Objective

This study aimed to investigate the effects of different ultrasound-combined pretreatments on the microwave drying efficiency and dried product quality of star anise,identify the optimal pretreatment method and its underlying mechanism,and thereby provide reference for the green,efficient,and energy-saving drying processing of star anise.

Method

Fresh star anise was subjected to three pretreatments as raw materials:ultrasound (US),ultrasound combined with steam blanching (US-SB),and ultrasound combined with freeze-thaw treatment (US-FT),with samples directly subjected to microwave drying without pretreatment served as the control (CK). Effects of the three pretreatments on the microwave drying characteristics,physicochemical quality,and antioxidant activity of star anise were systematically analyzed,and the underlying mechanisms were further elucidated through microstructural observation and low-field nuclear magnetic resonance analysis.

Result

All three pretreatments disrupted the cellular structure of star anise and increased the number of water-migration pathways. Compared with the CK group [drying time of 261.67 min and specific energy consumption of 14.60 (kW·h)/kg],the drying times of the US,US-SB,and US-FT groups were reduced by 8.92%,46.50%,and 32.48% respectively,while their specific energy consumptions decreased by 13.08%,50.00%,and 33.56% respectively. The US-SB group exhibited the highest drying efficiency. However,polyphenol oxidase (PPO) activity was almost completely inactivated,resulting in no significant difference in the redness value (a*) of dried product compared with the CK group (P>0.05,the same below). Moreover,its volatile oil content (6.59%),shikimic acid content (8.20%),and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate (53.95%) were significantly lower than those of the CK group (P<0.05,the same below). PPO activity in the US and US-FT groups was significantly higher than that in the CK group,and the a* values significantly increased by 15.74% and 15.64% compared with CK group respectively,demonstrating a brighter red color;the DPPH radical scavenging rates increased significantly by 22.28% and 25.53% compared with CK group respectively;no significant differences in volatile oil or shikimic acid contents were observed between these two groups and the CK group. The volume shrinkage rate of US-FT group (41.88%) showed no significant difference with the CK group,whereas those of the US (45.78%) and US-SB (54.18%) groups were significantly increased. Low-field nuclear magnetic resonance analysis showed that the US and US-FT pretreatments increased the free water peak area and promoted water migration,whereas the US-SB pretreatment reduced free water and increased immobilized water. Correlation analysis revealed that a* had an extremely significant positive correlation with the DPPH radical scavenging rate (P<0.01,the same below),while the volume shrinkage rate showed extremely significant negative correlations with volatile oil content,shikimic acid content,and DPPH radical scavenging rate.

Conclusion

Consi-dering both drying efficiency and product quality,ultrasound combined with freeze-thaw pretreatment exhibits the best overall performance,which can shorten drying time,reduce energy consumption,improve star anise color,effectively retain volatile oil,shikimic acid,and antioxidant activity,without increasing volume shrinkage ratio. This technology provides a feasible approach for the green drying and processing of star anise.

目的

探究不同超声波联合预处理对八角微波干燥效率及干制品品质的影响,明确最优预处理方式及其作用机制,为八角绿色、高效、节能干燥加工提供参考依据。

方法

以新鲜八角为原料,设超声波(US)、超声波联合蒸汽漂烫(US-SB)及超声波联合冻融(US-FT)3种预处理方式,以不经任何预处理直接进行微波干燥为对照(CK),系统分析3种预处理对八角微波干燥特性、理化品质及抗氧化活性的影响,并结合微观结构与低场核磁共振技术揭示其作用机制。

结果

3种预处理均能破坏八角细胞结构,增加水分迁移通道。与CK组[干燥时间261.67 min,单位能耗14.60 (kW·h)/kg]相比,US、US-SB和US-FT组干燥时间分别缩短8.92%、46.50%和32.48%,单位能耗分别降低13.08%、50.00%和33.56%。US-SB组干燥效率最高,但其多酚氧化酶(PPO)活性几乎被完全钝化,导致干制品红度值(a*)与CK组无显著差异(P>0.05,下同),且挥发油含量(6.59%)、莽草酸含量(8.20%)及1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率(53.95%)均显著低于CK组(P<0.05,下同)。US组和US-FT组PPO活性显著高于CK组,a*较CK组分别显著提高15.74%和15.64%,色泽更红亮,DPPH自由基清除率较CK组分别显著提高22.28%和25.53%,而挥发油和莽草酸含量与CK组无显著差异。US-FT组体积收缩率(41.88%)与CK组无显著差异,而US组(45.78%)和US-SB组(54.18%)显著提高。低场核磁共振分析结果显示,US和US-FT预处理增加八角自由水峰面积,促进水分迁移;US-SB预处理则导致自由水减少、不易流动水增加。相关分析结果显示,a*与DPPH自由基清除率呈极显著正相关(P<0.01,下同),体积收缩率与挥发油含量、莽草酸含量及DPPH自由基清除率呈极显著负相关。

