[Objective] To study the effects of the glycosyltransferase WekO involved in O-antigen synthesis on the biological characteristics of avian pathogenic Escherichia coli (APEC). [Methods] The mutant strain ΔwekO of APEC O1 was constructed by Red homologous recombination, and the complementary strain CΔwekO was then constructed. The lipopolysaccharide (LPS) profile of each strain was identified by silver staining. Simultaneously, the growth rate and swimming motility were measured. The reactivity of each strain with rabbit anti-O1 serum was determined by Western blotting. The ability of ΔwekO to form biofilms was measured by the crystal violet staining method. DF-1 cells were used to evaluate the adhesion and invasion of ΔwekOin vitro. Subsequently, chicks were selected as an animal model to evaluate the pathogenicity of ΔwekO. [Results] The mutant strain ΔwekO and the complementary strain CΔwekO were constructed. The LPS profile of ΔwekO was incomplete compared with that of the wild-type strain. The mutant lacked O-antigen bands and showed no reactivity to anti-O1 serum. There was no significant difference in growth rate between different strains (P>0.05). However, the motility and biofilm formation capabilities of ΔwekO decreased (P<0.001). Additionally, ΔwekO demonstrated weakened adhesion to DF-1 cells (P<0.001) and demonstrated weakened invasion to DF-1 cells (P<0.01). Also, ΔwekO reduced pathogenicity to 7-day-old chicks (P<0.05). [Conclusion] The deletion of wekO results in impaired O-antigen synthesis, incomplete LPS profile, loss of flagellar and biofilm formation capabilities, and reduced pathogenicity of APEC. These findings are highly significant for improving the understanding of the role of glycosyltransferases involved in APEC O-antigen synthesis.

