[Objective] Bacillus velezensis SH-1471 (CCTCC No. M 2022923, Patent No. ZL 2022 1 1479280.X) is a strain that can control soil-borne diseases of crops and promote soil nutrient conversion and crop growth. This study aims to explore its potential biological activity and the optimum fermentation conditions, so as to promote the industrialization and commercial development of this strain.[Methods] A strain SH-1471 was identified based on the morphological, physiological, and biochemical characteristics and the 16S rRNA gene andgyrB-based phylogenetic trees. PCR was carried out to detect antibiotic synthesis genes in the strain. The inhibitory spectrum of the strain was measured by the plate confrontation assay and the fermentation liquid inhibition test. The abilities of the strain to produce enzymes, solubilize phosphorus and potassium, fix nitrogen, and secrete siderophoresin vitro were measured. The fermentation conditions were optimized by single factor tests and response surface methodology withOD600 value and inhibition rate as indicators. The growth-promoting effect of the fermentation liquid on tomato plants and the control effect of the fermentation liquid on tomatoFusarium wilt before and after optimization were determined by indoor pot experiments.[Results] Strain SH-1471 was identified asB.velezensis, carrying the antibiotic synthesis genessrfA,fenB,ituA,ituD, andbymA. It had strong antagonistic effects on 8 pathogenic microorganisms such asFusarium oxysporum,Alternaria alternate, andExserohilum turcicum. Moreover, the strain was capable of producing protease and cellulase, solubilizing phosphorus, fixing nitrogen, and secreting siderophores. The optimal medium formula for the fermentation of SH-1471 was composed of 17.92 g/L sucrose, 16.95 g/L soybean powder, 2.88 g/L magnesium sulfate, and 5.0 g/L yeast extract. The optimum fermentation conditions were pH 7.5, 33 ℃, 220 r/min, and 20–24 h. After optimization, the inhibition rate,OD600, and potted control efficiency of the fermentation liquid reached 94.08% (increasing by 15.6%), 3.28 (increasing by 36.7%), and 93.8%, respectively. Moreover, strain SH-1471 significantly improved plant height, stem circumference, root length, root weight, and fresh weight and dry weight of the aboveground part of tomato seedlings.[Conclusion] B.velezensis SH-1471 carries rich genes for antibiotic synthesis, has the abilities of producing protease and cellulase, solubilizing phosphorus, fixing nitrogen, and secreting siderophores, and demonstrates strong inhibitory effects on a variety of pathogens. It can significantly reduce the incidence of tomatoFusarium wilt and improve the agronomic traits of tomato seedlings. Therefore, the strain has broad application prospects in the biocontrol of plant diseases and the promotion of plant growth.

【目的】贝莱斯芽孢杆菌(Bacillus velezensis) SH-1471是一株兼具防控作物土传病害、促进土壤养分转化以及促进作物生长等功能的菌株(保藏编号:CCTCC No. M 2022923,专利号:ZL 2022 1 1479280.X)。挖掘其潜在的生物活性并探究其最适发酵条件,是推进该菌株产业化和商业化开发的有效手段之一。【方法】结合形态学、分子生物学、16S rRNA基因和gyrB基因对菌株SH-1471进行分类地位鉴定;利用PCR技术,对菌株抗生素合成基因检测;使用平板对峙试验和发酵液抑菌试验测定菌株抑菌广谱性;并测定其体外产酶、解磷、解钾、固氮及产铁载体能力;以菌株发酵液OD600值和抑菌率为指标,通过设计单因素试验和响应面优化试验,探究菌株的最佳发酵配方和最佳发酵条件;采用室内盆栽试验测定优化前后菌株发酵液对番茄植株的促生效果及其对番茄枯萎病的防治效果。【结果】经鉴定,SH-1471为贝莱斯芽孢杆菌(Bacillus velezensis),具有srfAfenBituAituDbymA等抗生素合成基因,对番茄枯萎病菌(Fusarium oxysporum)、烟草赤星病菌(Alternaria alternate)和玉米大斑病菌(Exserohilum turcicum)等8种病原微生物具有良好的拮抗效果;且具有产蛋白酶、纤维素酶、解磷、固氮和分泌铁载体的能力。菌株SH-1471最适培养配方为:蔗糖17.92 g/L、豆粉16.95 g/L、硫酸镁2.88 g/L、酵母膏5.0 g/L;最适发酵条件为pH 7.5、温度33 ℃、转速220 r/min、发酵时间20‒24 h;优化后,抑菌率提升至94.08%,提高15.6%,发酵液OD600提升至3.28,提高36.7%,盆栽防效提升至93.8%;此外,菌株SH-1471可显著提高番茄幼苗的株高、茎围、根长、根重、地上部位鲜重和地上部位干重等农艺性状。【结论】贝莱斯芽孢杆菌SH-1471具有丰富的抗生素合成基因,同时具有产蛋白酶、纤维素酶、解磷、固氮和分泌铁载体等生物活性,对多种病原物具有良好的抑制效果,可显著降低番茄枯萎病的发病率,并对番茄幼苗农艺性状具有显著促进作用,在植物病害生物防治方面以及促进植物生长方面具有广阔的应用前景。

