[Objective] To isolate and identify a strain of foot-and-mouth disease virus (FMDV) from bovine oropharyngeal fluid (OPF) and subsequently analyze the amino acid mutations in antigenic proteins and the proliferative properties of the isolated virus. [Methods] RT-PCR and sequencing were performed to identify the FMDV-positive OPF samples. The FMDV strain was isolated from bovine OPF samples by cell inoculation and identified by indirect immunofluorescence assay. The whole genome sequence of the strain was obtained by fragment amplification and sequencing. The amino acid sequences were compared to reveal the amino acid mutations in the antigenic proteins of the strain. The proliferative properties of the isolated FMDV strain were analyzed by the plaque forming assay and one-step growth curve. [Results] A FMDV strain O/FMDV/OP/2022/B was successfully isolated from bovine OPF, belonging to serotype O and Ind-2001 lineage. Amino acid mutations were identified in the antigenic proteins VP1, VP2, and VP3 of O/FMDV/OP/2022/B. In addition, the plaque of O/FMDV/OP/2022/B in BHK-21 cells was smaller than that of the vaccine strain O/FMDV/HN/93. The one-step growth curves showed that O/FMDV/OP/2022/B had significantly lower replication titer than O/FMDV/HN/93 in BHK-21 cells. [Conclusion] A strain of FMDV was isolated from bovine OPF, and it displayed amino acid mutations in antigenic proteins and had lower proliferative capacity than the vaccine strain O/FMDV/HN/93. This study contributes to a better understanding of the mechanism of persistent FMDV infection in the bovine oropharynx and provides a reference for the prevention and control of FMD.

