OBJECTIVE To investigate the secondary metabolites and bioactivities of the mangrove Rhizophora stylosa derived fungus Diaporthe sp. SCSIO 41011. METHODS Various modern chromatographic methods were subjected to isolate and purify the fermentation products of the strain. The chemical structures of the obtained compounds were identified by nuclear magnetic resonance technology as well as literature data. The antibacterial activity, NF-κB luciferase inhibitory activity, and antioxidant activity of the compounds were determined. The antioxidant activity of the compounds was determined by 1,1-diphenyl-2-picrylhydrazy (DPPH) and ferric reducing ability of plasma (FRAP). RESULTS Seven compounds (1-7) were isolated from the fermentation products and were identified as nigrosporapyrone D (1), vermopyrone (2), tyrosol (3), 2-(2-hydroxyethyl)phenol (4), 3-phenylpropane-1,2-diol (5), 4-hydroxyphenethyl acetate (6), 2-hydroxyphenethyl acetate (7). The chemical structure of 2 was confirmed by X-ray single-crystal diffraction method for the first time herein. At the concentration of 1 mg·mL-1, the FRAP values of compounds 3, 6, and 7 were 208.75, 160.03, and 162.75 μmol·L-1, respectively. CONCLUSION Two pyranones (1 and 2) and five phenolic compounds (3-7) are characterized from the fungus, meanwhile compounds 1, 2, 4, 5, and 7 are isolated from the Diaporthe genus for the first time. And compounds 3, 6, and 7 show iron ion reduction/antioxidant capacity. This study would enrich the structural diversity of secondary metabolites of Diaporthe sp., and lays a foundation for further study on the natural products of mangrove endophytic fungi.

目的 开展红树植物红海榄(Rhizophora stylosa)来源真菌Diaporthe sp. SCSIO 41011的次级代谢产物及其生物活性研究。方法 综合运用多种现代色谱分离技术对菌株发酵产物进行分离纯化,结合核磁共振波谱技术和文献数据对比鉴定化合物的化学结构,测定化合物的抑菌活性、核因子κB(NF-κB)荧光素酶抑制活性,并采用1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazy, DPPH)、铁离子还原能力(ferric reducing ability of plasma, FRAP)测定化合物的抗氧化活性。