【目的】贝莱斯芽孢杆菌(Bacillus velezensis) SH-1471是一株兼具防控作物土传病害、促进土壤养分转化以及促进作物生长等功能的菌株(保藏编号:CCTCC No. M 2022923,专利号:ZL 2022 1 1479280.X)。挖掘其潜在的生物活性并探究其最适发酵条件,是推进该菌株产业化和商业化开发的有效手段之一。【方法】结合形态学、分子生物学、16S rRNA基因和gyrB基因对菌株SH-1471进行分类地位鉴定;利用PCR技术,对菌株抗生素合成基因检测;使用平板对峙试验和发酵液抑菌试验测定菌株抑菌广谱性;并测定其体外产酶、解磷、解钾、固氮及产铁载体能力;以菌株发酵液OD600值和抑菌率为指标,通过设计单因素试验和响应面优化试验,探究菌株的最佳发酵配方和最佳发酵条件;采用室内盆栽试验测定优化前后菌株发酵液对番茄植株的促生效果及其对番茄枯萎病的防治效果。【结果】经鉴定,SH-1471为贝莱斯芽孢杆菌(Bacillus velezensis),具有srfAfenBituAituDbymA等抗生素合成基因,对番茄枯萎病菌(Fusarium oxysporum)、烟草赤星病菌(Alternaria alternate)和玉米大斑病菌(Exserohilum turcicum)等8种病原微生物具有良好的拮抗效果;且具有产蛋白酶、纤维素酶、解磷、固氮和分泌铁载体的能力。菌株SH-1471最适培养配方为:蔗糖17.92 g/L、豆粉16.95 g/L、硫酸镁2.88 g/L、酵母膏5.0 g/L;最适发酵条件为pH 7.5、温度33 ℃、转速220 r/min、发酵时间20‒24 h;优化后,抑菌率提升至94.08%,提高15.6%,发酵液OD600提升至3.28,提高36.7%,盆栽防效提升至93.8%;此外,菌株SH-1471可显著提高番茄幼苗的株高、茎围、根长、根重、地上部位鲜重和地上部位干重等农艺性状。【结论】贝莱斯芽孢杆菌SH-1471具有丰富的抗生素合成基因,同时具有产蛋白酶、纤维素酶、解磷、固氮和分泌铁载体等生物活性,对多种病原物具有良好的抑制效果,可显著降低番茄枯萎病的发病率,并对番茄幼苗农艺性状具有显著促进作用,在植物病害生物防治方面以及促进植物生长方面具有广阔的应用前景。

[Objective] Bacillus velezensis SH-1471 (CCTCC No. M 2022923, Patent No. ZL 2022 1 1479280.X) is a strain that can control soil-borne diseases of crops and promote soil nutrient conversion and crop growth. This study aims to explore its potential biological activity and the optimum fermentation conditions, so as to promote the industrialization and commercial development of this strain.[Methods] A strain SH-1471 was identified based on the morphological, physiological, and biochemical characteristics and the 16S rRNA gene andgyrB-based phylogenetic trees. PCR was carried out to detect antibiotic synthesis genes in the strain. The inhibitory spectrum of the strain was measured by the plate confrontation assay and the fermentation liquid inhibition test. The abilities of the strain to produce enzymes, solubilize phosphorus and potassium, fix nitrogen, and secrete siderophoresin vitro were measured. The fermentation conditions were optimized by single factor tests and response surface methodology withOD600 value and inhibition rate as indicators. The growth-promoting effect of the fermentation liquid on tomato plants and the control effect of the fermentation liquid on tomatoFusarium wilt before and after optimization were determined by indoor pot experiments.[Results] Strain SH-1471 was identified asB.velezensis, carrying the antibiotic synthesis genessrfA,fenB,ituA,ituD, andbymA. It had strong antagonistic effects on 8 pathogenic microorganisms such asFusarium oxysporum,Alternaria alternate, andExserohilum turcicum. Moreover, the strain was capable of producing protease and cellulase, solubilizing phosphorus, fixing nitrogen, and secreting siderophores. The optimal medium formula for the fermentation of SH-1471 was composed of 17.92 g/L sucrose, 16.95 g/L soybean powder, 2.88 g/L magnesium sulfate, and 5.0 g/L yeast extract. The optimum fermentation conditions were pH 7.5, 33 ℃, 220 r/min, and 20–24 h. After optimization, the inhibition rate,OD600, and potted control efficiency of the fermentation liquid reached 94.08% (increasing by 15.6%), 3.28 (increasing by 36.7%), and 93.8%, respectively. Moreover, strain SH-1471 significantly improved plant height, stem circumference, root length, root weight, and fresh weight and dry weight of the aboveground part of tomato seedlings.[Conclusion] B.velezensis SH-1471 carries rich genes for antibiotic synthesis, has the abilities of producing protease and cellulase, solubilizing phosphorus, fixing nitrogen, and secreting siderophores, and demonstrates strong inhibitory effects on a variety of pathogens. It can significantly reduce the incidence of tomatoFusarium wilt and improve the agronomic traits of tomato seedlings. Therefore, the strain has broad application prospects in the biocontrol of plant diseases and the promotion of plant growth.

