    华盛顿大学蛋白质设计研究所David Baker团队提出一种新的膜蛋白增溶策略。研究人员设计出一类名为WRAPs的蛋白质“外衣”,可贴合并包裹膜蛋白的疏水表面,让原本离不开膜环境的膜蛋白在水溶液中保持稳定。2026年7月2日,相关研究成果发表于《Science》。
    WRAPs全称为Water-solu‐ble RFdiffused Amphipathic Pro‐teins,即水溶性RF扩散两亲性蛋白。所谓“两亲性”,可以理解为一面亲油、一面亲水。WRAPs的疏水面朝内,贴住膜蛋白原本与脂质相互作用的区域;亲水面朝外,与水接触。这样一来,膜蛋白像被包进一件合身外套,疏水表面被遮住,整体就能在水中保持可溶和稳定。这种设计思路的关键,是让WRAPs在膜蛋白疏水区域周围形成屏障,从而在不使用去垢剂的情况下维持其稳定性和水溶性。
    与传统外加增溶试剂不同,WRAPs本身可以通过基因编码。研究人员先根据目标膜蛋白的已知结构,或AlphaFold等工具预测出的结构,设计与其疏水表面匹配的WRAPs;随后把WRAPs与目标膜蛋白一同在大肠杆菌中表达。蛋白质翻译出来后,WRAPs会直接结合膜蛋白,使其不必先进入细胞膜,再被去垢剂提取出来。换句话说,原本需要“从膜里洗出来”的蛋白,现在可以直接以可溶形式进入后续纯化流程。
    研究团队在多类膜蛋白上验证了这一方法,包括β-桶外膜蛋白和多次跨膜螺旋蛋白。预印本阶段的同行评议摘要显示,WRAPs可用于单体和寡聚β-桶外膜蛋白,也可用于螺旋多次跨膜蛋白;包裹后的目标蛋白仍能保留结合功能和酶活性,并表现出更高稳定性。
    这项技术的一个重要应用场景是疫苗研发。梅毒螺旋体外膜蛋白长期被视为潜在疫苗抗原,但这类蛋白难以稳定表达和表征。研究团队将WRAPs用于多种梅毒螺旋体外膜蛋白,获得了可溶版本,为后续抗原结构研究、免疫评价和诊断试剂开发提供了新的入口。
    不过,WRAPs并不意味着去垢剂会立刻退出膜蛋白研究。它更像是为一部分难处理膜蛋白提供了新的实验路线。同行评议意见也提醒,WRAPs可能改变目标蛋白的稳定性和构象动态,因此在研究高度依赖构象变化的受体、通道或转运蛋白时,仍需仔细验证其功能是否真正接近天然状态。
