Objective To prepare reference material (RM) for positive serum specific for genotype Ⅱ African swine fever virus (ASFV) for serological detection, quality control, and proficiency testing (PT). Methods The anti-serum collected from specific pathogen-free (SPF) swine immunized with inactivated genotype Ⅱ ASFV was used as raw material for the preparation of RM. Indirect enzyme-linked immunosorbent assay (iELISA) was employed to evaluate the purity, specificity, homogeneity, and stability of RM. In addition, RM was characterized by nine laboratories and applied in clinical trials by three laboratories. Results A total of five hundred bottles of RM for positive serum specific for genotype Ⅱ ASFV strain HLJ/18 were successfully prepared. The results indicated that the RM we prepared was pure, homogenous, and free of exogenous virus contamination, showing good specificity, homogeneity, and stability. The RM was stable for at least 18 months when it was stored at -20 ℃ and for at least 7 days at 4 ℃, 25 ℃, and 37 ℃. The characterization by the nine laboratories showed that the RM was positive for antibodies against genotype Ⅱ ASFV. Conclusion The positive serum specific for genotype Ⅱ ASFV strain HLJ/18 has successfully been prepared, providing critical material for ASF detection and diagnosis.

目的 研制基因Ⅱ型非洲猪瘟病毒(African swine fever virus, ASFV)阳性血清定性标准样品,为该病的血清学检测、质控及实验室检测能力验证等工作提供实物标准。 方法 以灭活的基因Ⅱ型ASFV (HLJ/18株)免疫无特定病原体(specific pathogen-free, SPF)猪所获得的阳性血清为原料制备标准样品,采用间接酶联免疫吸附试验(indirect enzyme-linked immunosorbent assay, iELISA)等方法对该标准样品的纯净性、特异性、均匀性和稳定性等进行检验与评估,并委托9家实验室进行协作定值,3家实验室进行试用。 结果 成功研制出500瓶基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品,该标准样品纯净、特异、均匀,在-20 ℃条件下可稳定保存至少18个月,在4、25和37 ℃均可至少稳定保存7 d。9家实验室协作定值结果一致,均判定为ASFV抗体阳性。 结论 本研究成功研制了基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品,为该病检测相关工作提供了关键材料。

目的 研制基因Ⅱ型非洲猪瘟病毒(African swine fever virus, ASFV)阳性血清定性标准样品,为该病的血清学检测、质控及实验室检测能力验证等工作提供实物标准。 方法 以灭活的基因Ⅱ型ASFV (HLJ/18株)免疫无特定病原体(specific pathogen-free, SPF)猪所获得的阳性血清为原料制备标准样品,采用间接酶联免疫吸附试验(indirect enzyme-linked immunosorbent assay, iELISA)等方法对该标准样品的纯净性、特异性、均匀性和稳定性等进行检验与评估,并委托9家实验室进行协作定值,3家实验室进行试用。 结果 成功研制出500瓶基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品,该标准样品纯净、特异、均匀,在-20 ℃条件下可稳定保存至少18个月,在4、25和37 ℃均可至少稳定保存7 d。9家实验室协作定值结果一致,均判定为ASFV抗体阳性。 结论 本研究成功研制了基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品,为该病检测相关工作提供了关键材料。

Objective To prepare reference material (RM) for positive serum specific for genotype Ⅱ African swine fever virus (ASFV) for serological detection, quality control, and proficiency testing (PT). Methods The anti-serum collected from specific pathogen-free (SPF) swine immunized with inactivated genotype Ⅱ ASFV was used as raw material for the preparation of RM. Indirect enzyme-linked immunosorbent assay (iELISA) was employed to evaluate the purity, specificity, homogeneity, and stability of RM. In addition, RM was characterized by nine laboratories and applied in clinical trials by three laboratories. Results A total of five hundred bottles of RM for positive serum specific for genotype Ⅱ ASFV strain HLJ/18 were successfully prepared. The results indicated that the RM we prepared was pure, homogenous, and free of exogenous virus contamination, showing good specificity, homogeneity, and stability. The RM was stable for at least 18 months when it was stored at -20 ℃ and for at least 7 days at 4 ℃, 25 ℃, and 37 ℃. The characterization by the nine laboratories showed that the RM was positive for antibodies against genotype Ⅱ ASFV. Conclusion The positive serum specific for genotype Ⅱ ASFV strain HLJ/18 has successfully been prepared, providing critical material for ASF detection and diagnosis.

