[Objective] To investigate the effects of luxS deletion on the biofilm formation and stress tolerance of Listeria monocytogenes and to elucidate the role of luxS in regulating environmental adaptability of this pathogen. [Methods] With Listeria monocytogenes ATCC 19112 as the parental strain, a luxS-deleted mutant, a complemented strain, and an empty-vector control strain were constructed. Growth curve analysis, environmental stress tolerance assays and biofilm formation assays were performed to comparatively analyze phenotypic differences among the wild-type, mutant, and complemented strains. [Results] Compared with the wild-type strain, the luxS-deleted mutant exhibited reduced adaptability under adverse conditions, including low temperature, high osmotic pressure, oxidative stress, and acid stress. Its biofilm-forming ability decreased after 24 h and 48 h of incubation (P<0.01), accompanied by a reduction in the production of extracellular polymeric substances in the biofilm matrix. Further transcriptional analysis revealed that the expression levels of agrA, agrB, lmo2504, and sigB were downregulated (P<0.05), whereas that of ladR was upregulated (P<0.05) following luxS deletion. Under disinfectant stress, MTT assay showed that the metabolic activity of the luxS-deleted strain was lower than that of the wild-type and complemented strains after 48 h of biofilm cultivation (P<0.05). After 72 h, no significant differences were observed among the strains. [Conclusion] These findings demonstrate that luxS plays a critical role in regulating the environmental adaptability of L. monocytogenes and provide a novel theoretical basis for the development of future control and prevention strategies targeting this pathogen.

【目的】 研究单核细胞增生李斯特菌(Listeria monocytogenes, LM) luxS基因缺失对生物被膜形成及应激耐受性的影响,探讨luxS基因在单核细胞增生李斯特菌环境适应性调控中的作用。 【方法】 以ATCC 19112为亲本株,构建luxS基因缺失株、回补株及空载对照株。通过生长曲线测定、环境适应能力和生物被膜形成能力试验对比分析标准株、缺失株与回补株的表型差异。 【结果】 与标准株相比,luxS基因缺失株在低温、高渗透压、氧化及酸胁迫等不利条件下环境适应性显著减弱;生物被膜在培养24 h和48 h后形成能力显著降低(P<0.01),胞外聚合物生成量也显著减少。转录水平分析结果显示,luxS缺失后agrAagrBlmo2504sigB表达水平均显著下调(P<0.05),而ladR基因表达水平显著上调(P<0.05)。在消毒剂胁迫条件下,MTT法检测结果表明luxS基因缺失株在生物被膜培养48 h后细胞代谢活性显著低于标准株和回补株(P<0.05),而培养72 h后各菌株间差异不显著。 【结论】 本研究表明luxS基因在调控单核细胞增生李斯特菌环境适应性中发挥重要作用,可为该病原防控策略的设计提供新的理论依据。

【目的】 研究单核细胞增生李斯特菌(Listeria monocytogenes, LM) luxS基因缺失对生物被膜形成及应激耐受性的影响,探讨luxS基因在单核细胞增生李斯特菌环境适应性调控中的作用。 【方法】 以ATCC 19112为亲本株,构建luxS基因缺失株、回补株及空载对照株。通过生长曲线测定、环境适应能力和生物被膜形成能力试验对比分析标准株、缺失株与回补株的表型差异。 【结果】 与标准株相比,luxS基因缺失株在低温、高渗透压、氧化及酸胁迫等不利条件下环境适应性显著减弱;生物被膜在培养24 h和48 h后形成能力显著降低(P<0.01),胞外聚合物生成量也显著减少。转录水平分析结果显示,luxS缺失后agrAagrBlmo2504sigB表达水平均显著下调(P<0.05),而ladR基因表达水平显著上调(P<0.05)。在消毒剂胁迫条件下,MTT法检测结果表明luxS基因缺失株在生物被膜培养48 h后细胞代谢活性显著低于标准株和回补株(P<0.05),而培养72 h后各菌株间差异不显著。 【结论】 本研究表明luxS基因在调控单核细胞增生李斯特菌环境适应性中发挥重要作用,可为该病原防控策略的设计提供新的理论依据。

[Objective] To investigate the effects of luxS deletion on the biofilm formation and stress tolerance of Listeria monocytogenes and to elucidate the role of luxS in regulating environmental adaptability of this pathogen. [Methods] With Listeria monocytogenes ATCC 19112 as the parental strain, a luxS-deleted mutant, a complemented strain, and an empty-vector control strain were constructed. Growth curve analysis, environmental stress tolerance assays and biofilm formation assays were performed to comparatively analyze phenotypic differences among the wild-type, mutant, and complemented strains. [Results] Compared with the wild-type strain, the luxS-deleted mutant exhibited reduced adaptability under adverse conditions, including low temperature, high osmotic pressure, oxidative stress, and acid stress. Its biofilm-forming ability decreased after 24 h and 48 h of incubation (P<0.01), accompanied by a reduction in the production of extracellular polymeric substances in the biofilm matrix. Further transcriptional analysis revealed that the expression levels of agrA, agrB, lmo2504, and sigB were downregulated (P<0.05), whereas that of ladR was upregulated (P<0.05) following luxS deletion. Under disinfectant stress, MTT assay showed that the metabolic activity of the luxS-deleted strain was lower than that of the wild-type and complemented strains after 48 h of biofilm cultivation (P<0.05). After 72 h, no significant differences were observed among the strains. [Conclusion] These findings demonstrate that luxS plays a critical role in regulating the environmental adaptability of L. monocytogenes and provide a novel theoretical basis for the development of future control and prevention strategies targeting this pathogen.