【目的】 探讨糖基转移酶WekO基因对禽致病性大肠杆菌(avian pathogenic Escherichiacoli, APEC)生物学特性的影响。 【方法】 以O1血清型APEC为亲本株,利用Red同源重组技术构建wekO基因缺失株(ΔwekO)和回补株(CΔwekO)。通过硝酸银染色和蛋白质免疫印迹(Western blotting)分析细菌脂多糖(lipopolysaccharide, LPS)图谱及血清反应性。同时,检测ΔwekO的生长速率、运动能力,并采用结晶紫染色法评估其生物被膜形成能力。此外,利用鸡成纤维细胞(DF-1)测定ΔwekO的体外黏附和侵袭能力,并以萧山蛋鸡为动物模型评估其致病力。 【结果】 成功构建了缺失株ΔwekO和回补株CΔwekO;与野生株相比,ΔwekO的LPS图谱不完整,O-抗原梯状条带缺失,且与O1血清无反应。在生长速率方面,ΔwekO与野生株无显著差异(P>0.05),但其运动能力和生物被膜形成能力显著降低(P<0.001),对DF-1细胞的黏附率显著降低(P<0.001)、侵袭率显著降低(P<0.01),对7日龄雏鸡的致病力也显著降低(P<0.05)。 【结论】 本研究表明,wekO基因缺失导致APEC的O-抗原合成受阻,进而影响LPS的完整性。此外,wekO基因缺失还导致细菌的鞭毛形成和生物被膜构建能力丧失,致病力显著下降。这些结果对于深入理解参与APEC O-抗原合成的糖基转移酶的生物学功能具有重要意义。

【目的】 探讨糖基转移酶WekO基因对禽致病性大肠杆菌(avian pathogenic Escherichiacoli, APEC)生物学特性的影响。 【方法】 以O1血清型APEC为亲本株,利用Red同源重组技术构建wekO基因缺失株(ΔwekO)和回补株(CΔwekO)。通过硝酸银染色和蛋白质免疫印迹(Western blotting)分析细菌脂多糖(lipopolysaccharide, LPS)图谱及血清反应性。同时,检测ΔwekO的生长速率、运动能力,并采用结晶紫染色法评估其生物被膜形成能力。此外,利用鸡成纤维细胞(DF-1)测定ΔwekO的体外黏附和侵袭能力,并以萧山蛋鸡为动物模型评估其致病力。 【结果】 成功构建了缺失株ΔwekO和回补株CΔwekO;与野生株相比,ΔwekO的LPS图谱不完整,O-抗原梯状条带缺失,且与O1血清无反应。在生长速率方面,ΔwekO与野生株无显著差异(P>0.05),但其运动能力和生物被膜形成能力显著降低(P<0.001),对DF-1细胞的黏附率显著降低(P<0.001)、侵袭率显著降低(P<0.01),对7日龄雏鸡的致病力也显著降低(P<0.05)。 【结论】 本研究表明,wekO基因缺失导致APEC的O-抗原合成受阻,进而影响LPS的完整性。此外,wekO基因缺失还导致细菌的鞭毛形成和生物被膜构建能力丧失,致病力显著下降。这些结果对于深入理解参与APEC O-抗原合成的糖基转移酶的生物学功能具有重要意义。

[Objective] To study the effects of the glycosyltransferase WekO involved in O-antigen synthesis on the biological characteristics of avian pathogenic Escherichia coli (APEC). [Methods] The mutant strain ΔwekO of APEC O1 was constructed by Red homologous recombination, and the complementary strain CΔwekO was then constructed. The lipopolysaccharide (LPS) profile of each strain was identified by silver staining. Simultaneously, the growth rate and swimming motility were measured. The reactivity of each strain with rabbit anti-O1 serum was determined by Western blotting. The ability of ΔwekO to form biofilms was measured by the crystal violet staining method. DF-1 cells were used to evaluate the adhesion and invasion of ΔwekOin vitro. Subsequently, chicks were selected as an animal model to evaluate the pathogenicity of ΔwekO. [Results] The mutant