【目的】 分离、鉴定一株牛源口咽液中的口蹄疫病毒(foot-and-mouth disease virus, FMDV),分析其抗原蛋白氨基酸变异及增殖特性。 【方法】 采用RT-PCR及基因测序技术鉴定FMDV阳性牛口咽液样本,通过细胞接种从阳性样本中分离病毒,并利用间接免疫荧光法对分离病毒进行鉴定。通过PCR分段扩增及序列测定获得分离FMDV的全基因组,进一步通过序列比对分析分离FMDV的抗原蛋白氨基酸变异,通过蚀斑形成试验和一步生长曲线评价分离FMDV的增殖特性。 【结果】 成功从牛口咽液中分离、鉴定出一株FMDV,命名为O/FMDV/OP/2022/B。该病毒属于O型Ind-2001谱系,其抗原蛋白VP1、VP2和VP3的氨基酸残基发生变异。在BHK-21细胞上,O/FMDV/OP/2022/B毒株形成的蚀斑显著小于疫苗毒株O/FMDV/HN/93的蚀斑,且其一步生长曲线明显低于O/FMDV/HN/93。 【结论】 本研究从牛口咽液中分离出一株FMDV,发现其抗原蛋白氨基酸发生变异,且增殖能力显著低于疫苗毒株O/FMDV/HN/93。该研究有助于从病毒学角度深入理解FMDV持续感染牛口咽组织的机制,并为口蹄疫的防控提供了参考依据。

【目的】 分离、鉴定一株牛源口咽液中的口蹄疫病毒(foot-and-mouth disease virus, FMDV),分析其抗原蛋白氨基酸变异及增殖特性。 【方法】 采用RT-PCR及基因测序技术鉴定FMDV阳性牛口咽液样本,通过细胞接种从阳性样本中分离病毒,并利用间接免疫荧光法对分离病毒进行鉴定。通过PCR分段扩增及序列测定获得分离FMDV的全基因组,进一步通过序列比对分析分离FMDV的抗原蛋白氨基酸变异,通过蚀斑形成试验和一步生长曲线评价分离FMDV的增殖特性。 【结果】 成功从牛口咽液中分离、鉴定出一株FMDV,命名为O/FMDV/OP/2022/B。该病毒属于O型Ind-2001谱系,其抗原蛋白VP1、VP2和VP3的氨基酸残基发生变异。在BHK-21细胞上,O/FMDV/OP/2022/B毒株形成的蚀斑显著小于疫苗毒株O/FMDV/HN/93的蚀斑,且其一步生长曲线明显低于O/FMDV/HN/93。 【结论】 本研究从牛口咽液中分离出一株FMDV,发现其抗原蛋白氨基酸发生变异,且增殖能力显著低于疫苗毒株O/FMDV/HN/93。该研究有助于从病毒学角度深入理解FMDV持续感染牛口咽组织的机制,并为口蹄疫的防控提供了参考依据。

[Objective] To isolate and identify a strain of foot-and-mouth disease virus (FMDV) from bovine oropharyngeal fluid (OPF) and subsequently analyze the amino acid mutations in antigenic proteins and the proliferative properties of the isolated virus. [Methods] RT-PCR and sequencing were performed to identify the FMDV-positive OPF samples. The FMDV strain was isolated from bovine OPF samples by cell inoculation and identified by indirect immunofluorescence assay. The whole genome sequence of the strain was obtained by fragment amplification and sequencing. The amino acid sequences were compared to reveal the amino acid mutations in the antigenic proteins of the strain. The proliferative properties of the isolated FMDV strain were analyzed by the plaque forming assay and one-step growth curve. [Results] A FMDV strain O/FMDV/OP/2022/B was successfully isolated from bovine OPF, belonging to serotype O and Ind-2001 lineage. Amino acid mutations were identified in the antigenic proteins VP1, VP2, and VP3 of O/FMDV/OP/2022/B. In addition, the