结果 从该菌大米发酵产物中共分离鉴定了7个聚酮类化合物, 分别为黑孢霉吡喃酮D (nigrosporapyrone D,1)、马鞭吡喃酮 (vermopyrone,2)、酪醇 (tyrosol,3)、2-(2-羟基乙基)苯酚 [2-(2-hydroxyethyl)phenol,4]、3-苯基丙烷-1,2-二醇 (3-phenylpropane-1,2-diol,5)、4-羟基苯乙基乙酯 (4-hydroxyphenethyl acetate,6)、2-羟基苯乙基乙酯 (2-hydroxyphenethyl acetate,7)。首次通过X-ray单晶衍射技术证实了化合物2的化学结构。在1 mg·mL-1质量浓度下,化合物3、6和7的FRAP值分别为208.75、160.03和162.75 μmol·L-1结论 从该菌中共分离获得2个吡喃酮类化合物(1~2)和5个苯(酚)类化合物(3~7),其中,化合物1、2、4、5和7为首次从该属中分离得到。化合物3、6和7具有一定的铁离子还原/抗氧化能力。本实验丰富了红树中间座壳属Diaporthe sp.真菌次级代谢产物的结构多样性,为进一步研究红树内生真菌天然产物奠定基础。

目的 开展红树植物红海榄(Rhizophora stylosa)来源真菌Diaporthe sp. SCSIO 41011的次级代谢产物及其生物活性研究。方法 综合运用多种现代色谱分离技术对菌株发酵产物进行分离纯化,结合核磁共振波谱技术和文献数据对比鉴定化合物的化学结构,测定化合物的抑菌活性、核因子κB(NF-κB)荧光素酶抑制活性,并采用1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazy, DPPH)、铁离子还原能力(ferric reducing ability of plasma, FRAP)测定化合物的抗氧化活性。结果 从该菌大米发酵产物中共分离鉴定了7个聚酮类化合物, 分别为黑孢霉吡喃酮D (nigrosporapyrone D,1)、马鞭吡喃酮 (vermopyrone,2)、酪醇 (tyrosol,3)、2-(2-羟基乙基)苯酚 [2-(2-hydroxyethyl)phenol,4]、3-苯基丙烷-1,2-二醇 (3-phenylpropane-1,2-diol,5)、4-羟基苯乙基乙酯 (4-hydroxyphenethyl acetate,6)、2-羟基苯乙基乙酯 (2-hydroxyphenethyl acetate,7)。首次通过X-ray单晶衍射技术证实了化合物2的化学结构。在1 mg·mL-1质量浓度下,化合物3、6和7的FRAP值分别为208.75、160.03和162.75 μmol·L-1结论 从该菌中共分离获得2个吡喃酮类化合物(1~2)和5个苯(酚)类化合物(3~7),其中,化合物1、2、4、5和7为首次从该属中分离得到。化合物3、6和7具有一定的铁离子还原/抗氧化能力。本实验丰富了红树中间座壳属Diaporthe sp.真菌次级代谢产物的结构多样性,为进一步研究红树内生真菌天然产物奠定基础。

OBJECTIVE To investigate the secondary metabolites and bioactivities of the mangrove Rhizophora stylosa derived fungus Diaporthe sp. SCSIO 41011. METHODS Various modern chromatographic methods were subjected to isolate and purify the fermentation products of the strain. The chemical structures of the obtained compounds were identified by nuclear magnetic resonance technology as well as literature data. The antibacterial activity, NF-κB luciferase inhibitory activity, and antioxidant activity of the compounds were determined. The antioxidant activity of the compounds was determined by 1,1-diphenyl-2-picrylhydrazy (DPPH) and ferric reducing ability of plasma (FRAP). RESULTS Seven compounds (1-7) were isolated from the fermentation products and were identified as nigrosporapyrone D (1), vermopyrone (2), tyrosol (3), 2-(2-hydroxyethyl)phenol (4), 3-phenylpropane-1,2-diol (5), 4-hydroxyphenethyl acetate (6), 2-hydroxyphenethyl acetate (7). The chemical structure of 2 was confirmed by X-ray single-crystal diffraction method for the first time herein. At the concentration of 1 mg·mL-1, the FRAP values of compounds 3, 6, and 7 were 208.75, 160.03, and 162.75 μmol·L-1, respectively. CONCLUSION Two pyranones (1 and 2) and five phenolic compounds (3-7) are characterized from the fungus, meanwhile compounds 1, 2, 4, 5, and 7 are isolated from the Diaporthe genus for the first time. And compounds 3, 6, and 7 show iron ion reduction/antioxidant capacity. This study would enrich the structural diversity of secondary metabolites of Diaporthe sp., and lays a foundation for further study on the natural products of mangrove endophytic fungi.

红树林植物(mangrove)是一类盐生植物(耐盐植物),主要生长在热带和亚热带潮间带的潮汐和咸沼泽中[1-2],是海洋与陆地间动态转换的特殊生态系统[3]。红树植物拥有独特的生长环境,高盐、高温、潮湿、潮汐活动频繁等特殊条件能够产生丰富多样、结构复杂新颖且具有活性的天然物质,如抗氧化、抗肿瘤、抗菌、抗炎活性和调节细胞生长等活性[4-6],受到天然产物化学家的广泛关注。间座壳属(Diaporthe sp.)真菌属于子囊菌门、粪壳菌纲、黑腐皮壳科,是一类常见的内生真菌,广泛存在于自然界中,研究表明,该属真菌是抗肿瘤、抗菌、抗氧化等活性代谢产物的潜在来源之一[7]。因此,开展红树植物红海榄来源真菌活性代谢产物的研究具有重要意义。
近年来,本团队从北部湾来源真菌中分离鉴定了系列混源萜类、聚酮类、蒽醌类等结构新颖且某些活性显著的化合物[8-11]。前期从海南红树植物红海榄(Rhizophora stylosa)来源真菌Diaporthe sp. SCSIO 41011中分离获得系列新颖具肾癌细胞毒活性的氯代嗜氮酮类化合物[12],从该菌株的剩余流分分离获得2个吡喃酮类化合物(1~2)和5个苯(酚)类化合物(3~7),其中,化合物1、2、4、5和7为首次从该属中分离得到,并且首次通过X-ray单晶衍射技术证实了化合物2的化学结构,见图1
高压灭菌锅(MLS-3781L-PC,普和希健康医疗器械上海有限公司);超净工作台(ZYJ-S, 苏州安泰空气技术有限公司);纯水机(优普,成都优普仪器设备有限公司);大型旋转蒸发仪(Hei-VAP Industrial,德国Heidolph公司);中压制备色谱仪(瑞士BUCHI公司);薄层色谱及柱色谱用硅胶(青岛海洋化工厂);高效液相色谱仪(Prominnence LC-2030C3D, 日本岛津公司);YMC-Pack ODS-A色谱柱(10 mm×250 mm,5 μm,日本YMC公司);96孔细胞板(美国Corning公司);酶标仪(SYNERGY H1,Bio Tek公司);分析纯化学试剂(广州化学试剂厂);色谱纯试剂(美国默克公司);1,1-二苯基-2-三硝基苯肼 (DPPH)自由基(上海阿拉丁生化科技股份有限公司);抗坏血酸(上海阿拉丁生化科技股份有限公司);(E)-3-(4-甲基苯基)磺酰基丙烯腈(BAY11-7082,纯度≥98%,德国Merck KGaA公司);脂多糖(L2880, 西格玛奥德里奇公司)。
耐甲氧西林金黄色葡萄球菌(methicillin resistant Staphylococcus aureus, MRSA),铜绿假单胞菌(Pseudomonas aeruginosa ATCC9027)和表皮葡萄球菌(S. epidermidis ATCC 12228)3株测试病原菌购自北京北纳创联生物技术研究院,菌株保藏于广西中医药大学海洋药物研究院菌种库。
