OBJECTIVE To establish a method for the transformation of some commonly used pesticide limit standards in Chrysanthemi Flos based on ‘GB 2763 National Food Safety Standard-Maximum Residue Limits for Pesticides in Food’. METHODS The indexes of Chrysanthemi Flos (dry) in GB 2763 were sorted out, and the quasi-transformation pesticides were determined as pymetrozine, imidacloprid, boscalid and fluazinam according to the transformation principle. The UPLC-MS/MS method was used to establish the transformation method of Chrysanthemi Flos, and the methodological verification was carried out. RESULTS The four pesticides had good linear relationship in the range of 1-50 ng·mL-1, the correlation coefficients were 0.999 9, the average recovery was 77.9%-95.5%, and the RSD was 1.7%-6.8%. The overall detection rate of the samples was 60%, and the pesticide with the highest detection rate was imidacloprid. The maximum detectable amount was 1 mg·kg-1 (imidacloprid), and the results did not exceed the maximum residue limit specified in GB 2763. The risk assessment system of pesticide residues in traditional Chinese medicine was used to evaluate the chronic risk of the four pesticide limit values to be transformed. The results showed that the chronic risk of fluazinam was greater than 1, and the chronic risk quotients of the other three pesticides were less than 1. Therefore, fluazinam is not converted temporarily, and only pymetrozine, imidacloprid and boscalid are converted. CONCLUSION The three limit standards of pymetrozine, imidacloprid and boscalid in Chrysanthemi Flos were transformed. The established limit standard transformation method is simple and accurate, which is of great significance for standardizing the use of pesticides and medicinal safety in the process of chrysanthemum planting.

目的 建立基于《GB 2763食品安全标准 食品中农药最大残留限量》的菊花中部分常用农药限量标准转化方法。方法 对GB 2763中菊花(干)的指标进行梳理,并根据转化原则,确定拟转化指标为吡蚜酮、吡虫啉、啶酰菌胺和氟啶胺,采用超高效液相色谱-串联质谱(UPLC-MS/MS)方法,建立菊花限量标准转化方法,并进行方法学验证。结果 4种农药在1~50 ng·mL-1内线性关系良好,相关系数均为0.999 9,平均回收率为77.9%~95.5%,相对标准偏差(RSD)为1.7%~6.8%;结果样品总体检出率为60%,检出率最高的农药为吡虫啉;检出量最大的为吡虫啉(1 mg·kg-1),结果均未超过GB 2763规定的最大残留限量。采用中药中农药残留风险评估系统对拟转化的4个农药限量值的慢性风险进行评价,结果氟啶胺的慢性风险商大于1,其余3个农药的慢性风险商均小于1,故氟啶胺暂不转化,仅对吡蚜酮、吡虫啉和啶酰菌胺进行转化。结论 对菊花中吡蚜酮、吡虫啉和啶酰菌胺3个指标限量标准进行转化,建立的限量标准转化方法操作简单,结果准确,对于规范菊花种植过程中农药的使用和药用安全有重大意义。