非洲猪瘟(African swine fever, ASF)是由非洲猪瘟病毒(African swine fever virus, ASFV)感染家猪和野猪引发的一种急性、热性、烈性且高度接触性的传染病,其发病率和病死率可高达100%[1-2]。该病对我国乃至世界养猪业危害极大,其临床症状主要表现为高热、皮肤和组织器官出血、关节肿胀、脾脏肿大且充血变黑等[3-4]。ASF被世界动物卫生组织(World Organisation for Animal Health, WOAH)列为法定通报疫病,在我国被列为一类动物疫病[5-6]
自2018年ASF传入我国以来,已对我国养猪业构成持续威胁,甚至严重影响猪肉食品供应[7-8]。根据ASFV衣壳蛋白(p72)编码基因B646L的差异,研究者将ASFV分为24种基因型[9]。当前,我国ASFV的优势流行毒株为重组毒株,HLJ/18株作为2018年首次发现的早期代表性毒株,具有重要的流行病学意义和研究价值[10]。目前,世界范围内尚无安全有效的ASF商品化疫苗,因此在ASF防控体系中血清学检测对于病原感染溯源以及未来疫苗免疫评估等至关重要,其标准化程度直接影响检测结果的准确性、可靠性、一致性和科学性。虽然我国已建立以GB/T 18648—2020《非洲猪瘟诊断技术》[11]为核心的诊断技术标准,并推荐使用包括间接酶联免疫吸附试验(enzyme-linked immunosorbent assay, ELISA)、竞争ELISA等[12]在内的多种血清学检测方法,但在实际应用中仍面临巨大困境和挑战,即缺乏与我国ASFV流行毒株相匹配的阳性血清标准样品。这导致了一系列问题,例如:不同实验室使用的内部对照品/质控品来源不同、效价参差不齐;商品化试剂盒性能评估缺乏统一的阳性基准;实验室间检测能力比对和验证等缺乏可靠的评判依据,难以科学、准确地评价检测方法的敏感性和特异性。此外,实物标准的缺乏阻碍了上述国家标准等文字标准的有效实施和应用,这一问题已成为制约兽医实验室ASF相关检测质量提升的重要因素之一。
本研究选取在我国早期ASF疫情中具有代表性的基因Ⅱ型ASFV HLJ/18株作为研究对象,按照国家标准样品研制相关技术流程,研制基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品,以提升我国兽医实验室ASF诊断检测质量控制和标准化建设水平,这对完善我国ASF综合防控技术体系具有重要的现实意义。
本标准样品制备所用的基因Ⅱ型ASFV (HLJ/18株)由中国农业科学院哈尔滨兽医研究所分离、鉴定并保存,该毒株的GenBank登录号为MK333180.1;40日龄无特定病原体(specific pathogen free, SPF)猪购自中国农业科学院哈尔滨兽医研究所试验动物基地;猪骨髓巨噬细胞(bone marrow-derived macrophages, BMDMs)和猪肺泡巨噬细胞(porcine alveolar macrophages, PAMs)均分离自上述SPF猪;家兔抗ASFV p72多克隆抗体由中国农业科学院哈尔滨兽医研究所制备。
异硫氰酸荧光素(fluorescein isothiocyanate, FITC)标记的山羊抗家兔IgG,Millipore Sigma公司;T.G培养基,广州捷信生物技术有限公司;TSB培养基,北京沃格东方科技有限公司;非洲猪瘟病毒ELISA抗体检测试剂盒(包被抗原为大肠杆菌表达的ASFV p30重组蛋白),北京金诺百泰生物技术有限公司。
