OBJECTIVE To establish a universal method for determining the refractive index increment(dn/dc)of sodium hyaluronate(SH),laying the foundation for achieving accurate determination of SH molecular weight and molecular weight distribution. METHODS Various instruments, mobile phases,and samples were used to study the dn/dc of SH. RESULTS SH was more stable in 0.1 mol·L-1 sodium nitrate solution,and the relative standard deviation(RSD)of dn/dc was less than 1.0%(n=3). CONCLUSION The newly established determination method is simple,fast,and accurate,solving the problem of difficult determination of SH dn/dc both domestically and internationally for many years.It also provides new methods and ideas for determining the dn/dc of other polymer substances.

目的 建立玻璃酸钠(sodium hyaluronate,SH)折光指数增量(dn/dc值)的通用测定方法,为实现SH相对分子质量与相对分子质量分布的准确测定奠定基础。方法 采用不同仪器、流动相和样品对SH的dn/dc值进行研究。结果 SH在0.1 mol·L-1硝酸钠溶液中更稳定,dn/dc值的相对标准偏差(RSD)均小于1.0%(n=3),建立了dn/dc值的准确测定方法。结论 新建测定方法简便、快速、准确,解决了SH的dn/dc值测定困难的问题,也为其他高分子物质相对分子质量与相对分子质量分布的精准测定提供了新的方法。

目的 建立玻璃酸钠(sodium hyaluronate,SH)折光指数增量(dn/dc值)的通用测定方法,为实现SH相对分子质量与相对分子质量分布的准确测定奠定基础。方法 采用不同仪器、流动相和样品对SH的dn/dc值进行研究。结果 SH在0.1 mol·L-1硝酸钠溶液中更稳定,dn/dc值的相对标准偏差(RSD)均小于1.0%(n=3),建立了dn/dc值的准确测定方法。结论 新建测定方法简便、快速、准确,解决了SH的dn/dc值测定困难的问题,也为其他高分子物质相对分子质量与相对分子质量分布的精准测定提供了新的方法。

OBJECTIVE To establish a universal method for determining the refractive index increment(dn/dc)of sodium hyaluronate(SH),laying the foundation for achieving accurate determination of SH molecular weight and molecular weight distribution. METHODS Various instruments, mobile phases,and samples were used to study the dn/dc of SH. RESULTS SH was more stable in 0.1 mol·L-1 sodium nitrate solution,and the relative standard deviation(RSD)of dn/dc was less than 1.0%(n=3). CONCLUSION The newly established determination method is simple,fast,and accurate,solving the problem of difficult determination of SH dn/dc both domestically and internationally for many years.It also provides new methods and ideas for determining the dn/dc of other polymer substances.