近年来贝莱斯芽孢杆菌(Bacillus velezensis)在农业病虫害防控方面受到广大学者的关注,但部分研究主要以不同菌株资源的分离鉴定以及抑菌活性分析为目的,缺乏对菌株系统的研究,总体呈现优良的菌株资源匮乏,且菌株的发展前景受功能单一、活性较低以及定殖能力偏弱等条件的限制[1]。此外,微生物制剂对病害的防治效果受活菌数和抑菌活性物质含量的影响,而各异的生理特性和生境条件偏好性往往决定菌株的最适生长条件不同[2-3]。因此,靶向已有高活性菌株,多方位挖掘其生物活性,并探究其最适生长条件,不仅可为植物病害的生物防控提供优良的菌株资源,而且可促进菌株的规模化和商业化应用。
贝莱斯芽孢杆菌是革兰氏阳性好氧细菌,菌体呈杆状,能形成芽孢,广泛分布于自然界的水体、土壤、空气、植物根系、植株表面和动物肠道等[4]。近年来,国内外有关贝莱斯芽孢杆菌的报道越来越多,研究主要集中在促进植物生长、拮抗病原菌、诱导植物强化细胞壁、积累防御酶和产生抗菌物质等防御反应等方面[5]。研究表明,贝莱斯芽孢杆菌对番茄早疫病原菌(Alternaria solani)和马铃薯疮痂病原菌(Streptomyces galilaeus)具有良好的拮抗活性[6]。菌株JS25R可产生挥发性物质(如禾烷酮、2-壬酮和2-壬醇等)抑制谷镰刀菌的菌体生长和孢子萌发,从而降低小麦赤霉病的发病率[7]。菌株YB15可通过分泌β-葡聚糖酶等抑菌活性物质,从而对病原真菌起到拮抗作用[8]。综上所述,贝莱斯芽孢杆菌在防治植物病害方面具有极大的潜力,但部分菌株功能单一,同时也缺乏对菌株多样化的功能的进行开发利用。此外,菌株的抑菌活性通常与菌株生物量、活性物质产量以及菌株定殖能力的强弱相关[9]。发酵条件优化旨在营造一个菌株生长的适宜环境,促进菌株生长,提高菌株发酵液抑菌活性物质的含量,从而提高菌株的防治效果[10]。不同菌株具有不同的生理特性和生境条件偏好性,最适发酵温度、发酵时间、转速以及碳源、氮源和无机盐均有差异,如解淀粉芽孢杆菌L009在温度28 ℃时,发酵72 h发酵液含菌量最大[11]。解淀粉芽孢杆菌GM-1-2的最适发酵温度为28 ℃,且发酵时间仅为30 h[12]。暹罗芽孢杆菌的最适碳源为乳糖,最适氮源为酵母提取物[13],而贝莱斯芽孢杆菌HC-01的最适碳源为麦芽糖,最适氮源为蛋白胨[14]。因此,针对不同菌株探索其适宜发酵条件,是将功能菌株应用到生产、充分发挥其生防功能的关键步骤。
优良的菌种资源匮乏是制约植物病害生物防控的发展限制性因素之一,加强优良菌株的筛选和改良及以现有菌株为靶标,深入挖掘和评价菌株多样化的生物活性,优化其生产工艺是打破该壁垒的有效措施[4]。本研究以现有高活性贝莱斯芽孢杆菌SH-1471为研究对象,该菌分离自健康烟草根际土壤,本身具有极强的定殖能力,基于PCR技术检测菌株抗生素合成基因类别,并利用平板对峙法以及底物降解法等活性筛选模型,挖掘菌株对病原微生物的生防活性,溶磷、解钾和固氮等促生长活性以及产铁载体、蛋白酶和纤维素酶等活性,并进一步对其最适发酵条件进行优化,旨在为农用微生物制剂的发展提供高效且功能多样化的菌株资源,且为该菌株的开发以及推广利用奠定基础。
功能菌株SH-1471由云南省农业科学院农业环境资源研究所于健康烟草根际土壤分离与保存,并于2022年6月20日保藏于中国典型培养物保藏中心(保藏编号:CCTCC No. M 2022923)。
文中涉及的指示病原物番茄枯萎病菌(Fusarium oxysporum)、烟草赤星病菌(Alternaria alternate)、玉米大斑病菌(Exserohilum turcicum)、草果茎点霉属叶斑病菌(Phoma matteuciicola)、草果枝枯病菌(Diaporthe eres)、烟草炭疽病菌(Colletotrichum micotianae)、烟草黑胫病菌(Phytophthora parasitica)和油菜菌核病菌(Sclerotinia sclerotiorum)由云南省农业科学院农业环境资源研究所分离、鉴定与保存。