单核细胞增生李斯特菌(Listeria monocytogenes, LM)简称单增李斯特菌,是一种重要的食源性致病菌,能够穿过血脑屏障在孕妇、新生儿等易感人群中引发李斯特菌病(listeriosis),常表现为脑膜炎或败血症,具有高致死率[1]。LM广泛存在于自然环境、食品原料及冷链运输环境中,对环境适应力较强,可在加工设备表面形成生物被膜,使其在食品加工场所中长期存留并造成食品交叉污染,是食品安全领域重点防控的病原微生物之一[2]
细菌环境适应性是指细菌通过调节生理状态和基因表达以应对外界不利条件、维持生存与生长的能力。LM的生存能力与环境适应性密切相关,在其生长繁殖过程中需应对低温、高渗透压、氧化应激及酸性胁迫等不利条件,这些因素可直接或间接影响细胞膜结构完整性、代谢通路及蛋白质功能,进而抑制菌体的生长与存活[3]。LM可通过调控基因表达及代谢通路对环境胁迫进行生理调节以维持细胞稳态,使该菌在食品产业链中持续存在且难以彻底清除,进一步增加了其污染几率与食源性感染的风险。
群体感应(quorum sensing, QS)系统是细菌通过分泌和感知信号分子以调节基因表达的重要机制。在AI-2型QS系统中,luxS基因可编码合成AI-2的关键酶LuxS,介导细菌细胞间信号传递,从而调控生物被膜形成、应激响应及环境适应等相关生理过程[4]。Ebenberger等[5]发现霍乱弧菌(Vibrio cholerae)可通过种内通讯调控QS系统关键因子HapR影响生物被膜的形成,从而增强其环境适应性和长期存活能力;Li等[6]通过缺失猪链球菌(Streptococcus suis) 2型中luxS基因,结合应激试验与转录组分析发现,调控铁代谢、酸及氧化应激的相关基因在细菌环境适应性及生理代谢中发挥关键作用;此外,有研究报道副溶血性弧菌(Vibrio parahaemolyticus) luxS基因缺失后可影响其多种生物学特性[7]。上述研究表明,luxS基因在多种细菌的生理调控过程中具有重要作用。近年来,QS系统在LM中的调控作用逐渐受到关注,多项研究表明QS系统可通过调控LM生物被膜形成能力和运动性等表型影响其毒力相关功能表达,而这些表型与细菌对环境胁迫的适应能力密切相关[8-9]。然而,现有研究主要集中于QS系统对单一环境因素或单一表型的影响,关于luxS基因在多种与食品加工相关环境胁迫条件下调控LM环境适应性的具体作用机制尚未见系统性研究。基于此,本研究以LM的luxS基因为研究对象,探讨其在低温、高渗透压和酸胁迫等不利环境中的适应能力及生物被膜形成等生物学特性中的作用,旨在为进一步解析luxS基因在LM环境适应过程中的功能提供理论依据。
单核细胞增生李斯特菌参考菌株ATCC 19112、原核表达载体pET-32a(+)、穿梭载体pKSV7及pSET2由甘肃农业大学兽医公共卫生学实验室保存;大肠埃希氏菌DH5α感受态细胞购自北京擎科生物科技股份有限公司。
同源重组连接酶、限制性核酸内切酶EcoR I,TaKaRa公司;细菌基因组DNA/RNA抽提试剂盒、质粒小量提取试剂盒、DNA胶回收试剂盒,Omega公司;Green Taq Mix、反转录试剂盒、qPCR MasterMix、DNA Marker,南京诺唯赞生物科技股份有限公司;脑心浸出液肉汤培养基、壮观霉素、氯霉素,北京索莱宝科技有限公司;BCA蛋白浓度测定试剂盒,上海碧云天生物技术股份有限公司;3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, MTT],TCI公司;总多糖含量测试盒,上海优选生物科技有限公司。
高速低温离心机,HITACHI公司;凝胶成像系统、电穿孔仪,Bio-Rad公司;梯度基因扩增仪,Eppendorf公司;生物安全超净工作台,苏州安泰空气技术有限公司;超声波细胞粉碎机,宁波新芝生物科技有限公司。
根据参考菌株ATCC 19112基因组序列(https://genomes.atcc.org/genomes/da1bf206779541b2),使用Primer Premier 5.0软件设计基因缺失及回补引物,并对部分引物的5′端添加酶切位点(下划线标注),引物序列信息见表1。通过在线工具Primer Quest-design qPCR assays (https://sg.idtdna.com/pages/tools/primerquest)设计实时荧光定量PCR引物,引物序列信息见表2,所有引物均由北京擎科生物技术有限公司合成。
