OBJECTIVE To establish an UHPLC-MS/MS method for the determination of six alkaloids in Tianma capsule. METHODS Phenomenex Kinetex C18 (2.1 mm×100 mm, 2.6 μm) column was used for separation. The mobile phase consisted of methanol and 0.1% formic acid (containing 2.5 mmol·L-1 ammonium acetate) solution (gradient elution). The flow rate was 0.3 mL·min-1 and the column temperature was maintained at 35 ℃. Electrospray positive ion source was used for mass spectrometry detection by multiple reaction monitoring mode. RESULTS The liner range of calibration curve was 79.38-3 969 ng·mL-1 (r=0.999 9), 18.60-929.8 ng·mL-1 (r=0.999 8), 38.20-1 910 ng·mL-1 (r=0.999 9), 1.264-252.8 ng·mL-1(r=0.999 3), 1.196-239.2 ng·mL-1(r=1.000 0) and 1.268-253.6 ng·mL-1(r=1.000 0)for benzoylmesaconine, benzoylaconine, benzoylhypaconine, mesaconitine, hypaconitine and aconitine. The RSD of repeatability test (content) was 0.5%-3.2%. The average recoveries was in the range of 98.3%-105.3%. RSD was 1.0%-3.4%. CONCLUSION The method has good sprcificity, sensitivity and accuracy, and it is suitable for the analysis of six alkaloids in Tianma capsule.

目的 建立超高效液相色谱串联质谱(UHPLC-MS/MS)法同时测定天麻胶囊中苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱6种生物碱的含量。方法 采用Phenomenex Kinetex C18色谱柱(2.1 mm×100 mm, 2.6 μm);流动相为甲醇和体积分数0.1%甲酸水(含2.5 mmol·L-1乙酸铵)溶液,梯度洗脱,流速为0.3 mL·min-1,柱温为35 ℃;采用电喷雾离子源,以多反应离子监测模式进行质谱检测。结果 苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱分别在79.38~3 969 ng·mL-1(r=0.999 9)、18.60~929.8 ng·mL-1(r=0.999 8)、38.20~1 910 ng·mL-1(r=0.999 9)、1.264~252.8 ng·mL-1(r=0.999 3)、1.196~239.2 ng·mL-1(r=1.000 0)和1.268~253.6 ng·mL-1(r=1.000 0)内有良好的线性关系。重复性试验结果,各化合物含量RSD为0.5%~3.2%,平均回收率为98.3%~105.3%,RSD为1.0%~3.4%。。结论 该方法专属性好,灵敏度高,准确可靠耐用,可用于天麻胶囊中6种生物碱的含量测定。

目的 建立超高效液相色谱串联质谱(UHPLC-MS/MS)法同时测定天麻胶囊中苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱6种生物碱的含量。方法 采用Phenomenex Kinetex C18色谱柱(2.1 mm×100 mm, 2.6 μm);流动相为甲醇和体积分数0.1%甲酸水(含2.5 mmol·L-1乙酸铵)溶液,梯度洗脱,流速为0.3 mL·min-1,柱温为35 ℃;采用电喷雾离子源,以多反应离子监测模式进行质谱检测。结果 苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱分别在79.38~3 969 ng·mL-1(r=0.999 9)、18.60~929.8 ng·mL-1(r=0.999 8)、38.20~1 910 ng·mL-1(r=0.999 9)、1.264~252.8 ng·mL-1(r=0.999 3)、1.196~239.2 ng·mL-1(r=1.000 0)和1.268~253.6 ng·mL-1(r=1.000 0)内有良好的线性关系。重复性试验结果,各化合物含量RSD为0.5%~3.2%,平均回收率为98.3%~105.3%,RSD为1.0%~3.4%。。结论 该方法专属性好,灵敏度高,准确可靠耐用,可用于天麻胶囊中6种生物碱的含量测定。

OBJECTIVE To establish an UHPLC-MS/MS method for the determination of six alkaloids in Tianma capsule. METHODS Phenomenex Kinetex C18 (2.1 mm×100 mm, 2.6 μm) column was used for separation. The mobile phase consisted of methanol and 0.1% formic acid (containing 2.5 mmol·L-1 ammonium acetate) solution (gradient elution). The flow rate was 0.3 mL·min-1 and the column temperature was maintained at 35 ℃. Electrospray positive ion source was used for mass spectrometry detection by multiple reaction monitoring mode. RESULTS The liner range of calibration curve was 79.38-3 969 ng·mL-1 (r=0.999 9), 18.60-929.8 ng·mL-1 (r=0.999 8), 38.20-1 910 ng·mL-1 (r=0.999 9), 1.264-252.8 ng·mL-1(r=0.999 3), 1.196-239.2 ng·mL-1(r=1.000 0) and 1.268-253.6 ng·mL-1(r=1.000 0)for benzoylmesaconine, benzoylaconine, benzoylhypaconine, mesaconitine, hypaconitine and aconitine. The RSD of repeatability test (content) was 0.5%-3.2%. The average recoveries was in the range of 98.3%-105.3%. RSD was 1.0%-3.4%. CONCLUSION The method has good sprcificity, sensitivity and accuracy, and it is suitable for the analysis of six alkaloids in Tianma capsule.