功能筛选培养基:解无机磷选择培养基,解钾选择培养基,固氮选择培养基,营养琼脂(nutrient agar, NA)培养基,营养肉汤(nutrient broth, NB)培养基,马铃薯葡萄糖水培养基(potato dextrose broth, PDB),马铃薯葡萄糖琼脂(potato dextrose agar, PDA)培养基,脱脂牛奶培养基,纤维素培养基,果胶培养基,铬天青(chrome azurol sulphonate, CAS)琼脂双层培养基[15]
优化初始培养基:培养基A (g/L):葡萄糖5.00,鱼粉10.00,氯化钠5.00、酵母膏5.00。酵母蛋白胨蔗糖培养基(yeast sucrose peptone medium, YSP) (g/L):蛋白胨10.00,酵母浸粉5.00,蔗糖20.00。培养基D (g/L):玉米淀粉3.00,硫酸铵10.00,磷酸二氢钾15.00,蔗糖10.00。牛肉膏酵母葡萄糖培养基(nutrient yeast beef paste dextrose medium, NYBD) (g/L):牛肉膏8.00,酵母浸粉5.00,葡萄糖10.00。营养琼脂培养基(NA) (g/L):牛肉膏3.00,蛋白胨10.00,氯化钠5.00。细菌基础培养基(LB) (g/L):蛋白胨10.00,酵母浸粉5.00,氯化钠10.00。121 ℃灭菌30 min。
抗生素合成基因PCR检测,参照濮永瑜等[16]的试验方法进行。抑菌广谱性测定:(1) 平板对峙试验:以上述10种病原真菌为指示病原物,在PDA培养基中心接种5 mm的病原菌菌饼,十字交叉在距离其25 mm处接种菌株,3个重复。(2) 发酵液抑菌试验:将菌株SH-1471接种于NB培养基,30 ℃、180 r/min培养24 h,4 ℃、8 000 r/min冷冻离心5 min,使用0.22 μL无菌滤膜滤除菌体,得无菌发酵液,将其与PDA以1:4的比例混合均匀,制成带毒平板后取5 mm的病原菌菌饼接种于平板中央,3个重复。以未接菌株SH-1471为对照,置于25‒28 ℃的恒温培养箱黑暗条件培养5–7 d,记录并计算抑制率。
抑制率(%)=[对照组菌落直径(cm)‒处理组菌落直径(cm)/对照组菌落直径(cm)‒0.5]×100。
功能测定:将菌株分别点接到脱脂牛奶培养基、纤维素培养基、果胶培养基、解钾选择培养基和固氮选择培养基上,37 ℃培养48 h,若有透明圈生成则具有相应活性,使用CAS双层平板检测菌株产铁载体能力[17]
种子液制备:挑取斜面保存的菌株SH-1471菌落接种于100 mL NB液体培养基中,于28 ℃、180 r/min振荡培养24 h后备用。
分别制备培养基A、培养基D、YSP、NYBD、NB和LB液体培养基各100 mL,以1%的接种量接种SH-1471种子液,于28 ℃、180 r/min振荡培养24 h,3个重复,测量菌株发酵液的OD600值。将菌株发酵液8 000 r/min离心2 min,并使用0.22 μm的无菌滤膜滤除菌体,采用发酵液抑菌试验,验证菌株发酵液对番茄枯萎病菌的抑制效果,结合菌株发酵液OD600值与发酵液对病原菌的抑制率筛选出最适培养基种类。
以1.2.2筛选出的最佳发酵培养基靶标,设置单因素试验,分别加入碳源:麦芽糖(maltose)、乳糖(lactose)、葡萄糖(glucose)、蔗糖(saccharose)和淀粉(starch);氮源:氯化铵(NH4Cl)、酵母浸粉(yeast extract)、硝酸铵(NH4NO3)、甘氨酸(glycine)、蛋白胨(peptone)、硝酸钠(NaNO3)和豆粉(bean flour);无机盐:碳酸钾(K2CO3)、磷酸氢二钾(K2HPO4)、碳酸钙(CaCO3)、氯化钠(NaCl)、磷酸二氢钠(NaH2PO4)和硫酸镁(MgSO4)。于28 ℃、180 r/min振荡培养24 h后,测量菌株发酵液OD600值和发酵液抑制率(同1.2.2),以确定最佳碳源。