菌株SCSIO 41011分离自海南省三亚市的一株红树植物红海榄(Rhizophora stylosa)的新鲜组织中,菌种保存于广东省微生物菌种保藏中心(GDMCC 60671)。菌株鉴定及培养发酵参考本团队前期的研究工作[12]
发酵结束后,向大米培养基中加入2倍量的乙酸乙酯,切成小块后超声提取,真空减压蒸馏浓缩后获得粗浸膏50 g。采用中压正相硅胶柱色谱(石油醚-二氯甲烷-甲醇体系)对粗浸膏进行梯度洗脱,通过薄层色谱分析,经检识合并得到流分Fr.1~Fr.9。通过HPLC-UV分析发现流分Fr.5中化合物峰较为丰富,主要对该流分进行分离。采用中压反相(ODS)色谱(甲醇-水体系)梯度洗脱对Fr.5进行分离获得5个子流分Fr.5-1~5-5。流分Fr.5-1继续采用半制备高效液相色谱(CH3OH-H2O=40∶60, 2 mL·min-1)纯化得到化合物3(tR=9 min,11 mg)、4(tR=13 min,2.5 mg)、5(tR=14 min,14 mg)、6(tR=21 min,52 mg)和7(tR=33 min,6 mg)。流分Fr.5-2继续采用半制备高效液相色谱(CH3OH-H2O=45∶55, 2 mL·min-1)纯化得到化合物1 (tR=9 min, 3 mg)和2 (tR=11 min, 9 mg)。
参考文献[13]方法并修改,称取化合物,用二甲基亚砜(dimethyl sulfoxide,DMSO)溶解并配制成初始质量浓度为5 mg·mL-1的母液。称取DPPH自由基1.97 mg,溶解在25 mL的无水乙醇中, 配成物质的量浓度为0.2 mmol·L-1的溶液,-20 ℃下避光保存备用。设置给药组,空白对照组和空白组,每个化合物及各对照组均设置3次平行。给药组加入25 μL样品溶液与100 μL DPPH乙醇溶液,空白对照组加入25 μL空白溶剂与100 μL DPPH乙醇溶液,空白组加入125 μL DPPH乙醇溶液,避光反应30 min。以抗坏血酸为对照,用酶标仪在检测波长为517 nm的条件下检测吸光度,计算DPPH清除率。阳性对照药是质量浓度为100 μg·mL-1的抗坏血酸。
参考文献[14]方法并稍作改动测定化合物的抗氧化活性。样品用DMSO配制成初始质量浓度为5 mg·mL-1的母液,以FeSO4·7H2O作为标准品绘制标准曲线,得到回归方程:y=0.006x+0.038 5(r2=0.995 4)。其中:y为吸光度值;x为FeSO4的浓度(μmol·L-1)。阳性对照药是质量浓度为50 μg·mL-1的抗坏血酸。
参考文献[15]方法,采用滤纸片琼脂扩散法对单体化合物进行初筛。用DMSO溶解化合物并配制成初始质量浓度为5 mg·mL-1的母液,将3株病原菌(MRSA, Pseudomonas aeruginosa,Staphylococcus epidermidis)分别接种至经高温灭菌的LB液体培养基中并于37 ℃恒温、180 r·min-1条件下振荡培养4 h。往已灭菌的LB固体培养基(360 mL)中加入菌液(360 μL),摇匀后立即倒板。分别吸取各待测化合物的母液2 μL至无菌滤纸片上,将平板均匀地分为四等分,随后用镊子将滤纸片按顺序贴在LB平板上,37 ℃恒温条件下静置培养12 h后观察抑菌圈大小。阳性对照药是质量浓度为1 mg·mL-1(甲醇溶解)的氨苄西林。
参考文献[16]方法,采用荧光素酶报告基因技术测试化合物对脂多糖(LPS)诱导核因子κB(NF-κB)荧光素酶报告基因表达的影响。
化合物1为白色晶体(甲醇), 分子式为C8H8O41H-NMR(700 MHz,methanol-d4) δH:6.86(1H,d,J=2.3 Hz,H-5), 5.89(1H,d,J=2.3 Hz,H-3), 3.91(3H,s,9-OMe), 2.48(3H,s,H3-8)。13C-NMR(175 MHz,methanol-d4) δC: 192.1(C,C-7), 172.0(C,C-4), 164.8(C,C-2), 155.9(C,C-6), 105.7(CH,C-5), 94.2(CH,C-3), 57.4(CH3,C-9), 25.7(CH3,C-8)。以上数据与文献[17]对照基本一致,因此确定化合物1为nigrosporapyrone D。