结论

综合干燥效率与产品品质,超声波联合冻融预处理效果最优,可在缩短干燥时间、降低能耗的同时,改善八角色泽,有效保留挥发油、莽草酸及抗氧化活性,且不增加体积收缩率。该技术为八角绿色干燥加工提供了可行方案。

目的

探究不同超声波联合预处理对八角微波干燥效率及干制品品质的影响,明确最优预处理方式及其作用机制,为八角绿色、高效、节能干燥加工提供参考依据。

方法

以新鲜八角为原料,设超声波(US)、超声波联合蒸汽漂烫(US-SB)及超声波联合冻融(US-FT)3种预处理方式,以不经任何预处理直接进行微波干燥为对照(CK),系统分析3种预处理对八角微波干燥特性、理化品质及抗氧化活性的影响,并结合微观结构与低场核磁共振技术揭示其作用机制。

结果

3种预处理均能破坏八角细胞结构,增加水分迁移通道。与CK组[干燥时间261.67 min,单位能耗14.60 (kW·h)/kg]相比,US、US-SB和US-FT组干燥时间分别缩短8.92%、46.50%和32.48%,单位能耗分别降低13.08%、50.00%和33.56%。US-SB组干燥效率最高,但其多酚氧化酶(PPO)活性几乎被完全钝化,导致干制品红度值(a*)与CK组无显著差异(P>0.05,下同),且挥发油含量(6.59%)、莽草酸含量(8.20%)及1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率(53.95%)均显著低于CK组(P<0.05,下同)。US组和US-FT组PPO活性显著高于CK组,a*较CK组分别显著提高15.74%和15.64%,色泽更红亮,DPPH自由基清除率较CK组分别显著提高22.28%和25.53%,而挥发油和莽草酸含量与CK组无显著差异。US-FT组体积收缩率(41.88%)与CK组无显著差异,而US组(45.78%)和US-SB组(54.18%)显著提高。低场核磁共振分析结果显示,US和US-FT预处理增加八角自由水峰面积,促进水分迁移;US-SB预处理则导致自由水减少、不易流动水增加。相关分析结果显示,a*与DPPH自由基清除率呈极显著正相关(P<0.01,下同),体积收缩率与挥发油含量、莽草酸含量及DPPH自由基清除率呈极显著负相关。

结论

综合干燥效率与产品品质,超声波联合冻融预处理效果最优,可在缩短干燥时间、降低能耗的同时,改善八角色泽,有效保留挥发油、莽草酸及抗氧化活性,且不增加体积收缩率。该技术为八角绿色干燥加工提供了可行方案。

Objective

This study aimed to investigate the effects of different ultrasound-combined pretreatments on the microwave drying efficiency and dried product quality of star anise,identify the optimal pretreatment method and its underlying mechanism,and thereby provide reference for the green,efficient,and energy-saving drying processing of star anise.

Method

Fresh star anise was subjected to three pretreatments as raw materials:ultrasound (US),ultrasound combined with steam blanching (US-SB),and ultrasound combined with freeze-thaw treatment (US-FT),with samples directly subjected to microwave drying without pretreatment served as the control (CK). Effects of the three pretreatments on the microwave drying characteristics,physicochemical quality,and antioxidant activity of star anise were systematically analyzed,and the underlying mechanisms were further elucidated through microstructural observation and low-field nuclear magnetic resonance analysis.