玻璃酸钠(sodium hyaluronate,SH),也称为玻尿酸钠或透明质酸钠,系从鸡冠或微生物发酵液中提取的酸性黏多糖,由D-葡萄糖醛酸和N-乙酰-D-氨基葡萄糖双糖单位构成的糖胺聚糖钠盐[1]。因具有黏弹性高、生物相容性强及无毒副作用等优点[2],被广泛用于眼科、关节疾病、软组织修复、药物递送系统与组织工程支架等临床医药领域,近年来,在靶向治疗方面的应用也越来越多[3-15]
不同相对分子质量SH的生物活性存在较大差异,甚至与同一受体结合会表现出完全相反的生物效应[16-19]。在医药领域,为保证SH的安全有效性,欧洲药典11.0(EP11.0)[20]、日本药局方18版(JP18)[21] 及我国的国家药品标准WS1-(x072)2011Z[1] 均收录了不同用途SH相对分子质量与相对分子质量分布检查项,方法主要为黏度法和体积排阻色谱法(size exclusion chromatography,SEC),但上述方法都存在一定的局限。黏度法专属性差,不能给出相对分子质量分布信息;SEC法没有对照品,不同品牌市售对照品会导致测定误差较大[22-23]
分子排阻色谱-多角度激光光散射联用法(SEC-MALLS)不需要对照品,能同时获得相对分子质量与相对分子质量分布、分子尺寸、构造形态、高分子支化度等信息[24-25],是一种能够全面掌握SH关键质量属性的方法。该方法的关键是准确测定折光指数增量(dn/dc值)后计算高分子摩尔质量[26] 。而SH的dn/dc值测定操作烦琐,不易找到平台峰,重现性差,导致目前国内外相对分子质量标准一直无法统一和提高。
本研究通过优化获得了可准确测定国内外多种不同相对分子质量范围的SH dn/dc值的溶液体系;并通过比较SH在不同流动相、方法和仪器的结果差异,建立了采用梯度混合仪自动化测定dn/dc值的方法,能快速、准确地测定其相对分子质量与相对分子质量分布,为标准提高奠定基础,为国内外产品的安全有效性提供数据支持。
高效液相色谱仪(LC-20AD泵,DGU-20A3脱气装置,SIL-20AC自动进样器,日本岛津公司);Calypso Ⅱ梯度混合仪、多角度激光光散射仪(DAWN HELLOS Ⅱ)、DNDC仪(Optilab T-rEX)(美国怀雅特公司);Mittler XPE205电子天平(美国梅特勒集团);CALYPSO 2.4.1.6和ASTRA 8.1.0.14采集及处理软件。
国内外不同相对分子质量范围的SH样品10个,见表1(A1,B2~B6,C7~C8,D9~D10);硝酸钠、硫酸钠、氯化钠为分析纯;屈臣氏纯净水;Proclin 300(色谱纯);归一化样品右旋糖酐(SIGMA公司)。
流动相:0.2 mol·L-1氯化钠溶液(称取氯化钠11.9 g,加屈臣氏水溶解至1 000 mL,双层0.22 μm滤膜过滤,再加0.2 mL Proclin 300,超声10 min,即得)、0.1 mol·L-1硫酸钠溶液(另取硫酸钠14.2 g,同0.2 mol·L-1氯化钠溶液方法制备)、0.1 mol·L-1硝酸钠溶液(另取硝酸钠8.5 g,同0.2 mol·L-1氯化钠溶液方法制备)。
供试品溶液制备:精密称取A1样品约25 mg(按干燥品计),置50 mL量瓶中,分别加上述3种流动相5 mL,手动振荡使样品漂浮,再次加约40 mL流动相静置过夜,使样品充分溶胀,定容至刻度,混匀。用移液枪分别取该溶液0、1、2、3、4、5、6 mL,流动相6、5、4、3、2、1、0 mL,配成7个浓度梯度,依次注入已用归一化对照品校正的DAWN HELLOS Ⅱ,连接Optilab T-rEX作为浓度源,平行进样2次,通过ASTRA软件计算其结果。
精密称取SH(按干燥品计)各20 mg,置50 mL量瓶中,分别加0.1 mol·L-1硫酸钠溶液、0.1 mol·L-1硝酸钠溶液5 mL,手动振荡使样品漂浮,再次加约40 mL流动相静置过夜,使样品充分溶胀,定容至刻度,混匀。分别称取该溶液0、1、2、3、4、5、6 mL,流动相6、5、4、3、2、1、0 mL,配成7个浓度梯度,依次注入Optilab T-rEX,通过ASTRA软件计算其结果。
供试品溶液制备同“2.2.1”项,流动相为0.1 mol·L-1硝酸钠溶液。
色谱条件及采集方法:pump 1接供试品溶液,pump 3接流动相,分6个线性梯度,进样体积为2 mL,流速为1.0 mL·min-1,延迟时间60 s,Optilab T-rEX检测温度为35 ℃,平行测定3次;同时用CALYPSO和ASTRA软件进行数据的采集与处理,其中CALYPSO 软件直接用Optilab T-rEX作为浓度源进行计算,ASTRA软件采用理论浓度代入进行计算得到dn/dc值。
为评估SH在不同流动相中的溶解状态,本研究引入了第二维里系数(A2)对SH在溶液中的状态进行表征。A2表征了高分子“链段”与溶剂分子之间的相互作用,是检验溶剂良性程度的指标,A2越大,“链段”与溶剂分子间的扩张作用越强,“链段”越舒展,证明当前溶剂为更良溶剂[27]。本研究采用Zimm图法测定了0.5 mg·mL-1的SH在0.2 mol·L-1氯化钠溶液、0.1 mol·L-1硫酸钠溶液和0.1 mol·L-1硝酸钠溶液中的A2,结果见表2图1。所测SH在0.1 mol·L-1硝酸钠溶液中的A2最大,可见0.1 mol·L-1硝酸钠溶液为更适合SH的良溶剂。
国内外法定标准中均使用0.2 mol·L-1氯化钠溶液做溶剂。研究团队之前已经采用“2.2.1”项下的方法测定了SH在0.2 mol·L-1氯化钠溶液中的dn/dc值,实验发现进口企业不同批次间结果一致,企业之间甚至同一国产企业不同批次间差异较大[22]。为证明0.1 mol·L-1硝酸钠溶液为SH的更良溶剂,采取称重法测定SH样品分别在0.1 mol·L-1硫酸钠溶液、0.1 mol·L-1硝酸钠溶液中的dn/dc值,结果见表3。结果发现,10种样品测得结果差异大小与表2中各溶剂的A2大小相互验证,说明溶剂的良性程度与SH在溶液中dn/dc值的稳定性程度一致,所以选择0.1 mol·L-1硝酸钠溶液作为后续实验的流动相。
分别称取2份A1样品连续测定5次结果的相对标准偏差(RSD)<1.0%,该方法重复性良好。重复性测定结果见表4
线性结果见图2,拟合度r均大于0.999 0,ASTRA和 CALYPSO 软件分别为0.999 5、0.999 6,表明SH在0.025 9~0.492 3 mg·mL-1内线性良好。
Calypso Ⅱ测定结果见表5,同一样品平行测定3次RSD均小于1.0%,理论浓度和示差折光检测器作为浓度源所得结果误差小于1.2%,10种样品间的RSD为1.4%,证明Calypso Ⅱ梯度混合法在0.1 mol·L-1硝酸钠溶液中准确性和重复性良好,与称重法所得结果相互印证,可以替代称重法测定SH的dn/dc值。
实验采用的Calypso Ⅱ梯度混合仪可以自动制备浓度梯度进样,无需称重计算即可获得准确的溶液浓度,国内外暂无使用该仪器进行SH的dn/dc值测定的相关报道。对比称重法需最少称重配制6个浓度溶液后手动进样的烦琐操作,新建方法实现了自动化分析,仅需配制1个原浓度为0.4 mg·mL-1的SH溶液,就能通过Calypso Ⅱ梯度混合仪获得dn/dc值,简便、准确、重复性良好,见图3。但该方法对测定交联SH dn/dc值的适用性还有待考察。
本研究通过测定3种流动相中SH的A2大小和dn/dc值在不同流动相中的稳定性数据相互佐证,确定0.1 mol·L-1硝酸钠溶液做流动相,获得了可稳定测定dn/dc值的溶液体系;新建Calypso Ⅱ梯度混合仪自动化测定方法简便、快速、准确,重复性良好,解决了SH的dn/dc值测定繁琐的问题。目前国内外对SH dn/dc值的研究较少,通过测定A2来挑选合适溶解体系的相关研究尚属空白。通过本研究和所建方法能为高分子药物的相对分子质量测定和产品的标准提高提供新的依据。