分别配制上述优化的培养基100 mL,以pH:4.5、5.5、6.5、7.5和8.5,温度:21、24、27、30、33、36和39 ℃,转速:140、160、180、200和220 r/min为指标,设置单因素试验,以1%的接种量接种SH-1471种子液,振荡培养24 h后,测定不同培养条件下细菌生长情况(OD600),以及发酵液对番茄枯萎病菌的抑制率(同1.2.2),以确定最佳初始pH、温度和转速。
基于上述试验所得的最适配方条件,以菌株发酵液OD600为指标,选取蔗糖浓度(5、10、15、20和25 g/L)、豆粉浓度(5、10、15、20和25 g/L)和MgSO4浓度(0.5、1.0、1.5、2.0和2.5 g/L)进行优化,使用Design-Expert 13软件进行试验设计与分析,以进一步得到适合菌株SH-1471的最适发酵条件。
基于单因素试验和响应面优化结果,以菌株发酵液的OD600值为指标,测定不同发酵时间对菌株发酵液生物量的影响,在0‒24 h内每隔2 h测量一次,24 h后每隔12 h测量一次,直至120 h,并根据测定结果绘制菌株生长曲线。
病原菌经PDB培养基28 ℃、180 r/min培养5‒7 d后,稀释成1.5×107 CFU/mL孢子悬浮液;将菌株SH-1471用优化后的培养条件和优化前的培养条件,分别培养20‒24 h,稀释成2×108 CFU/mL菌悬液。采用灌根法接种菌液,用玻璃棒在番茄苗根茎约3 cm处扎孔,深约5 cm,每株100 mL病原菌悬液,待病原菌定殖3 d后接入等量的功能菌株菌悬液,每个处理10株,3个重复。以CK1 (灭菌培养基)与CK2 (病原菌+灭菌培养基)为对照,30 d后观察并记录发病情况,同时测量番茄植株株高、茎围、根长、根重和地上部鲜重和地上部干重。
将盆栽在恒温条件下培养30 d,观察并记录植株生长和发病情况。各处理发病情况按照番茄枯萎病病情分级标准[18],0:无症状;1:1片或2片叶子变黄;2:3片或者4片真叶变黄叶片萎蔫下垂;3:5片或6片真叶变黄或真叶萎蔫下垂;4:全株严重萎蔫以致枯死。

参照《伯杰氏系统分类手册》[19]和《常见细菌系统鉴定手册》[20]中的方法对菌株SH-1471进行形态学鉴定和生理生化特性检测。
将菌株SH-1471接种至NB培养基中,33 ℃、220 r/min振荡培养,当菌株处于对数生长期时结束培养。采用TaKaRa MiniBEST Bacteria Genomic DNA Extraction Kit Ver.3.0试剂盒提取菌株SH-1471的基因组DNA,采用16S rRNA基因序列27F (5′-AGAGTTTGATCC TGGCTCAG-3′)、1492R (5′-TACGGYTACCTT GTTACGACTT-3′)和gyrB基因序列gyrbBF2 (5′-GGGGTCTACTGCTTCACCAA-3′)、gyrbBR2 (5′-TTGTCCGGGTTGTACTCGTC-3′)对菌株SH-1471进行PCR扩增。PCR反应体系(25 μL):细菌DNA模板2 μL,2×Taq PCR Master Mix 12.5 μL,上、下游引物(10 μmol/L)各1 μL,ddH2O 8.5 μL。PCR反应条件:94 ℃ 5 min;94 ℃ 60 s,53 ℃ 60 s,72 ℃ 2 min,35个循环;72 ℃ 7 min,4 ℃保存。扩增产物经琼脂糖凝胶电泳纯化回收后,委托北京擎科生物科技有限公司测序。测序结果经BLAST搜索(https://blast.ncbi.nlm.nih.gov/Blast.cgi)后与GenBank数据库中相关种属的基因序列进行比较分析,选用同源性较高的模式菌株序列作为参比对象,用ClustalX 1.8软件进行多序列比对,计算供试菌株与参比菌株序列的相似性。系统发育分析时排除碱基缺失位点,采用邻接法(neighbor-joining analysis)用MEGA 7.0构建供试菌株与参比菌株之间的系统发育树。其中,bootstrap值设定为1 000,其余均为默认值。
