    半番鸭是番鸭和家鸭的杂交后代,羽色是其重要包装性状,白羽性状可以保持胴体的美观度,并满足市场羽绒的加工需求。本研究采集8周龄半番鸭5个黑羽个体以及5个白羽个体背腹部毛囊组织,提取总RNA后进行转录组测序,比较分析不同羽色组别间的转录表达谱,对筛选到的差异表达基因进行GO和KEGG富集分析,qRT-PCR验证挑选的DEGs表达水平。以黑羽个体为对照组,黑羽组与白羽组间共鉴定到383个差异表达基因,其中219个上调基因和164下调基因,qRT-PCR结果显示,转录组测序结果准确可靠。GO富集分析结果表明,上述差异基因主要参与内皮素受体信号通路,其中TYR、KIT、MC1R、EDNRB2、ASIP、TYRP1和WNT8A基因显著影响黑色素的形成。KEGG富集分析结果表明,有7个差异表达基因与黑色素生成有关。结合GSEA分析和蛋白网络互作,其中TYR、KIT、MC1R、EDNRB2、ASIP、TYRP1和WNT8A基因与黑色素发生通路相关的差异基因被特别关注,表明这些基因与半番鸭的羽色发育有密切相关。本研究为了解半番鸭羽色发育的差异提供了依据,为其他家禽的羽色研究提供了参考。
