OBJECTIVE To establish a method for preparing total saponins from Schizocapsa plantaginea Hance and establish a corresponding quality control method. METHODS The total saponins were extracted from the Schizocapsa plantaginea Hance tubers using a water bath reflux method, followed by purification of the crude extract through solvent extraction and macroporous resin. Subsequently, the purified product of total saponins of Schizocapsa plantaginea Hance(SSPHs) was prepared, and its quality was assessed using thin-layer chromatography, UV-visible spectrophotometry, and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS) combined method. RESULTS The developed preparation method resulted in a yield of total saponin products from the Schizocapsa plantaginea Hance tuber ranging from 0.29% to 2.81%, with a purity from 57.30% to 77.02%. After detection, saponin substances such as Schizocapsa Plantaginea Hance Ⅰ(SSPH Ⅰ) and Schizocapsa plantaginea Hance Ⅱ(SSPH Ⅱ) were found in the total saponin products. The content of SSPH Ⅰ was 10.77-85.60 mg·g-1 and the content of SSPH Ⅱ was 0.19-4.75 mg·g-1. CONCLUSION The established preparation method is simple, efficient, and environmentally friendly, with high product quality. The established qualitative and quantitative detection method can effectively reflect the quality of SSPHs and provide method reference for the quality control of SSPHs.

目的 建立从裂果薯药材中制备总皂苷的方法,并建立总皂苷的质量控制方法。方法 采用水浴回流的方法从裂果薯块茎粉末中提取总皂苷粗提取物,用溶剂萃取和大孔树脂对粗提物进行富集纯化,制成裂果薯总皂苷(SSPHs),分别建立薄层色谱法、紫外可见分光光度法和超高效液相色谱-串联质谱(UHPLC-MS)联用对SSPHs进行质量评价。结果 所建立的制备方法从裂果薯块茎中制备总皂苷制品得率0.29%~2.81%,总皂苷纯度57.30%~77.02%,经检测其中含有裂果薯皂苷 Ⅰ(SSPH Ⅰ)和裂果薯皂苷 Ⅱ(SSPH Ⅱ)等皂苷类物质,SSPH Ⅰ含量为10.77~85.60 mg·g-1,SSPH Ⅱ含量为0.19~4.75 mg·g-1结论 所建立的制备方法简单、高效、环保,产品品质高,所建立的定性、定量检测方法能有效反映SSPHs的质量情况,可为裂果薯总皂苷的质量控制提供方法参考。

OBJECTIVE To establish a method for preparing total saponins from Schizocapsa plantaginea Hance and establish a corresponding quality control method. METHODS The total saponins were extracted from the Schizocapsa plantaginea Hance tubers using a water bath reflux method, followed by purification of the crude extract through solvent extraction and macroporous resin. Subsequently, the purified product of total saponins of Schizocapsa plantaginea Hance(SSPHs) was prepared, and its quality was assessed using thin-layer chromatography, UV-visible spectrophotometry, and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS) combined method. RESULTS The developed preparation method resulted in a yield of total saponin products from the Schizocapsa plantaginea Hance tuber ranging from 0.29% to 2.81%, with a purity from 57.30% to 77.02%. After detection, saponin substances such as Schizocapsa Plantaginea Hance Ⅰ(SSPH Ⅰ) and Schizocapsa plantaginea Hance Ⅱ(SSPH Ⅱ) were found in the total saponin products. The content of SSPH Ⅰ was 10.77-85.60 mg·g-1 and the content of SSPH Ⅱ was 0.19-4.75 mg·g-1. CONCLUSION The established preparation method is simple, efficient, and environmentally friendly, with high product quality. The established qualitative and quantitative detection method can effectively reflect the quality of SSPHs and provide method reference for the quality control of SSPHs.

