Equine coronavirus (ECoV) is an emerging virus attacking the gastrointestinal tract in horses, and the infected adult horses mainly present fever, abdominal pain, and diarrhea. In 1975, ECoV infection first appeared in the United States, and since then it has been prevalent in many countries and regions. Only one recombinant strain of ECoV was isolated from the small intestine of a donkey experiencing diarrhea in Shandong Province, China. [Objective] Understanding the genetic composition, genetic relationship, and biological characteristics of ECoV strains in China can lay a foundation for unveiling the epidemic status and genetic evolution trend of ECoV and provide materials for the research and development of products for the prevention and control of ECoV. [Methods] RT-PCR was employed to detect the stool samples from a horse experiencing diarrhea in Huangpi District, Wuhan City, Hubei Province. The virus was isolated from the positive samples and verified by indirect immunofluorescence assay (IFA) with monoclonal antibodies targeting the S1 protein of ECoV. Based on the whole genome sequencing results of the isolate, the phylogenetic analysis and sequence alignments of the whole genome, N gene, and NS2 gene were performed. [Results] An ECoV strain was successfully isolated and named ECoV-JL. Transmission electron microscopy (TEM) showed that the isolated virus particles were spheroidal and had a capsule membrane and a typical spiroid structure of coronaviruses. The tissue culture infectious dose 50% (TCID50) of ECoV-JL reached a peak of 106.16 TCID50/mL 72 h post infection in HRT-18 cells. ECoV-JL strains could infect three human cell lines: HRT-18 (human ileocecal cancer cells), Caco-2 (human colorectal adenocarcinoma cells), and Huh7 (human liver cancer cells). The genome sequence of ECoV-JL and the ECoV genome sequences in GenBank showed the similarity within the range of 97.9%–99.0%. ECoV-JL was in a separate branch of the phylogenetic tree and far related to other strains, which indicated that ECoV-JL might be derived from recombination mutations. The NS2 gene presented more mutations, and the difference in NS2 gene was the main reason for the poor homology between ECoV-JL and other strains. [Conclusion] We isolated and identified an ECoV strain from the stool samples of horses with diarrhea and named it ECOV-JL. The study about the biological characteristics and phylogenetic relationship of this strain reflected the characteristics of the epidemic strains in Hubei Province, providing a clue for the epidemic status and evolution trend of ECoV in China.

马冠状病毒病是由马冠状病毒(equine coronavirus, ECoV)引起的一种马新发胃肠道病毒病,成年马感染后主要出现发烧、腹痛和腹泻等症状。1975年,马冠状病毒感染首次在美国出现,此后在多个国家和地区均有流行,此前我国仅从山东腹泻驴的小肠样品中分离得到了一株重组马冠状病毒。【目的】了解ECoV中国毒株的基因组成、亲缘关系以及生物学特性,可以为我国ECoV流行现状和遗传演化趋势提供依据,为ECoV防控产品的研发提供材料。【方法】对湖北省武汉市黄陂区腹泻马匹的粪便样品进行RT-PCR检测,对检测阳性样品进行病毒分离,并利用靶向ECoV S1蛋白的单克隆抗体通过间接免疫荧光试验(indirect immunofluorescence assay, IFA)对分离的病毒进行验证。根据ECoV-JL株全基因组测序结果,对全基因组、N基因和NS2基因进行了基因组系统发育分析和同源性比较。【结果】成功分离到一株ECoV,并命名为ECoV-JL。透射电镜(transmission electron microscopy, TEM)观察分离到的病毒颗粒呈球状,且具有囊膜和冠状病毒典型的纤突结构。该分离株感染HRT-18细胞72 h后病毒滴度可到达峰值,半数组织培养感染剂量(tissue culture infectious dose 50%, TCID50)约为106.16 TCID50/mL。ECoV-JL毒株可以在人回盲肠癌(human ileocecal cancer-18, HRT-18)细胞、人结直肠腺癌(human colorectal adenocarcinoma-2, Caco-2)细胞和人肝癌(human liver cancer cells, Huh7)细胞上稳定传代。ECoV-JL株与GenBank中现有的ECoV全基因组序列相似性为97.9%−99.0%,系统发育分析发现ECoV-JL株属于单独的演化分支,与其他毒株的亲缘关系较远,说明ECoV-JL株可能是重组变异而来,其中NS2基因突变较多,NS2基因编码的差异是造成ECoV-JL株与其他毒株同源性较差的主要原因。【结论】本研究从腹泻马的粪便样品中成功分离并鉴定了一株ECoV,将其命名为ECoV-JL株,对该毒株生物学特性和亲缘关系的研究反映了湖北地区流行毒株的特点,为我国ECoV流行现状和演化趋势提供重要依据。