天麻胶囊由天麻、羌活、独活、杜仲、牛膝、粉萆薢、附子(制)、当归、地黄和玄参共10味药材制成 [1]。其功能为祛风除湿,舒筋通络,活血止痛。临床主要用于肢体拘挛,手足麻木,腰腿酸痛。天麻胶囊目前执行标准为国家食品药品监督管理局国家药品标准,标准中仅对天麻、羌活、独活和当归进行了鉴别,对其他药味未进行质量控制。天麻胶囊处方中附子(制)具有散寒止痛的作用,附子(制)中同时含有发挥药效作用的单酯型生物碱和具有毒性的双酯型生物碱,该类成分如果控制不当,则严重影响天麻胶囊的有效性以及安全性。
《中国药典》2020版一部[2]中,采用高效液相色谱法对附子中的单酯型生物碱和双酯型生物碱进行测定,但是该方法样品处理较繁琐,分析时间长,且因天麻胶囊中附子生药量较低,灵敏度无法达到要求。文献中关于附子中生物碱含量测定的报道已有不少[3-8],但是样品前处理均较复杂。本研究建立了基于超高效液相色谱串联质谱(UHPLC-MS/MS)的含量测定方法,对天麻胶囊中的苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱进行测定。该方法样品前处理简单、灵敏度高。同时对样品中生物碱的含量限度进行了探讨,以期对天麻胶囊的有效性及安全性进行有效控制。
LCMS-8050型超高效液相色谱-串联四级杆质谱仪(日本Shimadzu公司);Milli-Q型超纯水仪(美国Millipore公司);XPE205型电子天平(北京赛多利斯公司)。
苯甲酰新乌头原碱(批号:111795-201805,含量93.1%)、苯甲酰乌头原碱(批号:111794-202006,含量99.5%)、苯甲酰次乌头原碱(批号:111796-201906,含量97.2%)、乌头双酯型生物碱对照提取物(批号:112029-201601,新乌头碱含量31.7%,次乌头碱含量30.0%,乌头碱含量31.8%)(中国食品药品检定研究院);甲醇为分析纯;流动相用甲醇和甲酸均为色谱纯;乙酸铵为色谱纯;实验用水为Milli-Q系统制备。
天麻胶囊:共收集到来自23家生产企业的101批样品,企业名称按自编号A1~A23命名。
天麻胶囊附子(制)缺味样品:企业提供的按照生产工艺生产的缺附子天麻胶囊样品。
取苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱和乌头双酯型生物碱对照提取物适量,精密称定,加体积分数50%甲醇溶液(含0.1 mol·L-1盐酸)分别制成每1 mL含苯甲酰新乌头原碱0.8 μg、苯甲酰乌头原碱0.2 μg、苯甲酰次乌头原碱0.4 μg、新乌头碱、次乌头碱和乌头碱各0.2 μg的混合对照品溶液。取苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱和乌头双酯型生物碱对照提取物适量,精密称定,加体积分数50%甲醇溶液(含0.1 mol·L-1盐酸)分别制成苯甲酰新乌头原碱质量浓度为79.38~3 969 ng·mL-1、苯甲酰乌头原碱质量浓度为18.60~929.8 ng·mL-1、苯甲酰次乌头原碱质量浓度为38.20~1 910 ng·mL-1、新乌头碱质量浓度为2.045~204.5 ng·mL-1、次乌头碱质量浓度为1.935~193.5 ng·mL-1和乌头碱质量浓度为2.051~205.1 ng·mL-1的系列溶液。
取本品2粒内容物,研细,精密称定,置具塞锥形瓶中,精密加入体积分数50%甲醇溶液(含0.1 mol·L-1盐酸)50 mL,称定质量,超声处理30 min,放冷,用体积分数50%甲醇溶液(含0.1 mol·L-1盐酸)补足减失的质量,摇匀,滤过,取续滤液,即得。
Phenomenex Kinetex C18色谱柱(2.1 mm×100 mm,2.6 μm);流动相:甲醇(A)-0.1%甲酸水溶液(含2.5 mmol·L-1的乙酸铵),梯度洗脱(0~12 min,33%A,12~25 min,33%→45%A),流速为0.3 mL·min-1,柱温为35 ℃。
