OBJECTIVE To investigate the effect of polyphyllin Ⅱ (PPⅡ) on apoptosis of osteosarcoma cells and its mechanism. METHODS Osteosarcoma (OS) U2OS and HOS cell lines were selected as the research object. CCK-8 method was used to detect the effect of PPⅡ on cell viability. Colony forming assay was used to detect the effect of PPⅡ on colony formation ability. EdU incorporation assay was used to detect the effect of PPⅡ on DNA synthesis. Annexin V-FITC/PI staining assay was used to detect the effect of PPⅡ on cell apoptosis. JC-1 assay was used to determine the effect of PPⅡ on the mitochondrial membrane potential (MMP). The protein expression levels of B-cell lymphoma-2 (Bcl-2), Bcl-2 associated X protein (Bax), cysteinyl aspartate specific proteinase 3 (Caspase 3), Caspase 7, Caspase 9, cleaved cysteinyl aspartate specific proteinase 3 (cleaved Caspase 3), cleaved Caspase 7, cleaved Caspase 9, poly (ADP-ribose) polymerase (PARP), cleaved poly (ADP-ribose) polymerase (cleaved PARP), protein kinase R like endoplasmic reticulum kinase (PERK), eukaryotic translation initiation factor 2α (eIF2α), C/EBP homologous protein (CHOP), activating transcription factor 4 (ATF4), p-PERK, and p-eIF2α were analyzed by Western blot. RESULTS PPⅡ significantly inhibited the viability of OS cells in a time- and dose-dependent manner, dose-dependently inhibited the colony formation and DNA synthesis, significantly promoted apoptosis rate, decreased MMP (P<0.05 or P<0.01), and increased Bax/Bcl-2 protein ratio and the protein expression levels of cleaved Caspase 3, cleaved Caspase 7, cleaved Caspase 9, cleaved PARP, p-PERK, p-eIF2α, ATF4, CHOP (P<0.05 or P<0.01). CONCLUSION PPⅡ promotes OS cell apoptosis by regulating mitochondrial pathway and mediating endoplasmic reticulum (ER) stress.

目的 探讨重楼皂苷Ⅱ(polyphyllin Ⅱ,PPⅡ)对骨肉瘤(osteosarcoma,OS)细胞凋亡的影响及作用机制。方法 选取OS的U2OS和HOS细胞株作为研究对象。细胞计数试剂盒-8(CCK-8)法检测PPⅡ对细胞活力的影响;克隆形成实验检测PPⅡ对细胞克隆形成能力的影响;5-乙炔基-2'-脱氧尿苷(EdU)渗入实验检测PPⅡ对细胞DNA合成的影响;膜联蛋白V-异硫氰基荧光素/碘化丙啶(annexin V-FITC/PI)双染法检测PPⅡ对细胞凋亡率的影响;JC-1荧光探针检测PPⅡ对线粒体膜电位变化的影响;Western blot法检测PPⅡ对B淋巴细胞瘤2(B-cell lymphoma-2,Bcl-2)、Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)、半胱氨酸天冬氨酸蛋白酶3(cysteinyl aspartate specific proteinase 3,Caspase 3)、Caspase 7、Caspase 9、切割型半胱氨酸天冬氨酸蛋白酶3(cleaved cysteinyl aspartate specific proteinase 3,cleaved Caspase 3)、cleaved Caspase 7、cleaved Caspase 9、多腺苷二磷酸多聚酶[poly (ADP-ribose) polymerase,PARP]、切割型多腺苷二磷酸多聚酶[cleaved poly (ADP-ribose) polymerase,cleaved PARP]、蛋白激酶R样内质网激酶(protein kinase R like endoplasmic reticulum kinase,PERK)、真核生物翻译起始因子2α(eukaryotic translation initiation factor 2α,eIF2α)、C/EBP同源蛋白(C/EBP homologous protein,CHOP)、激活转录因子4(activating transcription factor 4,ATF4)、p-PERK和p-eIF2α蛋白表达水平的影响。