参照细菌全基因组DNA抽提试剂盒说明书提取LM ATCC 19112基因组DNA。以提取的基因组DNA为模板,使用高保真DNA聚合酶分别以引物对(luxS-1/luxS-2)和(luxS-3/luxS-4)进行PCR扩增,获得luxS基因的上下游同源臂。PCR反应体系(15 µL):Green Taq Mix 7.5 µL,上、下游引物(10 µmol/L)各1 µL,DNA模板1 µL,ddH2O 4.5 µL。PCR反应条件:95 ℃预变性5 min;95 ℃变性30 s,55 ℃退火30 s,72 ℃延伸1 min,共30个循环;72 ℃终延伸10 min。产物经凝胶电泳检测条带大小正确后,使用试剂盒进行切胶回收纯化。随后,以纯化后的上游和下游同源臂片段为模板进行SOE-PCR扩增,反应体系及条件同上,扩增产物经纯化。将温敏型穿梭质粒pKSV7于37 ℃条件下EcoR Ⅰ酶切2 h,反应产物同样进行纯化。将上述2种纯化产物通过同源重组连接酶连接,连接产物经热激法转化至DH5α感受态细胞中,涂布于含氯霉素的抗性平板筛选阳性克隆。挑取单菌落,以pKSV7-F和pKSV7-R引物进行PCR鉴定,将扩增条带大小为1 171 bp的重组质粒送北京擎科生物科技股份有限公司测序验证。比对无误后,将构建成功的重组质粒命名为pKSV7-ΔluxS
将pKSV7-ΔluxS电转化至已制备好的LM感受态细胞中,于42 ℃含氯霉素的双重选择压力下连续传代进行同源重组。之后将菌株转接至30 ℃无抗生素培养基中连续传代约10代,使质粒丢失。经菌液PCR鉴定条带大小正确且确认稳定遗传后,命名为缺失株ΔluxS
以ATCC 19112基因组DNA为模板,使用CluxS-F和CluxS-R引物进行PCR扩增,反应体系及条件同1.4节。将PCR产物与经酶切的穿梭载体pSET2连接,并转化至DH5α感受态细胞中。经PCR鉴定并测序验证质粒构建正确后,将测序正确的重组质粒电转至制备好的缺失株ΔluxS感受态细胞中。挑取单菌落,以pSET2-F和pSET2-R进行菌液PCR验证。将条带大小正确的菌株命名为回补株CΔluxS
使用试剂盒提取ATCC 19112、ΔluxS和CΔluxS的总RNA,测定浓度后参照试剂盒说明书反转录为cDNA。以gyrB为内参基因,采用RT-qPCR检测各菌株中目的基因luxS的转录水平变化。RT-qPCR反应体系(20 µL):2×SYBR Green Pro Taq HS Premix 10 µL,上、下游引物(10 µmol/L)各0.4 µL,cDNA模板1 µL,ddH2O 8.2 µL。RT-qPCR反应条件:95 ℃ 100 s;95 ℃ 10 s,60 ℃ 30 s,共40个循环;95 ℃ 90 s;60 ℃ 60 s;以0.5 ℃/s速率逐步升温至95 ℃,持续采集荧光信号以绘制溶解曲线;37 ℃反应10 s,使反应体系降至室温。每个菌株的内参基因和目的基因均设3个生物学重复。
参照1.5节方法重新构建带有His标签的回补株及阳性对照luxS-pET-32a(+)重组蛋白表达载体,以缺失株作为阴性对照,进行Western blotting验证。将上述3株菌分别收集菌体并处理样品,经SDS-PAGE后转至PVDF膜,室温封闭1 h后依次以抗His标签一抗及HRP标记二抗孵育,采用ECL化学发光法显色,曝光成像后观察目的条带。
将空载穿梭质粒pSET2分别电转至ATCC 19112和ΔluxS感受态细胞中,30 ℃培养24-48 h,在含壮观霉素的BHI平板上筛选获得含pSET2空载质粒的菌株。电转成功的阳性克隆分别命名为标准株空载对照LM19112-pSET2和缺失株空载对照ΔluxS-pSET2,后续分别简称为LM19112-pSET2和ΔluxS-pSET2。