strain ΔwekO and the complementary strain CΔwekO were constructed. The LPS profile of ΔwekO was incomplete compared with that of the wild-type strain. The mutant lacked O-antigen bands and showed no reactivity to anti-O1 serum. There was no significant difference in growth rate between different strains (P>0.05). However, the motility and biofilm formation capabilities of ΔwekO decreased (P<0.001). Additionally, ΔwekO demonstrated weakened adhesion to DF-1 cells (P<0.001) and demonstrated weakened invasion to DF-1 cells (P<0.01). Also, ΔwekO reduced pathogenicity to 7-day-old chicks (P<0.05). [Conclusion] The deletion of wekO results in impaired O-antigen synthesis, incomplete LPS profile, loss of flagellar and biofilm formation capabilities, and reduced pathogenicity of APEC. These findings are highly significant for improving the understanding of the role of glycosyltransferases involved in APEC O-antigen synthesis.

禽致病性大肠杆菌是一种肠道外致病性大肠杆菌(extraintestinal pathogenic Escherichiacoli, ExPEC),感染该菌可引起禽大肠杆菌病,导致家禽肝周炎、气囊炎、心包炎、腹膜炎、关节炎等多种疾病[1]。APEC可感染各年龄段的多种家禽[2]。4-6周龄的肉鸡更容易受到APEC感染,而蛋鸡在整个生长和产蛋期均会受到APEC的威胁,尤其是在产蛋高峰期前后,给养殖行业造成了严重的经济损失[3]。禽致病性大肠杆菌与人尿道致病性大肠杆菌(uropathogenic Escherichia coli, UPEC)、新生儿致脑膜炎大肠杆菌(neonatal meningitis-causing Escherichia coli, NMEC)在基因组上存在相似性,可能会通过污染家禽产品进而导致人类尿道感染和脑膜炎,具有公共卫生风险,危害人类健康[4-5]
脂多糖(lipopolysaccharide, LPS)是革兰氏阴性菌细胞壁外层的组成成分,具有刺激宿主Toll样受体4 (Toll-like receptor 4, TLR4)引发炎症反应、抵抗抗生素杀伤等多种生物学功能[6]。LPS由3个组分组成:脂质A、核心多糖和O-抗原[7]。APEC的血清型以O-抗原结构的不同来分型[8],其中O1、O2和O78血清型常从全球鸡大肠杆菌感染中分离出来[9]。O-抗原是革兰氏阴性菌的重要毒力因子[10],其结构不完整或缺失会导致许多病原体对血清的敏感性增高以及致病力降低[11]。此外,O-抗原可以在细菌表面形成一层保护性屏障[12],帮助细菌抵抗宿主免疫系统的攻击[13],尤其是对补体系统的抵抗能力。这种抗补体作用能够增强细菌在宿主内的存活率,从而提高细菌的毒力[14]
O-抗原合成的基因通常以基因簇的形式存在于基因组中[15]。在大多数大肠杆菌和志贺氏菌中,O-抗原基因簇位于2个管家基因galFgnd之间[16]。APEC O1血清型的O-抗原基因簇由galF-rmlB-rmlD-rmlA-rmlC-wzx-mnaA-fdtC-wekM- wzy-wekN-wekO-gnd编码[17]。本研究前期通过生物信息学分析发现,wekO基因编码一种糖基转移酶,该酶负责催化甘露糖(β-l-ManNAc)和鼠李糖(β-d-GlcNAc)形成二糖[β-d-ManNAc-(1→2)-l-Rha]。然而,wekO基因在APEC O1中的作用尚未明确。因此,本研究聚焦于wekO基因的部分生物学功能,进一步完善糖基转移酶在APEC O1中的生物学作用。