结果 PPⅡ显著性抑制OS细胞活力,并呈时间和剂量相关性;抑制细胞克隆形成和DNA合成;显著性增加细胞凋亡率,降低线粒体膜电位(P<0.05或P<0.01);显著性提高Bax/Bcl-2蛋白比值,升高cleaved Caspase 3、cleaved Caspase 7、cleaved Caspase 9、cleaved PARP、p-PERK、p-eIF2α、ATF4和CHOP蛋白表达水平(P<0.05或P<0.01)。结论 PPⅡ通过线粒体和内质网应激途径诱导OS细胞凋亡。

OBJECTIVE To investigate the effect of polyphyllin Ⅱ (PPⅡ) on apoptosis of osteosarcoma cells and its mechanism. METHODS Osteosarcoma (OS) U2OS and HOS cell lines were selected as the research object. CCK-8 method was used to detect the effect of PPⅡ on cell viability. Colony forming assay was used to detect the effect of PPⅡ on colony formation ability. EdU incorporation assay was used to detect the effect of PPⅡ on DNA synthesis. Annexin V-FITC/PI staining assay was used to detect the effect of PPⅡ on cell apoptosis. JC-1 assay was used to determine the effect of PPⅡ on the mitochondrial membrane potential (MMP). The protein expression levels of B-cell lymphoma-2 (Bcl-2), Bcl-2 associated X protein (Bax), cysteinyl aspartate specific proteinase 3 (Caspase 3), Caspase 7, Caspase 9, cleaved cysteinyl aspartate specific proteinase 3 (cleaved Caspase 3), cleaved Caspase 7, cleaved Caspase 9, poly (ADP-ribose) polymerase (PARP), cleaved poly (ADP-ribose) polymerase (cleaved PARP), protein kinase R like endoplasmic reticulum kinase (PERK), eukaryotic translation initiation factor 2α (eIF2α), C/EBP homologous protein (CHOP), activating transcription factor 4 (ATF4), p-PERK, and p-eIF2α were analyzed by Western blot. RESULTS PPⅡ significantly inhibited the viability of OS cells in a time- and dose-dependent manner, dose-dependently inhibited the colony formation and DNA synthesis, significantly promoted apoptosis rate, decreased MMP (P<0.05 or P<0.01), and increased Bax/Bcl-2 protein ratio and the protein expression levels of cleaved Caspase 3, cleaved Caspase 7, cleaved Caspase 9, cleaved PARP, p-PERK, p-eIF2α, ATF4, CHOP (P<0.05 or P<0.01). CONCLUSION PPⅡ promotes OS cell apoptosis by regulating mitochondrial pathway and mediating endoplasmic reticulum (ER) stress.

