OBJECTIVE To identify Bambusae Caulis In Taenias and its adulterants using DNA barcoding,ensure the safety andefficacy of clinicalapplications and provide a referencefor the development and quality evaluation of Bambusae Caulis In Taenias-containing drugs. METHODS By using accurately original plants bamboo samples as a reference, and based on the screening of universal barcodes, variable regions or loci within the chloroplast genome were identified to design genus-specific DNA barcodes for the molecular identification of commercial Bambusae Caulis In Taenias. RESULTS ITS2 region effectively identified Phyllostachys edulis and allowed for the preliminary identification of other species at the genus level. Additionally, 17 genus-specific DNA barcodes were designed, enabling the precise identification of Phyllostachys nigra var. henonis, Bambusa beecheyana var. Pubescens and Bambusa textilis. Among 31 batches of commercial Bambusae Caulis In Taenias, only 1 batch was authenticated as Bambusa beecheyana var. Pubescens, while the others were all adulterants, including 19 batches were identified as Phyllostachys edulis, 8 as Bambusa textilis, and 3 as other species. Furthermore, several samples were found to be mixtures of different species. CONCLUSION The method established in this study provides an accurate tool for the identification of Bambusae Caulis In Taenias original plants, laying a foundation for quality control and market regulation.

目的 应用DNA条形码对竹茹及其混伪品进行准确鉴定,保证临床用药安全性和有效性,为含竹茹药物的研发与质量评价提供参考。方法 以基原准确的竹子样本为参照,在筛选通用条形码的基础上,寻找其叶绿体基因组的变异区域或位点,设计特异性DNA条形码序列,对市售竹茹样品进行分子鉴定。结果 内转录间隔区2(ITS2)序列可以鉴定毛竹(Phyllostachys edulis),且在属水平上初步鉴定其他物种,设计的17个特异性DNA条形码分别能鉴定淡竹(Phyllostachys nigra var. henonis)、大头典竹(Bambusa beecheyana var. Pubescens)及青皮竹(Bambusa textilis)。31批市售竹茹样品仅有正品(大头典竹)1批,其他均为伪品,包括毛竹19批,青皮竹8批,其他基原3批,同时存在不同基原混杂的情况。结论 本研究建立的方法可对竹茹基原进行准确鉴别,为竹茹的质量控制及市场监管奠定基础。

目的 应用DNA条形码对竹茹及其混伪品进行准确鉴定,保证临床用药安全性和有效性,为含竹茹药物的研发与质量评价提供参考。方法 以基原准确的竹子样本为参照,在筛选通用条形码的基础上,寻找其叶绿体基因组的变异区域或位点,设计特异性DNA条形码序列,对市售竹茹样品进行分子鉴定。结果 内转录间隔区2(ITS2)序列可以鉴定毛竹(Phyllostachys edulis),且在属水平上初步鉴定其他物种,设计的17个特异性DNA条形码分别能鉴定淡竹(Phyllostachys nigra var. henonis)、大头典竹(Bambusa beecheyana var. Pubescens)及青皮竹(Bambusa textilis)。31批市售竹茹样品仅有正品(大头典竹)1批,其他均为伪品,包括毛竹19批,青皮竹8批,其他基原3批,同时存在不同基原混杂的情况。结论 本研究建立的方法可对竹茹基原进行准确鉴别,为竹茹的质量控制及市场监管奠定基础。

OBJECTIVE To identify Bambusae Caulis In Taenias and its adulterants using DNA barcoding,ensure the safety andefficacy of clinicalapplications and provide a referencefor the development and quality evaluation of Bambusae Caulis In Taenias-containing drugs. METHODS By using accurately original plants bamboo samples as a reference, and based on the screening of universal barcodes, variable regions or loci within the chloroplast genome were identified to design genus-specific DNA barcodes for the molecular identification of commercial Bambusae Caulis In Taenias. RESULTS ITS2 region effectively identified Phyllostachys edulis and allowed for the preliminary identification of other species at the genus level. Additionally, 17 genus-specific DNA barcodes were designed, enabling the precise identification of Phyllostachys nigra var. henonis, Bambusa beecheyana var. Pubescens and Bambusa textilis. Among 31 batches of commercial Bambusae Caulis In Taenias, only 1 batch was authenticated as Bambusa beecheyana var. Pubescens, while the others were all adulterants, including 19 batches were identified as Phyllostachys edulis, 8 as Bambusa textilis, and 3 as other species. Furthermore, several samples were found to be mixtures of different species. CONCLUSION The method established in this study provides an accurate tool for the identification of Bambusae Caulis In Taenias original plants, laying a foundation for quality control and market regulation.