将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS在含壮观霉素的BHI平板上划线复苏,挑取单菌落接种至培养基中,37 ℃、200 r/min恒温振荡培养过夜。使用紫外分光光度计将各菌株菌悬液的OD600值调整为0.5,按1:100比例将菌液接种至30 mL含壮观霉素的BHI液体培养基中,继续于37 ℃、200 r/min条件下振荡培养;另取菌液于4 ℃条件下静置培养。37 ℃条件下接种3 h后开始测定OD600值,此后每隔1 h测定1次,直至细菌生长曲线进入平台期;4 ℃条件下自开始培养后每隔约12 h测定1次OD600值,直至曲线进入平台期[10]。每组设3个平行样本,独立重复3次。
挑取LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS单菌落接种至含壮观霉素的BHI液体培养基中,37 ℃、200 r/min培养6 h至OD600值为0.6-0.8,8 000 r/min离心2 min收集菌体沉淀,以无菌1×PBS缓冲液重悬菌体并洗涤3次,再稀释调节OD600值至0.5。取100 μL调节好的菌液分别加入900 μL含10 mmol/L H2O2的培养基BHI (pH 3.0)中,混匀后于37 ℃孵育30 min。倍比稀释后进行平板涂布,同时设置未应激处理对照组,统计菌落数。试验重复3次。应激存活率计算如公式(1)所示。

参考郭骞等[11]方法进行抗高渗应激试验,将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS于37 ℃、200 r/min条件下培养过夜,次日将各菌株菌悬液OD600值调整至0.5,按1:100比例接种至含5% NaCl的BHI液体培养基中,于37 ℃、200 r/min条件下振荡培养,每隔1 h测定1次OD600值,直至菌液生长达到平台期。每组设3个平行样本,独立重复3次。
采用半固体琼脂平板法测定菌株运动性。将37 ℃、200 r/min培养过夜的LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS菌液OD600值调节至0.5,取2 μL菌液点接于含0.3%琼脂的BHI半固体培养基中央,于30 ℃静置培养24 h。培养结束后,通过测定菌落向外扩散形成的泳动直径评价菌株运动能力,每组设3个平行样本,独立重复3次。
将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS分别于37 ℃、200 r/min培养过夜,次日按1:100转接至新鲜培养基中,37 ℃、200 r/min培养5 h至OD600值为0.5。以含壮观霉素的BHI培养基按1:100比例稀释备用。在96孔细胞培养板中每孔加入200 μL稀释菌液,置于37 ℃恒温培养箱中静置培养24、48、72 h。
生物被膜定量采用结晶紫染色法[12]。培养结束后,小心弃去孔内培养上清液,以无菌1×PBS缓冲液轻柔洗涤3次,彻底去除未黏附的浮游细菌。每孔加入200 μL甲醛固定15 min,通风处干燥10 min。加入0.5%结晶紫染色液染色10 min,吸弃染液,以蒸馏水反复冲洗数次并充分晾干。每孔加入200 μL 90%乙醇溶液溶解结晶紫,以微孔板分光光度计于OD595处测定吸光度值。每组设3个平行样本,并独立重复3次。
参考朱黄琛之等[13]方法,将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS划线活化单菌落,37 ℃、200 r/min培养5 h至OD600值为0.5,按1:100稀释备用。提前在12孔细胞培养板中放置无菌细胞爬片,每孔加入1 mL稀释菌液,并设阴性对照。37 ℃培养48 h和72 h后,吸弃上清液,以无菌1×PBS缓冲液冲洗3次,取出细胞爬片置于含3 mL 1×PBS缓冲液的离心管中,冰浴条件下40 W超声3 min,4 ℃、8 000 r/min离心30 min。将上清液经0.45 μm滤膜过滤,收集胞外聚合物样品备用。
采用总多糖含量测定试剂盒测定胞外多糖含量,预先绘制标准曲线。吸取200 μL样品、100 μL试剂一和500 μL浓硫酸混匀,90 ℃水浴20 min,冷却后取200 μL加入96孔板,于OD490处测定吸光度值,根据标准曲线计算样品浓度(μg/mL)。每组设3个平行样本,独立重复3次。