马冠状病毒病是由马冠状病毒(equine coronavirus, ECoV)引起的一种马新发胃肠道病毒病,成年马感染后主要出现发烧、腹痛和腹泻等症状。1975年,马冠状病毒感染首次在美国出现,此后在多个国家和地区均有流行,此前我国仅从山东腹泻驴的小肠样品中分离得到了一株重组马冠状病毒。【目的】了解ECoV中国毒株的基因组成、亲缘关系以及生物学特性,可以为我国ECoV流行现状和遗传演化趋势提供依据,为ECoV防控产品的研发提供材料。【方法】对湖北省武汉市黄陂区腹泻马匹的粪便样品进行RT-PCR检测,对检测阳性样品进行病毒分离,并利用靶向ECoV S1蛋白的单克隆抗体通过间接免疫荧光试验(indirect immunofluorescence assay, IFA)对分离的病毒进行验证。根据ECoV-JL株全基因组测序结果,对全基因组、N基因和NS2基因进行了基因组系统发育分析和同源性比较。【结果】成功分离到一株ECoV,并命名为ECoV-JL。透射电镜(transmission electron microscopy, TEM)观察分离到的病毒颗粒呈球状,且具有囊膜和冠状病毒典型的纤突结构。该分离株感染HRT-18细胞72 h后病毒滴度可到达峰值,半数组织培养感染剂量(tissue culture infectious dose 50%, TCID50)约为106.16 TCID50/mL。ECoV-JL毒株可以在人回盲肠癌(human ileocecal cancer-18, HRT-18)细胞、人结直肠腺癌(human colorectal adenocarcinoma-2, Caco-2)细胞和人肝癌(human liver cancer cells, Huh7)细胞上稳定传代。ECoV-JL株与GenBank中现有的ECoV全基因组序列相似性为97.9%−99.0%,系统发育分析发现ECoV-JL株属于单独的演化分支,与其他毒株的亲缘关系较远,说明ECoV-JL株可能是重组变异而来,其中NS2基因突变较多,NS2基因编码的差异是造成ECoV-JL株与其他毒株同源性较差的主要原因。【结论】本研究从腹泻马的粪便样品中成功分离并鉴定了一株ECoV,将其命名为ECoV-JL株,对该毒株生物学特性和亲缘关系的研究反映了湖北地区流行毒株的特点,为我国ECoV流行现状和演化趋势提供重要依据。

Equine coronavirus (ECoV) is an emerging virus attacking the gastrointestinal tract in horses, and the infected adult horses mainly present fever, abdominal pain, and diarrhea. In 1975, ECoV infection first appeared in the United States, and since then it has been prevalent in many countries and regions. Only one recombinant strain of ECoV was isolated from the small intestine of a donkey experiencing diarrhea in Shandong Province, China. [Objective] Understanding the genetic composition, genetic relationship, and biological characteristics of ECoV strains in China can lay a foundation for unveiling the epidemic status and genetic evolution trend of ECoV and provide materials for the research and development of products for the prevention and control of ECoV. [Methods] RT-PCR was employed to detect the stool samples from a horse experiencing diarrhea in Huangpi District, Wuhan City, Hubei Province. The virus was isolated from the positive samples and verified by indirect immunofluorescence assay (IFA) with monoclonal antibodies targeting the S1 protein of ECoV. Based on the whole genome sequencing results of the isolate, the phylogenetic analysis and sequence alignments of the whole genome, N gene, and NS2 gene were performed. [Results] An ECoV strain was successfully isolated and named ECoV-JL. Transmission electron microscopy (TEM) showed that the isolated virus particles were spheroidal and had a capsule membrane and a typical spiroid structure of coronaviruses. The tissue culture infectious dose 50% (TCID50) of ECoV-JL reached a peak of 106.16 TCID50/mL 72 h post infection in HRT-18 cells. ECoV-JL strains could infect three human cell lines: HRT-18 (human ileocecal cancer cells), Caco-2 (human colorectal adenocarcinoma cells), and Huh7 (human liver cancer cells). The genome sequence of ECoV-JL and the ECoV genome sequences in GenBank showed the similarity within the range of 97.9%–99.0%. ECoV-JL was in a separate branch of the phylogenetic tree and far related to other strains, which indicated that ECoV-JL might be derived from recombination mutations. The NS2 gene presented more mutations, and the difference in NS2 gene was the main reason for the poor homology between ECoV-JL and other strains. [Conclusion] We isolated and identified an ECoV strain from the stool samples of horses with diarrhea and named it ECOV-JL. The study about the biological characteristics and phylogenetic relationship of this strain reflected the characteristics of the epidemic strains in Hubei Province, providing a clue for the epidemic status and evolution trend of ECoV in China.

