articleAbstract=本研究旨在克隆鉴定乌蒙乌骨鸡酪氨酸酶(TYR)基因完整编码区并进行生物信息学分析,为明确其影响乌骨鸡乌质特性的分子机制提供参考。以乌蒙乌骨鸡血液基因组DNA为模板,分别在鸡TYR基因5个外显子两端设计5对引物,通过PCR、克隆及测序拼接方法获得TYR基因完整编码区序列(Coding DNA Sequence,CDS),基于生物信息学方法综合分析该基因的结构特征,利用DNAStar 2.0和MEGA 7.0软件分别分析12个鸡形目和6个雁形目TYR基因CDS的相似性与系统发育关系。结果表明,乌蒙乌骨鸡TYR基因全长3 686 bp,包括276 bp 5'UTR、261 bp 3'UTR,部分内含子1~4长度分别为339、45、355、415 bp,外显子1~5长度分别为819、217、148、182、224 bp,CDS长度为1 590 bp,编码529个氨基酸。TYR蛋白分子式为C 2700 H 4071 N 739 O 784 S 29 ,分子质量为60.36 kD,总原子数为8 323,理论等电点(PI)为6.03,属于不稳定亲水性蛋白。乌蒙乌骨鸡TYR蛋白存在1个信号肽模块、1个酪氨酸酶基因家族核心结构域、1个表皮生长因子样结构域及1个跨膜螺旋结构域。序列相似性及分子系统发育关系分析均显示乌蒙乌骨鸡与沐川乌骨鸡、白来航鸡和原鸡等原鸡属物种的亲缘关系最近,与雁形目物种亲缘关系较远。本研究成功克隆获得乌蒙乌骨鸡TYR基因CDS全长序列,编码蛋白包括多个功能结构模块,研究结果可为乌骨鸡乌质性状分子机理解析及鸡形目物种进化探究提供理论依据。, 本研究旨在克隆鉴定乌蒙乌骨鸡酪氨酸酶(TYR)基因完整编码区并进行生物信息学分析,为明确其影响乌骨鸡乌质特性的分子机制提供参考。以乌蒙乌骨鸡血液基因组DNA为模板,分别在鸡TYR基因5个外显子两端设计5对引物,通过PCR、克隆及测序拼接方法获得TYR基因完整编码区序列(Coding DNA Sequence,CDS),基于生物信息学方法综合分析该基因的结构特征,利用DNAStar 2.0和MEGA 7.0软件分别分析12个鸡形目和6个雁形目TYR基因CDS的相似性与系统发育关系。结果表明,乌蒙乌骨鸡TYR基因全长3 686 bp,包括276 bp 5'UTR、261 bp 3'UTR,部分内含子1~4长度分别为339、45、355、415 bp,外显子1~5长度分别为819、217、148、182、224 bp,CDS长度为1 590 bp,编码529个氨基酸。TYR蛋白分子式为C 2700 H 4071 N 739 O 784 S 29 ,分子质量为60.36 kD,总原子数为8 323,理论等电点(PI)为6.03,属于不稳定亲水性蛋白。乌蒙乌骨鸡TYR蛋白存在1个信号肽模块、1个酪氨酸酶基因家族核心结构域、1个表皮生长因子样结构域及1个跨膜螺旋结构域。序列相似性及分子系统发育关系分析均显示乌蒙乌骨鸡与沐川乌骨鸡、白来航鸡和原鸡等原鸡属物种的亲缘关系最近,与雁形目物种亲缘关系较远。本研究成功克隆获得乌蒙乌骨鸡TYR基因CDS全长序列,编码蛋白包括多个功能结构模块,研究结果可为乌骨鸡乌质性状分子机理解析及鸡形目物种进化探究提供理论依据。