竹茹为禾本科植物青秆竹(Bambusa tuldoides Munro)、大头典竹[Sinocalamus beecheyanus (Munro) McClure var. Pubescens P. F. Li]或淡竹[Phyllostachys nigra(Lodd.) Munro var. henonis(Mitf.)Stapf ex Rendle]的茎秆的干燥中间层。具有清热化痰,除烦,止呕的功效[1]。我国为竹类资源大国,有50多属500种竹种[2],据报道,国内很多地区存在使用慈竹(Bambusa emeiensis)、刚竹(Phyllostachys bambusoides)、毛竹(Phyllostachys edulis)等加工为竹茹使用的现象[3-5]。其他近缘植物如粉单竹(Bambusa chungii)、麻竹(Dendrocalamus latiflorus)、青皮竹(Bambusa textilis)等也常与竹茹基原植物混淆[6]。由于竹类植物形态特征的相似性与复杂性,仅靠形态特征来进行基原鉴定困难重重,也很容易发生分类上的混乱[7],对以此为原料加工的竹茹进行基原鉴定则困难更大。目前,竹茹的基原鉴定相关研究不足。
DNA条形码 (DNA barcode)是指生物体内能够代表该物种的、标准的、有足够变异的、易扩增且相对较短的DNA片段,DNA条形码鉴定技术具有技术简便、鉴定准确及使用方便等优势[8]。随着高通量测序技术的发展,叶绿体全基因组序列数据已经成为植物物种鉴定和系统发生学研究的重要工具,显著增加了可用于物种分辨的信息量,从而提高了物种鉴定的准确性和可靠性[9]。有文献利用核基因及叶绿体基因序列对竹亚科部分物种进行DNA条形码鉴定[10-12],但缺少对竹茹的3种法定基原物种的鉴定。Luo等[13]利用psbA-trnH序列从采购的竹茹药材中鉴定出青秆竹(Bambusa tuldoides)、淡竹(Phyllostachys glauca)及其他基原,但未鉴定出《中国药典》2020年版竹茹法定来源品种大头典竹(Bambusa beecheyana var. Pubescens)和淡竹(Phyllostachys nigra var. Pubescens)。本研究通过收集基原准确的竹类植物样本,筛选、设计DNA条形码并建立竹茹及其混伪品的分子鉴定方法,以实现对市售竹茹样品的基原鉴定,为竹茹临床用药的安全性、有效性提供保障。
采集12批四种基原的新鲜竹子(表1),根据原植物形态并经华南植物园倪静波工程师鉴定。收集31批市售竹茹药材及饮片样品(表2),经暨南大学药学院马志国教授鉴定符合《中国药典》2020年版一部竹茹项下的性状规定。所有样品保存于暨南大学岭南传统中药研究中心。