骨肉瘤(osteosarcoma,OS)是一种易发于儿童和青少年的常见原发性恶性肿瘤,具有隐匿性、侵袭性强和易转移的特点,有统计显示约80%患者确诊时已发生病灶转移,术后效果差[1];同时由于肺转移和对化疗的耐药性,导致患者的生存率低、总体预后差[2]。因此,研发治疗OS高效、低毒的药物显得尤为迫切。
重楼为百合科植物云南重楼[Paris polyphylla Smith var. yunnanensis (Franch.) Hand.-Mazz.]或七叶一枝花[Paris polyphylla Smith var. chinensis (Franch.) Hara]的干燥根茎,具有清热解毒、消肿止痛、凉肝定惊的功效[3]。重楼中主要含有甾体皂苷类、胆甾烷醇类、C21甾体化合物等多种化学成分,现代研究表明,甾体皂苷是重楼抗肿瘤的主要活性成分,对肺癌、胃癌、肝癌等多种恶性肿瘤均有良好的抗肿瘤作用。重楼皂苷Ⅱ(polyphyllin Ⅱ,PPⅡ)属于甾体皂苷中的薯蓣皂苷,对黑色素瘤、乳腺癌和肝癌等多种恶性肿瘤具有抑制细胞增殖、迁移侵袭和诱导细胞凋亡等作用[4],然而关于PPⅡ在OS中的作用及内在分子机制的相关研究还未见报道。因此,本研究以OS细胞为研究对象,从线粒体和内质网应激途径的角度探讨PPⅡ对OS细胞凋亡的影响及其可能的分子机制,以期为PPⅡ的开发利用和OS的治疗提供实验依据。
PPⅡ(质量分数≥98%,批号50773-42-7,成都仪睿生物科技有限公司);细胞计数试剂盒-8(CCK-8,批号917P016,北京索莱宝科技有限公司);膜联蛋白V-异硫氰基荧光素/碘化丙啶凋亡检测试剂盒(AnnexinV-FITC/PI,批号027E1210LA,南京诺唯赞生物科技有限公司);半胱氨酸天冬氨酸蛋白酶3抗体(Caspase 3,批号3)、cleaved Caspase 3抗体(批号22)、多腺苷二磷酸多聚酶抗体(PARP,批号15)、cleaved PARP抗体(批号13)、Caspase 7抗体(批号2)、cleaved Caspase 7抗体(批号3)、Caspase 9抗体(批号6)、cleaved Caspase 9抗体(批号2)、蛋白激酶R样内质网激酶抗体(PERK,批号6)、真核生物翻译起始因子2α抗体(eIF2α,批号9)、p-eIF2α抗体(批号6)、C/EBP同源蛋白抗体(CHOP,批号14)(美国CST公司);Bcl-2相关X蛋白抗体(Bax,批号GR3204974-1)、B淋巴细胞瘤2抗体(Bcl-2,批号GR3254866-1)(英国abcam公司);p-PERK抗体(批号#45R6406,北京博奥森生物技术有限公司);内质网应激抑制剂4-苯基丁酸(4-PBA,批号S359202,美国selleck公司);凋亡抑制剂Z-VAD-FMK(批号091521220310)、5-乙炔基-2'-脱氧尿苷染色试剂盒(EdU,批号102920201123)、JC-1检测试剂盒(批号102920201123)(上海碧云天生物技术有限公司)。
上皮形态骨肉瘤细胞系(U2OS)和成纤维细胞及上皮样骨肉瘤细胞系(HOS)(中国科学院上海细胞库)。
MR-96A型酶标仪(深圳迈瑞公司);YB3002型分析天平(上海力能公司);Western电泳仪电源(上海天能公司);Bio-rad ChemiDoc XRS+成像系统(美国Bio-rad公司);BX53型荧光电子显微镜(日本Olympus/奥林巴斯公司);FacsCanto I型流式细胞仪(美国BD公司)。
