实现碳达峰碳中和目标,是我国应对全球气候变化、推动经济社会绿色转型的重大部署。在此背景下,如何高效固定并转化二氧化碳等温室气体,从源头重塑工业生产的碳流路径,已成为学术界与产业界共同攻关的核心命题[1-2]。合成生物学作为一门旨在通过“设计-构建-测试-学习”循环重构生命系统的颠覆性技术,正为这一命题提供全新的解题思路:将微生物细胞从传统的“碳排放大户”改造为“碳固定工厂”乃至“高值创造平台”,实现从CO2到燃料、材料、食品与化学品的直接转化。这不仅是生物制造范式的深刻变革,更是衔接“碳中和”目标与“可持续生物经济”的关键技术桥梁[3-4]
CO2的生物固定与转化,在路径上可分为“直接路线”与“间接路线”两大层次[5]。直接路线即以CO2为唯一碳源的蓝细菌、产乙酸菌、氢氧化细菌等光合自养或化能自养微生物体系,通过天然或人工固碳途径将CO2直接同化为生物质或目标产物[6-7]。间接路线则通过热化学催化或电化学先将CO2还原为甲醇、甲酸、甲烷、乙酸等C1、C2等中间体,再以这些“低碳原料”作为微生物发酵的碳源与能源进行深加工[8-9]。2条路线各有优势:直接路线碳原子经济性最高、步骤最短,但受限于CO2分子本身的热力学稳定性与固碳酶的动力学瓶颈;间接路线引入了“化学-生物”耦合的灵活性,可借助可再生能源电力将CO2高效转化为易于储存和运输的C1、C2等化合物,再发挥微生物合成复杂分子的独特优势,实现“电/化学能-生物合成”的级联放大。甲醇、甲烷、乙酸等低碳原料,尤其是CO2来源的“绿色甲醇” “电制甲烷”和“电制乙酸”,正是联结这2条路线的重要物质纽带,承载着“可再生能源电力→化学储能→生物转化→高值产品”的完整价值链[10]
为了集中展示该领域的最新进展,《微生物学报》特推出“合成微生物与碳中和:从碳固定到高值创造”专刊。本专刊邀请国内该领域活跃的研究团队撰稿,共收录15篇综述与4篇研究论文,围绕“基础与前沿研究” “关键技术与路径”及“高值创造与应用” 3个维度,系统梳理了从固碳元件挖掘、途径设计重构到产业化路径探索的科学脉络与技术前沿。以下按三大板块对本专刊的主要内容进行梳理与评述。
对微生物固碳本质的深刻理解,是开展一切工程化改造的基石。在本方向收录的多篇综述中,研究者不仅审视了天然固碳体系的固有局限,更从催化机制、能量经济学及系统代谢等层面提出了突破方向。
固碳酶的催化机制与工程化改造是提升碳固定效率的首要环节。自然界虽然进化出多条固碳途径,但核心固碳酶,如卡尔文循环中的RubisCO,普遍存在催化速率慢或底物特异性低等“进化缺陷”,严重制约了CO2的生物同化效率。重庆大学王丹和中国科学院天津工业生物技术研究所王钦宏团队在综述中指出,打破这一瓶颈的关键在于从结构生物学视角深入认识酶的静态构象与动态构象变化对催化效率的决定性作用。近年来,理性设计、定向进化与高通量筛选等蛋白质工程策略的迭代发展,特别是AI辅助预测工具的引入,使得针对固碳酶的催化口袋重塑与底物特异性改造成为可能。该文还介绍了Greenase等[11]固碳酶数据库在新型酶元件挖掘中的支撑作用,并通过RubisCO、碳酸酐酶、甲醛裂合酶及多酶级联系统等典型案例,展示了从单酶优化到多酶体系集成的工程化路径。
如果说固碳酶是碳固定的“引擎”,那么能量供给则是决定引擎转速的“燃料”。能量-碳流的系统失配是固碳效率难以实现量级提升的深层瓶颈。北京化工大学刘子鹤团队创造性地提出了能量转化链条框架,将固碳过程统一为“能量输入-电子传递-碳固定输出”的连续网络,并建立了涵盖能量输入效率、电子利用效率及能量-碳转化效率的定量评价体系。这一视角将研究焦点从单一的碳流路径拉升至全局能量经济学高度,揭示了辅因子失配与电子传递损耗对碳通量的根本性限制。在工程层面,该综述系统总结了底盘宿主选择、外源供能模块构建及内源能量网络重构等策略,并提出了多尺度能量-碳流协同设计框架,为实现能量供给与固碳需求的精准匹配提供了可操作的理论基础[12]