DF-1细胞及质粒pKD46 (30 ℃培养,氨苄霉素抗性)、pKD3 (37 ℃培养,氯霉素抗性)、pCP20 (30 ℃培养,氨苄霉素抗性)、pSTV28 (37 ℃培养,氯霉素抗性)均由本实验室保存;高保真PCR扩增酶(KOD One PCR Master Mix)和DNA连接酶(ligation high v2)购自东洋纺(上海)生物科技有限公司;基因组DNA提取试剂盒购自生工生物工程(上海)股份有限公司;高纯度质粒小提中量试剂盒购自上海惠凌生物技术有限公司;限制性内切酶Kpn I、BamH I购自New England Biolabs公司;大肠杆菌O1血清型诊断血清购自天津生物芯片技术有限责任公司;DF-1细胞培养基(Dulbecco’s modified eagle medium, DMEM)购自赛默飞世尔科技(中国)有限公司;胎牛血清(fetal bovine serum, FBS)购自苏州依科赛生物科技股份有限公司;HRP标记的山羊抗兔IgG二抗、大肠杆菌DH5α感受态均购自武汉爱博泰克生物科技有限公司;试验动物萧山鸡购自杭州萧山种鸡(萧山鸡)有限公司。本研究动物实验经浙江农林大学实验动物伦理委员会审核,伦理编号为ZAFUC202477。
本研究基于GenBank数据库收录的APEC O1全基因组序列(登录号:GCF_902880315.1) ,以及质粒pKD3和pSTV28序列设计引物。其中,wekO基因上下游同源臂分别使用wekO-UF/UR和wekO-DF/DR引物对进行扩增,pKD3质粒的氯霉素抗性基因片段通过wekO-CF/CR引物对进行扩增。基因敲除株使用wekO-outF/outR引物对进行验证,wekO基因启动子区使用PwekO-Kpn I-F/PwekO-R引物对进行扩增,wekO基因编码区(coding DNA sequence, CDS)使用wekO-F1/wekO-BamH I-R引物对进行扩增。此外,PwekO-Kpn I-F/wekO-BamH I-R引物对用于启动子区与编码区的融合扩增,回补质粒使用M13-F/R引物对进行鉴定,回补株使用wekO-inF/inR引物对进行验证。本研究所用引物序列如表1所示。
吸取OD600为0.8的APEC O1菌液2 mL,按照基因组DNA提取试剂盒说明书提取APEC O1基因组。基因组提取完成后使用多功能酶标仪检测浓度,并将基因组用于试验或置于-20 ℃冰箱保存。
根据Red同源重组原理构建wekO基因缺失株[18]。以APEC O1基因组为模板,扩增wekO基因上下游同源臂(约500 bp),以pKD3质粒为模板,扩增氯霉素抗性基因(约1 000 bp)。按照表1中引物进行PCR扩增,PCR反应体系(50 μL):KOD One PCR Master Mix 25 µL,上、下游引物(10 µmol/L)各2.5 µL,DNA模板1 µL,ddH2O 19 µL。PCR反应条件:98 ℃预变性5 min;98 ℃变性10 s,58 ℃退火5 s,68 ℃延伸20 s,35个循环;68 ℃终延伸5 min。将上述3个片段进行Overlap PCR,获得wekO upstream-chloramphenicol fragment-wekO downstream融合片段。第2次PCR反应体系(50 μL):KOD One PCR Master Mix 25 µL,上、下游引物(10 µmol/L)各2.5 µL,3个片段DNA模板各1 µL,ddH2O 17 µL。PCR反应条件:98 ℃预变性5 min;98 ℃变性10 s,58 ℃退火5 s,68 ℃延伸1 min,35个循环;68 ℃终延伸5 min。将该融合片段纯化回收后,电转化至APEC O1 (pKD46)感受态细胞中,通过同源重组将氯霉素抗性片段替代wekO基因,并通过氯霉素抗性筛选阳性克隆。将阳性克隆传代纯化后制备感受态,电转化2 μg pCP20质粒以消除氯霉素抗性片段,并通过氨苄霉素抗性筛选缺失株。经37 ℃传代纯化后获得缺失株,命名为ΔwekO