采用电喷雾离子源(ESI),正离子模式;扫描模式为多反应离子监测(MRM);雾化气流量3.0 L·min-1;加热气流量6.0 L·min-1;接口温度300 ℃;接口电压4.0 kV;DL管温度300 ℃;加热块温度400 ℃;干燥气流量14 L·min-1;各化合物检测离子对及相关参数设置见表1
考察了天麻胶囊中其他药味的干扰。取天麻胶囊缺味样品,按照“2.1.2”项下方法制备供试品溶液,按照“2.2”项下分析条件进行分析。结果样品中其他药味对各生物碱测定无干扰,方法专属性良好,结果见图1
取“2.1.1”项下配制的系列浓度溶液,按照“2.2”项下方法进样分析,记录各化合物峰面积,以进样量为横坐标X,峰面积为纵坐标Y,绘制标准曲线,进行线性回归,计算回归方程及相关系数(r)。结果苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱分别在79.38~3 969、18.60~929.8、38.20~1 910、1.264~252.8、1.196~239.2和1.268~253.6 ng·mL-1内呈良好的线性关系。各化合物线性方程及相关系数见表2
取“2.1.1”项下混合对照品溶液,连续进样6次,计算苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱和乌头碱峰面积相对标准偏差(RSD),结果各化合物峰面积RSD分别为0.6%、0.6%、1.4%、0.9%、1.5%和1.6%,表明方法精密度良好。
取天麻胶囊,按“2.1.2”项下方法,平行制备样品6份。结果天麻胶囊中苯甲酰新乌头原碱的含量为50.97 μg·g-1,苯甲酰乌头原碱含量为9.07 μg·g-1,苯甲酰次乌头原碱的含量为77.30 μg·g-1,新乌头碱含量为0.46 μg·g-1,次乌头碱含量为4.09 μg·g-1,乌头碱含量为0.28 μg·g-1。各化合物RSD均小于3.3%,表明方法重复性良好。
采用加样回收率的方法。根据重复性测定结果,取天麻胶囊,分别加入苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱和乌头双酯型生物碱对照品溶液适量,按“2.1.2”项下方法,平行制备样品6份,计算回收率。结果天麻胶囊中各化合物平均回收率在98.3%~105.3%,RSD小于3.4%。表明方法准确性良好,见表3
取同一份天麻胶囊供试品溶液,分别在制备0、3、6、9、12、16、18、24 h时进样分析,计算苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱在各时间点峰面积的RSD。RSD分别为1.8%、1.3%、2.9%、2.6%,3.4%、3.6%。结果表明,本品在24 h内稳定。
本实验仅对作为限度检查的新乌头碱、次乌头碱和乌头碱进行检测限及定量限的考察。取线性最低浓度点对照品溶液进样分析,记录各化合物信噪比,以信噪比为3和10时相应的对照品溶液浓度计算检测限及定量限,结果见表4
取天麻胶囊样品,按“2.1.2”项下方法制成供试品溶液,并按“2.2”项下色谱条件测定含量,结果见表5。《中国药典》2020年版一部附子项下,生物碱的含量限度分别以单酯型生物碱总量和双酯型生物碱的总量制定,故本实验最终按照单酯型生物碱总量和双酯型生物碱总量进行统计分析。单酯型生物碱总量为苯甲酰新乌头原碱、苯甲酰乌头原碱和苯甲酰次乌头原碱含量总和,双酯型生物碱总量为新乌头碱、次乌头碱和乌头碱的含量总和。
采用SPSS软件对数据进行统计,绘制天麻胶囊中单酯型生物碱和双酯型生物碱箱式图,结果见图2。从图3中可以看出,不同企业单酯型生物碱和双酯型生物碱总量差异均较明显,同一生产企业不同批次样品间差异也较大。如企业A15,其单酯型生物碱在不同批次间含量差异超3倍。企业A12不同批次间双酯型生物碱含量差异接近4倍。
