)诱导小鼠睾丸间质细胞TM3氧化损伤和凋亡的作用及其分子机制。采用150μmol/L H 2 O 2 处理TM3细胞8 h诱导氧化损伤与凋亡,以5μmol/L RES干预24 h,采用比色法检测细胞培养液中乳酸脱氢酶(LDH)、细胞中谷胱甘肽过氧化物酶(GSH-Px)及丙二醛(MDA);通过DCF-DA探针法检测活性氧(ROS)的产生;通过流式细胞术分析细胞凋亡,并利用ELISA检测孕酮含量;进一步通过real-time PCR检测基质金属蛋白酶(MMP)-2/9 mRNA的表达,并利用明胶酶谱和ELISA检测细胞上清中MMP-2/9的水平;最后通过Western blot检测SIRT1/AMPK通路部分自噬分子的表达。结果显示:在H 2 O 2 诱导的TM3细胞中,RES下调其LDH、MDA及ROS水平,显著提高GSH-Px活性,减少细胞凋亡,并促进睾酮的分泌;同时RES显著下调MMP-2/9 mRNA的表达及其酶原与活性形式;此外,RES通过激活AMPK的磷酸化及沉默信息调节因子1(SIRT1)表达,促进自噬标志物LC3-I向LC3-II的转化,且SIRT1和AMPK抑制剂均可逆转此效应。综合以上,RES通过SIRT1/AMPK通路调控自噬并抑制MMP-2/9活性,缓解H 2 O 2 诱导的TM3细胞氧化损伤与凋亡。, authors=蒙丽舟, 张晓春, 陈指龙, 谷婷, 袁安文, 试验旨在研究白藜芦醇(RES)对过氧化氢(H 2 O 2 )诱导小鼠睾丸间质细胞TM3氧化损伤和凋亡的作用及其分子机制。采用150μmol/L H 2 O 2 处理TM3细胞8 h诱导氧化损伤与凋亡,以5μmol/L RES干预24 h,采用比色法检测细胞培养液中乳酸脱氢酶(LDH)、细胞中谷胱甘肽过氧化物酶(GSH-Px)及丙二醛(MDA);通过DCF-DA探针法检测活性氧(ROS)的产生;通过流式细胞术分析细胞凋亡,并利用ELISA检测孕酮含量;进一步通过real-time PCR检测基质金属蛋白酶(MMP)-2/9 mRNA的表达,并利用明胶酶谱和ELISA检测细胞上清中MMP-2/9的水平;最后通过Western blot检测SIRT1/AMPK通路部分自噬分子的表达。结果显示:在H 2 O 2 诱导的TM3细胞中,RES下调其LDH、MDA及ROS水平,显著提高GSH-Px活性,减少细胞凋亡,并促进睾酮的分泌;同时RES显著下调MMP-2/9 mRNA的表达及其酶原与活性形式;此外,RES通过激活AMPK的磷酸化及沉默信息调节因子1(SIRT1)表达,促进自噬标志物LC3-I向LC3-II的转化,且SIRT1和AMPK抑制剂均可逆转此效应。综合以上,RES通过SIRT1/AMPK通路调控自噬并抑制MMP-2/9活性,缓解H 2 O 2 诱导的TM3细胞氧化损伤与凋亡。