用二甲基亚砜(DMSO)作为溶剂,PPⅡ溶解于DMSO配制成母液,各组DMSO溶剂浓度不超过体积分数0.1%。药物分装储存于-20 ℃冰箱中避光保存。
U2OS和HOS细胞用含体积分数10%胎牛血清、100 u·mL-1青霉素、100 μg·mL-1链霉素的DMEM高糖培养基,于37 ℃、5% CO2的培养箱中培养,细胞密度长至80%~90%时消化传代,用于后续实验。
将对数生长期的U2OS和HOS细胞以每孔5×103个接种至96孔板中,待细胞密度长至60%~70%时加入不同浓度的PPⅡ(0.625、1.25、2.5、5、10、20、40 μmol·L-1),同时设置溶剂对照组,继续培养24、48 、72 h后,每孔加入10 μL CCK-8溶液再培养2 h,用酶标仪在450 nm波长处测定每个孔的吸光度(A)值,每组设3个复孔检测细胞活力(公式1)。

对于研究Caspase抑制剂Z-VAD-FMK对PPⅡ抑制细胞活力作用的影响时,给药前加入终浓度10 μmol·L-1 Z-VAD-FMK孵育1 h,其余操作同上。
对于研究内质网应激抑制剂4-PBA对PPⅡ抑制细胞活力作用的影响时,给药前加入终浓度2 mmol·L-1 4-PBA孵育1 h,其余操作同上。
将对数生长期的U2OS和HOS细胞以每孔1 000个接种于6孔板中,37 ℃、5% CO2培养箱中孵育过夜,加入含不同浓度的PPⅡ(5、10、20 μmol·L-1)的新鲜培养基,设置溶剂对照组,继续培养24 h后,换成正常的新鲜培养基继续培养,每隔2~3 d换1次液。14 d后,6孔板中出现肉眼可见的克隆体时终止培养,弃去培养液,用PBS清洗2次、4%多聚甲醛固定30 min、结晶紫染色液染色20 min、PBS缓慢洗去染色液,置于空气中干燥后拍照。
将对数生长期的U2OS和HOS细胞以每孔3×105个接种于激光共聚焦平皿中,37 ℃、5% CO2培养箱中孵育过夜,加入含不同浓度的PPⅡ(5、10、20 μmol·L-1)的新鲜培养基,设置溶剂对照组,给药18 h后,将50 μmol·L-1的EdU溶液加入培养基中继续孵育6 h。弃去上清液,用磷酸盐缓冲液(PBS)清洗2次、4%的多聚甲醛固定30 min、加入Apollo荧光染料避光孵育30 min、PBS清洗2次,加入4',6-二脒基-2-苯基吲哚(DAPI)染色液继续孵育30 min、PBS清洗2次后,激光共聚焦进行拍照检测。
将对数生长期的U2OS和HOS细胞以每孔3×105个接种于6孔板中,37 ℃、5% CO2培养箱中孵育过夜,加入含不同浓度的PPⅡ(5、10、20 μmol·L-1)的新鲜培养基,设置溶剂对照组。给药24 h后,离心收集细胞,加入500 μL Binding buffer重悬细胞,每个样品管加入5 μL Annexin V-FITC和 5 μL碘化丙啶(PI),室温下避光孵育15 min,流式细胞仪进行检测。
将对数生长期的U2OS和HOS细胞以每孔3×105个接种于6孔板中,37 ℃、5% CO2培养箱中孵育过夜,加入含不同浓度的PPⅡ(5、10、20 μmol·L-1)的新鲜培养基,设置溶剂对照组。给药24 h后,离心收集各孔细胞,用500 μL提前配好的JC-1工作液重新悬浮细胞沉淀,室温下避光孵育20 min,用Incubation buffer 洗涤细胞2次,流式细胞仪进行检测。
