Supernatant,CFS)对沙门氏菌的抑菌机制,本研究采用琼脂扩散法评估粪肠球菌CFS的抑菌效果,96孔板法绘制沙门氏菌的生长抑制曲线,通过扫描电镜观察菌体超微结构的变化,转录组测序分析沙门氏菌差异基因的表达。结果表明:CFS对沙门氏菌抑菌圈约为16.30 mm,30%(体积分数)终浓度CFS抑制共培养体系中沙门氏菌的生长(P<0.001)。电镜结果显示处理组沙门氏菌菌体出现胞膜塌陷、表面褶皱等形态学损伤。转录组学测序结果显示,与对照组相比,处理组有1 520个差异表达基因,其中747个基因上调,773个基因下调,差异基因显著富集于碳水化合物代谢、氨基酸生物合成及双组分信号系统等通路。研究结果提示CFS可通过破坏沙门氏菌细胞膜完整性、干扰碳水化合物代谢(三羧酸循环、丙酮酸代谢)以及下调磷酸转移酶系统和有氧呼吸过程中关键基因的表达,来抑制沙门氏菌细胞的能量供应从而发挥抑菌作用,可为开发新型替抗制剂提供理论基础。, authors=李培键, 龙金桃,     为探究益生性粪肠球菌无细胞上清液(Cell-Free Supernatant,CFS)对沙门氏菌的抑菌机制,本研究采用琼脂扩散法评估粪肠球菌CFS的抑菌效果,96孔板法绘制沙门氏菌的生长抑制曲线,通过扫描电镜观察菌体超微结构的变化,转录组测序分析沙门氏菌差异基因的表达。结果表明:CFS对沙门氏菌抑菌圈约为16.30 mm,30%(体积分数)终浓度CFS抑制共培养体系中沙门氏菌的生长(P<0.001)。电镜结果显示处理组沙门氏菌菌体出现胞膜塌陷、表面褶皱等形态学损伤。转录组学测序结果显示,与对照组相比,处理组有1 520个差异表达基因,其中747个基因上调,773个基因下调,差异基因显著富集于碳水化合物代谢、氨基酸生物合成及双组分信号系统等通路。研究结果提示CFS可通过破坏沙门氏菌细胞膜完整性、干扰碳水化合物代谢(三羧酸循环、丙酮酸代谢)以及下调磷酸转移酶系统和有氧呼吸过程中关键基因的表达,来抑制沙门氏菌细胞的能量供应从而发挥抑菌作用,可为开发新型替抗制剂提供理论基础。
