OBJECTIVE To screen one Monascus purpureus stain that has high yield of lovastatin and provide potential strains for the development of red yeast rice(RYR) industry. OBJECTIVE Twelve RYR samples were collected, and Monascus sp. was isolated from these samples using traditional isolation technology and identified by morphology and ITS sequencing. All M. purpureus strains were screened and their total lovastatin contents [including lactone lovastatin(C24H36O5) and acid lovastatin(C24H38O6)] were determined by high performance liquid chromatography(HPLC). Soft SPSS 25.0 was used for variance analysis. RESULTS A total of 35 Monascus sp. strains were isolated and 23 of them were identified at specie level, including 11 M. purpureus strains, 10 M. ruber strains and 2 M. aurantiacus strains. There was significant difference(P<0.05) in lovastatin production among 11 M. purpureus strains. Among them, SM-1 isolate produced the highest amount of total lovastatin, with the value reaching (4.652±0.001) mg·g-1, followed by M4-1, F1-3, and F5-3 strains with the value reaching (2.903±0.033) and (3.108±0.009) mg·g-1, respectively. M4-5 isolate produced the lowest amount of lovastatin with the value of (2.712±0.014) mg·g-1. CONCLUSION Fungal strain is chaotic in the fermentation of RYR on the market. A high-yielding lovastatin strain SM-1 is screened and can be used a potential strain for RYR production.

目的 筛选高产洛伐他汀的紫色红曲霉菌种,为红曲产业发展提供菌种参考。方法 收集红曲资源12份,采用传统分离法进行红曲霉的分离,结合形态学与ITS测序进行菌种鉴定,进一步选取紫色红曲霉菌株,采用高效液相色谱法(HPLC)测定其代谢产物中总洛伐他汀[包括内酯式洛伐他汀(C24H36O5)和酸式洛伐他汀(C24H38O6)]的含量,用SPSS 25.0软件进行方差分析。结果 从12个产地中共分离得到红曲霉属(Monascus sp.)菌株35株,其中23株鉴定到种,包括紫色红曲霉(M. purpureus)11株,红色红曲霉(M. ruber)10株,橙色红曲霉(M. aurantiacus)2株。11株紫色红曲霉菌株发酵产物中洛伐他汀含量存在显著差异(P<0.05),其中SM-1菌株总洛伐他汀含量最高,达(4.652±0.001) mg·g-1;其次为M4-1、F1-3,分别为(2.903±0.033) (3.108±0.009) mg·g-1,最低菌株为M4-5,仅有(2.712±0.014)mg·g-1结论 市售红曲米发酵所用红曲霉繁杂;筛选得到一株高产洛伐他汀的菌株SM-1,可作为红曲米发酵的备选菌株。

目的 筛选高产洛伐他汀的紫色红曲霉菌种,为红曲产业发展提供菌种参考。方法 收集红曲资源12份,采用传统分离法进行红曲霉的分离,结合形态学与ITS测序进行菌种鉴定,进一步选取紫色红曲霉菌株,采用高效液相色谱法(HPLC)测定其代谢产物中总洛伐他汀[包括内酯式洛伐他汀(C24H36O5)和酸式洛伐他汀(C24H38O6)]的含量,用SPSS 25.0软件进行方差分析。结果 从12个产地中共分离得到红曲霉属(Monascus sp.)菌株35株,其中23株鉴定到种,包括紫色红曲霉(M. purpureus)11株,红色红曲霉(M. ruber)10株,橙色红曲霉(M. aurantiacus)2株。11株紫色红曲霉菌株发酵产物中洛伐他汀含量存在显著差异(P<0.05),其中SM-1菌株总洛伐他汀含量最高,达(4.652±0.001) mg·g-1;其次为M4-1、F1-3,分别为(2.903±0.033) (3.108±0.009) mg·g-1,最低菌株为M4-5,仅有(2.712±0.014)mg·g-1结论 市售红曲米发酵所用红曲霉繁杂;筛选得到一株高产洛伐他汀的菌株SM-1,可作为红曲米发酵的备选菌株。

OBJECTIVE To screen one Monascus purpureus stain that has high yield of lovastatin and provide potential strains for the development of red yeast rice(RYR) industry. OBJECTIVE Twelve RYR samples were collected, and Monascus sp. was isolated from these samples using traditional isolation technology and identified by morphology and ITS sequencing. All M. purpureus strains were screened and their total lovastatin contents [including lactone lovastatin(C24H36O5) and acid lovastatin(C24H38O6)] were determined by high performance liquid chromatography(HPLC). Soft SPSS 25.0 was used for variance analysis. RESULTS A total of 35 Monascus sp. strains were isolated and 23 of them were identified at specie level, including 11 M. purpureus strains, 10 M. ruber strains and 2 M. aurantiacus strains. There was significant difference(P<0.05) in lovastatin production among 11 M. purpureus strains. Among them, SM-1 isolate produced the highest amount of total lovastatin, with the value reaching (4.652±0.001) mg·g-1, followed by M4-1, F1-3, and F5-3 strains with the value reaching (2.903±0.033) and (3.108±0.009) mg·g-1, respectively. M4-5 isolate produced the lowest amount of lovastatin with the value of (2.712±0.014) mg·g-1. CONCLUSION Fungal strain is chaotic in the fermentation of RYR on the market. A high-yielding lovastatin strain SM-1 is screened and can be used a potential strain for RYR production.