收集细胞,提取总蛋白,采用BCA法测定各样本蛋白浓度。配制体积分数8%~12%聚丙烯酰胺凝胶后,上样、电泳、转膜后,质量分数5%脱脂奶粉封闭1 h,4 ℃一抗孵育过夜(目的蛋白抗体稀释比例1∶1 000,内参β-actin抗体稀释比例1∶3 000)。用TBST缓冲溶液清洗3次,每次10 min,二抗室温孵育1 h后,继续用TBST缓冲溶液清洗3次,每次10 min。用增强型化学发光试剂(ECL)显色,Bio-RAD曝光仪曝光,采集不同时段的显色条带。采用Image J软件对蛋白条带的灰度值进行分析,以β-actin为内参,以相对灰度值表示目标蛋白的表达。
实验结果用$ \bar{x} \pm s$表示,实验数据用GraphPad Prism 8.3软件对数据进行统计分析,两组间均数比较采用t检验,多组间均数比较采用One-Way ANOVA检验,以P<0.05为差异具有统计学意义。
PPⅡ(0.625、1.25、2.5、5、10、20、40 μmol·L-1)分别作用细胞24、48、72 h,CCK-8法评估细胞活力,与对照组比较,PPⅡ显著抑制U2OS和HOS细胞活力,呈时间和剂量依赖性,且作用24 h时U2OS细胞对PPⅡ较为敏感(图1)。PPⅡ对U2OS细胞在24、48、72 h的IC50值分别为7.56、5.66、2.97 μmol·L-1,对HOS细胞在24、48、72 h的IC50值分别为10.57、4.87、1.87 μmol·L-1。参考以上结果,确定5、10、20 μmol·L-1作为PPⅡ的给药剂量,并选择作用24 h作为PPⅡ的给药条件。
克隆形成实验是测定单个细胞增殖能力的有效方法,细胞克隆能力强也是肿瘤细胞的一大特点[5]。与对照组比较,PPⅡ(5、10、20 μmol·L-1)组细胞克隆数目和大小均显著减少,且呈剂量相关性,表明PPⅡ能够抑制U2OS和HOS细胞的克隆形成能力,即抑制U2OS和HOS细胞增殖,见图2
EdU是一种胸腺嘧啶核苷类似物,在细胞增殖期代替T渗入正在复制的DNA中,通过检测EdU标记的细胞多少反映细胞的增殖情况[6]。EdU试剂盒进一步验证药物对细胞增殖能力的影响,用不同浓度的PPⅡ分别作用后,U2OS和HOS细胞中EdU阳性细胞数显著低于对照组,表明PPⅡ抑制OS细胞增殖的机制可能与抑制DNA合成有关,见图3
磷脂酰丝氨酸(phosphatidylserine,PS)外翻是细胞凋亡的典型特征[7]。为了确定PPⅡ是否具有促OS细胞凋亡的作用,Annexin V-FITC/PI双染色后用流式细胞仪检测。U2OS细胞对照组、5、10、20 μmol·L-1 PPⅡ组早期凋亡率分别是1.51%、5.53%、7.54%、13.40%,晚期凋亡率分别是3.20%、21.70%、25.90%、47.60%,总凋亡率分别是4.71%、27.23%、33.44%、61.00%。HOS细胞对照组、5、10、20 μmol·L-1 PPⅡ组早期凋亡率分别是2.08%、8.76%、11.30%、16.70%,晚期凋亡率分别是3.56%、14.9%、27.10%、46.60%,总凋亡率分别是5.64%、23.66%、38.40%、63.30%。随着PPⅡ浓度的增加,细胞早、晚期凋亡率逐渐增加。与对照组相比,不同浓度PPⅡ组细胞总凋亡率显著增加(P<0.05或P<0.01),且呈浓度相关性,见图4
研究表明,线粒体功能异常和细胞凋亡密切相关,且在细胞凋亡途径中发挥重要作用。细胞凋亡早期,线粒体通透性转换孔的开放增加,进而导致线粒体膜电位的下降[8]。JC-1是一种广泛用于检测线粒体膜电位的理想荧光探针。与对照组相比,PPⅡ显著性降低红色荧光与绿色荧光的比值(P<0.05或P<0.01),引起U2OS和HOS细胞线粒体膜电位的下降,且呈剂量相关性,见图5