裂果薯属于广西特色中草药,拉丁学名Schizocapsa plantaginea Hance,属于百合目,蒟蒻属科,裂果薯属植物,别名水田七、水萝卜、屈头鸡等,多生长在水沟边和田埂上,主要分布于广西、广东、云南、贵州以及湖南等地,在东南亚地区也有分布。药材资源丰富,民间常用于清热解毒、止咳化痰、凉血散瘀等,也可外用治疗跌打损伤,疮疡中毒。裂果薯属广西民族药中草药和区域性特色中药材,已有一定种植规模,《广西壮族自治区壮药材质量标准》[1]已建立了裂果薯药材的质量标准,但总皂苷的质量标准未见报道。本课题组研究中发现,裂果薯中皂苷类化合物具有明显抑制肝癌细胞增殖的作用[2-3]。其中,裂果薯皂苷Ⅰ(SSPH Ⅰ)和裂果薯皂苷Ⅱ(SSPH Ⅱ)抑制肝癌细胞增殖的作用强于总皂苷,说明SSPH Ⅰ和SSPH Ⅱ可能是裂果薯皂苷抗肿瘤的有效成分[4-5]。Sun等[6]研究发现,裂果薯总皂苷(SSPHs)通过丝裂原活化蛋白激酶信号传导从而抑制人肝癌细胞生长。Zhou等[7]研究发现,SSPHⅠ通过诱导肝癌细胞的活性氧(ROS)积累和ERK1/2信号通路抑制自噬并诱导细胞凋亡。Zhou等[8]研究发现,SSPHⅠ还通过抑制MAPK/ERK1/2和PI3K/AKT/mTOR信号通路抑制上皮细胞-间充质转化以及非小细胞肺癌的侵袭和迁移。目前研究表明,SSPHⅠ还可通过调节铁代谢诱导HepG2细胞的铁死亡,这对肝癌的治疗具有重要意义[9]。大孔树脂性质稳定,不溶于酸、碱及有机溶剂,用于分离纯化皂苷化合物已有较多报道[10]。SSPHs纯化目前多用大孔树脂,总皂苷成分复杂,纯化过程易受到大孔树脂种类、上样和洗脱工艺的影响,导致产物质量差异较大。本研究对SSPHs的大孔树脂纯化工艺进行研究,并以SSPH Ⅰ和SSPH Ⅱ作为检测指标建立相应的质量评价方法,为深入研究裂果薯的药用价值提供科学依据。
SSPH Ⅰ对照品(自制,纯度97.5%);SSPH Ⅱ对照品(自制,纯度98.0%);无水乙醇(成都市科隆化学品有限公司,分析纯);体积分数95%乙醇、石油醚、乙酸乙酯、正丁醇、高氯酸、冰乙酸、氨水、香草醛均为分析纯;薄层层析硅胶板(青岛海洋化工厂分厂,50 mm×100 mm);甲醇、乙腈均为色谱纯;超纯水;大孔树脂(型号: AB-8、D101、HPD100、HPD400、ADS7,东鸿化工有限公司)。
裂果薯药材信息见表1,经广西中医药研究院中药研究所黄云峰教授鉴定为蒟蒻薯科裂果薯属植物裂果薯(Schizocapsa plantaginea Hance)的干燥块茎。
岛津UV2700型紫外-可见分光光度计(日本岛津公司);Waters ACQUITYH-Class超高效液相色谱仪(美国沃特世科技有限公司);Qda质谱检测器(美国沃特世科技有限公司);TNF-50型多功能提取浓缩回收机组(青州市精诚医药装备制造有限公司);HH-S型数显恒温水浴锅(金坛市医疗仪器厂);AR-224CN型万分之一电子天平(美国奥豪斯仪器有限公司);KQ500VDE型双频数控超声波清洗器(昆山市超声仪器有限公司)。
称取裂果薯药材,粉碎,过2号筛,据文献报道的提取和萃取工艺条件[4],按药材质量(g)-溶液体积(mL)=1∶10加入体积分数80%乙醇溶液,80 ℃回流提取4次,每次2 h,合并提取液、浓缩蒸干,即得醇提物。用适量纯化水将醇提物制成混悬液,石油醚、乙酸乙酯、水饱和正丁醇依次萃取醇提物,将正丁醇部位萃取液浓缩蒸干,即得SSPHs粗品,备用。SSPHs粗品用适量水溶解后上样到大孔树脂吸附后,用不同浓度乙醇洗脱,洗脱液浓缩蒸干,即得SSPHs,制备流程图见图1
取SSPH Ⅰ对照品适量,精密称定,用甲醇溶解,制成0.102 mg·mL-1的对照品溶液。
取SSPHs粗提物约10 mg,精密称定,用甲醇溶解,制成1 mg·mL-1的供试品溶液。