采用BCA法测定胞外蛋白含量,预先绘制标准曲线。吸取20 μL样品与200 μL BCA工作液混匀,加入96孔板,37 ℃静置30 min,于OD562处测定吸光度值,根据标准曲线计算样品浓度(μg/mL)。每组设3个平行样本,重复3次。
通过在线工具Primer Quest-design qPCR assays设计实时荧光定量PCR引物。提取LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS的总RNA,测定浓度后参照试剂盒说明书反转录为cDNA。以gyrB为内参基因,生物被膜相关基因(agrAagrBlmo2504)、应激反应基因(sigB)及PadR家族转录调控蛋白基因(ladR)为目的基因,采用RT-qPCR检测各基因转录水平变化。RT-qPCR反应体系及条件同1.6.1节,每个菌株的内参基因和目的基因均设3个生物学重复,独立重复3次。
将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS分别于37 ℃培养过夜,次日按1:100转接至新鲜培养基中培养,待OD600值调整至1.0后,以含壮观霉素的BHI培养基按1:1 000比例稀释备用。将0.2% HCHO溶液、0.5% I2溶液和2% NaOH溶液以ddH2O进行连续2次倍比稀释,各管浓度如表3所示。将不同浓度的消毒剂在96孔板中各加10 μL,每孔加入100 μL制备好的菌液,混匀后于37 ℃恒温培养箱中培养24 h后观察结果。
参考陈月桃[14]方法,采用MTT还原法测定消毒剂处理后生物被膜的代谢活性。将LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS在96孔板中分别培养48 h和72 h以形成生物被膜,弃去培养液并以PBS洗涤3次。随后分别加入含MIC和5×MIC浓度HCHO、I2和NaOH的BHI液体培养基,于37 ℃静置培养24 h。处理结束后洗涤去除残留消毒剂,每孔加入MTT溶液(0.5 mg/mL)避光孵育3 h,弃去上清后每孔加入DMSO溶解生成的甲瓒结晶,于OD570处测定吸光值。生物被膜细胞代谢活性计算如公式(2)所示。

所有试验数据均采用GraphPad Prism 8.0软件进行处理与作图。采用t检验分析数据。统计学显著性水平设定如下:P<0.05表示差异显著,标记为*;P<0.01表示差异高度显著,标记为**;P<0.001表示差异极显著,标记为***;ns表示无显著差异(P>0.05)。
以标准株ATCC 19112、缺失株ΔluxS和回补株CΔluxS菌液为模板,通过引物luxS-1和luxS-4对其进行PCR扩增,标准株ATCC 19112 PCR产物条带大小为1 439 bp,缺失株ΔluxS条带大小为971 bp,较标准株减少468 bp (图1A,泳道1和泳道2),回补株条带大小与缺失株一致(图1A,泳道2和泳道3);使用引物CluxS-F和CluxS-R对其扩增时,得到标准株与回补株条带大小均为468 bp (图1B,泳道1和泳道3),缺失株无条带(图1B,泳道2)。
同时,采用RT-qPCR检测luxS基因转录水平,结果显示缺失株表达量接近0,回补株表达水平明显高于标准株,约为标准株的12倍(图1C)。上述结果表明缺失株ΔluxS和回补株CΔluxS构建成功。Western blotting结果显示,目的蛋白理论分子量约为18 kDa,阴性对照组在18 kDa处未检测到明显目的蛋白条带,而阳性对照和带有His标签的回补株在18 kDa处均检测到明显的特异性条带(图1D),表明luxS基因已在回补株中成功表达,回补株构建成功。
将空载质粒pSET2分别电转入ATCC 19112和ΔluxS感受态细胞中,以pSET2-F和pSET2-R引物进行PCR鉴定,均获得约214 bp的条带,分别为LM19112-pSET2 (图2,泳道1和泳道2)和ΔluxS-pSET2 (图2,泳道3和泳道4)。LM19112-pSET2和ΔluxS-pSET2作为CΔluxS的空载对照株,用于排除pSET2质粒本身对CΔluxS表型的干扰。
