articleAbstract=本试验旨在研究妊娠后期日粮中添加亮氨酸(Leu)对新生仔猪能量代谢的调控机制。选取200头妊娠70的健康长×大母猪(胎次为4~5胎),随机分为对照组、0.40%Leu组、0.80%Leu组和1.20%Leu组。试验期从母猪妊娠第70天开始,至母猪分娩结束。结果显示:妊娠84 d,0.40%Leu组和0.80%Leu组母猪血浆高密度脂蛋白(HDL)降低(P<0.01);妊娠98 d,0.40%Leu组甘油三酯(TG)下降(P<0.01);妊娠110 d,各Leu组TG均降低(P<0.01),0.40%Leu组母猪血浆葡萄糖(GLU)升高(P<0.05);3个处理组新生仔猪脐带血总胆固醇(TC)、HDL升高(P<0.05),背最长肌TG降低(P<0.05);0.80%Leu组新生仔猪背最长肌中SIRT-1和PGC-1α mRNA表达升高(P<0.05),各Leu组p-AMPK蛋白表达下降(P<0.05);0.40%Leu组和0.80%Leu组新生仔猪ACACA、GPAM的mRNA和FASN蛋白表达上调(P<0.05);0.40%Leu组新生仔猪ACSL1、CPT-1和FABP3 mRNA表达上调(P<0.05),而0.80%Leu组CD36和FATP4表达下降(P<0.05);0.40%Leu组SREBP1、PPARα和PPARγ表达下调(P<0.05);0.40%Leu组PEPCK的mRNA和0.80%Leu组GLUT1蛋白和ACL 本试验旨在研究妊娠后期日粮中添加亮氨酸(Leu)对新生仔猪能量代谢的调控机制。选取200头妊娠70的健康长×大母猪(胎次为4~5胎),随机分为对照组、0.40%Leu组、0.80%Leu组和1.20%Leu组。试验期从母猪妊娠第70天开始,至母猪分娩结束。结果显示:妊娠84 d,0.40%Leu组和0.80%Leu组母猪血浆高密度脂蛋白(HDL)降低(P<0.01);妊娠98 d,0.40%Leu组甘油三酯(TG)下降(P<0.01);妊娠110 d,各Leu组TG均降低(P<0.01),0.40%Leu组母猪血浆葡萄糖(GLU)升高(P<0.05);3个处理组新生仔猪脐带血总胆固醇(TC)、HDL升高(P<0.05),背最长肌TG降低(P<0.05);0.80%Leu组新生仔猪背最长肌中SIRT-1和PGC-1α mRNA表达升高(P<0.05),各Leu组p-AMPK蛋白表达下降(P<0.05);0.40%Leu组和0.80%Leu组新生仔猪ACACA、GPAM的mRNA和FASN蛋白表达上调(P<0.05);0.40%Leu组新生仔猪ACSL1、CPT-1和FABP3 mRNA表达上调(P<0.05),而0.80%Leu组CD36和FATP4表达下降(P<0.05);0.40%Leu组SREBP1、PPARα和PPARγ表达下调(P<0.05);0.40%Leu组PEPCK的mRNA和0.80%Leu组GLUT1蛋白和ACL mRNA表达增加(P<0.05)。本试验条件下,妊娠后期日粮中添加0.4%、0.8%Leu可提高新生仔猪背最长肌中脂肪酸和GLU的氧化代谢,降低TG合成提高能量代谢水平,为其促进蛋白质沉积提供能量。