精密吸取上述对照品溶液0.1、0.2、0.4、1、3 mL,挥尽乙醇,精密加入5%香草醛-冰乙酸溶液0.2 mL和高氯酸0.8 mL,摇匀,60 ℃水浴加热20 min,取出,冰水浴冷却10 min,精密加入5 mL冰乙酸,摇匀。在452 nm波长处测定吸光度,以吸光度为纵坐标,浓度为横坐标绘制标准曲线,外标法测定SSPHs含量。
静态吸附实验:分别称取预处理过的5种型号的大孔树脂:AB-8、D101、HPD100、HPD400、ADS7型树脂各1 g置于50 mL具塞三角瓶中,分别加入1 g总皂苷粗提物,用水溶解后,密封,摇床上震摇12 h,振速80 r·min-1,抽滤,将抽滤后的树脂用适量蒸馏水洗去残留药液,洗涤液与滤液合并,按“2.2”项下方法测定合并液中总皂苷的含量,按公式1计算吸附率。
吸附率(%)=(上样量-洗涤液和滤液中总皂苷量)/上样量×100%
静态解析实验:将上述吸附饱和的树脂分别转移至50 mL锥形瓶中,加入体积分数95%乙醇35 mL,密封,摇床上震摇12 h,振速80 r·min-1,抽滤,按“2.2“项下方法测定滤液中总皂苷的含量,按公式2计算解析率。

取处理好的AB-8型号大孔树脂装柱4根,每根装量15 g,分别取质量浓度为0.2、0.1、0.05、0.01 g·mL-1的总皂苷粗提物水溶液21、42、84、420 mL,以48 mL·h-1流速上样,收集流出液,再用120 mL纯净水洗脱,合并流出液和水洗液,分别测定合并液中总皂苷含量,计算吸附率。
取处理好的AB-8型号大孔树脂装柱4根,每根装量15 g,取0.2 g·mL-1的总皂苷粗提物水溶液20 mL上样,上样流速分别为24、48、72、96、120 mL·h-1,收集流出液,再用120 mL水洗脱,合并流出液和水洗液,分别测定合并液中总皂苷含量,计算吸附率。
取处理好的AB-8型号大孔树脂15 g,装柱,取质量浓度为0.2 g·mL-1的总皂苷粗提物水溶液,以72 mL·h-1的流速上样,分段收集流出液,每10 mL为1份,分别测定流出液中总皂苷含量,以未吸附的总皂苷浓度为纵坐标,份数为横坐标绘制泄露曲线。
取处理好的AB-8型号大孔树脂15 g,装柱,取0.2 g·mL-1的总皂苷粗提物水溶液21 mL,以72 mL·h-1的流速上样,待吸附完全后,用纯净水120 mL洗脱后,再依次用体积分数10%、20%、30%、50%、60%、70%、95%的乙醇以48 mL·h-1流速洗脱120 mL,收集洗脱液,分别测定洗脱液中总皂苷的含量并计算总皂苷纯度。
取处理好的AB-8型号大孔树脂15 g,装柱,取0.2 g·mL-1的总皂苷水溶液21 mL,以72 mL·h-1的流速上样,待吸附完全后,用纯净水120 mL洗脱,再依次用体积分数30%乙醇溶液240 mL、70%乙醇溶液192 mL洗脱,洗脱流速48 mL·h-1,分段收集各浓度乙醇洗脱液,每24 mL收集1份,分别测定洗脱液中总皂苷含量并计算总皂苷纯度。
按考察的最佳工艺制备裂果薯总皂苷,平行3份,分别收集洗脱液,测定总皂苷含量并计算纯化前后总皂苷纯度。
取SSPH Ⅰ对照品适量,精密称定,用甲醇溶解,制成1.02 mg·mL-1的SSPH Ⅰ对照品溶液;取SSPH Ⅱ对照品适量,精密称定,用甲醇溶解,制成1.36 mg·mL-1的SSPH Ⅱ对照品溶液。
取SSPHs适量,精密称定,用甲醇溶解,制成5 mg·mL-1的SSPHs供试品溶液。
取上述供试品溶液和SSPH Ⅰ、SSPH Ⅱ对照品溶液各5 μL,点于同一硅胶G薄层板上,以正丁醇-乙酸乙酯-水=4∶1∶5(上层)为展开剂,展开,喷以体积分数10%硫酸乙醇溶液,105 ℃加热至斑点显色清晰,在可见光和365 nm紫外光下检视。
不同品牌薄层板的考察:按“2.4.3”项下方法,在其他条件相同的情况下分别对MERCK和青岛海洋的薄层色谱板进行层析考察。