冻干机,Christ公司;酶标检测仪,Bio-Tek公司;倒置荧光显微镜,徕卡显微系统(上海)贸易有限公司。
将SPF猪安乐死后取其四肢骨头,破骨制备BMDMs,用该细胞培养基因Ⅱ型ASFV (HLJ/18株),经连续3次传代后收集病毒,采用GB/T 18648—2020中的荧光PCR方法进行鉴定[11],并测序验证。本研究中所有感染性非洲猪瘟病毒操作以及相关动物实验均在中国农业科学院哈尔滨兽医研究所国家动物疫病防控高级别生物安全实验室完成。本研究有关非洲猪瘟病毒活动资质由农业农村部畜牧兽医局授权,所有动物实验均获得中国农业科学院哈尔滨兽医研究所生物安全委员会审核批准,编号为220113-04-GT。
将基因Ⅱ型ASFV (HLJ/18株)进行10倍系列稀释后接种于96孔板中培养的PAMs,每个稀释度接种8孔,同时设置空白对照。接种后培养7 d,采用免疫荧光试验(immunofluorescence assay, IFA)进行检测,观察并记录出现绿色荧光的孔数,根据Reed-Muench法计算病毒的滴度。
在100 mL上述基因Ⅱ型ASFV (HLJ/18株)培养液中加入β-丙内酯,使其体积分数达到0.1%,置于4 ℃灭活72 h。取灭活的病毒液,按体积分数为1.0%接种PAMs,在37 ℃、5% CO2培养箱中培养5 d后收集细胞培养物,冻融1次后盲传3代。取盲传样品按1%接种PAMs,在37 ℃、5% CO2培养箱中培养7 d后进行IFA检测,以确保病毒灭活完全。检测抗体为兔抗ASFV p72多克隆抗体,二抗为FITC标记的山羊抗兔IgG。
将上述灭活的基因Ⅱ型ASFV (HLJ/18株)与弗氏完全佐剂等量(体积比)混合乳化制备免疫原,经肌内注射途径免疫2头SPF猪,2 mL/头,免疫3次,每次间隔3周。最后一次免疫1周后,通过前腔静脉采集血液,分离血清。采用GB/T 18648—2020《非洲猪瘟诊断技术》中的间接免疫荧光试验(indirect immunofluorescence assay, IIFA)和间接ELISA方法测定血清效价[11]。所有动物实验均在中国农业科学院哈尔滨兽医研究所动物实验中心进行。
将采集的基因Ⅱ型ASFV (HLJ/18株)阳性血清与含有小牛血清和环糊精等组分的冻干保护剂按1:9 (体积比)的比例充分混合,分装到西林冻干玻璃瓶中,0.5 mL/瓶。将样品先置于-80 ℃冰箱预冻2 h后,迅速转移到冻干机中,按设定的冻干程序进行冻干。将制备好的标准样品置于-20 ℃冰箱中冻存备用。
肉眼观察制备好的标准样品的外观物理性状,然后随机抽取3瓶,每瓶中加入0.5 mL PBS (pH 7.2),轻轻摇晃使其充分溶解,并观察复溶效果。
将上述制备好的标准样品分别接种于T.G培养基中,在25 ℃和37 ℃条件下培养,同时在TSB培养基中于25 ℃条件下培养7 d,每日观察是否有菌落生长。同时,按照《中国兽药典》(2020版)中“3308支原体检验法”进行支原体检验[13]
采用随机分层抽样方式,抽取15瓶所制备的标准样品,采用国家标准GB/T 18648—2020中的间接ELISA方法进行检测[11],每瓶重复检测2次,采用单因素方差分析对标准样品的均匀性进行分析。
对所制备的标准样品在不同温度和时间条件下的稳定性进行监测和评估。模拟运输稳定性(短期稳定性):将标准样品分别在-20、4、25、37 ℃保存,分别在-20、4、25 ℃条件第7天以及37 ℃条件下第1、3、5、7天进行检测。每个温度和时间节点各取样3瓶,每瓶重复检测2次,对检测结果进行分析。长期稳定性:将标样于-20 ℃保存,在第0、2、6、12、18月随机抽样3瓶同上检测,每瓶重复检测2次,监测一年以上,对检测结果进行分析。冻融稳定性:随机抽取3瓶标准样品进行0、5、10次反复冻融,每瓶重复检测2次,计算均值,进行稳定性分析。稳定性监测所用的方法为GB/T 18648—2020中的间接ELISA方法[11]