plaque of O/FMDV/OP/2022/B in BHK-21 cells was smaller than that of the vaccine strain O/FMDV/HN/93. The one-step growth curves showed that O/FMDV/OP/2022/B had significantly lower replication titer than O/FMDV/HN/93 in BHK-21 cells. [Conclusion] A strain of FMDV was isolated from bovine OPF, and it displayed amino acid mutations in antigenic proteins and had lower proliferative capacity than the vaccine strain O/FMDV/HN/93. This study contributes to a better understanding of the mechanism of persistent FMDV infection in the bovine oropharynx and provides a reference for the prevention and control of FMD.

口蹄疫(foot-and-mouth disease, FMD)作为一种传染性极强的重大动物疫病,主要感染猪、牛和羊等偶蹄类动物,其历史可以追溯到1 546年。Frosch等发现引起FMD的病原物质比细菌更小,随后的研究证实这种传染因子为口蹄疫病毒(foot-and-mouth disease virus, FMDV)。FMDV属于小RNA病毒科(Picornaviridae),口疮病毒属(Aphthovirus),基因组全长8 500 nt左右,编码的结构蛋白为VP1、VP2、VP3和VP4,编码的非结构蛋白为L、2A、2B、2C、3A、3B、3C和3D。FMDV有7种血清型,每种血清型又分为多种拓扑型和谱系。近年来,我国主要流行O型和A型FMDV,其中A型FMDV的报道越来越少,目前流行的FMDV多为O型。O型FMDV流行毒株复杂,流行的谱系包括O/CATHAY、O/Mya-98、O/Ind-2001和O/PanAsia,这种多谱系FMDV的共同流行加剧了病毒变异,使病毒逐渐进化出免疫逃逸能力,严重威胁我国FMD的防控工作[1]
在自然感染中,FMDV主要通过呼吸道感染易感动物,极少量的感染性病毒颗粒就足以引发FMD[2]。牛被认为是最容易通过空气传播途径感染FMDV的动物,仅需10 TCID50即可感染FMDV[3-4]。因此,FMD流行时牛的症状最明显,可作为疫情的“指示器”。FMDV的原发性及持续性感染部位定位于牛的鼻咽和软腭背侧的上皮细胞。在感染后的24-48 h,FMDV从感染部位进入血液,随后扩散到全身不同组织和细胞中进行复制,同时机体出现发热[5],可观察到明显的FMD临床症状,患病动物的分泌物、排泄物及组织中可检测到FMDV的存在[6]。有一定比例的牛在急性感染FMDV痊愈后无法完全清除体内的病毒,发展为FMDV携带者或FMDV持续性感染者,在口咽部周期性分泌低水平的FMDV,而无任何临床症状。1959年,Van Bekkum等[7]提出FMDV持续性感染的概念,即感染FMDV 28 d后仍然能够在无临床症状动物的口咽液(oropharyngeal fluid, OPF)中检测到FMDV的存在。综上所述,牛的OPF中只有在急性感染FMDV后或者成为无症状的FMDV持续性感染者时,才会携带FMDV。由于从无症状动物的OPF中分离出FMDV较为困难,致使国内关于此种FMDV分离和特性研究的报道屈指可数。
