% A;流速1.0 mL·min-1;柱温30 ℃;检测波长248 nm。理论板数按法卡林二醇峰计算应不低于20 000。
取对照品适量,加甲醇分别制成每1 mL含法卡林二醇、人参炔醇1.4 mg、含花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素100 μg的溶液,作为各对照品储备液。取上述各对照品储备液适量,制成每1 mL含法卡林二醇、人参炔醇各50 μg,含花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素各2 μg的混合对照品溶液,即得。
取北沙参粉末(过四号筛)约2 g,精密称定,置具塞锥形瓶中,精密加入甲醇25 mL,称定质量,超声处理(功率400 W,频率40 kHz)40 min,放冷,再称定质量,用甲醇补足减失的质量,摇匀,滤过,取续滤液,即得。
取北沙参粉末(过四号筛)约2 g,精密称定,制备供试品溶液,重复进样6次,选择峰形对称且影响值较大的6号法卡林二醇峰为参照峰,测得各共有峰的相对保留时间相对标准偏差(RSD)均<2%,相对峰面积RSD均<3%,表明仪器精密度良好。
取北沙参粉末(过四号筛)约2 g,精密称定,平行取样9份,制备供试品溶液,测定,以6号法卡林二醇峰为参照峰,测得各共有峰的相对保留时间RSD均<2%,相对峰面积RSD均<3%,表明此方法重复性良好。
取供试品溶液,分别于0、3、6、9、12、18、24 h测定,以6号法卡林二醇峰为参照峰,测得各共有峰的相对保留时间RSD均<2%,相对峰面积RSD均<3%,表明供试品溶液在24 h内稳定性良好。
取混合对照品溶液及各批次供试品溶液,测定,记录色谱图,并导入《中药色谱指纹图谱相似度评价系统(2012 版)》,采用中位数法和多点校正,设置时间窗宽度为0.1 min,进行匹配,生成对照指纹图谱,并进行相似度计算。
对照品溶液制备、供试品溶液制备、除检测波长外色谱条件同指纹图谱法。在200~400 nm进行光谱扫描,法卡林二醇、人参炔醇均在204 nm波长附近有最大吸收,且与杂质峰分离良好;花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素均在248 nm波长附近有最大吸收,兼顾方法简便与各成分高灵敏度,采用同一色谱条件,204及248 nm双波长测定,同时完成7个成分的含量测定。
取“2.1.2”项下各对照品储备液,加甲醇制成每1 mL含法卡林二醇、人参炔醇各280 μg,含花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素各20 μg的混合溶液,用甲醇稀释为系列浓度并测定。以对照品质量浓度为横坐标(X),相应峰面积为纵坐标(Y),绘制标准曲线(表3)。
精密吸取“2.2.2”项下对照品溶液10 μL,连续测定6次,法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素峰面积RSD分别为1.0%、0.4%、0.6%、0.4%、0.2%、0.4%、0.5%,表明仪器精密度良好。
取同批样品(批号C20111705),按取样量1、2、3 g分别各取样3份,按“2.2.3”项下方法制备供试品溶液,测定。测得法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素的含量平均值分别为0.68、0.18、6.91、2.13、4.38、1.62、4.49 μg·g-1,RSD值分别为0.7%、0.9%、1.2%、1.5%、0.8%、0.7%、0.7%,表明方法重复性良好。
称取已知含量的北沙参样品(批号C20111705)9份,0.5、1、1.5 g各3份,精密称定,按对照品与样品含量比1∶1加入法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素对照品,制备供试品溶液并测定,计算回收率。结果显示,法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素的平均回收率分别为102.5%、100.6%、97.0%、100.6%、97.1%、99.8%、98.1%,RSD值分别为1.3%、1.8%、1.1%、0.5%、1.6%、1.2%、1.0%。表明方法准确度良好。
取同一供试品溶液(批号C20111705),分别于0、3、6、9、12、18、24 h进样测定, 测得法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素峰面积,RSD值分别为0.7%、0.4%、1.3%、1.9%、1.3%、1.5%、1.9%,表明供试品溶液在24 h内稳定。