在37 ℃条件下LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS均在7-13 h处于对数生长期,13 h后进入平台期,生长趋势无明显差异(P>0.05,图3A)。在4 ℃条件下,3株菌均在6-9 d处于对数生长期,9 d后进入平台期;此外,ΔluxS-pSET2的生长速率较LM19112-pSET2显著降低(P<0.01或P<0.001,图3B)。结果表明,在4 ℃低温培养条件下luxS基因缺失可显著影响细菌的生长能力。
经氧化应激和酸应激处理后,将菌液稀释至10-4梯度进行平板计数,根据菌落计数结果计算存活率。结果显示,ΔluxS-pSET2的存活率为(3.56±0.88)%,显著低于LM19112-pSET2的(9.97±1.54)%和回补株CΔluxS的(9.47±1.65)% (P<0.01,图4A),表明缺失luxS基因可使LM对氧化环境的耐受能力下降。同样地,在pH 3.0胁迫条件下,ΔluxS-pSET2的存活率为(14.79±1.07)%,极显著低于LM19112-pSET2的(44.77±1.87)%和回补株CΔluxS的(44.03±3.60)% (P<0.001,图4B),表明luxS基因缺失显著降低了菌株对酸胁迫的耐受能力。
图5所示,在5% NaCl条件下,应激早期ΔluxS-pSET2和LM19112-pSET2对高渗透压胁迫的耐受性差异不显著(P>0.05);而在对数生长期后期及平台期,ΔluxS-pSET2的耐受能力显著低于LM19112-pSET2 (P<0.05),推测luxS基因可能在细菌密度较高时对高渗透压环境产生影响。
通过测定LM19112-pSET2、ΔluxS-pSET2和回补株CΔluxS在BHI半固体培养基上的运动圈直径评价其运动能力。如图6所示,各菌株的运动圈直径均约为3 mm,菌株间差异不显著(P>0.05),说明luxS基因缺失对LM运动能力的影响不明显。
结晶紫染色法检测结果如图7所示,各菌株生物被膜量随培养时间延长而增加。培养24 h和48 h后,ΔluxS-pSET2的生物被膜量均显著低于LM19112-pSET2和回补株(P<0.01);而培养72 h后,3株菌之间差异不显著(P>0.05)。
采用苯酚-硫酸法检测生物被膜胞外多糖含量,通过葡萄糖标准曲线计算胞外多糖含量。如图8所示,缺失luxS基因后,培养48 h胞外多糖量较标准株显著减少(P<0.01,图8B);而培养72 h后,3株菌之间的胞外多糖含量无明显差异(P>0.05,图8B)。
采用BCA法检测缺失luxS基因后对生物被膜胞外蛋白的影响,通过蛋白质标准曲线计算胞外蛋白含量。如图9所示,培养48 h后缺失株的胞外蛋白量较标准株显著减少(P<0.01,图9B);培养72 h后3株菌之间的胞外蛋白含量无明显差异(P>0.05,图9B)。如图7-9所示,培养前48 h,luxS缺失株相较标准株与回补株胞外多糖及胞外蛋白含量均明显降低;而培养72 h后差异不显著。其变化趋势与生物被膜量一致,表明luxS基因可能通过影响胞外聚合物的合成而调控生物被膜的形成。
RT-qPCR结果如图10所示,与LM19112-pSET2相比,ΔluxS-pSET2中群体感应系统调控基因agrAagrB的表达量均显著下调(P<0.05),生物被膜相关基因lmo2504的表达水平也明显降低(P<0.05)。结合生物被膜测定结果,ΔluxS-pSET2生物被膜形成能力显著减弱,二者变化趋势一致。此外,应激调控基因sigB在ΔluxS-pSET2中的表达水平同样低于标准株和回补株(P<0.05),这与应激耐受性试验中ΔluxS-pSET2在氧化应激、酸胁迫及高渗胁迫条件下存活率均明显降低有关。ladR基因在ΔluxS-pSET2中表达水平显著上调(P<0.05),LadR为PadR家族转录调控蛋白,参与细菌碳代谢及环境适应性相关基因的调控,该结果与已有报道中luxS缺失后ladR表达水平上调的现象一致[15]。上述结果表明,luxS基因缺失可通过调控agrAagrBlmo2504sigBladR的表达影响生物被膜的形成及环境应激反应。
选用0.2% HCHO、0.5% I2和2% NaOH 3种消毒剂测定3株菌的MIC值。如表4所示,3株菌对同种消毒剂的MIC值相近,表明缺失luxS基因对消毒剂的MIC值无显著影响。