天然固碳途径的多样性为合成生物学改造提供了丰富的“设计蓝图”。中国科学院微生物研究所朱华伟团队系统回顾了自然界已发现的6条天然固碳途径,包括卡尔文循环、还原性三羧酸循环、Wood-Ljungdahl途径、3-羟基丙酸循环、3-羟基丙酸/4-羟基丁酸循环以及二羧酸/4-羟基丁酸循环,并重点阐述了合成生物学在天然途径改造与人工途径设计方面的重要进展。该文指出,尽管天然途径在固碳速率、氧耐受性和能量效率等方面各有利弊,但尚需系统优化和设计以达到工业应用的全部要求。与此同时,人工途径的设计(如CETCH循环、巴豆酰辅酶A/乙醛酸循环等)为打破天然途径的进化束缚提供了全新可能[13]
在具体底盘微生物的研究层面,产乙酸菌与嗜甲烷菌作为两类天然固碳能力突出的微生物,其研究进展备受关注。中国科学院青岛生物能源与过程研究所李福利团队系统综述了产乙酸菌通过Wood-Ljungdahl途径高效同化CO/CO2的生化基础。Wood-Ljungdahl途径以其高碳原子经济性著称,该途径中2个CO2分子分别通过甲基支路与羰基支路进入乙酰辅酶A,整个过程不伴随碳损失,是自然界碳原子利用效率最高的固碳途径之一。该文重点介绍了近年来在产乙酸菌能量守恒机制以及代谢流调控方面的突破性进展。值得关注的是,以CRISPR-Cas为代表的先进遗传操作系统的开发,正有力推动产乙酸菌从生产乙酸、乙醇等天然代谢产物向合成丁醇、3-羟基丁酸等高附加值化学品的“定制化底盘”转变[14]。西安交通大学费强团队则聚焦嗜甲烷菌,展示了多组学技术如何加速该领域的范式变革,从单一基因功能鉴定迈向系统代谢过程的精准解析,尤其是在好氧嗜甲烷菌碳同化通量分配、厌氧甲烷氧化新型代谢路径的发现以及菌群间跨界互作与电子传递机制的阐明等方面取得了重要进展[15]
在对固碳元件与底盘代谢有了深入认知之后,合成生物学的核心驱动力在于“重构”与“创制”。本方向收录的论文涵盖了光合体系、氢能驱动、人工途径设计及合成群落等多个技术维度,展现了从单一途径改造到系统级整合的技术演进脉络。
光合生物制造被视为最接近自然碳中和逻辑的技术路径,它直接利用太阳能驱动CO2固定与产物合成,无需外加有机碳源或化学能源。中国科学院青岛生物能源与过程研究所栾国栋团队综述了以蓝细菌为底盘的光驱固碳细胞工厂构建策略。蓝细菌凭借高效的光合能力、清晰的遗传背景与成熟的操作体系,已成为光合生物制造领域最具潜力的模式底盘。该文从2个维度展开论述:在光合固碳效率提升方面,重点探讨了内源途径优化、外部材料赋能及底盘挖掘重塑等策略;在碳流定向与产物合成方面,围绕单菌株代谢工程和多菌株模块化协同,系统分析了碳资源精准导向的实现路径[16]
为了突破光合作用的光能转换效率限制,氢能驱动固碳提供了另一种技术思路。中国科学院天津工业生物技术研究所朱之光团队综述了以氢氧化细菌(hydrogen-oxidizing bacteria, HOB)为代表的氢能驱动固碳底盘细胞的代谢基础与研究进展。HOB是一类能以H2为电子供体、O2为电子受体、CO2为碳源的化能自养微生物,其固碳过程与“绿氢”耦合后可在不依赖光能的条件下实现CO2固定;该文详细介绍了代表性HOB菌株的代谢特征、遗传工具箱构建进展以及代谢通路重构与微生物-材料耦合等工程改造策略[17]
人工固碳途径的设计集中体现了合成生物学“造物致用”的核心理念。中国科学院天津工业生物技术研究所王钰团队系统总结了以CO2、甲醇和甲酸为一碳底物的人工同化途径研究进展。相较于天然途径,人工途径在设计理念上追求更短的反应步骤、更高的能量效率与更简洁的调控网络。代表性案例包括以甲醛为节点的缩合循环、以乙酰辅酶A为节点的体外多酶途径等。该文重点围绕一碳同化途径的设计原则、关键酶元件的创制改造以及人工途径在微生物底盘中的构建与应用三大核心环节展开论述,并前瞻性地讨论了人工智能辅助的新途径与新元件设计方向[18]