Result

All three pretreatments disrupted the cellular structure of star anise and increased the number of water-migration pathways. Compared with the CK group [drying time of 261.67 min and specific energy consumption of 14.60 (kW·h)/kg],the drying times of the US,US-SB,and US-FT groups were reduced by 8.92%,46.50%,and 32.48% respectively,while their specific energy consumptions decreased by 13.08%,50.00%,and 33.56% respectively. The US-SB group exhibited the highest drying efficiency. However,polyphenol oxidase (PPO) activity was almost completely inactivated,resulting in no significant difference in the redness value (a*) of dried product compared with the CK group (P>0.05,the same below). Moreover,its volatile oil content (6.59%),shikimic acid content (8.20%),and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate (53.95%) were significantly lower than those of the CK group (P<0.05,the same below). PPO activity in the US and US-FT groups was significantly higher than that in the CK group,and the a* values significantly increased by 15.74% and 15.64% compared with CK group respectively,demonstrating a brighter red color;the DPPH radical scavenging rates increased significantly by 22.28% and 25.53% compared with CK group respectively;no significant differences in volatile oil or shikimic acid contents were observed between these two groups and the CK group. The volume shrinkage rate of US-FT group (41.88%) showed no significant difference with the CK group,whereas those of the US (45.78%) and US-SB (54.18%) groups were significantly increased. Low-field nuclear magnetic resonance analysis showed that the US and US-FT pretreatments increased the free water peak area and promoted water migration,whereas the US-SB pretreatment reduced free water and increased immobilized water. Correlation analysis revealed that a* had an extremely significant positive correlation with the DPPH radical scavenging rate (P<0.01,the same below),while the volume shrinkage rate showed extremely significant negative correlations with volatile oil content,shikimic acid content,and DPPH radical scavenging rate.

Conclusion

Consi-dering both drying efficiency and product quality,ultrasound combined with freeze-thaw pretreatment exhibits the best overall performance,which can shorten drying time,reduce energy consumption,improve star anise color,effectively retain volatile oil,shikimic acid,and antioxidant activity,without increasing volume shrinkage ratio. This technology provides a feasible approach for the green drying and processing of star anise.