化合物2为无色针状结晶(甲醇), 分子式为C9H10O41H-NMR(700 MHz,chloroform-d4) δH: 7.07(1H,s,H-5), 3.95(3H,s,9-OMe), 2.54(3H,s,H3-8), 2.00(3H,s,H3-10)。13C-NMR(175 MHz,chloroform-d4) δC:191.8(C,C-7),164.2(C,C-4),163.5(C,C-2),153.1(C,C-6),109.8(C,C-3),98.2(CH,C-5),56.8(CH3,C-9), 26.0(CH3,C-8),9.5(CH3,C-10)。 以上数据与文献[18]对照基本一致,因此确定化合物2为vermopyrone。本实验首次通过X-ray单晶衍射技术确证了化合物2的化学结构(图2), 单晶数据如下:C9H10O4, Mr=182.17, 晶体尺寸0.2×0.2×0.1 mm3,正方晶形, a=15.867 6(6)Å,b=15.990 7(7)Å,c=6.793 6(4)Å,α=90°,β=90°,γ=90°,V=1 723.77(14)Å3,T=(-173.25±0.6)℃, 空间群 Aea2, Z=8,μ(CuKα)=0.943 mm-1, 收集到3 554个反射, 1 193个独立反射(Rint=0.039 0)。最终R1值为0.073 7[I>2σ(I)],wR(F2)的最终值为0.195 9[I>2σ(I)],最终R1值为0.074 8(所有数据), wR(F2)的最终值为0.196 9(所有数据), F2 的拟合优度为1.123。 以上数据已上传到剑桥晶体数据中心(CCDC1901459)。
化合物3为白色固体(甲醇),分子式为C8H10O21H-NMR (700 MHz,methanol-d4) δH:7.03 (2H,d,J=8.5 Hz,H-1,5),6.70 (2H,d,J=8.5 Hz,H-2,4),3.68 (2H,t,J=7.2 Hz,H2-8),2.71 (2H,t,J=7.2 Hz,H2-7)。13C-NMR (175 MHz,methanol-d4) δC:156.7 (C,C-6),130.9 (C,C-3),130.8 (CH,C-2,4),116.1 (CH,C-1,5),64.5 (CH2,C-8),39.4 (CH2,C-7)。以上数据与文献[19]对照基本一致,因此确定化合物3为tyrosol。
化合物4为黄色油状(甲醇),分子式为C8H10O21H-NMR (700 MHz,methanol-d4) δH:7.07 (1H,dd,J=7.3,1.7 Hz,H-4),7.01 (1H,td,J=7.3,1.7 Hz,H-6),6.76~6.72 (2H,overlapped,H-1,5),3.74 (2H,t,J=7.0 Hz,H2-8),2.83 (2H,t,J=7.0 Hz,H2-7)。13C-NMR (175 MHz,methanol-d4) δC:156.6 (C,C-2),131.8 (CH,C-4),128.4 (CH,C-6),126.6 (C,C-3),120.5 (CH,C-5),116.0 (CH,C-1),63.1 (CH2,C-8),35.1 (CH2,C-7)。以上数据与文献[20]对照基本一致,因此确定化合物4为2-(2-hydroxyethyl)phenol。
化合物5为无色油状(甲醇),分子式为C9H12O2,[α ${]}_{D}^{25}$+22(c 0.1 g·100 mL-1,methanol-d4)。1H-NMR (700 MHz,methanol-d4) δH:7.26 (2H,overlapped,H-6,8),7.24 (2H,overlapped,H-5,9),7.17(1H,m,H-7),3.81(1H,m,H-2),3.50(1H,dd,J=11.2,4.5 Hz,H-1a),3.45 (1H,dd,J=11.2,6.2 Hz,H-1b),2.82 (1H,dd,J=13.7,5.6 Hz,H-3a),2.68 (1H,dd,J=13.7,7.6 Hz,H-3b)。13C-NMR (175 MHz,methanol-d4) δC:140.1 (C,C-4),130.4 (CH,C-6,8),129.2 (CH,C-5,9),127.1 (CH,C-7),74.5 (CH,C-2),66.5 (CH2,C-1),40.9 (CH2,C-3)。以上数据与文献[21]基本一致,因此确定化合物5为3-phenylpropane-1,2-diol。为进一步确定化合物5的立体构型,本实验采用基于含时密度泛函理论-电子圆二色谱(TDDFT-ECD)理论的量子计算方法验证了其构型[16]。利用Spartan' 14软件对化合物进行构象搜索,进而选择玻尔兹曼分布大于1%的低能构象,利用Gaussian 16软件计算其ECD谱图,计算ECD数据经紫外偏移矫正及SpecDis 1.71软件处理[16],结果发现化合物5的实测ECD曲线与R-5构型对应的计算ECD曲线趋势基本一致(图3),故确定其构型为R