红曲是由曲霉科真菌紫色红曲霉(Monascus purpureus)菌株,接种于稻米(去皮种仁)上,人工培养制成[1-2]。红曲为药食同源,具有健脾消食,活血化瘀,以及降血糖、降血脂、降血压等功效[3]。在目前的红曲成品中,常出现红曲发酵菌种混杂且退化、生产过程不规范的问题。红曲霉属是一个多功能的丝状真菌类群,包括从毛红曲霉(M. pilosus)、紫色红曲霉(M. purpureus)、红色红曲霉(M. ruber)、烟色红曲霉(M. fuliginosus Sato)、发白红曲霉(M. albidus)、锈色红曲霉(M. rubiginosus)、变红红曲霉(M. rubropuncatatus Sato)、橙色红曲霉(M. aurantiacus)等[4]。紫色红曲霉M. purpureus属于小型丝状腐生真菌,可产生的有益次生代谢产物有天然色素、洛伐他汀、γ-氨基丁酸等[3],在食品、药品领域应用广泛。洛伐他汀具有抑制胆固醇合成、保护肾脏、抗动脉粥样硬化、预防癌症等功能[5];洛伐他汀中含有一个与HMG-CoA结构相似的基团,能竞争性抑制 HMG-CoA还原酶活性,制约胆固醇的过量合成[6]。2020年版《中国药典》红曲质量标准规定C24H36O5(内酯式)不得少于0.22%。现有的对红曲中洛伐他汀的研究表明固态发酵产物中洛伐他汀的含量比液态发酵高[7-8]。另外,在不同生产条件下,同种菌株洛伐他汀含量有所不同是由于培养条件的变化导致菌种生产能力差异,而在相同条件下,同种菌株洛伐他汀产生能力不同则是因为菌株本身在代谢过程中基因表达差异不同。
因此,本研究采用传统分离法进行红曲菌株的分离,结合形态学和内源转录间隔区(internal transcribed spacer,ITS)序列分析,筛选出不同的红曲霉菌株从而获得目标紫色红曲霉菌株,利用HPLC测定,筛选得到高产洛伐他汀的紫色红曲霉菌株。
2019年6月至2019年7月,收集浙江、福建、山东、湖北、云南等省红曲米资源12份,保存于浙江中医药大学中药资源教研室,见表1
酵母浸膏(上海博微生物科技股份有限公司);琼脂粉(纯化,杭州百思生物技术有限公司);DNA iso(9770A)、6×Loading Buffer(9156)、Marker(1000 bp)均购于宝日医生物技术(北京)有限公司; TE 缓冲液(N634,河北品科研生物科技有限公司);核酸染料[翌圣生物科技(上海)有限公司];乙腈、磷酸(色谱级);洛伐他汀对照品(B20806-20 mg,上海源叶生物科技有限公司)。
霉菌恒温培养箱(LMI-475-N,上海龙跃仪器设备有限公司);pH计(FE28,梅特勒-托利多仪器(上海)有限公司);立式压力蒸汽灭菌器(YXQ-LS-75SⅡ,上海博讯实业有限公司医疗设备厂);离心机(5810R)、超低温冰箱(900 Series)均购于艾本德中国有限公司;PCR 仪(4375786,赛默飞世尔科技(中国)有限公司);电泳仪(DYY-8C,北京六一生物科技有限公司);凝胶成像仪(GenoSens 1850,上海勤翔科学仪器有限公司);电热鼓风干燥箱(DHG-9080A)、电热恒温水浴锅(HWS-24)均购于上海哈伟实业有限公司;高速粉碎机(DXF-06D,温岭市林大机械有限公司);高效液相色谱仪(Waters Alliance e2695,美国Waters公司);数控超声波清洗器(KQ-300DV,昆山市超声仪器有限公司);超纯水仪(Milli-Q,美国Millipore公司)。