Zhang等研究表明马冠状病毒(equine coronavirus, ECoV)通过粪口传播,成年马患病后多表现为发烧、嗜睡、厌食、腹痛和腹泻[1]。1975年,Bass等在美国马群中发现了40多例以水样腹泻、发烧和淋巴组织病变为特征的传染性疾病,通过血清学研究和电子显微镜观察,将这些症状归因于马的冠状病毒感染,标志着马中首次出现ECoV感染[2]。直至2000年,Guy等首次在美国北卡罗来纳州腹泻马驹的粪便中分离到一株ECoV病毒[3]。近期,ECoV在亚洲(日本、韩国)和欧洲的一些国家也有报道[2,4]。目前,我国对于ECoV病毒感染的报道较少,仅在山东省某驴场腹泻病驴的小肠中鉴定并分离到一株ECoV病毒,证实了该病毒已经开始在中国传播[5]
ECoV感染症状温和,多表现为胃肠道症状,临床症状不具有特征性,部分马感染后表现为无症状,但用qPCR方法可从粪便中检测出马冠状病毒。目前该病原诊断主要依靠血清学实验、PCR检测和病毒分离鉴定。Tsunemitsu等、Pusterla等、Fielding等和Nemoto等分别建立了RT-PCR和qPCR方法检测ECoV[6-9]。虽然我国仅有一例马冠状病毒的报道,但是仇钰等用自主研发的酶联免疫吸附试验(enzyme-linked immuno sorbent assay, ELISA)检测方法检测了711份国内马血清样品,其中阳性血清13份(1.83%)[10],该结果说明马冠状病毒可能以隐性感染的方式存在于我国的马群中。马冠状病毒病还没有有效的疫苗或抗病毒药物,因此预防该疾病只能通过实施检疫、严格隔离协议和保持环境卫生等措施来实现。ECoV的成功分离对于开发治疗或预防ECoV相关有效抗病毒药物或疫苗,以及进行各种ECoV研究至关重要。
ECoV是单股正链,不分节段,有囊膜的RNA病毒,属于冠状病毒科正冠状病毒亚科,包含一个单链正链RNA基因组,大小为27−32 kb。病毒RNA具有5′端帽结构,其后是一个先导RNA序列和200−400 bp的未翻译区(untranslated region, UTR)核苷酸;3′端有一个200−500 bp的核苷酸非翻译区(UTR)和ploy(A)尾。分别编码2个复制酶多聚蛋白、5个结构蛋白(血凝素酯酶、纤突、囊膜、膜蛋白和核衣壳)和4个辅助蛋白(NS2、p4.7、p12.7和I)。有研究表明编码的2个复制酶多聚蛋白被由3个病毒编码的蛋白酶分解为16个非结构蛋白(nsp1−16)[1]
本研究采集湖北地区马场腹泻马的粪便样品,经分离鉴定后确定为ECoV感染,并将分离得到的毒株命名为ECoV-JL株,进一步对分离株进行了生物学特性和遗传进化分析,本研究结果为湖北地区可能流行的ECoV毒株提供了参考,为我国ECoV研究提供了重要材料。
本研究采用的粪便样品来源于湖北省武汉市黄陂区腹泻病马,粪便样品于2022年8月寄送本实验室;HRT-18、Caco-2、HeLa-R19、Huh7、HEK293T、NPTr、PK-15和Vero-CCL81细胞均由实验室保存。
胶回收试剂盒购自BioFlux公司;PrimeSTAR HS DNA Polymerase、DL2000 DNA Mmaker购自TaKaRa公司;胎牛血清(fetal bovine serum, FBS)购自BI公司;Dulbecco改良的Eagle培养基(Dulbecco’s modified eagle medium, DMEM)、胰蛋白酶购自Gibco公司;鼠源抗ECoV-S1蛋白的杂交瘤细胞由本实验室制备保存;辣根过氧化物(horseradish peroxide, HRP)标记的羊抗鼠IgG二抗购自ABclonal公司;DyLightTM 594山羊抗小鼠IgG (H+L)二抗购自赛默飞世尔科技公司。