采用在行业内具有一定权威性和资质条件的9家实验室进行协作定值,并安排3家实验室进行试用。所有实验室均采用统一规定的定值方案进行协作定值,要求有2个或2个以上实验技术人员独立进行操作,且尽可能使用不同的仪器设备。每人采用GB/T 18648—2020中所述的间接ELISA和IIFA这2种方法分别对5份标准样品进行协作定值检测[11]。实验数据经统计分析后出具协作定值报告和试用报告,返回本实验室进行确认和汇总分析。
本研究将基因Ⅱ型ASFV (HLJ/18株)在10层细胞工厂中进行培养,最终获得约2 000 mL细胞培养病毒液。荧光定量PCR检测结果为ASFV阳性,测序验证正确,与预期HLJ/18株基因序列一致。采用IFA对ASFV (HLJ/18株)细胞培养病毒液进行病毒滴度测定,结果显示培养的ASFV毒价为107.50 TCID50/mL。
取100 mL上述ASFV (HLJ/18株)培养物,加入β-丙内酯,使β-丙内酯在该培养物体系中的体积分数达到0.1%,将该混合物置于4 ℃条件下灭活72 h。灭活处理完成后,将样品在PAMs上培养和连续盲传3代,随后采用IFA方法对盲传后的样品进行检测,结果为阴性,未观察到特异性荧光,表明该ASFV (HLJ/18株)已被完全灭活。
将上述灭活的ASFV (HLJ/18株)与弗氏完全佐剂充分乳化后免疫SPF猪,3次免疫后采集血液,分离血清。将收集的血清采用IIFA进行检测,结果显示,1:400-1:12 800系列稀释的血清样品均检测到特异性绿色荧光,而1:25 600稀释的血清及阴性对照无特异性荧光(图1),表明所制备的基因Ⅱ型ASFV (HLJ/18株)血清IIFA效价为1:12 800。
将上述血清采用间接ELISA方法进行检测,结果显示所制备的基因Ⅱ型ASFV (HLJ/18株)阳性血清ELISA效价为1:6 400 (表1)。
将标准样品和冻干保护剂充分混合后,采用表2中冻干程序进行冻干,共制备500瓶标准样品。将所制备的标准样品置于-20 ℃冰箱中冻存,后续按2.2节标准样品的检验和评估方法,对其物理性状、复溶效果等指标进行质量检定,确保所制备的标准样品合格。
冻干后的标准样品外观呈白色、疏松、块状物(图2A)。加入PBS (pH 7.2)后,该标准样品迅速溶解,复溶后液体澄清,呈淡红色,无浑浊和沉淀(图2B)。结果表明,所制备的标准样品外观性状和复溶效果良好。按同样技术工艺共制备500瓶该标准样品(图2C)。
将上述制备好的标准样品在T.G培养基中分别于25 ℃和37 ℃条件下培养,同时在TSB培养基中在25 ℃条件下培养7 d,每日观察是否有菌落生长,结果无菌落生长。此外,支原体检测结果为阴性。结果表明所制备的标准样本中无支原体及其他微生物污染。
按照GB/T 15000.3—2023《标准样品工作导则 第3部分:标准样品定值和均匀性与稳定性评估》[14]要求对标准样品的均匀性进行检验。采取随机分层方式抽取15瓶标准样品,采用GB/T 18648—2020[11]中的间接ELISA方法进行检测,每个样品重复检测2次,采用单因素方差分析法对检测结果进行分析。检测结果经统计分析,结果如表3所示,P=0.146>0.05,表明标准样品各瓶单元之间无显著差异。根据自由度(degree of freedom, df)及显著性水平,可由表查临界值的F值,算得F< [F0.05 (14,15)=2.46],表明组内与组间差异无统计学意义,样品是均匀的。