在单一菌株面临代谢负担过重、碳流分配失衡等瓶颈时,合成微生物群落提供了“分散化”的系统解决方案。中国科学院天津工业生物技术研究所黄志勇/韩一凡团队综述了合成微生物群落在CO2转化体系中的应用进展。其核心设计理念是通过理性组合不同功能微生物实现“代谢分工”,将固碳模块、产物合成模块和能量供给模块分配至不同菌株,以分担代谢负担、提升固碳效率与产物多样性。该文系统介绍了光能驱动、氢能驱动以及化能驱动等3类合成群落模式的设计原则与应用案例[19]
此外,北京化工大学陈必强团队提出的混合碳源协同代谢策略,从碳原子利用经济性的角度拓宽了固碳技术的研究视野。该文指出,传统发酵工艺中,糖类底物在中心碳代谢中约有1/3的碳以CO2形式损失。通过理性耦联糖类有机底物与甲醇、甲酸、合成气等C1碳源或乙酸等C2碳源,可构建底物碳架互补、能量供给协同及还原力平衡的混合营养代谢系统,实现碳得率的跨越式提升[20]
技术与路径的最终落脚点在于应用。本专刊收录的综述与研究论文展示了如何将工业尾气、CO2衍生的低碳原料转化为高价值产品,并开始触及经济可行性与产业化路径等深层问题。
需要特别指出的是,专刊中大量工作涉及以CO2来源的低碳原料为底物的生物转化。甲醇可通过CO2加氢大规模制备,已成为连接“电化学还原”与“生物合成”的核心枢纽;甲烷可通过CO2与绿氢的Sabatier反应合成(即“电制甲烷”),也可直接来源于厌氧消化产生的沼气;乙酸则可通过CO2电化学还原或产乙酸菌的气体发酵获得。上述低碳原料承载着“可再生能源→化学能载体→生物制造”的价值链跃升,是间接固碳路线不可或缺的物质基础。
甲醇生物制造在本专刊中获得了2篇综述的集中关注。南京工业大学陈可泉团队从宏观视角比较了天然与非天然甲醇同化路径的优势与局限,总结了以天然甲基营养菌为底盘以及在大肠杆菌、谷氨酸棒杆菌等工业模式菌中导入异源甲醇同化路径的设计策略。该文指出,甲醇具有储存运输方便、还原力高、可通过CO2加氢大规模制备等优势,是理想的非粮生物制造原料,并梳理了近年来利用甲醇合成有机酸、氨基酸、萜类、聚羟基脂肪酸酯等高值化学品的代表性案例[21]。中国农业科学院北京畜牧兽医研究所罗会颖团队则聚焦于扭脱甲基杆菌AM1这一模式底盘,系统探讨了其甲醇代谢途径的调控特征与适配的合成生物学工具,盘点了通过代谢工程改造AM1实现甲醇向重组蛋白、可降解材料、有机酸和萜类化合物等产物转化的研究成果[22]
甲烷生物转化利用方面,西安交通大学费强团队在基础研究板块中已系统论述了嗜甲烷菌的代谢机制与组学研究成果。嗜甲烷菌以甲烷为唯一碳源和能源,其关键酶——甲烷单加氧酶可在常温常压下将甲烷氧化为甲醇,这一反应在化学工业中需要高温高压条件才能实现。在应用层面,嗜甲烷菌已被探索用于生产聚羟基脂肪酸酯、单细胞蛋白、虾青素等高值产品。该综述指出,通过单细胞多组学与人工智能的深度融合,嗜甲烷菌细胞工厂有望在碳减排与低碳生物合成的双重目标下发挥更大作用[15]
乙酸转化利用方面,乙酸既是产乙酸菌气体发酵的主要天然产物,也可通过CO2电化学还原高效制得。乙酸作为C2底物,其碳架可直接进入乙醛酸循环或TCA循环,绕过糖酵解途径中的碳损失步骤。中国科学院天津工业生物技术研究所戴宗杰团队在单细胞蛋白生产的相关论述中指出,乙酸是一种高效的低碳底物,其同化效率优于甲醇和CO2,且对多数微生物的细胞毒性较低。重点聚焦单细胞蛋白(single cell protein, SCP),系统综述了以CO2、甲醇、乙酸等低碳底物生产SCP的研究进展。该文指出,低碳原料来源广泛、成本低廉、碳足迹小,但其同化效率与蛋白高效积累仍受底盘细胞适配性不足、代谢通量分配失衡以及能量供给效率低下的制约。该文从底盘挖掘与理性改造、代谢工程驱动以及工艺优化3个层面系统梳理了最新进展,指出多种基于低碳原料的SCP生产工艺正逐步向中试及规模化应用推进[23]
气体发酵是最直接利用CO2/CO工业尾气的技术路线,也是本专刊“高值创造与应用”板块的重要组成部分。北京首钢朗泽科技股份有限公司晁伟团队系统综述了利用工业尾气、生物质气化气等原料进行气体发酵生产燃料与化学品的产业化进展。该文分析了三代生物质发酵原料的应用特性,阐明了气体发酵在碳原子经济性与非粮化方面的技术优势;全面梳理了原料气预处理、气液传质强化、多级连续发酵、原位产物回收等核心工艺的技术要点。文章还梳理了国内外产业化案例,标志着气体发酵技术已跨越中试门槛、步入商业化验证阶段[24]