Bcl-2家族蛋白分为Bax等促凋亡蛋白和Bcl-2等抗凋亡蛋白,而Bax/Bcl-2比例失衡改变线粒体膜通透性,激发细胞色素c等凋亡因子从线粒体释放出来,进而诱导凋亡小体的形成,激活Caspase家族蛋白,产生Caspase级联反应,诱发细胞发生线粒体凋亡[9-11],因此本实验进一步考察了PPⅡ对凋亡相关蛋白的表达。Western blot结果发现,与对照组相比,PPⅡ组Bcl-2蛋白表达显著降低,Bax蛋白表达和Bax/Bcl-2蛋白比值显著升高(P<0.05或P<0.01),且呈剂量依赖性,见图6
Western blot结果表明,PPⅡ显著性促进Caspase 3、7、9及PARP的剪切活化,即显著性促进cleaved Caspase 3、7、9和cleaved PARP的蛋白表达水平(P<0.05或P<0.01),且呈剂量依赖性,进一步明确了PPⅡ通过激活Caspase级联反应诱导OS细胞凋亡,见图7
Caspase抑制剂10 μmol·L-1 Z-VAD-FMK处理U2OS细胞后,再给予20 μmol·L-1 PPⅡ处理,测定细胞活力的变化。与PPⅡ组相比,PPⅡ+Z-VAD-FMK组细胞活力显著性升高(P<0.05),表明Z-VAD-FMK能部分逆转PPⅡ抑制的细胞活力,见图8。综上,PPⅡ对OS细胞的部分增殖抑制作用是由Caspase依赖的凋亡途径介导。
内质网应激是细胞凋亡的重要调控信号,过度的内质网应激会诱导细胞凋亡[12]。与对照组相比,PP Ⅱ组的p-PERK、p-eIF2α、ATF4和CHOP的蛋白表达水平显著性升高,且呈剂量相关性(P<0.05或P<0.01),表明PP Ⅱ可以激活U2OS和HOS细胞的内质网应激,见图9
内质网应激抑制剂2 mmol·L-1 4-PBA处理U2OS细胞后,再给予20 μmol·L-1 PPⅡ处理,进一步探讨内质网应激对PPⅡ诱导的细胞凋亡的影响。与PPⅡ组相比,PPⅡ+4-PBA组细胞凋亡率显著降低(P<0.01),见图10;PPⅡ+4-PBA组cleaved Caspase 3、cleaved PARP蛋白表达水平显著降低(P<0.05或P<0.01),见图11;PP Ⅱ +4-PBA组细胞活力显著升高(P<0.05),见图12。综上,PP Ⅱ 对OS细胞的部分增殖抑制作用和凋亡促进作用由内质网应激途径介导。
OS是一种恶性程度极高的原发性恶性骨肿瘤,常发于儿童和青少年,预后差,致残率和死亡率高[13]。目前,西医临床治疗OS的首选治疗手段是手术,由于OS易发生转移,多数患者确诊时已发生转移,单纯手术治疗的生存率低[14]。此外,有报道手术加放化疗虽然可以取得短期临床疗效,但易发生骨髓抑制、胃肠道症状等副作用[15]。中医认为,OS为“石痈”“骨瘤”“肉瘤”等,对其病因归结为“虚、寒、痰、瘀、毒”5个方面,证型分为痰瘀互结证、邪毒留滞证、气血两亏证及脾肾亏虚证[15]。中医药的整体思维和辨证施治观念,能够增强机体的免疫力、减轻手术的并发症、降低放化疗副作用,与西医药互补协同作用,达到提高患者生存质量、延长患者生存期的作用。研究发现,中药单体提取物或中成药制剂在抗肿瘤方面具有多靶点、多途径的作用,能够通过多条途径调节OS,使中医药在OS治疗方面有着广阔的发展前景[16]