将标准样品在-20、4、25、37 ℃ 条件下,分别在-20、4、25 ℃第7天及37 ℃下第1、3、5、7天进行稳定性监测。每个温度和时间节点各取样3瓶,每瓶重复检测2次,对检测结果分别进行t检验统计分析。结果显示,标准样品在-20、4、25、37 ℃环境中经检测和对检测结果进行统计分析后,结果P值均大于0.05,表明标准样品在不同温度及时间条件下无显著差异,能满足实际测量的需求。本标准样品在-20、4、25、37 ℃下均可稳定保存7 d (表4)。
本研究贮存条件为-20 ℃,样品放置数量应足够检测至第18个月,检测时间为第0、2、6、12、18个月,每个时间点随机抽取3瓶标准样品,每瓶重复检测2次,计算均值,进行稳定性分析,采用国标GB/T 18648—2020中的间接ELISA方法进行检测[11]。对所制备的ASFV (HLJ/18株)阳性血清定性标准样品在-20 ℃贮存条件下稳定性检测结果进行一元线性回归分析,计算得到斜率(β1)为0.008,t0.95,3×s (β1)=3.182×0.007=0.022,数据线性回归分析结果判断依据|β1|<t0.95,3×s(β1),认为斜率无显著差异,能满足实际测量的需要(表5图3)。结果表明,本标准样品在-20 ℃可稳定保存至少18个月,本研究将持续监测和评估该标准样品的稳定性。
将实验组与对照组间进行不同组间独立样本t检验:冻融0次与冻融10次后(表6),首先进行莱文方差等同性检验,当P值>0.05时,表明两组方差差异不显著,即方差齐性,t=-1.556,自由度(df)=10,P=0.151>0.05,因此认为反复冻融最大次数10次后,该标准样品的检测值与对照组中标准样品的检测值不存在显著差异,可认为本标准样品能经受10次反复冻融操作。
将所研制的标准样品通过“泡沫箱+冰袋”的运输方式寄往9家具有一定检测资质[如已通过中国合格评定国家认可委员会(China National Accreditation Service for Conformity Assessment, CNAS)认可或已获得中国计量认证(China Metrology Accreditation, CMA)资质]的实验室进行协作定值。9家实验室检测结果均为非洲猪瘟病毒抗体阳性,测得的效价均为1:6 400,与本实验室所测得的效价一致。此效价仅作为标准样品研制过程中的参考依据,用于制备环节的质量控制和稳定性监测。从选取的3家实验室试用的检测结果显示,所有寄送的标准样品试用检测效价均为1:6 400,说明该标准样品的均一性良好,产品质量稳定,可应用到实际检测工作中。
国务院印发的计量发展规划(2013-2020年)通知中指出,要鼓励自主研发标准物质,填补部分领域标准物质空白[15]。基于目前我国因严重缺乏ASFV阳性血清标准品(标准样品或标准物质)而导致不同实验室间检测能力验证和比对等工作难以开展以及相关检测文字标准难以有效实施等现实问题和实际需求,本研究根据GB/T 15000—2023《标准样品工作导则》[14]和GB/T 18648—2020《非洲猪瘟诊断技术》[11]等国家标准,成功研制了针对基因Ⅱ型非洲猪瘟病毒ASFV (HLJ/18株)的阳性血清定性标准样品。该样品适用于ASFV血清学检测中的特异性鉴定、室内与室间质控、试剂性能评价及检测能力验证等多项工作环节,是目前国内乃至国际范围内首个针对基因Ⅱ型ASFV且具备明确溯源性与规范研制流程的阳性血清标准样品。