化合物6为淡黄色油状(甲醇),分子式为C10H12O31H-NMR (700 MHz,methanol-d4) δH:7.03 (2H,d,J=8.5 Hz,H-2,6),6.72 (2H,d,J=8.5 Hz,H-3,5),4.18 (2H,t,J=7.1 Hz,H2-8),2.80 (2H,t,J=7.1 Hz,H2-7),1.99 (3H,s,H3-10)。13C-NMR (175 MHz,methanol-d4) δC:172.9 (C,C-9),157.0 (C,C-4),130.8 (CH,C-2,6),129.9 (C,C-1),116.2 (CH,C-3,5),66.5 (CH2,C-8),35.1 (CH2,C-7),20.8 (CH3,C-10)。以上数据与文献[22]对照基本一致,因此确定化合物6为4-hydroxyphenethyl acetate。
化合物7为无色油状(甲醇),分子式为C10H12O31H-NMR (700 MHz,methanol-d4) δH:7.07 (1H,dd,J=7.0,1.4 Hz,H-6),7.01 (1H,td,J=7.0,1.4 Hz,H-4),6.74 (2H,overlapped,H-3,5),4.24 (2H,t,J=7.0 Hz,H2-8),2.91 (2H,t,J=7.0 Hz,H2-7),1.99 (3H,s,H3-10)。13C-NMR (175 MHz,methanol-d4) δC:173.1 (C,C-9),156.7 (C,C-2),131.8 (CH,C-6),128.8 (CH,C-4),125.3 (C,C-1),120.4 (CH,C-5),115.9 (CH,C-3),65.1 (CH2,C-8),30.8(CH2,C-7),20.8 (CH3,C-10)。以上数据与文献[23]对照基本一致,因此确定化合物7为2-hydroxyphenethyl acetate。
所有化合物均进行了抗氧化活性、抑菌活性及NF-κB荧光素酶抑制活性测试。1 mg·mL-1质量浓度下,测定化合物1~7的DPPH清除率,结果表明,化合物均无明显活性;1 mg·mL-1质量浓度下,化合物3、6和7的FRAP值分别为208.75、160.03和162.75 μmol·L-1,表明化合物3、6和7具有一定的铁离子还原/抗氧化能力(抗坏血酸:50 μg·mL-1质量浓度下,FRAP值为133.92 μmol·L-1)。同时发现所有化合物对3个病原菌均未显示明显抑菌圈,表明化合物无抑菌活性,并且所有化合物均未具有明显的NF-κB荧光素酶抑制活性。
本实验对红树植物红海榄(Rhizophora stylosa)来源真菌Diaporthe sp. SCSIO 41011的次级代谢产物进行分析研究,分离得到2个吡喃酮类化合物(1~2)和5个苯(酚)类化合物(3~7),并且化合物1、2、4、5和7为首次从该属中分离得到。利用核磁共振技术以及文献数据对照的方法鉴定其化学结构,测试化合物1~7的抗氧化活性(DPPH法和FRAP法)、抑菌活性及NF-κB荧光素酶抑制活性,其中化合物3、6和7(1 mg·mL-1)具有一定的铁离子还原/抗氧化能力。与阳性对照相比,该类化合物的抗氧化活性较弱,从化学结构角度来讲,可能是由于化合物所含酚羟基数量较少或者酚羟基难以被氧化为醌式结构,后续将考虑通过结构优化增强化合物的活性。据文献[18]报道,化合物2对喉癌细胞Hep2具有一定的抑制作用,其IC50值为100 μg·mL-1。化合物3能够抑制稻瘟霉[24],也具有自由基清除活性[25];化合物4能够抑制铜绿假单胞菌P. aeruginosa的生长,最小抑菌浓度(MIC)值为35 μg·mL-1[26],化合物6能够抑制子宫颈癌HeLa细胞增殖[27],且对白色念珠菌(Candida albicans)的生长有明显抑制作用,其抑菌圈直径为25 mm[22]
聚酮类化合物是海洋真菌天然产物中最为常见类型之一,也是间座壳属的主要代谢产物[7],结构类型丰富多样,具有抗肿瘤、抗炎、抗菌、抗氧化等生物活性,受到化学家和药理学家广泛关注[28]。团队前期从该菌株中分离得到10个嗜氮酮类化合物[12],继续对该菌株剩余流分进行研究,从中分离得到7个吡喃酮类或苯(酚)类化合物,说明该菌株具有产生丰富聚酮类代谢产物的潜力,后续将继续利用培养调控策略深入挖掘聚酮类化合物结构多样性。其次,化合物1~7作为重要生源砌块,为嗜氮酮类化合物的生物合成推导提供重要依据。此外,该类化合物的活性筛选及文献报道数据说明部分化合物具有弱肿瘤细胞毒活性,以及对病原菌具一定选择性抑制作用,后续将利用乙酰胆碱酯酶等酶活或延缓衰老线虫模型继续对该类化合物进行活性筛选,挖掘潜在药用价值。本实验丰富了菌株Diaporthe sp.次级代谢产物的结构多样性,为红树植物共附生真菌天然产物研究奠定基础。
