Beta,ESR2)参与了绵羊的卵泡发育,但其调控机理尚不清楚。本研究以和田羊为试验对象,以NCBI数据库中绵羊ESR2基因启动子区2 871 bp序列为模板进行克隆测序,通过在线软件预测ESR2启动子区转录因子结合位点、CpG岛和调控元件,并以ESR2基因启动子区2 871 bp序列为模板构建6个连续缺失的荧光素酶载体,利用双荧光素酶报告基因系统筛选ESR2核心启动子区。本研究成功克隆了和田羊ESR2基因启动子区2 871 bp的序列,通过软件预测发现,和田羊ESR2基因启动子区存在4个潜在的转录起始位点、67个转录因子结合位点、1个TATA-box以及2个CpG岛。双荧光素酶活性检测结果表明ESR2基因启动子区位于L7(-527~-156)。本文研究结果为进一步探究绵羊ESR2基因启动子对绵羊卵泡发育的调控提供了新线索。, authors=谷若怀, 黄巧艳, 李伟, 孙慧萍, 朱乐潇, 邢凤,,     雌激素受体β基因(Estrogen Receptor Beta,ESR2)参与了绵羊的卵泡发育,但其调控机理尚不清楚。本研究以和田羊为试验对象,以NCBI数据库中绵羊ESR2基因启动子区2 871 bp序列为模板进行克隆测序,通过在线软件预测ESR2启动子区转录因子结合位点、CpG岛和调控元件,并以ESR2基因启动子区2 871 bp序列为模板构建6个连续缺失的荧光素酶载体,利用双荧光素酶报告基因系统筛选ESR2核心启动子区。本研究成功克隆了和田羊ESR2基因启动子区2 871 bp的序列,通过软件预测发现,和田羊ESR2基因启动子区存在4个潜在的转录起始位点、67个转录因子结合位点、1个TATA-box以及2个CpG岛。双荧光素酶活性检测结果表明ESR2基因启动子区位于L7(-527~-156)。本文研究结果为进一步探究绵羊ESR2基因启动子对绵羊卵泡发育的调控提供了新线索。