清热解毒类中药重楼对邪毒留滞证OS具有治疗的作用。研究表明,PPⅡ是重楼的主要抗肿瘤药效成分,对黑素瘤、非小细胞肺癌、乳腺癌、胃癌及膀胱癌等有抑制细胞增殖、诱导细胞凋亡的作用[17-21],然而关于PPⅡ对OS细胞的作用机制还未有报道。因此,本研究首次考察了PPⅡ对体外培养的OS细胞U2OS和HOS的细胞增殖和细胞凋亡的影响。CCK-8结果显示,PPⅡ时间和剂量依赖性抑制OS细胞U2OS和HOS的细胞活力,且对U2OS细胞在24、48、72 h的IC50值分别为7.56、5.66、2.97 μmol·L-1,对HOS细胞在24、48、72 h的IC50值分别为10.57、4.87、1.87 μmol·L-1。单克隆形成实验结果显示,PPⅡ显著降低细胞克隆形成数,从而抑制细胞的克隆形成。EdU渗入实验结果显示,PPⅡ剂量依赖性抑制EdU阳性细胞数。综上,PPⅡ对OS细胞有抑制细胞增殖的作用。
细胞凋亡,又称Ⅰ型程序性细胞死亡,是维持细胞内环境稳定、调控细胞组织器官发育、对环境因素改变产生的应答反应,是细胞主动实施的结束细胞生命的正常死亡过程。同时,凋亡是引起肿瘤细胞在对抗抗肿瘤活性化合物时发生的一种主要死亡过程[22]。细胞膜中的磷脂酰丝氨酸(PS)由膜内侧翻向外侧,是细胞凋亡的早期信号[7]。为了验证PPⅡ引起的细胞增殖抑制作用与细胞凋亡是否有关,本实验采用Annexin V-FITC/PI双染法检测PS外翻,结果显示,PPⅡ剂量依赖性增加细胞凋亡率,提示PPⅡ可以诱导OS细胞发生细胞凋亡。
细胞凋亡途径主要有外源性凋亡途径、内源性凋亡途径(又称为线粒体途径)[23]。线粒体是内源性凋亡途径的调控中心,其功能受以Bax和Bcl-2为主要影响因子的Bcl-2家族蛋白的靶向调节。细胞受到凋亡信号刺激时,诱导线粒体损伤,Bax/Bcl-2比值的高低决定细胞是否开启凋亡途径[24]。当Bax/Bcl-2升高时,引起线粒体膜通透性和膜电位改变,刺激细胞色素c从线粒体释放至胞浆中,激活Caspase 9酶原剪切活化为cleaved Caspase 9,从而进一步激活下游的Caspase 3活化为cleaved Caspase 3,最终导致细胞凋亡[25-27]。本研究考察了PPⅡ是否可以引起线粒体损伤。结果发现,PPⅡ显著性降低线粒体膜电位、升高Bax/Bcl-2蛋白表达比例,并进一步诱导了Caspase家族凋亡相关蛋白的显著性剪切活化。综上,PPⅡ可以诱导OS细胞发生线粒体介导的Caspase依赖的内源性凋亡途径(图13)。
内质网应激初期,PERK发生自磷酸化形成同源二聚体[28],二聚化的PERK通过磷酸化eIF2α,减弱蛋白翻译,从而减少蛋白质在内质网腔内的合成。ATF4基因在磷酸化eIF2α作用下翻译增强。ATF4能够激活抗氧化应激作用和氨基酸合成转运蛋白等靶基因的表达[29]。研究结果显示,PPⅡ通过上调U2OS和HOS细胞的p-PERK、p-eIF2α、ATF4、CHOP蛋白表达水平激活内质网应激。CHOP是内质网应激诱导细胞凋亡的标志性蛋白,PERK-eIF2α-ATF4通路是CHOP重要调控因素。过度的内质网应激通过诱导CHOP的表达调控Bcl-2基因家族蛋白的表达,提高线粒体对细胞凋亡信号的敏感性,诱导细胞凋亡[30]。内质网应激抑制剂4-PBA下调OS细胞的内质网应激水平后,结果发现PPⅡ所诱导的细胞凋亡率升高、cleaved Caspase 3和cleaved PARP蛋白高表达、细胞活力抑制得到部分缓解。因此,内质网应激途径可以部分介导PPⅡ对OS细胞的增殖抑制作用和凋亡促进作用。同时,作为线粒体途径的上游参与PPⅡ诱导的细胞凋亡。
综上所述,PPⅡ能够抑制OS细胞的增殖能力,诱导OS细胞凋亡。其作用机制可能是通过介导线粒体和内质网应激途径诱导细胞凋亡。本研究从线粒体和内质网应激途径方面揭示了PPⅡ抗OS的活性,为探索OS的治疗提供了新思路。