【研究意义】八角(Illicium verum Hook. f.)是一种兼具经济、食用和药用价值的重要香辛料(Xu et al.,2023Zou et al.,2023王乙淋等,2024陈赛赛等,2025郭向阳,2025),我国南方地区产量占全国80%以上。新鲜八角采收时湿基含水量高达75%左右,采收后若不及时干燥,易发生黑化、霉变及有效成分流失,从而导致严重的采后损失(Ding et al.,2020)。干燥是保障八角品质的关键加工环节。传统自然晾晒与热风干燥虽应用广泛,但存在干燥时间长、产品品质不稳定和香气损失大等问题(Zhang et al.,2025b)。部分从业者甚至违规使用硫磺熏蒸等不当加工方式,严重影响了八角的食用安全与产业升级(Wu et al.,2026)。因此,研发绿色、高效、安全的八角干燥技术具有现实紧迫性。【前人研究进展】近年来,不少学者围绕八角干燥技术开展了较多研究,旨在寻找替代传统自然晾晒和热风干燥的高效方法。微波干燥因加热迅速、节能环保、易于控制等优势,已在八角及其他药食同源植物干燥中受到关注。吴钊龙等(2022)研究表明,间歇微波干燥可显著缩短黄精片干燥时间,但单一微波处理易导致局部过热和品质劣变。李晨曦(2023)优化八角微波热泵联合干燥工艺,指出单一微波干燥存在有效成分损失较大、色泽不均等问题,仅依靠微波技术难以兼顾高效干燥与品质保持。Wu等(2026)比较自然晾晒、热风干燥、真空冷冻干燥、红外干燥和微波干燥对八角品质的影响,发现微波干燥效率最高,但对挥发油的保留效果不如真空冷冻干燥和热风干燥。为了改善干燥效果,引入适宜的预处理技术成为提升微波干燥效率和产品品质的有效策略。在八角加工中,杀青(漂烫)通常被视为干燥前或干燥初期的重要预处理环节,其主要目的在于通过短时热处理或物理作用破坏细胞膜结构、促进水分迁移及调控多酚氧化酶(PPO)等重要酶活性,从而改善干制品色泽并提高干燥效率(Zhang et al.,2026)。金建龙等(2021)以热风温度和杀青/不杀青为变量研究八角热风干燥过程,发现杀青处理可缩短干燥时间,改善干制八角色泽和复水性,但同时会增加皱缩率。Zhang等(2025b)指出,漂烫后干燥是八角加工中延长货架期和利于挥发油提取的重要策略,但不适宜的热处理可能造成挥发油损失、水溶性活性成分溶出及组织结构塌陷。由此可见,八角杀青效果不仅取决于酶失活程度,还与细胞结构破坏、水分迁移、色泽形成及活性成分保留等密切相关。Zhang等(2020)仇徐亮等(2024)Wang等(2024)研究发现,超声波等非热预处理能够保护热敏性成分,却难以显著提升干燥效率。宫许林(2023)研究不同预处理(烫漂、冻融、超声波)对八角热风干燥特性的影响,发现冻融预处理可提高干燥速率,但对八角的色泽改善有限,烫漂预处理虽能钝化酶活性,但导致部分香气成分损失。Zhang等(2026)系统比较了热/非热预处理对八角干燥品质的调控作用,指出热烫处理可抑制酶促褐变,但会造成水溶性莽草酸溶出。超声波、冻融等非热预处理虽不同于传统高温杀青,但可通过空化效应、机械振动或冰晶破坏等方式改变八角细胞结构,提高组织通透性并促进水分扩散,因此也可能产生一定的“类杀青”效果。Zhang等(2025a)比较了超声辅助电解水解冻、超声辅助去离子水解冻、热水漂烫等预处理方式对八角干燥效果的影响,结果发现,超声辅助电解水解冻可通过增强细胞结构破坏、促进水分迁移并调控PPO和过氧化物酶(POD)活性,缩短干燥时间,同时促进八角色泽形成和特征香气成分释放。近年来,联合预处理策略因兼具不同技术优势而受到广泛关注。已有研究表明,超声波联合冻融可实现紫甘蓝脆片的快速脱水和品质改善(虎鑫等,2025),超声波联合漂烫能够显著提高辣椒干燥效率并促进活性成分保留(曹玉丹等,2026)。【本研究切入点】目前,虽然已有超声波、冻融、漂烫等预处理在八角干燥中应用的相关研究,但将超声波分别与蒸汽漂烫、冻融联合应用于八角微波干燥的研究尚未见详细报道,其对八角水分含量及分布、微观结构、有效成分含量及抗氧化活性的影响尚不明确,作用机制亟待阐明。【拟解决的关键问题】比较超声波、超声波联合蒸汽漂烫及超声波联合冻融3种预处理方式对八角微波干燥特性、理化品质及抗氧化活性的影响,并结合低场核磁共振技术与微观结构分析揭示其内在机制,以期为八角绿色、高效、节能干燥加工提供参考依据。
新鲜八角由广西藤县古龙镇泗洲村八角种植基地提供,挑选大小均匀、色泽一致、成熟度相同的八角,清洗干净后备用。采用水分测定仪测得新鲜八角的初始湿基平均含水量为75.60%。
主要试剂:邻苯二酚、磷酸盐缓冲液(国药集团化学试剂有限公司);聚乙烯吡咯烷酮(上海麦克林生化科技股份有限公司);1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除能力检测试剂盒(苏州格锐思生物科技有限公司)。主要仪器设备:Webox-A6型微波干燥机(株洲市微朗科技有限公司)、JG-28K20超声波清洗仪(浙江九工环保设备有限公司)、NMI20型核磁共振成像分析仪(上海纽迈电子科技有限公司)、1510型全波长酶标仪(美国ThermoFisher Scientific公司);CR-10Plus型色彩色差计(日本柯尼卡美能达公司)、XSP-BM-3CB型光学显微镜(上海彼爱姆光学仪器制造有限公司)、S-3400N扫描电镜(日本Hitachi公司)。
根据前期预试验的优化结果,设超声波(US)、超声波联合蒸汽漂烫(US-SB)、超声波联合冻融(US-FT)3个预处理组和1个对照(CK)组。各组别的具体操作流程如下:CK组精确称取500 g八角样品,不经任何预处理直接进行微波干燥;US组精确称取500 g八角样品,置于超声波仪器中,在400 W、40 kHz的条件下处理20 min;US-SB组将经US预处理后的八角样品置于100 ℃蒸锅中进行蒸汽漂烫处理10 min,结束后在室温环境中自然冷却至室温;US-FT组将经US预处理后的八角样品放入冷冻柜中,在-20 ℃条件下冷冻24 h,结束后在室温环境中自然解冻至室温。
将经CK、US、US-SB 及US-FT预处理后的八角样品分别均匀平铺于微波干燥托盘上,单层不重叠为原则,铺放厚度控制在1.5~2.0 cm。预试验结果表明,功率密度过低会导致干燥时间延长、能耗增加,功率密度过高则易造成局部过热、色泽劣变及挥发性成分损失;综合干燥效率与品质保持效果,微波功率密度设定为0.9 W/g。干燥设备运行过程中采用设备自带转盘旋转方式进行加热,干燥过程中每隔20 min取样称重,直至八角的干基含水量≤0.13 g/g结束干燥,试验重复3次。
将预处理后的新鲜八角切为1 mm×1 mm小方块,取约0.5 mm切片,用0.05%甲苯胺蓝染色10 min,于10倍光学显微镜下观察,分析细胞结构变化。
将干燥后的八角样品切成1 mm×1 mm小方块,采用扫描电子显微镜进行观察。样品固定于覆有一层导电胶的样品台上,轻轻按压确保黏附牢固;随后对样品进行喷金处理,在8.0 kV加速电压、350倍放大条件下观察(Liu et al.,2025)。
参照Zhang等(2026)的方法测定八角PPO活性。
采用GB 5009.3—2016《食品安全国家标准 食品中水分的测定》中的直接干燥法测定八角干基含水量,并根据公式进行计算:

公式中,M为八角干基含水量(g/g),mt为干燥至t时刻的八角质量(g),m为八角干物质质量(g)。
根据公式计算八角的干燥速率(吴钊龙等,2023):

公式中,DR为干燥速率[g/(g·min)],Δt为时间间隔(min),MtMt+Δt分别为tt+Δt时刻的八角干基含水量(g/g)。
以去除1 kg八角水分所消耗的电能表示,单位为(kW·h)/kg,由电能表测定(黄纪民等,2023)。
将干燥过程的八角放入低场核磁共振仪专用玻璃试管中,并用CPMG脉冲序列测定T2,序列参数根据盘喻颜等(2021)的方法稍作修改,设为采集次数6次、回波数16000、回波时间0.2 ms。通过CPMG脉冲序列反演得到T2谱图,对各峰面积进行统计。
MRI参数根据盘喻颜等(2021)的方法稍作修改,主频SF1=22 MHz,偏移频率 O1=366645.85 Hz,90°脉冲宽度P1=3200 μs,180°脉冲宽度P2=3200 μs。
将不同预处理后的八角干制品粉碎,采用色差计测定明度值(L*)、红度值(a*)和黄度值(b*)3项颜色指标(覃海波等,2025)。
采用小米置换法测定八角体积,置换介质为小米,根据公式计算体积收缩率(林芳等,2025):

公式中,SR为体积收缩率(%),V0为新鲜八角体积(mm3),Vt为干燥后八角体积(mm3)。
参照Shi等(2021)的方法并略作修改进行测定,根据公式计算八角挥发油含量:

公式中,VO为八角挥发油含量(%),m1为提取所得挥发油质量(g),m2为用于提取的八角质量(g)。
参照Shi等(2021)的方法并略作修改进行测定。称取4.0 g干燥八角,加入200 mL去离子水,置于电热板上缓慢加热至沸腾;随后调小火力保持微回流5 h,直至接收器中挥发油体积不再增加,停止加热。蒸馏结束后,将收集的八角挥发油与烧瓶中的水提液一并转移,用去离子水定容至250 mL。精确移取1 mL上述溶液至100 mL容量瓶中,用去离子水定容至刻度;再吸取1 mL该稀释液至10 mL容量瓶中,用甲醇定容至刻度。采用酶标仪于213 nm波长下测定最终溶液的吸光度。测定前,溶液经0.45 μm滤膜过滤,收集滤液作为待测样液,根据公式计算八角莽草酸含量:

公式中,SA为莽草酸含量(%),c为莽草酸质量浓度(μg/mL),v为提取液体积(mL),k为稀释倍数,m为称取的八角质量(g)。
采用试剂盒测定八角的DPPH自由基清除能力。
每组试验设3次重复,采用SPSS 17.0进行数据统计分析,通过单因素方差分析(One-way ANOVA)结合邓肯多重极差检验比较各组间的差异显著性,采用Origin 2021绘图。
光学显微镜观察结果(图1)显示,CK组八角细胞壁与细胞膜结构完整,细胞排列整齐、轮廓清晰可辨,未受外界物理或化学因素的破坏;US预处理后八角细胞结构出现轻微变形;US-SB和US-FT预处理对细胞结构损伤更为明显,其中US-SB预处理导致细胞严重变形、塌陷,US-FT预处理后细胞虽发生明显变形,但损伤程度相对较轻,细胞结构完整性优于US-SB组。
扫描电子显微镜观察结果(图2)显示,US预处理后细胞孔隙数量增加,孔径明显增大,US-SB和US-FT预处理均导致细胞结构不同程度破损。上述结构变化有利于形成更多的水分迁移通道,为后续水分快速扩散和脱除提供结构基础。
为了阐明八角色泽形成与氧化反应相关的酶学机制,测定PPO活性。由图3可知,不同预处理组八角的PPO活性存在显著差异(P<0.05,下同),US组和US-FT组PPO活性分别为270.67和209.33 U/(mL·min),较CK组分别显著提高111.46%和63.54%;US-SB组PPO活性接近0 U/(mL·min),几乎完全被钝化,显著低于其他各组。
图4可知,与CK组相比,各预处理组八角的干燥时间与单位能耗均显著降低。CK组干燥时间为261.67 min,单位能耗为14.60 (kW·h)/kg;与CK组相比,US组干燥时间缩短8.92%,单位能耗降低13.08%;US-SB组干燥时间缩短46.50%,单位能耗降低50.00%;US-FT组干燥时间缩短32.48%,单位能耗降低33.56%。
图5-A所示,各预处理组的干燥曲线较CK组更为陡峭,其中US-SB组下降速度最快,US-FT组次之。由图5-B可知,八角的干燥过程仅包含加速和降速2个阶段,且以降速阶段为主导。此外,各预处理组的初始干基含水量与CK组相比存在差异,CK组初始干基含水量为3.10 g/g,US组和US-FT组分别增至3.44和3.12 g/g,而US-SB组降至2.89 g/g。
图6表1可知,不同预处理条件下八角的低场核磁共振T2反演谱均存在4个明显的弛豫峰:T21(0.14~0.32 ms)、T22(1.98~5.47 ms)、T23(16.23~55.18 ms)和T24(345.82~1258.32 ms),分别对应结合水、不易流动水、自由水、油脂相关质子。结合水、不易流动水、自由水、油脂相关质子的弛豫峰面积分别记为A21、A22、A23和A24。与CK组相比,各预处理组八角的弛豫峰位置均发生明显移动;US组和US-FT组的各峰整体右移,A23显著增大;相比之下,US-SB组八角的弛豫峰则呈现向左移动的趋势,A23减小,A22则显著增大。各组之间A21无显著差异(P>0.05,下同)。
图7可知,不同预处理条件下,八角在干燥过程中的T2曲线变化趋势基本一致。随着干燥时间的延长,各预处理组的反演谱均出现不同程度的左移。其中,T23和T24对应的信号幅值下降,T22信号幅值呈先升高后下降的变化趋势,T21的信号幅值变化幅度极小。与CK组相比,各预处理组八角的信号幅值下降速度更快,反演谱图左移的速率更为明显,其中US-SB组变化最明显。
图8可知,不同预处理条件下八角中的水分均以自由水为主,其峰面积(A23)占总峰面积的90%以上,其次为不易流动水(A22),结合水(A21)和油脂相关质子(A24)所占比例低。在干燥初期,A23急剧下降,A22和A21呈先上升后下降的变化趋势,A24则持续下降;当干燥进行至终点时,流动性最强的自由水几乎被完全去除,部分不易流动水因未能及时转化为自由水而留存在八角内部,结合最牢固的结合水及油脂相关质子则未被去除。因此,干燥末期八角内部仅含有油脂、结合水和少量不易流动水。