此前,本实验室和北京海关(现中国海关科学技术中心)等单位分别制备了非洲猪瘟病毒p72蛋白基因(B646L)质粒样品,并应用于非洲猪瘟病毒核酸检测能力验证工作[6]。然而,该项标准物质仅针对B646L基因,无法用于ASFV其他基因检测的质控,且该标准物质类型为质粒,不能对核酸抽提过程进行质控。按照GB/T 15000《标准样品工作导则》[14]和GB/T 18648—2020《非洲猪瘟诊断技术》[11]等标准,本实验室成功研制了基因Ⅰ型ASFV (SD/DY-Ⅰ/21株)阳性血清定性标准样品[17]。然而,该标准样品仅针对基因Ⅰ型ASFV,与本研究所用的基因Ⅱ型ASFV (HLJ/18株)毒株在基因型上不同,2个毒株免疫后所制备的血清交叉反应性很小,因此不能将基因Ⅰ型ASFV毒株(如SD/DY-Ⅰ/21株)所制备的阳性血清定性标准样品用于基因Ⅱ型ASFV (如HLJ/18株)血清学检测。因此,2种标准样品在靶向毒株基因型上不同,分别适配不同基因型ASFV相关血清学检测需求。本研究制备了基因Ⅱ型ASFV阳性血清定性标准样品,较好地完善了ASFV血清学检测用标准样品的覆盖体系。
通过检索全国标准信息公共服务平台、国际标准物质数据库(Code d’Indexation des Matériaux de Référence/International database for certified reference materials, COMAR)等国内外权威标准物质/标准样品相关数据库,结果显示,目前国内外均无针对基因Ⅱ型ASFV血清学检测所需的阳性血清对照或质控品,也无相关有证标准样品(CRM)供应或研制项目立项。此前相关研究多集中于ASFV核酸标准品的研制[18]和应用,但涉及ASFV活病毒操作及动物免疫的血清标准品研制鲜有报道。因此,本研究所研制的基因Ⅱ型ASFV (HLJ/18株)阳性血清定性标准样品填补了我国ASFV血清类标准品研制领域空白,对推动我国兽医实验室ASF检测标准化具有重要意义。
阳性血清标准样品可用于校准ELISA等血清学检测方法,提高这些方法的灵敏度和特异性[19]。尽管如此,标准样品研制需遵循严格的技术流程,整个研制过程均要做到可清晰溯源、可查证、可重复。本标准样品的整个研制过程均可清晰溯源。在原料溯源方面,本研究免疫用ASFV毒株由中国农业科学院哈尔滨兽医研究所分离鉴定,GenBank登录号为MK333180.1,该毒株已用于ASF候选疫苗研制,该病毒株的生物学特性(如毒力、致病性、传播特点)等背景信息清晰,可溯源。因此,本研究所用HLJ/18株病毒具有明确的基因组背景,为制备高特异性标准样品提供了可靠的原料基础。在定值方法溯源方面,在研制过程中,本研究严格遵循GB/T 15000系列标准[14]以及GB/T 18648—2020《非洲猪瘟诊断技术》[11]等国家标准,完成了从病毒培养、灭活验证、动物免疫、样品制备等关键步骤,并系统开展了均匀性、稳定性评估及多实验室协作定值等工作,所用的定值方法均可溯源到国家标准,可靠性高。鉴于ASFV的高致病性和生物安全风险(需在BSL-3或ABSL-3实验室操作),其阳性血清制备对实验设施和技术人员资质要求极为严格。本研究依托具备相应生物安全资质的专业实验室平台完成,这一条件保障了制备过程的规范性与安全性,也意味着此类标准样品难以在普通实验室随意制备,凸显了本研究所研制的标准样品的稀缺性和重要价值。相比之下,国外针对ASFV的标准样品/标准物质研究也多集中于核酸标准品,而涉及活病毒操作及动物免疫的血清标准品研制报道同样罕见。