罗布麻(Apocynum venetum L.)为夹竹桃科罗布麻属植物,其干燥叶是维吾尔族习用药材,兼具药用与茶饮价值,现代药理证实其具有降压、抗氧化、抗炎等多种活性[12]。此前,对其抗炎活性的研究大多聚焦于多糖、三萜类成分[3-4],而罗布麻叶醇提物(the extract of Apocynum venetum leaves,AVLE)主要活性成分为黄酮、酚酸类,与上述成分存在本质差异[5]。目前,针对AVLE抗炎分子机制的研究少见报道。因此,本研究以脂多糖(lipopolysaccharide,LPS)诱导RAW264.7巨噬细胞的体外炎症模型,探究AVLE的抗炎活性及分子机制,为罗布麻叶在抗炎领域的研究与开发提供科学依据。
RAW264.7小鼠单核巨噬细胞系,由新疆医科大学惠赠。
罗布麻叶,阿勒泰戈宝茶股份有限公司;穿心莲内酯(andrographolide,AG)购自MCE公司;COX-2抗体(12282S),p-JNK抗体(4668S),JNK抗体(9252S),p-ERK1/2抗体(9106S),ERK1/2抗体(4696S),p-p38 MAPK抗体(4511S),p38 MAPK抗体(9212S),Nrf2抗体(12721S)均购自CST公司;HO-1抗体(Abcam,ab68477);iNOS抗体(Affinity Biosciences,AF0199);多功能酶标仪(Molecular Devices,SpectraMax M5)。
罗布麻叶经50%乙醇回流提取2次,每次1 h,经HPD300树脂纯化,50%乙醇洗脱后浓缩干燥,制得备用。
实验分为Control组、AG组(5 µmol·L-1)和AVLE组(31.25、62.5、100、125、200、250 µg·mL-1),CCK-8法检测细胞存活率,确定无毒给药浓度。
实验分为Control组、LPS组、AG组(5 µmol·L-1)、AVLE组(31.25、62.5、100、125、200 µg·mL-1)。细胞孵育24 h后,AVLE预处理1 h,除Control组外各组均加入1 µg·mL-1 LPS培养16 h;Griess法检测NO含量,ELISA试剂盒检测TNF-α、IL-6、IL-1β含量。
细胞内ROS检测采用DCFH-DA方法。
Western blot法检测iNOS、COX-2、MAPK信号通路及Nrf2信号通路相关蛋白表达。
免疫荧光法观察Nrf2核转位,ImageJ 1.48软件半定量分析。
SPSS 17.0软件进行统计分析。数据以(x±s)表示,两组间比较采用t检验,多组间比较采用单因素方差分析(ANOVA),检验水准α=0.05。
与Control组比较,AVLE≤200 µg·mL-1时对细胞活力无明显影响(P > 0.05),后续实验选取200 µg·mL-1及以下无毒浓度。
与Control组比较,LPS组细胞中NO、TNF-α、IL-6、IL-1β和ROS的含量明显升高(P < 0.01);与LPS组比较,AVLE可明显降低细胞中NO、TNF-α、IL-6、IL-1β和ROS的含量(P < 0.01或P < 0.05)。
与Control组比较,LPS组iNOS、COX-2蛋白表达明显上调(P < 0.01);与LPS组比较,AVLE各剂量组iNOS、COX-2蛋白表达均明显下调(P < 0.01)。
与Control组相比,LPS组的p-ERK/ERK、p-p38/p38比值明显上调(P < 0.01);与LPS组比较,AVLE各剂量组p-ERK/ERK、p-p38/p38的比值均明显下调(P < 0.01或P < 0.05)。
与Control组比较,LPS组Nrf2、HO-1蛋白表达明显上调(P < 0.01);与LPS组比较,AVLE各剂量组Nrf2、HO-1蛋白表达均明显上调(P < 0.01)。
与LPS组相比,AVLE各剂量组对Nrf2核转位具有明显的促进作用(P < 0.01),提示AVLE可诱导细胞中Nrf2发生核转位,进而增强细胞的抗氧化抗炎能力。
MAPK由ERK、p38和JNK三种蛋白激酶分子构成,在炎症研究领域扮演着极其重要的角色[6]。本研究结果表明,AVLE可通过降低p-p38和p-ERK蛋白的表达来抑制炎症反应,然而对p-JNK蛋白表达的影响不显著。这种通路选择性可能与AVLE所含多成分协同作用密切相关,且不同活性成分对MAPK通路各亚型的调控效应本身存在明显差异[7-8]。Nrf2是氧化应激核心转录因子,能启动抗氧化基因表达,清除ROS并阻断炎症级联反应。本研究中,AVLE能够降低细胞内ROS含量,激活Nrf2通路并促进Nrf2核转位,实现抗氧化与抗炎协同作用。综上,AVLE在体外细胞水平具有明显抗炎活性,其机制与抑制炎症介质释放、调控MAPK及Nrf2信号通路密切相关。后续应进一步开展体内炎症模型实验研究,深入验证其抗炎作用,为其临床应用提供更充分的实验依据。