与CK组相比,各预处理组八角中水分的脱除速率明显更快,其中US-SB组峰面积下降最快,US-FT组次之,US组相对较慢。将4个弛豫峰的峰面积相加,得到总峰面积(A),其变化可直接反映八角中总水分和油脂的脱除程度。
图9可知,经拟合分析,在不同预处理条件下,八角的总峰面积与其干基含水量之间呈良好的线性正相关,R2≥0.977。
图10可知,随着干燥进行,在不同预处理条件下八角氢质子密度图像的亮度均逐渐降低,八角中心区域伪彩色图像最先由红绿色转为蓝色,随后蓝色区域向外围扩展。与CK组相比,各预处理组八角伪彩色图像的面积缩减速度明显加快,其中US-SB组图像面积缩小最快,US-FT组次之,US组相对较慢。
图11可知,在b*方面,3个预处理组与CK组均无显著差异;在L*方面,3个预处理组均显著高于CK组(50.86),其中US-SB组最高(57.49),US组(53.04)与US-FT组(54.47)无显著差异;在a*方面,US组和US-FT组较CK组分别显著提高15.74%和15.64%,而US-SB组与CK组无显著差异。
体积收缩率是评价干燥样品原始形状保持程度的关键指标,收缩率越高,表示结构变形越大、形态保持性越差,反之亦然。由图12可知,US组(45.78%)和US-SB组(54.18%)的八角体积收缩率显著高于CK组(42.51%),US-FT组(41.88%)与CK组无显著差异。
图13可知,CK组八角挥发油和莽草酸含量分别为7.81%和9.65%,US组八角干制品挥发油和莽草酸含量分别为7.63%和9.57%,US-FT组分别为7.55%和9.52%,US-SB组分别为6.59%和8.20%;与CK组相比,US组和US-FT组干燥后的八角挥发油和莽草酸含量均无显著差异,而US-SB组2项指标含量均显著降低。
图14可知,US组(80.41%)和US-FT组(82.55%)八角的DPPH自由基清除率较CK组分别显著提高22.28%和25.53%,而US-SB组(53.95%)较CK组显著降低17.96%。
为了更直观地呈现干燥后八角色泽、体积收缩率、生物活性成分及抗氧化活性之间的相关性,采用Pearson相关性分析法对7项指标进行分析,并绘制相关性热图。由图15可知,L*与挥发油含量、莽草酸含量呈极显著负相关(P<0.01,下同);a*与DPPH自由基清除率呈极显著正相关;体积收缩率与挥发油含量、莽草酸含量、DPPH自由基清除率均呈极显著负相关。
本研究对比分析US、US-SB及US-FT预处理组对八角微波干燥特性、微观结构、理化品质及抗氧化活性的影响,结果表明,3种预处理均可有效破坏八角细胞组织结构,提高细胞膜通透性,从而形成更多水分迁移通道,最终显著缩短干燥时间、降低单位能耗、提高干燥速率。其中,US-SB组干燥速率最快,而US-FT组干燥效率与产品品质综合表现最优。
本研究发现,所有预处理组均显著缩短干燥时间、降低单位能耗,其效率排序为US-SB>US-FT>US>CK。这一结果的根本原因在于预处理对八角微观结构的破坏,增强了水分迁移能力。光学显微镜和扫描电子显微镜图像证实,US、US-SB和US-FT预处理均导致八角的细胞发生不同程度的变形、塌陷或孔隙增多,与Wang等(2024)在桑葚、虎鑫等(2025)在紫甘蓝中的观察结果一致。
低场核磁共振分析进一步揭示了预处理对八角水分分布的影响规律。不同预处理条件下八角中水分均以自由水为主(占比>90%)。US预处理通过声波空化效应,不仅物理性地破坏细胞壁,还可能改变细胞壁多糖的结构(Wang et al.,2024),从而降低水分流动的阻力,表现为T2谱图右移和A23显著增大。表明部分结合较弱的不易流动水甚至结合水转化为移动性更强的自由水,为后续快速脱除打下基础。US-FT预处理的增效机制则更为复杂。冷冻过程中形成的胞内冰晶对细胞器及细胞膜造成不可逆的机械损伤(Osae et al.,2024),解冻后细胞结构变得松散,形成了类似海绵的多孔结构。低场核磁共振分析显示,US-FT预处理同样显著增加了A23比例,并增大了其弛豫时间T23,证实水分子活动增强。这种结构变化是US-FT组干燥效率显著优于US组的直接原因。