在定值过程溯源方面,本实验室以及9家参与协作定值的实验室所用的仪器设备均已经过正规专业机构校准(具有校准证书),所用的试剂均为商品化合规试剂,试剂盒均有生产文号。此外,本实验室以及参与协作定值的大多数实验室均已通过CNAS认可或CMA认证,或多次参加全国兽医实验室检测能力比对且结果良好,参加定值的实验室人员大多具有本单位实验室检测工作上岗证。因此,本研究所制备的阳性血清标准样品在原料、定值方法、定值过程等方面均可清晰溯源,具有背景明确、特异性强、效价高(协作定值效价达1:6 400)、物理性状良好、无生物安全风险及支原体污染等优点,可与现行ELISA和IIFA方法配套使用[20]
对比国内外相关研究,本研究研制的阳性血清标准样品在核心性能指标上展现出显著优势。在兽医领域,对于动物病原核酸标准样品研制的报道较常见,但针对血清类标准样品研制的报道相对较少,特别是对于要求必须在生物安全三级(biosafety level 3, BSL-3)实验室进行实验操作的高致病性病原微生物。在动物病毒阳性血清标准样品研制领域,目前主要有三大痛点:一是均匀性控制难度大,在禽白血病病毒(avian leucosis virus, ALV)、猪瘟病毒(classical swine fever virus, CSFV)等血清标准品研制中,单元间抗体效价变异系数多在5%-8%之间[21],甚至超过10%,难以完全满足国家标准GB/T 15000.3对样品均匀性的严格要求;二是溯源性描述不足甚至缺失,部分研究资料中未明确提供病毒原料的背景信息(如毒株名称、基因组GenBank登录号等),而且在制备流程中缺乏系统规范的记录,导致其无法实现全程溯源[22];三是适用场景局限,部分血清质控品仅能匹配单一检测方法,兼容性较差。本研究中所研制的标准样品经均匀性检验,结果显示均匀性良好,单元瓶之间在效价上无显著差异,满足标准样品研制的技术要求,这一良好的均匀性确保了不同分装单元在作为质控品使用时结果的可比性[23]。稳定性研究显示:该标准样品在-20 ℃条件下可稳定保存至少18个月,在4、25、37 ℃条件下均可稳定保存7 d;经10次反复冻融后抗体效价仍保持稳定,远优于某些动物病毒血清标准样品,例如ALV[24]和鸡法氏囊病病毒(infectious bursal virus, IBDV)[25]血清标准样品通常仅耐受3-4次冻融,表明该标准样品具备良好的实用性和重复使用性。协作定值方面,本研究联合9家具有一定检测资质或权威检测能力的单位(包括省级动物疫病预防控制中心、科研院所以及海关系统等)开展协作定值,结果一致性强,保证了定值结果在技术能力和管理体系上的可靠性与权威性。这种多家实验室协作模式是国际公认的、用于保证标准样品定值结果准确性和溯源性的最佳实践之一。这种模式相较于单一实验室定值,有效规避了个体实验差异带来的系统误差,提升了定值结果的准确性和溯源性,这也是本研究成果具备推广应用价值的重要基础。现有部分动物病毒血清标准样品的研制采用单一实验室定值方式,定值结果的权威性和认可度相对较低,难以满足全国范围内实验室检测标准化的需求。
尽管如此,本研究仍存在一定的局限性。目前工作主要集中于样品本身的制备与基本验证,尚未系统开展与口蹄疫、猪瘟、猪繁殖与呼吸综合征等其他常见猪病血清标准样品的交叉反应试验及诊断性能(如灵敏性、特异性)的全面评价[26]。这些不足可能影响其在复杂临床背景下的推广适用性。未来将持续开展该标准样品的稳定性监测,补充交叉反应性和诊断性能验证,推动该标准样品纳入国家动物疫病监测体系与实验室能力比对项目,并加强宣传推广,提升其在国内外的认可度与应用水平,为我国动物疫病防控工作提供有力技术支撑。