分离纯化培养基为马铃薯葡萄糖琼脂培养基(potato dextrose agar,PDA培养基)。种子液培养基的配制参考已有文献[9]进行改良:称取50 g葡萄糖(5%)、10 g蛋白胨(1%)、10 g酵母浸膏(1%)、1 g KH2PO4(0.1%)、0.01 g FeSO4·7H2O(0.001%)、0.5 g MgSO4·7H2O(0.05%),纯水定容至1 L(大米营养液在此基础调pH至4.0),分装于250 mL三角瓶中(每瓶100 mL),用8层纱布和一层牛皮纸封口,棉线绳扎紧,经121 ℃高温灭菌30 min,洁净工作台内紫外放凉,待接菌。大米培养基:取过60目筛的籼米米粉135 g和米糠15 g(减少黏性,发酵完成后筛去)置于650 mL大组培瓶中,混合均匀后加入54 mL营养液(0.36 mL·g-1,营养液即种子液培养基基础调pH至4.0),再次混合均匀,用8层纱布和一层牛皮纸封口,棉线绳扎紧,经121 ℃高压灭菌40 min,洁净工作台内紫外放凉,即得。
将不同来源的红曲米研磨成粉和红曲米粉分别少量均匀撒于PDA 培养基中,用两层封口膜封紧,置于霉菌恒温培养箱(28 ℃)中培养3~5 d,待白色绒毛菌丝长出后,洁净工作台内操作挑取少许菌丝接种于PDA培养基上培养1 周,后转移纯化培养3次即可在培养基上出现形态单一的菌落。
采用 TakaRa DNAiso 9770A 试剂盒对分离出的单一真菌进行基因组 DNA 的提取。采用通用引物ITS4和ITS5进行PCR扩增,扩增反应体系(50 μL):DNA样本2 μL,ITS4 1 μL,ITS5 1 μL,2×Tap PCR MasterMix 25 μL,dd H2O 21 μL。PCR 扩增反应条件具体如下:95 ℃预变性5 min;95 ℃变性30 s,55 ℃退火30 s,72 ℃延伸 1 min,30个循环后在72 ℃下稳定延伸 5 min,终止反应,-20 ℃保存。吸取扩增产物5 μL经琼脂糖凝胶电泳检测合格后送至生工生物工程(上海)股份有限公司进行测序。将基因测序结果与BLAST 工具软件进行比对,选取同源性较高的菌株序列并下载作为参考序列比对,利用 MEGA 7.0软件分析,以邻位连接法构建系统发育树[10]
选取分离鉴定得到的且保藏生长状态较好的11株紫色红曲霉菌株,分别为M1-7、M2-4、M4-1、M4-5、F1-3、F4-4、F4-7、F5-5、SM-1、HQ-W、YY1~3进行菌种活化。用经过高温灭菌的打孔器(6 mm)在活化好的菌种平板上打出菌饼,接种到液体种子液培养基中(每150 mL 1个菌饼),用8层纱布和1层牛皮纸封口,橡皮筋扎紧,30 ℃,160 r·min-1,避光恒温振荡培养3 d,期间注意观察颜色及黏稠度的变化。大米培养基放凉(<40 ℃),接入20%种子液(0.2 mL· g-1),敲打松散混合均匀后用8层纱布和1层牛皮纸封口,橡皮筋扎紧,变温培养,培养箱内30 ℃避光静置培养3 d,后降温至26 ℃稳定6 h,敲散培养基呈松散状态,继续培养至35 d。期间每隔5 d敲打松散(无需打开发酵瓶,防污染)。固态发酵期间,每隔5 d洁净工作台内取样一次(5、10、15、20、25、30、35 d),60~65 ℃烘4~5 h至恒重,研磨粉碎后过60目筛,密封于封装袋中备用。