分别称取各批次北沙参样品粉末约2 g,精密称定,制备供试品溶液,测定,以外标一点法分别计算各样品中法卡林二醇、人参炔醇、花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素的含量。
为最大限度避免不同带皮程度影响产地差异分析,从均一性更高的残皮≤1/3的样品中选取河北、内蒙古、山东与非主产地样品色谱图各15批,生成对照指纹图谱,并规定了18个指纹峰,指纹图谱见图2。采用液相色谱-质谱联用结合对照品比对法,共指认出2、4、5、8、9、11、16号共有峰,分别为花椒毒素、佛手柑内酯、欧前胡素、异珊瑚菜素、异欧前胡素、法卡林二醇和人参炔醇,结果见表4,其中与佛手柑内酯、异珊瑚菜素、异欧前胡素对应的色谱峰在部分样品中缺失,缺失批次分别为4批、8批与1批,样品涉及各个产地。
60批不同产地样品相似度在0.814~0.996之间,其中53批相似度大于0.900,7批相似度在0.814~0.899之间,分别为内蒙古产区样品1批、山东产区样品3批、非主产区样品3批,河北产区样品相似度均高于0.90,且样品间相对差异最小。
北沙参7个指标成分含量结果见表5。190批样品中4批未检测到花椒毒素,27批未检测到佛手柑内酯,36批未检测到异珊瑚菜素,5批未检测到异欧前胡素,均为残皮≤1/3的样品。
将190批样品的7个指标成分含量测定结果导入SIMCA14.1软件进行HCA,结果(图3)显示,残皮程度为1(残皮≤1/3)与残皮程度为2(1/3<残皮≤2/3)等级的北沙参分别聚为一类。
采用SIMCA14.1软件对190批北沙参样品数据进行主成分分析,以7个指标成分的含量为变量,由于各成分含量存在一定差异,将各成分量乘以相应系数进行修正,具体系数:法卡林二醇为1、人参炔醇为1、花椒毒素为50、佛手柑内酯为100、欧前胡素为100、异珊瑚菜素为100、异欧前胡素为100。通过主成分分析图(图4),可发现残皮程度为1的样品在原点左侧聚集,残皮程度为2的样品在原点右侧聚集,与聚类分析结果一致。
在PCA的基础上,利用OPLS-DA进一步对不同带皮程度的北沙参样品进行分析,并寻找差异性成分,结果见图5。OPLS-DA模型图中残皮程度为1的样品基本分布在中线右侧,残皮程度为2的样品分布在中线左侧,表明190批样品被较好地分为2类。1批样品游离于中心椭圆外,与其他样品不聚集,分析原因为北沙参样品带皮程度划分方式为目测,存在一定误差。R2X值为1.00,R2Y值为0.87,Q2值为0.86,数值均大于0.5,证明模型的解释率和预测能力良好。
通过置换次数为200次的置换检验,R2的截距为-0.0172,小于0.4,Q2的截距为-0.105,小于0.05,所有R2Q2的左边值均小于最右边数值,证明没有过拟合现象,可用于判断样品间的差异。
以变量重要性投影值(VIP)>1为标准筛选差异标志物,按影响程度排序为人参炔醇、法卡林二醇。
在对北沙参各产地指纹图谱比对基础上,为排除不同带皮程度北沙参对含量结果的交互影响,筛选出带皮程度为1的样品,比较产地间指纹图谱相似度,3个主产地与非主产地样品平均相似度均高于0.9,其中河北产地的样品相似度均在0.9以上,山东、内蒙古与主产地的样品部分批次相似度低于0.9,质量一致性相比河北产地略低。各产地均有部分批次样品存在缺失色谱峰现象,证明峰缺失现象主要与带皮程度相关,与产地相关性较小。结果表明,排除带皮程度影响的不同产地北沙参样品无显著差异,且河北产区北沙参质量一致性高。
现行质量标准中北沙参产地加工分为去皮后干燥与直接干燥两种,市场中未见直接干燥的北沙参,且北沙参表面不平整,去皮时不易完全去除,多带有比例不同的残皮,外观差异显著。北沙参指标成分含量结果显示,部分残皮≤1/3的样品存在成分缺失现象,与指纹图谱结果相对应。结合化学模式识别方法,聚类分析可明显区分不同带皮程度的北沙参样品,主成分分析结果与聚类分析结果一致。OPLS-DA可将残皮程度≤1/3与>1/3的样品明显区分,表明带皮程度对北沙参主要成分含量影响显著。并通过VIP值筛选出两个显著性差异标志物人参炔醇和法卡林二醇,可作为北沙参质量评价指标。
本研究通过HPLC指纹图谱,明确不同产地间北沙参质量无明显差异,河北产区北沙参质量一致性最高。并以7种指标成分的含量探究不同带皮程度对北沙参质量的影响,残皮较少与较多的北沙参质量差异显著,并筛选出两个指标性差异成分,为规范北沙参产地加工方式提供了依据,并为后续北沙参质量评价研究提供了技术支持。因本次样品收集于全国流通市场,未收集到不去皮北沙参,且带皮程度为目测划分,拟在后续研究中采用同一批样品自制不同加工方式的北沙参样品,深入研究加工方式与北沙参药效的内在关系。