根据GenBank上NC99株全基因(登录号:EF446615),利用NCBI网站Pick Primers工具针对N基因设计一对特异性引物EcoV-F (5′-GC CGACCAGTCCGACCAG-3′)和EcoV-R (5′-TCA GAGTTATCTTCGGTGAGGG-3′)。
无菌采取约1 g粪便,加入1 mL DMEM研磨混匀,反复冻融3次,4 ℃、8 000 r/min离心10 min,取上清液经0.45 µm的过滤器过滤后于−80 ℃保存。
取上述病料上清液200 μL与1 mL TRIzol裂解液混合,室温静置5 min,加入200 μL氯仿后混合均匀,冰上静置10 min。4 ℃、12 000 r/min离心10 min收集上清,与等体积异丙醇混匀,冰上静置10 min,4 ℃、12 000 r/min离心10 min。弃去上清,加入1 mL 95%乙醇洗涤,7 600 r/min离心风干。最后,将RNA溶解在不含RNase的水中,将提取出的RNA保存在–80 ℃。将总RNA反转录为cDNA,用N基因特异性引物进行扩增,预期片段大小为1 256 bp,PCR反应条件:94 ℃ 5 min;94 ℃ 15 s,56 ℃ 30 s,72 ℃ 60 s,扩增35个循环。
将过滤后的悬液接种于HRT-18细胞,放入37 ℃,含5% CO2培养箱中培养。吸附1 h后,加入含1%血清的DMEM,随后每5 d使用显微镜检查培养物以观察细胞病变效应(cytopathic effect, CPE),如发现病变,将剩余上清接种HRT-18细胞进行传代培养,盲传3代并通过RT-PCR、间接免疫荧光试验(indirect immunofluorescence assay, IFA)及透射电镜(transmission electron microscopy, TEM)观察进行进一步鉴定。
HRT-18细胞经0.01的病毒感染复数(multiplicity of infection, MOI)感染96 h后收集病毒液,12 000 r/min离心30 min后通过0.45 µm的过滤器过滤。加入30%的蔗糖,30 000 r/min超速离心2 h,用PBS重悬后再次用20%、30%、45%、60%的蔗糖溶液梯度超速离心脱糖。再次重悬后将重悬液滴加在覆有支持膜的铜网上,5 min后从铜网边缘吸去多余液体,室温风干,滴加2%磷钨酸(pH 7.2)负染2 min,用滤纸吸去多余的染色液,室温风干,干燥后于透射电镜(电压200 kV)下观察。
提取由本实验室保存的靶向S1蛋白的杂交瘤细胞RNA,并且用罗氏反转的方法将总RNA反转录为cDNA,方法同1.5。以cDNA为模板,设计引物扩增得到本株单克隆抗体的轻、重链序列,并构建表达质粒,利用293F细胞表达,经SDS-PAGE鉴定,将成功表达的抗体命名为S1-mAb。
将JL以0.1的病毒感染复数(MOI)感染HRT-18细胞,感染后96 h收取细胞样品。PBS洗涤1次,4%多聚甲醛室温固定15 min,0.1% Triton X-100透化15 min,5%牛血清白蛋白(bovine serum albumin, BSA)封闭2 h,4 ℃ EcoV-S1-mAb孵育过夜。用PBST洗涤3次后,用DyLightTM 594山羊抗小鼠IgG (H+L)在37 ℃下孵育45 min。最后用PBST洗涤细胞3次,用4′, 6-二氨基-2-苯基吲哚(4′, 6-diamidino-2-phenylindole, DAPI)染色液孵育7 min。倒置荧光显微镜下观察图像。
提前24 h以5×103 cell/mL的细胞密度将HRT-18细胞铺于96孔细胞板中准备感染,将JL株进行连续10倍倍比稀释,然后将稀释好的病毒加入各细胞孔中,每孔100 μL,每个稀释度重复8个孔,同时设置不加病毒的正常细胞作对照。37 ℃培养96 h后弃去上清,采用间接免疫荧光试验进行TCID50的测定,步骤同1.9。最后用Reed-Muench的方法计算TCID50