目的 建立基于《GB 2763食品安全标准 食品中农药最大残留限量》的菊花中部分常用农药限量标准转化方法。方法 对GB 2763中菊花(干)的指标进行梳理,并根据转化原则,确定拟转化指标为吡蚜酮、吡虫啉、啶酰菌胺和氟啶胺,采用超高效液相色谱-串联质谱(UPLC-MS/MS)方法,建立菊花限量标准转化方法,并进行方法学验证。结果 4种农药在1~50 ng·mL-1内线性关系良好,相关系数均为0.999 9,平均回收率为77.9%~95.5%,相对标准偏差(RSD)为1.7%~6.8%;结果样品总体检出率为60%,检出率最高的农药为吡虫啉;检出量最大的为吡虫啉(1 mg·kg-1),结果均未超过GB 2763规定的最大残留限量。采用中药中农药残留风险评估系统对拟转化的4个农药限量值的慢性风险进行评价,结果氟啶胺的慢性风险商大于1,其余3个农药的慢性风险商均小于1,故氟啶胺暂不转化,仅对吡蚜酮、吡虫啉和啶酰菌胺进行转化。结论 对菊花中吡蚜酮、吡虫啉和啶酰菌胺3个指标限量标准进行转化,建立的限量标准转化方法操作简单,结果准确,对于规范菊花种植过程中农药的使用和药用安全有重大意义。

OBJECTIVE To establish a method for the transformation of some commonly used pesticide limit standards in Chrysanthemi Flos based on ‘GB 2763 National Food Safety Standard-Maximum Residue Limits for Pesticides in Food’. METHODS The indexes of Chrysanthemi Flos (dry) in GB 2763 were sorted out, and the quasi-transformation pesticides were determined as pymetrozine, imidacloprid, boscalid and fluazinam according to the transformation principle. The UPLC-MS/MS method was used to establish the transformation method of Chrysanthemi Flos, and the methodological verification was carried out. RESULTS The four pesticides had good linear relationship in the range of 1-50 ng·mL-1, the correlation coefficients were 0.999 9, the average recovery was 77.9%-95.5%, and the RSD was 1.7%-6.8%. The overall detection rate of the samples was 60%, and the pesticide with the highest detection rate was imidacloprid. The maximum detectable amount was 1 mg·kg-1 (imidacloprid), and the results did not exceed the maximum residue limit specified in GB 2763. The risk assessment system of pesticide residues in traditional Chinese medicine was used to evaluate the chronic risk of the four pesticide limit values to be transformed. The results showed that the chronic risk of fluazinam was greater than 1, and the chronic risk quotients of the other three pesticides were less than 1. Therefore, fluazinam is not