US-SB预处理实现了最高的干燥效率,其机制是物理损伤与高温物理作用的叠加。蒸汽漂烫使细胞膜蛋白变性,彻底瓦解了细胞室的阻隔作用;同时,高温使细胞内空气和水分急剧膨胀,进一步撕裂细胞结构。虽然低场核磁共振分析显示US-SB组八角的A23下降,但并非初始自由水减少,而是由于剧烈的结构破坏使得水分在干燥前或初始干燥阶段就已迅速大量蒸发,导致在第一次核磁共振测量时,样品中的自由水含量已显著低于其他组。US-SB组的弛豫时间T23显著左移也印证了水分子的生存环境发生剧变,移动性降低,可能是因为细胞结构的严重塌陷反而限制了部分水分的流动性,但并不影响US-SB预处理在微波场中整体脱水效率的优势。
颜色是决定干制品外观品质的关键因素之一。本研究中,US预处理和US-FT预处理八角的a*显著高于CK组,这一变化与其PPO活性的变化密切相关。PPO活性测定结果显示,US组和US-FT组的PPO活性显著高于CK组。酶活性升高的主要原因是超声波通过空化效应破坏细胞结构,促进酶与底物从细胞内释放;此外,超声波引发的机械振动作用还可能诱导酶构象发生变化,从而表现出更高的催化活性(Zhang et al.,2025a)。PPO活性的增强促进了干燥过程中酚类物质的酶促氧化,生成醌类及其聚合产物,从而提升八角的红亮色泽,与Zhang等(2026)的研究结果一致,即热/非热预处理可通过结构改性和酶活性调控协同改善八角干燥品质。相比之下,US-SB组的PPO活性最低,几乎完全被钝化,活性值接近0 U/(mL·min),主要是因为蒸汽漂烫能有效灭活PPO,从而抑制酶促褐变反应(韩彦辉,2023)。因此,US-SB组八角的a*与CK组无显著差异,色泽改善效果不及US组和US-FT组。
在活性成分保留方面,US-FT组展现了显著优势。该预处理既能有效保留挥发油和莽草酸,又能提升抗氧化活性,与虎鑫等(2025)研究紫甘蓝脆片干燥特性得出的结果类似,表明超声波联合冻融预处理能够在提高干燥效率的同时,较好地保护热敏性成分。挥发油和莽草酸并非决定八角抗氧化活性的唯一因素。本研究中,US组和US-FT组的挥发油及莽草酸含量与CK组无显著差异,但DPPH自由基清除率显著高于CK组,说明预处理对抗氧化活性的提升可能更多地与细胞结构破坏后抗氧化物质释放效率提高有关。US预处理的空化效应和US-FT预处理形成的冰晶损伤均可增加细胞膜通透性,促进多酚、黄酮等抗氧化成分从细胞内或结合态结构中释放;同时,US和US-FT预处理缩短了干燥时间,降低了长时间微波干燥过程中活性物质的氧化降解。因此,即使挥发油和莽草酸含量与CK组差异不显著,US组和US-FT组仍表现出更强的DPPH自由基清除能力。后续研究可进一步测定总酚、总黄酮及特征抗氧化成分,以深入阐明八角抗氧化活性变化的物质基础。相比之下,US-SB组虽然干燥效率最高,但其高温处理过程导致了挥发油的挥发与分解,以及水溶性莽草酸的浸出,与Ghosh等(2012)的研究结果一致,即高温漂烫会显著降低水溶性活性成分的保留率。
此外,US-FT组在显著提高八角干燥效率的同时,并未导致其体积收缩率的增加。这可能是由于冻融过程中形成的均匀冰晶对细胞结构起到了支撑作用,减轻了后续干燥过程中的组织塌陷(Osae et al.,2024)。这一特性对于保持八角饱满的外形具有重要意义。
综上所述,US-FT组能够兼顾干燥效率与产品品质,在缩短干燥时间、降低能耗的同时,改善色泽、保留活性成分并提升抗氧化活性,是一种高效、绿色的八角加工技术。本研究仅考察了3种超声波联合预处理对八角微波干燥效率和基础品质的影响,未进一步分析预处理对八角特征香气成分及贮藏稳定性的作用,后续可结合气相色谱—质谱联用、电子鼻及中试生产研究,深入解析US-FT预处理对八角风味轮廓的调控机制并评估其产业化可行性。
综合干燥效率与产品品质,超声波联合冻融预处理效果最优,可在缩短干燥时间、降低能耗的同时,改善八角色泽,有效保留挥发油、莽草酸及抗氧化活性,且不增加体积收缩率。该技术为八角绿色干燥加工提供了可行方案。
