27.0软件对SNP位点与柯乐猪的繁殖性状进行关联性分析;使用在线软件RNAfold、SHEsis、STRING等对SNP位点进行生物信息学分析。结果发现PML基因第2外显子的g.59407427 A>C、第7内含子的g.59444512 G>A以及第8内含子的g.59444894 A>G。连锁不平衡分析显示g.59407427 A>C和g.59444894A>G之间存在强连锁关系(r 2 >0.88)。单倍型分析显示共存在4种单倍型和8种双倍型,群体优势双倍型为H1H1(AAAAGG)。进一步与繁殖性状关联分析显示,3个SNP对总产仔数、产活仔数、初生窝重、断奶仔猪数和断奶窝重均有显著影响,双倍型H2H3(AAAGAG)个体的产活仔数、初生窝重、断奶窝重和断奶仔猪数均高于其他双倍型,为群体有利双倍型;g.59407427 A>C没有导致氨基酸序列的改变,但mRNA二级结构发生了变化,影响了结构稳定性。在PML蛋白互作蛋白网络中主要互作蛋白DAXX、RARA、RUNX1、SUMO1、SPI1与免疫功能、发育调控和抗病性密切相关,为后续研究基因功能和调控机制提供了方向。本研究发现了3个对柯乐猪繁殖性状有显著影响的SNP,可作为柯乐猪繁殖性状选育的遗传标记进一步研究。, authors=付红梅1 , 张游宇2 , 试验选取240头健康的经产柯乐猪,利用PCR扩增和Sanger测序法筛选PML基因的SNP;使用SPSS 27.0软件对SNP位点与柯乐猪的繁殖性状进行关联性分析;使用在线软件RNAfold、SHEsis、STRING等对SNP位点进行生物信息学分析。结果发现PML基因第2外显子的g.59407427 A>C、第7内含子的g.59444512 G>A以及第8内含子的g.59444894 A>G。连锁不平衡分析显示g.59407427 A>C和g.59444894A>G之间存在强连锁关系(r 2 >0.88)。单倍型分析显示共存在4种单倍型和8种双倍型,群体优势双倍型为H1H1(AAAAGG)。进一步与繁殖性状关联分析显示,3个SNP对总产仔数、产活仔数、初生窝重、断奶仔猪数和断奶窝重均有显著影响,双倍型H2H3(AAAGAG)个体的产活仔数、初生窝重、断奶窝重和断奶仔猪数均高于其他双倍型,为群体有利双倍型;g.59407427 A>C没有导致氨基酸序列的改变,但mRNA二级结构发生了变化,影响了结构稳定性。在PML蛋白互作蛋白网络中主要互作蛋白DAXX、RARA、RUNX1、SUMO1、SPI1与免疫功能、发育调控和抗病性密切相关,为后续研究基因功能和调控机制提供了方向。本研究发现了3个对柯乐猪繁殖性状有显著影响的SNP,可作为柯乐猪繁殖性状选育的遗传标记进一步研究。