不同展开温度的考察:按“2.4.3”项下方法,分别在4和25 ℃条件下进行层析考察。
不同展开湿度的考察 按“2.4.3”项下方法,分别对相对湿度在55%和83%条件下进行层析考察。
不同点样量的考察:按“2.4.3”项下方法,取供试品溶液分别点样5 μL和2 μL,对照品溶液点样5 μL,进行层析考察。
分别取SSPH Ⅰ和SSPH Ⅱ对照品适量,精密称定,用甲醇溶解,制成含SSPH Ⅰ2.3 mg·mL-1 、SSPH Ⅱ1.22 mg·mL-1的对照品储备溶液。精密移取SSPH Ⅰ标准储备溶液7 mL,SSPH Ⅱ标准储备溶液0.5 mL置于同一25 mL量瓶中,用甲醇溶解并定容至刻度,制成含SSPH Ⅰ644 μg·mL-1,SSPH Ⅱ24.4 μg·mL-1的混合对照品储备溶液。
取SSPHs 10 mg,精密称定,置于10 mL量瓶中,加甲醇溶解并定容至刻度,制成1 mg·mL-1的溶液,使用0.22 μm微孔滤膜滤过,取续滤液作为供试品溶液。
取“2.3.1”项下SSPHⅠ、SSPHⅡ对照品储备溶液,分别注入高分辨率质谱仪中,测定SSPHⅠ、SSPHⅡ离子的精确质量数。
色谱柱:Waters BEH C18色谱柱(2.1 mm×100 mm,1.7 μm);流动相:乙腈(A)-0.1%氨水(B);梯度洗脱程序(A:0~1 min,50%~50%;1~2 min,50%~85%;2~4.5 min,85%~60%;4.5~6 min,60%~60%;6~6.5 min,60%~50%;6.5~10 min,50%~50%);流速0.35 mL·min-1;进样体积5 μL;柱温40 ℃;质谱条件:ESI离子源;负离子模式检测;毛细管电压为800 V;锥孔电压为15 V;采用选择性离子检测(SIM)模式:0~10 min,1 029.52监测SSPH Ⅰ;0~10 min,867.47监测SSPH Ⅱ;探头温度为600 ℃;采样速率每秒10点;载气N2;载气流速10 L·min-1
精密吸取“2.5.1”项下混合对照品储备溶液5,4,3,2,1 mL,0.5 mL置于25 mL量瓶中,分别加入甲醇至刻度,摇匀。使用0.22 μm微孔滤膜滤过,取续滤液作为混合对照品溶液。按“2.5.4”项下检测方法分别进样分析,以各成分的浓度ρ(μg·mL-1)为横坐标,峰面积y为纵坐标进行线性回归。
精密度试验:取同一份混合对照品溶液,按“2.5.4”项下检测方法连续进样6次,记录各成分的峰面积并计算相对标准偏差(RSD)值。
稳定性实验:取云南SSPHs,按“2.5.2”项下方法制成供试品溶液,每间隔2 h进样1次,记录各成分的峰面积并计算RSD值,观察样品在12 h内的稳定性。
重复性实验:取云南SSPHs,按“2.5.2”项下方法制成供试品溶液,平行操作6份,按“2.5.4”项下检测方法进样测定,记录峰面积,并计算含量和RSD值。
加样回收试验:取云南SSPHs约5 mg,分别加入SSPH Ⅰ、SSPH Ⅱ含量为39.77、1.89 μg·mL-1的混合对照品,按“2.5.2”项下方法制成供试品溶液,平行操作6份,按“2.5.4”项下检测方法进样,记录峰面积,并计算加样回收率和RSD值。
日间精密度实验:由不同实验人员分别在第一天及第二天取云南SSPHs样品各6份,按“2.5.2”项下方法制成供试品溶液,按“2.5.4”项下检测方法进样,记录峰面积,并计算含量和RSD值。
色谱柱耐用性考察:精密称定广东SSPHs 13.6 mg,按“2.5.2”项下方法制成供试品溶液,使用3根不同厂家的C18色谱柱:色谱柱①:Waters BEH C18(2.1 mm×100 mm,1.7 μm)色谱柱②:Agilent Eclipse Plus C18 RRHD(2.1 mm×100 mm,1.8 μm)色谱柱③:Thermo Hypersil GOLD C18(2.1 mm×100 mm,1.9 μm),按“2.5.4”项下检测方法进样,记录峰面积,并计算含量和RSD值。