数据采用单因素方差分析,应用Duncan’s新复极差法检验处理间差异显著性。选择Excel和SPSS Statistics 20.0软件进行数据统计分析以及正交试验设计,并采用软件Origin 2021和软件GraphPad Prism 8制图。
对菌株进行PCR检测结果表明(图1),菌株SH-1471具有产srfA (1 300 bp)、fenB (1 600 bp)、ituA (1 047 bp)、ituD (647 bp)和bymA等抗生素合成基因,平板对峙试验以及发酵液抑菌试验结果表明,菌株SH-1471对番茄枯萎病菌、烟草赤星病菌和玉米大斑病菌等8种病原微生物具有良好的抑制效果(图2),且对番茄枯萎病菌的抑制率最高,为96.59%。功能筛选结果可知,菌株SH-1471还具有产蛋白酶、纤维素酶、解磷、固氮和分泌铁载体的能力(图3)。综上所述,该菌株不仅在作物病害生物防治方面具有极大的应用前景,在促进作物生长提供土壤肥力等方面均具有广阔的应用前景。
七种培养基对菌株SH-1471的生长的影响具有显著差异(图4A),培养24 h后,SH-1471在培养基A、LB、NYBD、YSP和NA中的OD600值分别为2.56、2.45、2.43、2.41和2.31,其中培养基A的OD600值显著高于其他配方;而培养基D和培养基YPG对菌株的生长促进作用最差,OD600分别为1.46和1.98。且培养基A条件下,对病原物的抑制率最高,达82.4%,故选择培养基A作为菌株SH-1471的基础培养基。
图4B可知,当添加蔗糖和葡萄糖为碳源时,菌株发酵液的OD600值最高,分别为2.62和2.44,以麦芽糖为碳源时,OD600仅为1.32;此外,以蔗糖为碳源时,菌株发酵液对番茄枯萎病菌的抑制率最高,达84.6%,综合发酵液OD600值与发酵液抑菌活性,以蔗糖作为菌株SH-1471的最佳碳源。对不同氮源筛选结果如图4C所示,当以豆粉为氮源时,菌株OD600最高,达2.65,其次为酵母浸粉,为2.49,而氯化铵最低,仅为0.76,而以豆粉作为氮源时,菌株发酵液对病原物的抑制率为84.9%。综合考虑,以豆粉作为菌株SH-1471的最佳氮源。如图4D所示,当以硫酸镁和磷酸氢二钾作为无机盐源时,菌株发酵液OD600值和抑制率均强于其他无机盐源;当以硫酸镁作为无机盐时,菌株发酵液OD600值为2.61,抑制率为85.2%;以磷酸氢二钾为无机盐时,菌株发酵液OD600值为2.45,抑制率为81.2%;而以碳酸钾为无机盐时,菌株发酵液OD600值最低为1.54,抑菌率为73.6%。综上所述,以硫酸镁作为菌株SH-1471的发酵培养基的最佳无机盐。
图5A所示,当pH为4.5和8.5时,发酵液浓度显著低于其他浓度梯度,OD600值仅为2.11和2.22,表明pH值过高与过低均会抑制菌株的生长。pH值为7.5时,发酵液OD600值最高为2.65、发酵液对病原物的抑菌率最高为88.6%。综上所述,菌株SH-1471的最适pH值7.5。由图5B可知,当温度为33 ℃时,菌株发酵液OD600值最高为2.67,抑菌率为86.5%。因此,确定菌株的最适发酵温度为33 ℃。由图5C可知,不同转速对菌株发酵液OD600值和抑菌率具有不同的影响,当转速为220 r/min和200 r/min时发酵液OD600值相似,但转速为220 r/min时,抑菌率较大,故以此为菌株发酵的最适转速。