无论是否接种FMD疫苗,FMDV在OPF中持续存在的发生率约为50%[8],这意味着疫苗免疫不能清除OPF中持续存在的FMDV。监测显示,非洲水牛能够将OPF中周期性分泌和脱落的FMDV传播给同居的牛和黑斑羚。将FMDV携带动物的OPF注射到牛和猪中,能够引起牛和猪感染FMDV[9],表明OPF中持续存在的FMDV具有感染性,可以作为一种引发FMD流行的潜在传染源。OPF中持续存在的FMDV不仅会增加FMD在种群中的传播风险,而且这些持续携带FMDV的动物可能成为中间宿主,促使病毒在不同物种之间传播。此外,口咽部位持续存在的FMDV面临宿主免疫系统较大的选择压力,其基因组易发生变异,可能使疫苗失去对变异株的有效保护,加大FMD的防控难度[10]。近期研究发现,OPF中持续存在的FMDV与急性感染FMDV可以发生重组,这可能更有利于FMDV的传播[11]。因此,OPF中持续存在的FMDV可能成为自然界中的重要传染源,是FMD难以彻底消灭的根源。研究OPF中持续存在的FMDV对于FMD的防控和清除具有重要的现实意义。本研究从田间收集的无症状牛OPF样品中分离出一株FMDV,测定了该毒株的全基因组序列,比对了抗原蛋白氨基酸变异,研究了其增殖特性,以期为从病毒角度深入研究FMDV持续感染牛口咽组织的机理提供材料和参考。
FMDV O/FMDV/HN/93由中国农业科学院兰州兽医研究所国家口蹄疫参考实验室保存。涉及FMDV活病毒的操作均在中国农业科学院兰州兽医研究所P3生物安全实验室进行。BHK-21细胞由中国农业科学院兰州兽医研究所宿主抗病毒感染及免疫生物学团队保存。
MEM培养基、2×MEM培养基、胎牛血清(fetal bovine serum, FBS)、0.25% EDTA的胰酶溶液均购自Gibco公司;磷酸盐缓冲液(phosphate buffered saline, PBS)购自白鲨生物科技有限公司;DNA marker购自宝日医生物技术(北京)有限公司;RNA Extraction Kit购自Omega公司;FITC标记的山羊抗鼠二抗和HRP标记的山羊抗兔/鼠二抗均购自武汉赛维尔生物科技有限公司;ECL超敏显色液购自上海碧云天生物技术股份有限公司;抗FMDV 3A蛋白鼠源单克隆抗体(3A24)由本团队制备保存[12-13]
根据GenBank已报道的FMDV基因组全长序列(GenBank登录号为MN095354.1、KJ825809.1、MN095364.1、OM562600.1、OK318520.1、KY444646.1、MT944983.1、KJ825803.1),设计扩增FMDV全基因组序列引物和VP1基因引物(表1),由北京擎科生物科技股份有限公司合成。
2022年国家口蹄疫参考实验室在主动监测中从某地无FMD临床症状的牛中采集到6份牛OPF。将牛OPF样品在12 000 r/min的条件下离心10 min,上清液转移到新的EP管中,利用RNA Extraction Kit提取样品中的总RNA。以提取的总RNA为模板,通过RT-PCR扩增FMDV VP1基因。将凝胶电泳正确的PCR产物送至北京擎科生物科技股份有限公司进行序列测定。由于我国流行O型、A型和Asia1型FMDV,从GenBank下载3种血清型FMDV的VP1基因序列,与测定的序列通过MEGA 11软件进行比对分析,鉴定测得的基因是否为FMDV VP1基因以及属于何种谱系。
使用0.22 μm滤器过滤FMDV阳性OPF,与1/4体积的三氯三氟乙烷(trichlorotrifluoroethane, TTE)混合,振荡均匀后3 000 r/min离心10 min,取上清液接种至生长状况良好的BHK-21细胞,显微镜下观察BHK-21细胞病变(cytopathic effect, CPE)情况。若细胞出现变圆、变大且呈倒挂葡萄串状,判定为细胞病变。收取病变样品,在BHK-21细胞中连续传代,直至CPE时间稳定,将传代样品贮存于‒80 ℃冰箱备用。
利用间接免疫荧光试验(immunofluorescence assay, IFA)鉴定收取的BHK-21细胞病变样品是否为FMDV。将生长状况良好的BHK-21细胞铺于6孔板中,当细胞密度达到80%左右时,加入1/10体积的病毒液。将6孔板置于37 ℃细胞培养箱中,孵育6 h后弃去细胞上清液,用PBS漂洗3次,加入4%多聚甲醛,室温固定15 min。弃去固定液后,加入冰冷的甲醇溶液,通透10 min,再用PBS漂洗3次。加入鼠源3A24单克隆抗体(1:200稀释),37 ℃孵育1 h,用PBS漂洗3次。加入FITC标记的山羊抗小鼠IgG二抗(1:200稀释),37 ℃孵育1 h,用PBS漂洗3次后,在荧光显微镜下观察绿色荧光情况并拍照。
按照RNA Extraction Kit操作说明书提取第7代传代样品的总RNA。基于设计的6对引物,采用RT-PCR方法分段扩增牛源OPF分离FMDV的全基因组。将PCR产物送至北京擎科生物科技股份有限公司测序。从NCBI上检索并下载Ind-2001谱系FMDV的氨基酸序列,比对分析O/FMDV/OP/2022/B抗原蛋白氨基酸变异情况。