在更具体的产品案例层面,本专刊收录了多篇研究论文展示了从低碳原料到高值化学品的多样化转化路径。中国科学院天津工业生物技术研究所朱蕾蕾团队以甲醛(甲醇氧化产物)为底物,通过多酶级联催化合成药物中间体L-苏糖醇,经系统优化后产率达166.76 mmol/L[25]。中国科学院天津工业生物技术研究所杨建刚团队设计了“体外多酶级联+全细胞催化”的酶-菌协同路线,先将甲醇转化为甘油,再在谷氨酸棒状杆菌中完成甘油到1,3-丙二醇的转化,总碳转化率达90.2%,展示了“化学-生物”级联策略在甲醇高值化中的有效性[26]。中国科学院青岛生物能源与过程研究所吕明/苏航团队以永达尔梭菌为底盘,异源表达脂肪酶基因,打通了从CO/CO2到乙酸乙酯的完整生物合成通路,通过“气-糖”混合营养发酵使乙酸乙酯产量提升至36.25 mg/L。这一工作尤其值得关注,它实现了从气体原料直接到酯类产品的“一步发酵”,将固碳与高值合成在单一菌株中偶联,代表了自养底盘定向改造的前沿方向[27]。江南大学邓禹团队以乙二醇为底物,通过全细胞催化合成乙醇酸。经条件优化,摇瓶产量达19.3 g/L;进一步紫外诱变结合生物传感器筛选,获突变株GSUV-7,5 L罐产量达117.6 g/L、转化率100%,为乙醇酸生物合成提供了高效菌株与工艺方案[28]
最后,沈群团队通过对CO2合成淀粉、蛋白、微藻、乙醇4条技术路径的碳足迹与技术经济性评估,从系统层面回答了“固碳技术是否经济可行”这一关键问题。研究发现,微生物碳固定与利用技术的减碳潜力高度依赖能量供给方式与产品价值定位,在碳税政策驱动下,CO2基乙醇有望在特定条件下实现与传统路线的成本平价;而合成生物学与电催化技术的深度融合正在重塑碳利用的经济性边界。这一分析视角提醒我们,从实验室的“科学正确”到工业界的“经济正确”,仍需克服能量供给效率、气液传质强化、产物分离纯化等系统性工程挑战[29]
站在“碳中和”目标的历史节点上,合成微生物技术正处于从“概念验证”迈向“规模化应用”的关键转折期。从碳固定的角度来看,专刊清晰地呈现了2条路线——直接固碳与间接固碳的并行发展与相互交融。直接固碳路线以产乙酸菌的气体发酵、蓝细菌的光合固碳为代表,追求最短的CO2到产品路径;间接固碳路线以甲醇、甲烷、乙酸等CO2来源的低碳原料为枢纽,发挥化学催化与生物转化的各自优势,在灵活性与产品多样性方面更具潜力。2条路线殊途同归,共同指向从碳固定到高值创造的完整价值链条。
本专刊收录的19篇文章,从固碳酶的微观催化机制到气体发酵工厂的宏观产业化布局,全方位勾勒了这一创新链条的现状与未来。我们相信,随着人工智能辅助的酶设计与途径预测、基因编辑工具的迭代升级以及电/光-酶耦合等跨学科技术的深度融合,合成微生物将不仅仅是实验室中的科学创造,更将成为驱动我国乃至全球碳中和进程的“绿色细胞工厂”。期待本专刊能为该领域的研究者提供有益的参考与启发,共同推动合成生物学驱动微生物创新为可持续发展贡献更大的力量。

王钦宏,男,中国科学院天津工业生物技术研究所研究员/博士生导师。曾任中国科学院天津工业生物技术研究所副所长,现任中国食品科学技术学会新质蛋白专业委员会副主任委员兼干事长、《生物工程学报》《中国生物工程杂志》副主编及mLife、《合成生物学》《微生物学报》等学术刊物编委。致力于通过合成生物学手段重塑微生物碳代谢网络,设计构建了系列芳香类分子、微生物蛋白高效细胞工厂。完成10余项技术许可或转让,推进产业应用。积极推动二氧化碳人工合成淀粉等碳固定与生物转化关键技术研发,相关成果入选国家“十三五”科技创新成就展及2021年度中国科学十大进展。在Science、Trends Biotech等期刊发表论文200余篇,申请专利90余项,获授权专利60余项。