按0.01的病毒感染复数(MOI)感染HRT-18细胞,于37 ℃细胞培养箱中培养,设置12−96 h共8个收毒时间点取样,对不同时间点收取的样品进行TCID50测定,96 h后用间接免疫荧光试验读取细胞病变孔数。应用Reed-Muench法计算每个时间点的病毒滴度TCID50,步骤同1.9,以收毒时间点为横坐标,TCID50的对数为纵坐标建立病毒的一步生长曲线。
依照5的病毒感染复数(MOI)将分离得到的病毒接种到不同的细胞系,包括HRT-18、PK-15、Caco-2、NPTr、Huh7、HeLa-R19、Vero-CCL81和HEK293T细胞。病毒感染96 h后,用间接免疫荧光试验观察病毒感染情况。
参照GenBank上已上传的ECoV毒株序列,在保守区域分段设计5对引物。PCR反应条件:95 ℃预变性5 min;95 ℃变性30 s,55 ℃退火30 s,72 ℃延伸45 s,共35个循环;72 ℃延伸5 min。扩增片段连接平末端载体pEASY-Blunt后,转化Escherichia coli DH10β感受态细胞。12 h后挑取单克隆菌落进行菌落PCR鉴定,阳性菌液送武汉擎科生物科技有限公司测序。
使用MEGA 7.0软件对新分离毒株和已经上传的ECoV参考毒株的全基因组,N基因和NS2基因进行对比,分析其核苷酸差异位点,绘制遗传进化树进行遗传演化分析。本研究中应用的GenBank上已经上传的ECoV参考序列见表1
将处理后的样品进行RT-PCR检测,结果显示,扩增得到1 000−2 000 bp之间的特异性条带,阴性对照样品中未检测到特异性的条带(图1A);经进一步测序分析,扩增的基因为ECoV N基因(图1B),表明粪便样本中存在马冠状病毒。
处理ECoV检测阳性粪便样本,用HRT-18细胞分离病毒。将病毒盲传至第三代,在感染后第4天,收集分离到的病毒,并感染HRT-18细胞,未在HRT-18细胞上观察到冠状病毒诱导的典型细胞病变,即细胞聚集和合胞体形成。
为了对分离到的病毒进行进一步检测,从实验室杂交瘤细胞中扩增得到抗体的轻、重链,并且用293F细胞表达,图2为间接免疫荧光试验(IFA)和SDS-PAGE结果。
图3所示,透射电镜观察结果显示,ECoV为球形病毒粒子,具有囊膜,囊膜上具有明显的纤突,呈现有短的棒状突起,直径约为100 nm,与报道的ECoV形态一致,属于冠状病毒的典型结构。
根据Reed-Muench法测定,增殖动态监测显示其接种HRT-18细胞后病毒滴度在24−72 h逐渐提高,在72 h病毒滴度达到峰值,约为106.16 TCID50/mL,随后开始缓慢下降(图4)。
在病毒感染不同细胞96 h后,收取细胞样品并进行IFA检测,检测结果说明,ECoV可以在3种人源细胞HRT-18细胞(人回盲肠癌细胞)、Caco-2细胞(人结直肠腺癌细胞)和Huh7细胞(人肝癌细胞)上稳定传代。细胞嗜性研究的间接免疫荧光试验结果显示,ECoV-JL株在HRT-18细胞上感染效率最高(图5)。
本研究分离得到的ECoV-JL株基因组全长30 892 bp,经二代测序和基因组序列比对,其结构大致为:ORF1ab (172−21 560 bp)、NS2 (21 575−22 411 bp)、HE (22 423−23 694 bp)、S (23 709−23 800 bp)、E (28 357−28 611 bp)、M (28 626−29 318 bp)、N (29 328−30 668 bp)和I (29 389−30 009 bp)。对ECoV-JL株全基因组核苷酸序列进行分析,ECoV-JL株与GenBank中现有的ECoV全基因组序列相似性为97.9%−99.0%。ECoV-JL株与GenBank中上传的10株ECoV均具有97%以上的遗传相似性。