基于待测化合物的结构特点,对甲醇和含有不同比例盐酸的甲醇溶液的提取效率进行了比较,结果含有0.1 mol·L-1盐酸的甲醇溶液提取效率最高;同时比较了不同取样量和不同超声时间的提取效果,最终将取样量定为2粒(约0.5 g),超声时间定为30 min。
本实验比较了不同流动相组成,包括甲醇-0.1%甲酸溶液、乙腈-0.1%甲酸溶液、甲醇-0.1%甲酸(含2.5 mmol·L-1甲酸铵)溶液、乙腈-0.1%甲酸(含2.5 mmol·L-1乙酸铵)溶液的分离效果,结果甲醇-0.1%甲酸(含2.5 mmol·L-1甲酸铵)溶液分离效果较好,色谱峰峰形佳,出峰时间适宜。
《中国药典》2020年版一部中,附子各炮制品的单酯型生物碱总量和双酯型生物碱总量限度一致,其中含单酯型生物碱总量以苯甲酰新乌头原碱、苯甲酰乌头原碱和苯甲酰次乌头原碱的总量计,不得少于0.010%,含双酯型生物碱以新乌头碱、和乌头碱的总量计,不得过0.020%。天麻胶囊处方中,附子(制)10 g,最终制成胶囊1 066粒,每粒天麻胶囊中所含附子(制)生药量为0.009 4 g。因附子为生粉入药,故单酯型生物碱转移率定为90%,双酯型生物碱为毒性成分,转移率暂定为100%,按照《中国药典》2020年版一部限度折算,天麻胶囊中,含单酯型生物碱总量每粒应不得低于0.80 μg;含双酯型生物碱总量每粒应不得过1.8 μg。
附子具有回阳救逆、补火助阳、散寒除湿的功效[2],其主要成分为双酯型生物碱(如乌头碱、新乌头碱和次乌头碱等)、单酯型生物碱(如苯甲酰新乌头原碱、苯甲酰乌头原碱和苯甲酰次乌头原碱等),以及醇胺类(如乌头原碱等)等[9]。其中双酯型生物碱具有较明显的强心作用,但是同时也具有很强的毒性,有效剂量接近致毒剂量,服用过量会导致中毒,甚至死亡[10]。单酯型生物碱具有镇痛和消炎作用,醇胺类化合物镇痛和消炎作用则较弱。生附子中双酯型生物碱含量较高,毒性较大,临床多用其炮制品。生附子经过清洗,浸泡胆巴,蒸煮等步骤,炮制为盐附子、黑顺片、白附片以及淡附片等。在炮制过程中,毒性成分双酯型生物碱水解成为单酯型生物碱,进一步水解成为醇胺类化合物[11]
对本次实验所有样品中的单酯型生物碱和双酯型生物碱含量进行统计分析(图3),结果部分企业的天麻胶囊样品中单酯型生物碱总量和双酯型生物碱总量均较高,部分企业则相反。因为附子为生粉入药,不存在提取工艺的差异,因此提示不同企业投料使用的附子饮片质量差异悬殊,间接表明目前市场上附子饮片质量良莠不齐。目前《中国药典》2020年版附子项下只规定了附子中双酯型生物碱的含量上限以及单酯型生物碱的含量下限,由于复杂的加工过程,附子饮片不同批次之间有效成分和有毒成分差别很大,这直接关系着附子的疗效和安全。因此,应尽快制订合理的附子饮片的质量等级标准,区分饮片不同的商品规格,以体现优质优价,引导产业良性发展[12]。此外,部分企业的天麻胶囊中,双酯型生物碱总量检出量极低,甚至低于检出限。由于双酯型生物碱为毒性成分,饮片生产企业为保证饮片安全,可能存在过度炮制的嫌疑。炮制虽是附子减毒增效的必要手段,但炮制过度则有效成分含量严重降低,影响药效。正如张景岳所言:“制之太过,则但用附子之名耳,效与不效无以应验,今人只知附子之可畏,而不知太熟之无用”。《中国药典》2020年版附子项下对于炮制方法描述较简略,对于蒸煮、泡胆巴等步骤的关键参数未进行规定,不同企业实际炮制过程中工艺各不相同,也进一步导致了市场上附子饮片质量差异较大。
本研究建立了基于UHPLC-MS/MS的含量测定方法,对天麻胶囊中的苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、新乌头碱、次乌头碱和乌头碱进行测定。对不同企业生产的101批天麻胶囊进行了测定,结果不同企业天麻胶囊中附子生物碱含量差异较大,质量一致性较差。同时反映了市场上的附子质量良莠不齐,需引起关注。
