如果说土壤是地球的皮肤,那么黑土地微生物组可能是最重要的皮肤,因为黑土地覆盖了全球约7%的陆地面积,且黑土所在国家的人口占世界总人口的25%[1]。微生物通常被认为是地球生态系统最重要的分解者[2],作物可持续产能的关键,地球元素循环的引擎,宜居环境的塑造者[3]。然而,长期高强度利用下,黑土地严重退化,黑土地微生物组失衡,直接威胁国家粮食安全。以我国为例,黑土地贡献了全国1/4的粮食产量和1/3的粮食调出量,土壤退化问题不可忽视[4]。因此,黑土地保护利用已连续15年被写列入中央一号文件等国家重大纲领性文件。2022年8月1日,《中华人民共和国黑土地保护法》正式实施,黑土地成为全球唯一由国家立法保护的土壤类型,黑土地保护利用意义重大。
从历史上看,土壤学正是脱胎于黑土地的农业生产研究。150年前,俄罗斯因干旱等自然灾害导致农业歉收、民不聊生,道库恰耶夫行程万里开展黑土地调查,出版了《俄罗斯黑钙土》,指导了农业生产,创立了土壤发生学,使土壤学脱离地质学而成为一门独立学科[5]
黑土研究孕育了土壤学,但黑土微生物组研究与黑土地位极不相称。近10年来,在Web of Science核心数据库中,土壤学论文数量高达58万篇,土壤微生物组论文8万余篇,而黑土地微生物组论文仅2 500余篇,占比不足0.5%。基于此,《微生物学报》策划出版“黑土地微生物组”专栏,聚焦微生物资源发掘、生物养分活化、黑土退化阻控、肥沃耕层培育及碳中和微生物生态等方向,重点介绍我国黑土地微生物组的研究进展。
近30年来,微生物学最颠覆性的突破常被认为是卡尔伍斯的“三域分类理论”[6]。2011年,Nature Reviews Microbiology中发表题为“And the winner should be…”的评述性文章,认为卡尔伍斯应获诺贝尔奖[7]。该理论的核心是:以核糖体rRNA为分子钟的系统发育分析表明,地球所有生物可分为细菌域、古菌域和真核生物域。基于这一理论,大量研究发现地球“最高可达99%的微生物难以培养”,甚至将这些难培养微生物与物理学暗物质类比,称为“微生物暗物质”[8],在农业、医药、工业和食品等领域具有巨大潜力[9]。然而,在缺乏微生物纯培养条件下,单纯的核糖体RNA序列甚至全基因组序列能否作为种质资源的证据,在微生物系统分类学领域仍然存在较大的争议[10];甚至有研究表明,99%微生物难培养并无明确证据[11]。耦合开展微生物富集培养和分子生态分析,或许是破解这一难题的关键策略。
黑土地被认为是地球上最肥沃的土壤,可能蕴藏着最高的微生物多样性,深度发掘其中关键养分元素转化微生物菌种资源具有重要意义。初始黑土发生发育通常经历3个阶段,包括岩漆、地衣和苔藓[12],其共同特征是固氮。我们推测,原始黑土发生初期,母质中氮素匮乏,植物凋落物投入后,其碳氮比约为30,而微生物碳氮比仅为5左右,微生物必须通过固氮克服氮素胁迫才能分解植物凋落物。同时,寒温带大陆性季风气候下,夏季炎热多雨,植被生长茂盛并产生大量凋落物,进一步刺激固氮微生物繁殖,形成大量生物有机氮。然而由于冬季漫长干冷,微生物休眠、活性低,导致有机氮积累多、分解少,形成了高肥力黑土。挖掘黑土地固氮微生物资源,解析其植物促生功能及其分子机制,有望为合成微生物制剂研发,健康黑土定向培育、盐碱地改良及可持续利用提供理论依据。
黑土地微生物组是一把双刃剑,既有正效应、又有负效应[13]。正效应主要表现为分解复杂有机质、释放无机养分供给作物生长。我国黑土地每年产生粮食约1 750亿kg,其中绝大部分N、P、K养分均来自土壤微生物的转化,特别是外源有机物料的输入。然而,黑土地微生物群落是一种状态,而非是一种结果,这些微生物群落生态演化历经数千年甚至上万年的复杂过程,微生物组形成的环境条件不可复制,发生过程不可逆转,真实重演其发育演化不具有现实操作性[14]。因此,利用空间换时间策略,通过研究土壤剖面微生物群落构建机制,结合长期定位试验,包括施肥和耕作管理等不同模式,则可能准确解析土壤有机质转化的微生物调控机制。合成生物学技术(如CRISPR基因编辑、细胞表面展示)为氮磷高效养分元素转化的合成微生物生态调控策略提供了新机遇。
微生物负效应的典型示例则包括酸化和连作障碍等。大量施用氮肥促进微生物硝化过程,导致土壤酸化,引起土壤结构恶化,加剧黑土地退化和作物减产[13]。经典的理论大多聚焦细菌、古菌和全程氨氧化菌对硝化过程的生态与分子调控机制。然而,长期以来由于技术的局限,黑土地主要活性微生物类群对酸化过程的适应机制,土壤属性如有机氮质量和数量变化特征及其对特定功能微生物的定向选择机制仍不清楚[15]。同时,长期种植导致黑土中富集了大量的作物病原微生物,对农业可持续发展造成威胁,对此类问题也不能忽视。深入发掘耐酸和嗜酸等关键功能微生物资源,通过开展“从上到下”和“从下到上”的迭代微生物功能筛选策略,可为黑土地酸化和连作障碍防控提供新思路。
《中华人民共和国黑土地保护法》将黑土地定义为:黑龙江省、吉林省、辽宁省、内蒙古自治区的相关区域范围内具有黑色或者暗黑色腐殖质表土层,性状好、肥力高的耕地。根据我们的初步估算,黑土区3 590万hm2耕地中,土壤养分含量有机氮约1 600亿kg、有机磷800亿kg、钾9 600亿kg。若无外源投入,黑土氮素养分约23年即可能被完全耗竭,无法维持作物生长。外源养分投入后的元素转化与作物吸收几乎完全依赖微生物。随着先进技术的快速发展,微生物学已与土壤学、植物微生物组、合成生物学、计算生物学、生态学等交叉融合,有望催生跨学科研究新范式,为培育健康黑土、调控地上-地下互作机制、实现微生物群落-养分循环-土壤健康-作物高产稳产的动态平衡与可持续利用提供重要科技支撑。
黑土地被誉为“耕地中的大熊猫”。然而,没有微生物,就没有黑土地。期望“黑土地微生物组”专栏能够起到抛砖引玉的作用,为未来黑土地微生物组研究提供有益启示。