以APEC O1基因组为模板,扩增wekO基因启动子和CDS序列。将启动子和CDS片段进行Overlap PCR扩增,参照表1中引物扩增DNA片段。PCR反应体系(50 μL):KOD One PCR Master Mix 25 µL,上、下游引物(10 µmol/L)各2.5 µL,启动子和CDS序列DNA模板各1 µL,ddH2O 18 µL。PCR反应条件:98 ℃预变性5 min;98 ℃变性10 s,58 ℃退火5 s,68 ℃延伸30 s,35个循环;68 ℃终延伸5 min。使用限制性内切酶Kpn I和BamH I对融合片段及pSTV28载体进行双酶切。酶切产物纯化后,使用ligation high v2进行连接。将连接产物热转化至大肠杆菌DH5α感受态中,通过氯霉素抗性筛选阳性质粒。提取阳性质粒后,获得回补质粒pSTV28-wekO。将2 μg回补质粒电转化至ΔwekO缺失株感受态中,经PCR验证与测序鉴定后获得wekO回补菌株,命名为CΔwekO。验证时的PCR反应体系(20 μL):KOD One PCR Master Mix 10 µL,上、下游引物(10 µmol/L)各0.5 µL,DNA模板1 µL,ddH2O 8 µL。PCR反应条件:98 ℃预变性5 min;98 ℃变性10 s,58 ℃退火5 s,68 ℃延伸20 s,35个循环;68 ℃终延伸5 min。
参照文献[19]方法制备细菌LPS样品。吸取OD600为0.8的各菌株菌液2 mL,转移至无菌EP管中,5 000 r/min离心5 min,弃上清。用无菌1×PBS洗涤2次后,加入150 μL菌体裂解液,100 ℃水浴10 min。待样品冷却至室温后,12 000 r/min离心10 min,取30 μL上清、270 μL上样缓冲液和3 μL蛋白酶K混合,37 ℃摇床酶解蛋白1 h,获得各菌株LPS样品,可上样电泳或置于-20 ℃冰箱保存。使用硝酸银染色法检测各菌株的LPS图谱。首先,采用SDS-PAGE技术对上述LPS样品进行分离电泳。电泳完成后,将分离胶置于固定溶液中,室温下缓慢振荡固定4 h。用ddH2O振荡洗涤15 min,重复3次。随后,使用高碘酸溶液室温摇床振荡氧化反应7 min,ddH2O振荡洗涤15 min,重复3次。使用银染液室温摇床振荡着色10 min,ddH2O振荡洗涤15 min,重复3次。最后,加入适量显色液,缓慢振荡直至出现黑色条带,记录并拍摄实验结果。同时,将SDS-PAGE胶进行转膜,使用Western blotting方法检测各菌株LPS与大肠杆菌O1血清型O因子诊断血清的特异性反应。
吸取OD600为0.8的各菌株菌液150 μL,转接至15 mL LB液体培养基中,混合均匀后吸取200 µL测量OD600并记录数值(记为0 h)。将菌液置于37 ℃、200 r/min摇床培养,每间隔1 h测量OD600并记录数值,直至12 h,绘制各菌株生长曲线。将1 μL OD600约为0.8的各菌液穿刺至半固体培养基中(穿刺深度约为培养基深度的1/3-2/3),在37 ℃恒温培养箱中分别静置培养6 h和12 h。测量每个菌株的运动圈直径并扫描平板,使用GraphPad Prism 8.0软件进行数据处理和统计学分析,以评估各菌株运动能力的差异。此外,对APEC O1、ΔwekO和CΔwekO单菌落进行醋酸铀染色后,放置在铜网上干燥30 min,然后在透射电镜(transmission electron microscope, TEM)下观察并拍摄鞭毛结构。
采用结晶紫染色法测定细菌的生物被膜形成能力[20]。吸取OD600为0.8的各菌株菌液20 μL,加入至含有180 μL无菌LB的96孔聚丙烯板中,并设置无菌LB作为阴性对照。在25 ℃恒温培养箱中,分别静置培养24 h和48 h后,弃去96孔聚丙烯板中的菌液,使用无菌1×PBS轻微吹打洗涤3次,60 ℃烘箱风干。随后,各孔加入200 μL 0.1%结晶紫进行染色30 min,使用无菌1×PBS洗涤至无结晶紫残留。放置60 ℃烘箱固定生物被膜30 min后,每孔加200 μL 95%乙醇振荡15 min,测定各孔OD595值。最后,使用GraphPad Prism 8.0软件进行数据处理和统计学分析,以评估各菌株生物被膜形成的差异。
参考文献[21]方法进行细胞感染试验。当DF-1细胞在12孔细胞板中生长至均匀单层(细胞密度约为4×105个/孔)时,首先使用37 ℃无菌1×PBS洗涤3次,加入1 mL不含血清的DMEM (含有4×107 CFU/mL的各细菌),确保感染复数(multiplicity of infection, MOI)为100:1,每组做3个重复。将DF-1细胞置于37 ℃培养箱中感染2 h后,移除原培养基。随后使用37 ℃的无菌1×PBS缓冲液轻微洗涤3次,以去除未附着的细菌。向每孔加入1 mL含0.5% Triton X-100的细胞裂解液,在4 ℃静置裂解10 min。最后,冰上反复吹打破碎细胞,将裂解液稀释103倍,梯度点板细菌计数,计算黏附率,如公式(1)所示。