按“2.2.2”项下方法制备不同产地SSPHs,按“2.2.3”项下方法测定含量,重复3次,计算总皂苷产率和纯度,结果见表2。不同产地样品中总皂苷纯度差异较大,广西、广东、云南产药材中SSPHs纯度在70.00%~77.02%之间,纯度没有明显差异,贵州产药材总皂苷纯度57.30%,与其他3个产地相比纯度差异较大。
AB-8型大孔树脂吸附率及解吸率均最高,见表3,因此选择AB-8型大孔树脂分离纯化SSPHs。
在相同上样量条件下,吸附率随着上样液质量浓度的增加而增加,表明在0.2 g·mL-1上样液质量浓度下,没有超过树脂的吸附容量,见图2。但上样质量浓度太高会变得较黏稠,不利于树脂柱上样,最终选择上样液质量浓度为0.2 g·mL-1
吸附率随着上样流速的增加,呈现先升高后下降的趋势,在72 mL·h-1时吸附率最高,见图3。可能是流速太慢造成大孔树脂堵塞,药液无法流过某些大孔树脂层表面就流出来,导致泄漏增加;流速过快时大孔树脂和样品接触时间短,样品不能在大孔树脂上充分吸附导致泄漏增加,因此最终选择上样流速为72 mL·h-1
当累计上样量达到40 mL时,泄漏量开始明显增大,说明树脂柱吸附量达到饱和,见图4。实际生产中,一般取最大上样量的70%作为实际上样量,确定上样量为21 mL。
采用体积分数10%、20%、30%乙醇洗脱,总皂苷含量较低,总皂苷纯度分别为54.30%、39.34%、48.19%,说明该洗脱液中含有许多非皂苷类物质;体积分数50%、60%、70%乙醇洗脱液中总皂苷含量较高,总皂苷纯度分别为71.21%、88.82%、70.74%,见图5,因此选择体积分数30%乙醇作为除杂溶剂,体积分数70%乙醇作为洗脱溶剂。
体积分数30%乙醇洗脱条件下,第6个流份以后,总皂苷含量几乎没有变化,见图6,纯度均低于50%,继续洗脱除杂效率也不会有明显提升;体积分数70%乙醇洗脱条件下,第4个流分以后,总皂苷已基本洗脱完全,见图7,且前4个流分纯度均高于70%,为节约溶剂,提高效率,确定体积分数30%乙醇用量为144 mL,体积分数70%乙醇用量为96 mL。
AB-8大孔树脂纯化后3批SSPHs的平均纯度为77.02%,纯化倍数1.78,见表4
两个厂家的硅胶G薄层色谱板分离效果均良好,见图8
4 ℃展开条件下拖尾变明显,但仍可见两个特征性斑点,不影响薄层色谱的鉴别,见图9
在83%和55%相对湿度条件下薄层色谱板分离效果均良好,见图10
由于SSPHs中SSPH Ⅱ含量较低,在点样2 μL时特征性斑点几乎不可见,故点样量在5 μL为宜,见图11
供试品色谱中,在对照品色谱相应的位置上显相同的荧光斑点。薄层色谱鉴别结果见图12
混合对照品质谱图结果见图13。样品和混合对照品结果见图14
SSPH Ⅰ和SSPH Ⅱ的线性关系良好,可用于含量测定,见表5
SSPH Ⅰ的峰面积RSD值为2.26%(n=6),SSPH Ⅱ的峰面积RSD值为2.04%(n=6),表明仪器精密度良好。
结果显示SSPH Ⅰ和SSPH Ⅱ的平均含量分别为68.37、2.02 μg·mL-1,RSD值分别为1.96%、2.90%(n=6),表明方法的重复性良好。
结果显示SSPH Ⅰ和SSPH Ⅱ的峰面积RSD值分别为2.88%、2.38%,表明样品在12 h内稳定性良好。
6批样品回收率范围在95%~105%之间,SSPH I和 SSPH II的平均回收率分别为100.80%及102.42%,RSD分别为1.80%、2.17%(n=6),见表6,表明方法准确度良好。
SSPH Ⅰ和SSPH Ⅱ含量均值分别为70.17和2.09 mg·g-1,RSD分别为1.61%和2.66%,表明日间精密度良好。