converted temporarily, and only pymetrozine, imidacloprid and boscalid are converted. CONCLUSION The three limit standards of pymetrozine, imidacloprid and boscalid in Chrysanthemi Flos were transformed. The established limit standard transformation method is simple and accurate, which is of great significance for standardizing the use of pesticides and medicinal safety in the process of chrysanthemum planting.

菊花为菊科植物菊(Chrysanthemum morifolium Ramat)的干燥头状花序,具有散风清热,平肝明目,清热解毒的功效,常用于风热感冒,头痛眩晕,目赤肿痛,眼目昏花,疮痈肿毒[1],属于药食同源品种。菊花中含有丰富的黄酮类、多糖类及挥发油类等活性成分和其他人体所需的微量元素,具有显著的抗炎抗氧化、降血脂、抗菌及抗肿瘤等功效[2-3],广泛应用于食品和中医药领域,市场需求量较大,故菊花通常采用大规模集约化种植栽培,但由于菊花种植对土壤要求较高,因而在种植过程中易发生大面积的病虫害,其主要病虫害有根腐病、霜霉病和褐斑病等,常需要使用农药进行防治[4]。课题组前期对菊花农药使用情况进行调研,发现使用频率较高的为啶虫脒、吡虫啉、吡唑醚菌酯、百菌清、多菌灵、苯醚甲环唑及高效氟氯氰菊酯等,除啶虫脒、吡虫啉及吡唑醚菌酯为登记使用的农药外,其余均为菊花未登记使用的农药,由于种植人员对农药的登记制度和农药信息不了解,较易出现乱用药、滥用药和误用药等情况,给菊花的使用安全带来了较大的隐患[5-7]
《中国药典》2020年版增加了植物类药材和饮片的禁用农药一致性标准要求,但未针对菊花种植过程中常用农药残留设置检查项目。前期文献[8-11]报道,菊花中吡虫啉、吡蚜酮等常用农药检出率较高,同时课题组前期对菊花中101种常用农药进行监测后发现,检出率超过40%的为多菌灵、吡虫啉、啶虫脒、吡唑醚菌酯、嘧菌酯和苯醚甲环唑等,提示应制定科学的方法和限量标准进行控制。本研究以菊花为研究对象,依据中国食品药品检定研究院前期提出的《GB 2763 食品安全国家标准食品中农药最大残留限量》中药品种限量标准转化原则[12],仅对GB 2763中规定菊花最大残留限量的农药进行转化,确定菊花中常用农药指标,建立相应指标的超高效液相色谱-串联质谱(UPLC-MS/MS)检测方法,并对多批次样品进行测定及风险评估,初步拟定菊花中常用农药的限量标准及相应的检测方法。
GB 2763-2021中规定了包括菊花鲜品和干品的农药最大残留限量(MRL),其中菊花(干)属于药用植物,《中国药典》2020年版一部中规定菊花于9~11月花盛开时分批采收,阴干或焙干,或熏、蒸后晒干。GB 2763-2021 及GB 2763-2021.1 中菊花(鲜)和菊花(干)中农药MRLs 共同规定了6种农药的MRL,其中菊花(干)为5种,分别为吡虫啉(imidacloprid)、氟啶胺(fluazinam)、井冈霉素(validamycin)、噻虫嗪(thiamethoxam)、吡蚜酮(pymetrozine)和啶酰菌胺(boscalid),见表1。从表1可见,菊花(干)的限量值除吡蚜酮外均大于菊花(鲜),可能与干燥后菊花(干)由于水分的减失,农药残留量较易增加相关。该6个指标有3个已登记在菊花项下,各农药的限量差距较大,最终根据前期调研和拟定的标准转化原则[12],对菊花(干)的MRL进行转化,由于井冈霉素检测灵敏度低,暂不转化,最终拟转化的农药为4个。
LC-MS 8050型三重四极杆液相色谱质谱联用仪[配有电喷雾离子源(ESI),日本岛津公司];分析天平(XPE205型,瑞士Mettler公司);全自动均质器(AH-30型,睿科仪器有限公司);旋转蒸发仪(R215型,瑞士BUCHI公司);Milli-Q advantage A10超纯水仪(美国Millipore公司);台式大容量冷冻离心机(德国Eppendorf公司)。丙酮、甲醇、乙腈、甲酸均为色谱纯;甲酸铵为色谱纯;35种农药混标溶液(100 mg·L-1,批号为S142905,阿尔塔科技有限公司);菊花样品52批(包括市售和基地样品)。
①混合对照品溶液的制备:精密量取农药混合对照品溶液1 mL,置20 mL量瓶中,用乙腈稀释至刻度,摇匀,即得。②空白基质溶液的制备:取空白基质样品,同供试品溶液的制备方法处理制成空白基质溶液。③基质混合对照溶液的制备:取适量上述对照品储备溶液,制备浓度约为50、20、10、5、2、1 ng·mL-1的基质混合对照品溶液。
