长期以来,科学家猜测早期生命可能依赖更精简的氨基酸体系,但尚无实验证据。Ile因结构与缬氨酸(Val)、亮氨酸(Leu)相似,在进化序列分析中保守性最低,常被Val替代,因此成为简化氨基酸“字母表”的理想目标。
该研究突破的关键,不在传统的逐基因编辑,而在于“让蛋白质自己学会重写”。研究团队引入蛋白质语言模型(protein language model,一类借鉴自然语言处理思想的生成式AI),对细胞中关键蛋白进行系统性重设计。模型通过学习海量蛋白质序列中的“语法”和“语义”,预测在缺失Ile的情况下,哪些替代氨基酸组合仍能维持蛋白结构与功能。
在此基础上,团队对大肠杆菌的核心生命机器——核糖体——动刀。核糖体承担着将遗传信息翻译为蛋白质的任务,其结构高度保守、对突变极为敏感。研究人员将其中全部382个Ile残基逐一替换为其他氨基酸,并通过AI辅助设计确保替换后的结构仍能正常折叠与运作。这一“全局替换”在传统蛋白工程中几乎不可想象。
更具挑战的是,核糖体的改变只是起点。整个细胞内成千上万的蛋白质都依赖统一的氨基酸编码体系。为此,研究团队通过多轮迭代设计与筛选,逐步构建出一个“去Ile”的蛋白质网络,使细胞整体代谢与生长过程能够自洽运行。最终获得的工程菌株,不仅能够存活,还表现出相对稳定的生理状态。
从科学意义上看,这项工作触及了生命系统的一个根本问题:氨基酸种类的“20种限制”究竟是化学必然,还是演化偶然?如果生命可以在19种甚至更少的氨基酸条件下运行,那么天然生物的组成规则,或许只是众多可行解中的一种。
当然,这一体系仍处于早期阶段。去除一种氨基酸后,细胞在极端环境下的适应性、长期进化稳定性等问题仍有待评估。但可以预见的是,随着模型能力的提升与实验技术的迭代,未来“可编程生命”的设计空间将被进一步打开。