采用MTT法测定不同消毒剂处理条件下生物被膜细胞的代谢活性。如图11所示,生物被膜培养48 h后,经3种消毒剂MIC浓度处理后缺失株ΔluxS-pSET2的细胞代谢活性在MIC和5×MIC条件下均低于标准株LM19112-pSET2 (图11A11C11E);且在5×MIC条件下,3株菌生物被膜细胞代谢活性均进一步下降,ΔluxS-pSET2较标准株与回补株差异显著(P<0.05或P<0.001)。培养至72 h后,3株菌在不同消毒剂及浓度处理下的生物被膜细胞代谢活性整体降低(图11B11D11F),但菌株间差异均不显著(P>0.05)。综上所述,在生物被膜培养48 h后,luxS缺失株在不同消毒剂处理条件下的细胞代谢活性均低于标准株及回补株,而培养72 h后菌株间差异不显著。结合生物被膜形成量试验结果,推测luxS基因缺失导致细菌生物被膜形成能力减弱,进而影响生物被膜细胞对外界消毒剂胁迫的整体耐受能力。
LM具有较强的环境耐受能力,能够在食品加工环境中的低温、高渗透压及氧化应激等多种不利条件下长期存活,对食品安全和公共卫生构成潜在威胁[16]。目前普遍认为,该菌对环境的适应能力主要依赖于各调控通路的精密协调,使其能够灵敏感知并响应外界信号[17]。QS系统是细菌在复杂环境中协调群体行为的重要调控机制,在环境适应性中发挥关键作用,主要通过感应细菌生长发育过程中分泌的信号分子浓度来判断群体密度,进而调控生物被膜形成、胞外基质合成、代谢调节及胁迫应答等生理过程[18]。然而,LM中相关研究较为匮乏。本研究通过缺失LM ATCC 19112标准株中的luxS基因,从不同温度下的生长发育、生物被膜形成能力及环境应激耐受性等方面对luxS基因缺失株的生物学表型进行了系统分析。结果表明,缺失该基因后在适宜条件下不影响LM的生长发育,但在低温环境、生物被膜形成早期、胞外基质合成,以及氧化、高渗透压、酸胁迫耐受和抗消毒剂胁迫过程中LM的生长发育均受到不同程度的影响,证明luxS基因在其中发挥着重要的调控作用。
LuxS/AI-2介导的QS系统广泛存在于革兰阳性和阴性细菌中,信号分子AI-2可在不同物种间传递信号,影响LM群体行为的调控[19]。其中LuxS作为合成AI-2的关键酶,当缺失luxS基因时可对细菌运动能力、抗菌药物敏感性及应激适应能力等方面产生影响[20],表明luxS并非单一的调控因子,而是在调控信号分子生成的同时也参与相关代谢网络的调节[21]。细菌的环境适应性通常体现在不同外界条件下维持自身生长和存活的能力,已有研究报道,LM低温生长依赖多种系统的协同作用,包括钴胺素、鞭毛基因及rRNA分子等[22-24]。本研究中,37 ℃条件下3株菌的生长曲线基本重合,但在4 ℃低温条件下缺失株生长能力明显低于标准株,表明LM在适宜温度下生长时并不依赖luxS基因,而在低温环境下该基因的缺失可显著抑制菌株生长,luxS基因可能参与了低温环境适应的调控过程。有研究发现,LM中LuxS/AI-2介导的QS系统可通过多种调控通路影响细菌运动性及多种生理功能的调节,有助于该菌在多重胁迫环境中维持生理活性和结构稳定性[25]。本研究通过不同条件下的应激存活试验发现,luxS基因缺失显著降低了pH 3.0和10 mmol/L H2O2条件下的存活率,这与张晓荟等[26]lmo2300的研究结果相似,即缺失该基因后可导致细菌在氧化压力下的存活率降低。在5% NaCl条件下,应激初期luxS基因缺失株与标准对照株耐受能力相近,而进入应激后期后,随着细胞密度升高,缺失株耐受能力明显减弱,这一特征与群体感应依赖密度进行调控的特性一致。胡沛等[27]通过缺失qseCluxS双基因发现,该菌株对宿主及环境的适应能力整体显著下降,包括毒力、荚膜产量和抗逆能力等多方面,表明QS系统相关基因在维持细菌生存竞争力中不可或缺。运动能力试验中3株菌未表现出显著差异,表明缺失luxS基因对细菌运动能力的影响较为有限。细菌运动性通常与鞭毛结构及其相关调控基因的表达密切相关[28],而luxS主要参与AI-2介导的群体感应信号合成,其调控作用更多体现在生物被膜形成、细胞间信号传递及环境适应性等方面,因此luxS基因缺失并未明显改变细菌的运动表型。