取供试品粉末5 g,精密称定,加氯化钠1 g,摇散,再加入乙腈50 mL,匀浆处理2 min(转速不低于12 000 r·min-1),离心(4 000 r·min-1),分取上清液,沉淀再加乙腈50 mL,匀浆处理1 min,离心,合并两次提取的上清液,减压浓缩至约近干,放冷,先用丙酮1 mL溶解,再用乙腈稀释至10.0 mL,摇匀,即得。
色谱条件:Kinetex C18(2.1 mm×10 cm,1.7 μm);以体积分数0.1%甲酸溶液(含5 mmol·L-1甲酸铵)为流动相A,以体积分数95%甲醇溶液(含5 mmol·L-1甲酸铵、体积分数0.1%甲酸)为流动相B,梯度洗脱(0~1 min,70%A;1~12 min,70%A→0%A;12~14 min,0%A);流速为0.3 mL·min-1,柱温40 ℃。
质谱条件:以三重四极杆串联质谱仪检测;离子源为电喷雾(ESI)离子源,依据表2选择正离子/负离子切换扫描模式。监测模式为多反应监测(MRM),各化合物保留时间、监测离子对、碰撞电压(CE)参考值见表2。为提高检测灵敏度,可根据保留时间分段监测各农药。
4种农药在1~50 ng·mL-1内呈现良好的线性关系(r=0.999 9),以信噪比约为3的溶液浓度作为方法检出限,信噪比约为10的溶液浓度作为方法定量限。方法检出限均不高于0.001 mg·kg-1,定量限为0.000 1~0.004 mg·kg-1,均能满足农药残留分析要求,结果见表3
取菊花药材粉末5 g,精密称定,一式12份,置100 mL聚苯乙烯具塞离心管中,分别精密加入混合标准品溶液(1 μg·mL-1)各0.5 mL 3份(高水平)、0.2 mL 6份(中水平兼重复性)、0.1 mL 3份(低水平),同供试品溶液制备方法制得加样回收试验供试品溶液。计算回收率及相应相对标准偏差(RSD)值,菊花低、中、高(10、20、50 ng·mL-1)平均回收率在70%~100%,且RSD值小于8%,结果见表4。结果表明,各农药成分在样品基质中测得回收率重复性良好,满足农药残留测定要求。
本研究分别从浙江、山西、安徽等地收集共52批样品,来源包括市场和种植基地,采用建立的方法对菊花中4种农药进行测定,结果见表5
《中国药典》2020年版四部“9302 中药有害残留物限量制定指导原则”中指出可以根据中药使用特点将我国食品安全国家标准转化为我国药品标准,但需要根据我国农药登记情况,结合不少于50批次的中药品种市场监测数据进行科学性和适用性验证[13]。故在进行GB 2763 菊花食品标准向药材标准转化时,需按照中药使用特点和我国膳食结构进行评估。基于文献[12]报道的中药中农药残留风险评估方法,采用点评估模式,对本次拟转化的菊花中4个农药限量标准的慢性风险进行评价,风险评价计算见公式1~2。
EXPc=(EF×Ed×I×MRL)/(AT×bw)×PF
HQc=EXPc×100/ADI
公式1中,EXPc为慢性膳食暴露量,单位为mg·kg-1 bw·d-1;EF为服用频率,以每年90 d计;Ed为一生的暴露年限,以20年计;AT为平均寿命天数,以365天/年×70年计;I为平均日消费量,单位为kg·d-1,根据《中国药典》2020年版规定菊花平均日消费量为7.5 g;MRL为菊花中该农药的最大残留限量值,单位为mg·kg-1;bw为平均体质量,单位为 kg,以60 kg 计;PF值为农药加工因子,即农药在中药加工过程中从原料到终产品的转移率。由于农药性质各异,在不同加工方式下其加工因子差距较大,故基于风险最大化考虑,本研究中PF 值设定为1。
公式2中,HQc 为慢性风险商,即慢性暴露量与健康指导值的比值,再乘以一定安全系数以评价其风险;ADI 为每日允许摄入量,单位为mg·kg-1 bw,参考GB 2763 标准中农药相关数值;100 为安全因子,表示每日由中药材及其制品中摄取的农药的量不大于日总暴露量(包括食物和饮用水)的1%,此数值参考《中国药典》2020年版四部“9302 中药有害残留物限量制定指导原则”农药限量制定安全因子数值。
通过以上公式计算菊花中4种农药MRL值的HQc,结果见表1。一般认为,HQc大于1,风险不可接受;HQc小于1,风险可接受。结果发现,氟啶胺的HQc大于1,其余3种农药的HQc均低于1。故氟啶胺暂不转化,仅对吡蚜酮、吡虫啉和啶酰菌胺进行转化。
通过菊花标准限量风险评估及对检测方法的科学性和准确性验证后,根据已拟定的转化原则[12],同时兼顾中药产业经济效益,将检测结果没有超限或超限率小于等于10%的农药品种转化为我国国家药品标准,超标率大于10%的,暂不予转化,但需向国家药监局及主要地政府提出产品风险预警。
根据以上原则,最终将GB 2763菊花中3个农药残留限量标准转化为菊花药品标准,分别为:吡蚜酮(0.1 mg·kg-1 )、吡虫啉(2 mg·kg-1 )、啶酰菌胺(30 mg·kg-1 )。