以疫苗毒株O/FMDV/HN/93为阳性对照,通过蚀斑形成试验分析分离FMDV的蚀斑表型。将生长状况良好的BHK-21细胞接种至6孔板中,待细胞长满后,用无血清MEM培养基以10倍梯度连续稀释O/FMDV/HN/93和分离的FMDV。向6孔板的每孔中分别加入200 μL不同稀释度的FMDV,置于37 ℃细胞培养箱中孵育1 h。加入2 mL黄芪胶混合液(2×MEM:1.2%黄芪胶=1:1,1% FBS,1%青链霉素),放入37 ℃细胞培养箱中培养48 h。弃去培养液后,加入2 mL预冷的固定液(50%丙酮,50%甲醇),置于-20 ℃固定1 h。弃去固定液,加入结晶紫溶液,室温染色过夜,清水冲洗后观察蚀斑表型,统计蚀斑数量,并计算2株FMDV的蚀斑形成单位(plaque forming unit, PFU)。
以疫苗毒株O/FMDV/HN/93为阳性对照,通过一步生长曲线分析分离FMDV的复制能力。将BHK-21细胞接种至12孔板中,待细胞长满后,以1 MOI (multiplicity of infection)的剂量分别接种O/FMDV/HN/93和分离的FMDV。置于37 ℃细胞培养箱中,分别在4、8、12和20 h收取样品,测定样品的PFU,绘制2株FMDV的一步生长曲线。
利用RT-PCR检测6份牛OPF中是否能够扩增出FMDV VP1基因。1%琼脂糖凝胶电泳结果显示,6个PCR产物中仅有1个在预期位置出现目的条带(图1)。将电泳产物胶回收后送至北京擎科生物科技股份有限公司进行序列测定,测得的序列与从GenBank下载的O型、A型和Asia1型FMDV VP1序列比对。MEGA 11的比对结果显示,PCR扩增产物为FMDV VP1基因,且属于O型Ind-2001谱系(图2)。上述结果表明检测到1份FMDV阳性的牛OPF,该OPF可用于分离FMDV。
利用BHK-21细胞从阳性OPF中分离FMDV。将PCR结果为阳性的牛OPF经过处理后接种至生长状况良好的BHK-21细胞,连续盲传3代,观察到BHK-21细胞出现明显的FMDV感染引起的CPE,即细胞变大、变圆,呈现倒挂葡萄串状(图3)。分离的FMDV传至第6代时,完全CPE时间稳定为12 h左右。大量制备第7代病毒样品,贮存于-80 ℃冰箱备用。
利用间接免疫荧光试验鉴定第7代样品。将第7代样品接种至BHK-21细胞,6 h后进行间接免疫荧光试验,使用的一抗为特异性识别FMDV 3A蛋白的鼠源单克隆抗体3A24。结果显示,接种第7代样品的BHK-21细胞与3A24单克隆抗体及山羊抗鼠FITC二抗作用后,出现明显绿色荧光,而对照细胞无绿色荧光(图4),表明从牛的OPF中成功分离到FMDV,将其命名为O/FMDV/OP/2022/B。
以第7代样品提取的总RNA为模板,利用RT-PCR分段扩增持续性感染FMDV O/FMDV/OP/2022/B的全基因组序列。如图5所示,使用设计的6对特异性引物(表1),扩增到6个与预期片段大小(2 666、1 808、1 724、393、1 439和1 745 bp)相符的目的片段。将6个PCR产物送至北京擎科生物科技股份有限公司测序并拼接出全基因组序列,O/FMDV/OP/2022/B的基因组全长为8 161 bp [不包括poly(C)和poly(A)序列],5′非编码区长1 076 bp,开放阅读框长6 996 bp,3′非编码区长89 bp。从NCBI数据库中下载33株Ind-2001谱系FMDV的抗原蛋白氨基酸序列,与O/FMDV/OP/2022/B的相应蛋白序列进行比对,发现O/FMDV/OP/2022/B在VP1、VP2和VP3蛋白上均发生了氨基酸残基突变,且多数突变位于VP1 (表2)。
通过蚀斑形成试验测定OPF分离株O/FMDV/ OP/2022/B和疫苗毒株O/FMDV/HN/93的蚀斑表型。结果显示(图6),2个毒株均可以在BHK-21细胞上形成大小混合蚀斑,但O/FMDV/OP/2022/B以小斑为主,平均蚀斑直径约为0.05 cm,而O/FMDV/HN/93几乎全为大斑,平均蚀斑直径约为0.5 cm。此外,与O/FMDV/HN/93相比,O/FMDV/OP/2022/B的蚀斑明显偏小。统计蚀斑数量后,计算出的O/FMDV/OP/2022/B和O/FMDV/ HN/93的病毒滴度分别为5.28×105 PFU/mL和1.70×108 PFU/mL。