黏附率=(a/c)×100%
式中:a为黏附细胞的细菌数,c为初始感染细菌总数。
侵袭试验的细胞培养方法和感染比与黏附试验相同。细菌37 ℃感染2 h后,首先使用37 ℃的无菌1×PBS缓冲液轻微洗涤3次,每孔加入1 mL DMEM培养基和2 μL 50 mg/mL庆大霉素,继续培养1 h后使用37 ℃的无菌1×PBS缓冲液轻微洗涤3次。随后向每孔加入1 mL含0.5% Triton X-100的细胞裂解液,4 ℃静置裂解10 min。最后,冰上反复吹打破碎细胞,将裂解液稀释102倍,梯度点板细菌计数,计算入侵率,如公式(2)所示。
入侵率=(b/c)×100%
式中:b为入侵细胞的细菌数,c为初始感染细菌总数。
参考文献[22]方法进行攻毒试验。选取7日龄萧山鸡共70只,随机分为7组,每组10只。采用气囊注射方式分别感染APEC O1、ΔwekO和CΔwekO 3组菌株,每组每只注射100 μL 107 CFU/mL的对应菌液,对照组注射100 μL无菌1×PBS。攻毒后正常饲养7 d,观察并记录死亡情况,整理数据绘制存活曲线,评估缺失株的毒力变化。
所有试验均进行3次生物学重复。试验结果使用GraphPad Prism 8.0软件进行t-test统计学分析,数据以平均值±SD表示。ns表示P>0.05,*表示P<0.05,**表示P<0.01,***表示P<0.001。使用Adobe Illustrator 2021软件进行排版作图。
为了研究wekO基因的部分生物学功能,本研究构建了wekO基因缺失株和回补株。利用Red同源重组技术构建wekO基因缺失株,使用wekO-outF与wekO-outR引物进行PCR扩增,结果显示条带大小为610 bp,而APEC O1的条带大小为1 384 bp,成功构建了ΔwekO (图1A)。以APEC O1基因组为模板,PCR扩增含启动子区域的wekO片段,并将纯化的PCR片段与pSTV28质粒进行酶切连接。将构建成功的回补质粒电转化至ΔwekO感受态细胞中,使用wekO-inF和wekO-inR引物进行鉴定,野生株和回补株的条带大小均为618 bp,而缺失株未扩增出条带。经测序验,结果证符合预期,成功构建了CΔwekO (图1B)。
wekO基因位于APEC O1的O-抗原基因簇中,负责单糖之间的连接。因此,本研究使用硝酸银染色法鉴定各菌株的LPS图谱,结果显示wekO缺失后LPS仅存在类脂A与单个短层O单元(图2A)。这表明wekO缺失导致O-抗原合成受阻,APEC O1无法合成完整的LPS。此外,本研究还通过Western blotting检测各菌株的LPS与大肠杆菌O1 O因子血清的反应能力,结果显示ΔwekO不能与O1 O因子血清特异性结合(图2B),说明ΔwekO由于缺失O-抗原导致LPS与O1血清不反应。上述结果表明wekO基因参与O-抗原合成。
通过测定细菌在LB培养基中12 h的OD600值,绘制细菌生长曲线。结果表明,wekO基因缺失不影响APEC O1在LB培养基中的生长(P>0.05) (图3A)。运动能力检测表明,细菌培养6 h时,APEC O1的运动圈直径为5.44 cm,ΔwekO的运动圈直径为0.23 cm,CΔwekO的运动圈直径为4.95 cm;细菌培养12 h时,APEC O1的运动圈直径为8.31 cm,ΔwekO的运动圈直径为0.94 cm,CΔwekO的运动圈直径为7.13 cm (图3B)。统计学分析表明,ΔwekO的运动能力相较于APEC O1显著降低(P<0.001) (图3C)。这说明wekO基因在细菌运动能力的形成中起重要作用。
本研究通过醋酸铀染色,利用透射电镜观察细菌鞭毛的形成情况。结果显示,在37 ℃条件下静置培养12 h后,ΔwekO菌株未观察到鞭毛,而野生株和回补株则可观察到明显的鞭毛形态(图4)。这表明,wekO基因缺失导致鞭毛合成受阻,进而使ΔwekO菌株的运动能力丧失。
通过结晶紫染色法测定各菌株的生物被膜形成能力。结果显示,在培养24 h和48 h后,96孔板内各菌株的生物被膜染色深浅不一,其中ΔwekO的颜色最浅,依次为CΔwekO和APEC O1 (图5A)。各菌株的OD595吸光值从低到高依次为ΔwekO、CΔwekO和APEC O1 (图5B)。统计学分析表明,ΔwekO的生物被膜形成能力显著低于APEC O1 (P<0.001),而回补株则恢复至野生株水平。这说明wekO基因参与细菌生物被膜的形成。