ECoV-JL株与美国报道的马冠状病毒NC99 (GenBank登录号:EF446615.1)相似性最高,一致性为99%;与来源于驴的ECoV 2021/464693和ECoV 2020/7881遗传亲缘性较低。全基因组的系统进化树分析表明,ECoV-JL株单独属于一个演化分支,所处的分支与ECoV NC99 (GenBank登录号:EF446615.1)毒株分支亲缘关系较近。然而,对N基因的分析结果表现出不同的结果,ECoV-JL株与ECoV 2021/464693 (GenBank登录号:OL770366.1)和ECoV 2020/7881 (GenBank登录号:OM937885.1)的相似性最高,亲缘关系与ECoV 2021/464693 (GenBank登录号:OL770366.1)毒株较近。
对基因组NS2同源性和系统进化树的分析结果表明,与GenBank中登记的其他毒株相比,NS2基因突变较多,与分离株同源性最低的是ECoV 2021/464693 (OL770366.1)和ECoV2020/7881 (OM937885.1)株,一致性仅为49.5%。根据NS2基因系统进化树分析,ECoV-JL株与ECoV NC99 (GenBank登录号:EF446615.1)和ECoV Tokachi09 (GenBank登录号:LC061272.1)同处一个大的分支,亲缘关系较近(图6)。
ECoV是一种主要发生于成年马匹的病毒性传染病,目前已经在多个国家流行,造成很大的经济损失。根据有限的文献,ECoV可能以无症状感染的形式在全球范围内的马中广泛分布[11]。2022年,Qi等[5]从山东省某驴场中分离得到一株马冠状病毒,并对该毒株进行了生物学分析,这也是我国第一次分离得到该病毒,结合之前的研究结果,认为马冠状病毒可能已经在中国流行,导致潜伏感染。本研究从马的粪便中分离得到了ECoV新毒株,并且对该毒株进行全基因组测序和病毒特性研究,由于ECoV感染HRT-18细胞后光学显微镜观察未见到典型的细胞聚集和合胞体现象,所以需要借助间接免疫荧光对分离毒株的病毒学特性展开研究,因此实验室原核表达并且纯化了ECoV-S1蛋白,制备了ECoV-S1蛋白的单克隆抗体,为检测ECoV感染的细胞提供了便捷的工具。
本研究首先通过RT-PCR检测成功鉴定到粪便样品中存在病毒,但是盲传至第三代细胞仍然未产生典型的细胞病变,认为可能是RT-PCR检测较为灵敏,但实际病毒含量较低,不足以产生典型的细胞病变,于是决定利用间接免疫荧光和电子显微镜观察进行进一步检测,最终确定样品中确实存在ECoV病毒。
分析全基因组测序结果,来自不同地理区域的毒株基因组序列存在显著差异,本研究分离得到的毒株基因组序列与在美国分离得到的ECoV NC99株的同源性最高,但是与中国距离最近的韩国和日本相似性较低。对全基因组进行系统进化树分析,发现ECoV-JL株属于单独的演化分支,与其他毒株的亲缘关系较远。这些结果均表明,研究分离的病毒产生了变异,这种变异可能是由其他毒株重组造成的,认为这些变异可能是由于远距离传播和不同地区实施的不同疾病预防措施造成的基因突变。
根据以往的研究,ECoV可以编码2种NS2基因,按照所编码的NS2基因长度可以分为837 nt和585 nt两种[1],本研究分离到的ECoV-JL株可以编码长度为837 nt的NS2基因,除此之外ECoV CH21、Tokyo2020、Tokachi09和NC99毒株也可以编码837 nt长度的NS2基因,与分离得到的ECoV-JL株同源性较高,一致性约为98%。NS2蛋白主要参与病毒的自我切割和亚细胞定位,与病毒的自噬密切相关,NS2基因编码的差异是造成ECoV-JL株与部分毒株同源性较差的主要原因。
本研究从湖北省某马场中鉴定并分离到了一株马冠状病毒,将其命名为ECoV-JL株,通过二代测序、生物学鉴定和遗传进化分析,该毒株的分离鉴定一定程度上反映了湖北地区流行毒株的特点,为我国ECoV流行现状和演化趋势提供重要依据。
