    西藏自治区是我国第二大牦牛主产区,独特的自然生态环境积累了丰富的牦牛遗传资源,全面认识西藏牦牛的起源与分化,对保护地方牦牛种质资源具有重要的现实意义。在mtDNA基因中,ATP合酶亚基6(ATP6)表现出对自然选择的高度敏感性,并且与在高海拔环境中探讨机体适应性分子机制有关。本研究成功扩增了来自16个西藏牦牛群体共367头个体的ATP6基因序列并完成测序,结果显示,所获得的ATP6基因序列富含A+T核苷酸,核苷酸多样性和单倍型多样性分别为0.001 81和0.534,表明西藏牦牛具有丰富的遗传多样性。在所分析的氨基酸序列中,疏水氨基酸占50.11%,亲水氨基酸占49.89%,且亮氨酸(占12.92%)与丝氨酸(占10.59%)占比较高。共获得70个多态性位点,包括52个单一突变位点和18个简约信息位点,共检测到67个单倍型,其中hap_2分布类群及数量最多,属高频单倍型。系统发育树显示,西藏16个牦牛类群聚为两大类,类乌齐牦牛与其他13个牦牛类群聚为一类,桑桑牦牛与桑日牦牛聚为一类。此外,对67个单倍型的网络关系分析发现存在2个不同的集群,这表明西藏牦牛可能存在2个不同的母系起源。本研究结果为牦牛母系起源提供了ATP6基因层面的分子证据,且与此前多基因研究结果相互印证,丰富了牦牛起源演化的证据体系,也为开展牦牛分子标记辅助育种及推动产业可持续发展提供了重要参考。