以ZORBAX Eclipse C18(4.6 mm×250 mm,5 μm)为色谱柱,乙腈(A)-0.1%磷酸(B)为流动相,梯度洗脱(1~5 min,60%~65%A;5~20 min,65%A;20~25 min,65%~90%A;25~30 min,90%~60%A;30~35 min,60%),体积流量为1.0 mL·min-1,柱温为28 ℃,检测波长238 nm,进样量为10 μL。对照品溶液及发酵产物样品溶液 HPLC 色谱图见图1
内酯式洛伐他汀标准品溶液:精密称取洛伐他汀标准品适量,置于10 mL量瓶中,用甲醇稀释至刻度,配制成浓度为206.4 μg·mL-1内酯式洛伐他汀标准品溶液。
酸式洛伐他汀标准品溶液[11]:精密称取内酯式洛伐他汀标准品2 mg,适量甲醇溶解,用0.2 mol·L-1NaOH溶液超声转化40 min,取出冷却后,用3 mol·L-1磷酸将pH调至7.7,加甲醇定容至10 mL量瓶,即得208.9 μg·mL-1酸式洛伐他汀标准品溶液。
供试品溶液的配制:精密称取经烘干粉碎后的发酵产物(过60目筛)0.5 g,置于25 mL具塞锥形瓶中,精密加入25 mL甲醇,密塞,称定质量,超声(300 W,40 KHz)45 min后,取出,放冷,再称定质量,加甲醇补足失重,摇晃均匀,离心(4 000 r·min-1)10 min,取上清液过0.45 μm微孔有机滤膜,取续滤液,即得,实验重复3次(n=3)。
将“2.6”项下各标准品溶液用甲醇进行稀释后,在“2.5”项下相对应成分色谱条件下进样测定,以标准品溶液质量浓度为横坐标(X),峰面积为纵坐标(Y)进行线性回归分析,可得到内酯式洛伐他汀、酸式洛伐他汀的线性回归方程,分别为Y=27 454X-3 352.9,Y=40 797X+70 300。各标准品溶液稀释至不同倍数后,在“2.5”项下相对应成分色谱条件下进样测定,采用信噪比法测定各指标成分的检测限(S/N=3∶1)及定量限(S/N=10∶1),内酯式洛伐他汀的检测限和定量限分别为87.87,293.5 ng·mL-1;酸式洛伐他汀的检测限和定量限分别为35.65,119.1 ng·mL-1。两个成分在一定的浓度范围内与峰面积的线性关系良好。
将“2.6”项下供试品处理溶液在“2.5”项下相对应成分色谱条件下进样测定,记录峰面积,计算样品中内酯式洛伐他汀、酸式洛伐他汀的含有量,并对其含有量差异比较分析。
应用SPSS 25.0软件处理数据,方差分析采用单因素ANOVA分析,聚类分析采用组间联接法[12]
应用传统分离法,并通过对菌落形态的观察,从红曲样品中共计分离真菌117株,纯化后获得单一菌落菌株35株,部分菌株在PDA培养基上的形态见图2
ITS序列比对和鉴定结果见表3,由表3可知,35株红曲霉菌株Blast比对结果相似度可达97%~100%,其中可鉴定到红曲霉属(M1-6、 M2-1、 M2-2、 M4-4、 F1-4、 F2-3、 F4-1、 F4-3、 F4-5、 F4-6、 F5-1、 F5-4)的菌株共计12株;紫色红曲霉(M1-7、 M2-4、 M4-1、 M4-5、 F1-3、 F4-4、 F4-7、 F5-5、 SM-1、 YY1-3、 HQ-W)共计11株,红色红曲霉(M4-2、 M4-3、 M4-4’、 M4-6、 M6-2、 M6-3、 M6-4、 F3-1、 F4-2、 F5-2)共计10株,橙色红曲霉(M1-8、 M2-3)共计2株。SM-1与Monascus属的4个参考序列形成支持强度为99%的分支,并且SM-1与M. purpureus(LT963493.1)形成支持强度为99%的末端分支。 部分红曲霉菌PCR扩增产物琼脂糖凝胶电泳图见图3,系统发育树见图4