已有研究报道,当LuxS/AI-2信号通路受到影响时生物被膜形成能力将明显下降,相关毒力表型也会受到抑制[29]。作为该通路核心的luxS基因不仅负责AI-2前体分子4,5-二羟基-2,3-戊二酮(4,5-dihydroxy-2,3-pentanedione, DPD)的合成,还通过参与甲硫氨酸循环将代谢稳态与群体感应过程相互连接[30]。因此,luxS的缺失不仅会导致AI-2浓度降低和细胞间通讯受阻,还可能通过改变代谢通路影响胞外多糖和胞外蛋白的合成,从而在一定程度上影响生物被膜的形成与结构稳定[31]。Zhang等[32]研究发现,缺失pgpH基因可影响生物被膜的形成,这可能与该基因改变生化代谢从而削弱基质合成有关。生物被膜的形成是一个动态过程,通常包括初始附着、微菌落形成、成熟及解离分散4个阶段,其中早中期阶段依赖于细菌表面结构及胞外聚合物的合成调控[33]。本研究结果显示,缺失株在培养24 h和48 h后生物被膜量显著降低,而培养72 h后3株菌差异不显著。进一步检测胞外聚合物生成量发现,培养48 h后缺失株相较于标准株和回补株胞外多糖及胞外蛋白含量均不同程度降低,表明luxS基因可能通过影响胞外聚合物的生成而调控生物被膜形成;而培养72 h后3株菌的胞外多糖和胞外蛋白含量未表现出显著差异,这可能与生物被膜在培养后期逐渐进入成熟阶段有关。胞外聚合物作为生物被膜结构的重要组成成分,其含量减少会直接影响细菌表面黏附及生物被膜结构的稳定性[34]。综上所述,luxS基因可能主要调控生物被膜形成的早期至中期阶段,当生物被膜进入成熟稳定阶段后,其结构维持更多依赖于其他调控系统,从而在一定程度上弥补luxS缺失带来的影响。此外,RT-qPCR结果显示,luxS缺失后QS系统调控因子agrAagrB、生物被膜相关基因lmo2504及应激调控相关基因sigB表达均下调,而ladR基因表达上调。ladR基因属于PadR家族转录调控蛋白,可影响生物被膜的形成,本研究中ladR表达水平上调可能是细菌在luxS缺失导致群体感应信号受扰后的一种适应性调节。Agr系统已被证实参与LM生物被膜形成的调控,其表达水平降低可能影响细菌间信号传递及胞外基质合成,从而抑制生物被膜早期结构的建立[35]。对于应激调控因子SigB,有研究表明其缺失时LM生物被膜形成能力下降,进而影响细菌在消毒剂环境下的生存能力[36]。这可能是本研究中缺失株生物被膜量减少及环境应激耐受能力减弱的原因之一。
鉴于生物被膜被认为是细菌抵御消毒剂的重要屏障[37],在明确luxS基因对生物被膜形成影响的基础上,本研究进一步评估了luxS基因在消毒剂耐受性中的调控作用。消毒剂胁迫试验中3株菌的最小抑菌浓度无显著差异,考虑到luxS基因主要通过QS系统参与生物被膜的构建,推测其对消毒剂的影响可能更多体现在生物被膜耐受性方面。基于此,采用MTT法评价经HCHO溶液、I2溶液和NaOH溶液3种消毒剂处理后各菌株生物被膜细胞的代谢活性。结果显示,生物被膜培养48 h消毒剂处理条件下3株菌均表现出细胞代谢活性下降,其中luxS缺失株较LM ATCC 19112标准株与回补株下降更为显著,提示luxS基因可能在维持生物被膜细胞代谢状态及环境适应能力方面发挥重要作用。LuxS是细菌群体感应系统的重要组成部分,通过参与AI-2信号分子的合成调控生物被膜形成及应激响应相关通路的基因表达。因此,luxS基因缺失可能导致相关调控网络改变,从而降低生物被膜细胞在消毒剂胁迫下的代谢活性与耐受能力。生物被膜培养72 h后,3株菌间代谢活性未表现出显著差异,推测可能与生物被膜进入成熟阶段有关。成熟的生物被膜由菌体细胞及其分泌的胞外聚合物基质共同构成,可形成致密的三维网络结构,不仅为被膜内菌体提供稳定的生存微环境,同时可在一定程度上阻碍消毒剂向生物被膜内部扩散[38]。结合本研究结果推测,luxS介导的调控网络可能通过影响生物被膜结构形成及应激相关基因的表达,进而调节细胞的代谢活性及其对外界胁迫的响应能力。上述结果表明,luxS基因在细菌应对外界环境压力、维持自身生存稳态的过程中发挥着重要作用。综上所述,本研究加深了对luxS基因在LM环境适应性调控机制中作用的认识,并为后续深入解析其分子调控网络奠定了理论基础。