基于上述配方优化结果,选取蔗糖浓度5–25 g/L、豆粉浓度5–25 g/L和MgSO4浓度0.5–2.5 g/L 3个影响因素,采用Design-Expert 13进行响应面优化试验设计,进行方差分析导出结果(表1),得到菌株发酵液OD600=3.02+ 0.10A+0.097B+0.05C‒0.051AB‒0.054AC+6.500E‒003BC‒0.13A2‒0.14B2‒0.093C2的回归方程。从表可以看出,F值为0.026 4,P < 0.05,达到极显著水平,失拟项差异不显著,相关系数为0.965 4,说明实测值与预测值具有较好的拟合度,试验误差较小。CV值为3.47%,该模型R2Adj相关系数为0.921 0,响应面变化可由该模型解释,且与实际情况拟合较好,可用于试验分析预测。ABACBCA2B2均为差异极显著(P < 0.05),其余项差异不显著。根据F值大小,可以得出试验各因素对菌株OD600值影响的顺序为:蔗糖 > 豆粉 > MgSO4。运用Design-Export 13软件分析得知菌株的最适条件为:蔗糖浓度为17.92 g/L、豆粉浓度为16.95 g/L、MgSO4浓度为2.88 g/L,预测值为3.06,在该条件下进行3次平行试验,试验结果分别为3.17、3.12和3.21,平均值为3.17,与预测值相近,表明优化结果可靠(图6)。
基于单因素试验结果和响应面优化结果,利用所得的最佳发酵配方及发酵条件进行菌株最适发酵时间优化,结果如图7所示,菌株SH-1471发酵液OD600值总体呈“S”型变化,0–8 h时,发酵液OD600值无显著变化,为菌株生长迟缓期,22 h时菌株发酵液OD600值最大,为3.37。因此,8–22 h为菌株的对数生长期,此时菌株生长最快。22–120 h菌株生长缓慢,趋于平稳期。120 h之后,呈下降趋势,为生长的衰亡期。
基于单因素试验结果和响应面优化结果,利用所得的最佳发酵配方及发酵条件进行连续3批发酵实验对比,结果如图8所示,优化后菌株发酵液的OD600达到3.28,相比于优化前提升36.7%;优化后对病原物的抑菌率达到94.08%,相比于优化前提升15.6%。综上所述,发酵优化试验结果可靠,对菌株的生长量及其对病原物的抑制率均具有较强的提高效果。
基于上述优化结果进行室内盆栽试验,30 d后测定各处理下的病情指数、发病率以及番茄幼苗的农艺性状。结果如表2图9所示,番茄幼苗经过接种处理30 d,无论优化前或优化后,菌株均能有效抑制番茄枯萎病的发生,只接病原菌(CK1)的对照处理发病较为严重,病情指数高达79.8。菌株SH-1471优化前的发酵液处理的番茄幼苗病情指数为4.6,防治效果为85.9%;优化后的发酵液处理的番茄幼苗病情指数为2.2,防治效果为93.8%;此外,菌株SH-1471对番茄幼苗的生长还具有一定的促生长作用,与对照相比,经菌株发酵液处理后,番茄幼苗的株高、茎围、根长、根重、地上部位鲜重和干重等农艺性状均得以显著提高。
菌株SH-1471孢子呈杆状,菌落中心呈乳白色,不产色素,菌落形状不规则,边缘呈放射状,菌落表面粘稠,略微凸起(图10)。生理生化鉴定结果表明,菌株革兰氏染色均为阳性,好氧生长,能产生芽孢,接触酶、硝酸还原反应、精氨酸双水解、V-P试验均为阳性,可分解淀粉、使明胶水化、可利用葡萄糖和甘露醇,不可利用柠檬酸盐,最低生长温度为3 ℃,生长含盐量范围0%‒9%,不具有产H2S和分泌吲哚的活性。
基于16S rRNA基因扩增,将菌株序列在NCBI上注册,菌株序列登录号为ON417363,经NCBI BLAST比对分析发现SH-1471分别与Bacillus velesiensis CR-502 (GenBank登录号:AY603658)和Bacillus siamensis KCTC 13613 (GenBank登录号:AJVF01000043)的同源性分别为99.8%和99.3%,利用邻接法(neighbour-joining)构建系统发育树,结果表明,SH-1471与Bacillus velesiensis CR-502在同一分支上(图11A)。基于gyrB基因扩增结果,经NCBI BLAST比对分析发现SH-1471分别与Bacillus velezensis KACC 13105 (GenBank登录号:JTKJ020000141)和Bacillus amyloliquefaciens DSM 7 (GenBank登录号:ATCC 23350)同源性最高,分别为100%和99.2%,且与菌株Bacillus velezensis KACC 13105位于系统发育树的同一分支(图11B),结合形态学鉴定结果以及16S rRNA和gyrB基因检测结果,最后将菌株SH-1471鉴定为贝莱斯芽孢杆菌(并保藏于中国典型培养物保藏中心保藏,保藏编号:CCTCC No. M 2022923)。