使用DF-1细胞评估ΔwekO菌株感染细胞的黏附与入侵能力。结果显示,ΔwekO菌株感染DF-1细胞的黏附率较APEC O1下降44.10% (图6A),侵袭率较APEC O1下降26.33% (图6B)。统计学分析表明,ΔwekO菌株对DF-1细胞的黏附率显著低于APEC O1 (P<0.001),侵袭率显著低于APEC O1 (P<0.01)。这说明wekO基因参与细菌的黏附与入侵。
将各菌株通过气囊接种至1周龄雏鸡体内,观察并记录7 d内雏鸡的存活情况。当攻毒剂量为107 CFU/mL时,攻毒野生株和回补株的雏鸡存活率均为10%,而ΔwekO菌株的存活率为60%。与野生株相比,ΔwekO菌株的毒力显著降低(P<0.05) (图7)。
APEC O1是我国田间APEC的主要流行血清型,给家禽业带来了严重的经济损失[23]。此外,与其他血清型相比,APEC与人类肠外致病性大肠杆菌基因组具有更高的相似性,存在人畜共患的风险[5]。近年来,APEC对多种抗生素(包括碳青霉烯类等关键药物)表现出耐药性,这使得治疗与防控工作变得更加困难[3]。O-抗原位于细菌外膜,是最先与药物或宿主相互作用的成分,因此研究APEC O1的致病机制尤为重要。O-抗原作为LPS的重要组成部分,其合成需要一系列负责糖基合成和修饰的基因参与。已有研究报道,缺失wzy (O-抗原聚合酶)[24]waaL (O-抗原连接酶)[25]均会影响细菌表型和毒力。曹启予等[26]发现,wekM (O-抗原糖基转移酶)参与细菌LPS合成及其对环境的适应。作为同基因簇基因,WekO基因注释与WekM一致,但WekO负责O-抗原中甘露糖与鼠李糖的连接。本研究对wekO基因进行敲除,分析突变株的部分生物学特性,结果表明缺失wekO基因会影响细菌鞭毛形成能力、生物被膜形成能力、细胞黏附入侵能力和致病力。
wekO位于APEC O1 O-抗原基因簇中,因此本研究首先通过银染法分析该基因是否参与O-抗原合成,从而影响APEC O1 LPS图谱的变化。结果显示,缺失wekO后无法形成长链O-抗原,LPS仅存在核心多糖和短链O单元。通过Western blotting试验发现ΔwekO无法检测到LPS条带,说明ΔwekO无法与O1型血清发生反应。上述结果表明,wekO基因在形成完整LPS及与O1型血清反应中发挥了重要作用。缺失该糖基转移酶导致糖单元之间连接受阻,进而影响O-抗原的合成,提示糖基转移酶WekO在O-抗原合成中发挥重要作用,但其参与O-抗原合成过程的具体步骤及负责的糖基转移过程还有待进一步研究。
本研究结果显示,wekO基因缺失导致细菌无法形成鞭毛,进而使细菌几乎丧失了运动能力,这与已知报道的大肠杆菌中O-抗原的缺失在泳动和群集运动中均显示出显著缺陷[27],以及奇异变形杆菌中O-抗原连接酶的缺失抑制了固体表面的群集运动[28]相符。研究表明,LPS突变可以通过荚膜多糖合成调控系统(regulation of capsular polysaccharide synthesis, Rcs)通路抑制鞭毛基因表达来损害细菌运动能力[29]。由此推测,wekO基因缺失可能导致LPS结构发生变化,进而影响Rcs系统对flhDC相关鞭毛基因的表达。此外,本研究结果还显示,wekO基因缺失可以导致细菌几乎丧失生成生物被膜的能力。研究表明,鞭毛在大肠杆菌生物被膜形成初始阶段发挥着重要作用[30]。鞭毛介导的运动可以使附着、分裂的细菌沿着非生物表面传播,寻找合适的生态位,随后运动性被抑制,细菌转化为静止状态并开始形成生物被膜[31]。由此推测,wekO基因缺失导致细菌失去运动能力,从而显著影响其生物被膜的形成。
本研究还对体外细胞黏附和侵袭能力进行了评估。细胞感染试验结果显示,ΔwekO对DF-1细胞的黏附率较APEC O1下降44.10%,侵袭率较APEC O1下降26.33%,说明wekO基因参与了细菌黏附侵袭宿主细胞的过程。细菌黏附和入侵宿主细胞的过程是介导微生物定殖和感染宿主发病的首要条件[32]。细菌的黏附和侵袭与O-抗原密切相关,例如缺失wzy基因后LPS结构改变导致APEC O2黏附和入侵能力显著下降[24]。由此推测,ΔwekO可能是因为改变LPS的结构,从而影响对宿主细胞的黏附和侵袭。同时,ΔwekO感染天然宿主萧山蛋鸡的致病力较APEC O1降低,说明wekO参与细菌毒力。由此推测,ΔwekO黏附侵袭能力降低导致细菌致病力下降。
综上所述,本研究针对O-抗原糖基合成酶基因wekO缺失的部分生物学特性进行了研究,结果显示ΔwekO的LPS图谱不完整,无法形成鞭毛,丧失了生物被膜形成能力,细胞黏附侵袭能力下降,细菌致病力降低。本研究进一步完善了O-抗原合成相关基因的生物学功能。