采用HPLC对11株紫色红曲霉(M. purpureus)的固态发酵产物进行内酯式及酸式洛伐他汀的测定,并计算总洛伐他汀的含有量,结果见表4,图5。由表4可知,11株紫色红曲霉中有4株(SM-1、M4-1、F1-3、M4-5)产洛伐他汀,其中,发酵35 d结束后,SM-1产总洛伐他汀含量最高,可达到(4.652±0.001) mg·g-1;产量居中的有M4-1、F1-3;产量最低的是M4-5,仅有(2.712±0.014) mg·g-1;其余7株紫色红曲霉(M1-7、 M2-4、 F4-4、 F4-7、 F5-5、 HQ-W、 YY1-3)不产洛伐他汀。利用SPSS 25.0软件分析,将上述4株固态发酵产生洛伐他汀的菌株分别在同一发酵时间段总洛伐他汀的产量做差异比较分析(表4)。由表4中可知,不同菌株紫色红曲霉在发酵时间段(5、10、15、20、25、30、35 d)上产洛伐他汀的量存在显著差异(P<0.05)。
本实验利用收集到的12个产地的红曲样品,从中分离得到红曲霉菌株35株,初步建立了红曲霉资源库。通过ITS测序得到2020年版《中国药典》所规定的红曲发酵菌种紫色红曲霉11株,并对11株紫色红曲霉进行固态发酵,利用HPLC测定发酵产物中总洛伐他汀的含量,筛选得到4株产洛伐他汀的菌株,分别为SM-1、M4-1、F1-3、M4-5,其中SM-1菌株产洛伐他汀的能力最强。在酸性条件下,酸式洛伐他汀能转化为内酯式洛伐他汀,但转化并不完全[13]。通过对它们的固体发酵产物中内酯式洛伐他汀与酸式洛伐他汀含量的比率进行分析,发现在发酵中后期(20~35 d),SM-1菌株的内酯式洛伐他汀与酸式洛伐他汀含量的比率均大于其他四株紫色红曲霉,如发酵35 d,SM-1内酯式洛伐他汀与酸式洛伐他汀含量的比率为8.79,而M4-1、M4-5、F1-3分别为5.63、5.87与6.62,较高的内酯式洛伐他汀与酸式洛伐他汀含量比例可能是SM-1高产的原因之一。
从分离鉴定的结果可知,部分红曲是由橙色红曲霉或红色红曲霉发酵制成,不符合2020年版《中国药典》规定的紫色红曲霉发酵规定,表明市售红曲发酵菌种较为繁杂,存在红曲伪品,如浙江宁波与福建屏南产地红曲,只分离得到了红色红曲霉M. ruber
传统红曲霉发酵大多采用固态发酵,Wu等[14]、Wu等[15]通过固态发酵实验筛选出的高产洛伐他汀的红曲菌株,洛伐他汀产量分别达17.90 mg·g-1、5.75 mg·g-1,均高于本试验SM-1菌株洛伐他汀的产量。经对比分析发现,Wu等所使用的为固态发酵培养基用乳酸调节了pH至5.0,并浸泡2 h,且种子液孢子浓度控制在每克106个(大米干质量),而本实验未调节pH及定量孢子浓度,这可能是SM-1菌株产量低的原因。Wu等所使用的固态发酵培养基中含有米粉和豆粉,比例为3:2,该培养基有利于洛伐他汀产量积累,原因可能培养基中豆粉含有丰富的蛋白质,降低了发酵基质中C/N值。而有研究表明,通过降低发酵基质中C/N值可以有效提高洛伐他汀的产量[16]。在下一步工作中,我们将优化SM-1发酵条件,包括培养基的配比与pH,以期提高SM-1洛伐他汀的含量。2020年版《中国药典》红曲质量标准规定C24H36O5(内酯式)不得少于0.22%,忽略了酸式洛伐他汀的存在。而纯天然发酵的红曲产品中,洛伐他汀是以内酯式与酸式两种形式存在,内酯式洛伐他汀需体内产生的羟基酯酶水解成酸式方能发挥药效,酸式洛伐他汀与HMG-CoA还原酶结构更为接近,具有更强的生物活性[17],因此,同时测定红曲中两种构型洛伐他汀的含量更能反映红曲产品的质量。