通过一步生长曲线分析O/FMDV/OP/2022/B和O/FMDV/HN/93毒株的复制能力。结果表明(图7),O/FMDV/OP/2022/B在不同时间点的复制滴度均低于O/FMDV/HN/93,表明O/FMDV/OP/2022/B的复制能力弱于O/FMDV/HN/93。
FMD对畜牧养殖业构成了巨大威胁,而OPF中长期存在的无症状FMDV潜伏感染,即FMDV持续性感染,使得FMD的防控变得更加复杂。多种因素影响FMDV持续感染易感动物的口咽组织,主要包括宿主和病毒两方面。宿主动物为了避免频繁产生炎症,上呼吸道黏膜免疫对外来刺激具有较高的耐受水平。当上呼吸道鼻咽黏膜未能建立起强有效的抗病毒免疫时,病毒可能会发生持续性感染。此外,当动物机体的天然免疫和获得性免疫能力偏低时,也不容易完全清除体内感染的FMDV。在病毒方面,国外有研究显示,持续感染口咽组织的FMDV具有独特特性,主要包括病毒复制能力降低、蚀斑减小、致病力降低和抗逆性增强等,还可能与病毒抗原变异导致的免疫逃逸有关[14]。然而,目前国内对OPF中FMDV的分离及特性研究较少,这不利于FMD的防控和根除。
本研究在国家口蹄疫参考实验室从田间无症状牛采集的OPF样品中分离出一株FMDV,命名为O/FMDV/OP/2022/B。溯源结果显示,采样牛在采样前2个月内未发生过FMD,采样后1个月内也未出现FMD临床症状。因此,从牛口咽液中分离出的O/FMDV/OP/2022/B可基本认定为持续性感染FMDV。为了增强适应性并在宿主体内存活,持续性感染FMDV在与宿主相互选择、相互进化的过程中会发生遗传变异。Biswal等[14]发现,FMDV在持续性感染形成过程中会出现基因突变,这种突变增加了病毒种群的多样性,也可能影响病毒的抗原性。另有研究确定,持续性感染FMDV的VP1蛋白发生V50A突变,可能影响VP1 G-H环的稳定性,也可能破坏其与SUMO1 (small ubiquitin-like modifier, SUMO)的结合[15]。目前已报道FMDV的抗原位点共有5个,分布在VP1、VP2和VP3衣壳蛋白上,分别为VP1 G-H环和C末端组成的抗原位点1,VP2的B-C环、E-F环和H-I环组成的抗原位点2,VP1的B-C环组成的抗原位点3,VP3的B-B结组成的抗原位点4,以及VP1 G-H环组成的抗原位点5。本研究测定了牛口咽分离FMDV O/FMDV/OP/2022/B的全基因组序列,并与NCBI数据库中的33株FMDV氨基酸序列进行比对,发现O/FMDV/OP/2022/B在VP1、VP2和VP3蛋白中均发生了氨基酸残基突变,且多数突变发生在VP1 (表2)。在13个氨基酸残基突变中,VP1 V150M和VP1 P158A突变位于抗原位点1内,VP2 N191T突变位于新鉴定的抗原位点2内,VP3 E131K突变位于新鉴定的抗原位点3内[16]。VP1 E198K突变位于抗原位点1的C末端附近,VP2 L137Q、VP3 Q76R和VP3 A174V突变可能与FMDV特异性抗体的相互作用相关[17-19],VP3 T99A和VP1 R172Q可能影响FMDV特异性抗体的产生[10],VP1 A96T突变可能影响FMDV的受体利用能力。由此可见,O/FMDV/OP/2022/B衣壳蛋白中高达71%的氨基酸残基突变与FMDV抗体的产生相关,推测O/FMDV/OP/2022/B在牛OPF中的存在可能与其逃逸宿主免疫清除的能力密切相关。
本研究还分析了口咽分离FMDV O/FMDV/OP/2022/B的蚀斑表型和一步生长曲线。蚀斑表型与FMDV的复制、毒力和宿主嗜性等特性相关。结果显示,O/FMDV/OP/2022/B的蚀斑显著小于疫苗毒株O/FMDV/HN/93的蚀斑,且O/FMDV/OP/2022/B的PFU也明显低于O/FMDV/HN/93。一步生长曲线与FMDV的复制能力相关,结果表明,O/FMDV/OP/2022/B的一步生长曲线明显低于O/FMDV/HN/93。上述结果表明,O/FMDV/OP/2022/B的增殖能力较弱,更容易与宿主细胞维持平衡,从而在OPF中持续性存在。本研究从OPF中成功分离到一株牛源FMDV,命名为O/FMDV/OP/2022/B,测定了其全基因组序列,并分析了其增殖特性,为深入研究FMDV持续感染牛口咽组织的机制以及FMD的防控提供了参考。