DN14改良十六烷基三甲基溴化铵(cetyltrimethylammonium bromide,CTAB)植物基因组DNA快速提取试剂盒(北京艾德莱生物科技有限公司);50×TAE缓冲液(北京索莱宝科技有限公司);MD101-02DL 2000 Plus Marker (南京诺唯赞生物科技股份有限公司);GR501-01核酸染料(南京诺唯赞生物科技股份有限公司);琼脂糖(上海翌圣生物科技有限公司);无酶水;无水乙醇(广州光华科技股份有限公司);β-巯基乙醇(上海麦克林生化科技有限公司);氯仿(广州化学试剂厂);P520-012型Phanta Flash Master Mix(Dye Plus)(南京诺唯赞生物科技股份有限公司)。
采用植物基因组DNA快速提取试剂盒法,按照说明书操作,提取基因组DNA,并由超微量紫外分光光度计检测浓度及纯度。
选用内转录间隔区(internal transcribed spacer,ITS)、内转录间隔区2(internal transcribed spacer 2,ITS2)、psbA-trnH、matK及rbcL等通用引物对基原准确的竹子样品进行鉴定,发现ITS2扩增序列可以准确鉴别毛竹,且在属水平上初步鉴定其他物种,其余引物的扩增序列无法准确鉴定基原。
为进一步鉴定竹茹基原,本研究从美国国家生物信息中心(NCBI)下载淡竹、大头典竹、青秆竹、青皮竹的叶绿体全基因组序列作为参考序列,利用Bioedit软件对上述序列进行多重序列比对,分析竹茹3种法定基原物种及非法定基原物种青皮竹的基因序列间差异,寻找变异区域或位点,并由Primer Premier 5软件针对该区域或位点两端的保守序列设计扩增引物。最终,筛选出17个变异区域或位点,包括淡竹1个,大头典竹9个,青皮竹7个,并设计对应的扩增引物,其中Bbp-Bt引物可同时鉴别大头典竹与青皮竹,具体见表3
PCR 反应体系为30 μL,包括2×Phanta Flaash Master Mix(Dye Plus)15 μL,引物(F,R)各2 μL,DNA模板含量为60~150 ng,ddH2O补足至30 μL。扩增程序为:98 ℃预变性2 min;98 ℃变性10 s,50 ℃退火5 s,72 ℃延伸5 s,33个循环;72 ℃延伸1 min。PCR产物经1%琼脂糖凝胶电泳检测后送往广州华大基因测序,拼接序列并进行Blast比对。
使用ITS2及表3中的Pnh、Bbp-Bt引物对采集的基原准确的竹子样品进行分子鉴定,测序结果显示,12批样品均与通过性状鉴定的基原相符(表4)。此外,分别选用一批大头典竹及青皮竹对设计的其他特异性DNA条形码进行验证,结果各特异性DNA条形码均能准确鉴定基原,表明ITS2及设计的各基原特异性DNA条形码可对四种基原的竹茹样品进行准确鉴定。
使用ITS2及表3中的Pnh、Bbp-Bt引物对31批市售竹茹样品进行分子鉴定,结果见表5。共鉴定出基原为大头典竹的正品1批;其余竹茹基原均非药典品种,包括毛竹19批、青皮竹8批、粉单竹1批、慈竹1批、毛竹与粉单竹混杂1批。
利用研究建立了市售竹茹及其混伪品的分子鉴定方法,对31批市售竹茹样品进行鉴定,仅鉴定出1批大头典竹属于《中国药典》2020年版规定的正品,因NCBI数据库中大头典竹命名已采用新的分类系统,本研究中大头典竹采用新分类系统的拉丁学名,即Bambusa beecheyana var. Pubescens;其余30批均为非药典法定基原,毛竹因分布广,很多地区用于竹茹的加工,占比高达61.2%,是市售竹茹中最主要的基原,青皮竹主要分布在两广地区,也是市售竹茹的一种主要基原,占比25.8%。此外,个别批次样品也存在不同基原混用的情况,可能是产地采收时竹子原料混淆造成的。可见,因竹茹原料来源复杂、质量控制标准低等原因,竹茹药材产地就地取材,缺少有效监管措施,造成当前市售竹茹品种混乱,非法定基原占比高,应引起有关部门重视。
叶绿体基因组中存在多个变异区域或位点,在筛选DNA条形码时,除了保证序列的变异度足以区分不同物种的需求外,还需要综合考虑其他因素,比如序列长度、PCR扩增成功率、测序成功率以及是否含有poly结构等[14]。本研究在筛选通用条形码的基础上,对叶绿体基因组序列进行全面分析,寻找变异区域或位点,并设计特异性DNA条形码序列,所建立的方法体系,为竹茹的准确鉴定提供了保障。目前已有研究利用单核苷酸多态性(SNP) 或插入缺失(Indel)标记鉴定物种[15-16],后续可在筛选的各基原特异性位点基础上,设计特异性扩增引物,通过多重位点特异性PCR或实时荧光定量PCR等技术鉴定市售竹茹真伪及掺伪情况。
本研究对竹茹法定基原中的淡竹、大头典竹及伪品毛竹、青皮竹、慈竹及粉单竹实现准确鉴定,另一法定基原青秆竹(Bambusa tuldoides)未成功鉴定。通过对采集的基原准确的青秆竹新鲜植物样品进行叶绿体全基因组测序后再次验证,发现其叶绿体全基因组无变异区域或位点,只能在属水平上对其鉴定,后续可考虑采用其他分子标记方法对青秆竹进行鉴定研究。目前,已将青秆竹叶绿体全基因组序列提交NCBI网站(登录号:PQ014193)。为提高竹茹样品的鉴定效率和准确性,建议先用ITS2引物对竹茹最常见的伪品毛竹进行鉴定,再根据样品所在属针对性地使用不同基原的特异性引物鉴定出具体物种,以达到快速、准确鉴定竹茹正品的目的。