近年来,贝莱斯芽孢杆菌在农业上的应用越发广泛。在已有的研究中,具有拮抗作用的贝莱斯芽孢杆菌常从水体、土壤、空气、植物根系、植株表面和动物肠道中分离得到[1]。本文的研究对象贝莱斯芽孢杆菌SH-1471,分离自健康烟草根际土壤,本身对自然环境具有良好的适应性,前人研究表明,贝莱斯芽孢杆菌可通过聚酮合酶合成途径、核糖体途径和非核糖体途径合成脂肽类化合物、聚酮类化合物、细菌素和抑菌蛋白等活性物质抑制病原物生长[21-23]。冯江鹏等[24]研究的贝莱斯芽孢杆菌JK3菌株基因组中含有srfAAbymBfenDbacAituC等脂肽类抗菌物质生物合成相关基因。与之类似,本研究PCR结果表明,菌株SH-1471具有srfAfenBituAituDbymA等基因,可通过合成多种脂肽类抗生素抑制真菌、细菌的生长,并诱导生物膜的形成。且贝莱斯芽孢杆菌SH-1471菌体以及无菌发酵液对番茄枯萎病菌均可起到良好的抑制效果,并可显著提高番茄幼苗的株高、根长以及根重等农艺性状。与陈万权等[25]分离自土壤的贝莱斯芽孢杆菌Jnb16的结果类似,该菌对水稻稻瘟病菌具有良好的抑制效果,且可有效促进水稻植株的生长。贝莱斯芽孢杆菌SH-1471除对番茄枯萎病菌具有良好的抑制效果,还对烟草赤星病菌、烟草炭疽病菌、玉米大斑病菌等病原微生物具有抑制效果。余水等[26]分离的贝莱斯芽孢杆菌MT310亦具有广谱抑菌性,对烟草赤星病菌、黄连根腐病菌和高粱炭疽病菌等植物病原真菌有明显抑制效果。
除生防作用外,贝莱斯芽孢杆菌被广泛应用在工业以及生物医学方面。贝莱斯芽孢杆菌SH-1471还具有产蛋白酶和纤维素酶活性,对难溶性蛋白、植物纤维等具有良好的分解作用,在饲料添加剂、洗涤剂和造纸领域具有良好的应用前景[27]。此外,菌株SH-1471具有解磷和固氮功能,可促进土壤中难溶性化合物的降解,增加土壤中有效磷和可溶性氮的含量,对作物发芽率和作物根系生长具有促进作用,且对提高土壤肥力以及磷肥的利用率具有重要的作用[28]。而菌株SH-1471的铁载体合成能力也是诱导植物系统抗性从而抵御病原菌的侵染的重要途径,可通过分泌铁载体来促进植物根部系统发育和营养物质的摄取来促进植物生长,诱导系统抗病性等。
此外,在菌株生长过程中,往往伴随着多种活性物质和芽孢的产生,二者决定着菌株的生防活性以及菌株的抗逆性,但其通常与发酵条件息息相关,因此对菌株的发酵条件进行优化决定着菌株的生物量大小以及抑菌活性强度[29]。前人研究表明,细菌在营养物质和外界条件都适宜的情况下,产生的抑菌物质往往较为丰富[30],枯草芽孢杆菌MC4-2的最适pH为6.0、转速210 r/min、温度32 ℃[31]。本研究在该条件下,菌株生物量及抑菌活性均得以显著提高。解淀粉芽孢杆菌HF-01的最适无机盐为硫酸镁,最适pH为微酸性至中性,培养温度28 ℃[32],与本研究结果类似。桑树内生甲基营养型芽孢杆菌(Bacillus methylotrophicus) XP-27最适无机盐为硫酸镁,培养温度为30 ℃[33],这与本研究类似,但其最适初始pH值为8.0,而本研究最适pH为7.5,可能是由于菌株长期生长环境的不同而导致的[34]。本研究通过室内摇瓶对菌株培养基配方、发酵条件进行优化,优化结果对菌株的生物量、拮抗活性及盆栽防效等均具有显著的促进作用。此外,平板对峙试验和室内盆栽试验表明,发酵条件优化后菌株SH-1471对番茄枯萎病菌的抑制作用和盆栽防效显著提高至94.08%,盆栽防效提高至93.8%。但缺乏大型的发酵设备以及田间实际防效进行验证,今后还需进行小试和中试,以验证并完善优化结果,进一步获得菌株的最适发酵罐发酵工艺,同时验证其是否还具有促生长和诱导植物抗性等作用,为贝莱斯芽孢杆菌SH-1471的产业化应用提供理论基础。