云南样品中SSPH Ⅰ和SSPH Ⅱ含量均值分别为85.73和3.76 mg·g-1,RSD分别为1.24%和1.49%,证明该方法耐用性良好。
取适量不同产地SSPHs,精密称定,用甲醇溶解,按2.3项下检测方法测定不同产地裂果薯总皂苷中SSPH Ⅰ和SSPH Ⅱ的含量,结果见表7
按“2.2.1”和“2.2.2”项下方法制备对照品溶液和SSPHs供试品溶液,按“2.2.3”项下方法显色后在400~800 nm波长下进行扫描,按紫外可见分光光度法下方法制备供试品和对照品,显色后在400~800 nm波长下进行扫描,结果对照品和供试品在452 nm处有最大吸收峰,故确定检测波长为452 nm,见图15。实验考察了方法的线性、精密度、重复性、加样回收率和稳定性,线性r2=0.999 9,平均加样回收率为100.60%,RSD为1.34%,其余方法学考察RSD均小于3%,符合含量测定的要求。
参照皂苷类成分常用展开系统,分别考察了3种展开剂系统,①正丁醇-乙酸乙酯-水=4∶1∶5(上层)、②三氯甲烷-甲醇-水=5∶1.5∶0.5(上层)、③三氯甲烷-甲醇-甲酸=4∶1∶0.25。结果②号展开剂中SSPH ⅠRf值太小,导致其未能与样品点完全分开;③号展开剂出现明显拖尾,导致SSPH Ⅱ斑点不清晰;①号展开剂SSPH Ⅰ、SSPH Ⅱ明显分开,虽有一些拖尾现象,但不影响SSPH Ⅰ与SSPH Ⅱ的鉴别,故采用正丁醇-乙酸乙酯-水=4∶1∶5(上层)作为展开溶剂。喷上显色剂后薄层色谱板加热超过7 min开始碳化,影响鉴别,加热时间短,供试品中SSPH Ⅱ在可见光下斑点不清晰,但在365 nm紫外光下可观察到斑点,因此本实验薄层色谱板均在365 nm紫外光下检视。
SSPH Ⅰ、SSPH Ⅱ属于甾体皂苷类,结构中仅含有一个孤立双键,在紫外波长下属于末端吸收,专属性和灵敏度较低,因此紫外检测器不太适合于SSPHs成分的含量测定。本实验采用单重四级杆质谱仪,相比于紫外检测器对待测物质具有更高的选择性和灵敏度,与其它质谱检测器相比使用方便、成本低廉,故本研究使用质谱测定裂果薯皂苷中 SSPH Ⅰ、SSPH Ⅱ含量。
预实验中,最初使用乙腈-水作为流动相,但峰形较差,难以满足含量测定的要求,考察了乙腈-0.1%甲酸、乙腈-5 mmol·L-1乙酸铵、乙腈-0.1%氨水流动相系统对色谱峰的影响,乙腈-0.1%氨水流动相系统色谱峰峰形良好,保留时间稳定,无干扰峰,没有拖尾现象,能够达到定量要求,因此选择乙腈-0.1%氨水作为流动相。皂苷类成分复杂,使用HPLC进行分析需要耗费较长时间,如重楼皂苷采用HPLC分析需要40 min以上的时间[14]。SSPH Ⅰ与SSPH Ⅱ极性接近,普通高效液相色谱条件难以有效分离两种成分,超高效液相色谱采用了更微小颗粒的固定相,超高压输液泵,因此可以采用更短的色谱柱,在检测速度、灵敏度和分离度上都有了明显的提升,10 min即可完成SSPHs的成分分析,大大提高了分析效率。
本研究中,确定最佳工艺:取质量浓度为0.2 g·mL-1的总皂苷粗提物水溶液以72 mL·h-1流速通过AB-8型大孔树脂,完全吸附后依次用120 mL的水和144 mL的体积分数30%乙醇洗脱,再用96 mL的体积分数70%乙醇洗脱,洗脱流速72 mL·h-1,收集体积分数70%乙醇洗脱液,减压干燥,即得SSPHs。紫外可见分光光度法测定不同产地SSPHs含量,计算得到不同产地SSPHs纯度为57.30%~77.02%。使用Waters BEH C18色谱柱,乙腈-0.1%氨水溶液为流动相,10 min内梯度洗脱,色谱峰峰形良好,系统适用性考察均符合要求。不同产地SSPHs中SSPH Ⅰ含量为10.77~85.60 mg·g-1,SSPH Ⅱ含量为0.19~4.75 mg·g-1。本研究测定了SSPHs及SSPHs中SSPH Ⅰ、SSPH Ⅱ的含量,并进行了比较分析,为裂果薯药材质量评价提供了科学依据。
